Ethnopharmacological Relevance

Pupalia lappacea is a medicinal plant found in savannah and woodland localities and forest path sides from Senegal to Southern Nigeria. It has been used in the management of diarrhoea in Nigerian traditional medicine. This study was designed to evaluate the antidiarrhoeal activity of the hydroethanolic leaf extract of Pupalia lappacea (PL).

Materials and methods

The antidiarrhoeal activity of PL was evaluated using the normal and castor oil-induced intestinal transit, castor oil-induced diarrhoea, gastric emptying and intestinal fluid accumulation tests in rodents.

Results

PL (100–400 mg/kg, p.o.) produced a significant dose-dependent decrease in normal and castor oil-induced intestinal transit compared with the control group (distilled water 10 ml/kg, p.o.). This effect was significantly (P<0.05) inhibited by pilocarpine (1 mg/kg, s.c.) but not by yohimbine (10 mg/kg, s.c.), prazosin (1 mg/kg, s.c.), or propranolol (1 mg/kg, i.p.). The extract produced a dose-dependent and significant increase in the onset of diarrhoea. PL (100–400 mg/kg) also reduced the diarrhoea score, number and weight of wet stools. The in-vivo antidiarrhoeal index (ADIin vivo) of 56.95% produced by the extract at the dose of 400 mg/kg was lower compared to that produced by loperamide 5 mg/kg (77.75%). However, PL (400 mg/kg) significantly increased gastric emptying in rats but significantly reduced the volume of intestinal content in the intestinal fluid accumulation test. Phytochemical analysis of the extract revealed the presence of alkaloids, saponins, and fixed oils and fats. The acute toxicity studies revealed that the extract is relatively safe when given orally; no death was recorded at a dose of 10 g/kg.

Conclusion

Results showed that the hydroethanolic leaf extract of Pupalia lappacea possesses antidiarrhoeal activity possibly mediated by antimuscarinic receptor activity.  相似文献

Ethnopharmacological relevance

The aqueous root extract of Sansevieraliberica (Agavaceae), SL, is used in Traditional African Medicine (TAM) for the treatment of diarrhoea. However, the scientific basis for this usage has not been established.

Aim of the study

To evaluate the antidiarrhoeal activity of SL using various pharmacological models.

Materials and methods

The intestinal transit, castor oil induced diarrhoea, enteropooling, and gastric emptying methods were used in this study.

Results

SL (25–400 mg/kg, p.o.) produced significant (P < 0.05) dose dependent reduction in propulsive movement in both the normal and castor oil induced intestinal transit tests in mice. Peak effect was elicited at 200 mg/kg but this effect was lower than that produced by morphine (10 mg/kg, s.c.). The effect of SL on castor oil induced intestinal transit was antagonized by isosorbide dinitrate, IDN (150 mg/kg, p.o.) but not by yohimbine (1 mg/kg, s.c.). In the castor oil induced diarrhoea test, SL significantly delayed the onset and decreased the frequency and severity of diarrhoea. The effect at 200 mg/kg was comparable to that of morphine and was reversed by IDN. SL at the dose of 200 mg/kg significantly reduced the volume of intestinal secretion induced by castor oil but produced no effect on gastric emptying.The extract was practically nontoxic administered p.o. The LD50 was 631 mg/kg given i.p. Phytochemical analysis revealed the presence of oils, reducing sugars, alkaloids, saponins, anthraquinones, and tannins in the extract.

Conclusion

The results obtained in this study suggest that the aqueous root extract of Sanseviera liberica possesses antidiarrhoeal property due to inhibition of gastrointestinal propulsion and fluid secretion, possibly mediated through inhibition of the nitric oxide pathway. This justifies the use of the plant extract in TAM for the treatment of diarrhoea.  相似文献

In our search for plants useful in the treatment of diarrhoea, we investigated the ethyl acetate extract of Baphia nitida (BN) using intestinal transit, enteropooling and gastric emptying tests in mice and rats. In the castor oil intestinal transit test, BN produced a significant (P<0.05) dose dependent decrease in propulsion with peristaltic index (PI) values of 56.85+/-6.76, 36.84+/-3.04 and 31.98+/-2.60%, respectively at doses of 100, 200 and 400mg/kg vs. 89.33+/-6.28% for control. The effect at 400mg/kg was significantly lower than that of morphine, 10mg/kg, s.c. (20.29+/-3.78%), and was antagonized by isosorbide dinitrate, IDN (150mg/kg, p.o.) but not by yohimbine (1mg/kg, s.c.). This effect was not potentiated by atropine (1mg/kg, s.c.). In the castor oil-induced diarrhoea test, BN produced a significant increase in onset of diarrhoea (103.40+/-8.74, 138.80+/-17.04 and 174.8+/-29.04min, 100 to 400mg/kg, vs. 47.60+/-8.76min for control and 226.10+/-12.57min for morphine). The severity of diarrhoea (diarrhoea score) was dose dependently reduced (19.00+/-2.26, 17.04+/-1.89, 15.00+/-2.05, 100 to 400mg/kg, vs. 31.40+/-2.11 for control and 7.7+/-2.2 for morphine). This effect was not antagonized by IDN or yohimbine. The effect on severity was, however, potentiated by atropine. BN also reduced the number and weight of wet stools but did not have any significant effect on intestinal fluid accumulation and gastric emptying. Results obtained suggest that the ethyl acetate extract of Baphia nitida is endowed with antidiarrhoeal activity possibly mediated by interference with the l-arginine nitric oxide pathway and synergistic with antagonistic action on muscarinic receptors.  相似文献

Aim of the study

To evaluate the antidiarrhoeal properties of Alstonia congensis leaves claimed to be effective for the treatment of diarrhoea by traditional healers during our ethnopharmacological investigation conducted in Kinshasa, Democratic Republic of Congo.

Materials and methods

The aqueous extract (decoction), and the 80% hot methanol extract (Soxhlet extraction) were obtained. This last extract was fractionated. The antidiarrhoeal activity was evaluated using castor oil and magnesium sulphate-induced diarrhoea in animals. The potential antibacterial activity of all samples was also assessed in vitro.

Results

At all oral doses of 100 and 200 mg/kg body weight, all A. congensis samples showed significant and dose-dependent antidiarrhoeal activity in treated Wistar rats characterised by significant increase of onset time and decrease of all other diarrhoeal parameters at various degrees compared to untreated groups in both models. At the highest oral dose of 200 mg/kg bodyweight, the 80% hot methanol and aqueous extracts produced 79.8±2.1% and 78.6±0.5%, and 75.0±2.1% and 71.4±2.1% inhibition of defecation and diarrhoea respectively against castor oil-induced diarrhoea, and 75.0±1.2% and 73.3±1.2% inhibition of diarrhoea respectively against magnesium sulphate-induced diarrhoea. The 80% hot methanol and aqueous detannified extracts showed low activity (42–47% inhibition of defecation and/or diarrhoea in both tests) suggesting that tannins may be responsible for the observed activity. At the same oral doses, the total alkaloid extract, the chloroform soluble fraction rich in alkaloids, the 80% methanol and the alkaline aqueous soluble subfractions produced more than 50% inhibition of defecation and/or diarrhoea in both tests. From the antibacterial testing in vitro, results indicated that all A. congensis samples exhibited an antibacterial activity mainly against bacteria implicated in diarrhoea with MIC and MBC values in the range of 15.6–500 μg/ml. The most active samples were the aqueous (decoction) and the 80% hot methanol dried extracts, the chloroform subfraction rich in alkaloids and the total alkaloid extract (MIC: 15.7–125 μg/ml, MBC: 31.2–250 μg/ml). Proteus varibilis was found to be the most resistant microorganism.

Conclusion

These reported results can partly support and justify the traditional use of extracts from Alstonia congensis leaves for the treatment of diarrhoea in tradittional medicine.  相似文献

The effect of the aqueous extract of Mezoneuron benthamianum (MB) on experimentally induced diarrhoea, intestinal propulsive movement (IPM) and intestinal fluid accumulation (enteropooling) were investigated in rats and mice. The extract (400, 800 and 1600 mg/kg, orally) produced a significant (p<0.05) and dose-dependent reduction in propulsion in the castor oil-induced intestinal transit in mice. The mean peristaltic index (%) for these doses of extract, control, (distilled water, 10 ml/kg, p.o.) and morphine, (10 mg/kg, s.c.) were 73.48, 69.34, 57.27, 89.93 and 31.56, respectively. The effect of the extract at the highest dose was significantly (p<0.05) lower than that of the standard drug. This effect was antagonised by yohimbine (1mg/kg, s.c.). In a dose-dependent manner, the extract delayed the onset of diarrhoea, produced a significant decrease in the frequency of defaecation, severity of diarrhoea and protected the mice treated with castor oil. Total diarrhoea scores were 12.0+/-0.63, 10.3+/-2.06, 8.5+/-2.15, 7.1+/-0.91 and 5.8+/-0.79 for control, extract (400, 800 and 1600 mg/kg) and morphine, respectively. The extract significantly decreased the volume (ml) of intestinal fluid secretion induced by castor oil (1.75+/-0.02 to 0.93+/-0.04) compared with 1.90+/-0.05 for control. The inhibitory effect on fluid accumulation by the extract was also attenuated by yohimbine (1.0 mg/kg). Preliminary phytochemical screening revealed the presence of flavonoids, tannins, cardiac glycosides, anthraquinones and saponins. Administration of the extract up to 2 g/kg (orally) did not produce any toxic effect in the acute toxicity studies in mice. The LD(50) of the extract when given intraperitoneally was 1021.31 mg/kg. The results obtained show that MB possesses anti-diarrhoeal activity due to its inhibitory effects on gastrointestinal propulsion and intestinal fluid accumulation. The antagonistic actions of yohimbine in the experiments suggest a role for the a(2)-adrenergic receptor system.  相似文献

目的: 分析木果楝Xylocarpus granatum果实中脂肪酸成分及相对含量。 方法: 采用气相色谱-质谱联用技术,对木果楝果实中的脂肪酸以甲醇回流提取,石油醚萃取,硫酸甲醇甲酯化后进行测定,用面积归一化法得出各脂肪酸的相对含量。 结果: 从木果楝中检出6种饱和脂肪酸和5种不饱和脂肪酸。其中饱和脂肪酸占总含量的17.63%,以棕榈酸含量最高(相对含量13.36%);不饱和脂肪酸占总含量的81.97%,以亚麻酸含量最高(相对含量73.86%)。 结论: 为木果楝的药用价值开发提供了科学依据。  相似文献

Xylocarpus granatum J. K?nig (Meliaceae), commonly known as 'dhundul', is a Bangladeshi mangrove tree, and well distributed in a number of other countries of south-east Asia, Australia and east Africa. Traditionally, X. granatum has been used as an astringent and febrifuge, and also for the treatment of fever, malaria, thrush, cholera, dysentery and diarrhoea in many countries including Bangladesh. Two limonoids, gedunin and 1alpha-hydroxy-1,2-dihydrogedunin, the latter being new, have been isolated from the bark of Xylocarpus granatum by reversed-phase preparative HPLC, and the structures were confirmed by spectroscopic means. The cytotoxic potential of gedunin has been evaluated by the Promega's CellTiter 96 non-radioactive cell proliferation assay using the CaCo-2 colon cancer cell line (IC(50) = 16.83 microM). A summary of the biological activities of gedunin reported to date is also presented.  相似文献

Ethnopharmacological relevance

Geranium incanum Burm. f. (Geraniaceae) is used in South Africa especially in rural communities by traditional medicine practitioners to treat diarrhoea. However, scientific evidence does not exist in any literature to corroborate the claim of therapeutic success of the plant species in diarrhoea.

Aim of study

The study intended to investigate the antidiarrhoeal activity of the leaf aqueous extract of Geranium incanum in mice.

Materials and Methods

Castor oil induced diarrhoeal test was used to assess the antidiarrhoeal activity of Geranium incanum. Gastrointestinal tract transit of charcoal meal test was used to assess the antipropulsive activity of the plant extract while the acute toxicity study and phytochemical analysis were carried out using well established protocols and methods.

Results

The antidiarrhoeal activity of Geranium incanum was investigated by studying the effect of leaf aqueous extract of the plant species on castor oil-induced diarrhoea in mice. The leaf aqueous extract of Geranium incanum significantly reduced faecal output in castor oil -induced diarrhoea and also significantly reduced the number of diarrhoeal episodes. Geranium incanum significantly delayed the onset of diarrhoea induced by castor oil and significantly reduced the number of animals exhibiting diarrhoea. Loperamide, a standard antidiarrhoeal drug, produced similar effects to the leaf aqueous extract of Geranium incanum on castor oil-induced diarrhoea. Both Geranium incanum and loperamide significantly reduced the intestinal propulsion of charcoal meal in mice. The phytochemical analysis of the leaves revealed the presence of tannins, saponins particularly steroidal saponin, and flavonoids. The LD50 of the plant species obtained was greater than 4000 mg/kg (p.o.).

Conclusion

The data obtained indicate that the leaf aqueous extract of Geranium incanum has both antidiarrhoeal and antipropulsive activities The data also show that the plant material given orally may be safe and/or non toxic in mice. However, further investigation on the acute toxicity and on the mechanism of the antidiarrhoeal effect of the plant species needs to be carried out.  相似文献

Ethnopharmacological relevance

The traditional use of Punica granatum has been reported to regulate urine discharge and controls the burning sensation of urine.

Materials and methods

Animals model of calcium oxalate urolithiasis was developed in male rats by adding ethylene glycol 0.75% in drinking water. The Punica granatum chloroform extract (PGCE) and Punica grantum methanol extract (PGME) orally at 100, 200 and 400 mg/kg, respectively, were administered along with ethylene glycol for 28 days. On 28 day, 24 h urine was collected from individual rats and used for estimation of urine calcium, phosphate and oxalate. The serum creatinine, urea and uric acid levels were estimated in each animal. The kidney homogenate was used for the estimation of renal oxalate contents. The paraffin kidney sections were prepared to observe the CaOx deposits.

Results

The ethylene glycol control (Gr.-II) had significant (P < 0.001 vs. normal) increase in levels of urine oxalate, calcium and phosphate, serum creatinine, urea and uric acid and renal tissues oxalates, as compared to normal (Gr.-I). The paraffin kidney sections show significant histopathological changes. The treatment of PGCE and PGME at 100, 200 and 400 mg/kg doses, significantly (P < 0.001 vs. control) decreased the urine oxalate, calcium and phosphate, renal tissue oxalates and serum creatinine, urea and uric acid, in EG induced urolithiasis after 28 days.

Conclusions

The PGCE and PGME at the doses of 400 mg/kg, found to be more effective in decreasing the urolithiasis and regeneration of renal tissues in male rats.  相似文献

Topical anti-inflammatory activity of Dalbergia lanceolaria Linn. bark ethanol extract was demonstrated in albino mice using TPA-, EPP- and AA-induced ear edema models. The systemic activity of extract was confirmed using acute and sub-acute anti-inflammatory models in albino rats. The ethanol extract exhibited significant systemic anti-inflammatory activity in Carrageenan-induced rat paw edema, by inhibition of histamine and prostaglandin phases of acute inflammation. The extract also showed significant activity against turpentine-induced exudative changes and no activity against granular tissue formation in cotton pellet-induced granuloma in albino rats. Thus, resultant anti-inflammatory activity might be due to effects on several mediators involving cyclo-oxygenase pathway resulting in prostaglandin formation and leukocyte migration from the vessels.  相似文献

The bark of M. urundeuva (Anacardiaceae) used in popular medicine as an antidiarrhoeal agent has been investigated for its effects on castor oil-induced diarrhoea and on intestinal motility. In rats, the ethanol extract of stem bark significantly inhibited the castor oil-induced diarrhoea, small intestinal transit and accumulation of fluid volume at an oral dose of 400 mg/kg. Furthermore, the plant extract caused marked inhibition of the contractile responses evoked by acetylcholine and histamine on guinea-pig ileum in vitro. These results point to a possible antidiarrhoeic effect of M. urundeuva bark extract since inhibition of intestinal motility and secretion can greatly control clinical diarrhoea. The present study was undertaken to evaluate the contraceptive efficacy of methanol extract of Dendrophthoe falcata Ettingsh (family-Loranthaceae), stem in male albino rats as reported in folk remedies. Adult proven fertile male rats were gavaged methanol extract of D. falcata stem at 50, 100 and 200mg/rat/day for 60 days. The activity was compared with standard drug, i.e. Lonidamine. On day 61 the animals were autopsied and the testes, epididymides, seminal vesicle and ventral prostate were dissected out and weighed. Sperm motility and density and serum testosterone level were assessed. The sperm motility and density were significantly reduced. The histoarchitecture of testes revealed degenerative changes in the seminiferous tubules, arrest of spermatogenesis at the stage of round spermatid. Serum testosterone levels were decreased significantly in all treatment groups. It is concluded that D. falcata methanol stem extract showed a significant effect on fertility in male rats as reported in folk remedies.  相似文献

Ethnopharmacological relevance

Dorstenia arifolia is a plant that has been used in the folk medicine to produce hypnotic, sedative and ansiolitic effects but the pharmacological properties have not yet been studied. In addition, the smoke of its rhizome is reputed to induce lethargic sensation.

Aims of the study

The present study investigated possible activities of the methanol extract (ME) of Dorstenia arifolia rhizome on the central nervous system (CNS).

Materials and methods

ME was tested for sedative, hypnotic and anticonsulsant effects using locomotor activity evaluation, pentobarbital-induced sleeping time and pentylenetetrazol (PTZ)-induced convulsion, respectively.

Results

Intraperitoneal administration of ME (10 and 50 mg/kg) significantly decreased locomotor activity from 205.2 ± 25.6 movements/min (DMSO) to 112.1 ± 18.4 (P < 0.05) and 114.9 ± 16.9 (P < 0.05), respectively. Flumazenil (10 mg/kg), an antagonist of GABAA receptor, prevented the ME-induced sedation. Treatment with ME (50 mg/kg) significantly increased the duration of pentobarbital-induced sleeping time from 41.0 ± 2.3 to 57.9 ± 2.9 min (P < 0.05). The latencies to seizures after intraperitoneal injection of PTZ was recorded and compared between groups. ME promoted a significant protection of PTZ-induced seizures and mortality in a dose-dependent manner.

Conclusions

Our findings indicate that ME of Dorstenia arifolia rizhome has pronounced central effects, and that the sedative and anticonvulsant activities may be related to a facilitation of the GABAergic transmission.  相似文献

Aim of the study

This study was aimed at investigating the antimicrobial activity of the methanolic extract (MMB) and compounds isolated from the stem bark of Morus mesozygia, namely 3β-acetoxyurs-12-en-11-one (1), moracin Q (2), moracin T (3), artocarpesin (4), cycloartocarpesin (5), moracin R (6), moracin U (8), moracin C (9), and moracin M (10).

Materials and Methods

The liquid microdilution assay was used in the determination of the minimal inhibitory concentration (MIC) and the minimal microbicidal concentration (MMC), against nine bacterial and two fungal species.

Results

The results of the MIC determination showed that the compounds 3, 4, 8 and 9 were able to prevent the growth of all tested microbial species. All other samples showed selective activities. Their inhibitory effects were noted on 90.9% studied organisms for the crude extract, 81.8% for compound 6, 72.7% for compound 10, 63.6% for compound 1, 54.5% for compound 5, and 45.5% for compound 2. The lowest MIC value of 39 μg/ml was obtained with the crude extract against Escherichia coli. The corresponding value for compounds (5 μg/ml) was registered with compound 9 on Shigella dysenteriae and compound 3 on E. coli, S. dysenteriae, Pseudomonas aeruginosa, Salmonella typhi and Bacillus cereus. The lowest MIC value (39 μg/ml) observed with the crude extract (on E. coli) was only eightfold greater than that of gentamycin used as reference antibiotic (RA) while the corresponding value (5 μg/ml) recorded with compounds 3 and 9 was equal to that of RA on the corresponding microorganisms.

Conclusions

The obtained results highlighted the interesting antimicrobial potency of M. mesozygia as well as that of the studied compounds, and provided scientific basis for the traditional use of this species.  相似文献

Ethnopharmacological relevance

The plant, Vismia rubescens (Guttiferae) is popularly used in Cameroon and in several parts of Africa as febrifugal and for the treatment of various microbial infections (skin diseases, diarrhoea and venereal diseases).

Aim of the study

This study was mapped out to evaluate the antimicrobial activities of the methanol extract and compounds from the stem bark of Vismia rubescens.

Materials and methods

Structures of the compounds obtained after column chromatography of the methanol-soluble fraction were determined by spectroscopy and in comparison with published data. The broth micro-dilution method was used to evaluate the antimicrobial activities against three bacteria species (Salmonella typhi, Stahylococcus aureus and Pseudomonas aeruginosa) and four yeast species (Candida albicans, Candida tropicalis, Candida parapsilosis and Cryptococcus neoformans).

Results

Chemical analysis of the methanol extract from the stem bark of Vismia rubescens yielded five known compounds 1,4,8-trihydroxyxanthone (1), 1,7-dihydroxyxanthone (2), physcion (3), friedelin (4) and friedelanol (5). The crude extract and compounds 1, 2 and 3 exhibited both antibacterial and antifungal activities that varied between the microbial species (MIC = 3.12–1000 μg/ml). Compounds 2 and 3 were the most active (MIC = 3.12–100 μg/ml) while Stahylococcus aureus and Pseudomonas aeruginosa were sensitive to all the tested compounds. The antimicrobial activity of this plant as well as that of compounds 1 and 2 is being reported here for the first time.

Conclusion

These results provide promising baseline information for the potential use of this plant as well as some of the isolated compounds in the treatment of skin diseases and diarrhoea.  相似文献

Ethnopharmacological relevance

Caesalpinia pyramidalis Tul. (Fabaceae), known as “catingueira”, has been used in folk medicine in the treatment of various disorders such as gastritis, heartburn, indigestion, and stomach ache. However, the gastroprotective properties of this species have not yet been studied.

Materials and methods

The ethanol extract of Caesalpinia pyramidalis inner bark was used in rats via oral route, at the doses of 30, 100, and 300 mg/kg. The antiulcer assays were performed using the ethanol- and nonsteroidal anti-inflammatory drug-induced ulcer models. Gastric secretion parameters (volume, pH, and total acidity) were also evaluated by the pylorus ligated model, and the mucus in the gastric content was determined. The anti-Helicobacter pylori activity of the ethanol extract of Caesalpinia pyramidalis was performed using the agar-well diffusion and broth microdilution methods.

Results

The ethanol extract (30, 100, and 300 mg/kg) produced dose dependent inhibition (P<0.01) on the ulcer lesion index, the total lesion area, and the percentage of lesion area in the ethanol-induced ulcer model. The ethanol extract (30, 100, and 300 mg/kg) also reduced (P<0.001) the ulcer index in the indomethacin-induced ulcer model. In the model ligature pylorus, the treatment with Caesalpinia pyramidalis ethanol extract failed to significantly change the gastric secretion parameters. However, after treatment with the ethanol extract of Caesalpinia pyramidalis (30, 100, and 300 mg/kg), there was a significant increase (P<0.05) in mucus production. The ethanol extract showed anti-Helicobacter pylori activity, with inhibition halos of 12.0±1.7 mm at 10,000 μg/mL. The minimum inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) values of the ethanol extract were of 625 and 10,000 μg/mL, respectively.

Conclusions

Collectively, the present results suggest that the ethanol extract of Caesalpinia pyramidalis displays gastroprotective actions, supporting the folkloric usage of the plant to treat various gastrointestinal disturbances.  相似文献

Punica granatum L. (Punicaceae) whole fruit extracts, have been used in Cuban traditional medicine as an effective drug for the treatment of respiratory diseases. This species showed interesting anti-viral activity, e.g. aqueous or hydroalcoholic extracts of whole fruits have proved highly active against the influenza virus. However, some toxic properties of this extract have also been reported and, to date, very little is known about its genotoxic properties. In the present study, the genotoxicity of a Punica granatum (pomegranate) whole fruit extract was assessed using different in vitro and in vivo assays that detect DNA damage at different expression levels. Results from reversion and gene-conversion test in microorganisms, sister chromatid exchanges, micronuclei and sperm-shape abnormality assays in mice, clearly showed that the hydroalcoholic extract of P. granatum whole fruits is genotoxic when tested both in vitro and in vivo.  相似文献

The leaves of Achyranthes aspera L. (Amarenthacea) has been used traditionally for the treatment of wound in various parts of Ethiopia. However, the plant has not been explored scientifically for its wound healing activity. Therefore, this study was designed to investigate the wound healing activity of methanol extract of Achyranthes aspera L. leaves in rats.

Materials and methods

Incision and excision wounds were inflicted on albino rats of either sex, under diethyl ether anesthesia. Group I served as positive control and was treated with 1% silver sulphadiazine, group II, III, IV treated with simple ointment containing 2.5%, 5% and 10% (w/w) methanol extract of the leaves of Achyranthes aspera L., respectively, whereas group V served as negative control and was treated with simple ointment. All the animals were treated topically once a day. Wound healing potential was assessed with excision and incision wound model. Excision wound model was used to assess the change in percentage contraction of wound, epithelization time, DNA content and histological features whereas rats inflicted with the incision wounds were used to determine breaking strength.

Results

Based on the results of percentage wound contraction, the DNA content and epithelization time, all groups of rats treated with methanol extract of the leaves of Achyranthes aspera L. showed significant (p<0.05) wound healing activity compared to group of rats treated with simple ointment (negative control) group. The difference in breaking strength was, however, significant (p<0.05) only for the 5% and 10% methanol extract of Achyranthes aspera (w/w) ointment treated groups. Histological evaluation showed well organized epidermal layer, increased number of fibrocytes, remarkable degree of neovascularization and epithelization which was comparable to the standard on the 21st day after treatment; especially in the 5% and 10% (w/w) extract treated group.

Conclusion

The present study provides a scientific rationale for the traditional use of the leaf extracts of Achyranthes aspera L. in the treatment of wound.  相似文献

Aim of the study

This study was designed to investigate the antimicrobial activities of the methanol extracts from the stem bark of Ficus ovata (FOB), fractions (FOB1–6) and compounds isolated following bio-guided fractionation [3-friedelanone (1), taraxeryl acetate (2), betulinic acid (3), oleanoïc acid (4), 2-hydroxyisoprunetin (5), 6,7-(2-isopropenyl furo)-5,2,4-trihydroxyisoflavone (6), Cajanin (7) and protocatechuic acid (8)].

Materials and Methods

The micro-dilution method was used for the determination of the minimal inhibition concentration (MIC) and the minimal microbicidal concentration (MMC) against fungi (two species), Gram-positive (three species) and Gram-negative bacteria (five species).

Results

The results of the MIC determinations indicated that the crude extract (FOB), fractions FOB2 and FOB4 as well as compound 5 were active on the entire studied organisms. Other samples showed selective activity, fractions FOB1, FOB3 and FOB5 being active against 50% of the tested microbial species while FOB6 was active on 40%. Compounds 8, 6, 2 and 7 prevented the growth of 80%, 70%, 50% and 20% of the organisms respectively. The lowest MIC value (156g/ml) observed with the crude extract was recorded on Streptococcus faecalis, Candida albicans and Microsporum audouinii. The corresponding value for fractions (39 μg/ml) was noted with FOB4 against Staphylococcus aureus, while that of the tested compounds (10 μg/ml) was observed with compound 8 on Microsporum audouinii. The results of the MMC determination suggested that the cidal effect of most of the tested samples on the studied microorganisms could be expected.

Conclusions

The overall results provided evidence that the studied plant extract, as well as some of the isolated compounds might be potential sources of new antimicrobial drug.  相似文献

The antimicrobial activity of the methanolic extract from the stem bark of Irvingia gabonensis (IGM), fractions and compounds isolated from IGM [3-friedelanone (1), betulinic acid (2), oleanolic acid (3), 3,3',4'-tri-O-methylellagic acid (4), 3,4-di-O-methylellagic acid (5) and hardwickiic acid (6)] was evaluated against Gram-positive bacteria (6 species), Gram-negative bacteria (13 species) and three Candida species using dilution methods for the determination of the minimal inhibition concentration (MIC) and the minimal microbicidal concentration (MMC). From the obtained results, IGM prevented the growth of all the species of microorganisms tested at a concentration limit of 312.50 microg/ml. Compounds 4-6 also inhibited the growth of all the tested microbial species while compounds 1-3 showed selective activities. The lowest MIC values (78.12 microg/ml) were obtained with IGM on 13 of the 22 microorganisms tested. The corresponding value of 1.22 microg/ml (4.26 microM) for compounds was recorded with compound 6 on Neisseria gonorrhoeae. The obtained results confirmed the use of Irvingia gabonensis in the treatment of bacterial and fungal infections.  相似文献
