Objective:To study the expressions and clinical significances of PCNA, EGFR, Bcl-2 and Bax in thymoma. Methods: The expressions of EGFR, PCNA, Bcl-2 and Bax in 46 cases of thymoma and 11 cases of normal thymus were detected with S-P immunohistochemistry. The results were analyzed with the pathologic indexes. Results: The positive rates of EGFR, Bcl-2 and Bax in normal thymus were 18. 2%, 9. 1%,and 18.2% respectively, while in thymoma were 71.7%, 41.3%, and 15.2% separately. The expression of EGFR in thymoma was significantly correlated with Masaoka staging and Levine classification. The survival rate of EGFR negative patients was significantly higher thanthat of EGFR positive patients (P<0. 01). PCNA labelling index was significantly higher in thymoma as (4.00±1.87)% than in normal thymus, (2.68±0. 62)% , which was significantly correlated with Levine classification. The expression of Bcl-2 in thymoma was also significantly correlated with Levine classification. The survival rate of Bcl-2 negative patients was significantly higher than that of Bcl-2 positive patients (P<0. 01). The expression of PCNA, EGFR, Bcl-2and Bax in thymoma had no correlation with histologic type and myasthenia gravis (MG) (P>0. 05). Conclusion: The expression of EGFR may be involved in the occurrence and development of thymoma. EGFR can be regarded as a supplementary predictor for Masaoka staging so as to predict the progression accurately.The expression of Bcl-2 may contribute to the occurrence of thymic carcinoma and Bcl-2 can be used as a biomarker identifying thymic carcinoma. 相似文献

Expression and Significance of Bcl-2, Bax, Fas and Caspase-3 in Different Phases of Human Hemangioma 总被引:4,自引:0,他引:4
Summary: The relationship between Bcl-2, Bax, Fas, caspase-3 and development of hemangioma and the molecular mechanism was investigated. By using immunohistochemical S-P method, proliferating cell nuclear antigen was detected. According to the classification of Mulliken in combination with PCNA expression, 27 cases were identified as proliferating hemangioma and 22 cases as involutive hemangioma. Five normal skin tissues around the tumor tissue served as controls. By using immunohistochemical technique, the expression of Bcl-2, Bax, Fax and Caspase-3 was detected. The cells expressing Bcl-2, Bax, Fax and cappase-3 were identified as hemangioma endothelia by immunohistochemical staining of Ⅷ factor. The average absorbance (A) and average positive area rate of Bcl-2, Bax, Fas and caspase-3 expression were measured by using HPIAS-2000 imaging analysis system. The results showed that the expression of Bcl-2 in the endothelia of proliferating hemangioma was significantly higher that in involutive degenerative hemangioma endothelia and vascular endothelia of normal skin tissue (P〈0.01). The expression of Bax, Fas and Caspase-3 in the endothelia of involutive hemangioma was obviously higher than in the endothelia of proliferating hemangioma and normal skin tissue (P〈0.01). The expression of BAx and Fas in endothelia of proliferating hemangioma was higher than in those of normal skin tissue (P〈0.05). It was suggested that Bcl-2, Bax, Fas and caspase-3 might be involved in the development and involution of hemangioma. Bcl-2 could promote the growth of hemangioma by inhibiting apoptosis of endothelia. Bax, Fas and caspase-3 promote the switch of hemangioma from proliferation to involution by inducing the apoptosis of hemangioma endothelia. 相似文献

Objective To study the interactive effect of job stress and genetic susceptibility (or gene polymorphism) on hypertension. Methods A cross-sectional epidemiological study was conducted in 452 workers from a thermal power plant in China. Extrinsic effort, occupational reward, and over-commitment were measured. Hypertensive patients were defined by three phases of screening, reexamination, and final diagnosis. β2-AR genotypes and allele frequencies at amino acid positions 16 (β2-AR-16: Arg→Gly) and 27 (β2-AR-27: Gln→Glu) were identified by PCR-RFLE Results Job stress was related with the prevalence of hypertension in males (P〈0.05), whereas no significant relationship was found in females (P〉0.05). Differences in genotypes and allele frequencies of the β2-AR-16 were statistically significant between the hypertension and control groups (P〈0.05), whereas those of β2-AR-27 were not (P〉0.05). The prevalence of hypertension was higher in individuals carrying Gly16 allele than in those carrying Arg16 allele of the high job stress group (P〈0.01 or 0.05). Conclusion High job stress and polymorphism of β2-AR-16 have an interactive effect on the prevalence of hypertension in male workers. 相似文献

目的:研究l20例乳腺癌中ER、PR、P53、HER-2及TOPⅡ表达和临床意义.方法:应用免疫组化SP法测得ER、PR、P53、H ER-2及TOPⅡ在120例乳腺癌中表达情况.结果:ER、PR、P53、HER-2及TOPⅡ的表达分别是:48(40.00%)、50(42.67%)、71(59.17%)、67(55.83%)、69(57.50%).P53的表达与HER-2呈正相关,1P53、HER-2的表达与ER,PR呈负相关(P <0.05);ER、PR的表达与肿瘤临床分期及淋巴结转移呈负相关.P53、HER-2的表达与肿瘤临床分期及淋巴结转移呈正相关(P<0.05).结论:TOPⅡ、HER-2是判断乳腺癌预后的参考指标,ER、PR、P53、HER-2及TOPⅡ联合检测对乳腺癌治疗方案及预后判断有重要临床意义. 相似文献

P53 Gene Mutation and Expression of MDM2, P53, P16 Protein and their Relationship in Human Glioma 总被引:4,自引:0,他引:4
To investigate the effect of P53 protein accumulation and p53 gene mutation in the pathogenesis of glioma and to study the role of MDM2, P53 and P16 protein in glioma formation and progression and their relationship with each other, LSAB immunohistochemical staining method and non-isotopic PCR-SSCP techniques were used to detect the expression of MDM2, P53 and P16 protein and p53 gene mutation in 48 cases of gliomas. The results showed that the positive expression rate of MDM2, P53 and the negative rate of P16 was 22.9 %, 41.7 % and 60.4 %, respectively. The latter two in high grade (grade Ⅲ , Ⅳ) gliomas had a significantly higher rate than in the low grade (grade Ⅱ ) gliomas. Moreover, the co-expression of MDM2 and P53 protein was confirmed in only 1 of 48 cases. No significant difference was found in the rate of the expression of MDM2 between high grade and low grade gliomas (P〉0.1) . PCR-SSCP results showed that mutation of 5 --8 exons of p53 gene was detected in 17 out of 48 cases (35.42 %) . Mutation was detected in 16 of 20 cases of positive p53 expression, and another one was detected in 28 cases of negative expression cases. The correlation between p53 mutation and p53 immunopositivity was observed in 89.6 % of the cases. P53 gene mutation and the level of MDM2, P53 and PI6 protein were not related to age, gender of the patients, tumor location and size. It is concluded that the mutation of p53 and deletion of p16 might play important roles in the tumorigenesis of gliomas and it was significantly associated with the grade of tumor differentiation. P53 protein accumulation can indirectly reflect p53 mutation. MDM2 amplification and overexpression might be an early event in the growth of human gliomas. 相似文献

The role that free radicals in general and oxygen radicals in particular play in carcinogenesis has attracted considerable attention in recent years. The oxygen radicals are undesirable but inevitable products of aerobic metabolism in the normal living cell. The cellular antioxidant defense system maintains an appropriate balance between necessary oxidative events and those that are excessive. When this critical balance cannot be maintained because of the overloading of the cellularredox system, oxygen radicals can induce cell damage. They can influence carcinogenesis by inducing DN A damage from direct oxidation or indirectly from DNA-binding products of lipid peroxidation. Oxygen radicals can induce conformational changes in the plasma membrane by lipid peroxidation and protein degradation, thus influencing membrane-associated cellular activities. They are capable of affecting membrane-bound protein kinases, growth factors and their receptors, and, therefore, signal transduction and oncogene activation. Cupric chloride (CuCl_2) and zinc chloride (ZnCl_2) were tested by the FETAX (frog embryo teratogenesis assay: xenopus) procedure.Results: In control embryos, the survival rate at 101h post-fertilization was>95%) and teratogenic rate was>7%. The teratogenic concentrations (EC_(10), EC_(50), EC_(90)) of Cu~(2+) were 0.9, 2.5 and 4.4μmol/L; the embryolethal concentrations(LC_(10), LC_(50), LC_(90)) of Cu~(2+) were 10, 22 and 40 μmol/L; the teratogenic index(TI=LC_(50)/EC_(50))for Cu~(2+) was 8.8. EC_(10),EC_(50),EC_(90) of Zn~(2+) were 6.7,40 and 70μmol/L: LC_(10), LC_(50), LC_(90) of Zn~(2+) were 410, 850 and 1080μmol/L, TI for Zn~(2+) was 21. TI(1.5) was considered the threshold for positive teratogenicity in the FETAX assay. Both metal ions were shown to be potential teratogens for xenopus(TI>1.5). Mortality and teratogenic rate in embryos exposed to Cu~(2+) and Zn~(2+) were shown concentration-response relation. Head-to tail lengths of the tadpoles, measured at 101h post-fertilization were negative correlation with the Cu~(2+) and Zn~(2+) concentrations (r=0.95). The minimum concentration to inhibit growth,(MCIG) was 10 μmol/ L for Cu~(2+) and 300 μmol/L for Zn~(2+).The anomalies observed in Cu~(2+) and Zn~(2+) exposed tadpoles primarily affected the face、eyes、heart、gut、notochord and fins. The most common facial anomaly was a beak-like deformity of the snout, eye malformations included incomplete closure of the choroid fissure, herniation of the selera. Cardioproptosis was frequent findings, hypertrophy and distention of the gut were commonly noted. Flexion was frequent at the tip of the notochord or at the midsection of the notochord, associated with broad, ruffled fins. The authors emphasize that caution and scientific judgment must be used in extrapolating the results of FETAX assays to predict teratogenic risks for man and other mammals. 相似文献
