目的:利用熔融法制备布洛芬Kollidon CL和Kollidon CL-SF固体分散体,提高布洛芬的体外溶出性质。方法:以Kollidon CL和Kollidon CL-SF为载体,利用熔融法制备难溶性药物布洛芬固体分散体,并进行体外溶出研究。采用差示扫描量热分析、粉末X射线衍射分析及扫描电镜观察对制备的固体分散体进行物相鉴别。结果:熔融法制备的布洛芬-Kollidon固体分散体中,布洛芬以无定形态存在,体外溶出速率显著提高,5 min时以Kollidon CL和Kollidon CL-SF为载体的固体分散体中布洛芬累积溶出度是67.1%和67.6%,分别是布洛芬原药的17.8倍和17.9倍。结论:固体分散体技术是提高布洛芬溶出性质的一种有效可行的方法。  相似文献

目的运用固体分散技术制备杨梅素固体分散体并提高其体外溶出速率。方法选用PEG6000和PVPK30为载体,采用溶剂法和溶剂-熔融法制备杨梅素固体分散体,采用紫外分光光度法进行含量测定,并进行溶解度、体外溶出试验。结果两种载体的固体分散体均能增加药物的溶解度和溶出速率,杨梅素在载体中以高度分散状态存在。结论以PVPK30为载体的杨梅素固体分散体体外溶解度和溶出速率明显提高。杨梅素固体分散体能显著提高杨梅素的溶出速率。  相似文献

目的:制备马来酸氟吡汀-PEG 6000固体分散体以加快药物的体外溶出速度。方法:以PEG 6000为药物载体,采用熔融法制备马来酸氟吡汀固体分散体,采用X-射线衍射法和差示扫描量热法(DSC)观察药物在载体中的存在状态。结果:马来酸氟吡汀以分子状态存在于固体分散体中;药物与载体的比例为1:4时,所制备的固体分散体具有最高的溶出度。结论:固体分散体能显著提高药物溶出度和溶出速率。  相似文献

目的将难溶性微管蛋白抑制剂SUD-35制备成固体分散体,以增加其溶解度及溶出速率。方法以聚乙二醇6000为载体,溶剂-熔融法制备SUD-35固体分散体。采用差示扫描量热分析与X-射线衍射观察药物在载体中的存在状态,并进行溶解度和体外溶出度研究。采用MTT法对SUD-35固体分散体对小鼠白血病L1210细胞药效进行测定。结果 SUD-35固体分散体中SUD-35的溶解度和溶出速率相对原料药和物理混合物均有明显提高,差示扫描量热分析与X-射线衍射结果显示SUD-35以无定型状态存在于固体分散体中,细胞药效结果显示SUD-35固体分散体对小鼠白血病L1210细胞增殖抑制率强于SUD-35纯药。结论聚乙二醇6000为载体制备SUD-35固体分散体,可显著提高SUD-35的溶解度及溶出速率。  相似文献

目的 :制备卡维地洛固体分散体 ,提高其溶解度和溶速率。方法 :以聚乙烯吡咯烷酮 (PVP)、聚乙二醇 -6000(PEG -6000)为载体 ,以溶剂法和熔融法制备固体分散体 ,并进行体外溶出度研究。结果 :载体比例越大 ,药物溶出愈快 ;载体比例愈小 ,差异愈显著。载体为PVP所制固体分散体的体外溶出行为总体优于载体为PEG -6000的固体分散体。结论 :本试验所制卡维地洛固体分散体能加速体外溶出 ,提高生物利用度 ,可用于制备高效制剂  相似文献

目的 将蒙花苷制备成固体分散体,提高其体外溶出度。方法 分别以不同辅料为载体,采用溶剂法、熔融法或溶剂–熔融法制备蒙花苷固体分散体,以蒙花苷的体外溶出度为指标,考察蒙花苷固体分散体的最佳处方和制备工艺。并采用差示热分析法和X射线粉末衍射法对蒙花苷固体分散体物相进行表征。结果 蒙花苷与聚乙二醇6000质量比为1∶9,采用熔融法制备的固体分散体的体外溶出最好。结论 以聚乙二醇6000为载体制备的蒙花苷固体分散体可以显著提高蒙花苷的体外溶出度,且以过饱和的固态溶液或无定型状态均匀分布在载体中。  相似文献

目的 制备茴三硫固体分散体,提高其溶解度和溶出速率.方法 以不同分子量不同比例的聚乙二醇为载体,以熔融法制备固体分散体,并进行体外溶出度研究和DSC扫描.结果 茴三硫与聚乙二醇可形成低共熔物,并使溶出度大大增加,载体比例越大,药物溶出愈快.结论 本试验所制茴三硫固体分散体能加速体外溶出,体外溶出与载体分子量无关,但与固体分散体载体比例有关。  相似文献

The aim of the study was in vitro evaluation of piroxicam solid dispersions containing hydroxypropyl methylcellulose acetate succinate (HPMCAS-LF, -HF) as a carrier. Binary (piroxicam-HPMCAS) and ternary (piroxicam-HPMCAS-Carbopol 940) solid dispersions were prepared by spray-drying method. The morphological characteristics were investigated by scanning electron microscopy. X-ray diffraction and differential scanning calorimetry were employed to study physical and chemical properties. In vitro release was studied using a flow-through cell technique. Studies of dissolution rate of piroxicam from solid dispersions were carried out in comparison with corresponding physical mixtures and drug alone. The dissolution profiles depend on the presence of Carbopol 940 in solid dispersions.  相似文献

In this study, solid dispersion for carvedilol was prepared by using spray-drying method. Solid dispersions were formulated with carvedilol and Eudragit RS and hydroxypropyl methylcellulose to control the dissolution rates of carvedilol. Scanning electron microscope was used to analyze surface of solid dispersion samples. Differential scanning calorimetry and powder X-ray diffraction were used to analyze the crystallinity of solid dispersions. Fourier transform infrared spectroscopy was used to analyze the change in chemical structure of solid dispersions. The release behavior of solid dispersion analyzed at simulated gastric fluid (pH 1.2) in in vitro study. The dissolution rate of carvedilol was higher than active pharmaceutical ingredient. In conclusion, we can control the dissolution rate by solid dispersion using biomedical polymers.  相似文献

目的:将难溶性药物间尼索地平制备成固体分散体,以增加其溶解度及体外溶出度。方法:以泊洛沙姆为载体,共沉淀法制备间尼索地平固体分散体。采用差示扫描量热分析(DSC)方法鉴别药物在载体中的存在状态,并进行溶解度和体外溶出度研究。结果:DSC显示间尼索地平与泊洛沙姆形成了低共熔物,间尼索地平原料药及其与泊洛沙姆不同比例的固体分散体(1∶3、1∶5、1∶7)的溶解度分别为0.89、4.50、15.35、23.03mg·L-1,120min时的累积溶出百分率分别为26.80%、38.57%、41.38%、45.92%,固体分散体的溶出度高于同比例的物理混合物。结论:以泊洛沙姆为载体制备间尼索地平固体分散体,可增加药物的体外溶出度和溶解度。  相似文献

Solid dispersions using Pluronic F-68 as a carrier were studied for improving the dissolution and bioavailability of ABT-963, a poorly water-soluble compound. The solid dispersions were prepared either by evaporation of the ethanol solutions containing ABT-963 and Pluronic, or by cooling the hot melt of the drug in the carrier. The dispersions were characterized using differential scanning calorimetry, powder X-ray diffractometry, scanning electron microscopy, elemental mapping, and by constructing the melting point phase diagram. In vitro dissolution and in vivo oral bioavailability in fasted dogs were compared for the solid dispersion and a conventional IR capsule formulation. Results showed that, at a composition of approximately 7.5%, ABT-963 formed a eutectic mixture with Pluronic F-68. Both the drug and the polymer were crystalline in the solid dispersion with a wide range of composition of each component. The solid dispersion substantially increased the in vitro dissolution rate of ABT-963. Dosing of the dispersion to fasted dogs resulted in a significant increase of oral bioavailability compared with the conventional IR capsule formulation. These results show that solid dispersion is a promising approach for developing ABT-963 drug products.  相似文献

目的:制备布洛芬固体分散体,以增加布洛芬的溶出度并掩盖其苦味。方法:取布洛芬原料药与丙烯酸树脂Eudragit EPO,以1∶1.5(w/w)混合,采用热熔挤出法制备布洛芬固体分散体。用差示扫描量热法和粉末X射线衍射法分析布洛芬在Eudragit EPO中的分散状态。测定固体分散体、物理混合物和市售布洛芬片剂的溶出度,并评价布洛芬固体分散体的掩味效果。结果:布洛芬晶体结构的特征峰在差示扫描量热和粉末X射线衍射图中消失。在磷酸盐缓冲液中,固体分散体的溶出速度大于物理混合物和布洛芬片。志愿者对布洛芬固体分散体的味觉评价优于物理混合物和布洛芬原料。结论:热熔挤出法制备的Eudragit EPO固体分散体能增加布洛芬的溶出度,并有明显的掩味效果。  相似文献

Ezetimibe (EZE), a water insoluble drug, depicts variable bioavailability. The objective of the present investigation was to improve dissolution characteristics of EZE, which might offer improved bioavailability. The solid dispersions were prepared using poloxamer 407 (L 127) and polyvinyl pyrrolidone by melt and solvent method, respectively. Phase solubility studies indicated linear relationship between drug solubility and carrier concentration. In vitro release studies revealed improvement in the dissolution characteristics of EZE in solid dispersions. Solid dispersion with L 127 gave better rate and extent of dissolution. The best fit model indicating the probable mechanism of drug release from solid dispersions was found to be Korsemeyer–Peppas. The results of characterization of solid dispersions by Fourier transform infrared spectroscopy, differential scanning calorimetry, and powder X-ray diffraction revealed reduction in drug crystallinity which might be responsible for improved dissolution properties. The tablets of solid dispersion, containing L 127 prepared by direct compression, exhibited better drug release as compared to marketed formulation.  相似文献

用溶剂法制备紫杉醇-PVP固体分散体,对其溶解度及体外溶出特性进行考察并对物相进行鉴定。采用溶剂法制备紫杉醇-PVP固体分散体,对固体分散体中紫杉醇的溶解度和溶出率进行测定,研究固体分散体的溶出性质。同时,利用差热分析(Differential scanning calorimetry,DSC)、粉末X衍射(X-ray powder diffractometry,PXRD)、扫描电镜(Scanning electron microscopy,SEM)等方法对其进行物相鉴定。采用SRB法对紫杉醇-PVP固体分散体对SKOV-3细胞药效进行测定。紫杉醇-PVP固体分散体中紫杉醇的溶解度和溶出速率相对其原料药和物理混合物均有了明显的提高;热差分析及粉末X衍射结果表明固体分散体中紫杉醇呈非结晶形式;扫描电镜下固体分散体中无紫杉醇晶体。细胞药效结果表明紫杉醇-PVP固体分散体的细胞药效强于紫杉醇纯药。采用溶剂法制备的紫杉醇-PVP固体分散体可显著提高紫杉醇的溶解度和溶出速度。  相似文献

目的:利用固体分散技术制备替硝唑固体分散体,增加替硝唑溶解度和溶出速度。方法:以聚乙二醇(PEG)为载体材料,采用溶剂-熔融法制成固体分散体,测定表观溶解度,进行体外溶出试验,并采用差示扫描量热(DSC)法鉴别药物在固体分散体中的存在状态。结果:替硝唑的溶出度和表观溶解度随PEG的比例不同而不同,且溶出度随载体用量增加而增加。固体分散体的DSC曲线中替硝唑药物的特征熔融峰消失。结论:所制得的固体分散体能明显提高替硝唑的溶出度和表观溶解度。  相似文献
