Aim of the study
In Nigerian folk medicine, water infusion of the dried seeds of Hunteria umbellata (K. Schum.) Hallier f. has a reputation for the local management of obesity and hyperlipidaemia. The present study is aimed at evaluating the anti-obesity and antihyperlipidaemic activities as well as the underlying mechanisms of action of the aqueous seed extract of Hunteria umbellata (HU) in normal, triton-induced, and olive oil-induced hyperlipidaemic rats.Materials and methods
Normal and olive oil-induced hyperlipidaemic, and triton-induced hyperlipidaemic rats were pre-treated with single, daily oral administration of 10 ml/kg of distilled water, 20 mg/kg of simvastatin, 50 mg/kg, 100 mg/kg and 200 mg/kg of HU in 10 ml/kg of distilled water for 28 days and 24 h. The effects of these drugs on % body weight change, feeding pattern, serum lipids, coronary artery risk index (CRI) and atherogenic index (AI) and Lee's index (LI) were investigated.Results
Oral pre-treatment with simvastatin and graded oral doses of HU significantly (p < 0.05) reduced the weight gain pattern and caused dose related (p < 0.05, p < 0.01 and p < 0.001) reductions in the serum lipids, CRI, AI and LI. Also, HU pre-treatment significantly improved triton-induced hepatic histological lesions.Conclusions
Results of this study showed that HU has both anti-obesity and antihyperlipidaemic effects which may partly be mediated via inhibition of intestinal lipid absorption and de novo biosynthesis of cholesterol. Thus, the results justify the ethnopharmacological use of the extract in the management of obesity and hyperlipidaemia. 相似文献2.

Polygonum aviculare L. has been used in traditional Korean medicine to treat obesity and symptoms associated with hypertension. The effectiveness or mechanism of Polygonum aviculare L. ethanol extract (PAE) on atherosclerosis disease has not been examined experimentally. This study investigated the protective effect of PAE in atherosclerotic mice.Materials and methods
ApoE KO mice were fed a Western diet (WD) alone or with PAE or a statin for 12 weeks, followed by analysis of bodyweight, serum lipid levels, and blood pressure. Staining of the aorta and adipose tissue, expression levels of adhesion molecules, and the MAPK pathway were also examined. Cell viability, NF-κB activity, and protein levels of adhesion molecules were assessed in vitro.Results
ApoE KO mice fed PAE (50 and 100 mg/kg) or statin (10 mg/kg) gained less body weight, and has less adipose tissue and lower serum lipid levels and blood pressures than the WD group. Aorta ICAM-1, VCAM-1, and NF-κB levels were decreased by PAE in a dose-dependent manner, consistent with the in vitro observations. PAE and statin decreased atherosclerotic plaque and adipocyte size versus the WD group. Furthermore, PAE decreased phosphorylation of MAPK pathway components in the aorta of PAE-treated mice, suggesting that PAE's anti-atherosclerotic effects are mediated via a MAPK pathway-dependent mechanism.Conclusions
PAE may protect against the development of atherosclerotic disease. The beneficial effects are associated with lowering bodyweight, serum lipids, blood pressure, adhesion molecular protein levels, atherosclerotic plaque, and adipocyte size, involving the MAPK pathway. 相似文献3.

Aim of the study
Seeds of Securigera securidaca are used for the treatment of disorders such as hyperlipidemia, diabetes, and epilepsy in Iranian folk medicine. The possible hypolipidemic and antioxidative effects of hydroalcoholic extract of S. securidaca seeds as well as the effect of the extract on vascular reactivity were investigated in hypercholesterolemic rats.Materials and methods
High-fat fed wistar rats received orally different doses of the extract for 20 days. At the end of the experiment vein blood and liver were collected to measure the lipid profile, lipid peroxidation, and antioxidative enzyme activities. The thoracic aorta was excised and used for isolated vessel preparation and histological study.Results
The extract produced significant reductions (p < 0.05) in the level of low-density lipoprotein (LDL) and triglyceride with concomitant reduction in lipid deposition in the liver. The extract also suppressed markedly (p < 0.001) the hypercholesterolemia-induced elevation of malondialdehyde levels both in serum and liver. In hypercholesterolemic rats the endothelium-dependent vasodilatation was improved significantly (p < 0.05) by 100 mg/kg/day of the extract. However, in histological study no atherosclerotic lesion was observed.Conclusion
These results suggest that S. securidaca seed in addition to decrease lipid levels and peroxidation, is able to improve vascular endothelium-dependent relaxation in hypercholesterolemia. 相似文献4.

Aim of the study
To investigate the effects of centipede acidic protein (CAP) on atherosclerotic rats and the mechanisms involved.Materials and methods
Male Sprague–Dawley rats were randomly divided into five groups: control, atherosclerotic, low-dose CAP (L-CAP), high-dose CAP (H-CAP) and simvastatin group (n = 12 in each group). Atherosclerotic model was established by a single dose of vitamin D3 and an atherogenic diet. Rats of H-CAP and simvastatin groups simultaneously received CAP or simvastatin daily for 6 weeks. At the completion of the experiment, the changes in lipid profile, hemorrheology, nitric oxide (NO), endothelin-1 (ET-1), malondialdehyde (MDA) and superoxide desmutase (SOD) were measured, and the histological changes in aorta and liver were observed.Results
Treatment of atherosclerotic rats with either low or high doses of CAP led not only to significant decreases in plasma total cholesterol, triglyceride, low density lipoprotein and increase in plasma high density lipoprotein, but also to improvement of the hemorrheologic abnormalities. On the other hand, CAP suppressed the lipid peroxidation, regulated the levels of ET-1 and NO. From the histopathological examination, treatment with CAP ameliorated the pathological changes in thoracic aorta and liver in atherosclerotic rats.Conclusions
These results suggest that CAP significantly suppress the development of atherosclerosis, improves the hemorrheological disturbances and histopathological changes in the atherogenic diet fed rat model. These effects may partly attribute to reverse of dyslipidemia, inhibition of lipid peroxidation, regulation of NO and ET-1 system. 相似文献5.

Aim of study
Paeonol, a major phenolic component of Moutan Cortex, is traditionally used as a Chinese herbal medicine in various diseases including hepatitis. Evidence shows that paeonol has anti-inflammatory, anti-tumor, and anti-atherosclerosis effects. However, the effect of paeonol on alcoholic liver injury remains obscure. The present investigation was designed to determine the effects of paeonol on alcohol-induced hepatic injury in mice.Materials and methods
The degree of alcoholic liver injury was evaluated biochemically by measuring serum markers and pathological examination. Real-time PCR and ELISA methods were used to check the expression of cytokines. Western blotting was used to check CYP 450 expression.Results
Treatment with paeonol significantly attenuated the level of serum aminotransferase, reduced the severe extent of hepatic cell damage, steatosis, and the infiltration of inflammatory cells in a model of alcoholic liver injury (P < 0.05). Interestingly, paeonol markedly decreased hepatic mRNA expression of lipogenic genes (P < 0.05) while had no effect on protein expression of hepatic CYP2E1. Furthermore, paeonol significantly decreased serum and tissue inflammatory cytokine levels, tissue lipid peroxidation, neutrophil infiltration and inhibited the apoptosis of hepatocytes (P < 0.05). Kupffer cells isolated from ethanol-fed mice produced high amounts of tumor necrosis factor alpha, whereas Kupffer cells from paeonol treatment ethanol-fed mice produced less tumor necrosis factor alpha (P < 0.05).Conclusions
These findings suggest that paeonol may represent a novel, protective strategy against alcoholic liver injury by attenuating hepatic steatosis, inflammatory response and apoptosis. 相似文献6.

Deer antler, traditionally used as a tonic and valuable drug in oriental medicine, has been considered to possess bone-strengthening activity and effectively used in bone diseases therapy.Aim of the study
The present study was designed to investigate therapeutic effect of antler extract on avascular necrosis of the femoral head (ANFH) induced by corticosteroids in rats.Materials and methods
Rats were intragluteally injected with dexamethasone at 50 mg/kg twice per week for 6 weeks to induce ANFH. Then the rats were treated with antler extract by oral gavage at 200 mg/kg, 400 mg/kg and 800 mg/kg once per day for 60 days. The concentration of hydroxyproline and hexosamine in serum was measured and the ultrastructure of femoral head was examined. In vitro, effect of the drug-containing serum of antler extract on proliferation and differentiation of primary osteoblasts were investigated by MIT assay, ALP activity assay and cell cycle analysis.Results
After treatment with antler extract, the degree of necrosis induced by dexamethasone was significantly reduced, hydroxyproline was significantly decreased, and hexosamine and the ratio of hexosamine/hydroxyproline were significantly increased. The drug-containing serum of antler extract promotes osteoblastic proliferation through regulation of cell cycle progression.Conclusions
The results suggest that antler extract has a positive curative effect on ANFH by promoting osteoblastic proliferation. 相似文献7.

The aim of the present study was to evaluate the effect of methanolic extract of the bark of Ficus amplissima (FAB) in streptozotocin-induced diabetic rats.Materials and methods
Oral administration of FAB bark at the doses of 50 mg/kg, 100 mg/kg and 150 mg/kg was studied in normal, glucose-loaded and STZ—induced diabetic rats.Results
The three doses caused significant reduction in blood glucose levels in all the models. The effect was more pronounced in 50 mg/kg and 100 mg/kg than 150 mg/kg. FAB also showed significant increase in serum insulin and body weight. The glycogen content in liver, skeletal muscle, total protein contents were markedly increased and marker enzymes of hepatic function of STZ-induced diabetic rats while there was significant reduction in the levels of serum triglyceride and total cholesterol. FAB also showed significant anti-lipid peroxidative effect in the pancreas of STZ-induced diabetic rats. The anti-diabetic effect of FAB was compared with glibenclamide, a well known hypoglycemic drug. Histological analysis showed the regenerative effect of FAB on the β-cells of diabetic rats.Conclusion
Results of this experimental study indicated that FAB possessed anti-diabetic and antioxidant activities. Hence it could be used as a natural source of antidiabetic (Type-I) and antioxidant drug. 相似文献8.

The ethanol extract of leaves of Leucas cephalotes was administered (150, 300 and 450 mg kg−1 bw) to diabetes induced (IDDM and NIDDM) rats and carbohydrate, lipid, antioxidant, urea and creatinine profiles were assessed.Results
All the three doses of extract decreased plasma glucose and lipid profiles and, improved the antioxidant status of both types of diabetic rats. The extract administration improved hepatic glycogen content and hexokinase activity, decreased glucose-6-phosphatase activity, blood urea, creatinine contents and decreased lipid peroxidation in diabetic rats. Of the three doses used, 450 mg kg−1 bw dose was found to be more potent in its effects comparable to those of glibenclamide and metformin.Conclusion

The effects of TSPN on apoptosis and expressions of caspase-1, caspase-3 and caspase-8 were studied after cerebral ischemia for 2 h followed by reperfusion for 46 h in rats.Materials and methods
Rats were subjected to transient middle cerebral artery occlusion (MCAO) model using the intraluminal thread. TSPN was administered intraperitoneally at 5 min before and 12 h, 24 h and 36 h after MCAO, respectively.Results
TSPN (at the dose of 25 mg/kg) significantly attenuated TUNEL-positive cells and reduced the expression of caspase-1 and caspase-3 compared to the model group, while it had no obvious effect on the expression of caspase-8.Conclusions

The aim of the current study was to validate and explore the folk use of Asteracantha longifolia Nees. (AL) (Leaf part) on pharmacological grounds using haloperidol induced iron deficiency anemia for the assessment of erythropoietic activity.Materials and methods
Determination of iron in plant extracts was carried out using spectrophotometric method. Plant extract was obtained from crude drugs using extraction with ethanol. In vivo study, haloperidol induced iron deficiency anemia model was used in experimental studies.Results
An administration of ethanolic extract of AL at the doses of 100 mg/kg and 200 mg/kg body weight, i.p., demonstrated a significant (P < 0.05) increase in erythrocyte count, haemoglobin count, serum iron and serum protein etc. This effect may be due to the presence of iron (622 μg/50 mg) in extract estimated by spectrophotometric method.Conclusion
An ethanolic extract of AL effectively restored the hematological parameters, serum iron and serum protein and normalized the microcytic (smaller in size), anisocytosis (disturbed shape) and hypochromic RBCs. These observations could justify the inclusion of this plant in the management of iron deficiency anemia due the presence of iron and other constituents as flavonoids, terpenoids, steroids, lupeol and betulin. 相似文献11.

To evaluate the antidiabetic activity of aqueous and ethanolic extracts of grains of Paspalum scrobiculatum Linn. (Poaceae) in alloxan induced diabetic rats.Materials and methods
Aqueous and ethanolic extracts (250 and 500 mg/kg body weight), were administered orally to male Wistar albino rats. Alloxan monohydrate was used to induce diabetes mellitus. Total phenolic content was estimated in the extracts. The parameters studied included oral glucose tolerance test, fasting blood glucose, serum insulin and glycated haemoglobin levels, liver glycogen content, serum lipid profile, and changes in body weights.Results
In oral glucose tolerance test, reduction of fasting blood glucose levels took place from 60 min of extract administration. The extracts produced a dose-dependent fall in fasting blood glucose (FBG). After 15 days of treatment with extracts the maximum reduction in FBG (35.14%) was observed in diabetic rats treated with ethanolic extract 500 mg/kg dose. A significant increase in serum insulin level was observed in the treated rats. Serum lipid levels were reversed towards near normal and a control in the loss of body weight was observed in treated rats as compared to diabetic control. The extract treatment also showed a significant increase in the liver glycogen and a significant decrease in glycated haemoglobin levels. The results demonstrate that Paspalum scrobiculatum possesses significant antidiabetic activity in diabetic rats.Conclusion
The results suggest that Paspalum scrobiculatum has antidiabetic activity, thereby justifying its traditional claim and augmenting it into the present day systems of medicine. 相似文献12.

Diabetes mellitus is a major metabolic disorder affecting a huge population all over the world. Artemisia species have been extensively used for the management of diabetes in folkloric medicine. The present study is designed to investigate the antidiabetic and antihyperlipidemic effects of aeriel parts of Artemisia indica.Materials and methods
Hydromethanolic crude extracts, chloroform, ethyl acetate and n-butanol fractions of aerial parts of Artemisia indica were tested for their antidiabetic potential in Streptozotocin (STZ) (50 mg/kg, i.p.) induced diabetic Sprague-Dawley rats. Blood glucose level, body weight, serum lipid profile and activities of liver enzymes were determined. The extracts were further subjected to preliminary phytochemical analysis.Results
A daily oral dose of hydromethanolic crude extracts (200 and 400 mg/kg b.w.) and chloroform fraction (200 mg/kg b.w.) of Artemisia indica for 15 days showed a significant reduction in blood glucose level which was comparable to that of the standard antidiabetic drug, glibenclamide (500 μg/kg, p.o.). Artemisia indica extracts also showed reduction in total cholesterol, triglycerides and low density lipoproteins as well as serum creatinine level, serum glutamate pyruvate transaminase (SGPT), serum glutamate oxaloacetate transaminase (SGOT) and alkaline phosphatase (ALP) in diabetic rats.Conclusion
According to the results Artemisia indica possesses hypoglycemic, antihyperlipidemic and valuable effects on liver and renal functions in diabetic rats, which seems to validate its traditional usage. 相似文献14.

The radix of Acorus calamus L. (AC) is widely used in the therapy of diabetes in traditional folk medicine of America and Indonesia, and we previously reported the insulin sensitizing activity of the ethyl acetate fraction of AC (ACE).Aim of the study
To investigate the insulin releasing and alpha-glucosidase inhibitory activity of ACE in vitro and in vivo.Materials and methods
Insulin releasing and alpha-glucosidase inhibitory effects of different fractions from AC were detected in vitro using HIT-T15 cell line and alpha-glucosidase enzyme. Furthermore, effects of ACE orally on serum glucose were detected in fasted and glucose/amylum challenged normal mice.Results
AC and ACE increased insulin secretion in HIT-T15 cells as gliclazide did. As in vivo results, ACE (400 and 800 mg/kg) significantly decreased fasting serum glucose, and suppressed the increase of blood glucose levels after 2 g/kg glucose loading in normal mice. In addition, ACE as a mixed-type inhibitor inhibited alpha-glucosidase activity in vitro with an IC50 of 0.41 μg/ml, and 100 mg/kg of it clearly reduced the increase of blood glucose levels after 5 g/kg amylum loading in normal mice.Conclusions
Apart from its insulin sensitizing effect, ACE may have hypoglycemic effects via mechanisms of insulin releasing and alpha-glucosidase inhibition, and thus improves postprandial hyperglycemia and cardiovascular complications. 相似文献15.

Artemisia absinthium L. has long been used as traditional herbal medicine in China, Europe and Pakistan for the treatment of gastric pain, cardiac stimulation, to improve memory and for the restoration of declining mental function.Aim of the study
The present study was designed to investigate the potential protective effects of Artemisia absinthium on cerebral oxidative stress and damage as well as behavioral disturbances induced by cerebral ischemia and reperfusion injury in rats.Materials and methods
Focal ischemia and reperfusion were induced by middle cerebral artery occlusion (MCAO) for 90 min, followed by 24 h reperfusion. MCAO led to significant rise in infarct size and lipid peroxidation, and depletion in glutathione content, superoxide dismutase and catalase activity in brain. Further, behavioral deficits like motor incoordination and impairment of short-term memory were also significantly impaired by MCAO as compared with sham group.Results
The brain oxidative stress and damage, and behavioral deficits were significantly attenuated by pre-treatment with the methanol extract of Artemisia absinthium (100 mg/kg and 200 mg/kg, p.o.).Conclusion
These findings suggested that Artemisia absinthium is neuroprotective and may prove to be useful adjunct in the treatment of stroke. 相似文献16.
Aim of the study
The aim of the study was to evaluate the antihyperglycaemic activity of aqueous (PPSB-AQE) and petroleum ether (PPSB-PEE) extract of stem bark Pongamia pinnata in alloxan induced diabetic mice.Materials and methods
Diabetes was induced in mice by alloxan (80 mg/kg, i.v.). After acute and subacute treatment serum glucose was determined. OGTT was performed in PPSB-PEE pretreated animals.Results
PPSB-PEE (25, 50, 100, 200 and 400 mg/kg) showed significant reduction in serum glucose level in acute and subacute studies. The antihyperglycaemic effects of PPSBPE (100, 200 and 400 mg/kg) showed onset at 2 h and peak effect at 6 h and the effect was sustained until 24th h with 400 mg/kg. In subacute study, antihyperglycaemic effect was observed on 21st day. In PPSBPE treated mice the body weight was not reduced in contrast to that in vehicle group. In OGTT, increased glucose utilization was observed.Conclusions
It is concluded that PPSB-PEE but not PPSB-AQE showed antihyperglycaemic activity. 相似文献17.

Fresh juice of Cyanodon dactylon known as ‘durva’ grass is employed in India as a rejuvenator and for wound healing.Aim of the study
To validate the traditional use of the herb through evaluation of DNA protective activity in vitro and immunomodulatory activity in vivo.Materials and methods
Fresh juice of the grass was prepared as indicated for use in traditional medicine and standardized for solid content. Its total phenol content was estimated by Folin–Ciocalteau method. Freshly prepared juice was investigated for its effect on doxorubicin-induced DNA damage in vitro. Its immunomodulatory activity was tested on balb/c mice by the humoral antibody response which was determined by haemagglutination antibody titer and spleen cell assay.Results
Fresh juice of Cyanodon dactylon of 1.46% (w/w) solid content had a phenolic content of 47 ± 0.33 mg/kg GAE. At doses equivalent to 50, 100 and 200 mg total solids/kg body weight the juice protected human DNA against doxorubicin-induced DNA damage as demonstrated in DNA spectral studies, where the ratio of absorbance of DNA at 260 and 280 nm in samples pretreated with the juice was 1.66, 1.53 and 1.63 respectively, while it was 1.37 for DNA treated with doxorubicin only. This indicates nucleic acid purity in the Cynodon dactylon treated samples. Oral administration of the juice at 250 and 500 mg/kg in balb/c mice increased humoral antibody response upon antigen challenge, as evidenced by a dose-dependent, statistically significant increase in antibody titer in the haemagglutination antibody assay and plaque forming cell assay.Conclusions
The present report demonstrated the DNA protective activity and immunomodulatory property of the fresh juice of Cynodon dactylon validating the traditional use of the herb as a ‘rasayana’ in ayurvedic system of medicine. 相似文献18.

Aims of the study
Fatty liver is the most common cause of abnormal liver function tests. We investigated the effect and its underlying mechanism of pomegranate flower (PGF), a traditional antidiabetic medicine, on fatty liver.Materials and methods
At the endpoint of treatment of male Zucker diabetic fatty (ZDF) rats with PGF extract (500 mg/kg, p.o. × 6 weeks), liver weight index, hepatic lipid contents (enzymatic colorimetric methods) and droplet accumulation (Oil Red O staining) were determined. Gene profiles (RT-PCR) were analyzed in the liver of ZDF rats and in human liver-derived HepG2 cell line.Results
PGF-treated ZDF rats showed reduced ratio of liver weight to tibia length, hepatic triglyceride contents and lipid droplets. These effects were accompanied by enhanced hepatic gene expression of peroxisome proliferator-activated receptor (PPAR)-alpha, carnitine palmitoyltransferase-1 and acyl-CoA oxidase (ACO), and reduced stearoyl-CoA desaturase-1. In contrast, PGF showed minimal effects on expression of genes responsible for synthesis, hydrolysis or uptake of fatty acid and triglycerides. PGF treatment also increased PPAR-alpha and ACO mRNA levels in HepG2 cells.Conclusion
Our findings suggest that this Unani medicine ameliorates diabetes and obesity-associated fatty liver, at least in part, by activating hepatic expression of genes responsible for fatty acid oxidation. 相似文献19.

To investigate the antidiabetic action of the aqueous extract of Cucumis trigonus fruits in the different models of rats.Materials and methods
The antidiabetic activity of aqueous extract of Cucumis trigonus fruit was evaluated by using normal and streptozotocin-induced-diabetic rats. The acute effect of aqueous extract was evaluated by administering 500 mg/kg p.o. to normoglycemic rats. In the chronic model, the aqueous extract was administered to normal and STZ-induced-diabetic rats at dose of 500 mg/kg p.o. per day for 21 days. Blood glucose levels and body weights were monitored at specific intervals and different biochemical parameters were also carried out.Results
The statistical data indicated the significant increase in the body weight, liver glycogen and serum insulin level and decrease in the blood glucose, glycosylated hemoglobin levels, total cholesterol and serum triglycerides. HDL cholesterol level was significantly increased when treated with the extract.Conclusion
The aqueous fruit extract of Cucumis trigonus has had beneficial effects in reducing the elevated blood glucose level and lipid profile of STZ-induced-diabetic rats. 相似文献20.
