背景:骨髓间充质干细胞可以在体内外诱导下分泌神经营养因子,还可向神经元方向转化,从而促进创伤脑组织再生修复,但骨髓间充质干细胞又存在生存周期短的问题而影响其发挥对损伤脑组织的保护作用。目的:观察骨髓间充质干细胞移植联合神经节苷脂治疗大鼠颅脑创伤的效果。方法:60只Wistar大鼠采用液压颅脑损伤仪制备大鼠重型颅脑创伤模型,建模24 h后按照随机表法随机分为3组:模型组经尾静脉注入1 mL DMEM培养液,移植组经尾静脉注入1 mL(1×1010 L-1)骨髓间充质干细胞悬液,联合组经尾静脉注入1 mL(1×1010 L-1)骨髓间充质干细胞悬液以及经腹腔注射神经节苷脂水溶液(30 mg/kg),1次/d,共3 d。伤后24 h,细胞移植后3 d及1,2,3,4 周依据Longa 5分法进行神经功能行为学评分;移植后3 d应用RT-PCR、Western blot检测创伤脑组织中水通道蛋白4基因和蛋白的表达;移植后1周,行苏木精-伊红染色观察创伤脑组织病理变化。结果与结论:移植后3 d及1,2,3,4 周,联合组神经行为学功能评分低于移植组(P < 0.05),移植组低于模型组(P < 0.05);移植后3 d,模型组创伤脑周围组织水通道蛋白4蛋白及mRNA的表达高于移植组(P < 0.05),移植组高于联合组(P < 0.05);苏木精-伊红染色观察到联合组脑组织恢复明显优于模型组和移植组(P < 0.05)。结果提示骨髓间充质干细胞移植联合神经节苷脂可以明显改善颅脑创伤大鼠的神经行为学功能。中国组织工程研究杂志出版内容重点:干细胞;骨髓干细胞;造血干细胞;脂肪干细胞;肿瘤干细胞;胚胎干细胞;脐带脐血干细胞;干细胞诱导;干细胞分化;组织工程   相似文献

背景:神经节苷脂是目前世界公认的惟一具有修复神经损伤,重塑神经网络的特效物质。单唾液酸神经节苷脂是神经节苷脂的一种。 目的:观察骨髓间充质干细胞移植并应用单唾液酸神经节苷脂治疗大鼠脑梗死的效果。 方法:Wistar大鼠应用线栓法建立大脑中动脉阻塞模型后随机分为3组,建模成功6 h后通过尾静脉注射浓度为1×1010 L-1的骨髓间充质干细胞悬液或细胞培养液1 mL,同时经腹腔注射单唾液酸神经节苷脂水溶液或生理盐水,1次/d,连续3 d。静脉移植后24 h,3 d及伤后1、2 周行Longa行为学评分,检测神经功能的损伤情况。治疗3 d后用RT-PCR、Western Blot检测脑组织中AQP4mRNA表达和蛋白合成的变化。2周后行BrdU免疫组化和苏木精-伊红染色观察移植细胞的存活和对组织的修复情况。 结果与结论:移植后1,2周,大鼠神经功能障碍评分细胞移植+单唾液酸神经节苷脂组低于细胞移植组,细胞移植组低于梗死组(P < 0.05);移植后3 d,脑梗死周围组织AQP4蛋白及其mRNA的表达梗死组高于细胞移植组,细胞移植组高于细胞移植+单唾液酸神经节苷脂组(P < 0.05);2周后BrdU免疫组化和苏木精-伊红染色切片中的神经元数量细胞移植+单唾液酸神经节苷脂组多于细胞移植组,细胞移植组多于梗死组(P < 0.05)。结果提示骨髓间充质干细胞移植的同时应用神经节苷脂治疗可明显改善脑梗死大鼠的神经学功能。  相似文献

BACKGROUND:Both of acupuncture and mesenchymal stem cell transplantation have therapeutic effects on cerebral palsy. OBJECTIVE:To explore the therapentic effect of acupuncture combined with placenta derived mesenchymal stem cells on rat cerebral palsy. METHODS:Thirty-five Wistar rats were equivalently randomized into five groups, including control group, model group, acupuncture group, transplantation group and combination group. Rat models of cerebral palsy were prepared in all groups except the control group. At 7 days after modeling, in the transplantation group, 10 μL placenta derived mesenchymal stem cells were transplanted into the right cerebral cortex and striatum of rats; rats in theacupuncture group were given acupuncture at Shuigou (DU26), Dazhui (DU14), Baihui (DU20), Guanyuan (RN4) and Qihai (RN6) points; rats in the combination group given acupuncture and transplantation treatment as described above; the model and control groups received no treatment. Then, after 7-day intervention, the body mass, neurological function and pathological changes of rat brain tissues were measured and observed, respectively.  . RESULTS AND CONCLUSION:The order of the growth rate of body mass was as follows: control group > combination group > acupuncture and transplantation groups > model group, and all groups had significant differences except the two groups of acupuncture and transplantation (P < 0.05); findings from holding tract, foot error and water finding in maze tests showed the neurological function was best in the control group, followed by the combination group, as well as acupuncture and transplantation groups, and the worst in the model group. All groups had significant differences except acupuncture and transplantation groups (P < 0.05). Besides, in the model group, cells in the brain and hippocampus decreased, arranging disorderly with fuzzy structure, and inflammatory cells appeared; pathological changes in the other groups were in between control group and model group, Additionally, brain tissues in the combination group exlibited better pathological changes than those in the acupuncture and transplantation groups, which were similar with the normal brain tissues. To conclude, acupuncture, placenta derived mesenchymal stem cells transplantation, or their combination can improve neurological function in rats with cerebral palsy, especially the combination treatment.  相似文献

背景:国内外已有实验动物和临床应用脐血间充质干细胞移植治疗神经系统遗传性疾病安全、有效的诸多报道。 目的:探讨脐血间充质干细胞移植治疗儿童进行性脊髓性肌肉萎缩症的可行性及效果。 方法:2010-01收治1例确诊为儿童进行性脊髓性肌肉萎缩症患儿,经药物及康复治疗无效,行脐血间充质干细胞移植治疗。移植途径采取首次静脉输注,后3次蛛网膜下腔注入,1次/周,每次细胞数量达(4~6)×107个,4次为1个疗程。治疗前和治疗后半年均需完善神经系统体检、实验室检查、肌酶、FIM评分、肌电图等。 结果与结论:移植6个月与移植前比较,肌酶下降,FIM评分由68分提高到93分,肌电图检查重收缩每10.0 ms所检肌运动单位增加两三个,双下肢肌力增加,生活自理能力改善。随访10个月,患者未出现明显的不良反应。提示脐血间充质干细胞移植治疗该例儿童进行性脊髓性肌肉萎缩症患儿有效,其神经功能恢复明显。  相似文献

背景:近年研究显示,脐血间充质干细胞的自我更新和多向分化潜能为细胞移植治疗提供了基础条件,而其免疫调节功能也极大地拓展了细胞治疗的方向和范围。 目的:就近期脐血间充质干细胞的免疫调节和细胞移植研究进行回顾分析。 方法:应用计算机以“umbilical cord blood-derived mesenchymal stem cells”为关键词检索Pubmed数据库,以“脐血间充质干细胞”为关键词检索知网数据库,时间限定为2008-01/2011-06,语言种类为“English”和汉语。通过阅读标题和摘要进行初筛,排除研究内容与此文无关的文献、重复性研究及Meta分析,选择近期发表或发表在权威杂志的30篇文献进行综述。 结果与结论:脐血间充质干细胞具有与骨髓间充质干细胞相似的自我更新和多向分化潜能。通过细胞移植技术,脐血间充质干细胞在糖尿病、神经退化性疾病如阿尔茨海默病和帕金森病等以及神经损伤后的修复治疗方面显示出很强的潜力。同时,脐血间充质干细胞又具有免疫调节作用,其可通过下调T细胞的增殖,降低免疫反应。利用该特点,脐血间充质干细胞在一些免疫性疾病,如移植物抗宿主病和狼疮性肾炎等疾病的细胞治疗方面也取得了积极的进展。  相似文献

背景:脐血间充质干细胞移植治疗新生动物脑血肿继发脑水肿的研究少见报道。 目的:探讨脐血间充质干细胞静脉移植治疗新生鼠实验性脑血肿继发脑水肿的可行性。 方法:健康新生7 d SD大鼠分为未进行干预的空白对照组、仅制作脑出血模型的脑血肿对照组和制作脑出血模型后给予脐血间充质干细胞治疗的移植组。3组大鼠均于移植后1周处死,提取脑组织进行指标检测。 结果与结论:脑血肿形成后,脑系数和脑组织含水量均明显增加,脐血间充质干细胞移植组明显小于脑血肿对照组(P < 0.01),与空白对照组比较差异无显著性意义(P > 0.05)。移植1周后,各组鼠死亡率无显著差异,均未见植入反应和其他任何负作用。提示,脐血间充质干细胞静脉移植治疗能有效减轻实验性脑血肿继发的脑水肿,对其具有明显的保护作用。其机制可能与脐血间充质干细胞能有效地透过血脑屏障,在病灶脑组织周围迁移、扩散、整合,从而在病灶脑组织存在并分泌大量保护性细胞因子有关。  相似文献

背景:大量研究证实脐血间充质干细胞体外培养可被诱导分化为表达神经元和神经胶质细胞表面标志物的细胞,对神经系统疾病起到替代修复的作用。 目的:观察人脐血单个核细胞移植治疗痉挛型小儿脑性瘫痪的疗效和安全性。 方法:收集正常足月剖腹产胎儿的脐血,经肝素抗凝,用淋巴细胞分离液分离脐血单个核细胞,采用差速贴壁法进行纯化,并经腰椎穿刺注入30例痉挛型脑性瘫痪患者体内。 结果与结论:移植后患者肢体的肌肉痉挛程度和关节活动度明显改善,与移植前相比差异有显著性意义(P < 0.05),移植后和随访过程中无不良反应和并发症。  相似文献

目的:探讨神经节苷脂治疗婴幼儿大脑性瘫痪(脑瘫)的临床疗效。方法:将84例脑瘫患儿随机分为治疗组和对照组,各42例。两组性别、年龄、临床分型、伴随疾病、头颅CT及发病因素等对比差异均无显著意义(P〉0.05)。两组均采用支持、对症、脱水和降颅压等常规治疗,并予以物理疗法、针灸、按摩、高压氧等综合治疗,治疗组患儿同时加用神经节苷脂2ml(20mg),加入5%葡萄糖注射液250m1中静脉滴注,每日1次,10d为1个疗程。停药20d后进入下一个疗程,连续治疗6个疗程。结果:与对照组相比,洽疗组治疗后发育商评分明显上升,头颅CT阳性率显著降低。治疗组总有效率为95%(40/42)与对照组的79%(33/42)相比差异有显著意义(P〈0.05)。结论:神经节苷脂治疗婴幼儿脑瘫的临床疗效确切。  相似文献

背景:干细胞局部移植可治疗神经系统疾病。 目的:观察脐血干细胞移植联合使用鼠神经生长因子及物理康复治疗小儿脑性瘫痪的临床效果。 方法:将90例小儿脑性瘫痪患儿根据是否接受干细胞治疗分为3组:脐血干细胞治疗组采用脐血干细胞+鼠神经生长因子+物理康复治疗,常规治疗组采用鼠神经生长因子+物理康复治疗,对照组未行有效治疗。 结果与结论:3组患儿入组前粗大运动功能测试量表各能区分值差异无显著性意义(P > 0.05)。治疗后3个月,脐血干细胞治疗组及常规治疗组粗大运动功能测试量表各能区中A、B、D能区分值较治疗前提高(P < 0.05),且脐血干细胞治疗组A、B能区分值均明显高于常规治疗组(P < 0.05),对照组粗大运动功能测试量表各能区分值无变化(P > 0.05)。表明脐血干细胞移植联合使用鼠神经生长因子、物理康复治疗治疗小儿脑瘫疗效优于鼠神经生长因子联合物理康复疗法。  相似文献

背景:干细胞具有修复和替代受损脑组织的潜力,被认为是一种新的治疗小儿脑性瘫痪的方法。 目的:用粗大运动功能测试量表88项及精细运动功能评估量表45项评价自体骨髓间充质干细胞移植对脑性瘫痪患儿运动功能的改善情况。 方法:对20例脑性瘫痪患儿进行自体骨髓间充质干细胞移植治疗,用粗大运动功能测试量表88项及精细运动功能评估量表45项对治疗前及治疗后1,3,6 个月时患儿的粗大及精细运动功能进行评估。 结果与结论:随访过程中有3例患儿失访。顺利完成研究的17例患儿治疗后 1,3,6个月的粗大运动功能测试量表88项总分及A(卧位与翻身)、B(坐位)、C(爬与跪)功能区得分,精细运动功能评估量表45项总分及B(上肢关节活动能力)、C(抓握能力)功能区得分均较移植前显著提高(P < 0.05),以治疗后1个月的提升速度最显著,且未见明显不良反应。提示自体骨髓间充质干细胞移植可改善脑性瘫痪患儿的粗大运动和精细运动功能,对治疗小儿脑性瘫痪是安全、有效、可行的。  相似文献

BACKGROUND:As propofol has a neuroprotective effect, and umbilical cord blood mesenchymal stem cells have a high differentiation potential, their combination will have a better therapeutic effect on cerebral ischemia-reperfusion injury. OBJECTIVE:To study the effect of propofol pretreatment combined with umbilical blood mesenchymal stem cell transplantation on cerebral ischemia-reperfusion injury in rats. METHODS:Sixty-three Sprague-Dawley rats were randomized into model, propofol, and combined group (n=21 per group). Rat models of cerebral ischemia-reperfusion injury were made using ligation of the middle cerebral artery occlusion in the three groups. Rats in the combined group were given 100 mg/kg propofol injection at 1 day before injury and injection of umbilical blood mesenchymal stem cells via the tail vein (0.5 mL, 2×109/L). RESULTS AND CONCLUSION:Compared with the model group, the neurological function was improved significantly in the propofol and combined group, especially in the latter one, presenting with a remarkable mitigation in brain injury and an increased level of survivn mRNA in the rat hippocampus. The content of serum malondialdehyde was lower but the activity of superoxide dismutase was higher in the combined group compared with the propofol group. These findings indicate that propofol pretreatment combined with umbilical blood mesenchymal stem cell transplantation has better therapeutic effects than propofol pretreamtnet alone for improving cerebral ischemia-reperfusion injury in rats.  相似文献

BACKGROUND:Sufentanil and propofol are both found to have good neuroprotective effects on neurological damage in clinical practice. OBJECTIVE:To investigate the effect of propofol combined with sufentanil in umbilical cord blood mesenchymal stem cells transplantation for the treatment of spinal cord injury. METHODS:Sixty-five Wistar rats were selected to make animal models of acute spinal cord injury using Allen’s method. Six hours after modeling, these rats were randomly assigned into combined group (injection of 2×107/L human umbilical cord blood mesenchymal stem cell suspension (0.5 mL) plus injection of 1.0-1.5 mg/kg propofol and 0.5 μg/kg sufentanil via the tail vein), stem cell group (injection of 2×107/L human umbilical cord blood mesenchymal stem cell suspension (0.5 mL) via the tail vein), or control group (injection of 30 μL of LDMEM containing 5% fetal bovine serum). S100β protein level in serum was detected in each group at 15 and 60 minutes after injection. Motor function of rat in each group was assessed by Basso Beattie Bresnahan (BBB) scoring and incline plane test at 1, 2, 4, 6, 8 weeks after modeling. Pathological changes of the spinal cord were observed at 4 weeks after modeling. Expression of vascular endothelial growth factor was detected using western blot assay at 1 and 2 weeks after modeling. RESULTS AND CONCLUSION:After 15 and 60 minutes of intervention, S100β protein level was lowest in the combined group followed by the stem cell and control groups (P < 0.05). At 2, 4, 6, 8 weeks after modeling, scores on the incline plane test and BBB were ranked as follows: combined group > stem cell group > model group (P < 0.05). At 4 weeks after modeling, severe damage to the spinal cord and few nerve fibers were found in the control group; spinal cord hyperplasia and a few of regenerated axons and PKH-26-positive stem cells appeared in the stem cell group; while in the combined group, there were a large amount of PKH-26-positive stem cells and nerve axon-like structures. At 1 and 2 weeks after modeling, the highest protein level of vascular endothelial growth factor was found in the combined group followed by the stem cell group and control group (P < 0.05). To conclude, these findings indicate that propofol and sufentanil in umbilical cord blood mesenchymal stem cell transplantation therapy can promote the recovery of hindlimb function after spinal cord injury, thereby promoting the functional recovery of rats from spinal cord injury.  相似文献

BACKGROUND:In recent years, it has been a hot topic that stem cell transplantation is used to improve cardiac insufficiency after acute myocardial infarction by inducing regeneration of cardiomyocytes in the infarction regions. OBJECTIVE:To observe the effect of rosuvastatin combined with umbilical cord blood mesenchymal stem cells transplantation on rat cardiac function after acute myocardial infarction. METHODS:Forty-five Sprague-Dawley rats were enrolled to prepare myocardial infarction models by ligaturing the left anterior descending coronary artery. Then they were equivalently divided into model group, transplantation group and combination group. At 7 days after modeling, rats in the combination group were given injection of 300 μL umbilical cord blood mesenchymal stem cells (15.0×108) via the tail vein and by gavage once a day for 28 days with 1 mg/kg rosuvastatin; rats in the transplantation group and model group were injected with 300 μL umbilical cord blood mesenchymal stem cell suspension through the tail veins or the same amount of LG-DMEM medium, respectively, followed by intragastrical administration of the same amount normal saline. At 5 weeks after modeling, indexes of cardiac function, level of plasma Lp-PLA2 and heat shock protein 70 in the infarction regions were detected by color Doppler ultrasound, enzyme-linked immunosorbent assay and western blot assay, respectively. In addition, pathological changes of myocardial tissues were observed using hematoxylin-eosin staining. RESULTS AND CONCLUSION:Left ventricular ejection fraction and left ventricular end-systolic pressure were significantly higher in the combination group than in the transplantation group as well as higher in the transplantation group than the model group (P < 0.05); compared with the transplantation group, left ventricular end-diastolic pressure was significantly decreased in the combination group, but significantly increased in the model group (P < 0.05); the number of cardiomyocytes in the infarction regions was significantly higher in the combination group than the other groups. Additionally, expression of heat shock protein 70 in the infarction regions was significantly increased in the combination group (P < 0.05). To conclude, rosuvastatin combined with umbilical cord blood mesenchymal stem cell transplantation can significantly improve rat cardiac function after myocardial infarction.  相似文献

BACKGROUND:With the depth understanding of mesenchymal stem cells, mesenchymal stem cells are found to exert a prominent effect on immune regulation and anti-inflammation. OBJECTIVE:To investigate the therapeutic effect of umbilical cord blood stem cell transplantation on endotoxin-induced hypertension in pregnant rats. METHODS:Twenty-four pregnant Sprague-Dawley rats were randomized into three groups with eight rats in each group: control, model and experimental groups. Endotoxin-induced hypertension models were made in the model and experimental groups. Meanwhile, rats were given intravenous injection of umbilical cord blood stem cell suspension (1 mL) in the experimental groups and the same volume of normal saline in the control and model groups. Therapeutic effects of umbilical cord blood stem cell transplantation were observed through detection of systolic blood pressure, urine protein level, serum white blood cell quantity and Ang II and ET-1 expression. RESULTS AND CONCLUSION:Compared with the control group, the systolic blood pressure, urine protein level and serum white blood cell quantity of rats were increased significantly in the model group, and over time, endotoxin continuously promoted these parameters in the model group. After cell transplantation, a significant reduction in systolic blood pressure, urine protein level and serum white blood cell quantity of rats was found in the experimental group compared with the model group (P < 0.05). After modeling, the expression levels of Ang II and ET-1 were decreased significantly, while these levels were increased significantly after cell transplantation (P < 0.05). These findings indicate that umbilical cord blood stem cell transplantation may have a certain therapeutic effect on gestational hypertension in rats, which may be realized by regulating the secretion of endothelial injury-related factors.  相似文献

BACKGROUND:Studies have shown that human umbilical cord mesenchymal stem cells can improve pulmonary ventilation function by reducing inflammations. OBJECTIVE:To observe the therapeutic effect of human umbilical cord mesenchymal stem cell transplantation on acute lung injury. METHODS:Thirty Sprague-Dawley rats were randomized into normal group, model group and experimental group. Rats in the latter two groups were used to establish animal models of acute lung injury by intratracheal instillation of lipopolysaccharide. One hour after modeling, rats in the experimental group were intratracheally administered human umbilical cord mesenchymal stem cell suspension (0.1 mL, 1×106 cells), and those in the other two groups were given normal saline in the same dose intratracheally. Twenty-four hours after treatment, the pathological changes of lung tissue were observed using hematoxylin-eosin staining; the wet and dry weight ratio of the lung tissue and the levels of serum interleukin-1 and interleukin-8 were detected. RESULTS AND CONCLUSION:Compared with the normal group, the wet and dry weight ratio of the lung tissue and the levels of serum interleukin-1 and interleukin-8 were significantly increased in the model group (P < 0.05), while compared with the model group, these levels were significantly decreased in the experimental group (P < 0.05). Hematoxylin-eosin staining results showed clear alveolar space structure with complete alveolar septum in the normal group. In the model group, the alveolar septum was markedly thickened, and there was visible pulmonary capillary hyperemia, edema, as well as a large amount of inflammatory cell infiltrations in the pulmonary capillaries and alveolar space. Edema fluid rich in proteins was observed in a part of the pulmonary alveoli, and an extensive transparent membrane formed in the alveolar space. In the experimental group, the alveolar structure was clear, but the alveolar septum became thickened, and red blood cells and a small amount of infiltrated inflammatory cells were leaked from the pulmonary interstitial tissue. In conclusion, human umbilical cord mesenchymal stem cell transplantation for treatment of acute lung injury can reduce inflammatory factor levels and alleviate lung injury.  相似文献

背景:药物预处理的延迟性保护作用成为近年来预处理领域的研究热点,舒芬太尼是一种非选择性阿片受体激动剂,有明显的心肌保护作用。目的:探讨舒芬太尼预处理联合脐血间充质干细胞移植对心肌梗死后损伤心肌的保护作用。方法:采用结扎左冠状动脉前降支的方式建立大鼠心肌缺血再灌注模型(缺血30 min,再灌注180 min),建模后随机分为缺血再灌注组,脐血间充质干细胞组,舒芬太尼+脐血间充质干细胞组,每组30只。缺血再灌注组于缺血再灌注前5 min经腹腔静脉注射1 mL生理盐水,脐血间充质干细胞组经腹腔静脉注射脐血间充质干细胞悬液1 m L,舒芬太尼+脐血间充质干细胞组在此基础上于冠脉阻断前10 min经腹腔静脉注射舒芬太尼10μg/kg,治疗后2周进行相关指标检测。结果与结论:(1)心肌梗死面积:舒芬太尼+脐血间充质干细胞组<脐血间充质干细胞组<缺血再灌注组,差异有显著性意义(P<0.05);(2)与缺血再灌注组比较,脐血间充质干细胞组血清肌酸激酶、乳酸脱氢酶及肌钙蛋白Ⅰ水平降低,一氧化氮水平增加,与脐血间充质干细胞组比较,舒芬太尼+脐血间充质干细胞组血清肌酸激酶、乳酸脱氢酶及肌钙蛋白I水平明显降低,一氧化氮水平增加更明显,差异有显著性意义(P<0.05);(3)Caspase-3蛋白表达:舒芬太尼+脐血间充质干细胞组<脐血间充质干细胞组<缺血再灌注组,差异有显著性意义(P<0.05);(4)血流动力学指标:与缺血再灌注组比较,脐血间充质干细胞组、舒芬太尼+脐血间充质干细胞组左心室舒张压升高,左心室舒张末压降低,差异有显著性意义(P<0.05);(5)与缺血再灌注组比较,脐血间充质干细胞组心肌组织病理学损伤程度减轻,舒芬太尼+脐血间充质干细胞组明显减轻;(6)结果表明,舒芬太尼预处理联合脐血间充质干细胞移植能够减轻大鼠心肌梗死程度,保护损伤的心肌。  相似文献
