Changes of Expressions of VEGF, bFGF, and Angiogenesis, and Effect of Benazepril, bFGF on Angiogenesis in Acute Myocardial Infarction Model of the Rabbits

Objective To explore the changes of vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF) and angiogenesis, and the effects of bFGF, angiotensin converting enzyme inhibiter(ACEI) benazepril on the angiogenesis in acute myocardial infarction (AMI) model of rabbits, and to provide a probable evidence for the treatment of AMI. Methods AMI model was established by ligating anterior descending branch of coronary artery of Japan-Sino hybridization white rabbits. The postoperative rabbits were randomly divided into 6 groups and each group was treated with different drugs. Groups 1 and 2 were treated with normal saline (NS) for 28 and 14 days (d), group 3 and 4 with bFGF for 28 and 14 d, groups 5 with benazepril for 14 d, and group 6 with benazepril and bFGF for 14 d respectively. The rabbits were killed on the 14th or 28th d and their hearts were excised, sectioned and stained with HE, Masson trichrome to observe VEGF, bFGF and CD34 under a microscope, which were quantified with a computer-assisted morphometry. Results Compared with group 1, the granulation tissue of infarction zone (IZ) in group 2 freshened up, and the capillary density (CD) in IZ was increased (P=0.002). The CD in the IZ as well as VEGF and bFGF in groups 3 and 4 were increased respectively (P=0.011-0.037). In group 5 the changes of VEGF and bFGF were not found in the IZ and the border zone (BZ) while CD was significantly increased (35.4% and 25.6%, P=0.036 and 0.037). Compared with group 2, the CD in the IZ and BZ of group 6 was significantly increased (63.4% and 44.3% P=0,007 and 0.007), meanwhile VEGF and bFGF were increased. Compared with group 5, only VEGF was increased. Conclusion Intravenous bFGF may increase VEGF and bFGF significantly, thus promoting the angiogenesis in the IZ and BZ in cardiac infarction as VEGF and bFGF are the potent angiogenic growth factors. Benazepril may promote angiogenesis in the IZ and BZ in cardiac infarction, but its mechanism is irrelative to the expression of VEGF and bFGF. The combination of benazepril and bFGF may promote, to some extent, the expression of VEGF and bFGF, but their effect on angiogenesis has not been found.

Job Stress, Gene Polymorphism of β2-AR, and Prevalence of Hypertension

Objective To study the interactive effect of job stress and genetic susceptibility （or gene polymorphism） on hypertension. Methods A cross-sectional epidemiological study was conducted in 452 workers from a thermal power plant in China. Extrinsic effort, occupational reward, and over-commitment were measured. Hypertensive patients were defined by three phases of screening, reexamination, and final diagnosis. β2-AR genotypes and allele frequencies at amino acid positions 16 （β2-AR-16： Arg→Gly） and 27 （β2-AR-27： Gln→Glu） were identified by PCR-RFLE Results Job stress was related with the prevalence of hypertension in males （P〈0.05）, whereas no significant relationship was found in females （P〉0.05）. Differences in genotypes and allele frequencies of the β2-AR-16 were statistically significant between the hypertension and control groups （P〈0.05）, whereas those of β2-AR-27 were not （P〉0.05）. The prevalence of hypertension was higher in individuals carrying Gly16 allele than in those carrying Arg16 allele of the high job stress group （P〈0.01 or 0.05）. Conclusion High job stress and polymorphism of β2-AR-16 have an interactive effect on the prevalence of hypertension in male workers.

Influence of vasectomy on cholesterol level and activities of CPK, GPT, AKP in serum of rabbits

采用动物模型对输精管结扎引起的血清胆固醇水平及一些血清酶活性的改变进行了研究，结果表明：①增龄可使血清胆固醇含量增高；②血清中肌酸磷酸激酶（CPK）、乳酸脱氢酶（LDH）、谷草转氨酶（GOT）、α-羟丁酸脱氢酶（HBDH）水平随年龄增长略有增高，但结扎组（VG）和假手术组（SOG）比较无差异；③血清酸性磷酸酶（ACP）水平，25月组明显高于6月组（P＜0．05），VG与SOG比较无明显差异（P＞0．05）；血清碱性磷酸酶（AKP），25月组明显低于6月组，VG与SOG无明显差异；血清谷丙转氨酶（GPT）与γ-谷氨酰转肽酶（γ-GD）的活性各组间无显著差异。提示：血清酶水平与衰老有关，输精管结扎对其无影响。

Radix Astragali(RA),a traditional Chinese medicine from the dried root of Astragalus species,is widely distributed throughout the temperate regions of the world.The major bioactive constituents of RA are triterpene glycosides,flavonoids,saponins,and alkaloids,and these compounds mostly exert pharmacological activities on the cardiovascular,immune,respiratory,and hepatic systems.This review summarizes the recent studies on RA and provides a comprehensive summary regarding the status of resources,ethnopharmacology,phytochemistry,pharmacology,toxicology,clinical application,and patent release of RA.We hope this review can provide a guidance for further development of therapeutic agents from RA.

The Effect of Calcitonin Gene Related Peptide on the MDA, T-SOD, T-ant-OX and the Expression of ICAM-1, NOS, TIMP-2 of the Endothelial Cells

Objective To investigate the protective and regulative function of calcitonin gene-related peptide （CGRP） on endothelial cells （EC）. Methods After the endothelial cells of the no diamide and diamide induced experimental groups were cultured with CGRP （ 1× 10.8 mol/L）, the supernate and cells were collected respectively. The supernate was used to detect the metabolic changes of MDA, T-ant-OX and the activity of T-SOD, and the cells were collected for determination of the mRNA expression of the intercellular adhesion molecule-1 （ICAM-1）, the tissue inhibitor of metalloproteinase-2 （TIMP-2） and the nitric oxide synthase （NOS） by RT-PCR and immunocytochemistry. Results CGRP can up-regulate the metabolic levels of T-SOD and T-ant-OX and the expression of TIMP-2 mRNA and NOS significantly, and down-regulate the metabolism of MDA and the mRNA expression of ICAM-1 after acted upon the EC of control and experimental groups. Conclusion The protective regulation of EC by CGRP can be realized through down-regulate lipid peroxidation injury to EC and the expression of ICAM-1 and up regulating the expression of TIMP-2 and NOS. It is suggested that CGRP might play a certain role in the prevention and treatment of atherosclerosis （AS）, especially significant in protection against early damage.     

The evaluation of IL-1β,IL-6,IL-8,TNF-α in diagnosis of bacterial bloodstream infection

目的 探讨白细胞介素(IL)-1β、IL-6、IL-8、肿瘤坏死因子α(TNF-α)及其联合检测在细菌性血流感染中的诊断价值.方法 采用化学发光方法检测144例细菌性血流感染患者和32例正常健康者的血清IL-1β、IL-6、IL-8、TNF-α水平.结果 革兰阴性菌和革兰阳性菌感染组的阳性率显著高于正常对照组(P<0.01);IL-6的敏感性和特异性均高于其他3项指标;4项指标的联合检测率高于单项指标的检测率.结论 IL-1β、IL-6、IL-8、TNF-α是参与细菌性血流感染的重要细胞因子;IL-6的诊断价值高于IL-1β、IL-8、TNF-α;联合检测有助于细菌性血流感染的检测.
