目的探讨细胞因子TNF-α、IL-6与IL-8在子宫内膜异位症(内异症)的表达及其与盆腔粘连程度的相关性。方法选取子宫内膜异位症患者50例,采用ELISA法分别测定手术前血清、术中腹腔液及术后3个月血清标本TNF-α、IL-6、IL-8含量,并分析与粘连的相关性。结果 50例内异症患者中,41例(80.2%)患者合并不同程度的盆腔粘连,内异症患者血清、腹腔液中TNF-α、IL-6及IL-8显著高于对照组(P<0.05)。内异症Ⅲ~Ⅳ期期患者腹腔液中TNF-α、IL-6及IL-8的水平显著高于Ⅰ~Ⅱ期,Ⅰ~Ⅱ期Ⅲ~Ⅳ期期患者TNF-α水平显著高于对照组(P<0.05);术后3个月血清TNF-α、IL-6及IL-8较术前明显下降(P<0.05)。盆腔粘连程度与腹腔液IL-6、IL-8、TNF-α均呈正相关(r=-0.457,r=0336,r=0263,P<0.05);术前血清细胞因子除IL-8(r=0.135,P>0.05),TNF-α、IL-6与粘连呈正相关(r=0.521,r=-0.470,P<0.05)。结论细胞因子TNF-α、IL-6及IL-8与内异症临床分期和治疗疗效密切相关,检测TNF-α、IL-6及IL-8患者血清的变化,可作为子宫内膜异位症术前评估、疗效评价、复发的重要指标。  相似文献

目的:探讨肾性继发性甲状旁腺功能亢进(SHPT)患者血清同型半胱氨酸(Hcy)、血管细胞黏附分子-1(VCAM-1)水平及其与认知功能的相关性。方法:选择2020年01月—2021年12月本院收治的发生轻度认知障碍的SHPT患者113例作为研究对象,选择同期未发生认知障碍的110例患者作为对照组,检测血清Hcy、VCAM-1水平,ROC曲线分析血清Hcy、VCAM-1对SHPT患者发生认知功能障碍的诊断价值,Logistics多因素回归分析影响SHPT患者发生认知功能障碍的危险因素。结果:与认知功能正常组比较,认知功能障碍组血液透析比例、血清iPTH、Scr、血磷、血钙、Hcy、VCAM-1水平显著升高,MoCA评分显著降低(P<0.05);血清Hcy、VCAM-1及联合诊断SHPT患者发生认知功能障碍的曲线下面积(AUC)分别为0.828、0.850、0.914;透析方式、血清iPTH、受教育年限、Hcy、VCAM-1是影响SHPT患者发生认知功能障碍的危险因素(P<0.05)。结论:SHPT患者血清Hcy、VCAM-1水平升高,与患者认知功能有关,是发生认知功能障碍的独...  相似文献

目的 :探讨子宫内膜异位症腹腔巨噬细胞释放血管内皮细胞生长因子( VEGF)的变化。方法 :将行诊断性腹腔镜检查的不育患者 45例 ,根据术后诊断分为子宫内膜异位症组 ( 2 5例 )和对照组 ( 2 0例 ) ,采用酶联免疫吸附测定检测腹腔液和腹腔巨噬细胞培养上清液中 VEGF的水平。结果 :子宫内膜异位症患者腹腔液及腹腔巨噬细胞培养上清液中 VEGF的水平均显著高于对照组 ( P<0 .0 1 )。结论 :腹腔巨噬细胞分泌能力增强可能是导致子宫内膜异位症患者腹腔液 VEGF水平增高的原因  相似文献

探讨早期肠内营养联合腹腔引流在改善重症急性胰腺炎(SAP)患者炎症、免疫及营养方面的应用价值。74例SAP患者按照随机数字表法分为对照组及观察组,各37例。对照组给予胰腺炎传统综合治疗,观察组在对照组的基础上给予早期肠内营养联合腹腔引流治疗。比较治疗前后两组患者炎症因子、免疫功能指标及营养指标水平变化。治疗后1、2周两组患者血清CRP、TNF-α、IL-6等炎性因子水平较治疗前降低(P<0.05或P<0.01),且观察组较对照组降低更明显(P<0.01);两组患者血清IL-10水平、CD3+、CD4+、CD4+/CD8+、IgA、IgG及血清白蛋白(ALB)、前白蛋白(PA)及转铁蛋白(TRF)等水平均较治疗前升高(P<0.05或P<0.01),且观察组显著高于对照组(P<0.05或P<0.01)。早期肠内营养联合腹腔引流治疗SAP可显著减轻机体炎症反应,改善患者免疫功能及营养状态,有助于患者康复。  相似文献

目的观察子宫内膜异位症(EMs)患者自然周期卵泡液中细胞因子的表达情况,探讨其与EMs的相关性。方法收集2013~2016年间在瑞士伯尔尼大学妇女医院生殖中心因EMs行自然周期IVF-ET或卵胞浆内单精子注射(ICSI)助孕治疗的17例患者为研究对象(EMs组),其中轻度EMs患者7例,中重度EMs患者10例。同时收集同期17例年龄匹配,因输卵管因素或男方因素在该中心行自然周期IVF/ICSI-ET助孕的患者做为对照组。在自然周期采卵日收集卵泡液,通过Bio-Plex技术检测卵泡液中白介素(IL)-1β、IL-6、IL-8、IL-15、IL-18、调节活化正常T细胞表达与分泌的趋化因子(RANTES)、肿瘤坏死因子(TNF)-α、血管内皮生长因子(VEGF)、白血病抑制因子(LIF)等9种细胞因子的浓度;ELISA法检测单核细胞趋化因子-1(MCP-1)和瘦素(Leptin)的浓度,分析两组间细胞因子浓度的差异及其与EMs的相关性。结果 IL-1β和IL-6在EMs患者卵泡液中的浓度显著高于对照组(中位数比较分别为0.8pg/ml vs.0pg/ml和9.1pg/ml vs.6.1pg/ml,P均0.05)。中重度EMs患者卵泡液中IL-1β浓度(1.44pg/ml)显著高于轻度EMs及对照组(分别为0.26pg/ml、0pg/ml),VEGF浓度(2 464pg/ml)亦显著高于轻度EMs及对照组(分别为1 268pg/ml、1 560pg/ml,P均0.05)。通过Spearman相关分析发现卵泡液中只有IL-1β和IL-6的浓度与EMs相关,大部分细胞因子相互之间存在较强的相关性。结论 EMs患者自然周期卵泡液中细胞因子微环境发生了变化,IL-1β和IL-6的浓度均异常增高,且大部分细胞因子相互之间存在较强的相关性,可能与EMs患者生育力降低有关。  相似文献

目的观察类风湿性关节炎(RA)患者血清及关节液SOCS-1、SOCS-3蛋白含量的变化,探讨其在炎症中的意义。方法 49例类风湿性关节炎患者根据疾病活动性分为活动期组(26例)及缓解期组(23例),采用ELISA法检测其血清及关节液SOCS-1、SOCS-3蛋白含量,并与20例健康体检者(对照组)进行比较。结果与对照组比较,RA患者中活动期组与缓解期组血清SOCS-1、SOCS-3蛋白含量均明显升高(P<0.01),其中活动期组血清SOCS-1、SOCS-3含量表达较缓解组明显升高(P<0.01、P<0.05);活动期组患者关节积液SOCS-1、SOCS-3蛋白表达较缓解期组明显升高(P<0.05);活动期组患者血清SOCS-1、SOCS-3蛋白水平均与血清ESR、CRP、RF呈正相关(P<0.01、P<0.05)。结论 SOCS 1/3可能参与了RA炎症反应的病理过程,且可能与RA病情活动性有关。  相似文献

目的 探讨术中腹腔灌洗液细胞学检测(peritoneal lavage cytology,PLC)和癌胚抗原蛋白测定(pCEA)对胃癌患者术后预测腹膜转移及判断预后的价值。方法 术中采集57例进展期胃癌患者和8例胃良性疾病患者的腹腔灌洗液,进行细胞学检测和放射免疫法测定pCEA水平。结果 胃癌患者腹腔液pCEA阳性率(43.9%)明显高于PLC(26.3%)(P<0.05),PLC和pCEA阳性率与肿瘤浸润深度和淋巴结转移密切相关(P<0.05~0.01)。与PLC相比,pCEA对胃癌术后预测腹膜转移有较高的敏感度(94.7%)和较低的假阴性率(5.3%)。PLC阳性组及pCEA阳性组术后1、3、5年生存率均明显低于PLC阴性组及pCEA阴性组(P<0.05)。多因素分析表明pCEA是影响胃癌患者生存率的独立预后因素。结论 对于胃癌患者术后腹膜转移的预测和预后判断,术中pCEA测定是一种较PLC更加灵敏、可靠的指标。  相似文献

目的探讨胫骨横向骨搬移术对糖尿病足患者血清中血管生成相关因子表达的影响。方法回顾分析2018年1月-12月收治的10例Wagner 4期糖尿病足患者临床资料,男5例,女5例;年龄51~70岁,平均59.2岁。糖尿病病程2~60个月,平均24.2个月;糖尿病足病程30~120 d,平均54.1 d。分别于术前1 d及术后1、4、11、18、28、35 d抽取外周静脉血,离心取血清采用ELISA法检测血管生成相关因子VEGF、bFGF、EGF、PDGF的表达水平。结果患者血清VEGF、bFGF及EGF表达水平均在术后11 d突然升高,11、18、28、35 d各因子表达水平均显著高于术前,差异有统计学意义(P<0.05);PDGF表达水平于术后18 d突然升高,18、28、35 d PDGF表达水平显著高于术前,差异有统计学意义(P<0.05)。结论胫骨横向骨搬移术治疗糖尿病足,早期可显著增加血清血管生成相关因子的表达,可能是其促进糖尿病足创面愈合的作用机制。  相似文献

目的探讨血清及卵泡液瘦素(leptin)水平与多囊卵巢综合征(PCOS)发病的关系和对体外受精-胚胎移植(IVF-ET)结局的影响。方法采用放射免疫法测定进行IVF-ET的20例PCOS患者(PCOS组)和20例非PCOS患者(对照组)卵泡液中leptin、胰岛素(INS)水平以及同期血清中leptin水平,PCOS患者经二甲双胍和口服避孕药(OC)预治疗三个月。结果PCOS组卵泡液及血清leptin水平均高于对照组(12.87±7.34 vs 10.10±5.15)、(13.65±7.75 vs 10.53±4.65),但差异无显著性,卵泡液及血清leptin水平均与卵泡液INS水平呈正相关(r=0.578、r=0.682,P均<0.01)。PCOS患者卵泡液低leptin组成熟卵细胞率高于高leptin组,差异有统计学意义(94%vs 86%,P<0.05),血清leptin水平与卵裂率呈负相关(r=-0.501,P<0.05)。结论PCOS患者体内leptin可能与卵泡液INS共同参与PCOS发生发展,leptin可能主要通过影响卵母细胞成熟、受精卵分裂而影响妊娠成功率。  相似文献

探讨IgA肾病(IgA nephropathy, IgAN)患者血清及尿液半乳糖缺陷IgA1(galactose-deficient IgA1, Gd-IgA1)水平与临床病理及肠黏膜屏障损伤指标的相关关系。酶联免疫吸附测定法检测血尿Gd-IgA1、炎性因子及肠黏膜屏障损伤指标水平。IgAN组45例, 健康对照组25例。结果显示IgAN组患者血清炎性因子肿瘤坏死因子α和白细胞介素6及血尿Gd-IgA1水平高于健康对照组人群(均P<0.05);Spearman秩相关分析结果显示尿Gd-IgA1水平与血肌酐、24 h尿蛋白量及肾脏损伤程度呈正相关(均P<0.05);IgAN组患者肠道黏膜屏障损伤指标细胞黏附分子1、D-乳酸、脂多糖、二胺氧化酶水平高于健康对照组人群(均P<0.05), 且与尿Gd-IgA1水平呈正相关(均P<0.05)。本研究结果提示尿Gd-IgA1水平与IgAN严重程度及肠道黏膜屏障损伤指标相关。  相似文献

BACKGROUND: Sepsis-induced cardiac dysfunction occurs commonly in critically ill patients and is associated with high mortality rates. Neutrophils play a central role in sepsis-induced lung and liver injury; however, the mechanism of sepsis-induced cardiac dysfunction remains unclear. Vascular cell adhesion molecule-1 (VCAM-1) has been implicated in neutrophil-mediated liver injury during endotoxemia and is also expressed in myocardium. The purposes of this study were to examine the temporal relationship of myocardial VCAM-1 expression with neutrophil accumulation during endotoxemia and to determine whether VCAM-1 mediates neutrophil accumulation and cardiac dysfunction during endotoxemia. METHODS: Mice were subjected to lipopolysaccharide (LPS; 0.5 mg/kg, intraperitoneally). Myocardial VCAM-1 expression and neutrophil accumulation were determined by immunofluorescence staining. Cardiac performance with or without VCAM-1 blocking antibody (5 mg/kg, intravenously) was determined by the Langendorff technique. RESULTS: LPS caused a time-dependent increase in both myocardial VCAM-1 expression and neutrophil accumulation. At 6 hours after LPS, the immunofluorescent intensity for VCAM-1 increased from 2.5 +/- 0.6 x 10(6) in saline solution controls to 19.9 +/- 3.5 x 10(6) (P <.05, analysis of variance), and neutrophil count increased from 2.4 +/- 1.7/mm(2) in saline solution controls to 13.0 +/- 2.5/mm(2) (P <.05). Left ventricular developed pressure was decreased maximally at 6 hours after LPS compared with saline solution controls (29.1 +/- 1.1 mm Hg vs 53.1 +/- 3.9 mm Hg; P <.05). Treatment with VCAM-1 monoclonal antibody abrogated both myocardial neutrophil accumulation and cardiac dysfunction during endotoxemia. CONCLUSIONS: LPS-induced myocardial dysfunction is associated with increased expression of VCAM-1 and with neutrophil accumulation. Blockade of VCAM-1 abrogates myocardial neutrophil accumulation and preserves cardiac function during endotoxemia, which supports a role for VCAM-1 as a therapeutic target for myocardial protection during sepsis.  相似文献

OBJECT: The aim of this study was to explore whether levels of intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) are elevated in the cerebrospinal fluid (CSF) and serum of patients after aneurysmal subarachnoid hemorrhage (SAH). METHODS: This prospective clinical study focused on 21 patients who had recently suffered an SAH due to aneurysmal rupture and 15 control patients with hydrocephalus who had no other central nervous system disease. Cerebrospinal fluid and serum samples obtained within the first 3 days and on the 5th and 7th days of SAH were assayed for ICAM-1 and VCAM-1 by using quantitative enzyme-linked immunosorbent assays. Levels of soluble forms of ICAM-1 (p = 0.00001) and VCAM-1 (p = 0.009) in the patients' CSF and those of ICAM-1 (p = 0.00001) and VCAM-1 (p = 0.00001) in their serum were found to be elevated after SAH compared with levels in the CSF and serum of control patients with hydrocephalus. In addition, when the authors compared the increased levels of adhesion molecules in the CSF and serum of patients after SAH, the only statistically insignificant difference that they found was between the levels of VCAM-1 in serum obtained on Days 5 and 7 after SAH (p = 0.27). CONCLUSIONS: Adhesion molecules are a group of macromolecules that may participate in the inflammatory process, a common pathway leading to vasospasm after SAH. Leukocyte adherence to the vascular endothelium, which is induced by adhesion molecules, has been believed to be the initial signal of the development of vasospasm. The authors have demonstrated the synchronized elevation of two adhesion molecules in both CSF and serum following aneurysmal SAH. Blocking of ICAM-1 as well as VCAM-1 by monoclonal antibodies post-SAH may provide a beneficial effect on vasospasm.  相似文献

An association between cytomegalovirus (CMV) infection and alloresponse has been suggested. CMV increases inflammation and adhesion molecule expression in graft, and induces cytokines and growth factors, linked with transplant vasculopathy and chronic rejection. We have investigated the gene expression of various inflammatory factors in the CMV-associated immune response and compared this with the immune response of acute rejection in liver transplants by using DNA microarray technology. Gene expression was studied at mRNA level in biopsies from liver transplant patients experiencing CMV infection or acute rejection. RNA extracted from liver grafts after reperfusion was used as control material. Among the strongly upregulated genes in the specimens obtained from liver transplants during CMV infection were IFN-gamma, caspases 1 and 3, granzymes A and B, TGF-beta receptors II and III, IL-10 receptor alpha, VCAM-1, TNF receptor, IL-4, TNF-alpha, IL-10, IL-2 receptor beta, IL-1beta, PDGF-receptor beta, vascular adhesion protein-1, TGF-beta2, and ICAM-1. In biopsies with acute liver allograft rejection, the most significantly upregulated genes were MHC class II, IFN-gamma, caspases 1 and 3, IL-2R beta and gamma, granzymes A and B, VLA-4, L-selectin, E-selectin, VCAM-1, and IL-1beta. Upregulated genes common for CMV and alloresponse were granzyme A and B, E-selection, IFN-gamma, VCAM-1, VLA-4, TNF, caspases 1, 3, and 8, and PDGF. Microarray analysis defined different entities in the immune responses of CMV infection and acute rejection. The differences and similarities of the gene expression profiles related to those in CMV infection and rejection may help to understand the intragraft immunologic events.  相似文献

BACKGROUND: Antineutrophil cytoplasmic autoantibodies (ANCA) play a role in the expression of adhesion molecules. Differences in ANCA test results in ANCA-associated vasculitis may provide differences in their renal expression. PATIENTS AND METHODS: We assessed the renal expression of ICAM-1 and VCAM-1 with monoclonal antibodies in 19 patients with ANCA-vasculitis: 7 microscopic polyangiitis, 5 Wegener's granulomatosis, 4 renal-limited vasculitis and 3 Churg-Strauss disease. Immunofluorescence and ELISA for myeloperoxidase (MPO) and proteinase 3 (PR3) were performed for ANCA-testing. 10 normal renal tissues were used as controls. RESULTS: The ANCA staining pattern was perinuclear in 14 patients, with MPO-ANCA 31 - 220 EU/ml, and cytoplasmic in 5, with PR3-ANCA 37 - 144 EU/ml. Abnormal tubular expression of ICAM-1 and VCAM-1 was seen in more than 80% of biopsies and abnormal expression of VCAM-1 in glomerular tuft was seen in 60%. Glomerular tuft stains of ++ or +++ for VCAM-1 were observed in 10% of renal biopsies from MPO-ANCA-GN patients, but in 60% of biopsies from PR3-ANCA-GN patients (Fi = 8.538, p = 0.03). IN CONCLUSION: De novo expression of VCAM-1 on glomerular tuft suggests that the endothelial cells play a role in ANCA-GN. De novo glomerular expression of VCAM-1 is associated more with ANCA directed against PR3 than with ANCA directed against MPO. Upregulated glomerular expression of VCAM-1 may reflect a higher histological activity in patients with PR3-ANCA, and supports the existence of specific immune activation mechanisms in the different serologic subgroups in ANCA-GN. The de novo tubular expression of ICAM-1 and VCAM-1 suggests that the epithelial cells may participate in adhesive interactions in ANCA-GN.  相似文献

BACKGROUND: The purpose of the present study was to investigate the relationship between serum concentrations of the immunoglobulin-like superfamily, selectins, hsCRP, and the development of DIND in patients with aneurysmal SAH. METHODS: Serum ICAM-1, VCAM-1, E-selectin, P-selectin, L-selectin, and hsCRP were measured in 33 patients with SAH who underwent aneurysmal clipping within 48 hours of the onset of symptoms. Serum samples were obtained during the early period (day 0) and the late period (day 7). RESULTS: The serum concentrations of ICAM-1 (P = .009), VCAM-1 (P = .0383) and hsCRP (P = .0014) during the early period were significantly higher in patients with SAH than in control patients. Further, serum hsCRP concentration during the late period was significantly higher in patients with SAH than in control patients (P = 0033). Finally, serum concentrations of ICAM-1, VCAM-1, and hsCRP during the early (P = .0055, P = .0266, and P = .0266) and late (P = .0423, P = .0041, and P = .0004) period were significantly higher in patients with DIND than in patients without DIND. CONCLUSIONS: Serum levels of ICAM-1, VCAM-1 and hsCRP during the early and late period following SAH correlate with DIND.  相似文献

利用间接免疫荧光和膜免疫印迹方法,观察了血管细胞粘附分子(VCAM-1)在人类狼疮肾炎和新月体肾炎患者肾组织及血清中的表达。结果表明:狼疮肾炎(Ⅱ、Ⅳ、Ⅴ型)和新月体肾炎患者沿肾小球毛细血管壁可见VCAM-1特异性荧光;肾小球系膜区VCAM-1表达呈现增强的趋势;与轻度系膜增生性肾炎比较,狼疮肾炎(Ⅳ、Ⅱ型)患者血清中VCAM-1水平显著增加(P<0.01)。提示VCAM-1在狼疮肾炎和新月体肾炎的发生发展中可能具有重要作用。  相似文献

Aim: To investigate the relationship between androgen level and the indexes indicating endothelial function in male patients with coronary heart disease (CHD). Methods: We registered the following data for 106 50-70-year-old men: age, weight, blood lipid, including total cholesterol, low density lipoprotein cholesterol, high density lipoprotein cholesterol and triglyceride, whether a smoker, sugar levels, blood pressure, free testosterone (FT), vascular cell adhesion molecule- 1 (VCAM- 1) and the intima-media thickness (IMT) of common carotid artery, common carotid diameter, maximum velocity in systolic phase, minimum velocity in diastolic phase and resistent index. Among the 106 men, 51 were patients with CHD. The relationships between FT level, VCAM-1 concentration and IMT were examined, respectively, using a stepwise linear regression technique among all the 106 men. Results: There was no statistical difference in terms of age, blood pressure, whether a smoker, sugar levels, HDL-C, minimum velocity in diastolic phase, resistent index between male CHD patients and controls; whereas results for weight, total cholesterol, low density lipoprotein cholesterol, triglyceride, VCAM- 1 and IMT of male CHD patients were higher than those of controls; FT level and maximum velocity in systolic phase were lower. It was found that among all the objects, FT level was inversely correlated with IMT and VCAM-1 concentration. 目的了解子宫内膜异位症(Em)患者的在位内膜在分泌RANTES及对单核细胞的趋化活性是否有别于非Em患者子宫内膜。方法白细胞介素(IL)-1β刺激培养Em与非Em患者子宫内膜基质细胞,分别在刺激培养后12、24、36、48、60、72、84和96 h取上清酶联免疫吸附试验(ELISA)检测RANTES水平;Boyden小室趋化实验分析刺激培养上清液对单核细胞的趋化活性及RANTES趋化单核细胞的能力。结果Em在位内膜从刺激培养60 h始,RANTES分泌量及趋化单核细胞的能力显著高于非Em在位内膜;Em在位内膜基质细胞刺激培养60 h上清所趋化的单核细胞中有55%是由RAN-TES趋化而来。结论在相似的培养条件下,Em患者的在位内膜基质细胞RANTES的表达模式和趋化单核细胞的活性不同于非Em患者子宫内膜。  相似文献
