ObjectiveTo study the in vitro antitrypanosomal activity, antioxidant property and phytochemical constituents of aqueous extracts of nine Nigerian medicinal plants.MethodsIn vitro antitrypanosomal activity test was carried out on aqueous extracts of dried leaves of Acacia albida (A. albida), Artemisia absinthium, Bryophyllum pinnatum, Gongronema latifolium, Holarrhena floribunda, Leptadenia hastata, Pericopsis laxiflora (P. laxiflora) and dried stem barks of A. albida and P. laxiflora. The phytochemical constituents and composition of the extracts and the in vitro antioxidant activity of the extracts were subsequently measured using the α,α-diphenyl-β-picryl-hydrazyl (DPPH) radical scavenging assay, Ferric reducing antioxidant power (FRAP) assay, thiobarbituric acid (TBA) activity assay and H2O2 radical scavenging activity assay.ResultsFrom the study, it was discovered that the stem bark extracts of A. albida and P. laxiflora were most active against both Trypanosoma evansi and Trypanosoma congolense. There was complete cessation of motility in both trypanosomes within 5 min at 40 mg/mL of the stem bark extract of A. albida and complete cessation of motility within 25 min and 40 min at 40 mg/mL with P. laxiflora stem bark extract for Trypanosoma congolense and Trypanosoma evansi, respectively. Quantitative analysis of the phytochemical constituents of the aqueous extracts of the plant parts such as alkaloids, saponins, flavonoids and phenols revealed that the stem barks of A. albida, P. laxiflora and leaves of Leptadenia hastata contained relatively high amount of all the phytochemicals quantified. The stem bark extracts of A. albida, P. laxiflora and leaves of Gongronema latifolium possess more scavenging capacity when compared to other extracts in relation to vitamin C, the reference antioxidant.ConclusionsThis study provides scientific evidence for the use of A. albida, and P. laxiflora for the treatment of trypanosomosis and diseases associated with oxidative stress.  相似文献

ObjectiveTo detect the in vitro total phenolics, flavonoids contents and antioxidant activity of essential oil, various organic extracts from the leaves of tropical medicinal plant Tetrastigma from Sabah.MethodsThe dry powder leaves of Tetrastigma were extracted with different organic solvent such as hexane, ethyl acetate, chloroform, butanol and aqueous methanol. The total phenolic and total flavonoids contents of the essential oil and various organic extracts such as hexane, ethyl acetate, chloroform, butanol and aqueous ethanol were determined by Folin - Ciocalteu method and the assayed antioxidant activity was determined in vitro models such as antioxidant capacity by radical scavenging activity using α, α-diphenyl- β-picrylhydrazyl (DPPH) method.ResultsThe total phenolic contents of the essential oil and different extracts as gallic acid equivalents were found to be highest in methanol extract (386.22 mg/g) followed by ethyl acetate (190.89 mg/g), chloroform (175.89 mg/g), hexane (173.44 mg/g), and butanol extract (131.72 mg/g) and the phenolic contents not detected in essential oil. The antioxidant capacity of the essential oil and different extracts as ascorbic acid standard was in the order of methanol extract > ethyl acetate extract >chloroform> butanol > hexane extract also the antioxidant activity was not detected in essential oil.ConclusionsThe findings show that the extent of antioxidant activity of the essential oil and all extracts are in accordance with the amount of phenolics present in that extract. Leaves of Tetrastigma being rich in phenolics may provide a good source of antioxidant.  相似文献

ObjectiveTo test antibacterial and antioxidant activity of the lemongrass plant Cymbopogon citratus (C. citratus) leaves extracted serially by the solvents (chloroform, methanol and water).MethodsThe plant leaves extracts were used for antibacterial activity on Bacillus subtilis, Pseudomonas aeruginosa, Proteus vulgaris, Staphylococcus aureus, Nocardia sp., Serratia sp., and Enterobacter aeruginosa microorganisms by the Kirby Bauer agar disc diffusion method. This study was carried out on lemongrass plant leaf extracts in different concentration of all solvents. The leaf extracts from different solvents were tested for their scavenging activity against the stable free radical DPPH in quantization using a spectrophotometric assay. Oxidative damage was induced in vitro by treating blood DNA and analyzing the effects of the leaf extracts.ResultsThe results showed that C. citratus extracts exhibited maximum zones of inhibition in chloroform, methanol and water extracts. It was Observed that the C. citratus extracts exhibited maximum zone of inhibition against Bacillus subtilis, Pseudomonas aeruginosa and Proteus vulgaris. Analyzed data in the present work suggested that antibacterial activity of C. citratus plant leaf extracts showed good results for Gram-positive and Gram-negative organisms. DPPH scavenging activity was highly elicited by the extract of C. citratus. Chloroform, methanol and water extracts of C. citratus leaves effectively decreased the extent of DNA damage.ConclusionsThe present study suggested that the lemongrass plant extracts could offer various health benefits.  相似文献

ObjectiveTo evaluate the antioxidant potential of different extract/fractions of Anthocephalus cadamba (A. cadamba) (Roxb.) Miq. (Rubiaceae) and study the tentative identification of their active constituents.MethodsThe extract/fractions were screened for antioxidant activity using various in vitro assays viz. DPPH assay, ABTS assay, superoxide anion radical scavenging assay, reducing power assay and plasmid DNA nicking assay. Total phenolic content of extract/fractions was determined by colorimetric method. An ultra–performance LC-electrospray-quadrupole-time of flight mass spectrometry method was used to analyse the active constituents of extract/fractions of A. cadamba.ResultsThe ethyl acetate fraction was found to be most active fraction in all the assays as compared to other extract/fractions. The IC50 value of ethyl acetate fraction (ETAC fraction) was 21.24 μg/mL, 1.12 μg/mL, 9.68 μg/mL and 57.81 μg/mL in DPPH assay, ABTS assay, reducing power assay and superoxide scavenging assay respectively. All the extract/fractions also showed the potential to protect the plasmid DNA (pBR322) against the attack of hydroxyl radicals generated by Fentońs reagent. The bioactive compounds were identified by UPLC-ESI-QTOF-MS, by comparing the mass and λmax with literature values.ConclusionsThe potential of the extract/fractions to scavenge different free radicals in different systems indicated that they may be useful therapeutic agents for treating radical-related pathologic damage.  相似文献

ObjectiveTo evaluate phytopharmacologically eugenol and two extract products of Ocimum gratissimum Linn. (O. gratissimum) (Labiaceae) on free radical scavenging and antioxidant activity.MethodsAqueous and methanol extract of fresh aerial part of O. gratissimum were prepared and eugenol (1-allyl-4-hydroxy-3-methoxybenzene) was isolated from fresh leaves and characterized by high performance liquid chromatography, fourier transform infrared spectroscopy, 1 h nuclear magnetic resonance. To establish the antioxidant potentiality of aqueous extract, methanol extract and eugenol, 1, 1-diphenyl-2-picrylhydrazyl radical, hydroxyl radical, nitric oxide scavenging activity, antioxidant activity by ferric thiocyanate and reducing power were measured in chemical system in vitro.ResultsSignificant (P<0.05) concentration-dependent free radical scavenging activity, antioxidant activity, and reducing power was observed by O. gratissimum products. Moreover, eugenol is more potent than the two extract products of O. gratissimum, but lower than potent antioxidant ascorbic acid.ConclusionsHence, O. gratissimum presents a potentially valuable source of natural antioxidant and bioactive material.  相似文献

ObjectiveTo investigate the in vitro antioxidant and antimicrobial activities along with phytochemical screening of organic and aqueous extracts of leaf and stem of Suaeda maritima (Dumort), a mangrove associate from Bhitarkanika of Odisha, India.MethodsAntioxidant activity of the crude extracts was evaluated in terms of total antioxidant capacity, total phenol content, ascorbic acid content, DPPH radical scavenging, metal chelating, nitric oxide scavenging, and reducing power etc. The antimicrobial activity of the plant was determined by agar well diffusion method along with MIC and MBC carried out by microdilution techniques against 10 gram positive and gram negative human pathogenic bacteria. The qualitative and quantative phytochemical screening were carried out by standard biochemical assays.ResultsOut of the seven antioxidant bioassays, both the leaf and stem extracts were found to posses strong antioxidant properties of 70 % to 92 % for phenol, total antioxidant capacity, DPPH free radical scavenging activity and fairly good ascorbic acid content, metal chelating (1.33 %-22.55 %), reducing power (0.01-0.12) and nitric oxide scavenging (0.84 %-66.99 %) activities. Out of the four extracts evaluated for antimicrobial activity, two leaf extracts such as acetone and ethanol showed promising activity against four pathogenic bacteria and one stem methanol extracts against one pathogenic bacteria when compared with amoxcycillin as standard. The MIC and MBC values of the antimicrobial extracts ranged between 2.5 to 5.0 mg/mL. Screening of phytochemicals showed presence of carbohydrates, protein, tannins, alkaloids and flavonoids in comparatively higher amount than other phytochemicals tested.ConclusionsThe present study reveals the presence of potential antioxidants and antimicrobial properties in the plant extract which could be exploited for pharmaceutical application.  相似文献

ObjectiveTo elucidate the antioxidant activity and total phenolic content (TPC) of ethyl acetate extracts of endophytic fungi isolated from Eugenia jambolana by three different antioxidant assays.MethodsTwenty one different endophytic fungal extracts were screened for presence of various phytochemicals, TPC and in vitro antioxidant activity. TPC was tested by Folin-Ciocalteau reagent based assay. DPPH free radical scavenging, hydrogen peroxide scavenging and reducing power assays were used to evaluate the antioxidant activity.ResultsAlkaloids, phenols, flavonoids, saponins, and terpenes were the main phytochemicals presents in all 21 endophytes. A significant positive correlation was found between antioxidant activity and TPC in fungal extracts. There is 36% endophytic extracts having high phenolic content exhibited potent antioxidant activity. Chaetomium sp., Aspergillus sp., Aspergillus peyronelii and Aspergillus niger strain showed the highest antioxidant activity ranging from 50% to 80% having 58 mg/g to 60 mg/g GAE total phenolics. Ascorbic acid used as a standard showed 90% reducing potential.ConclusionsThe results reveal that metabolites produced by endophytic fungi isolated from Eugenia jambolana can be a potential source of novel natural antioxidant compounds.  相似文献

ObjectiveTo determine the phenolic content in Codariocalyx motorius root extract and to evaluate its antioxidant properties using various in vitro assay systems.MethodsThe antioxidant activity was evaluated based on scavenging of 1,1-diphenyl-2-picrylhydrazyl, hydroxyl radicals, superoxide anions, nitric oxide, hydrogen peroxide, peroxynitrite, reducing power and by inhibition of lipid peroxidation which was estimated in terms of thiobarbituric acid reactive substances.ResultsThe root extract of the Codariocalyx motorius (C. motorius) exhibited potent total antioxidant activity that increased with increasing amount of extract concentration, which was compared with standard drug such as quercetin, butylated hydroxytoluene, tocopherol at different concentrations. The different concentrations of the extracts showed inhibition on lipid peroxidation. In addition, the extracts had effective reducing power, free radical scavenging, super oxide anion scavenging, nitric oxide scavenging, lipid peroxidation, and total phenolic content depending on concentration. High correlation between total phenolic contents and scavenging potential of different reactive oxygen species (r2=0.831–0.978) indicated the polyphenols as the main antioxidants.ConclusionsCodariocalyx motorius (C. motorius) root possess the highly active antioxidant substance which can be used for the treatment of oxidative stress-related diseases.  相似文献

ObjectiveTo investigate the in vitro antioxidant and antiproliferative activity of rhizome extracts of Rhodiola imbricata (R. imbricata) in HT-29 human colon cancer cell line.MethodsThe successively extracted rhizome of R. imbricata using various solvents was analyzed for their total phenolics, tannins and flavonoid contents. In vitro antioxidant activity was evaluated by employing different assays, including DPPH, ABTS radical scavenging assays, FRAP, phosphomolybdenum reduction assay, superoxide anion, hydroxyl radical scavenging activities and metal chelating ability.ResultsAcetone and methanol extracts recorded higher phenolic content and showed comparable antioxidant activity with standard reference. Additionally, they also inhibited the proliferation of HT-29 cells upon treatment at higher concentration (200 μg/mL) (acetone and methanol, 84% and 84%, respectively). On examination acetone extract exhibited antiproliferative activity in a concentration dependent manner whereas, methanol extract showed both dose dependent and time dependent inhibitory activity.ConclusionsThe results obtained justify the traditional usage of R. imbricata from their promising antioxidant activity.  相似文献

ObjectiveTo investigate the antihyperglycemic and antioxidant properties of the petroleum Ether, chloroform and methanol extract ofSaraca asoca (Roxb.) De Wild (S. asoca) leaves in mice.MethodsSwiss albino mice were made diabetic by a single dose of streptozotocin (150 mg/kg i.p.). Blood glucose levels and body weights of mice were measured using on weekly intervals i.e. day 0, 7, 14 and 21 after daily administration of all extracts at doses of 250 and 500 mg/kg. Other biochemical parameters such as serum cholesterol, triglycerides, HDL-cholesterol, VLDL-cholesterol, LDL-cholesterol, urea, creatinin and proteins levels were also measured at the end of study. The effects of all extracts were also noticed on vital organs e.g. liver, kidney and pancreas. The antioxidant potential of all extracts was also determined by DPPH and H2O2 radical scavenging in vitro methods.ResultsOral administration of the extracts for 21 days caused a significant (P <0.01) reduction in blood glucose levels in diabetic mice. Among all the extracts, methanol extract showed better results. The body weight of diabetic animals was also improved after daily administration of extracts. All the extracts also improved other altered biochemical parameters associated with diabetes. Furthermore, the extracts have favorable effects on the histopathological changes of the pancreas, liver and kidney in STZ induced diabetic mice. All the extracts showed significant (P <0.05) antioxidant activity at the dose of 500 µg/ml.ConclusionsS. asoca possesses antihyperglycemic and antioxidant properties as well improves body weight, liver profile, renal profile and total lipid levels. It can justify folklore uses of the plant in diabetes.  相似文献

ObjectiveTo evaluate antioxidant and radical scavenging activities of organic extracts from fruit, roots and aerial parts of Fagonia cretica.MethodsShed dried and powdered plant parts were initially extracted in methanol and subsequently partitioned in n-hexane, chloroform, ethyl acetate and 1-butanol successively. Antioxidant and radical scavenging potential of the methanol extracts and the fractions of each part were evaluated using total phenolic contents (TPC) and total flavonoid contents (TFC), 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radical cation radicals scavenging, reducing power (potassium ferricyanide-trichloroacetic acid system), ferric ion reducing antioxidant potential, lipid peroxidation inhibition activity (linoleic acid system) and total antioxidant activity (phosphomolybdate) assays.ResultsTPC and TFC values for methanol extracts and various fractions ranged from 0.23-4.30 mg/L gallic acid equivalents and from 30-545 mg/L rutin equivalents, respectively. Overall, methanol extracts and all the fractions of root and aerial parts showed higher TPC and TFC values. Methanol extracts and aqueous fractions of root and aerial parts and the n-butanol fraction of root showed lower EC50 values for 2,2-diphenyl-1-picrylhydrazyl scavenging than the other plant extracts. The 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging, total antioxidant potential and ferric ion reducing antioxidant potential values confirmed the presence of potent antioxidant principles in the methanol extract of roots. In general, all the extracts/fractions and especially those of root showed high antioxidant and radical scavenging activities.ConclusionsThe crude methanol extract of root can be explored further for in vivo studies. This study revealed the potent antioxidant potential of Fagonia cretica and its prospective efficacy against various reactive oxygen species-mediated diseases.  相似文献

ObjectiveThe aim of the present study was to investigate the in vitro antioxidant assays and in vivo non enzymatic antioxidant potential of aerial parts of Pergularia daemia methanolic extract (PDME) on 7, 12-dimethylbenz(a)anthracene (DMBA) induced hamster buccal pouch (HBP) carcinogenesis.MethodsThe plant extract was tested for their total phenol and flavonoid content and radical scavenging effect such as 2, 2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS?+) radical decolorization assay, DPPH (2, 2-diphenyl, 2-picryl hydrazyl) radical scavenging, nitric oxide radical scavenging, reducing power assays.ResultsAccording to IC50values obtained, PDME contains more powerful antioxidant compounds than gallic acid. The level of plasma vitamin C and the levels of Vitamin E and GSH in the plasma and erythrocyte lysate were decreased while the buccal tissue levels of vitamin E and GSH were increased in DMBA induced animals. Pretreatment with P.daemia (200 mg/kg bw) to DMBA induced animals significantly (P < 0.05) increased the levels of plasma and erythrocyte vitamin C, vitamin E and GSH where as the levels of vitamin E and GSH in the buccal tissue were increased compared to DMBA alone induced animals.ConclusionsThe PDME exhibited strong antioxidant activity in free radical scavenging assays and hamster buccal pouch carcinogenesis.  相似文献

ObjectiveTo compare the antioxidative effects of the methanolic leaf extracts of Ficus religiosa (F. religiosa), Chromolaena odorata (C. odorata), Cynodon dactylon (C. dactylon) and Tridax procumbens (T. procumbens) as well as the contents of antioxidants in the extracts.MethodsTotal phenol and total flavanoid contents were measured according to the standard procedures. The total antioxidant capacity was determined using the phosphomolybdenum method. Reducing power was determined by the potassium ferricyanide reducing method. The free radical scavenging activity was measured by 2,2′-Diphenyl-1-picrylhydrazyl (DPPH) assay.ResultsQuantitative phytochemical analysis of total phenol content showed that C. odorata had the highest content of phenolic compounds significantly followed by F. religiosa, T. procumbens and C. dactylon. As for the total flavanoids content, F. religiosa had the highest content, followed by C. odorata, T. procumbens and C. dactylon. Study on the total antioxidant capacity revealed that F. religiosa, C. dactylon and C. odorata showed higher total antioxidant capacity. T. procumbens showed the lowest capacity. Meanwhile, T. procumbens and C. odorata have the highest reducing power activity followed by F. religiosa and C. dactylon. The results of DPPH radical scavenging activity indicated that T. procumbens induced the largest elevation as the concentration of its extract increased, followed by C. odorata and F. religiosa and C. dactylon.ConclusionsThe present study demonstrates the antioxidative capacity of all the four plant species. Of all the plants, C. odorata, a perennial weed plant showed potentially a high antioxidant activity, with higher phenolic and flavonoids contents. The data suggest that C. odorata can be best utilized in developing bioantioxidants.  相似文献

ObjectiveTo evaluate the anti-oxidant and anti-inflammatory activity of leaf extracts and fractions of Mangifera indica in in vitro conditions.MethodsIn vitro DPPH radical scavenging activity and lipoxygenase (LOX) inhibition assays were used to evaluate the anti-oxidant and anti-inflammatory activities respectively. Methanolic extract (MEMI), successive water extract (SWMI) and ethyl acetate fraction (EMEMI), n-butanol fraction (BMEMI) and water soluble fraction (WMEMI) of methanolic extract were evaluated along with respective reference standards.ResultsIn in vitro DPPH radical scavenging activity, the MEMI, EMEMI and BMEMI have offered significant antioxidant activity with IC50 values of 13.37, 3.55 and 14.19 μg/mL respectively. Gallic acid, a reference standard showed significant antioxidant activity with IC50 value of 1.88 and found to be more potent compared to all the extracts and fractions. In in vitro LOX inhibition assay, the MEMI, EMEMI and BMEMI have showed significant inhibition of LOX enzyme activity with IC50 values of 96.71, 63.21 and 107.44 μg/mL respectively. While, reference drug Indomethacin also offered significant inhibition against LOX enzyme activity with IC50 of 57.75. Furthermore, MEMI was found to more potent than SWMI and among the fractions EMEMI was found to possess more potent antioxidant and anti-inflammatory activity.ConclusionsThese findings suggest that the MEMI and EMEMI possess potent anti-oxidant and anti-inflammatory activities in in vitro conditions.  相似文献

ObjectiveTo investigate the hepatoprotective and antioxidant activities of Alchornea cordifolia (A. cordifolia) leaf extract.MethodsVarious solvent fractions of the methanol extract of the leaf of the plant A. cordifolia Mull. Arg (Fam: Euphorbiaceae) were evaluated for hepatoprotective activity by carbon tetrachloride-induced liver damage in rats. The degree of protection was measured by using biochemical parameters such as serum glutamate oxalate transaminase (SGOT/AST), serum glutamate pyruvate transaminase (SGPT/ALT), alkaline phosphatase (ALP) and total bilirubin. The in vitro antioxidant activity of the extract was also evaluated by the 1, 1-diphenyl- 2-picrylhydrazyl (DPPH) free radical scavenging assay. The extract was subjected to preliminary phytochemical screening.ResultsThe ethyl acetate and chloroform fractions, at a dose of 300 mg/kg, produced significant (P<0.05) hepatoprotection by decreasing the activities of the serum enzymes and bilirubin while there were marked scavenging of the DPPH free radicals by the fractions. The effects were comparable to those of the standard drugs used for the respective experiments, silymarin and ascorbic acid. Alkaloids, flavonoids, saponins and tannins were detected in the phytochemical screening.ConclusionFrom this study, it was concluded that the plant of A. cordifolia possesses hepatoprotective as well as antioxidant activities and these activities reside mainly in the ethyl acetate and acetone fractions of methanol leaf extract.  相似文献

ObjectiveTo investigate the antioxidant activity of marine actinobacteria.MethodsThe content of total phenolics, the level of antioxidant potential by DPPH radical scavenging activity, metal chelating activity, FRAP method, β carotene assay and NO scavenging activity in extract were determined.ResultsIn all the methods the extract exhibited good scavenging activity except NO scavenging activity. The IC50 values of marine actinobacteria extract on DPPH radical were found to be 41.09 μg/mL. The zone of color retention was 12 mm in β-carotene bleaching assay. DNA protective efficiency of the extracts was also studied using UV-photolysed H2O2-driven oxidative damage to pBR322. HPLC analysis identified some of the major phenolic compounds in extracts, which might be responsible for the antioxidant potential and cyto-protection. It showed a 100% cytotoxic effect in brine shrimp lethality assay within 10 mins. The novel actinobacteria was identified as Streptomyces LK-3 (JF710608) through 16S rDNA Sequencing.ConclusionsThe results obtained suggest that the extracts bear anti-cancer metabolites and could be considered as a potential source for anti-cancer drug development.  相似文献

ObjectiveTo evaluate the antioxidant and anti proliferative potential of different solvent extract of micropropagated and naturally grown plants of Leptadenia reticulata against various cancer cell lines.MethodsIn this study different extract were tested for cytotoxicity against human breast adenocarcinoma cell line MCF-7, human colon adenocarcinoma grade II cell line HT-29 and non cancer skeletal muscle cell line L6 through 3-(4, 5–dimethyl thiazol–2–yl)–5–diphenyl tetrazolium bromide assay. The total antioxidant potential was estimated by three different antioxidant model diphenylpicrylhydrazyl free radical scavenging activity, H2O2 scavenging activity and FeCl3 reducing activity.ResultsThe ethyl acetate extract of both naturally grown plant and tissue cultured plant exhibited significant cytotoxicity with IC50 values of 21 µg/mL, 26 µg/mL and 22 µg/mL; 20 µg/mL, 30 µg/mL and 18 µg/mL respectively against three cell lines. The diphenylpicrylhydrazyl free radical scavenging activity was found to be highest with IC50 value of 267.13 µg/mL in ethyl acetate extract. The methanolic extract exhibited moderate antioxidant activity with IC50 value of 510.15 µg/mL. A highly positive correlation was observed between the antioxidant potential and cytotoxic activity of the plant.ConclusionsThe strong cytotoxicity of ethyl acetate extract revealed anti carcinogenic potential of the plant which supports its traditional use as medicine. The present investigation is new to literature till date and will provide better scientific basis for future pharmacological, in vivo studies and novel source of pure bioactive compounds having anti cancer properties in this plant.  相似文献

Objective:To evaluate in vitro antioxidant and apoptotic activities of Crperus rotundas(C.rotundus).Methods:The phytochemical study and the antioxidant activities of both methanol and aqueous extracts from C.rotundus aerial part were determined.In addition,these extracts were also investigated for their cytotoxic and apoptotic activities.The major compound of the methanol extract was isolated.Both metlianol and aqueous extracts(300,150,and 50μg/mL)were evaluated for their antioxidant activity by the xanthine/xanthine oxidase assay system.However,16,8,and 4 mg/mL of each extract were tested to investigate their OH.formation scavenging potential.Aqueous extract(800,400.and 200μg/mL)and melhunol extract(350,175,and 88μg/mL)were tested against lipid peroxidation,induced by 75μM H_2O_2,The cytotoxicity(by MTT assay)and cell DNA fragmentation of both extracts were evaluated Inwards K562 and L1210 cell lines.The major compound was obtained from the butanol fraction of methanol extract and its structure was determined by KMN spectroscopic analysis.Results:The methanol and aqueous extracts showed respectively,88%and 19%inhibition of xanthine oxidase activity.Vet.the same extracts inhibited lipid peroxidation by 61.5%and 42.0%.respectively.Roth extracts inhibited OH.formation by 27.1%and 25.3%,respectively.Only methanol cxtract induced DNA degradation.Orientin was determined as the major compound isolated from the butanol fraction of metlianol extract.Conclusions:It appears that C.rotundus extracts exhibit a potential use as a natural antioxidant and an apoptosis inducer.  相似文献

ObjectiveTo elucidate the phytochemical content and antioxidant activity of selected wild edible Nigerian mushroom species.MethodsPhytochemical screening was carried out using standard methods while 1,1-Diphenyl picryl hydrazyl (DPPH) radical and reductive power assays were used to evaluate the in vitro antioxidant properties of the selected edible Nigerian mushroom species.ResultsThe result obtained revealed the presence of alkaloids, cardiac glycosides, saponins, flavonoids, terpenes, steroids, tannins and phenols in the selected mushrooms extracts. The extract of Pleutorus ostearus showed a significantly (P<0.05) higher total phenol and flavonoid content of (248.80±7.63) mg/g and (42.63±0.63) mg/g respectively compared to other mushroom extracts. Cantherale cibarus had the most significant (P<0.05) amount of alkaloids [(135.57±0.27) mg/g] and saponins [(150.41±0.50) mg/g] when compared to other extracts while the tannin content [(170.56±0.74)] mg/g was highest in the mushroom Temitomyces robustus. All mushroom extracts scavenged DPPH radical in a dose dependent manner. However, Lactarus deliciousus had the highest DPPH scavenging activity compared to the other mushroom extracts. Pleutorus ostearus and Lactarus deliciousus had better reductive power than other mushroom extracts concentrations used.ConclusionsThe mushroom species analysed have been shown to be good sources of antioxidants and other phytoconstituents, thus it can be used in the management of oxidative stress induced diseases.  相似文献

ObjectiveTo evaluate the antioxidant activity of aqueous extract ofMoringa oleifera (M. oleifera) young leaves by in vivo as well as in vitro assays.MethodsIn vitro study included estimation of total phenolic, total flavonol, total flavonoid and total antioxidant power (FRAP assay). In addition, in vivo study was done with the identified most effective dose of 200 mg/kg of its lyophilized powder on normal and diabetic rats. Its effect on different oxidative free radical scavenging enzymes,viz, superoxide dismutase (SOD), catalase (CAT), glutathione-S-transferase (GST), lipid peroxide (LPO) contents were measured.ResultsSignificant increase in activities of SOD, CAT, GST while, a decrease in LPO content was observed. Whereas, total phenolic, flavonoid and flavonol contents in the extract were found to be 120 mg/g of GAE, 40.5 mg/g of QE and 12.12 mg/g of QE, respectively. On the other hand, FRAP assay results ofM. oleifera leaves was (85.00±5.00)μM/g of extract powder.ConclusionsThe significant antioxidant activities ofM. oleifera leaves from both in vivo as well as in vitro studies suggests that the regular intake of its leaves through diet can protect normal as well as diabetic patients against oxidative damage.  相似文献
