The expression of Aurora B in normal endometria and endometrial carcinomas and its relation with clinicopathologic parameters of endometrial carcinomas were investigated.Streptavidin-biotin peroxidase(SP) immunohistochemical technique was used to detect the expression of Aurora B in 10 cases of normal proliferative phase endometria,10 cases of normal secretory phase endometria and 72 cases of endometrial carcinomas respectively.According to the 1988 International Federation of Gynecology and Obstetrics(FIGO) grade,there were 37 patients in grade 1,23 in grade 2 and 12 in grade 3 respectively.According to the FIGO stage,there were 59 patients in stage Ⅰ-Ⅱ and 13 patients in stage Ⅲ-Ⅳ.Aurora B was expressed in both normal proliferative phase endometria,secretory phase endometria and endometrial carcinomas,but its positive labeling index(PLI) in proliferative phase endometria was significantly higher than that in secretory phase endo-metria(P<0.01) and endometrial carcinomas(P<0.01).The PLI of Aurora B was lower in tumors with well differentiation(G1),low surgical staging(Ⅰ-Ⅱ),and ≤1/2 myometrial invasion than that in tumors with moderate and low differentiation(G2-G3),higher surgical staging(Ⅲ-Ⅳ),and >1/2 myometrial invasion(all P<0.01).Aurora B exerts its functions in the replication of normal endometrial glandular cells;Expression of Aurora B is significantly correlated with biologic behavior of endo-metrial carcinoma,indicating that Aurora B may be a promising prognostic factor in endometrial carcinoma.  相似文献

Objective To determine the number of goblet cells, the change of MUC5AC expression in chronic obstructive pul monary disease (COPD) patients and the relationship of smoking with goblet cell, MUC5AC, and lung function. Methods Eighteen patients undergoing lung resections for a solitary peripheral carcinoma were classified by lung function as having COPD. Twenty patients with normal lung function served as the control group. Normal lobe bronchioles far away from the lesion site were taken for paraffin section. Goblet cells were identified by AB/PAS staining and the ex pressionof MUC5AC in the paraffin's section was tested by immunohistochemistry. Results Goblet cell hyperplasia was observed in the COPD group. The positive rate of goblet cell in COPD group (0.20% ± 0.10%) was significantly higher than that in the normal lung function group (0.13% ± 0.06%, P < 0.05). The positive rate of MUC5AC expression in the COPD group (0.27% ± 0.09%) was higher than that in the normal lung function group (0.20% ± 0.10%, P <0.05).The positive rate of goblet cell in smokers (27.93% ± 9.00%) of the COPD group and normal lung function group was higher than that in non-smokers (17.70% ± 9.37%, P < 0.05), while MUC5AC expression had no significant difference between smokers and non-smokers (17.88% ± 6.44% and 10.88% ± 7.10%, respectively). Conclusion For COPD patients with declined lung function, there were goblet cell hyperplasia and increased expres sion of MUC5AC. MUC5AC expression up-regulation may due to goblet cell hyperplasia. Smoking may be an important factor for goblet cell hyperplasia.  相似文献

Objective: To investigate the clinical significance of survivin in endometrial carcinoma and to investigate the relationship between the expression of survivin and Ki-67. Methods: Immunohistochemical S-P (streptavidin-biotin-peroxidase complex)method was performed to detect the expression of survivin and Ki-67 antigen in 15 cases of normal endometrium, 21 cases of endometrial simple and complex hyperplasia, 22 cases of endometrial atypical hyperplasia, and 61 cases of endometrial carcinoma. Results: Survivin was hardly detected in some normal endometrium in the proliferative phase and in the secretory phase. However, the level of survivin expression in atypical hyperplasia endometrium(72.73%)was higher than that in normal en- dometrium (7.14%)(P 〈 0.05), including simple and complex hyperplasia (42.38%)(P 〈 0.01 ), and was lower than that in endometrial carcinoma(90.17%)(P 〈 0.05). Moreover, significant correlation was present between the expression of survivin and the characteristics of endometrial carcinoma, including clinical stage, histological grade and the presence of invasion to myometrium (P 〈 0.05). In addition, Ki-67 antigen expression was positively correlated with survivin expression in all specimen. Ki-67 labeled indexes (LIs)in hyperplasia endometrium were significantly lower than those in atypical hyperplasia endometrium and endometrial carcinoma (P 〈 0.01 ), while there was no significant difference in Ki-67 LIs between atypical hyperplasia endometrium and endometrial carcinoma(P 〉 0.05). There was no significant relationship between Ki-67 LIs and the characteristics of endometrial carcinoma, including histological grade, clinical stage or the invasion to myometrium(P 〉 0.05). Conclusion: Survivin may participate in the onset and progression of endometrial carcinoma through inhibiting apoptosis and promoting proliferation. Survivin expression is correlated with the malignant degree and prognosis of tumor. Ki-67 is also associated with carcinogenesis and progression of endometrial carcinoma. The results suggest that survivin could be a diagnostic and prognostic marker for endometrial carcinoma and might provide pathways to treat the patients with recurrent or refractory or rudimental endometrial carcinoma.  相似文献

Objective: To observe the effect of the artesunate (ART) on cellular proliferation in vitro, to search for the possible anti-tumor mechanism of ART on endometrial carcinoma at the molecular level and to provide the experimental and theoretical foundations for the clinical applications of ART. Methods: The cell proliferation was observed by microscope; MTT was used to examine the effects of ART on proliferation of HEC-1B cells, and flow cytometric analysis was used to detect cell cycle and apoptosis. The human endometrial carcinoma HEC-1B cells were conventionally cultured; ART was administered with a concentration of 40 μg/ml before the total RNA were extracted, mRNA expression of Survivin, Caspase-3, N-Cadherin, E-Cadherin, Fibronectinl and Cox-2 were detected using RT-PCR. Results: ART reduced proliferation in human endometrial carcinoma cell line HEC-1B in a dose- and time-dependent effect. The cells of G0/G1 stage were significantly increased (P〈0.05), but the cells of G2/M stages were significantly decreased (P〈0.05), so it has shown that the cell cycle was probably blocked in G0/G1 stage. After intervention with ART at 20 and 80 μg/ml for 48 h, cellular apoptosis rate respectively was (36.42±0.77)% and (11.77±0.58)%, and the difference was statistically significant compared with the control ([6.64±0.191%, P〈0.01). The expression of Cox-2 mRNA in the ART group was lower than those of control group, yet the expression of Caspase-3 and E-Cadherin mRNA in the ART group was higher than those of control group. Conclusion: ART can inhibit HEC-1B cell growth and proliferation in a dose- and time-dependent manner. Furthermore, ART can induce apoptosis in a dose-dependent manner. ART is able to downregulate Cox-2 mRNA expression and to upregulate E-Cadherin and Caspase-3 mRNA expression. So we can conclude that ART could induce the endometrial carcinoma HEC-1B cell apoptosis and inhibit tumor cell proliferation.  相似文献

In this experiment lung carcinoma was induced by crocidolite plus benzo(a) pyrene in rat.From the cancer models,we observed that the incidence(16.0%) of lung carcinomas was lower,and the survival time(376days)of the first case of carcinoma and the mean survival time(758days)of the rats with carcinoma were higher in the group of rats drinking 2% green tex extract for life than in the positive group(without drinking green tex extract).The mortality ratio(0.5047)was smaller in the experimental group than in the positive control group,and the survival curve of the experimental group significantly raised up,in comparison with the positive group.  相似文献

Objective To investigate the expression of Survivin mRNA in lung cancer tissue microarray (TMA) by fluorescence in situ hybridization (FISH) method, and determine the role and significance of it in lung cancer genesis and progress. Methods The expression of Survivin mRNA was detected by FISH method and TMA technology. Fifty-four cases of lung cancer and 10 cases of normal lung tissue were examined. Results Survivin mRNA was expressed in 66.7% (36/54) of lung cancer; the positive ratio of lung cancer was significantly higher than that of normal lung tissue (0/10; x2= 15.238, P < 0.05). The positive ratio of Survivin mRNA was significantly higher in poor differentiated cancer (20/24, 83.3%) than moderate and well differentiated cancer (16/30, 53.3%; x2= 5.40, P <0.05). The positive ratio of Survivin mRNA was significantly higher in group with lymph node metastasis (27/32, 84.4%) than without lymph node metastasis (9/22, 40.9%; x2= 11.084, P < 0.05). The positive ratio of Survivin mRNA was significantly higher in stage Ⅲ-Ⅳ(12/13, 92.3%) than stage Ⅰ - Ⅱ (24/41, 58.5%; x2= 5.066, P < 0.05). Conclusion Survivin mRNA highly expresses in lung cancer, which is related to the progress and malignant behavior. Survivin may play a promoting role in lung cancer genesis and progress and provide a basis for estimating prognosis and treatment.  相似文献

目的 研究孤啡肽在子宫腺肌病患者组织中的表达与患者痛经程度的关系.方法 收集吉林大学第二附属医院妇产科2007年12月至2008年6月,年龄41~52岁行全子宫切除术的子宫标本共55例,分为病例组(子宫腺肌病组)34例和对照组(子宫肌瘤组)21例,采用免疫组织化学方法检测病例组的在位内膜、异位内膜及对照组正常子宫内膜及肌层组织中孤啡肽的表达,并初步探讨孤啡肽表达与患者痛经关系的临床意义.结果 孤啡肽在病例组病灶及在位内膜组织中表达的计分分别为7.2±1.6及7.1±1.7,对照组在肌层及内膜组织中表达的计分分别为2.6±1.2及2.7±2.0,P<0.05;孤啡肽在病例组痛经组病灶及在位内膜组织中表达计分为7.5±1.5,7.4±1.5,在无痛经组病灶及在位内膜组织中表达的计分分别为5.1±1.2,5.0±1.2,P<0.05;孤啡肽的表达在病例组重度痛经组肌层及在位内膜组织表达最高计分分别为8.5±1.2,8.4±1.3,无痛经组计分分别为5.1±1.2,5.0±1.2,轻度痛经组计分分别为6.9±1.4,6.8±1.4,后两者比较差异无统计学意义(P>0.05),中度痛经、轻度痛经及无痛经组在位内膜及肌层组织中的表达逐渐降低.结论 孤啡肽在子宫腺肌病中的高表达可能是引起痛经的因素之一;患者的痛经程度与在位内膜的孤啡肽表达呈正相关,与病灶组织的孤啡肽表达也呈正相关;动态检测在位内膜中孤啡肽的表达,以监测子宫腺肌病的发展变化.
Abstract:
Objective To study the expression of nociceptin in adenomyosis tissue and understand its relationship with the severity of dysmenorrhea. Methods The myometrial specimens were collected from 55 patients undergoing hysterectomy at our hospital during December 2007 and June 2008. Their mean age was 41 -52 years old. There were 34 adenomyosis patients in case group and 21 other patients in control group. Immunohistochemical staining was employed to detect nociceptin in eutopic endometrium and ectopic endometrial tissues in case group and normal endometrium and myometrial tissues in control group. Results The expression of nociceptin was significantly higher in the eutopic endometrial tissue ( 7. 1 ± 1.7 ) of case group than those in endometrial(2.7±2.0 ) and myometrial tissues of control group ( P < 0.05 ); The expression of nociceptin was higher in the eutopic endometrial tissue (7.4±1.5) of dysmenorrhea group than that of non-dysmenorrhea group ( 5.0 ± 1.2 ) in case group ( P < 0.05 ); The expression of nociceptin in eutopic endometrium and myometrium was the highest in severe dysmenorrhea group ( 8.4 ±1.3 ). And the non-dysmenorrhea and mild dysmenorrheal groups had no significant differences [(5.0 ± 1.2 ) vs (6.8 ± 1.4)]. And it decreased gradually in moderate dysmenorrhea, mild dysmenorrhea and nondysmenorrhea groups. Conclusion A high expression of nociceptin in adenomyosis may be one of the casual factors of dysmenorrhea. The severity of dysmenorrhea is positively correlated with the expression of nociceptin in eutopic endometrial tissue in case group ( r = 0.515, P = 0.034) and ectopic endometrium tissue. And the endometrial expression of nociceptin may be monitored dynamically to track the development of adenomyosis.  相似文献

Objective: To study the effect of TP chemotherapy regimen combined with Kanglaite in the treatment of cervical carcinoma and its effect on the serum level of CYFRA21-1 and CK19. Methods: 74 patients with cervical carcinoma were admitted in our hospital from October 2015 to May 2017 and were randomly divided into observation group and control group according to random digital table method, 60 cases in each group. The control group was treated with TP chemotherapy and the observation group was treated with TP chemotherapy regimen combined with Kanglaite. The clinical efficacy, the quality of life, serum CYFRA21-1 and CK19 levels and adverse reactions were compared between the two groups before and after treatment. Results: After treatment, the clinical effective rate(83.78%) in the observation group was significantly higher than that of control group (62.16%), u=4.390,P=0.036, with statistical significance(P<0.05). The KPS scores of the two groups were significantly decreased, and the KPS scores in the observation group were significantly higher than those in the control group(u=6.538,P=0.000). After treatment, the serum levels of CYFRA21-1 and CK19 were significantly decreased in both groups, and the levels in the observation group were significantly lower than those in the control group (u = 6.659, 8.647, P=0.000). The adverse reactions were mainly myelosuppression, gastrointestinal reaction and phlebitis. The incidences of myelosuppression and gastrointestinal reaction of grade III-IV in the observation group were significantly lower than that in the control group, while the incidence of phlebitis was significantly higher in the observation group than in the control group ((u = 2.160, 2.000, 4.16, P = 0.030, 0.045, 0.041, respectively). Conclusion: The effect of TP chemotherapy regimen combined with Kanglaite on cervical carcinoma is significantly. It can effectively reduce serum CYFRA21-1 and CK19 levels, improve the quality of life and safety, which is worth popularizing in clinical application.  相似文献

To observe the relationship between TCM Syndrome-type and insulin resistance (ISR) in coronary heart disease (CHD). Methods: Fifty patients were divided into 3 groups according to the Syndrome Differentiation-typing in TCM, the Heart blood stasis (HBS) Syndrome group, the Phlegm-Turbid stagnation (PTS) Syndrome group and both Qi-Yin Deficiency (QYD) Syndrome group. The fasting blood glucose (FBG), fasting blood insulin (Ins), insulin antibody (IAA), islet cell antibody (ICA), glutamic acid decarboxylase antibody (GAD-Ab) and related blood lipid parameters in patients were determined and insulin sensitive index (ISI) was calculated simultaneously. Then the above-mentioned data were compared with those determined in 20 healthy control subjects. Results: The levels of FBG and Ins in CHD group were higher than those in the healthy control group significantly (P<0.05), but ISI level was obviously lower (P<0.01). Moreover, the positive percentage of IAA (40%) was higher in CHD group than that in the control group (5%) significantly (P<0.01). Comparison between the 3 TCM Syndrome-type groups and the control group showed that ISI level in HBS and PTS group was obviously lower than that in the control and the QYD (P<0.05) respectively, and the IAA positive percentage in the former 2 groups (50%, 47.37%) was higher than that in the latter two (5%,P<0.01 and 15.38%, P<0.05) markedly. While Ins level increased only in the HBS group (P<0.05). Besides, patients of HBS and PTS Syndrome were accompanied by lipid metabolic disturbance. Conclusion: ISR presents in part of CHD patients particularly in those with HBS and PTS Syndrome, which was partly due to the existence of serum IAA in patients.  相似文献

目的 探讨雌激素对缺氧/复氧诱导新生大鼠皮质神经元损伤的保护作用及其机制.方法 将培养7 d的大鼠皮质神经元随机分为三组,A组为正常对照组,B组采用缺氧/复氧(H/R),C组采用雌激素(17βE2)预处理加H/R处理,各组在H/R后0、1、3、6、12、24 h各时间点,以TUNEL法比较各组凋亡细胞,免疫组化方法比较各组高迁移率族蛋白B1(HMGB1)、核因子-κB(NF-κB)表达.结果 ①B组凋亡神经元明显多于正常对照组(H/R后3~24 h),C组凋亡细胞数目显著少于B组,三组比较差异有统计学意义.②B组HMGB1、NF-κB的表达较正常对照组明显增加,C组HMGB1、NF-κB表达较B组明显减少.结论 雌激素可使H/R后神经元HMGB1、NF-κB表达降低,抑制神经元凋亡,提高其存活率,这可能是其脑保护作用的机制之一. Abstract: Objective To explore the protective effects of estrogen on injured neurons induced by H/R and the mechanisms of that. Methods The cortical neurons cultured for 7 days were randomly divided into group A (normal control group), group B (H/R alone), group C (pretreatment with Estrogen -17βE2 and H/R). Then the apoptotic neurons were count by TUNEL, and the expression of HMGB1,NF- κB was observed by immunocytochemical technique, on each time point after reoxygenation 0,1,3,6,12,24 h of each group. Results ①The number of apoptotic neurons in group B was more than that in control group after H/R 3-24 h, and was less in group C than in group B. ②Compared with group A, the expression of HMGB1 and NF-κB was higher in group B,and was lower in group C than in group B. Conclusions Estrogen could decrease the expression of HMGB1 and NF-κB to inhibit neuronal apoptosis after H/R, which may be one of the mechanisms in which estrogen exerts its neuro-protective effect.  相似文献

目的 研究螺内酯对大鼠心肌梗死后血清转化生长因子-β1(TGF-β1)的影响.方法 结扎SD大鼠冠状动脉前降支,建立心肌梗死模型,将术后存活24 h大鼠随机分为两组:螺内酯(MIS)组给予螺内酯20 mg/(kg·d)溶于2 ml生理盐水灌胃,连续4周;心肌梗死(MI)对照组给予等体积的生理盐水,穿针但不结扎前降支的作为假手术组,给予等体积的生理盐水,4周末行血流动力学测定,同时断尾取血测血清TGF-β1含量.结果 与假手术组相比,4周末AMI组血清TGF-β1含量显著升高(P<0.01),左室舒张末压(LVEDP)显著增加(P<0.01),左室内压最大收缩速率(+dp/dtmax)显著下降(P<0.01).与AMI组相比,4周末螺内酯组血清TGF-β1含量显著下降(P<0.01),LVEDP显著下降(P<0.01),而+dp/dtmax显著增加(P<0.01).结论 螺内酯可能通过抑制TGF-β1产生而改善心功能,抑制心肌纤维化. Abstract: Objective To observe the effects of aldactone on blood serum TGF - β1 in acute myocardial infarction rats. Methods Rats surviving 24 h after AMI induced by left anterior descending branch ligation were randomized to aldactone and control group compared with sham - operated group. The aldactone group was given aldactone 20 mg/( kg · d) , the MI control group and sham -operated group were given an equal volume of saline, continued for 4 weeks. At the end of 4th week,the hemodynamics change was detected, then tails to take blood and measure the serum TGF - β1.Results Compared with the sham - operated rats,the serum TGF - β1 level and the LVEDP of the AMI rats were significantly elevated at the end of the 4th week ( P <0.01 ) , while the + dp/dtmax were significantly reduced ( P <0.01 ) compared with the AMI control group, the serum TGF - β1 level and the LVEDP of the aldactone group rats decrease significantly at the end of the 4th week (P < 0.01 ) ,while the + dp/dtmax were significantly elevated ( P < 0.01 ). The difference had statistical significance. Conclusions Aldactone may improve the cardiac fibrosis and inhibit myocardial fibrosis by inhibiting the production of the TGF - β1.  相似文献

目的 研究凝血酶敏感蛋白-1(TSP-1)对人肝癌细胞株HCCLM3凋亡的体外诱导作用及机制.方法 采用流式细胞术检测TSP-1及其受体CD36、CD47诱导HCCLM3的细胞凋亡率,应用电镜分析作用后的形态变化,逆转录聚合酶链反应(RT-PCR)分析作用后HCCLM3细胞Caspase-3 mRNA表达的变化.结果 TSP-1组凋亡率[(12.44±0.72)%]显著高于对照组[(4.31±0.29)%]和CD47阻断组[(4.99±0.12)%],P<0.01.CD36阻断组[(9.99±0.57)%]高于对照组或CD47阻断组,低于TSP-1组(P<0.01).电镜观察:对照组和C1D47阻断组细胞生长旺盛.TSP-1组和CD36阻断组细胞凋亡率增加,细胞呈现各种凋亡的表现.TSP-1组Caspase-3 mRNA的表达TSP-1组(0.652±0.024)和CD36阻断组(0.615±0.020)显著高于对照组(0.398±0.033)和CD47阻断组(0.432±0.019),P<0.01.结论 TSP-1可诱导人肝癌细胞株HCCLM3的凋亡,TSP-1与受体CD47结合后上调Caspase-3的表达可能是作用途径之一. Abstract: Objective To study the effect and mechanism of thrombospondin- 1 on the apoptosis of human hepatocarcinoma cell HCCLM3. Methods Detecting the role of TSP-1, CD36, CD47 on apoptosis rate of HCCLM3 by the flow cytometry and the change of cell ultractructure by transmission electron microscope,then detecting the influence of TSP-1, CD36, CD47 on the expression of Caspase-3 in HCCLM3. Results The apoptosis rate was respectively significantly higher in TSP-1 group [(12. 44 ±0. 72)%] than that in control group [(4. 31 ±0. 29)%] or CD47-blocked group[(4. 99 ±0. 12)%],P < 0. 01. The apoptosis rate was was respectively significantly higher in CD36 -blocked group [(9. 99 ±0. 57)%]than that in control group and CD47 -blocked group,was lower than in the TSP-1 group(P <0. 01). The morphological changes were: grow vigorously and many divided nuleus in control group and CD47- blocked group. Apoptosis cell increased in TSP- 1 group and CD36 -blocked group: less cellular module in cytoplasm, many nucleus pieces and concentrate. The expression of Caspase-3 were higher in TSP-1 group (0. 652 ± 0. 024) and CD36-blocked group (0. 615 ± 0. 020) than that in control group (0. 398 ±0. 033)and CD47-blocked group (0. 432 ± 0. 019) (P < 0. 01). Conclusions TSP-1 can induce apoptosis of HCCLM3 cell line through combined with CD47 which can increase the expression Caspase -3.  相似文献

目的 观察阻塞性睡眠呼吸暂停低通气综合征(OSAHS)患儿行腺样体扁桃体切除手术前、后血清中炎性因子的变化,并评估手术的影响.方法 将45例行扁桃体腺样体切除术的OSAHS患儿作为治疗组,另选45例性别和年龄等与治疗组相匹配的健康儿童为对照组.用超敏ELISA法检测血清α-肿瘤坏死因子(TNF-α)和白介素-6(IL-6),用胶乳增强免疫比浊法检测血清中超敏C-反应蛋白(hs-CRP).治疗组手术治疗后6个月复查上述指标.结果 治疗组术前血清TNF-α、IL-6和hs-CRP水平均高于对照组(P<0.01);经手术治疗后1年,上述指标均低于术前水平(P<0.01);TNF-α、IL-6和hs-CRP水平与AHI呈正相关,与LSaO2呈负相关.结论 手术治疗可有效逆转OSAHS患儿血中TNF-α、IL-6和Hs-CRP的水平,可以将其作为手术治疗效果的评价指标. Abstract: Objective To observe the changes of circulating tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6) and high-sensitivity C-reactive protein (hs-CRP) levels in children with obstructive sleep apnea-hypopnea syndrome (OSAHS) before and after operation,and to assess the effect of surgical treatment on the levels of these inflammatory factors. Methods Forty-five children with OSAHS underwent adenoidectomy and tonsillectomy were selected as OSAHS group and matched for age and sex to 45 health children control group. Serum levels of TNF-α and IL-6 were detected by high sensitivity ELISA. The hs-CRP was measured by latex-enhanced turbidometry. The serum levels of these factors in OSAHS group before operation were compared with those of the control group and OSAHS group 6 months after operation. Results Serum levels of TNF-α, IL-6, and hs-CRP in OSAHS group were higher than those in the control group (P<0.01), and the levels in OSAHS group 6 months after operation were significantly lower than those before operation (P<0.01). All the serum parameters were correlated positively with AHI, and negatively with LSaO2. Conclusions Operative treatment can effectively correct circulating serum levels of TNF-α,IL-6 and hs-CRP in children with OSAHS. The changes of TNF-α,IL-6 and hs-CRP levels can be a valuable measure in treatment follow-up.  相似文献

目的 构建含有胰腺-十二指肠同源盒-1(Pdx-1)的真核表达载体,并研究其在大鼠骨髓间充质干细胞(MSCs)中的表达情况.方法 克隆Pdx-1基因,构建含Pdx-1的真核表达载体pEGFP-Pdx-1及pcDNA3.1-Pdx-1,转染MSCs,倒置荧光显微镜及逆转录聚合酶链反应(RT-PCR)检测MSCs中Pdx-1的表达.结果 测序结果 表明克隆的Pdx-1基因序列正确,构建的含Pdx-1的真核表达载体能有效的转染MSCs,并检测到该基因mRNA的表达.结论 完成Pdx-1基因克隆,成功构建含有Pdx-1基因真核表达载体,并在转化株中检测到该基因的表达,为糖尿病的细胞替代及基因治疗提供基础. Abstract: Objective To construct eukaryotic expression vector containing the pancreatic-duodenum homeobox-1(Pdx-1)and to study the expression in rat bone marrow-derived mesenchymal stem cells (MSCs). Methods Cloned Pdx-1 gene and constructed containing Pdx-1 eukaryotic expression vector pEGFP-Pdx-1 and pcDNA3.1-Pdx-1, transfected MSCs, detected the Pdx-1 expression in MSCs by invert fluorescence microscope and RT-PCR.Results Sequencing results showed that the cloned Pdx-1 gene sequence was correct, the eukaryotic expression vector containing Pdx-1 could effectively transfected MSCs and detected the gene expression at mRNA level.Conclusions The completion of Pdx-1 gene is cloned successfully and constructed containing Pdx-1 gene eukaryotic expression vector, detected the expression of the gene in the stable transfected MSCs,making preparations for the cell replacement and gene therapy of diabetes.  相似文献

目的 检测不同宫颈病变中Bmi-1的表达,探讨其与宫颈癌各临床病理因素的关系及意义.方法 以宫颈癌、慢性宫颈炎组织为材料,采用免疫组化染色法检测Bmi-1蛋白表达水平,并用原位杂交法检测Bmi-1 mRNA表达水平.结果 免疫组化结果显示,Bmi-1蛋白在宫颈癌组织中的表达明显高于慢性宫颈炎组织(P<0.05).原位杂交结果显示,宫颈癌组织的Bmi-1 mRNA表达水平明显高于慢性宫颈炎组织(P<0.05).Bmi-1的表达与宫颈癌肿瘤分级、临床分期等密切相关(P<0.05),而与患者的年龄、病理分型等无关(P>0.05).结论 Bmi-1在宫颈癌组织的表达状态与其发生发展关系密切,可作为反映宫颈癌生物学行为的有效指标之一. Abstract: Objective To investigate the expression and its clinicopathologic significance of Bmi-1 in different cervical diseases. Methods Immunocytochemistry was used to examined Bmi-1 protein expression in cervical carcinoma and chronic cervicitis tissue. The mRNA expression of Bmi-1 was examined by in situ hybridzation. Results Immunocytochemistry demonstrated that the levels of Bmi-1 protein in cervical carcinoma samples were significantly higher than those in chronic cervicitis tissues(P<0.05).In situ hybridration results shows that the levels of Bmi-1 mRNA in cervical carcinoma samples were significantly higher than those in chronic cervicitis tissues(P<0.05).Expression of Bmi-1 was correlated with tumor grading and clinical stage(P<0.05). On the other hand, it was not correlated with age or pathological type(P>0.05). Conclusions Overexpression of Bmi-1 in cervical carcinoma enhances carcinogenesis and invasion. It's possible that Bmi-1 serves as a good prognostic factor to indicate biologic behavior of cervical carcinoma.  相似文献

目的 探讨慢性阻塞性肺疾病(COPD)患者血清瘦素水平变化及其与炎性反应的关系,了解瘦素在COPD患者炎症反应过程中的作用及其意义.方法 选择COPD患者共58例,COPD急性加重期(Ⅰ组)30例,COPD缓解期(Ⅱ组)28例,正常对照组28例.测定和计算各组的多项营养指标,包括身高、体重、体重指数(BMI)、理想体重百分比(NM%)、体脂百分比(fat%).用酶联免疫吸附试验法(ELISA)测定58例COPD患者和28例正常人的血清瘦素、肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)、超敏C-反应蛋白(hs-CRP)水平及肺功能,分析瘦素与各项参数的相关性.结果 ①COPD患者的各营养指标:BMI、NM%、fat%均显著低于正常对照组(P<0.01).②COPD I组血清瘦素、TNF-α、IL-8、hs-CRP均显著高于正常组与COPDⅡ组(P<0.01);COPDⅡ组血清瘦素低于正常组(P<0.01),TNF-α、IL-8、hs -CRP高于对照组(P<0.01);③COPD患者的瘦素含量与BMI、NM%、fat%、TNF-α、IL-8、hs -CRP水平显著正相关.结论 血清瘦素在COPD急性加重期明显高于缓解期,并与营养参数、TNF-α、IL-8、hs -CRP呈显著正相关,提示瘦素可能与COPD的疾病本身和全身炎症反应有关,可作为COPD急性加重期的炎症标志物. Abstract: Objective To investigate the expression and significance of serum leptin in patients with chronic obstructive pulmonary disease(COPD). Methods Thirty COPD patients in acute exacerbations phase (group I), 28 COPD patients in stationary phase(group Ⅱ) and 28 healthy controls were studied. Body height, body mass,body mass index (BMI), percentage of normal body mass (NM%), percentage of body fat(fat%) were determined.The serum leptin、TNF-α、IL-8、hs-CRP levels were examined by ELISA. The correlation between leptin and other parameters was analysed. Results ①BMI,NM% fat% in COPD patients significantly lower than those in healthy controls (P<0.01). ②Serum leptin、TNF-α、IL-8、hs-CRP level was significantly higher in group I than that in healthy controls and in group II (P<0.01); Serum leptin level was significantly lower in group II than that in healthy controls (P<0.01); Serum TNF-α、IL-8、hs-CRP level was higher in healthy controls (P<0.01). ③The leptin level was associated positively with BMI,NM%, fat%. TNF-α、IL-8、hs-CRP level of COPD. Conclusions The leptin level of COPD in acute is higher than that in stationary, and the levels were correlated to nutritional parameters,TNF-α,IL-8,hs-CRP.The result suggests that leptin may play a role in systemic inflamination of COPD, the leptin is an inflaming mark of the actue exacerbation phase in patients with COPD.  相似文献

目的 初步探讨特应性皮炎(AD)小鼠中Th1/Th2 类细胞因子失衡对脾淋巴细胞神经生长因子(NGF) mRNA 表达的影响.方法 用卵清蛋白经皮肤致敏建立小鼠AD模型, ELISA测定血清总IgE水平.分离脾淋巴细胞并培养.逆转录-聚合酶链反应(RT-PCR)半定量法测定细胞在刀豆素A(ConA)刺激下表达NGF mRNA的基础水平及γ-干扰素(IFN-γ)和白细胞介素-4(IL -4)干预后NGF mRNA表达改变.结果 ①AD小鼠脾淋巴细胞在基础状态下具有表达NGF mRNA的功能,在12、24、36、48 h时基础NGF mRNA表达量随着时间的延长逐渐升高,IL-4组(50 μg/L)随着时间的延长基础NGF mRNA表达量逐渐升高,并高于同时间空白对照组(P均<0.01). 相反, IFN-γ组(25 μg/L)的NGF mRNA水平随时间逐渐下降,并均低于同时间空白对照组(P均<0.01).②干预24 h时,随着IL-4干预浓度的增加,NGF mRNA的表达水平亦升高,并且分别与前一梯度低浓度干预组比较差异有统计学意义(P均<0.01),随着IFN-γ干预浓度的增加, NGF mRNA的表达水平却逐渐减低,分别比前一梯度低浓度干预组有显著降低(P均<0.01).结论 在特应性皮炎中Th2类细胞因子IL-4可上调淋巴细胞NGF mRNA表达,Th1类细胞因子IFN-γ可下调淋巴细胞NGF mRNA表达,两者均呈时间和浓度依赖性,Th1 / Th2类细胞因子免疫失衡可能通过调控NGF mRNA 表达间接促进其诱导神经源性炎症. Abstract: Objective To explore the effect of Th1 / Th2 cytokines on the expression of nerve growth factor (NGF) in splenic lymphocytes in atopic dermatitis mice.Methods Four AD mice were sensitized and challenged with ovalbumin to establish AD model, and serum IgE level was measured by ELISA.The mouse splenic lymphocytes were isolated and cultured with ConA. The expressions of NGF mRNA were detected by RT-PCR, and were observed after the lymphocytes were exogenously added with interferon -γ(IFN-γ) or interleukin-4 (IL-4). Results ①The lymphocytes of the AD model stimulated by ConA in vitro expressed NGF mRNA in a time-dependent manner. After the lymphocytes had been cultured with IL-4 for 12 h, 24 h, 36 h, and 48 h, 50μg/L IL-4 upregulated the expressions of NGF mRNA in a time-dependent manner and all the NGF mRNA expressions were significantly higher than the basal values at the same time (P all<0.01), however,25μg /L IFN-γ downregulated the expressions of NGF mRNA in a time-dependent manner and all the NGF mRNA expressions were significantly lower than the basal values at the same time (P all<0.01).②The lymphocytes of the AD model stimulated by ConA in vitro also expressed NGF mRNA in a dose-dependent manner.After 0,10, 25,50, and 100 μg /L IL-4 had been added for 24 h, IL-4 upregulated the expressions of NGF mRNA in a dose-dependent manner and the NGF mRNA expressions were all significantly higher than the values of the lower dose IL -4 (P all <0.01), however,after 0, 1, 10, 25,and 50μg /L IFN-γ had been added for 24 h, IFN-γ downregulated the expressions of NGFmRNA in a dose-dependent mannerand all the NGF mRNA expressions were significantly lower than the values of the lower IFN-γ dose (P all<0.01).Conclusions In the splenic lymphocytes of atopic dermatitis mice, IL -4, one of the Th2 cytokines, can upregulate the expressions of NGF, IFN-γ, one of the Th1 cytokines, can downregulate the expressions of NGF both in a time-dependent manner and in a dose-dependent manner. Th1 /Th2 cytokine immune imbalance may indirectly induce the skin neurogenic inflammation by regulating the NGF mRNA expression.  相似文献

目的 研究食管鳞状细胞癌(ESCC)中缺氧诱导因子-1α(HIF-1α)、6-氧-甲基鸟嘌呤-DNA甲基转移酶(MGMT)蛋白的表达及其与食管癌病理特征的关系.方法 采用免疫组织化学(SP)法检测58例食管鳞癌组织中HIF-1α、MGMT蛋白的表达,并分析其与临床病理各因素之间的关系.结果 58例患者的ESCC组织中HIF-1α、MGMT蛋白均有表达,阳性表达率分别为51.4%、39.6%,MGMT 蛋白的表达与食管鳞癌的分化程度密切相关(P<0.05),而与性别、年龄、分期及淋巴结转移无关.HIF-1α蛋白的表达与性别、年龄、分期、分化程度以及淋巴结转移均无关(P均>0.05).HIF-1α与MGMT 蛋白表达无明显相关关系(P>0.05).MGMT阳性表达患者生存时间(OS)较阴性组明显延长(P<0.05),而HIF-1α蛋白表达与OS无明显相关(P>0.05).结论 HIF-1α蛋白在食管鳞癌组织中呈高表达,MGMT与其分化程度相关,两者检测对判断预后有重要意义. Abstract: Objective To investigate the expression of hypoxia-inducible factor-1α (HIF-1α) and O~6-methylguanine DNA methytransferase(MGMT) in esophageal squamous cell carcinoma(ESCC) tissue and its relation with the clinicopathalogy features. Methods The expression of HIF-1α, MGMT in 58 cases of ESCC were detected by immnohistochemical staining. Results The positive rates of HIF-1α,MGMT in ESCC tissue were 51.4%,39.6%, respectively. The positive expression rates of HIF-1α and MGMT were not correlated with the degree of sex,age,clinical stage and differentiation(P>0.05), but MGMT expression was correlated with differentiation and overall survival time (P<0.05). The expression of HIF-1α was not correlated with that of MGMT (P>0.05), and HIF-1α was not correlated with overall survival time. Conclusions HIF-1α, MGMT play important roles in esophageal carcinoma,MGMT was correlated with the differentiation of esophageal carcinoma, respectively, which may be used as important indexes to evaluate the prognosis of esophageal carcinoma.  相似文献

目的 应用RNA干扰(RNA interference)技术,抑制沉默小凹蛋白-1(caveolin-1)基因的表达,观察小凹蛋白-1在骨髓间充质干细胞(MSCs)分化为神经元样细胞的作用.方法 构建大鼠caveolin-1 siRNA(small interfering RNA),然后传染大鼠MSCs,诱导分化并在倒置显微镜下进行形态学观察,用MTT比色法检测细胞存活率,采用免疫细胞化学染色法检测Nestin、 NSE(神经元标记物) 、GFAP(神经胶质细胞标记物)的表达,设诱导未传染组和传染对照组作为对照研究.结果 用caveolin-1 siRNA传染MSCs后,caveolin-1基因表达消失,传染后24 h和3 d MSCs细胞存活率下降,与诱导未传染组和传染对照组比较差异有统计学意义(P<0.01),传染caveolin-1 siRNA的MSCs向神经元样细胞分化效率显著提高,诱导24 h后NSE阳性细胞率为(75.5±2.5)%,6 d后达(81.6±3.5)%,且未见GFAP的表达,与诱导未传染组和传染对照组比较差异有统计学意义(P<0.01).结论 caveolin-1 siRNA抑制caveolin-1基因的表达,可提高MSCs向神经元样细胞的分化效率. Abstract: Objective To investigate the role of caveolin-1 in bone marrow mesenchymal stem cells(MSCs) differentiating into neuron-like cells by transfecting MSCs with small interfering RNA(siRNA). Methods Construct caveolin-1 siRNA and then transfect into MSCs cultured in vitro. Cell growth was observed by an inverted phase contrast microscope. The survival ratio of MSCs was determined by MTT before transfection,24 hour, 3 days after transfection,respectively. MSCs was induced to differentiate into neuron-like cells by-ME. The neural cell specific marker Nestin,NSE,GFAP were determined by immunocytochemistry stain technique. Results Expresion of caveolin-1 vanished by tranfection of caveolin-1 siRNA,survival ratio of MSCs by transfection decreased and there was significant differences between no-transfection and transfection-controlling group after 24 hours and 3 days of transfection(P<0.01).The differentiation efficiency of MSCs transfected by caveolin-1 siRNA into neuron-like cells and the expression of NSE were increased significantly than no-transfection and transfection-controlling group, The percentage of positive NSE was(75.5±2.5)% after 24 hours and(81.6±3.5)% after 6 days, there was no expression of GFAP. There was significant differences between no-transfection and transfection-controlling group(P<0.01). Conclusions The differentiation efficiency of MSCs increased by suppressing caveolin-1 expression.  相似文献
