目的 观察脑卒中后抑郁(post-stroke depression,PSD)大鼠海马原位增殖新生细胞的存活和分化.方法 采用左侧大脑中动脉阻塞(MCAO)联合慢性不可预见温和应激刺激(chronicmild stress,CMS)及孤养法建立PSD模型,将雄性SD大鼠分为假手术、脑卒中、CMS和PSD组.每组均为18只.采取免疫组织化学、荧光双标染色及共聚焦成像动态检测,比较各研究组大鼠左侧海马齿状回溴脱氧尿苷嘧啶(BrdU)及其与神经元特异性核蛋白(NeuN)或胶质纤维酸性蛋白(GFAP)共表达.结果 与脑卒中组(232.2±8.6、123.7±2.6、136.2±2.6)相比,PSD组大鼠左侧(伤侧)海马齿状回BrdU+细胞数在脑梗死后第21(156.2±2.5)、30(70.2±2.0)和45天(81.2±1.1)均明显减少(t=28.83、52.2、62.08,均P<0.01),但仍高于假手术组.与脑卒中组(79.3%±2.8%、87.7%±4.6%)相比,PSD组大鼠左侧(伤侧)海马齿状回BrdU+/NeuN+细胞比例在脑梗死后第30(69.0%±3.4%)和45天(78.3%±2.4%)均明显减少(t=5.871、4.403,均P<0.01).BrdU+/GFAP+细胞比例在脑梗死后30和45 d均明显增加(t=4.226、8.945,P<0.01).结论 PSD大鼠脑卒中后海马齿状回原位增殖的新生细胞存活降低,分化为神经元的比例下降,胶质细胞比例增加.  相似文献

目的 观察大鼠实验性脑出血后内源性神经前体细胞的增殖、迁移、分布和在出血灶周边的分化。方法 将成年SD大鼠随机分为正常组、假手术组和脑出血组;脑出血组大鼠通过立体定向术向脑内注入自体动脉血制成脑尾壳核出血模型,并按不同的再喂养时间(1、3、7、14及30 d)分为5个亚组。手术后腹腔注射5-溴脱氧核苷尿嘧啶标记新生的内源性神经前体细胞。采用免疫组化单标观察内源性神经前体细胞(BrdU阳性细胞)的增殖、迁移和分布,免疫荧光双标观察内源性神经前体细胞在出血灶周边的分化情况。结果 与正常组和假手术组相比,脑出血组大鼠的BrdU阳性细胞数显著增加,并在7~14d达高峰;BrdU阳性细胞主要分布于室管膜下层、海马齿状回、脉络丛、胼胝体腹侧、出血灶周边区、外侧隔核、斜角带、缰核和大脑皮层等处。免疫荧光双标显示在脑出血灶周边区可见BrdU/GFAP、BrdU/NSE及BrdU/Nestin三种双标阳性细胞;BrdU/Nestin双标阳性细胞随着脑出血后时间的推移逐渐减少,而BrdU/GFAP、BrdU/NF-200双标阳性细胞则增多。结论 脑出血可诱导内源性神经前体细胞增殖,并向出血灶周边区迁移,进一步分化出神经元和胶质细胞,这可能是脑出血后神经结构重塑和功能恢复的重要物质基础。  相似文献

目的研究不同程度的性发作对成年大鼠空间学习记忆的影响。方法采用氯化锂和匹罗卡品联合诱导大鼠不同程度的癫模型(轻型和重型)。于造模后第6d给所有大鼠腹腔注射5-溴脱氧尿苷嘧啶(BrdU+)标记海马齿状回增殖的内源性神经前体细胞;用免疫组化方法观察各组大鼠注射BrdU+后第1d和第28d齿状回BrdU+阳性细胞数以及第28d的BrdU+/神经元核性蛋白(NeuN+)阳性细胞数及分布情况;利用Morris水迷宫评价大鼠的学习记忆功能。结果与正常组及轻型组比较,在各个时间点重型组海马齿状回BrdU+细胞数均增加(P<0.05),28d时BrdU+/NeuN+细胞数相应增多,但其占BrdU+细胞数的比例明显下降(P<0.05)。28d时重型组大鼠的学习记忆功能较正常组及轻型组明显下降(P<0.05)。结论严重的癫发作造成大鼠对空间学习记忆功能的损害,可能与其刺激大鼠海马齿状回内源性神经前体细胞增殖水平,抑制其分化为新生的成熟神经元有关。  相似文献

目的研究卡马西平对成年癫大鼠海马齿状回内源性神经前体细胞增殖的影响。方法采用氯化锂和匹罗卡品联合诱导大鼠癫模型,将癫大鼠随机分为癫对照组和癫卡马西平组,正常大鼠随机分为正常对照组和正常卡马西平组。癫对照组和正常对照组给以蒸馏水灌胃,同时癫卡马西平组和正常卡马西平组给予卡马西平灌胃。于灌胃后第6d腹腔注射5-溴脱氧尿苷嘧啶(BrdU)标记海马齿状回的内源性神经前体细胞的增殖情况;用免疫组化方法观察各组大鼠在注射BrdU后第1d、第7d齿状回BrdU阳性细胞数量的表达。结果①注射BrdU后第1d,癫对照组大鼠海马齿状回BrdU阳性细胞数较正常对照组明显增加(P<0.01),癫卡马西平组大鼠海马齿状回BrdU阳性细胞数较癫对照组减少(P<0.05);②注射BrdU后第7d,癫对照组大鼠海马齿状回BrdU阳性细胞数较正常对照组明显增加(P<0.01),癫卡马西平组大鼠海马齿状回BrdU阳性细胞数较癫对照组明显减少(P<0.05)。结论卡马西平抑制癫大鼠海马齿状回内源性神经前体细胞增殖。  相似文献

神经干细胞移植促进脑出血大鼠神经功能恢复的实验研究   总被引:2,自引:0,他引:2  
目的探讨脑内定向移植神经干细胞(NSCs)治疗脑出血的可行性和机制。方法采用在成年大鼠中立体定向纹状体内注入新鲜自体血液制作实验性脑出血模型,术后1d采用立体定向将在体外扩增、纯化并经BrdU标记的神经干细胞移植入脑出血部位,通过神经功能缺损评分来观察移植后大鼠神经行为学改善情况,并通过双标GFAP、NeuN、BrdU免疫组化来检测移植入脑的NSCs。结果移植的NSCs迁移至血肿周围区,并分化为神经元(≈10%BrdU阳性细胞)和星形胶质细胞(≈75%);与对照组比较,NSCs移植组在移植后1周有更好的神经功能缺损评分,而且此疗效可维持长达8周。结论移植的NSCs可在脑出血的大鼠脑中存活、迁移,并促进功能恢复。立体定向移植神经干细胞可改善实验性脑出血大鼠的神经功能。  相似文献

目的观察胶质细胞源性神经营养因子(GDNF)对局灶性脑缺血大鼠皮质和尾壳核神经干细胞(NSCs)增殖分化以及nNOS阳性神经元分布的影响,探讨GDNF和NO对内源性NSCs增殖分化影响的作用机制。方法制作右侧局灶性脑缺血模型,左侧脑室注射GDNF,5-溴脱氧尿嘧啶(BrdU)标记DNA合成期(S期)细胞,免疫组化观察正常组、假手术组、脑缺血组、生理盐水组和GDNF组大鼠局灶性脑缺血90min后再灌注时间分别为3d、7d、14d、21d、28d的BrdU/nNOS、BrdU/NeuN、BrdU/GFAP双标阳性细胞。结果GDNF组较脑缺血组和生理盐水组行为学恢复明显;脑缺血后皮质和尾壳核BrdU和nNOS阳性细胞增多,BrdU/nNOS(38.23%±6.87%)、Br—dU/NeuN(52.38%±13.29%)、BrdU/GFAP(13.51%±8.78%)双标细胞阳性率明显增加,与其它组比较均有显著性差异(P〈0.05)。结论局灶性脑缺血激活内源性NSCs,GDNF可调节NO释放,促进NSCs增殖、分化。  相似文献

目的 观察学习记忆能力减退老年大鼠海马齿状回神经细胞的增殖。方法 Morris水迷宫筛选出学习记忆能力减退大鼠 (痴呆组 )与正常大鼠 (对照组 )。BrdU标记海马齿状回增殖细胞。TUNEL法标记DNA片段原位检测凋亡细胞。计数海马颗粒细胞层与海马门的BrdU阳性细胞与凋亡细胞数。结果 痴呆组与对照组大鼠相比 ,海马颗粒细胞层BrdU阳性细胞显著增加 (P <0 0 1) ,而凋亡细胞数无显著差异 (P >0 0 5 )。海马门BrdU阳性细胞数及凋亡细胞数均无显著差异 (P >0 0 5 )。结论 学习记忆能力减退的老年大鼠海马齿状回神经细胞增殖能力减低。  相似文献

目的 探讨丰富环境对颞叶癫(痫)大鼠齿状回新生细胞分化和存活的影响及其相关分子机制.方法 成年Wistar 大鼠随机分为4组:假手术组、丰富环境+假手术组、癫(痫)组、丰富环境+癫(痫)组,各组均n=15.大鼠侧脑室注射海人酸制作颞叶癫(痫)模型.丰富环境干预30 d后,应用免疫荧光技术观察大鼠海马齿状回的新生细胞分化和存活情况,用Western blot方法检测各组海马脑源性神经营养因子(BDNF)、cAMP应答元件结合蛋白(pCREB)、蛋白激酶A(PKA)表达水平.结果 丰富环境+假手术组、丰富环境+癫(痫)组齿状回新生细胞标记物(BrdU)和新生成熟神经细胞标记物(BrdU/NeuN)阳性细胞数分别多于假手术组、癫(痫)组(P<0.05),而新生星形胶质细胞BrdU/GFAP阳性细胞数无统计学意义,并且丰富环境+假手术组、丰富环境+癫(痫)组海马BDNF和pCREB蛋白表达水平分别高于假手术组、癫(痫)组(P<o.05),而PKA蛋白表达水平无增高.结论 丰富环境可能通过增强pCREB/BDNF通路促进成年颞叶癫(痫)大鼠海马齿状回的神经发生.  相似文献

目的 观察血管性认知障碍小鼠模型中,缺血性炎性损伤对室管膜下区及海马齿状回少突胶质细胞 再生分化的影响,为血管性认知障碍的缺血性炎症机制提出新的损伤途径。 方法 成年雄性CD1小鼠随机分为模型组和假手术组,每组24只,模型组采用双侧颈动脉反复缺 血再灌注法制备血管性认知障碍小鼠模型。造模后4~6 d连续腹腔注射5 -溴脱氧尿嘧啶核苷 (bromodeoxyuridine,BrdU)(150 mg/kg)标记新生细胞,分别于术后14 d和28 d每组随机取一半小鼠脑 组织进行脑切片免疫组化、免疫荧光双标共聚焦检测,标记脑组织室管膜下区和海马区的少突胶质 细胞、星形胶质细胞及神经元,观察新生少突胶质细胞增殖及分化情况,并观察星形胶质细胞的增 生活化情况。 结果 造模后14 d和28 d室管膜下区新生细胞(BrdU阳性细胞)在模型组较假手术组明显增加(P均 <0.001),造模28 d模型组新生神经元(BrdU/NeuN阳性细胞)较假手术组显著增加(P<0.001)。与假 手术组相比较,术后28 d模型组海马齿状回少突胶质细胞祖细胞显著增多(P<0.001);少突胶质细 胞前体细胞显著减少(P =0.006)。造模后28 d模型组海马齿状回新生星形胶质细胞(BrdU/GFAP阳性 细胞)较假手术组显著增加(P =0.015)。 结论 血管性认知障碍小鼠内源性新生细胞增殖区室管膜下区与海马齿状回区均存在新生细胞反 应性增生的情况。新生细胞区分化的主要细胞为星形胶质细胞,而少突胶质细胞分化障碍,可能是血 管性认知障碍患者影像学常见皮层下白质病变的重要原因。  相似文献

目的研究成年大鼠脑梗死后自体神经干细胞的原位增殖、分化及其可塑性。方法雄性Wistar大鼠共90只,分对照组(n=10)、脑梗死后1d组(n=16)、脑梗死后3d组(n=16)、脑梗死后7d组(n=16)、脑梗死后14d组(n=16)、脑梗死后28d组(n=16)。用免疫组织化学方法动态检测BrdU、GFAP、NeuN、PSA-NCAM的表达。BrdU确定神经干细胞的增殖,GFAP、NeuN确定神经干细胞的分化,PSA-NCAM确定神经干细胞的可塑性。结果与对照组相比,大鼠海马BrdU 细胞数在脑梗死后1d组开始增加,7d组达到高峰,28d组接近正常水平;BrdU /GFAP 细胞数在脑梗死前后无明显变化;BrdU /NeuN 细胞数在脑梗死后14d组开始增加,28d组最多;BrdU /PSA-NCAM 细胞数在脑梗死后7d组开始增加,14d组达到高峰,28d组开始下降,但仍高于对照组,大约占同期BrdU阳性细胞数60%。结论大鼠脑梗死激活自体神经干细胞原位增殖,大多数增殖的神经干细胞分化成神经元并且具有可塑性。  相似文献

Summary Beginning from the observation that Scots living in England have much higher rates of mental hospital admission than do the English, several hypotheses are proposed to account for this. Much of the excess in rates of mental illness is accounted for by those diagnosed as having alcohol-related disorders and behaviour and personality problems. The results of an examination of offical statistics in the two countries enabled some explanations to be offered. It was found that rates of admissions to mental hospitals are higher in Scotland than in England but not as high as those found among Scots migrants who have a much higher rate of readmission to hospitals than either of the other groups. In fact, if first admissions only are considered the rates of admission in Scotland are not only higher than rates for English natives but also higher than for Scottish migrants. It seems that Scots living in England are somewhat less likely to become mental patients than Scots in Scotland but that once they do achieve this status they are very much more likely to be readmitted on subsequent occasions. It was concluded that there might be two fairly distinct groups of migrants from Scotland to England who have different backgrounds, different reasons for migrating and different psychological characteristics. On the one hand there are stable, economically motivated migrants who move south for definite employment related reasons and who show few psychological symptoms. While on the other hand there is a group of migrants who perhaps have psychological problems and who move more in hope than expectation without definite prospects and who account for the high rates of mental hospital admission found in Scottish migrants.  相似文献

Prostacyclin release from rat isolated perfused hearts and from dog coronary circulation was studied by measuring immunoreactive 6-keto-PGF1 alpha (6-keto-PGF1a) in heart perfusate and in plasma obtained from the great cardiac vein respectively. Continuous infusion of arachidonic acid at constant concentration in isolated perfused hearts induced an increased prostacyclin release. This release showed a rapid peak within 10 min and a subsequent decrease. Low-flow ischemia induced an increased perfusate concentration of 6-keto-PGF1a but, considering the decreased flow, prostacyclin release was actually reduced. During the whole period of ischemia (60 min) prostacyclin release was constant. In open-chest anesthetized dogs 6-keto-PGF1a concentration in the great cardiac vein was increased after ligation of the left anterior descending coronary artery. A prolonged period of coronary occlusion (4.5 hours) resulted in a progressive rise of prostacyclin release. 6-keto-PGF1a determinations in the femoral vein and in the aorta did not show relevant variations during the observation period.  相似文献

Summary Sudanophilic lipid accumulation is a characteristic feature of periventricular leukomalacia (PVL) of infants. At least two types of lipid-containing cells have been identified, one being the macrophage, the other the pre-myelin glial cell. A third type of lipid-containing cell has been seen in two monkeys with spontaneous PVL. Electron microscopically this cell appears to be an astrocyte.
