The action of Gly-Tyr-NH2,(GY-NH2) and Gly-Tyr-LYS(GYK) on ^125I-LH binding,cAMP accumulation and progesterone production was investigated.Incubation of rat luteal cells for 2.5h with GH-NH2 and GYK at dosage of 0.2mmol/L caused a significant inhibition of basal and gonadotropin-stimulated steroidogenesis.GYNH2 and GYK were also foumnd to reduce cAMP formation in response to hCG.The activity of adenylate cyclase of luteal cells was inhibitd by 0.2mmol/LGY-NH2 and GYK.GY-NH2 and GYK at a concentration of 0.2mmol/L were not found to have an inhibitory effect on 8Br-cAMP-stimulated progesterone production.GY-NH2 and GYK did not affect ^125I-LH binding to LH receptors on the luteal cellsurface.These results suggest that GY-NH2 and GYK inhibit steroidogenesis at the step of gonadotropin-stimulated c AMP for mation in luteal cells.Adenylate cyclase in luteal cells was also inhibited.  相似文献

The photodynamic activity of berberine on astomach cancer cell line was studied and revealedthat the cell growth activity was not inhibited whe-ther the drug was added to the culture medium alone(the final concentration of drug does not exceed10μg/ml of medium) or the culture was illuminatedby means of a black light lamp (wave length 320-450nm,20W) alone.If both are combined,and themedium is changed but without the drug afterillumination,are could be observed a strong in-hibition of cell growth.The inhibitory effect isdirectly proportional to the concentration of drug.  相似文献

This paper presented the effects of systemic fluconazole therapy via intravenous (IV) and oral (PO) administrations on the adhesion of Candida albicans (C. albicans) to the huccal epithelial ceils (BEC) from five treated patients with three candidosis, one mucornlycosis and one sporotrichosis and at the same time,an analysis of the cell surface proteins involving candidal adherent receptor in the BEC of the patients in the course of 7 days were exposed to ^3H-leucine radiolabaled C. atbicans for in vitro eandidal adherent assay,and the BEC from first intake day and the last intake day of the patients were extracted by dithiothreitol(DTT)-iodoacetamide treatment for SDS-PAGE. These results indicate that the systemic iluconazole therapy resuks in the inhibitory effect of candldal adhesion to BEC of treated patients to prevent them from oral candidosis for a prolonged time, which is based on the absent surface protein (35KDa) of the BEC.  相似文献

Objective:To understand the mechanism of the hypertherjia and harringtonine in purging of leukemia cells In vitro.telomerase activity of HL-60 cells treated by hyperthermia(42℃ for one hour) and different concentrations of Harringtonine were investigated .Methods Using telomeric Repeats Amplification Protocol(TRAP)and ELISA techniques to analyze the telomerase activity of HL-60 cells,Results:Our results showed that harringtonine inhibited the telomerase activity of HL-60 cells in a dosage related manner,Moreover,the telomerase activity of HL-60 cells was significantly decreased after the hyperthermia treatment as compared with untreated cells.Conclusion:The effect of the hyperthermia and Harringtonine on purging leukemia cellsIn vitro may be mediated by down regulation of telomerase activity of tumor cells.  相似文献

Using continuous cultures of ECC_2/HNstrain of P.falciparum,we found that Qing-haosu(QHS)at a concentration of 1×10~(-8)M hadno appreciable effect on the growth of the para-sites,while QHS at 1×10~(-8)M markedly inhibit-ed their growth and multiplication.This indi-cated the possibility of QHS exerting direct ef-fect on the erythrocytic stage of P.falciparumin vitro.Electron microscopy showed morpho-logical abnormalities appearing in the asexualforms of P.falciparum following exposure toQHS at 1×10~(-7)M or 5×10~(-7)M for 2 hours or 1hour respectively.The main pathological chan-ges were:(1)injuries to the membrane struc-ture,such as widening of the intermembranousspace between the nuclear membrane and thelimiting membrane,swollen endoplasmic re-ticulum,etc.;(2)formation of autophagic va-cuoles containing no pigment.For comparisonwith QHS,the action of CQ on the ultrastruc-ture of the parasite was also investigated anddiscussed.  相似文献

Objective To establish a smoke-induced chronic bronchitis rat model and evaluate the patho-logical change semi-quantitatively, and study the characteristics of the inflammatory cells in the bronchoalveolar lav-age fluid (BALF) in various stages. Methods Chronic bronchitis sequential rat model was established by passively inhaling smoke mixture. Experiments were performed in 30 young male Sprague-Dawley rats, which comprised 5 groups in random, i.e.,4 chronic bronchitis model groups and 1 control group. After stained with hematoxylin and eosin, the specimens were studied by semi-quantitative method to evaluate the morphologic changes in various stages. Meanwhile, the inflammatory cells of the BALF and the activity of myeloperoxidase (MPO) of lung tissue were analysed. Results During the process of the chronic bronchitis, the pathologic score was increasing as time went on, and the typical morphologic changes of chronic bronchitis emerged in the group 7 weeks. 用黄体生成素释放激素(LHRH)和三种对离体假孕大鼠黄体细胞分泌孕酮有较强抑制作用的LHRH类似物(LHRH-A),观察了它们对离体培养的人早孕黄体细胞分泌孕酮的影响。实验发现LHRH和LHRH-A对人早孕黄体细胞分泌孕酮有微弱的直接抑制作用。抑制作用的大小与外加人绒毛膜促性腺激素(hCG)浓度有关。低浓度时(hCG=1IU/ml),抑制作用相对较明显,最大抑制率为19.19%(P=0.05)。但此抑制作用可部分被高浓度的(hCG=50IU/ml)所取消。  相似文献

目的:观察血小板活化因子(PAF)对离体培养的大鼠卵巢黄体细胞孕酮分泌的影响,初步探讨其可能的作用机制.方法:原代细胞培养和放免技术.结果:PAF、hCG、Forscolin、PMA组与对照组相比差异均有统计学意义(P<0.05),PAF与hCG、Forscolin、PMA交互作用均有显著性(P<0.01).结论:PAF能明显促进离体培养的大鼠卵巢黄体细胞孕酮的分泌,并且受hCG及PKA、PKC信号转导通路的调节.  相似文献

目的:探索大鼠骨髓间充质干细胞(mesenchymal stem cell,MSCs)在体外定向分化为神经元样细胞的方法并研究其死亡的影响因素.方法:分离纯化大鼠骨髓间充质干细胞并用2-巯基乙醇诱导分化为神经元样细胞,使用免疫组化方法鉴定其神经元特异性巢蛋白(Nestin)的表达,通过MTT、免疫组化及形态学方法观察丝裂原活化蛋白激酶激酶1/2(mitogen-activated protein kinase kinase 1/2,MEK1/2)阻断剂U0126及缩胆囊肽(cholecystokinin,CCK)对细胞转化及活性的影响.结果:诱导后MSCs分化为具有典型的神经元形态的细胞,免疫组化实验显示Nestin呈强阳性反应,同时免疫组化及MTT方法显示U0126可以显著提高细胞Nestin的表达强度和神经元样细胞的活性,CCK虽可轻度增强神经元样细胞的活性,且CCK可延长已分化神经元的存活时间,但其作用不具统计学意义.结论:大鼠MSCs在体外可定向诱导分化为神经元样细胞,且通过阻断MEK1/2信号通路可以提高其阳性分化率和细胞活性,提示大鼠MSCs诱导分化的神经元样细胞的死亡机制可能涉及MEK1/2信号通路.CCK可能具有对已分化神经元的保护作用.  相似文献

作者采用放射免疫分析法,观察酪氨酸类似物酪氨酰肼对离体培养大鼠黄体细胞3β-羟-甾体脱氨酶(3β-HSD)活性的影响,同时还观察了酪氨酰肼在拮抗hCG诱导孕酮生成作用中对3β-HSD活性的影响。结果显示:(1)0.2mmol/L酪氨酰肼明显抑制大鼠黄体细胞3β-HSD活性。(2)底物浓度较低时,酪氨酰肼对3β-HSD活性具有显著增强作用,随着底物浓度的增加,它的作用渐弱且转为抑制。(3)酪氨酰肼可明显抑制hCG诱导的3β-HSD活性。  相似文献

目的探讨H2S对大鼠肝星形细胞(HSC)增殖和氧应激的影响。方法用铁超载的方法复制HSC增殖模型,通过绘制生长曲线以及CCK-8细胞计数法检测不同浓度的H2S对500μmol/L次氮基三乙酸铁(FeNTA)作用下的HSC增殖的影响。通过不同浓度的H2S作用于500μmol/LFeNTA培养的HSC,检测6h、12h、24h细胞上清液和裂解液中H2S、SOD活力、MDA含量,观察H2S对HSC氧应激反应的影响。结果同对照组比较,一定浓度的H2S对HSC增殖有抑制作用。在FeNTA诱导的氧应激反应中,一定浓度范围的H2S可有效提高SOD的活性和降低MDA含量。结论H2S对HSC增殖有较强抑制作用,对FeNTA诱导的氧应激具有保护作用。  相似文献

应用PMSG-hCG诱发兔黄体产生并用胶原酶-DNAase混合液分散黄体细胞方法,观察了黄体细胞存活率以及机能的变化,结果表明,该法制得的黄体细胞其存活率高达94%,孕酮分泌功能旺盛,提示黄体细胞结构完整,机能健全。可利用此细胞模型对激素的合成及影响因素加以研究。  相似文献

本文报道了用BCG致敏的大鼠耳部皮内注射PPD后所致的迟发型过敏反应的动态变化、浸润细胞的核型及细胞毒活性。DTH耳肿胀程度及浸润细胞的数目在PPD注射后渐增,第24~48h最明显,此后渐退。单克隆抗体染色法及荧光激活细胞分类仪定量研究,确定浸润细胞的T细胞亚群,主要为T辅助细胞和T诱导细胞,也有T抑制细胞和T杀伤细胞,短期~(51)C_r释放试验研究发现浸润细胞毒活性较低,进而在混合细胞培养中证实DTH细胞内有抑制脾自然杀伤细胞的抑制细胞。  相似文献

本实验以形态和功能相结合的方法应用细胞培养、扫描电镜、透射电镜及放射免疫等多种技术,研究了锗-132对体外培养大鼠腺垂体细胞的作用。Ge-132作用于细胞后,放免测定细胞上清液中生长激素(GH)、催乳激素(LTH)。Ge-132浓度为0.01mmol/L组与1mmol/L组比较GH、LTH浓度增高显著;实验组促甲状腺激素(TSH)、卵泡刺激素(FSH)、黄体生成素(LH)较对照组明显降低,且随着Ge-132浓度的增高,激素TSH、PSH、LH浓度降低更为显著。电镜观察腺垂体嗜酸性细胞里分泌功能旺盛相,嗜碱性细胞呈分泌功能不活跃状态。  相似文献
