As one of the main components of risk assessment, exposure assessment plays a key role in evaluating risk. Many different scenarios can be developed to estimate the risk from exposure to chemicals such as persistent organic pollutants (POPs). The US Food and Drug Administration (FDA)'s Center for Food Safety and Applied Nutrition (CFSAN) is primarily interested in POPs as humans may be exposed to these compounds through food. Examples of POPs found in food include dioxins, polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers, and some pesticide chemicals. This overview discusses various sources of data that CFSAN has used to estimate dietary exposure to POPs, and provides an example of a recent calculation of an estimate for dietary exposure for consumers in the USA to dioxins in the food supply.  相似文献

Previous research suggests that exposure to persistent organic pollutants (POPs) increases the risk of chronic diseases such as hypertension. We identified the zip codes of more than 800 waste sites contaminated with POPs and other pollutants, based on which we classified zip codes of upstate New York into three groups: "POPs sites", zip codes containing hazardous waste sites with POPs; "other waste sites", zip codes containing hazardous waste sites but not with POPs; and "clean sites", zip codes without any known hazardous waste sites. Age, gender, race, and zip code of residence of patients diagnosed with hypertension (ICD-9 codes 401-404) were identified using the New York Statewide Planning and Research Cooperative System (SPARCS) for the years 1993-2000. A generalized linear model, the negative binomial model, was used to assess the effect of living in a zip code with a hazardous waste site on the discharge rate of hypertension. After control for the aforementioned covariates, we found a statistically significant elevation of 19.2% (95% CI = 8.5%, 31%) in hypertension discharge rate for "POPs sites" and a 10% elevation in discharge rates for "other waste sites" as compared to "clean sites". In a subset of "POPs sites" where people have higher income, smoke less, exercise more and have healthier diets, there was still a 13.9% elevation of hypertension discharge rate as compared to "clean sites". The results support the hypothesis that living near hazardous waste sites, particularly sites containing POPs, may constitute a risk of exposure and of developing hypertension.  相似文献

Persistent organic pollutants include some organo-metals, such as methylmercury; lipophilic halogenated organics, such as dioxins, polychlorinated biphenyls, chlorinated pesticides, and polybrominated flame retardants; and perfluorinated compounds used as repellants. These compounds are resistant to degradation both in the environment and in the human body and tend to bioaccumulate within the food chain. Persistent organic pollutants cause a variety of adverse health effects, including cancer, immune system suppression, decrements in cognitive and neurobehavioral function, disruption of sex steroid and thyroid function, and at least some of them increase the risk of chronic diseases, such as hypertension, cardiovascular disease, and diabetes. Some compounds are byproducts of industry and combustion. Although the manufacture and use of most man-made chemicals has been reduced in recent years, the levels currently present in the population are still associated with an elevated risk of human disease. Others are still manufactured and used. These are dangerous chemicals that have contaminated even areas remote from the industrialized world, such as the polar regions.  相似文献

Feathers as a nondestructive biomonitor for persistent organic pollutants   总被引:1,自引:0,他引:1  
Keratinous tissues of mammals and humans, such as hair, have been used to determine the exposure to polychlorinated biphenyls (PCBs) and organochlorine pesticides. In the present study, we investigated the accumulation of persistent organic pollutants (POPs; PCBs, p,p'-DDT, p,p'-dichlorodiphenyldichloroethane [DDD], p,p'-dichlorodiphenyldichloroethylene [DDE], hexachlorobenzene [HCB], oxychlordane, trans-nonachlor, polybrominated diphenyl ethers, and polybrominated biphenyls [PBBs]) in feathers of the great tit (Parus major). The accumulation of POPs in feathers and fat samples of 27 adult great tits collected in April 2000 and December 2002 was compared with regard to possible temporal and spatial differences. Polybrominated diphenyl ethers, PBBs, trans-nonachlor, and HCB could not be quantified in feathers. Most PCB congeners and DDTs (p,p'-DDE, p,p'-DDD, and p,p'-DDT) could be determined in feathers, although concentrations in feathers were markedly lower (sigmaPCB concentration range, 28.2-87.1 ng/g) than concentrations in fat (sigmaPCB concentration range, 1244-10074 ng/g). Feathers had relatively higher levels of low-chlorinated PCBs (tetra- and pentachlorinated biphenyls) and p,p'-DDD compared to levels in fat, presumably reflecting levels in blood. Persistent organic pollutant concentrations did not increase with the age of the feather, suggesting that exogenous or aerial depositions of POPs onto the feather surface had no, or only minimal, influence on levels in feathers. For almost all compounds studied, POPs in feathers and fat were significantly positively correlated in samples collected during the breeding season but not in samples collected during the winter. This temporal difference may have been caused by seasonal changes in lipid reserves. Although further research is required, feathers appear to be a promising new, nondestructive biomonitor for PCBs and DDTs in avian wildlife.  相似文献

Environmental pollution is ubiquitous and can pose a significant threat to wild populations through declines in fitness and population numbers. To elucidate the impact of marine pollution on a pelagic species, we assessed whether toxic contaminants accumulated in black-footed albatross (Phoebastria nigripes), a wide-ranging North Pacific predator, are correlated with altered physiological function. Blood samples from adult black-footed albatrosses on Midway Atoll, part of the Hawaiian (USA) archipelago, were analyzed for organochlorines (e.g., polychlorinated biphenyls [PCBs] and chlorinated pesticides), trace metals (silver, cadmium, tin, lead, chromium, nickel, copper, zinc, arsenic, selenium, and total mercury), and a sensitive physiological marker, peripheral white blood cell immune function (mitogen-induced lymphocyte proliferation and macrophage phagocytosis). We found a positive significant relationship between organochlorines, which were highly correlated within individual birds (p < 0.001, r > 0.80, Spearman correlation for all comparisons; PCBs, 160 +/- 60 ng/ml plasma [mean +/- standard deviation]; DDTs, 140 +/- 180 ng/ml plasma; chlordanes, 7.0 +/- 3.6 ng/ml plasma; hexachlorobenzene, 2.4 +/- 1.5 ng/ml plasma; n = 15) and increased lymphocyte proliferation (p = 0.020) as well as percentage lymphocytes (p = 0.033). Mercury was elevated in black-footed albatrosses (4,500 +/- 870 ng/ml whole blood, n = 15), and high mercury levels appeared to be associated (p = 0.017) with impaired macrophage phagocytosis. The associations we documented between multiple contaminant concentrations and immune function in endangered black-footed albatrosses provide some of the first evidence that albatrosses in the North Pacific may be affected by environmental contamination. Our results raise concern regarding detrimental health effects in pelagic predators exposed to persistent marine pollutants.  相似文献

The ratification of the Stockholm Convention on Persistent Organic Pollutants is a departure point for several considerations on their presence in environmental matrices. In this paper, the Italian situation is presented with emphasis on the most toxic compounds--chemicals with dioxin-like toxic action, and the activity in the area of interest of the Istituto Superiore di Sanità. Biotic, as well as abiotic matrices have been taken in consideration and, where possible, a comparation between their contamination levels with the levels reported on similar matrices by other European countries has been reported.  相似文献

The uptake of polychlorinated biphenyls (PCBs) was studied in four species of marine algae. A novel experimental system to establish and maintain constant dissolved concentrations of PCBs was employed. Headspace sampling was used to verify that the freely dissolved concentrations remained constant with time. The headspace analysis also allowed sorption to dissolved organic carbon (DOC) to be quantified for all but the most lipophilic PCB congeners. Equilibration with the dissolved phase was rapid for three of the four algae species (<1 d for the majority of congeners). Organic carbon-normalized algae/water partition coefficients (KAlgW) were similar for three of the four species, but were lower by a factor of 10 to 20 for Phaeodactylum tricornutum. The KAlgW values of the first three species were similar to the octanol/water partition coefficient (Kow) for those PCB congeners for which DOC sorption could be quantified. These KAlgW values also agreed well with organic carbon-normalized bioconcentration factors for PCBs in suspended particulate matter (BCF(SPM)) sampled in Baltic Sea surface water during the summer.  相似文献

Schafer and Kegley bring up the important issue of excessive chemicals exposure of children. However, they do not consider in depth the "cumulative and simultaneous exposures faced by children, (.) moving beyond the chemical-by-chemical approach of the past", as quoted from the US Environmental Protection Agency national agenda to protect children from environmental health threats. Existing evidence for contamination by many substances beyond those dealt with in the article calls for additional protective measures. These could include an extra margin of exposure by a factor of 10 to cover cumulation of chemicals, for adults and children alike.  相似文献

The concentrations of a suite of persistent organic chemicals were measured in multiple media in 10 child day care centers located in central North Carolina. Five centers served mainly children from low-income families, as defined by the federal Women, Infants, and Children (WIC) assistance program, and five served mainly children from middle-income families. The targeted chemicals were chosen because of their probable carcinogenicity, acute or chronic toxicity, or hypothesized potential for endocrine system disruption. Targeted compounds included polycyclic aromatic hydrocarbons (PAHs), pentachloro- and nonyl-phenol, bisphenol-A, dibutyl and butylbenzyl phthalate, polychlorinated biphenyls (PCBs), organochlorine pesticides, the organophosphate pesticides diazinon and chlorpyrifos, and the herbicide 2,4-dichlorophenoxyacetic acid (2,4D). Sampled media were indoor and outdoor air, food and beverages, indoor dust, and outdoor play area soil. Concentrations of the targeted compounds were determined using a combination of extraction and analysis methods, depending on the media. Analysis was predominantly by gas chromatography/mass spectrometry (GC/MS) or gas chromatography with electron capture detection (GC/ECD). Concentrations of the targeted pollutants were low and well below the levels generally considered to be of concern as possible health hazards. Potential exposures to the target compounds were estimated from the concentrations in the various media, the children's daily time-activity schedules at day care, and the best currently available estimates of the inhalation rates (8.3 m(3)/day) and soil ingestion rates (100 mg/day) of children ages 3-5. The potential exposures for the target compounds differed depending on the compound class and the sampled media. Potential exposures through dietary ingestion were greater than those through inhalation, which were greater than those through nondietary ingestion, for the total of all PAHs, the phenols, the organophosphate pesticides, and the organochlorine pesticides. Potential exposures through dietary ingestion were greater than those through nondietary ingestion, which were greater than those through inhalation, for those PAHs that are probable human carcinogens (B2 PAH), the phthalate esters, and 2,4D. For the PCBs, exposures through inhalation were greater than those through nondietary ingestion, and exposures through dietary ingestion were smallest. Differences in targeted compound levels between the centers that serve mainly low-income clients and those that serve mainly middle-income clients were small and depended on the compound class and the medium.  相似文献

Phytoplankton and bacteria play an important role on the biogeochemical cycles of persistent organic pollutants (POPs). However, experimental data and quantitative knowledge of the kinetics of uptake and depuration of most POPs by bacteria and phytoplankton are scarce. In the present paper, a procedure to predict the sorption kinetics to bacteria and phytoplankton is developed. The prediction method is the combination of a mechanistic model for sorption and quantitative structure-activity relationships relating bioconcentration factors and membrane permeability to the chemical physical-chemical properties. The model consists of two compartments where the first compartment is the cellular surface and the second compartment is the cell biomass or matrix. Equations for estimating uptake and depuration rate constants into the matrix and adsorption and desorption rate constants onto the surface are obtained. These expressions depend on the physical-chemical properties of the chemical, the environmental temperature, the microorganism size, and species-specific quality of organic matter. While microorganism shape has a secondary influence on uptake dynamics, microorganism size and chemical hydrophobicity arise as the key factors controlling the kinetics of POP incorporation into bacteria and plankton. Uptake, depuration, adsorption, and desorption rate constants are reported for POPs such as polychlorinated biphenyls (PCBs), polycyclic aromatic hydrocarbons (PAHs), polychlorinated dioxins and furans (PCDD/Fs), and POPs of emerging concern, such as polybrominated diphenyl ethers (PBDEs). Finally, implications of uptake and depuration dynamics on the biogeochemical cycling of POPs are discussed.  相似文献

Bans and regulation in many countries have reduced the environmental levels of many persistent organic pollutants (POPs). Despite these declines, there is still evidence of exposures in a range associated with adverse health effects. This seeming paradox is a result of the realisation that levels once presumed safe can cause subtle but important health effects. For example, levels of PCBs now shown to affect human brain development are nearly one million-fold lower than levels previously believed safe. These findings add urgency to efforts to globally eliminate these chemicals, and carry lessons for persistent pollutants that are still in widespread use today, such as the polybrominated diphenyl ethers (PBDEs) and synthetic musks.  相似文献

OBJECTIVE: We sought to study the concentrations of certain persistent organic pollutants with endocrine-disrupting properties in cases with prostate cancer and controls with benign prostate hyperplasia. METHODS: Adipose tissue was obtained from 58 cases and 20 controls. RESULTS: The median concentration among controls was used as cut-off in the statistical analysis. In the total material, a greater-than median concentration of PCB congener 153 yielded an odds ratio (OR) of 3.15 and 95% confidence interval (CI) of 1.04-9.54 and one chlordane type, trans-chlordane, yielded OR 3.49 (95% CI = 1.08-11.2). In the group of case subjects with PSA levels greater than the median level of 16.5 ng/mL, PCB 153 was OR 30.3 (95% CI = 3.24-284), hexachlorobenzene OR = 9.84 (95% CI = 1.99-48.5), trans-chlordane OR = 11.0 (95% CI = 1.87-64.9), and the chlordane-type MC6 OR = 7.58 (95% CI = 1.65-34.9). The grouping of PCBs according to structural and biological activity was found to produce significantly increased risks for enzyme and phenobarbital-inducing PCBs and lower chlorinated PCBs in the case group with PSA levels greater than 16.5 ng/mL. CONCLUSIONS: These chemicals might be of etiologic significance but need to be further investigated. The biological relevance of the arbitrary cut-off point of PSA is unclear.  相似文献

According to the National Implementation Plan on persistent organic pollutants (POP) for the years 2004-2020 the situation on POP was actualised in Latvia. The aim of the study was to assess the contamination of breast milk with polychlorinated biphenyls (PCB), polychlorinated dibenzodioxins, dibenzofurans (PCDD/PCDF) and some pesticides. Breast feeding mothers (together 30) were selected from probably polluted area, and a control group on the basis of WHO criteria for donor selection. The concentration of POP in milk was detected by gas chromatography with HRGC/HRMS, HRGC/ECD and GC/ECD detection after extraction of milk fat and purification. The results of the survey show comparability of both groups in age, weight, habits, occupation as well as age of the children and the weight at birth and at the time of collecting the samples. PCB were not observed in 15.9%, PCDD/PCDF in 6.6% and pesticides in 14% analyses of the target group and in 26.3%, 7% and 8.6% analyses correspondingly of the control group. The concentration of 18 detected PCB, 17 detected PCDD/PCDF and 8 chlorinated pesticides and its metabolites varied in wide range and the differences of mean values within groups were not statistically significant (p>0.05). The concentration of POPs corresponds to lowest levels detected in European countries. The concentration of dioxine-like PCBs and PCDD/PCDF according to toxicity equivalents does not exceed the WHO accepted level of 15 TEQ pg/g milk fat.  相似文献

Exposure to environmental pollutants may contribute to the development of coronary heart disease (CHD). We determined the ZIP codes containing or abutting each of the approximately 900 hazardous waste sites in New York and identified the major contaminants in each. Three categories of ZIP codes were then distinguished: those containing or abutting sites contaminated with persistent organic pollutants (POPs), those containing only other types of wastes ("other waste"), and those not containing any identified hazardous waste site ("clean"). Effects of residence in each of these ZIP codes on CHD and acute myocardial infarction (AMI) hospital discharge rates were assessed with a negative binomial model, adjusting for age, sex, race, income, and health insurance coverage. Patients living in ZIP codes contaminated with POPs had a statistically significant 15.0% elevation in CHD hospital discharge rates and a 20.0% elevation in AMI discharge rates compared with clean ZIP codes. In neither of the comparisons were rates in other-waste sites significantly greater than in clean sites. In a subset of POP ZIP codes along the Hudson River, where average income is higher and there is less smoking, better diet, and more exercise, the rate of hospitalization for CHD was 35.8% greater and for AMI 39.1% greater than in clean sites. Although the cross-sectional design of the study prevents definite conclusions on causal inference, the results indirectly support the hypothesis that living near a POP-contaminated site constitutes a risk of exposure and of development of CHD and AMI.  相似文献
