近年来 ,人们认为CD4 4作为细胞表面黏附分子与整合跨膜糖蛋白 ,在肿瘤发生、发展及转移中起着十分重要的作用 ,尤其是CD4 4基因拼接体———CD4 4v6更为引人注目[1] 。本文应用免疫组化方法 ,探讨CD4 4v6在大肠癌组织中的表达及其与淋巴转移程度的关系。一、材料与方法1.材料 :4 3例大肠癌组织为我院手术切除的新鲜标本 ,大肠腺瘤性息肉 19例与正常大肠黏膜 16例 ,均为本院内镜室在肠镜下采集的标本。三种组织分别送病理学检查与冷冻包埋剂 (OCT)包埋 ,液氮速冻 ,- 70°C冰箱保存待用。2 .免疫组化染色 :①试剂 :第一抗体为…  相似文献

目的 探讨大肠癌组织中CD44v6的表达,分析其与大肠癌临床病理指标的关系。方法采用高敏感性催化信号放大免疫组化技术,检测78例大肠癌组织、20例癌旁组织和20例正常大肠组织中CD44v6表达水平。结果大肠癌组织中CD44v6阳性表达率为83.3%,癌旁组织中为40%,正常组织中为阴性,两两比较,P〈0.01;CD44v6阳性表达与大肠癌的临床分期、分化程度、淋巴结转移及远处转移和患者预后有关(P〈0.05)。结论CD44v6的表达与大肠癌的侵袭、转移有关,并在大肠癌发生、发展中发挥重要作用。大肠癌组织中CD44v6的表达情况可作为判定其预后的新生物学指标。  相似文献

Purpose This study was conducted to evaluate the prognostic significance of CD44v5 and CD44v6 in resectable colorectal cancer.Materials and methods Membranous CD44v5 and CD44v6 levels were measured by an immunoenzymatic assay in tumors and surrounding mucosal samples obtained from 105 patients with resectable colorectal carcinomas.Results There were no significant differences of CD44v5 levels between tumors [median: 3.2 (range: 0.9–83.5) ng/mg protein) and surrounding mucosal samples (3 (3–146.2) ng/mg protein]. However, tumor samples showed significantly higher CD44v6 levels [19.5 (2.2–562.9) ng/mg protein] than mucosal samples [5 (5–230) ng/mg protein] (P=0.0001). Patients with higher CD44v5 or CD44v6 content in tumor samples had a considerably shorter relapse-free survival (P<0.05, for both). Patients with a higher CD44v6 content also had a shorter relapse-free and overall survival in the multivariate analysis (P<0.05).Conclusion The results of this study suggest a role of CD44v5 and CD44v6 in colorectal cancer progression. Membranous CD44v levels in primary tumors, measured by immunoenzymatic assay, may contribute to a more precise prognostic estimation in patients with resectable colorectal cancer.Supported by grants from ISCIII Red de Centros de Cancer RTICCC (C03/10) and Obra Social Cajastur  相似文献

目的探讨大肠癌患者外周血CD44表达变化的意义.方法以流式细胞仪对26例大肠癌患者外周血CD44表达进行免疫荧光检测,通过自身对照分析术前、术后变化,并与正常人(n=20)及大肠良性肿瘤患者(n=17)进行对比研究,t检验处理数据.结果大肠癌患者外周血CD44细胞(205%±45%)明显高于正常人(18%±11%)及大肠良性肿瘤患者(24%±14%,P<001),后二者差别不明显(P>005),已发生转移的大肠癌患者外周血CD44含量(229%±48%)较无转移者(161%±33%)明显增高(P<001),无转移组根治手术后外周血CD44含量迅速下降(29%±18%)至正常人水平,已有转移者变化不明显(P>005).结论CD44水平可作为大肠恶性肿瘤负荷及转移的指标.  相似文献

PURPOSE: This retrospective study was undertaken to evaluate immunohistochemically the expression of CD44 standard protein and CD44v5 and CD44v6 isoforms in colorectal adenomas and early invasive cancers developing within adenomas as possible markers characterizing colorectal polyps with a more aggressive biologic potential. METHODS: Archival tissues of 81 consecutive locally resected colorectal polyps, comprising 57 colorectal adenomas and 24 carcinomas-in-adenomas, were stained immunohistochemically with the use of commercially available mouse monoclonal antibodies: SFF-2 for CD44 standard protein, VFF-8 for CD44v5, and VFF-7 for CD44v6. RESULTS: Sixtythree percent of the colorectal polyps were positive for CD44 standard protein, 59 percent were positive for CD44v5, and 27 percent were positive for CD44v6. Ninetythree percent of the low-grade adenomas were CD44 standard protein-positive, in contrast to 50 percent of the high-grade adenomas and only 42 percent of the carcinomas-in-adenomas (Kendall's Tau =–0.42;P<0.0001). CD44v6 expression was more frequently found in early invasive cancers (54 percent) than in high-grade adenomas (25 percent) and low-grade adenomas (7 percent). This difference also was statistically significant (Kendall's Tau-b =0.39;P=0.00003). Surprisingly, a downregulation of CD44 standard protein expression was observed in the adenoma tissue adjacent to carcinomas (62 percent) and areas with high-grade atypia (71 percent), compared with low-grade adenomas (93 percent; Kendall's Tau-b =–0.28;P=0.004). CONCLUSIONS: Our data suggest that CD44 standard protein and CD44 isoform v6 expression differs considerably in benign and malignant colorectal polyps. 目的:探讨大肠癌组织RAB5A和CD44v9表达的意义.方法:应用SP免疫组化法检测43例大肠癌病理蜡块中RAB5A和CD44v9的蛋白质表达;取 60例大肠癌患者新鲜组织标本,提取RNA并逆转录合成cDNA,应用Real-time PCR法检测组织样本中RAB5A和CD44v9的基因表达;分析CD44v9和RAB5A的基因与蛋白质表达量与大肠癌分化及转移程度的相关性.结果:RAB5A蛋白表达阳性率为100%,阳性表达位于胞质和胞膜;CD44v9表达阳性率为 86.7%,阳性表达位于胞质;荧光定量PCR结果经熔解曲线分析各样本扩增产物的Tm值一致并与阳性对照的Tm值吻合.RAB5A cDNA 的表达量在高分化和中分化大肠癌组织中低于低分化大肠癌组织(P<0.01),在无淋巴结转移的大肠癌组织中低于有淋巴结转移的组织(P<0.01),而CD44v9 cDNA的表达趋势与 RAB5A相同;RAB5A与CD44v9的蛋白和基因表达均呈正相关(x2=14.532,P<0.01).结论:RAB5A和CD44v9的表达与大肠癌的分化和转移有关,可能在跨膜信号传导和基质侵袭中共同发挥作用.  相似文献

BACKGROUNDOxidized low-density lipoprotein (ox-LDL), which is abnormally increased in the serum of colorectal cancer (CRC) patients consuming a high-fat diet (HFD), may be one of the risk factors for the development of CRC. Ox-LDL exerts a regulatory effect on macrophages and may influence CRC through the tumor microenvironment. The role of ox-LDL in CRC remains unclear.AIMTo investigate the role of ox-LDL through macrophages in HFD associated CRC.METHODSThe expression of ox-LDL and CD206 was detected in colorectal tissues of CRC patients with hyperlipidemia and HFD-fed mice by immunofluorescence. We stimulated the macrophages with 20 μg/mL ox-LDL and assessed the expression levels of CD206 and the cytokines by cell fluorescence and quantitative polymerase chain reaction. We further knocked down LOX-1, the surface receptor of ox-LDL, to confirm the function of ox-LDL in macrophages. Then, LoVo cells were co-cultured with the stimulated macrophages to analyze the CD44 and CD133 expression by western blot.RESULTSThe expression of ox-LDL and the CD206 was significantly increased in the stroma of colorectal tissues of CRC patients with hyperlipidemia, and also upregulated in the HFD-fed mice. Moreover, an increased level of CD206 and decreased level of inducible nitric oxide synthase were observed in macrophages after ox-LDL continuous stimulation. Such effects were inhibited when the surface receptor LOX-1 was knocked down in macrophages. Ox-LDL could induce CD206+ macrophages, which resulted in high expression of CD44 and CD133 in co-cultured LoVo cells.CONCLUSIONOx-LDL stimulates CD206+ macrophages to upregulate CD44 and CD133 expression in HFD related CRC.  相似文献

BACKGROUND AND AIMS: In colorectal cancer patients' mortality is largely influenced by spreading of tumour cells from the primary tumour site and subsequent metastasis formation. CD44 is an adhesion molecule and represents a highly variable family of isoforms. The isoform CD44v6 has been associated with metastatic spread and poor prognosis in animal models and several human cancers. Results of immunohistological studies in primary colorectal cancer are mostly retrospective and contradictory. The aim of our prospective study was to assess the controversial role of CD44v6 as a prognostic factor in colorectal cancer. METHODS: In 93 patients we analysed tumour CD44v6 expression in prospectively sampled stage I-IV colorectal adenocarcinomas using RT-PCR and Southern blotting. The prognostic value of the CD44v6 expression was assessed using univariate and multivariate analysis. RESULTS: CD44v6 expression was found in 47 % of the cases. CD44v6 expression failed to show any association with the clinical or histological variables examined. CD44v6 expression did not correlate with survival in long-term follow-up. The most important prognostic factor in this cohort was tumour stage. CONCLUSIONS: Changes in CD446 expression level do not predict tumour spread or patient survival in colorectal cancer.  相似文献

目的 研究老年大肠癌患者外周血CD44v5、CD44v6表达水平与血行转移的关系及化疗对其影响。方法 应用流式细胞仪检测 49例老年大肠癌患者手术前外周血CD44变异体 (CD44v5、CD44v6)表达水平 ,与健康老年人及非老年大肠癌患者进行对照 ,并在应用化疗后观察黏附分子表达水平的变化。结果 老年大肠癌组外周血CD44v5、CD44v6的表达水平分别为 1 0 71± 5 44、1 4 62± 6 34 ,明显高于健康对照组 (P <0 0 1 ) ,与非老年大肠癌组比较无显著性差异 (P >0 0 5)。CD44v5、CD44v6的表达水平与老年大肠癌患者肿瘤大小、肿瘤浸润深度及淋巴结转移相关 ,老年大肠癌组肿瘤≥ 5cm、肿瘤浸润深度≥深肌层、淋巴结转移阳性患者外周血CD44v5、CD44v6的表达水平分别高于肿瘤 <5cm、肿瘤浸润深度 <深肌层、淋巴结转移阴性患者 (P <0 0 1、P <0 0 5、P <0 0 1 )。老年大肠癌患者化疗后外周血CD44v5、CD44v6表达水平较化疗前明显下降 (P<0 0 1 )。结论 黏附分子具有介导老年人大肠癌细胞血行转移的作用 ,化疗可降低老年大肠癌患者外周血CD44v5、CD44v6的表达水平 ,具有抗黏附和肿瘤转移作用  相似文献
