DNA histograms of exponentially growing EMT-6 cells in vitro were obtained at regular time intervals after exposure at 300 rads of x rays or one hour of heat shock at 43 degrees C. After either treatment, about 70% of the cells retained clonogenicity. Irradiation induced the expected block in the G2 + M phase, resulting in an increase of cells in that phase during the first 5 hours after exposure. Heat shock caused a block in both the S and G2 +M phases.  相似文献

目的 探讨用抗体封闭热休克蛋白70(heat shock protein 70,HSP70)后对星形胶质细胞反应性增生的影响. 方法 原代培养并纯化星形胶质细胞,采用划痕损伤法构建细胞损伤模型,通过培养液内加入HSP70抗体来进行干预.在不同时间点对损伤对照组和损伤干预组进行胶质纤维酸性蛋白(GFAP)免疫细胞化学染色,观察细胞增生情况及形态学变化,并用RT-PCR检测GFAP mRNA的表达. 结果 与对照组相比,干预组细胞面积、突起数目及突起长度均减小(P<0.05或0.01),GFAP mRNA的表达也明显降低(P<0.05). 结论 星形胶质细胞损伤后,HSP70有促进星形胶质细胞反应性增生的作用.用抗体封闭HSP70后,能够一定程度上抑制星形胶质细胞的反应性增生.  相似文献

The purpose of the current research was to test the hypothesis that exercise induced leukocyte heat shock protein (HSP) expression is increased during periods of intensified exercise training. Seven male endurance cyclists carried out tests of maximal oxygen consumption and endurance capacity. These standard exercise tests were carried out prior to and following 6 days of prescribed intensified training. Sampled leukocytes were examined for Hsp27 and Hsp70 expression using a Fluorescence Activated Cell Scanner (EPICS XL, Coulter). During a period of overreaching, as signified by a drop in time to fatigue following the intensified training period (p = 0.02), the number of extracellular Hsp27 positive granulocytes increased in response to the VO(2)max test. Acute, intracellular HSP responses were observed in both baseline and overreached conditions. The present study showed that a period of intensified training caused adaptations in the acute heat shock protein exercise response, reflected by a greater increase of cell surface HSP positive leukocytes following heavy training.  相似文献

In forensic casework, evidence regarding the type of cell material contained in a stain can be crucial in determining what happened. For example, a DNA match in a sexual offense can become substantially more incriminating when there is evidence supporting that semen cells are present.Besides the question which cell types are present in a sample, also the question who donated what (association) is very relevant. This question is surprisingly difficult, even for stains with a single donor. The evidential value of a DNA profile needs to be combined with knowledge regarding the specificity and sensitivity of cell type tests. This, together with prior probabilities for the different donor-cell type combinations, determines the most likely combination.We present a Bayesian network that can assist in associating donors and cell types. A literature overview on the sensitivity and specificity of three cell type tests (PSA test for seminal fluid, RSID saliva and RSID semen) is helpful in assigning conditional probabilities. The Bayesian network is linked with a software package for interpreting mixed DNA profiles. This allows for a sensitivity analysis that shows to what extent the conclusion depends on the quantity of available research data. This can aid in making decisions regarding further research.It is shown that the common assumption that an individual (e.g. the victim) is one of the donors in a mixed DNA profile can have unwanted consequences for the association between donors and cell types.  相似文献

热休克蛋白65增强小鼠对HBV DNA 疫苗免疫反应的研究   总被引:3,自引:0,他引:3  
目的:观察热休克蛋白65的真核表达载体(pHSP65)对HBV DNA疫苗诱导BALB/c小鼠(H-2d)免疫应答的调节作用。方法:肌内注射空载体pcDNA3,HBV DNA疫苗加HSP65佐剂(pHBVS2S pHSP65)或不加佐剂(pHBVS2S);ELISA法测定血清抗HBs抗体;ELISPOT检测分泌IFN-γ的脾淋巴细胞;4 h51Cr释放法检测小鼠脾细胞CTLs活性。结果:HBV DNA佐剂组免疫小鼠抗HBsAg抗体滴度虽高于不加佐剂组,但无显著性差异;其IgG1/IgG2 a的比例不同于多肽免疫组,二者分别为0.395与10。佐剂组小鼠分泌IFN-γ的脾淋巴细胞量是不加佐剂组的2~3倍。CTLs细胞杀伤活性(E∶T=100)佐剂组与不加佐剂组分别为:(53.68±7.5)%、(42.81±7.7)%,差异显著(P<0.05)。结论:HBV DNA疫苗具有较强的免疫原性,能够诱导机体产生特异性的抗体及CTLs反应;HSP65佐剂能够有效提高小鼠对DNA疫苗的细胞免疫应答,有望成为DNA疫苗的免疫佐剂。  相似文献

Protein synthesis in Saccharomyces cerevisiae after exposure to ultraviolet light (UV) was examined by two-dimensional gel electrophoresis of pulse-labelled proteins. The synthesis of 12 distinct proteins was induced by treatment with UV doses of 10-200 J/m2. The induced proteins differed in the minimum dose necessary for induction, the maximum dose at which induction still occurred and the constitutive level present in unirradiated cells. A chemical mutagen, 4-nitroquinoline-1-oxide, induced synthesis of the same proteins. Induction after UV treatment was observed in seven different yeast strains, including three mutants deficient in DNA repair. Synthesis of five of the proteins was also induced by brief heat shock treatment. These five proteins may be members of a family of proteins whose synthesis is regulated by two different pathways responding to different types of stress.  相似文献

目的 观察热休克蛋白70(HSP70)对急性肺损伤的保护作用.方法 64只SD大鼠按随机数字表法分为假加热正常对照组(Sham组)、中暑组(HS组)、中暑加谷氨酰胺处理组(HS+GLN组)和中暑加槲皮素处理组(HS+QU组),每组16只.Sham组大鼠置于温度23℃,湿度55% ± 5%环境中,其余3组大鼠置于模拟热气候动物舱(舱内温度39℃,相对湿度65%)内.监测大鼠直肠温度、收缩压和脉率,比较各组大鼠热应激反应的差异.以收缩压从峰值开始下降作为中暑的开始,之后将大鼠移至常温复温.分别在中暑恢复期0h和6h处死大鼠,每个时间点8只,留取支气管肺泡灌洗液(BALF)后分离肺脏组织行组织学观察.采用酶联免疫法检测BALF中的IL-1β、TNF-α和IL-6浓度,以及肺组织匀浆中的HSP70浓度.结果 与HS组和HS+QU大鼠比较,HS+GLN组大鼠承受更多的热负荷后才发生HS(P<0.001),起病后的中位生存时间明显延长(P<0.001).与Sham组比较,HS组大鼠肺组织匀浆HSP70浓度呈时间依赖性升高(P<0.001);HS+GLN组HSP70浓度在各个时点与HS组比较均明显升高(P<0.001),而HS+QU组的HSP70表达则受到明显抑制时点与Sham组比较差异无统计学意义(P>0.05),但明显低于HS组和HS+GLN组(P<0.001).与HS组和HS+QU组比较,HS+GLN组大鼠BALF中的IL-1β、TNF-α和IL-6浓度明显降低(P<0.001),病理结果显示其肺损伤更轻(P<0.001),而HS+QU组则相反.结论 HSP70具有保护HS大鼠急性肺损伤的作用,其机制可能与增强大鼠热耐受能力和抑制HS大鼠肺部炎症相关.  相似文献

目的研究几种主要热休克蛋白(heat shock protein, HSP)在不同病理改变的宫颈组织中的表达.方法根据病理诊断,把478例宫颈活检标本分为宫颈癌组(63例)、宫颈上皮内瘤形成组(CIN组,106例)、宫颈炎组(293例)及正常宫颈组(16例).采用以特异性复合cRNA为内参照的定量RT-PCR法,检测各组标本的HSP70、HSP90α和HSP90β mRNA的表达.结果 (1)HSP70、HSP90α和HSP90β mRNA在宫颈浸润癌、CIN、宫颈炎及正常宫颈组织中的表达量均呈现阶梯式下降,差异均有极显著意义(P<0.01).(2)HSP90β mRNA在晚期宫颈癌中的表达比早期表达高(P<0.05);HSP70和HSP90β mRNA在低分化宫颈癌中的表达比高分化表达高(P<0.05);而HSP70、HSP90α和HSP90β mRNA在不同组织类型宫颈癌中的表达均无显著性差异(P>0.05).结论热休克蛋白在宫颈癌的发生和发展中起着重要的协同作用.HSP70和HSP90α与宫颈癌细胞的转化和增殖密切相关,HSP90β可能参与细胞的分化.  相似文献

目的 探讨采用不同功率胆管内射频消融时,毗邻消融靶区的胆管组织热休克蛋白(heat shock protein,HSP)70表达的差异及意义。方法 15个新鲜离体猪肝随机分为3组(胆囊及胆总管中上段完整保留),将收集的人胆汁注入猪肝胆道系统内,然后将射频消融导管经胆总管引入肝门区胆管内。所有猪肝均消融120 s,第1、2、3组分别采用10 W、12 W、14 W功率消融。消融后取距远端消融电极1cm、2cm处胆管组织,采用免疫组化S-P法检测15例猪肝共30例胆管组织中HSP70的表达。结果 ①距远端消融电极1cm处胆管组织HSP70表达:1级染色4例(26.7%),2级染色6例(40%),3级染色5例(33.3%);距远端消融电极2 cm处胆管组织HSP70表达:1级染色10例(66.7%),2级染色4例(26.7%),3级染色1例(6.6%);②消融区毗邻胆管(距远端消融电极1cm)HSP70表达强度与消融功率呈正相关(r=0.755,P<0.001);距远端消融电极1cm、2cm处胆管组织HSP70表达强度差异有统计学意义(Z=-2.334,P<0.05)。结论 胆管内射频消融可导致消融区毗邻胆管组织HSP70表达,随着消融功率的增加,毗邻消融区胆管组织内HSP70表达强度亦随之增加;远离消融区域胆管(距离远端消融电极≥2cm)HSP70表达无明显增多。  相似文献

目的 探讨不同 Gz重复暴露后,大鼠不同脑区热休克蛋白-70(HSP70)表达强度和时程的变化规律。及其与正加速度致脑损伤的相关性。方法 将雄性SD大鼠随机分为对照组、 2Gz、 6Gz和 10Gz暴露组。分别在暴露后6h、1d、2d、4d和6d处死取脑,利用HE和免疫组织化学染色,观察HSP70在鼠脑不同部位的表达。及脑神经元形态学和血脑屏障通透性的变化,结果 G2重复暴露可诱导HSP70的表达。其表达强度,持续时间和部位随G值而异。低G值( 2Gz)下,表达强度弱,持续时间短,但范围较广;高G值( 10Gz)下,表达仅局限在下丘脑和梨状皮层等部位。呈中等强度反应;中等强度G值( 6Gz)下,表达范围广(在皮层、海马、丘脑等部位均有HSP70较强反应),持续时间长。结论 6Gz诱导的HSP70表达最强,持续时间最长。为研究热休克蛋白对正加速度致脑损伤的保护作用。提供了实验依据。  相似文献

目的 探讨热休克蛋白70(heat shock protein70,HSP70)在创伤愈合过程中的变化及其意义。方法 取大鼠伤后1天伤口液作为创伤修复启动微环境,刺激真皮多能干细胞,24小时后检测细胞表达HSP70含量的变化。同时制作单纯创伤和难愈创伤动物模型,通过免疫组化和图像分析方法观察伤后不同时相点伤口组织HSPT0含量变化。结果 创伤早期伤口液作用真皮多能干细胞后,HSP70表达显著增加。单纯创伤组大鼠伤后各时相点伤口局部HSP70表达均显著增加,尤以真皮组织增加明显,其峰值为伤后3天。难愈创伤组大鼠伤口局部HSP70含量在伤后各时相点亦显著增加,但与单纯创伤组相比,其含量在伤后各时相点均显著减少,尤以真皮组织表达减少更为明显,并且HSP70表达峰值显著延迟,为伤后7天。结论 HSP70不仅是一种应激保护分子,而且还是参与调节创伤愈合的一种重要分子。  相似文献

 目的探讨幽门螺杆菌(Helicobacter pylori,Hp)对消化性溃汤(Peptic ulcer,PU)患者热休克蛋白60(Heat shock protein 60,HSP60)的影响.方法观察HP清除率,运用Western斑点印迹技术对患者血清及胃黏膜匀浆HSP60进行检测,并将相关结果进行比较.结果HSP60含量治疗前与对照组比较,HP阳性者明显增高(P<0.01);治疗后HP阳性者仍明显高于HP阴性者及HP清除者(P<0.05),而后两者无明显差异.结论HP感染可促进HSP60表达,HP清除使HSP60表达能力下降,HP与HSP60存在关联.  相似文献

PURPOSE: This investigation was designed to examine the influence of creatine (Cr) supplementation on acute cardiovascular, renal, temperature, and fluid-regulatory hormonal responses to exercise for 35 min in the heat. METHODS: Twenty healthy men were matched and then randomly assigned to consume 0.3 g.kg(-1) Cr monohydrate (N = 10) or placebo (N = 10) for 7 d in a double-blind fashion. Before and after supplementation, both groups cycled for 30 min at 60-70% VO2(peak) immediately followed by three 10-s sprints in an environmental chamber at 37 degrees C and 80% relative humidity. RESULTS: Body mass was significantly increased (0.75 kg) in Cr subjects. Heart rate, blood pressure, and sweat rate responses to exercise were not significantly different between groups. There were no differences in rectal temperature responses in either group. Sodium, potassium, and creatinine excretion rates obtained from 24-h and exercise urine collection periods were not significantly altered in either group. Serum creatinine was elevated in the Cr group but within normal ranges. There were significant exercise-induced increases in cortisol, aldosterone, renin, angiotensin I and II, atrial peptide, and arginine vasopressin. The aldosterone response was slightly greater in the Cr (263%) compared with placebo (224%) group. Peak power was greater in the Cr group during all three 10-s sprints after supplementation and unchanged in the placebo group. There were no reports of adverse symptoms, including muscle cramping during supplementation or exercise. CONCLUSION: Cr supplementation augments repeated sprint cycle performance in the heat without altering thermoregulatory responses.  相似文献

A case of hypotensive shock associated with bradycardia that occurred after injection of nonionic monomeric contrast medium is described. Whether the pathogenesis of this type of reaction is solely vasovagal or is directly caused by the contrast medium is not known. The number of occurrences after administration of nonionic monomeric versus conventional contrast agents should be compared to solve this problem.  相似文献

目的比较寻常型银屑病患者皮损组织与健康人皮肤组织中热休克蛋白的表达情况。方法收集银屑病患者的皮损组织及健康人的皮肤组织,提取蛋白,进行二维凝胶电泳并分析比较。选择在银屑病皮损组织中明显差异表达的蛋白点,进行质谱和生物信息学分析。结果对选择的20个差异表达的蛋白质点进行质谱和生物信息学分析,鉴定了包括热休克蛋白(PHB,HSPβ1,CH60)在内的8个蛋白点的质谱。结论在银屑病患者皮损组织与正常组织中,差异表达的热休克蛋白可能是病理的标志物。  相似文献
