2-亚氨基-3-(β-羟基苯乙基)噻唑烷(H_(322))(Ⅰ)是驱虫药四咪唑的中间体。在生产中发现操作工人血压偏低,高血压患者血压下降。为此进行动物试验,发现具有明显的降压活性,并进行了临床观察。据报道,剂量120~150mg/天,分三次口服,疗效为60~70%,但作用时间短,疗效也不够满意。为此进行结构改造并观察其结构与降压作用关系。  相似文献

In searching for new antimalarial agents ten new compounds of 2,4-diamino-6-[N-(substituted benzyl)-N-(substituted aminomethyl)-amino]-quinazoline have been synthesized. Preliminary screening results showed that only one (Ⅳ6) of these compounds displayed a slight degree of antimalarial activity against plasmodium berghei in mice. The intermediate compound (Ⅰ2) showed suppressive effect on plasmodium berghei in mice and prophylactic activity against plasmodium gallinaceum in chicken.  相似文献

本文报道以N-[1-(对-溴苯甲酰甲基)-3-甲基-4-哌啶基]-N-丙酰苯胺(Ⅲ)为前体,以PdO/BaSO4作催化剂,用氚气进行卤—氚置换,氚化还原羰基的反应。反应产物经硅胶纸层析纯化后,用甲基橙比色法定量测定,得到N-{1-[β-羟基-β-氚-β-(对-氚苯基)乙基]-3-甲基-4-哌啶基}-N-丙酰苯胺(Ⅳ,[3H]F-7302),其比放射性为59 Ci/mM,放化纯度为98%。[3H]F-7302与小鼠脑内阿片受体的特异性结合在浓度为4.5×10-9M时达到饱和,解离常数Kd=1.25×10-9M,最大结合量Bmax=93.08×1012M/g蛋白,其特异性结合与非特异性结合比值达10~15。  相似文献

本文报道38个β-(5-硝基-2-呋喃)丙酰胺及其α,β-二溴取代衍生物的合成。这类化合物的制备是以相应的β-(5-硝基-2-呋喃)丙烯酰胺类化合物(Ⅰ)进行催化氢化或与溴加成而得。经动物筛选发现β-(5-硝基-2-呋喃)丙烯酰胺类的丙烯双键以氢饱和以后生成丙酰胺类化合物(Ⅱ),对感染日本血吸虫病的小白鼠完全失去治疗或预防作用。而以溴饱和双键的α,β-二溴化合物(Ⅲ)则仍然有较显著的杀虫作用。其中尤以β-(5-硝基-2-呋喃)-α,β-二溴丙酰异丙胺(Ⅲ11)和β-(5-硝基-2-呋喃)-α,β-二溴丙酰甘氨酸乙酯(Ⅲ24)最为显著,后者曾试用于临床,证明有一定疗效。  相似文献

Four stereoisomers of 2-amino-3-(1,2-dicarboxyethylthio) propanoic acid were prepared by reaction of L- and D-cysteine with fumaric acid. The absolute configuration of the diastereoisomer of 2-amino-3-(1,2-dicarboxyethylthio) propanoic acid from Amanita pantherina were assigned as (2R, 1'R) and (2R, 1'S) by analysis of the optical properties. Pharmacological tests showed that all of the four stereoisomers inhibited the depolarization of NMDA on spinal motorneurones in newborn rats, The inhibition intensity of L-A,D-A and D-B were higher than that of L-B.  相似文献

Synthesis of 35 new compounds of α-chloro-β-(5-nitro-2-furyl) acrylamides and 5-[α-chloro-β-(5-nitro-2-furyl ) vinyl]-oxadiazoles by known methods are reported. In preliminary test in mice 10 compounds were found to possess pronounced activity against Schistosomiasis japonica. Among these Ⅰ12, Ⅰ13, Ⅰ14, Ⅰ20, Ⅱ1 and Ⅱ8 were shown to be the most effective.  相似文献

目的 培养(S)-2-(Boc-氨基)-3-[(S)-2-氧代-3-吡咯烷基]丙酸甲酯单晶,并进行晶型表征和立体结构确证。方法 用醋酸乙酯-甲基叔丁基醚(1∶1)体系培养单晶,并采用热重法、差示扫描量热法、粉末X射线衍射和单晶X射线衍射分析。结果 制备得到无色块状晶体;样品熔点约为114 ℃;该晶胞属于正交晶系,空间群为P212121,分子式为C13H22N2O5,相对分子质量为286.33,晶体密度为1.152 mg/mm3,绝对构型为S,S构型。结论 确定了(S)-2-(Boc-氨基)-3-[(S)-2-氧代-3-吡咯烷基]丙酸甲酯的立体构型,培养的晶体为非水合物和溶剂化物。  相似文献

本文报道β-(4,5-二溴-2-呋喃)-及β-(5-硝基-4-溴-2-呋喃)丙烯酰胺及其酯类衍生物26个的合成。动物筛选结果表明;化合物Ⅲ6,Ⅲ8和Ⅲ13对感染日本血吸虫小白鼠有明显的治疗作用。化合物Ⅱ6有较明显的预防作用。  相似文献

目的研究R,S-1-(2-甲氧基苯基)-4-[3-(萘-1-氧基)-2-羟基丙基]哌嗪(naftopidil,NAF)在大鼠血浆中的代谢产物。方法用LC/MS法对大鼠口服NAF后的血浆样品进行分析,根据检测到的代谢产物与原形药的质谱行为及类似结构化合物的代谢规律,推测可能的代谢产物。合成对照品,通过比较代谢产物和合成对照品的色谱和质谱行为,对I相代谢物予以确认。通过质谱信息及酶水解的方法间接鉴定II相代谢物。结果大鼠血浆中发现I相代谢物:R,S-1-(2-羟基苯基)-4-[3-(萘-1-氧基)-2-羟基丙基]哌嗪(DMN)、R,S-1-(2-甲氧基-4-羟基苯基)-4-[3-(萘-1-氧基)-2-羟基丙基]哌嗪(PHN),R,S-1-(2-甲氧基苯基)-4-[3-(4-羟基萘-1-氧基)-2-羟基丙基]哌嗪(NHN)及II相代谢物:NAF和NHN与<>-D-葡糖醛酸形成的结合物。结论NAF在大鼠血浆中的主要代谢途径是苯环、萘环羟基化和苯环邻位甲氧基的脱甲基化。此外,萘羟化代谢物和原形药与内源性分子<>-D-葡糖醛酸形成结合物也是原形药的代谢方式之一。  相似文献

为了寻找心血管系统药物,以表雄酮为原料,合成了7个16,17位取代的目标化合物:2β-(4’-甲基-1’-哌嗪基)-3α-羟基-5α-雄甾-17-酮、2β-(4’-甲基-1’-哌嗪基)-3α,17β-二羟基-5α-雄甾烷、2β-(4’-甲基-1’-哌嗪基)-3α-羟基-16α-溴-5α-雄甾-17-酮、2β-(4’-甲基-1’-哌嗪基)-3α,16α-二羟基-17-氧-5α-雄甾烷、2β-(4’-甲基-1’-哌嗪基)-3α,16α-二羟基-17-肟-5α-雄甾烷、2β-(4’-甲基-1’-哌嗪基)-3α,16α-二羟基-17β-氨基-5α-雄甾烷和2β—(4’-甲基-1’-哌嗪基)-3α,17β-二羟基-16β-氨基-5α-雄甾烷。初步药理试验结果表明它们都有不同程度的抗心律失常活性。  相似文献

To study the chemical composition of the skin of Bufo bufo gargarizans, various chromatographic methods were used in the isolation procedures and the structures of isolated compounds were determined based on NMR and MS analysis. As a result, two new compounds were isolated from its ethanolic extract and characterized as N-[2-hydroxy-2-(4-hydroxyphenyl)ethyl]-N-methylurea (1) and 19-oxocinobufotalin 3-adipoylarginine ester (2), together with 11 known compounds. Isolated bufadienolides showed significant inhibition effect against human hepatocellular carcinoma cell line SMMC-7721 in vitro.  相似文献

Two new monoterpenoid glycosides, spicatoside A and spicatoside B, were isolated from the whole herbs of Mentha spicata L. which have anti-inflammatory and hemostatic activities. Their structures have been determined on the basis of spectral and chemical analysis. They are (+)-5-[1-(g -D-glucopyranosyloxymethyl)ethenyl]-2-methyl-2-cyclohexen-1-one (1), and (m )-5-{[2-( g -D-glucopyranosyloxy)-1-hydroxy-1-methyl]ethyl}-2-methyl-2-cyclohexen-1-one (2).  相似文献

3-Chloro-4-(chloromethyl)-5-hydroxy-2(5H)-furanone (CMCF), 3-chloro-4-methyl-5-hydroxy-2(5H)-furanone (MCF) and 3,4-dichloro-5-hydroxy-2(5H)-furanone (MCA) are chlorination byproducts in disinfected drinking water. These compounds are positive in genotoxicity tests in vitro. We have previously shown that 3-chloro-4-(dichloromethyl)-5-hydroxy-2(5H)-furanone (MX) can induce malignant transformed foci in the two-stage cell transformation assay in C3H 10T1/2 cells in vitro in both the initiation and promotion phases. In the present study we compared the effects of CMCF, MCF and MCA in the same assay. C3H 10T1/2 mouse embryonic fibroblasts were exposed to these chlorohydroxyfuranones (CHFs) at three different concentrations in the initiation phase or the promotion phase of the assay. In the latter experiments 3-methylcholanthrene (MC, 5 µg/ml) was used as the initiating chemical. The phorbol ester 12-O-tetradecanoylphorbol-13-acetate (TPA, 0.3 µg/ml) was used as a positive control promoter. At the end of the assay (6 weeks from the start), the transformation foci were counted and scored after fixation and staining of the cells. When added at the initiation phase of the assay on their own, CMCF and MCF, but not MCA, increased the transformation foci formation. TPA added in the promotion phase did not modify the responses of CMCF and MCF but TPA increased the number of foci in MCA-treated cells. When CHFs were added during the promotion phase to the MC-initiated cells, MCF and MCA enhanced the development of the transformation foci. The effect of CMCF was equivocal since at higher concentrations CMCF actually decreased the number of the MC-induced foci. Including the previous data for MX in this assay and considering the lowest active concentrations, the initiation activity of the foci formation decreased in the order MX >CMCF >MCF, i.e. with the decreasing number of chlorine atoms of the methyl group in the 4-position of the CHF molecule (two, one, and zero, respectively). In contrast, the activity in the promotion phase did not follow the same pattern. MX, MCF and MCA were all active over the same concentration range. Hence, in addition to MX, MCF and MCA may also possess some potential to promote tumor development.  相似文献

To explore secondary metabolites in deep-sea sediment metagenomic clone-derived Escherichia coli fermentation broth, different kinds of chromatography methods were used in the isolation procedures, while the structures of the isolated compounds were assigned based on the MS analysis and their 1H and 13C NMR spectra including 2D NMR techniques such as COSY, HMQC, and HMBC experiments. As a result, a novel compound was isolated and characterized as N-{1-[4-(acetylamino)phenyl]-3-hydroxy-1-(1H-indol-3-yl)propan-2-yl}-2,2-dichloroacetamide (1). In addition, eight known compounds were also obtained. Fatty acid amide hydrolase and monoacylglycerol lipase were used to screen analgesic activity, and the new compound showed analgesic activity to some extent in pharmacological test.  相似文献

本文报道了2,4-二氨基-6-N1,N2-二取代肼基-喹唑啉类衍生物的合成及其抗疟活性的研究。这类化合物的合成是以2,4-二氨基6-取代苄基氨基-喹唑啉为原料经亚硝化、还原成为2,4-二氨基6-(N1-取代苄基)—肼基喹唑啉,再与相应的醛缩合而成。此类化合物经伯氏鼠疟原虫抑制性治疗初筛表明有少数具有一定的效果。有11个化合物经约氏鼠疟原虫—斯氏按蚊系统病因性初筛有效。其中化合物Ⅱ1,7,8,11,15和Ⅲ1口服2.5mg/kg,连续3天,可使受试小鼠全部得到保护。  相似文献

Seven new aromatic acid derivatives (17), together with five known analogs, were isolated from the lateral roots of Aconitum carmichaelii. Structures of the new compounds were determined by spectroscopic and chemical methods as 4-methyl ( ? )-(R)-hydroxyeucomate (1), 4-butyl ( ? )-(R)-hydroxyeucomate (2), 4-butyl-1-methyl (+)-(R)-2-O-(4′-hydroxy-3′-methoxybenzoyl)malate (3), 1-butyl-4-methyl (+)-(R)-2-O-(4′-hydroxy-3′-methoxybenzoyl)malate (4), dimethyl (+)-(R)-2-O-(4′-hydroxy-3′-methoxybenzoyl)malate (5), dimethyl (+)-(R)-2-O-(4′-hydroxybenzoyl)malate (6), and methyl ( ± )-3-(4′-hydroxy-3′-methoxyphenyl)-3-sulfopropionate (7), respectively. Compounds 1 and 2 are 2-benzylmalates (eucomate derivatives), 36 belong to 2-O-benzoylmalates, and 7 is a rare phenylpropionate containing a sulfonic acid group. The absolute configurations of eucomate derivatives were confirmed by X-ray crystallographic analysis of 4-methyl eucomate (11).  相似文献

报道11个预期有血管紧张素转化酶抑制活性的N-(4-甲氧羰基-4-邻苯二甲酰亚氨基丁酰基)-N-取代甘氨酸(VII1~9)、脯氨酸(VII10)和焦谷氨酸(VIl11)的合成和鉴定。所有上述化合物以及与VⅡ1~9相应的叔丁酯(VI1~9)均未见文献报道。药理初试结果显示,化合物VII8,VII9和VI10均有明显降压活性。  相似文献
