目的 探讨P16蛋白表达与垂体腺瘤侵袭及复发的关系。方法 应用免疫组化染色技术检测57例垂体腺瘤中P16蛋白的表达水平,并对患者进行了5抻随访观察。结果 垂体腺瘤细胞中P16蛋白表达的阳性率为31.6%;侵袭性组中该蛋白表达的阳性率显著低于非侵袭性组(P<0.01)。复发组11例未见该蛋白表达。结论 P16蛋白的表达阳性率与垂体腺瘤激素分泌功能无明显的相关性,但其表达水平可作为临床评价垂体腺瘤的侵袭性和预测患者复发的指标。  相似文献

目的探讨人垂体腺瘤中垂体肿瘤转化基因(PTTG)蛋白的表达与肿瘤侵袭性和增殖程度的关系。方法采用免疫组织化学染色方法检测手术切除石蜡包埋的63例垂体腺瘤(侵袭组45例,非侵袭组18例)组织中PTTG蛋白的表达,染色增殖细胞核抗原(PCNA),同时计数组织内微血管数量(MVD)。结果 PTTG在侵袭性垂体腺瘤中的表达水平显著高于非侵袭性垂体腺瘤。侵袭性垂体腺瘤中PCNA标记指数和微血管密度也显著高于非侵袭性垂体腺瘤。相关分析显示PTTG表达与垂体腺瘤内PCNA标记指数和微血管密度呈正相关(P<0.05)。结论 PTTG在垂体腺瘤形成过程中起重要作用,并与垂体腺瘤的侵袭性和增殖程度密切相关。  相似文献

目的 探讨p5 3蛋白和p2 7蛋白在乳腺癌中的表达及意义。方法 应用免疫组化S -P法检测 6 7例乳腺癌中p5 3蛋白和p2 7蛋白的表达情况。结果  6 7例乳腺癌中p5 3蛋白和p2 7蛋白的阳性表达率分别为 5 8 3%和 5 9 7% ,p5 3蛋白的表达与乳腺腋窝淋巴结转移有相关性 (P <0 0 1) ,p2 7蛋白的表达也与乳腺腋窝淋巴结转移有关 (P <0 0 5 ) ,乳腺癌p2 7和p5 3蛋白表达之间无明显相关。结论 p5 3蛋白的过表达和p2 7蛋白的失表达均与乳腺癌发生发展相关 ,二者可作为判断乳腺癌预后的有用指标  相似文献

目的:研究突变型p53癌基因在垂体腺瘤中的表达。方法:应用免疫组化ABC法检测64例垂体腺瘤(复发组31例,非复发组33例),6例正常垂体、2例垂体瘤中p53蛋白表达阳性,非复发组中有2例p53蛋白表达阳性,两组差异有显著性。2例垂体癌p53蛋白强阳性表达,6例正常垂体无阳性表达。p53蛋白在不同激素类型垂体腺癌中的阳性表达率差异无显著性。结论:p53蛋白高表达与少数垂体腺癌在潜在恶性及复发有关,  相似文献

目的探讨胃肠道间质瘤中p16、p27和Ki67表达与临床预后的关系。方法根据核分裂象的多少、肿瘤体积的大小及有无浸润等将胃肠道间质瘤划分为良性、交界性和恶性,并运用免疫组化SP方法检测p16、p27和Ki67在胃肠道间质瘤中的表达,并统计分析其良性、交界性、恶性和复发死亡病例中的表达差异。结果p16、p27和Ki67的阳性表达率分别为48%、26%和24%。p16在良、恶性中的表达无明显差异,但在良性和交界性中的表达与复发和转移相关;p27低标记指数和Ki67高标记指数与复发和转移相关。结论p16、p27和Ki67在胃肠道间质瘤中的表达对判断预后有价值。  相似文献

目的 :检测CyclinD1 和p16、p2 7以及PCNA在脑胶质瘤中的表达状况 ,以探讨不同病理类型脑胶质瘤中各自的表达及其相关性。方法 :应用免疫组化S P法 ,检测 12例正常脑组织、5 8例脑胶质瘤组织中CyclinD1 、p16、p2 7及PCNA表达及其特征。结果 :CyclinD1 在由低度恶性胶质瘤向高度恶性胶质瘤转化中阳性表达逐渐增强 (χ2 检验 ,P <0 0 0 5 ,χ2 =5 1 6 7) ;而p16、p2 7阳性表达却是随着胶质瘤恶性程度的升高而降低 (χ2 检验 ,P值均 <0 0 0 5 ,χ2 分别为 15 4 1和 12 81)。CyclinD1 与PCNA呈正相关 ,rs =0 74 5 ;p16和p2 7与PCNA呈负相关 ,rs分别为 - 0 5 6 6和 - 0 6 12。结论 :脑胶质瘤中CyclinD1的表达程度对胶质瘤的细胞增殖活性起促进作用 ,而p16、p2 7的表达则起抑制作用。  相似文献

目的 研究p27^kip1、p16蛋白及增殖细胞核抗原(PCNA)在鼻咽癌(NPC)组织中的表达,探讨它们之间的关系及其与.NPC生物学行为及预后的相关性。方法应用免疫组织化学EnVision两步法检测66例鼻咽非角化性癌(NKC)组织和25例鼻咽黏膜慢性炎症(NP)组织中p27^kip1、p16蛋白及PCNA表达水平。结果(1).NKC组织中p27^kip1、p16阳性表达率分别为65%、68%;与NP组比较,差异有统计学意义(P<0.05)。(2)<、p16蛋白在NKC中的表达与NKC颅神经侵犯及治疗后5年生存率有关(P<0.05),与临床分期、淋巴结转移无关(P>0.05);PCNA表达与.NKC临床分期及治疗后5年生存率有关(P<0.05),与淋巴结转移、颅神经侵犯无关(P>0.05)。(3)p27^kip1、p16及PCNA阳性表达之间有相关性(P<0.05)。结论检测p27^kip1、p16蛋白及PCNA有助于综合评估NKC的预后。  相似文献

目录探讨p16蛋白表达异常与肺癌的关系。方法采用辣根酶标记链霉亲和素免疫组化方法,对35例肺癌组织及相应的癌旁正常肺组织标本和10例肺良性疾病肺组织标本进行研究。结果癌周正常肺组织、肺良性疾病肺组织p16蛋白阳性率均显著高于肺癌组织;临床I,II期p16蛋白阳性率显著高于临床Ⅲ、Ⅳ期;高、中、低分化肺癌细胞p16蛋白表达无显著性差异,男、女肺癌患者p16蛋白表达无显著性差异。结论肺癌的发生可能与p16基因表达产物缺失有关,p16基因的缺失在肺癌进展中起着一定的作用,有可能同时成为预测病程进展的指标之一。  相似文献

We studied 40 endocrinologically inactive pituitary adenomas by immunohistochemistry, electron microscopy, and cell culture in order to determine the incidence of gonadotropic adenomas and to classify nonfunctioning adenomas. Immunohistochemical studies using a large panel of monoclonal and polyclonal antibodies identified the following nonfunctioning adenomas: 20 gonadotropic adenomas, four silent corticotropic adenomas, one plurihormonal adenoma, and 15 nonsecreting adenomas. Among nonsecreting adenomas, ultrastructural study of 13 cases identified seven null cell adenomas and six oncocytomas. Silent corticotropic adenomas were classified into subtypes I, II, and III according to Kovacs and Horvath. Most often, gonadotropic adenomas displayed a varying number of oncocytic cells, characteristic secretory granules, and a prominent Golgi apparatus. Postembedding immunoelectron microscopy was performed on eight gonadotropic or nonsecreting adenomas, but this technique did not provide any additional information. Six gonadotropic adenomas and 10 so-called nonsecreting adenomas were studied in primary cell cultures. The six gonadotropic adenomas and seven of the 10 nonsecreting adenomas released gonadotropins in the culture medium. The use of in vitro results as a supplementary diagnostic criterion allowed classification of the 40 nonfunctioning adenomas as follows: 27 gonadotropic adenomas, four silent corticotropic adenomas, one plurihormonal adenoma, and eight nonsecreting adenomas. These results demonstrate a high proportion of gonadotropic adenomas among nonfunctioning adenomas (67.5%) and the usefulness of several techniques in characterizing this type of pituitary adenoma.  相似文献

CDKN2A/p16 inactivation is related to pituitary adenoma type and size   总被引:16,自引:0,他引:16  
p16 (CDKN2A, MTS1, INK4A) status at genomic and protein levels was analysed and correlated with clinico-pathological features in 72 pituitary adenomas. Methylation of CpG islands of promoter/exon 1 sequences was found in most gonadotroph, lactotroph, plurihormonal, and null cell adenomas (36 of 44, 82%), but it was rare in somatotroph (1 of 13 cases, 8%) and corticotroph adenomas (1 of 15 cases, 7%). Homozygous CDKN2A deletion was restricted to rare somatotroph (15%) and corticotroph adenomas (13%). Immunohistochemical p16 protein expression was observed in the normal adenohypophysis, whereas it was absent in 60 of 72 (83%) tumours and reduced in another ten (14%) tumours. Staining for p16 was only seen in 5 of 15 (33%) corticotroph, 3 of 13 (23%) somatotroph, 3 of 5 (60%) plurihormonal, and 1 of 19 (5%) null cell adenomas. p16 immunonegativity without CDKN2A methylation or deletion occurred in 22 tumours, including most somatotroph and corticotroph adenomas (15 of 28, 54%). Both CDKN2A alterations and p16 negativity were related to larger tumour size. Patients with p16-negative tumours were older than patients with p16-positive tumours. These data suggest that p16 down-regulation is common in all adenoma types. The mechanisms of p16 down-regulation probably involve CDKN2A methylation in most types, but remain to be determined in somatotroph and corticotroph adenomas. These findings also suggest that p16 down-regulation is usually not an initial event, but is acquired during adenoma progression.  相似文献

目的 探讨 p2 7、p5 3蛋白表达在口腔鳞癌发生发展中的意义。 方法 应用免疫组化S P法分别检测 9例口腔正常黏膜 ,11例单纯性增生、2 6例癌前病变及 5 4例鳞癌组织中p2 7、p5 3蛋白的表达。 结果 p2 7蛋白在口腔正常黏膜和单纯性增生组织中呈高表达 ,在癌前病变和鳞癌组织中高 (低 )表达率分别为 6 1 5 % (38 5 % )、2 5 9% (6 1 1% ) ,在鳞癌中阴性表达率为 13% ;p2 7蛋白的表达与鳞癌的组织分化程度、临床分期相关 (P <0 0 5 )。p5 3蛋白在正常黏膜、单纯性增生及轻、中度不典型增生中未见表达 ,在重度不典型增生和鳞癌中可见 2 8 6 %和 4 8 1%的阳性表达 ,二者差异有高度显著性 (P <0 0 1) ;在鳞癌中 p5 3蛋白表达与组织分化程度相关 (P <0 0 5 ) ;p2 7和p5 3表达在鳞癌中呈负相关 (P <0 0 1)。 结论 p2 7蛋白表达的减少在口腔鳞癌的发生发展中起着重要作用 ,并与其预后因素密切相关。p5 3蛋白的表达在癌前病变向鳞癌转变过程中起重要作用。综合分析 p2 7、p5 3表达有助于口腔鳞癌的早期诊断和患者预后的估计。  相似文献

p27 (Kip1) plays regulatory roles in the cell cycle by inhibiting the activity of cyclin dependent kinases (CDKs). This immunohistochemical study is aimed at elucidating the expression of p27 in human pituitary and in various types of pituitary adenomas in order to clarify its role in the regulation of proliferation. Sixteen normal pituitary glands and 179 human pituitary adenomas were used for immunohistochemical studies. The tissues were fixed in 10% formalin and embedded in paraffin. Indirect peroxidase method was performed after heat-induced antigen retrieval using a monoclonal antibody against p27 protein. p27 protein was expressed in the nuclei of all 16 normal human pituitary glands. p27 protein was also expressed in 128 of 179 cases of pituitary adenomas (71.5%). A marked decrease of p27 expression was noted in ACTH-secreting adenomas, 8/20 (40.0%), compared with other types of pituitary adenomas—GH-secreting adenomas, 35/46 (76.1%); PRL-secreting adenomas, 22/33 (66.7%); TSH-secreting adenomas, 8/11 (72.7%); and nonfunctioning adenomas, 55/69 (79.7%). These results suggest that p27 may play some role in the regulation of proliferation in all types of pituitary adenomas. The lower levels of p27 in ACTH-secreting adenoma is of particular interest with respect to the intermediate lobe-derived pituitary tumor developed in p27 knockout mice.  相似文献

AIMS: To determine if the cyclin dependent kinase inhibitors (CDKIs) p16 and p27 show reduced expression in the progression from benign to malignant melanocytic tumours, and to correlate these findings with patient prognosis. METHODS: Ninety-two melanocytic tumours were assessed for immunohistochemical expression of p16 and p27. These specimens included nine compound naevi, 10 dysplastic naevi, 17 thin (<1 mm) melanomas, 22 thick (>1 mm) melanomas, nine in-transit metastases, 13 lymph node metastases, and 12 soft tissue metastases. Clinicopathological information on the 39 patients with melanoma primaries was obtained from the Sydney Melanoma Unit database. The median follow up period was 43.3 months. RESULTS: A significant loss of expression of p16 and p27 was found with tumour progression. Positive expression of p27 was found in all compound and dysplastic naevi but only 43.6% of melanoma primaries. Expression of p27 was greater in lymph node and in-transit metastases (63.6%), but lower in soft tissue metastases (36.4%). Positive expression of nuclear p16 was evident in 73.7% of benign naevi, 28.2% of primary melanomas and 14.7% of metastatic melanomas. Neither p16 nor p27 expression was significantly correlated with overall survival, disease free survival or other clinicopathological markers. CONCLUSIONS: The CDKIs p16 and p27 are associated with tumour progression in melanoma, but do not reliably predict recurrence or survival.  相似文献

目的 :探讨 p2 7kip1 、PCNA及 p1 6与胃肠道类癌转移的关系。 方法 :采用免疫组化SABC法检测 2 5例胃肠道类癌组织中p2 7kip1 和PCNA蛋白表达 ,同时用PCR法检测 p1 6基因的缺失情况。 结果 :p2 7kip1 在胃肠道类癌阳性表达率为 64 % (1 6/ 2 5) ,PCNAⅠ~Ⅱ级和Ⅲ~Ⅳ级表达率分别为 56 %、44 %。 2种蛋白阳性表达率与分化程度无相关性 (P >0 0 5) ,与淋巴结转移相关 (P <0 0 5)。p2 7kip1 与PCNA表达负相关。p1 6基因纯合性缺失率为 48% (1 2 / 2 5) ,其缺失率与类癌分化程度和淋巴结转移无关。结论 :p2 7kip1 低表达、PCNA高表达与胃肠道类癌转移有关 ,胃肠道 ;类癌的发生与p1 6基因缺失有关 ,但可能为早期分子事件。  相似文献

目的 探讨p21、p27、p53和Rb蛋白表达在胃癌发生、发展中的作用.方法 应用免疫组化EnVision法检测111例胃癌,38例不典型增生、47例肠上皮化生、25例慢性萎缩性胃炎和53例正常胃黏膜组织中p21、p27、p53和Rb蛋白的表达.结果 p21和p27蛋白表达水平在不典型增生组织最高,其次是胃癌组织,与正常胃黏膜组织相比差异均有显著性(P<0.001).在胃癌组织中p21、p27、p53和Rb蛋白表达均高于正常胃黏膜组织(P均<0.01).p21、p27蛋白表达与胃癌的类型和分化相关(P均<0.01),p53表达水平与患者的年龄、性别、胃癌的类型和患者的生存率相关.女性患者Rb蛋白表达明显高于男性,差异有显著性(P<0.05).p21、p27、p53和Rb蛋白表达水平之间均呈正相关.结论 p21、p27、p53和Rb蛋白表达可以作为胃癌的辅助诊断指标,四种蛋白在判断胃癌的生物学行为上具有协同作用.  相似文献
