Using incision, excision and dead space wound models in rats, a study was conducted on the effect of histamine on wound healing. Exogenous histamine given either ip or locally was without any effect. Semicarbazide as (histamine synthesis inhibitor) suppressed healing process (breaking strength of skin incision wound), decreased breaking strength and hydroxyproline content of granulation tissue and delay in period of epithelization. On the other hand compound 48/80 (a promoter of histamine forming capacity) was found to promote wound healing. Exogenous histamine (topical) reversed the anti-healing effect of semicarbazide on incision and excision wounds.  相似文献

Cimetidine, an H-2 antagonist, is one of the most commonly prescribed drugs and is used in the management of peptic ulcer disease and prevention of stress ulcerations following major trauma or operations. It has been shown that histamine has a stimulatory effect on wound healing which is believed to be mediated through H-2 receptors. We hypothesized that cimetidine would have an inhibitory effect on wound healing. Fifty-two adult rats were divided into two groups: Group A was placed on a standard chow and Group B was given chow supplemented with cimetidine (300 mg/Kg diet), a dose equivalent to that clinically used in man and effective in inhibiting gastric hypersecretion in the rat. Ten days later, all rats underwent a dorsal skin incision under pentobarbital anesthesia and implantation of a polyvinyl alcohol sponge subcutaneously. Animals were maintained on their respective diets. Rats were killed with ether five and ten days postoperatively, the wounds were excised and breaking strength measured. Sponge hydroxyproline content was measured. No significant difference in breaking strength or sponge hydroxyproline content was found between the two groups. We thus conclude that cimetidine has no effect on wound healing in rats when used in doses equivalent to the clinical dose in man.  相似文献

Dermal substitution and wound healing are areas of medicine in which there have been many recent advances, but neither the commercially available products nor the products currently described in experimental studies are able to fully substitute for natural living skin. There is an overall consensus that to heal wounds, the substitution of connective tissue matrix, the main component of each wound, is necessary. Both artificial and natural polymers have been used to reconstitute dermis. Nowadays, collagen has been discovered again. Collagen is a natural substrate for cellular attachment, growth and differentiation, and promotes cellular proliferation and differentiation. Once dermis reconstruction is done, the covering of the wound surface with both in vitro expanded epidermis and autologous split-skin transplants is significantly easier and has an improved chance of success. Nowadays, many commercial and experimental products have been introduced to improve cutaneous wound healing. This review discusses some of both acellular and cell-containing products used in the treatment of skin wounds.  相似文献

Wound healing is divided into three phases: inflammatory, proliferative and remodeling. Mast cells participate in all these phases. The aim of the present study was to determine the effects of propolis on the population of mast cells in oral surgical wounds in comparison to the results obtained with dexamethasone. This study was prospective, in vivo, randomized, semiexperimental, quantitative and comparative animal. A circular surgical wound was made on the dorsum of the tongue of 90 hamsters divided into three experimental groups: topical application of 30% propolis alcoholic extract (Group 1); 0.1% dexamethasone in orabase cream (Group 2); and orabase cream alone (Group 3). Applications were performed every 12 h throughout the experiment. The postoperative times for killing of the animals were 1, 3, 7, 14 and 28 days. The Student's t test for independent samples was employed in the statistical analysis. In the inflammatory phase of healing, propolis caused a greater reduction in the number of mast cells on the edge and in the central region of the surgical wound in comparison to dexamethasone. Moreover, the number of mast cells on day 1 was lower in the central region of the wounds treated with the orabase cream alone in comparison to dexamethasone. In conclusion, the anti-inflammatory action of propolis mediated by mast cells was more effective than dexamethasone in the inflammatory phase of healing.  相似文献

Factors affecting the rate of healing of experimental skin wounds in rats have been investigated. The effectiveness of healing was measured by determining the tensile strengths of the incised skin after various time intervals. When the skin histamine content was lowered by treatment with polymyxin B or with compound 48/80, retardation of the healing process was evident from the reduced tensile strengths. When the skin 5-hydroxytryptamine content was lowered by treatment with reserpine, retardation of healing was also found. Heparin increased the rate of healing and more rapid healing was obtained by giving histamine before each dose of heparin. On the other hand, some glucocorticoids markedly inhibited the healing process. Of the constituents of the tissue mast cells, heparin appears to be more important than histamine and 5-hydroxytryptamine in promoting the healing of experimental skin wounds in rats.  相似文献

The wound healing effect of two calcium channel blockers, nifedipine and amlodipine was studied in rats using incision and excision wound models. In incision wound, two straight paravertebral skin thickness incision were made and on tenth day skin tensile strength was measured by using continuous water flow technique. In excision wound, circular piece of skin excised in its full thickness and wound contraction monitored by alternate day wound tracing and epithelisation period was monitored by noting the number of days required for escher to fall. Drugs enhanced the skin tensile strength in incision wound model. In excision wound model, wound contraction is increased on 4th and 16th day but epithlisation period was not significantly altered. In conclusion, calcium channel blockers can be used to enhance wound healing, especially if wound healing was suppressed by steroids.  相似文献

目的:探讨四妙君逸软膏促进创面愈合的作用及机制。方法:将60只大鼠随机分成空白基质组、重组人表皮生长因子组、龙珠软膏组及四妙君逸软膏高、中、低剂量组,每组10只,制备大鼠创伤模型。治疗从造模后d 1开始至创面完全愈合,观察术后各组大鼠创面的愈合情况,于术后d 3,d 7和d 14记录创面愈合率,在d 7和d 14取材。采用考马斯亮兰法检测大鼠肉芽组织中蛋白含量、免疫组化ABC法检测表皮生长因子受体(EGFR),最后观察创面愈合时间。结果:四妙君逸软膏能缩短创面愈合时间,提高创面愈合率及创面肉芽组织中蛋白含量和EGFR。结论:四妙君逸软膏能促进创面愈合,其机制之一是通过提高EGFR以及蛋白含量来促进创面愈合。  相似文献

Successful wound healing depends upon angiogenesis, and impaired angiogenesis is a hallmark of the chronic wounds encountered with diabetes and venous or arterial insufficiency. To intervene and improve wound closure, it is essential to investigate the effects of different natural remedies in wound healing. The chicken dorsum skin excisional wound assay was used to investigate the influence of different concentrations of aged garlic solution (AGS) on wound healing. Gross, histopathology, scanning electron microscopy (SEM) and computer-based three-dimensional (3D) image-probing techniques were utilized to determine the effects of AGS on wound closure, re-epithelialization, dermal matrix regeneration, and angiogenesis. Ninety chicks, aged 1 week and divided in 6 groups, were topically exposed to different concentrations of AGS for 6 days: control (group A), 1% (group B), 5% (group C), 10% (group D), 15% (group E), and skin lotion (group F). Different patterns, ranging from incomplete to almost complete wound closure, were observed among different groups with highly significant results (P?<?0.001) in group E. Histological investigations revealed a positive augment in the re-epithelialization of all AGS exposed wounds. An increase in the number of new loosely packed collagen and maturation of collagen bundles was observed in all treated wounds at days 4 and 6 post-wounding, respectively. Similar results were achieved through SEM of treated wounds. Histological investigations revealed the profuse dose-dependent neovascularization among AGS-treated wounds. Abbott curve, angular spectrum, and different parameters of 3D surface roughness of wounds were also measured for the precise quantification of angiogenesis. A very highly significant (P?<?0.001) increase in angiogenesis was observed among all treated groups. No significant change was observed among control and skin lotion–treated groups. These observations substantiate the beneficial use of AGS in the treatment of wounds. Additional studies are needed to study the specific wound-healing mechanisms of chemical, or group of chemicals, present in AGS.  相似文献

目的探讨抗生素骨水泥治疗创伤性胫骨骨髓炎患者的临床效果。方法 96创伤性胫骨骨髓炎患者,依据患者入院单双号分为观察组和常规组,每组48例。常规组患者给予局部抗生素灌注治疗,观察组患者给予局部抗生素骨水泥治疗。比较两组患者治疗效果、并发症发生率、骨缺损长度、软组织缺损面积及手术前后疼痛评分。结果观察组患者总有效率为97.92%,常规组总有效率为85.42%;观察组患者总有效率明显高于常规组,差异有统计学意义(P<0.05)。观察组患者骨缺损长度为(1.42±0.23)cm,软组织缺损面积为(0.76±0.25)cm^2;常规组患者骨缺损长度为(1.79±0.37)cm,软组织缺损面积为(0.99±0.48)cm^2;观察组患者骨缺损长度及软组织缺损面积均优于常规组,差异有统计学意义(P<0.05)。手术前,两组患者疼痛评分比较差异无统计学意义(P>0.05);手术后,观察组患者疼痛评分(2.47±0.79)分显著低于常规组的(3.85±0.98)分,差异有统计学意义(P<0.05)。观察组患者并发症发生率6.25%低于常规组的25.00%,差异有统计学意义(P<0.05)。结论对于创伤性胫骨骨髓炎患者在临床上可以根据患者病情发展的实际情况采取抗生素骨水泥治疗,一方面能够促进患者临床治疗效果的提升,另一方面还可有效减少骨缺损的长度、面积,降低并发症发生率,有较高的使用和推广价值。  相似文献

目的研究重组人神经生长因子(recombinant human nerve growth factor,rh NGF)对糖尿病大鼠合并难愈创面的治疗效果。方法腹腔注射链脲佐菌素(60 mg·kg~(-1))诱导大鼠糖尿病模型,并在大鼠背部造成一圆形烫伤创面,建立糖尿病合并难愈创面的动物模型。用药组rh NGF(3、6、12μg·kg~(-1))肌肉注射给药,每天1次,连续21 d。测定大鼠伤口面积,HE染色和透射电镜检查创面皮肤组织病理形态学变化。结果 rh NGF明显减小糖尿病大鼠烫伤创伤面积(P <0. 05); HE染色结果表明,rh NGF明显促进糖尿病大鼠烫伤创面纤维细胞和毛细血管再生;透射电镜结果表明,rh NGF对糖尿病大鼠烫伤创面皮肤的上皮细胞结构排列、细胞连接以及上皮下胶原等超微结构有明显改善作用。结论rh NGF对糖尿病大鼠烫伤性难愈创面具有促进愈合的作用。  相似文献

This study explored the feasibility of development of solubilized amniotic membrane extract (AME) as a potential wound healing substrate with improved efficacy. Bovine amniotic membrane was extracted using a mixture of acetic acid and 2-mercaptopropionic acid under sonication, which was followed by the frozen, and then lyophilized processes. The effects of AME on cell migration and growth properties were evaluated from 0 to 24 h of post injury using primary human foreskin fibroblast monolayer culture with one line scratch as an in vitro wound model. Its wound healing efficacy and scar preventive effects were investigated using whole thickness biopsy punch (8 mm) wound model obtained from rabbit ear. Intra dermal injections of AME fluid (10 μl of 1.2 μg/μl) on four wound sites were performed at 1 h pre injury, post 1, 2 and 3 day. The processes and levels of re-epithelialization and dermal regeneration were examined through histological assessment with H–E staining. In cell migration study conducted at 24 h post injury, AME (1.7 μg/ml) treated cells significantly increased wound closure with 54.9 % compared to control. Histological image analysis on AME treated wound sites at 36 days post injury showed properly developed epidermal basal cell layers and weave-like dermal collagen bundles, whereas those of untreated control skin showed over-proliferation of epidermis and aggregated collagen bundles with defected dermal regeneration. The results of this study verified the feasibility of dermal injections of freeze dried AME as a potential wound healing substrate which can promote epidermal and dermal regeneration, while avoiding undesirable hyper-proliferation of damaged tissue.  相似文献

目的观察富血小板血浆(PRP)在慢性创面愈合中的应用效果。方法选取2018年12月-2019年5月郑州人民医院收治的慢性创面患者60例,根据随机方法分为A、B组,每组30例。A组应用PRP处理,B组以碘伏敷料换药。比较2组创面愈合情况,治疗前后炎性因子分泌情况,并发症发生情况。结果 A组创面愈合率为93. 33%,高于B组的73. 33%(P <0. 05);治疗后,2组炎性因子(TNF-α、IL-6、IL-8)水平较治疗前均下降,且A组下降幅度大于B组(P <0. 01);A组并发症发生率为3. 33%,低于B组的20. 00%(P <0. 05)。A组伤口愈合时间、换药次数、疼痛评分均低于B组(P <0. 01)。结论相较于常规换药、包扎处理,利用PRP能促进慢性创面愈合,抗炎抑菌,预防多种并发症的发生,值得推广应用。  相似文献
