目的 :观察消痰散结方对体外培养的胃癌细胞粘附分子ICAM -1表达的影响 ,初步探讨消痰散结方抑制胃癌细胞转移的作用环节。方法 :制备消痰散结药物血清 ,体外培养MKN -4 5人胃癌细胞 ,拟消痰散结方药物血清干预培养细胞 ,采用免疫组化ABC法检测胃癌组织中I CAM -1的表达。结果 :中药组胃癌细胞中ICAM -1表达水平明显低于空白对照组。结论 :消痰散结方抑制胃癌细胞转移的环节可能和影响粘附分子的表达 ,从而影响细胞的粘附性机能有关  相似文献

目的:探讨中药消痰散结方对实验性裸鼠人胃癌血管生成的影响.方法:建立裸鼠人胃癌原位种植模型,将动物随机分为空白对照组、中药组和联合组(n=10),中药组予消痰散结方0.2 mL灌胃,联合组同时予5-FU溶液0.2 mL腹腔注射.第11周结束实验,动物行超声探查,利用TOMTEC imaging system对超声图像进行三维重建,计算肿瘤组织的三维超声血管容积指数(Ⅵ),处死动物,取瘤组织,Envision法免疫组化染色CD34,计数微血管密度(MVD).结果:与空白对照组(2.14±0.43 g)相比,中药组(1.12±0.38 g,P<0.05)和联合组(0.77±0.26 g)瘤质量明显减低(P<0.05);联合组比中药组瘤质量明显减低(P<0.05).中药组和联合组的血流较空白对照组明显减少.中药组和联合组Ⅵ和MVD与空白对照组相比.明显减低(Ⅵ:18.87%±3.74%,14.97%±3.61% vs 31.00%±4.06%,P<0.05;MVD:27.3±8.8,25.8±8.6 vs 49.3±15.3,P<0.05).结论:消痰散结方可以明显抑制裸鼠人胃癌原位种植瘤的血管生成.  相似文献

目的 建立兔颈总动脉血管吻合口狭窄模型,并用局部转染小凹蛋白(Caveolin-1)质粒,观察血管吻合口部位Caveolin-1对白细胞介素10(IL-10)和白细胞介素6(IL-6)表达的影响.方法 从南华大学动物中心取成年家兔32只,行颈总动脉血管吻合术,运用脂质体局部转染Caveolin-1质粒,随机分为4组:正常组(假手术组)、手术组、手术+空转染组、手术+转染组.分别于7天及28天取颈总动脉标本,通过HE染色观察内膜增生情况,并测量内膜与中膜面积比值(IA/MA);实时荧光定量PCR检测血管中Caveolin-1和IL-6、IL-10 mRNA的表达,免疫组织化学观察血管组织中IL-6、IL-10蛋白的表达.结果 HE染色显示,手术组及空转染组内膜明显增殖,管壁增厚,管腔缩小;而转染组未见明显内膜增殖及管壁增厚;从血管内膜/中膜面积比值发现,转染组比手术组明显降低;血管吻合手术后,抗炎因子IL-10和促炎因子IL-6都升高.当吻合口部位Caveolin-1基因高表达时,IL-10 mR-NA和蛋白表达都上调,而IL-6 mRNA和蛋白表达都减少.结论 Caveolin-1促使血管吻合口部位炎症因子的平衡向抑炎因子倾斜.  相似文献

目的探讨小鼠心肌梗死后血清细胞因子表达的变化与心肌炎症反应的关系。方法冠状动脉结扎法复制小鼠心肌梗死模型,采用ELISA方法测定结扎后不同时间点血清肿瘤坏死因子α、白细胞介素6和白细胞介素10浓度和心肌组织病理变化。结果心肌梗死后1h,小鼠血清中肿瘤坏死因子α和白细胞介素10浓度分别是82.4±27.6ng/L和47.7±22.3ng/L,与假手术组水平(40.4±9.7ng/L和23.4±4.3ng/L)比较均明显提高,差异有统计学意义(P<0.05),以后呈逐渐下降趋势;血清中白细胞介素6水平在心肌梗死后1h、2h、4h、1d、2d和5d分别为202.7±89.5ng/L、578.9±198.0ng/L、696.8±104.7ng/L、325.1±131.7ng/L、218.5±16.0ng/L和195.8±93.1ng/L,与假手术组水平(54.0±3.0ng/L)比较均有显著增高(P<0.05),病理切片显示梗死心肌内大量的炎性细胞浸润。梗死后第2至4周细胞因子表达下降,梗死区炎症细胞逐渐减少,心肌纤维化。结论肿瘤坏死因子α、白细胞介素6和白细胞介素10三种细胞因子参与了心肌梗死后心肌的炎症反应,在心肌梗死的发生发展过程中起重要作用。  相似文献

测定IDDM患者16例、NIDDM患者20例和正常对照组27例的外周血单个核细胞产生白细胞介素2(IL-2)和白细胞介素6(IL-6)的能力。结果:IDDM患者IL-2水平较正常对照组和NIDDM患者显著降低(p<0.001),IL-6的水平显著升高(P<0.001)。NIDDM患者IL-2和IL-6与正常对照组差异无显著性。提示IDDM患者IL-2和IL-6异常改变导致免疫功能紊乱。  相似文献

目的:比较罗格列酮和辛伐他汀对兔早期动脉粥样硬化白细胞介素(IL)-6、IL-8表达的影响,探讨罗格列酮抗动脉粥样硬化的作用机制. 方法:将36只雄性新西兰兔随机分为对照组、模型组、辛伐他汀组和罗格列酮组.采用皮下注射同型半胱氨酸硫内酯(HTL)及高脂饲养结合的方法建立兔动脉粥样硬化模型,酶联免疫吸附法(ELISA)检测血清中IL-6和IL-8的水平,免疫组化检测兔主动脉粥样硬化组织中IL-6和IL-8的表达. 结果:与对照组相比,其他3组血清及病变组织中IL-6、IL8表达均显著升高;与模型组比较,辛伐他汀和罗格列酮组IL-6、IL-8表达显著降低;罗格列酮组IL-6、IL-8表达较辛伐他汀组更低(P均<0.05). 结论:罗格列酮能通过抑制兔主动脉粥样硬化组织及血循环中IL-6、IL-8等炎症因子的表达,发挥其抗炎作用,拮抗动脉粥样硬化的发生及发展.  相似文献

目的:通过与C反应蛋白(CRP)检测对比,探讨抗炎因子白细胞介素(IL)-10、促炎因子IL-6与急性冠状动脉综合征(ACS)的关系。方法:冠心病患者78例,其中ACS患者40例(ACS组),稳定型心绞痛(SAP组)患者38例,33例胸痛综合征(CPS组)患者作为对照。所有患者均行冠状动脉造影检查。采用ELISA法检测血浆IL-6、IL-10水平,采用免疫比浊法测定CRP值。结果:与SAP组及CPS组相比,ACS组血浆IL-10水平明显降低,IL-6、CRP水平明显升高,差异有统计学意义;SAP组IL-10水平低于CPS组,IL-6、CRP水平高于CPS组,差异无统计学意义;血浆IL-10水平与IL-6及CRP水平呈负相关,IL-6水平与CRP水平正相关。结论:血浆IL-10水平降低和IL-6、CRP值升高对患者ACS的发生有预测价值。抗炎因子和促炎因子水平的失衡是导致斑块不稳定、急性冠状动脉事件发生的重要因素。  相似文献

目的探讨炎性因子白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素-6(interleukin-6,IL-6)对人冠状动脉平滑肌细胞(human coronary artery smooth muscle cell,HCASMC)表达妊娠相关血浆蛋白-A(pregnancy-associated plasma pro-tein-A,PAPP-A)的影响。方法应用20μg/L的IL-1β、10μg/L的IL-6各自刺激HCASMC,共同培养0、2、4、8、243、6 h后收集细胞。应用不同浓度的IL-1β(0、5、20、40μg/L)I、L-6(0、5、10、50μg/L)刺激HCASMC,共同培养6 h后收集细胞。应用实时定量聚合酶链反应的方法检测细胞内PAPP-A基因的表达量。结果在同剂量IL-1βI、L-6刺激下,PAPP-A的表达量在2 h时就开始发生上调,8 h达高峰,而后开始下降;在不同剂量IL-1βI、L-6刺激下,PAPP-A的表达量在实验剂量范围内随着剂量的加大呈上升趋势(IL-1β:r=0.972,P=0.000;IL-6:r=0.941,P=0.000)。结论炎性因子IL-1βI、L-6能促进HCASMC中斑块稳定相关标记物PAPP-A的表达,可能是炎症在急性冠状动脉综合征发生发展中的重要作用机制之一。  相似文献

白细胞介素6(IL-6)是一种多功能的细胞因子,主要在炎症急性期起介导作用,包括激活炎症细胞和淋巴细胞。正常时IL-6在血清中不能测出,但老年时其水平升高,能被测出,其原因可能是由于T细胞免疫调节功能的丧失和绝经期后雌激素低下,导致老年时IL-6基因表达调节丧失。IL-6可能对许多老年疾患的发病起作用,例如淋巴瘤、骨质疏松和AD。本文综述了IL-6的生物学特性及其与老年医学的密切关系。  相似文献

AIM: To determine the underlying mechanisms of action and influence of Xiaotan Sanjie (XTSJ) decoction on gastric cancer stem-like cells (GCSCs).METHODS: The gastric cancer cell line MKN-45 line was selected and sorted by FACS using the cancer stem cell marker CD44; the stemness of these cells was checked in our previous study. In an in vitro study, the expression of Notch-1, Hes1, Vascular endothelial growth factor (VEGF), and Ki-67 in both CD44-positive gastric cancer stem-like cells (GCSCs) and CD44-negative cells was measured by Western blot. The effect of XTSJ serum on cell viability and on the above markers was measured by MTT assay and Western blot, respectively. In an in vivo study, the ability to induce angiogenesis and maintenance of GCSCs in CD44-positive-MKN-45- and CD44-negative-engrafted mice were detected by immunohistochemical staining using markers for CD34 and CD44, respectively. The role of XTSJ decoction in regulating the expression of Notch-1, Hes1, VEGF and Ki-67 was measured by Western blot and real-time polymerase chain reaction.RESULTS: CD44+ GCSCs showed more cell proliferation and VEGF secretion than CD44-negative cells in vitro, which were accompanied by the high expression of Notch-1 and Hes1 and positively associated with tumor growth (GCSCs vs CD44-negative cells, 2.72 ± 0.25 vs 1.46 ± 0.16, P < 0.05) and microvessel density (MVD) (GCSCs vs CD44-negative cells, 8.15 ± 0.42 vs 3.83 ± 0.49, P < 0.001) in vivo. XTSJ decoction inhibited the viability of both cell types in a dose-dependent manner in vitro. Specifically, a significant difference in the medium- (82.87% ± 6.53%) and high-dose XTSJ groups (77.43% ± 7.34%) was detected at 24 h in the CD44+ GCSCs group compared with the saline group (95.42% ± 5.76%) and the low-dose XTSJ group (90.74% ± 6.57%) (P < 0.05). However, the efficacy of XTSJ decoction was reduced in the CD44- groups; significant differences were only detected in the high-dose XTSJ group at 48 h (78.57% ± 6.94%) and 72 h (72.12% ± 7.68%) when compared with the other CD44- groups (P < 0.05). Notably, these differences were highly consistent with the Notch-1, Hes1, VEGF and Ki-67 expression in these cells. Similarly, in vivo, XTSJ decoction inhibited tumor growth in a dose-dependent manner. A significant difference was observed in the medium- (1.76 ± 0.15) and high-dose XTSJ (1.33 ± 0.081) groups compared with the GCSCs control group (2.72 ± 0.25) and the low-dose XTSJ group (2.51 ± 0.25) (P < 0.05). We also detected a remarkable decrease of MVD in the medium- (7.10 ± 0.60) and high-dose XTSJ (5.99 ± 0.47) groups compared with the GCSC control group (8.15 ± 0.42) and the low-dose XTSJ group (8.14 ± 0.46) (P < 0.05). Additionally, CD44 expression was decreased in these groups [medium- (4.43 ± 0.45) and high-dose XTSJ groups (3.56 ± 0.31) vs the GCSC control (5.96 ± 0.46) and low dose XTSJ groups (5.91 ± 0.38)] (P < 0.05). The significant differences in Notch-1, Hes1, VEGF and Ki-67 expression highly mirrored the results of XTSJ decoction in inhibiting tumor growth, MVD and CD44 expression.CONCLUSION: Notch-1 may play an important role in regulating the proliferation of GCSCs; XTSJ decoction could attenuate tumor angiogenesis, at least partially, by inhibiting Notch-1.  相似文献

目的探讨银杏叶提取物(EGB)对实验性结肠炎大鼠肠组织大体形态组织学及炎性指标的影响。方法利用三硝基苯磺酸灌肠制备大鼠实验性结肠炎模型。实验设正常对照组、模型组、5-氨基水杨酸组、EGB组。4周后评价结肠组织大体形态和组织学评分,检测髓过氧化物酶(MPO)活性和IL-6、IL-6 mRNA表达。结果模型组大鼠结肠组织大体形态和组织学评分较对照组显著增加,MPO活性、IL-6、IL-6 mRNA表达水平显著升高;EGB组上述指标较模型组显著下降。结论EGB对大鼠实验性结肠炎有一定治疗作用,其作用机制可能与其降低结肠组织细胞因子IL-6水平有关。  相似文献

目的研究胃癌组织中IL-6及其受体(IL-6R)、增殖细胞核抗原(PCNA)的表达及临床意义。方法运用酶联免疫吸附实验(ELISA)技术半定量检测30例胃癌患者癌组织及其相应癌旁组织中IL-6、IL-6R的水平,免疫组化法(ABC法)检测PCNA的表达。结果胃癌组织中IL-6、IL-6R水平较癌旁组织显著增高;IL-6、IL-6R及PCNA的表达与胃癌临床生物学特征关系密切。结论IL-6、IL-6R和PCNA是反映胃癌细胞恶性程度和浸润转移的重要指标。  相似文献

To elucidate the role of interleukin (IL)-6 and IL-8 in the pathogenesis of gastric ulcer in Helicobacter pylori-positive gastritis, in situ hybridization using digoxigenin-labeled cDNA probes for both cytokines was performed. Immunogold silver staining was added to further improve the sensitivity of this non-radioactive hybridization. The biopsy specimens were taken from eight patients with active gastric ulcer before treatment, in all of whom H. pylori was positive. Macrophages (the putative producers of these cytokines) were determined by immunohistochemistry using anti-CD68 monoclonal antibodies (KP-1). IL-6 mRNA was most abundantly expressed in the epithelium and in the infiltrating cells in tissue adjacent to gastric ulcer. Quantitative analysis disclosed a significant increase in cells positive for IL-6 mRNA near the ulcer margin compared to cells in the surrounding tissue. In contrast, cells positive for IL-8 mRNA were observed in equal proportions and evenly in the epithelium and over the entire layer of the gastric mucosa regardless of the presence of gastric ulcer. The majority of infiltrating cells positive for both IL-6 and IL-8 mRNA were thought to be macrophages because of their morphologic features and their immunohistochemical reactivity to CD68. These findings strongly suggest that IL-6 is overexpressed at the margin of gastric ulcer in H. pylori-positive gastritis. Received Sept. 24, 1997; accepted Mar. 19, 1998  相似文献

采用生物学方法与分子杂交技术观察了人胎肾小球系膜细胞产生与表达白介素-6。结果显示系膜细胞上清液中白介素-6活性明显高于对照组(P<0.01),Northernblot信号扫描分析结果为10μg系胰细胞总RNA中白介素-6mRNA是22.93;30μg是84.44,提示人胎系膜细胞可自然产生白介素-6,并表达白介素-6mRNA。  相似文献

A 47-year-old woman was admitted because of hypermenorrhea. Transvaginal ultrasonography revealed an ovarian tumor and myoma uteri, and total hysterectomy with bilateral salpingo-oophorectomy was performed. Histology revealed signet-ring cell carcinoma in the right ovary. In order to find out the primary site of this tumor, gastroendoscopy was performed after the operation, and showed a IIc lesion in the lower body of the stomach; biopsy specimens showed signet-ring cell carcinoma similar to that in the right ovary. Total gastrectomy revealed that the lesion was an early gastric cancer confined to the mucosa, but there was lymphatic invasion slightly beneath the muscularis mucosa, with regional lymph node metastasis. In the light of a review of the seven cases of early gastric cancer with Krukenberg tumor previously reported, lymphatic metastasis seemed to be the most likely pathway of ovarian metastasis in early gastric cancers.  相似文献

目的 观察急性心肌梗死(AMI)患者人院外周血白细胞(WBC)总数、血浆白细胞介素-6(IL-6)对近期预后的影响。方法 首发AMI患者103例,通过检测WBC、IL-6,以发病后30天内是否发生心血管不良事件(包括心源性死亡、心力衰竭、心源性休克、恶性心律失常)为观察终点。结果 36例发生心血管事件组外周血WBC总数和IL-6显著高于67例无心血管事件组。AMI时外周血WBC总数和IL-6水平为发生心血管事件的预测因素。结论 WBC、IL-6与AMI患者近期预后相关。  相似文献
