The effect of the hot water extract of black tea (Camellia sinensis (L.) O. Kuntze, Theaceae) on ulceration induced by various ulcerogens and by cold restraint stress (CRS) was investigated in albino rats. While prior administration of tea extract for 7 days significantly reduced the incidence of ulcer, ulcer number and ulcer index produced by aspirin, indomethacin, ethanol, reserpine and CRS, it failed to inhibit the ulcers induced by serotonin and histamine. Tea extract also favourably altered the changes in acid and peptic activity of gastric secretion induced by aspirin, indomethacin, ethanol, reserpine and CRS. The observations suggest that the hot water extract of black tea possesses anti-ulcer activity, probably mediated through prostaglandins.  相似文献

Solanum nigrum, an herbal plant which is recommended in ayurveda for the management of gastric ulcers. Therefore, the purpose of the study was to investigate the antiulcer effect of Solanum nigrum fruits extract (SNE) on cold restraint stress (CRU), indomethacin (IND), pyloric ligation (PL) and ethanol (EtOH) induced gastric ulcer models and ulcer healing activity on acetic acid induced ulcer model in rats. The treatment with SNE at higher dose significantly inhibited the gastric lesions induced by CRU (76.6%), IND (73.8%), PL (80.1%) and EtOH (70.6%), respectively, with equal or higher potency than omeprazole. SNE showed concomitant attenuation of gastric secretory volume, acidity and pepsin secretion in ulcerated rats. In addition, SNE (200 and 400mg/kgb.w.) accelerated the healing of acetic acid induced ulcers after the treatment for 7 days. Further, to ascertain the antisecretory action, the effects of SNE on H(+)K(+)ATPase activity and plasma concentration of gastrin hormone in ulcerated rats were determined. SNE significantly inhibits H(+)K(+)ATPase activity and decreases the gastrin secretion in EtOH-induced ulcer model. The severity of the reaction of ulcerogen and the reduction of ulcer size by SNE was evident by histological findings. Toxicity studies of SNE have also been carried out for its safety evaluation. SNE, thus, offers antiulcer activity by blocking acid secretion through inhibition of H(+)K(+)ATPase and decrease of gastrin secretion. These results further suggest that SNE was found to possess antiulcerogenic as well as ulcer healing properties, which might also be due to its antisecretory activity.  相似文献

The ethanol extract of Toona ciliata Roemer (heart wood) was evaluated for its anti-ulcer activity against aspirin plus pylorous ligation induced gastric ulcer (antisecretory), HCl-ethanol induced ulcer (cytoprotective) and water immersion stress induced ulcer in rats. We found that Toona ciliata extract at a dose of 300mg/kg p.o. markedly decrease the incidence of ulcers in all the three models. Ethanol extract of Toona ciliata showed significant reduction in gastric volume, free acidity, total acidity and ulcer index. The plant extract also showed gastro protective activity (52.94%), whereas standard drug sucralfate showed 94.85%. Toona ciliata extract showed protection index 43.0% in water immersion stress induced ulcer, whereas standard drug omeprazole showed protection index 100%.  相似文献

ETHNOPHARMACOLOGICAL RELEVANCE: Traditionally fruits of Ficus glomerata Roxb (Family: Moraceae) are used to treat anemia and gastrointestinal disorders. AIM: Gastroprotective effect of 50% ethanolic extract of F. glomerata fruit (FGE) was studied in different gastric ulcer models in rats. METHODS: FGE (50, 100 and 200 mg/kg body weight) was administered orally, twice daily for 5 days for prevention from pylorus ligation (PL), ethanol (EtOH) and cold restraint stress (CRS)-induced ulcers. Estimation of H+K+ATPase activity and gastric wall mucous were performed in EtOH-induced ulcer and antioxidant enzyme activities in supernatant mitochondrial fraction of CRS-induced ulcers. RESULTS: FGE showed dose dependent inhibition of ulcer index in pylorus ligation, ethanol and cold restraint stress-induced ulcers. FGE prevents the oxidative damage of gastric mucosa by blocking lipid peroxidation and by significant decrease in superoxide dismutase, H+K+ATPase and increase in catalase activity. High performance thin layer chromatography (HPTLC) analysis showed the presence of 0.57% and 0.36% w/w of gallic acid and ellagic acid in FGE. CONCLUSIONS: Our results show that F. glomerata possess significant gastroprotective activity which might be due to gastric defence factors and phenolics might be the main constituents responsible for this activity.  相似文献

Allophylus serratus is known to possess various therapeutic properties. We evaluated the anti-ulcerogenic property of crude ethanolic extract of Allophylus serratus (AS) in different ulcer models in Sprague-Dawley rats. The extract at 400 mg/kg body weight, once daily, orally has a significant effect in cold restraint (CRU, 2 h cold restraint stress), aspirin (ASA, 150 mg/kg body weight, orally), alcohol (AL, 1 ml/200 gm of absolute alcohol) and pyloric ligation (PL, 4h ligation) induced gastric ulcer models as it showed protection index of 71.28, 62.50, 90.84 and 64.29% protection, respectively whereas, standard drug omeprazole (OMZ, 10mg/kg body weight) has shown protection index of 85.70, 74.99 and 74.99 in CRU, ASA and PL model respectively. Sucralfate (SUC, 500 mg/kg body weight) as a standard drug in AL model has 93.20% protection. Furthermore, AS has significantly decreased the free acidity (72.41%), total acidity (47.97%) and peptic activity (24.59%), respectively as well as has significantly increased the mucus secretion (29.41%). Conclusively the ulcer protective effect of AS may be due to its anti-secretory along with cytoprotective mechanism.  相似文献

The ulcer protective potential of an ethanol extract of Commiphora opobalsamum (L.) Engl. (Burseraceae) 'Balessan' was assessed against different acute gastric ulcer models in rats induced by necrotizing agents (80% ethanol, 0.2M NaOH and 25% NaCl), hypothermic restraint stress, pyloric ligation (Shay) and indomethacin. Balessan, 250 and 500 mg/kg administered orally (intraperitoneally in Shay rat model) showed a dose-dependent ulcer protective effects in all the above ulcer models. Besides, the extract offered protection against ethanol-induced depletion of stomach wall mucus and reduction in nonprotein sulfhydryl (NP-SH) concentration. Ethanol treatment also caused histopathological lesions of the stomach wall. Pretreatment with Balessan extract provided a complete protection of gastric mucosa through supporting both the offensive and defensive factors. Balessan extract was also showed a large margin of safety without any apparent adverse effects in rats.  相似文献

The mature fruits of Momordica charantia L. (Cucurbitaceae) are used externally for the rapid healing of wounds and internally for the treatment of peptic ulcers in Turkish folk medicine. For the evaluation of the latter activity, ethanol-induced ulcerogenesis model in rats was employed. The olive oil extract of the material as well as dried-powdered fruits in filtered honey showed significant and dose-dependent anti-ulcerogenic activity against this model. A potent and dose-dependent inhibitory activity was also observed by the administration of ethanol extract of the fruits. For the bioassay-guided fractionation, the material was first extracted with hexane and then by ethanol and both extracts were found active against the same ulcer model. Furthermore, ethanol extract of the fruits showed significant activity against HCl-EtOH induced ulcerogenesis in indomethacin-pretreated rats and diethyldithiocarbamate-induced ulcer models.  相似文献

Rosmarinus officinalis L. crude hydroalcoholic (70%) extract was evaluated for antiulcerogenic activity employing different experimental models. The crude hydroalcoholic extract (CHE) decreased the ulcerative lesion index produced by indomethacin, ethanol and reserpine in rats. No antisecretory activity was observed on pyloric ligation model. The previous administration of L-NAME, a NO-synthase inhibitor, did not reduce the antiulcerogenic activity of CHE in ethanol induced ulcer model, suggesting that the pharmacological mechanism has no relationship with nitric oxide (NO). Whereas when the animal groups were treated with indomethacin, using the same experimental model, CHE did not reduce the antiulcerogenic activity, suggesting that the pharmacological mechanism has no relationship with prostaglandins. The previous treatment with N-ethymaleimide, a thiol blocker, including mucosal nonprotein sulfhydryl groups, reduced the anitulcerogenic activity of CHE on ethanol induced ulcer model. This result suggests that the crude hydroalcoholic extract of R. officinalis L. has active substances that increase the mucosal nonprotein sulfhydryl groups content. In another hypothesis, the pharmacological mechanism could be attributed to the activity of antioxidant compounds found in the crude hydroalcoholic extract which can react with N-ethylmaleimide.  相似文献

Ethnobotanical studies have revealed that Bidens pilosa is used in the traditional management of wounds and chronic gastro-duodenal ulcers. This led us to screen the methanol, cyclohexane and methylene chloride extracts of the plant for anti-ulcerogenic activity using the HCl/ethanol gastric necrotizing solution. The methylene chloride extract, which showed the highest activity (100% inhibition) at a dose of 750 mg/kg compared with the methanol and cyclohexane extracts (41 and 46% inhibition, respectively), was further tested using the indomethacin-HCl/ethanol-, absolute ethanol- and pylorus ligation-induced ulcer methods. Pre-treatment with indomethacin significantly reduced the protective effect of the extract against HCl/ethanol solution to 31%. The extract had very little gastric mucosal protection against absolute ethanol (9.8% inhibition at 750 mg/kg) compared with the controls and neither reduced gastric acid secretion in vivo nor the acidity of gastric juice following in vitro incubation.  相似文献

目的: 对三九胃泰方抗胃溃疡的有效部位进行筛选。方法: 将SD大鼠分为正常对照组、模型组、阳性对照(西米替丁(0.1 g·kg-1)与三九胃泰颗粒剂(2 g·kg-1)组、石油醚萃取物组、醋酸乙酯萃取物组、正丁醇萃取组及水萃取组,按生药剂量均为2 g·kg-1ig给药7 d,采用乙醇造大鼠胃黏膜损伤模型,以胃溃疡抑制率、胃蛋白酶活力、血清中丙二醛(MDA)含量与超氧化物歧化酶(SOD)活力为检测指标,对不同极性溶剂(石油醚、醋酸乙酯、正丁醇与水)萃取部位的抗胃溃疡活性进行评价。结果: 三九胃泰方各极性萃取部位均能显著抑制乙醇致大鼠胃黏膜损伤(P<0.05),其中石油醚萃取部位胃溃疡抑制率最高(89.15%),高于西米替丁组(59.90%)及三九胃泰颗粒剂组(55.31%)。三九胃泰方各极性萃取部位均可显著降低大鼠胃蛋白酶活力,同时增强血清中SOD的活力(P<0.05)。结论: 三九胃泰方不同极性萃取部位均具有显著的抗胃溃疡作用,其中石油醚萃取部位活性最强。  相似文献

Ethnopharmacological relevance

Caesalpinia pyramidalis Tul. (Fabaceae), known as “catingueira”, has been used in folk medicine in the treatment of various disorders such as gastritis, heartburn, indigestion, and stomach ache. However, the gastroprotective properties of this species have not yet been studied.

Materials and methods

The ethanol extract of Caesalpinia pyramidalis inner bark was used in rats via oral route, at the doses of 30, 100, and 300 mg/kg. The antiulcer assays were performed using the ethanol- and nonsteroidal anti-inflammatory drug-induced ulcer models. Gastric secretion parameters (volume, pH, and total acidity) were also evaluated by the pylorus ligated model, and the mucus in the gastric content was determined. The anti-Helicobacter pylori activity of the ethanol extract of Caesalpinia pyramidalis was performed using the agar-well diffusion and broth microdilution methods.

Results

The ethanol extract (30, 100, and 300 mg/kg) produced dose dependent inhibition (P<0.01) on the ulcer lesion index, the total lesion area, and the percentage of lesion area in the ethanol-induced ulcer model. The ethanol extract (30, 100, and 300 mg/kg) also reduced (P<0.001) the ulcer index in the indomethacin-induced ulcer model. In the model ligature pylorus, the treatment with Caesalpinia pyramidalis ethanol extract failed to significantly change the gastric secretion parameters. However, after treatment with the ethanol extract of Caesalpinia pyramidalis (30, 100, and 300 mg/kg), there was a significant increase (P<0.05) in mucus production. The ethanol extract showed anti-Helicobacter pylori activity, with inhibition halos of 12.0±1.7 mm at 10,000 μg/mL. The minimum inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) values of the ethanol extract were of 625 and 10,000 μg/mL, respectively.

Conclusions

Collectively, the present results suggest that the ethanol extract of Caesalpinia pyramidalis displays gastroprotective actions, supporting the folkloric usage of the plant to treat various gastrointestinal disturbances.  相似文献

ETHNOPHARMACOLOGICAL RELEVANCE: Solanum torvum (Solanaceae) is a plant currently used in Cameroon ethnomedicine for the treatment of stomach ailments. AIM OF THE STUDY: The present study was undertaken to determine the anti-ulcer potential of the aqueous and methanol extracts from the leaves of Solanum torvum. MATERIALS AND METHODS: The aqueous and methanol extracts from the leaves of Solanum torvum were tested orally at the doses of 250, 500 and 750mg/kg, on gastric ulcerations experimentally induced by HCl/ethanol, indomethacin, pylorus ligation and stress. The fractionation of the methanol extract through silica gel column chromatography produced seven different fractions (A-G) which were tested orally at the dose of 100mg/kg against HCl/ethanol-induced ulceration. RESULTS: The methanol extract at the dose of 750mg/kg produced 98.12, 99.16, 98.70 and 96.03% inhibition when gastric ulcerations were induced by HCl/ethanol, indomethacin, pylorus ligation and stress, respectively. The aqueous extract at the same dose produced 96.55, 96.86, 98.63 and 98.63% inhibition on ulcerations induced respectively by HCl/ethanol, indomethacin, pylorus ligation and stress. All the fractions of the methanol extract significantly inhibited ulcer formation. Fraction F which contains flavonoids and triterpens was the most active and exhibited an inhibitory percentage of 84.74. Both extracts significantly increased mucus production and reduced gastric acid secretion. CONCLUSIONS: The aqueous and methanol extracts of the leaves of Solanum turvum possess anti-ulcerogenic properties that may be due to cytoprotective mechanism. These results support the ethnomedical uses of the plant in the treatment of gastric ulcer.  相似文献

OBJECTIVE: To evaluated the gastroprotective effects of standardized aqueous extract of Ziziphus jujuba(Z. jujuba) stem bark against acidified ethanol-induced gastric ulcers as well as anti helicobacter pylori activity of the plant extract in rats.METHODS: Five groups of rats were orally pre-treated with normal saline(0.9%) as ulcer group, 150 mg/kg of ranitidine as positive control group, 100, 200 and 400 mg of standardized extract solution as the experimental groups. Two hours later, acidified ethanol solution was given by gavages in order to induce of gastric ulcer. The antibacterial effect of extract against clinical strains of Helicobacter pylori(H. pylori) was evaluated through disc diffusion test.RESULTS: The ulcer group exhibited significantly severe mucosal injury as compared with ranitidine or extract group which shows significant protective action against gastric mucosal injury. The extract showed no effect on H. pylori.CONCLUSION: The present study indicates that Z.jujuba stem bark extract had a potential antiulcer activity which might be due to its protective activity, providing a direct, protective effect on the gastric mucosa. Our study showed that anti-H. pylori activity was not among gastroprotective mechanism of Z. jujuba. Further pre-clinical and clinical investigations for evaluating natural active agents and efficacy of this plant are recommended.  相似文献

Methanolic extract of Phyllanthus amarus Shum & Thonn (Euphorbiaceae) 50, 200, and 1000 mg/kg body weight significantly inhibited gastric lesions, induced by intragastric administration of absolute ethanol (8 ml/kg). Mortality, increased stomach weight, ulcer index, and intraluminal bleeding were reduced significantly by Phyllanthus amarus. Biochemical analysis indicated that reduced glutathione (GSH) of gastric mucosa produced by ethanol administration was significantly elevated by treatment with Phyllanthus amarus extract. Aqueous and methanol extracts of Phyllanthus amarus produced an inhibition of rat paw edema up to 42% compared to control in 3h and continued up to 8h. Anti-oxidant activity of the extract as well as presence of tannins in the extract may be responsible for these observed activities.  相似文献

目的观察白芨多糖对实验性小鼠和大鼠胃黏膜损伤的影响。方法采用水提醇沉的方法制备白芨多糖。采用冷水应激、乙醇诱导法制作小鼠胃溃疡模型,幽门结扎等方法制作大鼠胃溃疡模型。试验动物各分为5组:白芨多糖低剂量组、白芨多糖中剂量组、白芨多糖高剂量组、对照组和模型组,连续给药3 d,造模后处死动物并观察胃溃疡指数。结果白芨多糖对冷水应激小鼠胃黏膜损伤、乙醇诱导的小鼠胃黏膜损伤及结扎型大鼠胃黏膜损伤均具有保护作用。结论白芨多糖对实验性动物胃溃疡具有保护作用。  相似文献

Aim of the study

Momordica charantia L. (Cucurbitaceae) commonly known as ‘bitter gourd’ is a multi purpose herb cultivated in different parts of the world for its edible fruits. The objective of the present study was to evaluate the effect of standardized methanolic extract of Momordica charantia L. fruits on gastric and duodenal ulcers.

Materials and methods

The effect was evaluated in acetic acid induced chronic gastric ulcer, pylorus ligation induced gastric ulcer, ethanol induced gastric ulcer, stress induced gastric ulcer, indomethacin induced gastric ulcer and cysteamine induced duodenal ulcer model. The extract was administered orally at two different doses of 100 mg/kg and 500 mg/kg.

Results

The healing of acetic acid induced gastric ulcer was increased by both doses of the extract. In pylorus-ligated rats, the extract showed significant decrease in ulcer index, total acidity, free acidity and pepsin content and an increase in gastric mucosal content. The extract also reduced the ulcer index in stress induced, ethanol induced and indomethacin induced gastric ulcers and cysteamine induced duodenal ulcer.

Conclusion

The methanolic extract of Momordica charantia L. fruit increases healing of gastric ulcer and also prevents development of gastric ulcers and duodenal ulcers in rats.  相似文献

Lipophilic extracts of Ginkgo biloba L. leaves were tested for their possible role on rodent models of depression and stress. Lipophilic extracts of Ginkgo leaves (LEG) at (50 and 100 mg/kg, p.o.) exhibited dose dependent, significant antidepressant activity in the behavioral despair test and learned helplessness rodent model of depression. The activities were comparable to that of imipramine (15 mg/kg) and EGb 761 (50 mg/kg). In the cold immobilization stress induced gastric ulcer model of stress, only the LEG showed a significant reduction in the ulcer index. GC-MS characterization of this bioactive extract was found to be rich in a group of 6-alkyl salicylates (6-AS), along with a fatty alcohol, fatty acids and cardanols. The n-heptadecenyl salicylate represented 60% of the 6-AS. Notable was the absence of dihydroxy alkylphenols which are linked to allergic reactions similar to the urushiols present in poison ivy. In commercial products of Ginkgo, these dihydroxy phenols as well as the favorable 6-AS are removed during enrichment of flavonol glycosides and terpenic lactones. The current findings suggest that intact carboxylic acid groups containing 6-AS are the bioactive components of the lipophilic extract of Ginkgo leaves with antidepressant and antistress activities.  相似文献

AIM OF THE STUDY: Dodonaea viscosa Linn. (Sapindaceae) is used as a medicinal herb by the tribes of Shola forest regions of Western Ghats. It is used for headaches, backaches, stomach pain, piles and simple ulcers. The present study was performed to evaluate the gastroprotective effect and acute toxicity of this plant in various experimental models. MATERIALS AND METHODS: Studies were performed in two different models (ethanol and indomethacin induced gastric ulcer) in wistar rats. Gastric protection was evaluated by measuring the ulcer index, gastric glutathione assay, alkaline phosphate assay and histopathological studies. Gastric secretion studies were done by pyloric ligation experiment. RESULTS AND CONCLUSIONS: Water and ethanol extract (500 mg/kg body weight) showed moderate activity compared to hexane extract. Hexane extract of Dodonaea viscosa dose dependently inhibited ethanol induced gastric lesions, causing 90% protection at 500 mg/kg, 81% protection at 250 mg/kg, and 70% protection at 125 mg/kg and it also dose dependently inhibited indomethacin induced gastric lesions, causing 92% protection at 500 mg/kg, 77% protection at 250 mg/kg, and 52% protection at 125 mg/kg. Bio‐guided fractionation of the total alcoholic extract of Convolvulus austro‐aegyptiacus was screened for its anti‐ulcerogenic activity, using an absolute‐ethanol‐induced ulcer model at 500 and 1000 mg/kg doses. Two compounds were isolated from the butanol extract of C. austro‐aegyptiacus and identified by 1H and 13C nuclear magnetic resonance as scopoletin and scopolin. The isolated compounds (50 mg/kg) showed a remarkable anti‐ulcerogenic activity because they exhibited control‐ulcer protection by 16.7% and 90.8%, respectively. The acute toxicity study showed that the extract is highly safe; the median lethal dose (LD50) was more than 4000 mg/kg. Moreover, the obtained results were confirmed by the sub‐chronic toxicity because the rats that have been administered 1000 mg/kg of the extract for 15 consecutive days showed no alteration in the liver and kidney functions. Pradosia huberi is a medicinal plant very common in the Amazonian forest population. The research interest in this plant is justifiable because of its potential medicinal value in gastritis and gastric ulcer mentioned in local folk medicine. In this paper, we evaluated the acute toxicity and antiulcerogenic effect of a hydroalcoholic extract (HAE) obtained from Pradosia huberi barks in rodents. No acute toxicological sign or symptom was observed in animals treated with the highest dose (5000 mg/kg, p.o.) of Pradosia huberi. In the HCl/EtOH-induced gastric ulcer model, HAE demonstrated significant inhibition of the ulcerative lesion index by 73% (500 mg/kg) and 88% (1000 mg/kg), respectively, in relation to the control value (p<0.05). The gastric damage induced by absolute ethanol in rats was effectively reduced by 84, 88 and 81% (250, 500 and 1000 mg/kg) when compared with the control group (p<0.01). In the NSAID-induced lesion model, HAE also showed antiulcerogenic effect with decrease in gastric lesions of 56% (250 mg/kg), 57% (500 mg/kg) and 67 % (1000 mg/kg) when compared with animals treated with vehicle (p<0.05). In the gastric ulcer induced by pylorus ligature model, the administration of HAE by oral and intraduodenal routes inhibited the gastric lesion index by 79 and 52% (500 mg/kg), respectively. HAE administered orally or intraduodenally was able to change gastric juice parameters (pH, volume and acid output) as well as those treated with cimetidine. The treatment with HAE (p.o.) significantly increased gastric volume, the pH values and promoted reduced acid output (p<0.01). By comparative effect (intraduodenal and oral route), we observed that HAE was better for local activity in gastric mucosa than in systemic action. HAE also has a non-specific activity when found to be the inhibitor of intestinal motility (p>0.01). The mechanism of action of HAE did not seem to be related to the NO-inhibitor but showed the participation of endogenous sulphydryl group in the gastroprotective action.  相似文献
