Rabbits were fed skim milk-based diets containing by weight either 14% soybean oil, 28% soybean oil, 14% tallow or 28% tallow. A control group was fed a commerical stock diet. Plasma cholesterol concentration was monitored for 10 weeks, after which cholesterol concentrations of liver, skeletal muscle, adipose tissue, brain, heart, aorta, kidney, skin and intestinal mucosa were determined. Feeding soybean oil diets (relatively high content of polyunsaturated fat by acids) resulted in lesser concentrations of cholesterol in blood plasma than did feeding tallow diets (greater content of saturated fatty acids). Conversely, cholesterol contents were greater in skeletal muscle, adipose tissue, heart, aorta, kidney, skin and intestinal mucosa of rabbits fed soybean oil diets. Cholesterol contents of liver and brain were unaffected by type of dietary fat. Our results demonstrate that the degree of saturation of fat in a milk-based diet of rabbits will influence the partitioning of cholesterol between the blood and several other tissues and organs.  相似文献

To study effects of dietary protein source on plasma cholesterol homeostasis, six littermate pairs of 8-wk-old, castrated male pigs were assigned randomly to high fat diets containing either lean beef or soy protein isolate, with beef tallow as the major fat source in both diets. Diets were fed for 6 wk, with blood samples taken weekly and analyzed for lipid composition. At 6 wk, each pig was injected with [3H]cholesterol- and [14C]sucrose-labeled low density lipoproteins, and serial blood samples were taken. Pigs were then killed, and several tissues were sampled. Results show that dietary beef or soy protein, when fed with beef tallow as a fat source, has differential effects on rates of uptake of intact LDL by heart, M. sartorius muscle and subcutaneous adipose tissue, and on rates of uptake of LDL cholesterol by net free cholesterol exchange in subcutaneous and perirenal adipose tissues. Dietary protein source affected the composition of only two tissues analyzed. Pigs fed the beef-based diet had greater lipid concentrations in M. sartorius muscle, whereas pigs fed the soy protein-based diet had greater lipid concentrations in heart. Source of dietary protein did not influence in vivo kinetics of LDL cholesterol removal from blood plasma.  相似文献

Young, growing pigs were fed for 8 wk diets containing either soy protein isolate or ground beef as the main protein source and soybean oil or beef tallow as the main fat source to examine effects of types of dietary protein and fat on cholesterol distribution between blood plasma, plasma lipoproteins and body organs. Type of dietary protein did not influence concentrations of a) cholesterol in plasma, b) low-density-lipoprotein (LDL) cholesterol, c) high-density-lipoprotein (HDL) cholesterol or d) cholesterol in several organs of pigs. Pigs fed tallow had greater concentrations of plasma cholesterol and LDL and HDL cholesterol than did pigs fed soybean oil. Ratios of HDL to LDL were not altered by dietary fat. Cholesterol concentrations were greater in heart, skeletal muscle and viscera of pigs fed soybean oil than in those fed tallow. Plasma cholesterol varied reciprocally with cholesterol in several tissues. Thus, dietary beef and soy protein isolate had similar effects on cholesterol concentrations in plasma, LDL, HDL and organs, whether pigs consumed soybean oil or beef tallow as a major fat source. Soybean oil, however, exerted a hypocholesterolemic effect, irrespective of major source of dietary protein.  相似文献

The objective of the present study was to determine the combined effects of cod protein and fish oil on the modulation of triglyceride metabolism in rats, and to evaluate their potential mechanisms of action. Plasma and hepatic lipid concentrations, triglyceride (TG) secretion rates and postheparin plasma lipoprotein lipase (LPL) activity were determined in rats fed for 28 d diets varying in both protein (200 g/kg) and lipid (140 g/kg) sources: 1) casein-menhaden oil, 2) casein-beef tallow, 3) cod protein-menhaden oil or 4) cod protein-beef tallow. Menhaden oil feeding diminished hepatic TG concentrations (P = 0.02), hepatic TG secretion rates (P = 0.003) and triglyceridemia (P = 0.02) compared with beef tallow. Hepatic TG concentrations (P = 0.05) and TG secretion rates (P = 0.04) were reduced in rats fed cod protein compared with those fed casein. The protein source did not exert an independent effect on triglyceridemia, whereas the combination of cod protein and menhaden oil resulted in 50% lower plasma TG compared with the casein-beef tallow mixture, whereas the combination of menhaden oil and casein did not significantly decrease triglyceridemia compared with casein-beef tallow. Menhaden oil (P = 0.005) and cod protein (P = 0.03) also lowered plasma cholesterol concentrations in comparison with beef tallow and casein, respectively. This was associated with a reduction in hepatic cholesterol concentrations when rats fed cod protein were compared with those fed casein (P = 0.006). No diet effect was observed on postheparin plasma LPL activity, but the activity of hepatic triglyceride lipase was reduced in rats fed menhaden oil compared with those fed beef tallow. These findings show that both cod protein and menhaden oil exert independent and beneficial effects on lipid metabolism in rats.  相似文献

Male weanling rats were fed for 28 d a purified diet containing 20% (wt/wt) fat providing high levels of either saturated fat or alpha-linolenic acid or eicosapentaenoic and docosahexaenoic acids with or without 2% (wt/wt) cholesterol supplementation. Effect of diet on rate of desaturation of eicosatrienoic acid (20:3 omega 6) and lipid composition of liver microsomal membranes was examined. The desaturation of 20:3 omega 6 to arachidonic acid (20:4 omega 6) was higher in rats fed linseed oil and lower in rats fed fish oil than in control animals fed the beef tallow diet. The desaturation of 20:3 omega 6 was lower in rats fed beef tallow or linseed oil diets supplemented with cholesterol than in the respective unsupplemented diet. Inclusion of 2% (wt/wt) cholesterol in the fish oil diet failed to affect synthesis of 20:4 omega 6 from 20:3 omega 6. These in vitro changes in delta 5-desaturase activity are consistent with the diet-induced alterations observed in the fatty acid composition of microsomal membranes. Both free cholesterol and cholesterol ester in the microsomal membrane were higher in rats fed beef tallow or linseed oil diets supplemented with exogenous cholesterol than in the respective unsupplemented diet, and only free cholesterol was higher in rats fed the fish oil diet supplemented with cholesterol. Feeding fish oil appears to prevent both the inhibition of 20:4 omega 6 biosynthesis and the accumulation of cholesterol ester that were apparent when 2% cholesterol was added to either beef tallow or linseed oil diets.  相似文献

Most studies of the effects of dietary fat sources on plasma lipid components have used diets with extreme fat compositions; the current study was designed to more nearly mimic human dietary fat intake. Young growing pigs were fed diets containing either 20 or 40% of energy as soy oil, beef tallow or a 50/50 blend of soy oil and tallow. Different dietary fats did not affect concentrations of cholesterol, triacylglycerol or protein in plasma or major lipoprotein fractions. The concentration of phospholipid was less in plasma and in very low density lipoproteins with soy oil feeding than with tallow feeding. The weight percentage of cholesteryl ester in the low density lipoprotein fraction tended to be greater with 40% than with 20% tallow and tended to be less with 40% than with 20% soy oil. Phospholipid as a weight percentage of low density lipoprotein was least in pigs fed soy oil. Tallow feeding increased the percentage of myristic, palmitic, palmitoleic and oleic acids in plasma, relative to both other groups. Soy oil feeding increased the percentage of linoleic and linolenic acids. These moderate diets were not hypercholesterolemic, but they did alter plasma fatty acid composition and phospholipid concentrations in plasma and very low density lipoprotein.  相似文献

OBJECTIVE: We assessed the effect of dietary proteins isolated from beef, pork, and turkey meat on concentrations of cholesterol and triacylglycerols in plasma, lipoproteins, and liver and the composition of the microsomal membrane (fatty acids, phosphatidylcholine/phosphatidylethanolamine ratio) compared with that of casein and soy protein in rats. METHODS: Five groups of 12 rats each were fed semisynthetic diets for 20 d that contained 200 g/kg of proteins isolated from beef, pork, or turkey meat or, as controls, casein or soy protein. RESULTS: Rats fed beef, pork, or turkey proteins did not differ in cholesterol concentrations of plasma, lipoproteins, and liver and in composition of microsomal membrane from rats fed the casein diet. All groups fed a protein from an animal source had higher very low-density lipoprotein (VLDL) and liver cholesterol concentrations than did rats fed soy protein. However, rats fed pork protein had lower concentrations of triacylglycerols in liver, plasma, and VLDL and lower mRNA concentrations of sterol regulatory element binding protein-1 and glucose-6-phosphate dehydrogenase than did rats fed casein. However, concentrations of plasma and VLDL triacylglycerols in rats fed pork protein were not as low as those observed in rats fed soy protein. CONCLUSION: Proteins isolated from beef, pork, or turkey meat do not differ from casein in their effects on cholesterol metabolism. Pork protein decreases plasma triacylglycerol concentrations compared with casein but not compared with soy protein. The triacylglycerol-lowering effect of pork protein compared with casein is suggested to be caused by decreased hepatic fatty acid synthesis.  相似文献

This work was designed to investigate the effect of different lipid sources on ob gene expression and serum leptin levels in rats with two different feeding protocols: (1) free access to food; or (2) energy-controlled feeding. Male Wistar rats were fed diets containing 40 % energy as fat (olive oil, sunflower oil or beef tallow), for 4 weeks. In Expt 1 rats had free access to food, and in Expt 2 rats were fed a controlled amount of food (16 g/d, equivalent to 300 kJ/d). Insulin and leptin were determined by ELISA and ob mRNA by Northern blot. When rats had free access to food, ob mRNA levels were higher in animals fed either olive oil or sunflower oil than in those fed beef tallow. In marked contrast with feeding ad libitum, no differences were found among dietary fat groups in rats fed energy-controlled diets. When both feeding protocols were compared, free access to food induced an increased expression of ob mRNA in perirenal and/or epididymal adipose tissues from rats fed either olive oil or sunflower oil, but not from rats fed beef tallow. Dietary lipid type did not induce modifications in serum leptin concentrations. A tendency to higher serum leptin levels was observed more in rats with free access to food than in rats fed energy-controlled feeding. No differences were found in insulin levels. Dietary fat type importantly affects ob mRNA expression in rat white adipose tissue under hyperphagic conditions. Further study is needed in order to elucidate the mechanism underlying this effect.  相似文献

Growing pigs were fed diets, similar in protein and fat content, based on soy protein isolate and soybean oil (soy) or ground beef containing 21% fat (beef). Ground corn, vitamins and minerals completed the diets. Egg yolk was added to balance the intake of cholesterol. No significant differences were seen in total plasma cholesterol or liver cholesterol concentrations. Soy-fed pigs had significantly greater concentrations of cholesterol in the viscera excluding the liver (p<.001), aorta (p<.04), carcass (p<.001) and whole body (liver + other viscera + carcass) (p<.001). No evidence of fatty streaking or plaque formation was seen in Sudan IV-stained aorta segments of either treatment group. Although pigs fed the soy diet gained weight at a slower rate, their carcasses contained a significantly greater (p<.02) concentration of fat.Our data show that the soy diet, as compared to the beef diet, increased the proportion of fat in the carcass and increased tissue cholesterol deposition.  相似文献

Liver cholesterol concentrations were measured in mice after feeding for 30 days cholesterol-free, semipurified diets containing various sources of fat, carbohydrates or fiber. Olive oil produced significantly higher liver cholesterol concentrations than tallow, sunflowerseed oil and cocoa fat. In mice fed either fructose or sucrose liver cholesterol was significantly increased when compared with mice fed galactose or lactose. Dietary cellulose, when compared with pectin, did not influence liver cholesterol. The amount of fat in the diet, in the form of either corn oil or coconut fat, had no significant effect on liver cholesterol. It is concluded that the type of carbohydrate and fat in the diet are major determinants of liver cholesterol in mice.  相似文献

目的探讨大豆蛋白在正常饮食和高脂饮食状态下对大鼠脂代谢相关固醇调节元件结合蛋白(SREBP)基因表达水平的影响。方法 48只清洁级SD大鼠,按体重随机分为4组,分别喂饲含酪蛋白和大豆蛋白的正常饲料和高脂饲料28d后,脱颈椎处死动物,检测激素指标及基因表达水平。结果大豆蛋白组大鼠血清胰岛素水平和胰岛素/胰高血糖素比值显著低于酪蛋白组(P<0.05);大豆蛋白高脂组大鼠血清胰岛素水平显著低于酪蛋白高脂组(P<0.05);大豆蛋白组和大豆蛋白高脂组大鼠肝脏SREBP-1,2基因表达均分别显著低于酪蛋白组和酪蛋白高脂组(P<0.05);大豆蛋白组和大豆蛋白高脂组大鼠肝脏SREBP-1蛋白表达明显低于酪蛋白组和酪蛋白高脂组(P<0.05)。结论大豆蛋白可能通过影响胰岛素进而影响SREBP-1基因表达来调节血脂水平。  相似文献

In experiments with male Wistar rats, we measured the effects of nonpurified diets containing 9.1% added fat (beef tallow, native or randomized fish oil, native or randomized peanut oil) on apparent digestibility of total fat and individual fatty acids. We also investigated the effects of the diets on plasma contents of triglyceride, cholesterolesters and free and total cholesterol as well as on the fatty acid profiles of plasma and liver phospholipids. Randomization of fish oil or peanut oil had no significant effect on any of the lipid measurements. Fat digestibility was significantly lower in the rats fed beef tallow. Apparent absorption of 18:1(n-9) and polyunsaturated fatty acids was not dependent on the fatty acid profile of the dietary fat. Apparent absorption of 16:1(n-7) and saturated fatty acids was generally highest in the rats fed fish oil. Intake of fish oil or peanut oil significantly decreased plasma triglyceride content. Intake of fish oil resulted in substantially decreased contents of total cholesterol and cholesterolesters in plasma, but intake of peanut oil did not. Efficiency of conversion of 18:2(n-6) into 20:4(n-6) was inhibited by long-chain (n-3) fatty acids.  相似文献

To determine whether the long-term consumption of different amounts or types of fat by female rats affects the growth and development of their progeny, Wistar rats were fed from weaning either a low fat diet (4.5% by weight) or one of three high fat (32%) diets containing predominantly beef tallow (high saturated fat), corn oil (high polyunsaturated fat) or equal portions of tallow and corn oil (high mixed fat). Offspring were killed at birth or weaning. Weight of newborn pups was lower with maternal consumption of high polyunsaturated fat diets. Carcass composition of newborn pups and body weight of weanling rats was unaffected by maternal diet. The percentage of carcass lipid was greater in weanling rats from all high fat-fed dams. In both newborn pups and weanling rats, percent composition in carcass lipids of 16:0, 16:1 and 18:1 fatty acids generally decreased and 18:2 increased as the high fat maternal diet became more unsaturated. The consumption of diets high in polyunsaturated fatty acids prior to and throughout gestation thus seemed to have a transitory effect on reducing fetal growth in rats.  相似文献

Experiments were conducted to assess whether changing dietary fat composition altered phospholipid composition of rat testicular plasma membranes in a manner that altered receptor-mediated action of luteinizing hormone (LH)/human chorionic gonadotropin (hCG). Weanling rats were fed diets that provided high or low cholesterol intakes and that were enriched with linseed oil, fish oil or beef tallow for 4 wk. Feeding diets high in (n-3) fatty acids decreased plasma and testicular plasma membrane 20:4(n-6) content. A marked reduction of the 22:5(n-6) content and an increase in the 22:6(n-3) content of testicular plasma membrane was found only in animals fed fish oil. A decrease in binding capacity of the gonadotropin (LH/hCG) receptor in the plasma membrane, with no change in receptor affinity, was observed for animals fed either linseed oil or fish oil diets. Dietary treatments that raised plasma membrane cholesterol content and the cholesterol to phospholipid ratio in the membrane were associated with increased binding capacity of the gonadotropin receptor. Feeding diets high in 18:3(n-3) vs. those high in fish oil altered receptor-mediated adenylate cyclase activity in a manner that depended on the level of dietary cholesterol. Feeding diets high in cholesterol or fish oil increased basal and LH-stimulated testosterone synthesis relative to that in animals fed the low cholesterol diet containing linseed oil. It is concluded that changing the fat composition of the diet alters the phospholipid composition of rat testicular plasma membranes and that this change in composition influences membrane-mediated unmasking of gonadotropin receptor-mediated action in testicular tissue.  相似文献

The effects of feeding dietary and defatted oyster meat on lipid metabolism were investigated in rats by comparing measurements with those of casein and soybean protein. In the first experiment, male rats were fed 0.1% and 1% cholesterol-supplemented diets containing casein, oyster or soybean protein under the same dietary level of protein (20%). The concentrations of serum and liver cholesterol in the oyster group were significantly lower than those in the casein group for both the 0.1% and 1% cholesterol-supplemented diets. The cholesterol-lowering effect of oyster meat was more predominant than that of soybean protein. Feeding oyster meat significantly decreased the serum triglyceride concentration as compared to feeding casein for the 0.1% cholesterol-supplemented diets, and it reduced hepatic triglyceride concentration in both groups fed the 0.1% and 1% cholesterol-supplemented diets. The excretion of fecal total steroids was higher in the rats fed oyster meat than those fed casein or soybean protein for both the 0.1% and 1% cholesterol-supplemented diets. In the second experiment, the effects of defatted oyster on lipid metabolism were compared with casein and soybean protein in diets supplemented with cholesterol. The serum cholesterol concentration in the defatted oyster group was comparable to that in the other two groups, but the ratio of high-density lipoprotein-cholesterol to total cholesterol was higher in the defatted oyster group. The feeding of defatted oyster induced a lower liver cholesterol concentration as compared to casein and soybean protein. Serum and liver triglyceride levels were lower in the defatted oyster group than in the casein group. Defatted oyster accelerated the fecal excretion of both neutral and acidic steroids as compared to casein. Our results suggest that the feeding of oysters exerts a more potent hypolipidemic activity than soybean protein, and the effect may be ascribed to both lipid and non-lipid fractions in oyster.  相似文献

Isoenergetic diets containing 20% corn oil, 20% beef tallow, or an equal mixture of 10% corn oil and 10% beef tallow (mixed fat) were fed to 30 rats per diet for 28 weeks following weaning. DMBA [7,12-dimethylbenz(a)anthracene] was administered (1.75 mg/100 g body weight) in a single oral dose after 4 weeks of feeding. After 28 weeks, 70% of the rats fed corn oil had mammary tumors versus 47% for mixed fat and 30% for tallow. Diet had no effect on the number of tumors per tumor-bearing rat or the proportion of tumors that were adenocarcinomas. Other rats assigned to each of the three diets were killed at the time corresponding to DMBA administration for examination of hepatic mixed-function oxidase activity. NADPH cytochrome c reductase activity and cytochrome P-450 content were higher in rats fed corn oil or mixed fat rather than tallow. However, no significant differences in aryl hydrocarbon hydroxylase, glutathione transferase, and uridine-diphosphoglucuronide transferase activities were observed. The effects of dietary fat saturation on enzyme activity failed to show a clear association with DMBA carcinogenesis. In other rats assigned to the three dietary treatments for 4 or 16 weeks, lipid saturation did not change serum prolactin (PRL) concentrations during diestrus or proestrus. PRL secretion was examined following a provocative stimulus (perphenazine) in rats fed the experimental diets for 4 or 10-22 weeks. Although perphenazine increased serum PRL and depleted the pituitary of PRL, differences in dietary lipid saturation caused no significant changes in these indices. These data show that the incidence of mammary tumors in rats fed high fat diets (20% by weight) was greater in those fed corn oil compared to beef tallow. The effect of dietary lipid source on tumorigenesis was not associated with changes in carcinogen-metabolizing enzyme activity or PRL secretion.  相似文献

We investigated the relative effects of different dietary saturated fats on the size distribution, apolipoprotein (apo) and chemical composition of HDL in fasted rats. Male Sprague-Dawley rats (174 +/- 2 g) were fed diets containing 0.035% cholesterol and 16% fat (wt/wt) from corn oil (CO diet) or from 2% CO plus 14% butterfat (BF diet), beef tallow (BT diet), palm oil (PO diet) or coconut oil (CN diet) for 6 wk. Apparent lipid digestibility was significantly lower with the PO and BT diets vs. the CO, BF and CN diets. Plasma total cholesterol levels were significantly higher in rats fed the PO and BT diets than in rats fed the BF and CN diets but were not different among the PO-, BT- and CO-fed groups. Nondenaturing gradient gel electrophoresis immunoblot analysis indicated that HDL apo A-I and E resided on particles with significantly smaller modal diameters in rats fed all saturated fats compared with those fed the CO diet. Chemical analyses indicated that HDL generally contained proportionately less protein and more triglyceride, free cholesterol and apo E with saturated fat feeding than with CO diet feeding. Significantly higher plasma and VLDL triglyceride levels were noted with ingestion of the BT, PO or CN diet than with the CO diet. Butterfat feeding resulted in lower plasma triglycerides and HDL-esterified cholesterol than did feeding the other saturated fats. Very low density lipoprotein triglyceride concentrations were inversely correlated with HDL modal diameter of apo E containing lipoproteins (P less than 0.005). These data provide further evidence of the interrelationship of triglyceride and HDL metabolism and suggest that mechanisms independent of cholesterol ester transfer protein may mediate this response in rats.  相似文献

Although epidemiological studies have implicated red meat as increasing colon cancer risk, animal studies have generally not been supportive of such an effect. This study examined red meat components, such as beef protein and tallow, on markers of colon cancer risk. Rats administered dimethylhydrazine were fed either casein or beef protein as the protein source and soybean oil or tallow as the fat source in a 2 2 factorial design for 9 wk. There were fewer preneoplastic lesions [aberrant crypt foci (ACF)] and a greater apoptotic labeling index (P < 0.05) in the distal colonic mucosa of rats fed tallow compared with soybean oil. Fecal bile acid concentrations were significantly lower in rats fed tallow compared with soybean oil. There were no significant differences in mucosal cell proliferation. No significant effects were found due to protein source or to interactions between fat and protein sources for ACF, cell proliferation labeling indexes, or bile acid concentrations. However, there was a significant protein by fat source interaction for the apoptotic labeling index. The decreased number of ACF, decreased fecal bile acid concentration, and increased mucosal apoptosis with tallow consumption are not consistent with a role for this fat in increasing risk of colon cancer.  相似文献

Our previous work showed that rats consuming 20% (wt/wt) lard diets selected more protein and less carbohydrate than did rats fed 20% (wt/wt) soybean oil diets. To determine if this effect is specific to saturated fat source and to measure changes in fatty acid composition of synaptosomal phospholipids, male Wistar rats were fed a 20% (wt/wt) soybean oil or tallow diet with 24% protein and 48% carbohydrate for 2 wk. Rats then self-selected from two diets with the same fat previously fed but different protein and carbohydrate composition (5% protein, 67% carbohydrate and 55% protein, 17% carbohydrate). Tallow-fed rats selected more protein and less carbohydrate than did soybean oil-fed rats. Dietary fat also affected fatty acid composition of synaptosomal phospholipids, phosphatidylcholine being most affected and phosphatidylinositol most resistant. Later studies determined if selection differences were due to taste. When rats self-selected without prior exposure to diets, tallow-fed rats selected more protein and less carbohydrate than did soybean oil-fed rats during the last 2 wk but not the first 2 wk. When rats selected from diets with identical protein and carbohydrate composition but different dietary fat, rats consumed slightly greater amounts of the tallow diet at each protein level. The relative intake of tallow and soybean oil diets, however, did not vary significantly with dietary protein treatment. These results confirm that rats fed saturated fat diets select more protein and less carbohydrate than do rats fed polyunsaturated fat diets and suggest this difference is not due to taste factors.  相似文献

Five sources of dietary fiber were compared for their effect on blood and liver cholesterol. The effects of soybean fiber, rice bran (full fat), oat bran, barley bran and mixed bran on total blood cholesterol concentrations and liver cholesterol concentrations were measured in beef-fed C57BL/6 male mice. Each diet contained cooked beef, beef tallow, corn starch and 7% dietary fiber from one of the five fiber sources. A control group consumed a fiber-free diet. Dietary cholesterol was provided by the beef and beef tallow only. The experimental diets were fed for 3 wk; blood and liver were collected when the mice were 18 wk old. The liver cholesterol concentration in the rice bran-fed group was the lowest of the six diet groups and was significantly different than concentrations in the oat bran-fed group and the barley bran-fed group (P less than 0.05). The oat bran, mixed bran, and barley bran did not significantly lower blood cholesterol in the mice. Both the soybean fiber and rice bran diet groups had significantly lower total blood cholesterol than did the fiber-free controls (P less than 0.05).
