目的:改进2015年版中国药典土茯苓现有质量标准的含量测定方法。方法:通过改进供试品溶液的处理过程,优化操作步骤。结果:色谱条件为VP-ODS(250 mm×4.6 mm,5μm)色谱柱;以甲醇-0.1%冰醋酸溶液(39∶61)为流动相;检测波长为291 nm。改进方法的落新妇苷平均加样回收率为99.3%,RSD=1.1%(n=9)。落新妇苷进样量在0.047~9.400μg的范围内线性关系良好,测定结果与药典方法比较明显提高。结论:改进后的方法减少了实验过程中落新妇苷的损失,使结果更加准确,且操作较原方法简便。  相似文献

目的:建立微波协助提取法提取土茯苓中有效成分落新妇苷含量测定方法,比较微波协助提取法与药典法提取落新妇苷优势。方法:采用Dikma Diamonsil C18色谱柱(4.6 mm×150 mm,5μm),流动相为甲醇-0.1%冰醋酸水溶液(39∶61),检测波长291 nm,柱温30℃,流速1 mL.min-1。结果:微波提取时间10 min,提取温度60℃,提取溶剂60%乙醇。落新妇苷在0.04~0.8μg呈良好线性关系(r=0.999 9)。平均回收率99.80%(n=6)。结论:微波提取法较药典法提取落新妇苷更简便快速、结果准确,为一种符合环保绿色理念的含量测定方法,可用于检测土茯苓饮片中有效成分落新妇苷含量。  相似文献

目的:研究超声法提取土茯苓中落新妇苷及总黄酮的最佳工艺条件。方法:采用正交试验法研究落新妇苷和总黄酮的提取工艺,其中落新妇苷含量用HPLC法测定,总黄酮含量用紫外分光光度法测定。结果:土茯苓中落新妇苷最佳提取工艺为加入甲醇30mL,超声提取15min,超声3次。总黄酮的最佳提取工艺为料液比1∶50,加入50%乙醇,超声提取60min。结论:所建立的方法简便、准确、重复性好,可用于土茯苓药材中落新妇苷及总黄酮的含量测定及其质量控制。  相似文献

高效液相色谱法测定土茯苓药材中7种活性成分的含量   总被引:1,自引:0,他引:1  
建立同时测定土茯苓中(-)-表儿茶素、5-O-咖啡酰基莽草酸、新落新妇苷、落新妇苷、新异落新妇苷、异落新妇苷和黄杞苷含量的高效液相色谱法。采用Agilent Zorbax SB-C18色谱柱(250 mm×4.6 mm,5μm),柱温35℃,流动相为乙腈(B)-0.05%磷酸(A)梯度洗脱,流速1.0 m L·min-1,检测波长230 nm,对土茯苓、短柱肖菝葜、华肖菝葜药材进行分析。测定的84份土茯苓药材中(-)-表儿茶素、5-O-咖啡酰基莽草酸、新落新妇苷、落新妇苷、新异落新妇苷、异落新妇苷和黄杞苷的含量范围分别为trace~1.381,trace~9.913,trace~3.673,0.670 6~27.08,trace~1.181,trace~4.833,trace~2.754 mg·g-1。用建立的方法对土茯苓常见混淆品短柱肖菝葜、华肖菝葜进行分析,结果显示短柱肖菝葜、华肖菝葜药材中(-)-表儿茶素的含量范围分别为0.011 63~0.060 07 mg·g-1和0.015 83~0.085 85 mg·g-1;而其他6种成分均未检测到。该方法简便、准确,可作为土茯苓药材的质量控制方法。短柱肖菝葜和华肖菝葜的成分显著不同于土茯苓,不宜与土茯苓混用。  相似文献

目的:建立大鼠体内落新妇苷的SPE-HPLC分析方法,通过复方银屑灵与单味土茯苓中落新妇苷药代动力学参数的比较,探讨配伍对落新妇苷大鼠体内药代动力学行为的影响。方法:雄性SD大鼠分别灌胃银屑灵、土茯苓提取物后,于不同时间眼眶静脉丛采血,固相萃取(SPE)法处理。采用反相高效液相色谱法,以甲醇-乙腈-0.05%甲酸为流动相梯度洗脱;Phenomenex C18色谱柱,柱温24℃,流速0.8 mL.min-1。结果:落新妇苷在0.266~53.1 mg.L-1线性关系良好(r=0.996);提取回收率为79.0%~89.1%;用非隔室模型估算药物动力学参数t1/2,Cmax,AUC0-t,AUC0-∞,MRT,各参数具有统计学意义。结论:该法可用于落新妇苷的药代动力学研究;配伍可提高落新妇苷在大鼠体内的生物利用度。  相似文献

目的:考察断面红棕色及类白色的土茯苓醇提液是否具有抗炎作用?并探讨哪种断面颜色土茯苓抗炎效果更佳。方法:小鼠以尾静脉注射伊文思蓝,二甲苯涂腹部致炎,以腹部染料的渗出量,进行急性(渗透型)炎症抗炎作用的评价;以棉球肉芽肿法,进行慢性(增殖型)炎症抗炎作用的评价;大鼠液体石蜡致炎实验对炎性物质NO释放程度进行评价。结果:二甲苯致小鼠腹部毛细血管通透性影响试验中,断面红棕色及类白色的土茯苓(7g/kg)均能减少染料的渗出量,抑制率分别为21%和32%。而对棉球肉芽肿试验中,断面红、白二色的土茯苓均能减少肉芽肿的增生,二者高剂量组(10g/kg)抑制率分别为25%和24%,低剂量组(7g/kg)抑制率分别为37%和25%。在大鼠液体石蜡致炎试验中,断面红棕色及类白色的土茯苓(7g/kg)均能减少血清中NO的产生,抑制率分别为34%和62%。结论:断面红、白二色的土茯苓醇提液均具有抗炎作用。在急性(渗透型)炎症中断面类白色土茯苓组的抗炎效果要优于断面红棕色土茯苓组;在慢性(增殖型)炎症中断面红棕色土茯苓组抗炎效果要优于断面类白色土茯苓组。  相似文献

目的:确立土茯苓药材中阿魏酸、落新妇苷、白藜芦醇三种指标成分的含量。方法:应用高效液相色谱法(HPLC)测定,色谱条件为ODS-C18色谱柱(5μm,150x4.6mm),流动相为乙腈-0.05%磷酸水溶液(18:82),流速1.0mL/min,测定波长290nm。结果 :阿魏酸、落新妇苷、白藜芦醇分别在0.02088~0.1044ug、0.4284~2.142ug、0.0432~0.216ug范围内线性关系良好;样品中阿魏酸、落新妇苷、白藜芦醇评价含量分别为0.05964mg/g、0.01455mg/g、0.01839mg/g。结论:该方法操作简单、准确,可作为土茯苓质量控制方法。  相似文献

目的:建立PR-HPLC同时测定金刚藤中落新妇苷、新异落新妇苷、槲皮素-3-O-α-L-鼠李糖苷、黄杞苷4个黄酮类成分的含量。方法:Hypersil C18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈-0.04%磷酸水溶液,检测波长290 nm,流速0.7mL·min-1;分别采用外标测定法(以落新妇苷、新异落新妇苷、槲皮素-3-O-α-L-Rha、黄杞苷为对照品),一测多评法(以落新妇苷为内标),测定金刚藤中4个黄酮类化合物的含量。结果:外标法测定4个黄酮类成分在标准曲线范围内均呈现良好线性关系(r>0.999 9);平均加样回收率分别为:落新妇苷100.8%、新异落新妇苷100.3%、槲皮素-3-O-α-L-Rha101.4%、黄杞苷101.4%,RSD分别为1.8%,2.0%,1.4%,1.6%。一测多评法,新异落新妇苷、槲皮素-3-O-α-L-Rha、黄杞苷相对落新妇苷对照品的校正因子f分别为0.726,0.264,0.801,RSD分别为0.62%,0.18%,0.66%。两种方法测得的4个黄酮化合物含量相对偏差无显著差异。结论:本实验建立HPLC外标法同时测定金刚藤中4个黄酮化合物含量简便、准确。同时以落新妇苷对照品为内标的一测多评法测得金刚藤中4个黄酮化合物的含量与外标法没有显著性差异,可替代外标法用于金刚藤的质量分析,为全面评价金刚藤药材的质量提供了依据。  相似文献

目的:建立UPLC-MS法同时测定清热凉血方中落新妇苷、梓醇、绿原酸、没食子酸、对香豆酸5种成分的含量。方法:采用Waters Acquity BEH C_(18)(100 mm×2.1 mm,1.7μm)色谱柱;流动相为0.5%甲酸溶液-乙腈,梯度洗脱;ESI负离子扫描模式检测。结果:5种化合物在各自浓度范围内线性关系良好(R~2≥0.9990),平均加样回收率为99.59%~103.75%。清热凉血方中含量较高的成分是没食子酸、梓醇、落新妇苷;不同批次清热凉血方浸膏中各成分含量差异较小。结论:该方法灵敏度高、专属性强、稳定可靠、重复性良好,可用于清热凉血方的质量控制。  相似文献

目的:克隆金银花类药用植物4-二磷酸胞苷-2-C-甲基赤藓糖激酶(4-diphosphocytidyl-2-C-methyl-D-erythritol kinase,IspE)和4-羟基-3-甲基-2-邻苯基二磷酸还原酶(4-hydroxy-3-methylbut-2-enyl diphosphate reductase,IspH)基因,并对其基因序列、蛋白特性和转录活性进行分析、比较.方法:从忍冬Lonicera japonica转录组测序结果中分析获得了IspE,IspH基因.分别以忍冬、红白忍冬L.japonica var.chinensis、红腺忍冬L.hypoglauca和水忍冬L.dasystyla新鲜花蕾为材料,利用RT-PCR技术克隆获得了4种金银花类药用植物IspE和IspH基因的全长cDNA.运用生物信息学分析软件,预测编码蛋白的结构和功能,并通过RT-PCR检测IspE和IspH在忍冬、红白忍冬、红腺忍冬、水忍冬花蕾中的转录情况.结果:金银花类药用植物IspE基因含有1个完整的开放阅读框,长度为1 221 bp,编码406个氨基酸;IspH含有一个完整的开放阅读框,长度为1 380 bp,编码459个氨基酸.IspE和IspH均为非分泌蛋白,均定位于叶绿体中.RT-PCR分析结果表明在忍冬、红腺忍冬和水忍冬的花蕾中IspE,IspH基因的转录水平没有显著差异,但红白忍冬花蕾中IspE,IspH基因的转录水平均显著高于忍冬.结论:克隆获得忍冬、红白忍冬、红腺忍冬和水忍冬中IspE,IspH基因,并证实了其在不同金银花类药用植物中的表达,为进一步研究IspE,IspH基因对萜类化合物生物合成和花香气以及颜色的影响奠定了基础.  相似文献

Aim of the study

To investigate the activities of the 217 plant extracts in traditional medicine of the Brazilian Cerrado against protozoans and yeasts.

Materials and methods

Plant extracts were prepared by the method of maceration using solvents of different polarities. The growth inhibition of chloroquine-resistant Plasmodium falciparum strain (FcB1) was determined by measuring the radioactivity of the tritiated hypoxanthine incorporated. Activity against Leishmania (Leishmania) chagasi and Trypanosoma cruzi was measured by the MTT colorimetric assay. The antifungal tests were carried out by using the CLSI method. The active extracts were tested also by cytotoxicity assay using NIH-3T3 cells of mammalian fibroblasts.

Results

Two hundred and seventeen extracts of plants were tested against Plasmodium falciparum. The eleven active extracts, belonging to eight plant species were evaluated against L. (L.) chagasi, Trypanosoma cruzi, yeasts and in NIH-3T3 cells. The results found in these biological models are consistent with the ethnopharmacological data of these plants. The ethyl acetate extract of Diospyros hispida root showed IC50 values of 1 μg/mL against Plasmodium falciparum. This extract demonstrated no toxicity against mammalian cells, resulting in a significant selectivity index (SI) of 435.8. The dichloromethane extract of Calophyllum brasiliense root wood was active against Cryptococcus gattii LMGO 01 with MIC of 1.95 μg/mL; and Candida albicans ATCC 10231 and Candida krusei LMGO 174, both with MIC of 7.81 μg/mL. The same extract was also active against Plasmodium falciparum and L. (L.) chagasi with IC50 of 6.7 and 27.6 μg/mL respectively. The ethyl acetate extract of Spiranthera odoratissima leaves was active against Cryptococcus gattii LMGO 01 with MIC of 31.25 μg/mL, and against Plasmodium falciparum with IC50 of 9.2 μg/mL and Trypanosoma cruzi with IC50 of 56.3 μg/mL.

Conclusion

The active extracts for protozoans and human pathogenic yeasts are considered promising to continue the search for the identification and development of leading compounds.  相似文献

目的:建立四川藏羌地区五加属主要资源种类的HPLC指纹图谱,为鉴别其种类提供依据.方法:采用梯度洗脱方法,以Kromasil C18色谱柱(4.6 mm×150 mm,5μm),柱温30℃,以甲醇-0.5%磷酸水溶液为流动相.建立四川藏羌地区五加属药材的指纹图谱,并采用“中药色谱指纹图谱相似度评价系统”软件进行相似度的计算.结果:建立的方法重现性较好,并根据检测结果确定了12个共有特征峰.应用DPSv3.0统计软件对获得的HPLC数量化特征进行系统聚类分析(组间距离法,欧氏距离),聚类分析结果为S1与S2,S3红毛五加药材的不同药用部位,成分最为接近;其次是S5(糙叶藤五加茎皮);再其次分别是S4(蜀五加茎皮)、S8(香加皮药材)、S6(刺五加药材)和S7(五加皮药材).结论:所建立的HPLC指纹图谱对四川藏羌地区五加属主要资源种类的鉴别具有参考价值.  相似文献

Ethnopharmacological relevance

Arctium lappa and Tussilago farfara (Asteraceae) are two plant species used traditionally as antitubercular remedies. The aim of this study was (i) to screen Arctium lappa and Tussilago farfara extracts for activity against Mycobacterium tuberculosis and (ii) to isolate and identify the compound(s) responsible for this reputed anti-TB effect.

Materials and methods

The activity of extracts and isolated compounds was determined against Mycobacterium tuberculosis H37Rv using a high throughput spot culture growth inhibition (HT-SPOTi) assay.

Results

The n-hexane extracts of both plants, the ethyl acetate extract of Tussilago farfara and the dichloromethane phase derived from the methanol extract of Arctium lappa displayed antitubercular activity (MIC 62.5 μg/mL). Further chemical investigation of Arctium lappa led to the isolation of n-nonacosane (1), taraxasterol acetate (2), taraxasterol (3), a (1:1) mixture of β sitosterol/stigmasterol (4), isololiolide (5), melitensin (6), trans-caffeic acid (7), kaempferol (8), quercetin (9), kaempferol-3-O-glucoside (10). Compounds isolated from Tussilago farfara were identified as a (1:1) mixture of β sitosterol/stigmasterol (4), trans-caffeic acid (7), kaempferol (8), quercetin (9), kaempferol-3-O-glucoside (10), loliolide (11), a (4:1) mixture of p-coumaric acid/4-hydroxybenzoic acid (12), p-coumaric acid (13). All compounds were identified following analyses of their physicochemical and spectroscopic data (MS, 1H and 13C-NMR) and by comparison with published data. This is the first report of the isolation of n-nonacosane (1), isololiolide (5), melitensin (6) and kaempferol-3-O-glucoside (10) from Arctium lappa, and of loliolide (11) from Tussilago farfara. Amongst the isolated compounds, the best activity was observed for p-coumaric acid (13) (MIC 31.3 μg/mL or 190.9 μM) alone and in mixture with 4-hydroxybenzoic acid (12) (MIC 62.5 μg/mL).

Conclusions

The above results provide for the first time some scientific evidence to support, to some extent, the ethno-medicinal use of Arctium lappa and Tussilago farfara as traditional antitubercular remedies.  相似文献

Aim of the study

The in vitro antiplasmodial activity and cytotoxicity of methanolic and dichloromethane extracts from five Congolese plants were evaluated. The plants were selected following an ethnobotanical survey conducted in D.R. Congo and focusing on plants used traditionally to treat malaria. The in vivo antimalarial activity of aqueous and methanolic extracts active in vitro was also determined in mice infected by Plasmodium berghei berghei.

Materials and methods

The growth inhibition of Plasmodium falciparum strains was evaluated using the measurement of lactate dehydrogenase activity. The extracts (aqueous, CH3OH, EtOH and CH2Cl2) were prepared by maceration and tested in vitro against the 3D7 (chloroquine sensitive) and W2 (chloroquine resistant) strains of Plasmodium falciparum and against the human normal fetal lung fibroblasts WI-38 to determine the selectivity index. Some extracts were also used at the dose of 300 mg/kg to evaluate their activity in mice infected since 4 days by Plasmodium berghei.

Results

Two plants presented a very high activity (IC50 < 3 μg/ml). These plants were Strychnos icaja roots bark (MeOH and CH2Cl2) and Physalis angulata leaves (MeOH and CH2Cl2). One plant (Anisopappus chinensis whole plant, MeOH and CH2Cl2) presented a high activity (IC50 < 15 μg/ml). The extracts of Anisopappus chinensis and Physalis angulata showed also a good inhibition of parasitemia in vivo. Flavonoids, phenolic acids and terpenes were identified in these plants by a general phytochemical screening method.

Conclusion

Three plants showed a very interesting antiplasmodial activity (Anisopappus chinensis, Physalis angulata and Strychnos icaja) and one of them showed a good selectivity index (>10, Anisopappus chinensis). Anisopappus chinensis and Physalis angulata were also active in vivo.  相似文献

目的:分析比较柘树与构棘根及茎中花旗松素、柑桔素、槲皮素、山柰酚的含量,为品种鉴定与质量评价提供科学参考.方法:采用高效液相二极管阵列检测法(HPLC-DAD),色谱柱为Agilent C18(4.6 mm×150 mm,5μm),流动相为甲醇(A)-0.1%磷酸溶液梯度洗脱(0 ~ 15 min,35% ~50% A;15 ~30 min,50% ~65%A),检测波长290 nm(花旗松素、柑桔素)和365 nm(槲皮素、山柰酚),柱温30℃.结果:花旗松素、槲皮素的含量柘树根明显高于构棘根,柘树茎明显高于构棘茎,种间含量差异大;柑桔素、山柰酚的含量构棘根高于柘树根,柘树茎与构棘茎含量相当,种间含量差异小,同种药用部位间含量差异大.结论:4种成分的含量在柘树和构棘及其药用部位根和茎都存在一定的差异,在临床上不可混淆使用.  相似文献

Aim of the study

Antiplasmodial and analgesic activities of leaf extract and fractions of Acalypha wilkensiana were evaluated to ascertain the folkloric claim of its antimalarial and analgesic activities.

Materials and methods

The crude leaf extract (220–659 mg/kg) and fractions (chloroform and aqueous; 440 mg/kg) of Acalypha wilkensiana were investigated for antiplasmodial activity against chloroquine sensitive Plasmodium berghei infections in mice and for analgesic activity against chemical and heat-induced pains. The antiplasmodial activity during early and established infections as well as prophylactic activity were investigated. Chloroquine (5 mg/kg) and pyrimethamine (1.2 mg/kg) were used as positive controls. Thin films made from tail blood of each mouse were used to assess the level of parasitaemia of the mice. Analgesic activity of the crude extract was also evaluated against acetic acid, formalin and heat-induced pains.

Results

The extract and its fractions dose-dependently reduced parasitaemia induced by chloroquine sensitive Plasmodium berghei infection in prophylactic, suppressive and curative models in mice. These reductions were statistically significant (p < 0.001). They also improved the mean survival time (MST) from 16 to 22 days relative to control (p < 0.01–0.001). The activities of extract/fractions were incomparable to that of the standard drugs used (chloroquine and pyrimethamine). On chemically and thermally induced pains, the extract inhibited acetic acid and formalin-induced inflammation as well as hot plate-induced pain in mice. These inhibitions were statistically significant (p < 0.001) and in a dose-dependent fashion.

Conclusion

The antiparasitaemic and analgesic effects may in part be mediated through the chemical constituents of the plant.  相似文献

目的:对“蒂达”各品种不同基原植物药用合理性及其资源利用价值作出评价.方法:采用HPLC指纹图谱共有特征峰、结合SPSS聚类分析方法,评价“蒂达”不同品种基原植物的成分组成相似性.结果:“蒂达”不同品种涉及的来源有龙胆科獐牙菜属、花锚属、扁蕾属及肋柱花属共15种1变种33个样品,指纹图谱全谱相似度较差;以10个共有峰成分的相对面积聚类,其基原植物可分别聚为4个组;上述4个属的属间成分组成差异较大;其中獐牙菜属不同种间成分组成变化较大,而其他各属种间成分组成相似度较高.结论:上述龙胆科4属植物均含有具有保肝利胆生物活性的环烯醚萜类、(山)酮类及三萜类成分,藏医学将其归为“蒂达”一类药材使用具有一定的物质基础;鉴于“蒂达”类药材不同基原各品种间在成分组成上差异较大,分别制定各品种的HPLC指纹图谱,是鉴别和控制各品种质量的有效手段,可有效控制各基原品种质量.  相似文献

Aim of the study

Based on the recipe of the traditional anti-diabetic formula TZQ, we developed TZQ-F, a new formula including 8 fractions isolated from Red Paeony root, Mulberry leaf, Lotus leaf, Danshen root and Hawthorn leaf with a good quality assurance. The study was aimed at fraction preparation and effects of the fractions on abnormal glucose and lipid metabolism.

Materials and methods

The active fractions were obtained by macroporous resin, ion-exchange resin and polyamide resin column chromatographies. HPLC analyses were used for quality control. In vitro mechanism study included DPPH radical scavenging, AGEs formation inhibition, α-glucosidase inhibition and lipase inhibition, and rats on high-fat diet were used for in vivo study.

Results

In vitro mechanism study showed that among the 8 fractions, three of them had inhibition effects on intestinal disaccharase, three with inhibition effects on lipase, and five with effects of free radical scavenging. In vivo study showed that after 4 weeks of treatment, TZQ-F significantly decreased the levels of serum total cholesterol, TG, glucose, LDL-C and HDL-C in rats on high-fat diet. Consistent with the in vitro and in vivo results, histology study demonstrated that TZQ-F alleviated hepatic steatosis induced by high-fat diet.

Conclusions

TZQ-F possesses the potential regulation effects on abnormal glucose and lipid metabolism.  相似文献

目的:建立并比较野生和栽培品种防风中升麻苷、5-O-甲基维斯阿米醇苷HPLC测定方法和HPLC指纹图谱。方法:含量测定采用岛津C18色谱柱(4.6 mm×150 mm,5μm),流动相为甲醇-水(40∶60),检测波长254 nm,流速1.0 mL.min-1,柱温30℃。HPLC指纹图谱测定采用资生堂MGⅡC18(4.6 mm×250 mm,5μm)色谱柱,流动相为乙腈-水,梯度洗脱,检测波长254 nm,流速1.0 mL.min-1,柱温30℃。结果:升麻苷加样回收率为99.6%,RSD 0.72%(n=6);5-O-甲基维斯阿米醇苷加样回收率为102.6%,RSD 0.88%。野生与栽培品种升麻苷质量分数分别为(4.96±2.59),(3.61±1.82)mg.g-1;5-O-甲基维斯阿米醇苷质量分数分别为(3.91±2.09),(4.37±2.02)mg.g-1。野生与栽培品种升麻苷和5-O-甲基维斯阿米醇苷质量分数无显著性差异。在此条件下所建立的指纹图谱中防风各成分得到有效分离。结论:升麻苷、5-O-甲基维斯阿米醇苷HPLC测定方法简便,结果准确,HPLC指纹图谱分析可作为防风药材质量控制及分类评价的依据。  相似文献
