Centaurea species are used for the treatment of various ailments in the popular medicine in some countries. This study was designed to examine antioxidant potentials and fatty acid profiles of five Centaurea species from Turkey flora. Antioxidant properties of methanolic extracts from these species were evaluated by six different methods: phosphomolybdenum assay, free radical scavenging assay, β-carotene/linoleic acid test system, metal chelating activity, ferric and cupric reducing power. Total phenolic and flavonoid concentrations of each extract were also determined using the Folin–Ciocalteu reagent and aluminum chloride. The results of these assay showed a significant antioxidant capacity in all researched extracts. Centaurea cheirolopha extract, with the highest amount of total phenolic and flavonoids, showed the highest antioxidant activities in all assay, except for metal chelating. Fatty acid profiles of these species were examined by GC–FID and 30 fatty acids were identified. Palmitic, linoleic, oleic, and linolenic acid were detected as the main components. The results of the study indicated that the Centaurea species can be considered as a source of new natural antioxidants and unsaturated fatty acids for food, cosmetic and pharmaceutical industries.  相似文献

The fatty acids of Agaricus essettei, Agaricus bitorquis and Agaricus bisporus were investigated by using GC and GC-MS. The dominant fatty acids were found to be linoleic (61.82-67.29%) and palmitic (12.67-14.71%) acids among the 13 fatty acids detected in the oils. Total unsaturation for the oils was calculated as 77.50%, 77.44%, and 79.72%, respectively. In vitro antioxidant, anticholinesterase and antimicrobial activities were also studied. The ethyl acetate extract of Agaricus bitorquis showed the highest activity in β-carotene-linoleic acid, DPPH and ABTS+ assays, while the hexane extract of Agaricus bisporus exhibited the best metal chelating activity. The ethyl acetate and hexane extract of Agaricus bitorquis and the hexane extract of Agaricus essettei showed meaningful butyrylcholinesterase activity being close to that of galantamine. The extracts were found to be effective on Gram (+) bacteria, especially against Micrococcus luteus, Micrococcus flavus, Bacillus subtilis and Bacillus cereus. In conclusion, Agaricus bitorquis and Agaricus essettei demonstrated higher iron content, and better antioxidant, anticholinesterase and antimicrobial activities than those of Agaricus bisporus commonly consumed mushroom. Hence, Agaricus species, particularly Agaricus bitorquis might be useful as antioxidant agents and moderate anticholinesterase agents, and their extracts will probably be used for development of dietary foods, food products and additives.  相似文献

This study is designed for the determination of metal concentrations, antioxidant activity potentials and total phenolics of Amanita caesarea, Clitocybe geotropa and Leucoagaricus pudicus. Concentrations of four heavy metals (Pb, Cd, Cr, Ni) and five minor elements (Zn, Fe, Mn, Cu, Co) are determined. In the case of A. caesarea, Cr and Ni concentrations are found in a high level. Concentrations of the metals are found to be within safe limits for C. geotropa. In β-carotene/linoleic acid test, L. pudicus showed the highest activity potential. In DPPH system, A. caesarea showed 79.4% scavenging ability. Additionally, reducing power and chelating capacity of the mushrooms increased with concentration. The strongest super-oxide anion scavenger was A. caesarea. In the case of total phenolics, L. pudicus found to have the highest content.  相似文献

The chemical composition of the essential oil from Gnaphlium affine was determined, and its antimicrobial and antioxidant activities were evaluated. Twenty-four compounds, representing 94.95% of the amount of total oil, were identified by gas chromatography-mass spectrometry (GC-MS) analysis. Main constituents of the essential oil were found to be eugenol (18.24%), linalool (10.62%), trans-caryophyllene (8.86%), α-terpineol (5.97%), p-cymene (5.75%), hexadecanoic acid (5.63%), γ-cadinene (4.98%), δ-cadinene (4.22%), α-humulene (3.22%), and (−)-β-elemene (3.15%). The essential oil revealed a remarkable antimicrobial effect against the tested food-borne microorganisms with the MIC and MBC values in the ranges of 0.2-1.56 μg/ml and 0.39-3.13 μg/ml, respectively. The essential oil showed a potent antioxidant activity in ABTS radical scavenging, lipid peroxidation and reducing power assay. It was suggested that the essential oil from G. affine may be a new potential source as natural antimicrobial and antioxidant agents applied in food systems.  相似文献

This study aimed to explore the antioxidant and DNA protection abilities of hydroalcoholic extracts from fruits of Anadenanthera colubrina (ACHE), Libidibia ferrea (LFHE) and Pityrocarpa moniliformis (PMHE). These extracts were tested by five antioxidant methods (phosphomolibdenium and reducing power assays; superoxide, hydrogen peroxide and nitric oxide scavenging) and DNA protection capacity. Total phenolic content was measured by Folin-Ciocalteu method. ACHE exhibited the highest phenolic content (578 mg/g GAE), followed by LFHE (460 mg/g GAE) and PMHE (448 mg/g GAE). In phosphomolibdenium assay, ACHE showed 24.81% of activity in relation to ascorbic acid, whereas LFHE and PMHE had 21.08% and 18.05%, respectively. These plants showed high ability to inhibit reactive species tested with IC50 values ranged from 10.66 to 14.37 μg/mL for superoxide radical; 26.05 to 45.43 μg/mL for hydrogen peroxide; 178.42 to 182.98 μg/mL for reducing power; and 199.2 to 283 μg/mL for nitric oxide. Furthermore, these extracts had capacity to break the DNA damage induced by hydroxyl radicals. The antioxidant activity of these plants is related with their higher phenolic content and show that they may be used as source of bioactive compounds, relevant to the maintenance of oxidative stability of the food matrix, cosmetics and/or pharmaceutical preparations.  相似文献

The essential oil of aerial parts of Salvia lanigera Poir. (Lamiaceae) growing wild in Cyprus was obtained by hydrodistillation and was analysed by GC and GC-MS. A total of 67 compounds, representing 93.6% of the oil, were identified, and the major components were showed to be thymol (12.1%), hexadecanoic acid (6.0%), carvacrol and α-thujone (5.7%). The essential oil was assayed for its antioxidant and antimicrobial activities. Antimicrobial activity of the oil, evaluated using the broth dilution method, resulted higher against Gram-positive bacteria than the other referenced strains tested. Antioxidant activity of the oil was evaluated by using DPPH and FRAP methods together with three antioxidant standards, l-ascorbic acid, tert-butyl-4-hydroxy toluene (BHT) and gallic acid. The activity of the sample in both methods was higher than that of all of standards used at the same dose.  相似文献

The objective of this study was to characterize the chemical composition of the essential oil and extracts of Ptychotis verticillata. The antioxidative activities of this species were also evaluated to suggest it as a new potential source of natural antioxidants. Analysis of the chemical composition of P. verticillata essential oil from Morocco was carried out using GC and GC-MS. The oil was dominated by phenolic compounds (48.0%) with carvacrol (44.6%) and thymol (3.4%) as the main compounds. Plant phenolics constitute one of the major groups of components that act as primary antioxidant free radical terminators. The amounts of total phenolics and flavonoids in the solvent extracts (diethyl ether and ethyl acetate) were determined spectrometrically. Furthermore, the antioxidant activities of the essential oil and extracts were determined using a DPPH test system. The DPPH scavenging activity of extracts increased in the order ethyl acetate > ascorbic acid > diethyl ether > essential oil. Finally, a relationship was observed between the antioxidant activity potential and total phenolic and flavonoid levels of the extract.  相似文献

In vitro acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibitory and antioxidant activities of the aqueous and ethanol extracts of the leaves, ripe fruits, and unripe fruits of Juniperus communis ssp. nana, Juniperus oxycedrus ssp. oxycedrus, Juniperus sabina, Juniperus foetidissima, and Juniperus excelsa were investigated in the present study. Cholinesterase inhibition of the extracts was screened using ELISA microplate reader. Antioxidant activity of the extracts was tested by 2,2-diphenyl-1-picrylhydrazyl (DPPH) and superoxide radical scavenging, ferrous ion-chelating, and ferric-reducing antioxidant power (FRAP) assays. Total phenol and flavonoid contents of the extracts were determined spectrophotometrically. The extracts had low or no inhibition towards AChE, whereas the leaf aqueous extract of J. foetidissima showed the highest BChE inhibition (93.94 ± 0.01%). The leaf extracts usually exerted higher antioxidant activity. We herein describe the first study on anticholinesterase and antioxidant activity by the methods of ferrous ion-chelating, superoxide radical scavenging, and ferric-reducing antioxidant power (FRAP) assays of the mentioned Juniperus species.  相似文献

The dichloromethane, ethyl acetate, ethanol, and aqueous extracts of Cyclotrichium niveum (CN) and Thymus praecox subsp. caucasicus var. caucasicus (TP), Echinacea purpurea (EPU), and E. pallida (EPA) along with the essential oils of CN and TP were assessed for their anti-acetylcholinesterase (AChE) and antioxidant activities. AChE inhibition was estimated using spectrophotometric method of Ellman. Antioxidant activity was evaluated by 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and ferrous ion-chelating power tests. Ferric-reducing antioxidant power (FRAP) of CN and TP were also tested. CN essential oil was found to contain isomenthone (56.21%) and pulegone (19.76%). The ethyl acetate (83.11–87.98%) and dichloromethane (73.45–84.02%) extracts of CN showed the highest AChE inhibition. The ethyl acetate and ethanol extracts of TP exerted significant DPPH scavenger effect. The water extracts of CN and TP and the chloroform extract of the aerial parts of EPU displayed the highest ferrous ion-chelating effect. The leaf and flower essential oils of TP had the best FRAP.  相似文献

The members of Scutellaria L. (Lamiaceae) is known to be rich particularly in flavonoids and among them, S. baicalensis has been recorded to be used for memory-enhancing purpose. Therefore, we initiated a study to screen the methanol extracts prepared from the aerial parts of 33 Turkish Scutellaria species for their acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibitory activities, which are the key enzymes taking place in pathogenesis of Alzheimer’s disease. Besides, the methanol extracts were tested in vitro against another enzyme, tyrosinase, which is associated with melanin hyperpigmentation. 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical scavenger effect, ferrous ion-chelating ability, and ferric-reducing antioxidant power (FRAP) of the ethyl acetate and methanol extracts were also determined. AChE, BChE, and tyrosinase inhibition of the extracts were performed on ELISA microplate reader by spectrophotometric method. The extracts showed weak inhibition against AChE and BChE, while the best tyrosinase inhibition was caused by the methanol extract of S. brevibracteata subsp. subvelutina. The extracts had a very high DDPH radical scavenging effect and moderate antioxidant activity in ferrous ion-chelating and FRAP tests.  相似文献

Numerous plant species worldwide including Palicourea marcgravii and Tanaecium bilabiatum in Brazil cause sudden death and are known to contain monofluoroacetate (MFA). Other species in Brazil including some species traditionally assigned to Mascagnia but now properly called Amorimia species and other Palicourea species are reported to cause sudden death in livestock and are suspected to contain MFA due to the similarity of clinical signs. In this study, an HPLC-APCI-MS method to detect and quantify MFA was developed and was used to investigate plant material from field collections and/or herbarium specimens of Mascagnia, Amorimia, and Palicourea species suspected of causing sudden death. MFA was detected in Amorimia amazonica, Amorimia camporum, Amorimia exotropica, Amorimia pubiflora, Amorimia rigida, and Amorimia septentrionalis as well as Palicourea aeneofusca. MFA concentrations differ greatly between Palicourea species and Amorimia species, which may explain the incidence of poisoning and the amount of plant material required to cause sudden death between these taxa.  相似文献

The antioxidant properties of different extracts of Halenia elliptica was investigated by employing various established in vitro systems. The results showed that various extracts possessed strong antioxidant activity in vitro, and the 70% methanol extract (ME) had the strongest antioxidant activity. Based on our in vitro results, ME was used for investigating the antioxidant properties of H. ellipticain vivo. The liver and kidney of CCl4-intoxicated animals exhibited a significant decrease in superoxide dismutase (SOD), catalase (CAT) and glutathione (GSH) levels. Additionally, these organs exhibited a significant increase in the level of malondialdehyde (MDA). These changes were significantly reversed, in a dose-dependent manner, after treatment with ME and the standard treatment Vitamin E. Thus, it may be concluded that the ME possesses potent antioxidant properties, and might be valuable natural source of antioxidants that could be applicable to both the medical and food industries.  相似文献

The high prevalence (14 of 24 isolates) of enniatin-producing isolates from Western Australian Fusarium species isolated from pasture legumes associated with sheep feed refusal and rat deaths, and the high toxicity of their crude extracts to brine shrimp (Artemia franciscana) from a previous study warranted further investigation of this class of mycotoxin. Crude extracts from Fusarium acuminatum, Fusarium avenaceum, Fusarium tricinctum and Fusarium sambucinum, along with enniatins A, A1, B and B1 purified from a Western Australian strain of F. acuminatum using semi-preparative HPLC, were bioassayed using brine shrimp. All Fusarium isolates produced both enniatins B and B1, except for F. tricinctum WAC 8019, and 11 of the 17 isolates produced enniatin A1. Overall, all of the F. avenaceum isolates produced high amounts of enniatins, in particular enniatin B. One isolate of F. acuminatum (WAC 5715) and of F. tricinctum (WAC 11486) also produced high amounts of both enniatins B and B1. Only F. acuminatum WAC 5715 produced enniatin A among the tested isolates. All four purified enniatins A, A1, B, B1, individually and in combination, caused brine shrimp toxicity after 6 h of exposure, implicating that this emerging class of mycotoxin as a cause of the acute toxicity to brine shrimp observed. The mixture of all four enniatins was the most toxic to brine shrimp compared to purified individual enniatins, where the relative toxicity order was B > B1 > A1 > A. Enniatin B was the individual most toxic enniatin with some bioactivity at 5 μg/mL and almost 100% brine shrimp death at 50 μg/mL after 24 h of exposure. This study is the first report to confirm the acute toxicity of enniatins A, A1, B and B1 to brine shrimp, and also highlights the need for further investigation of the potential toxicity of these cyclic hexadepsipeptides to animals and humans.  相似文献

Water-soluble crude polysaccharide from a brown alga Turbinaria ornata (TCP) was screened for its antioxidant and anti-inflammatory potential. The major functional groups of polysaccharide were analyzed by Fourier Transmission-Infra Red (FT-IR). In vitro free radical quenching and total antioxidant activity of TCP was investigated by 1, 1-diphenyl-2-picryl hydrazyl (DPPH), nitric oxide (NO) scavenging, lipid peroxidation (LPO) inhibition and ABTS radical assay. Evaluation of anti-inflammatory activity of TCP was performed using carrageenan-induced paw edema in rats and vascular permeability test in mice. Phytochemical analysis of TCP showed the presence of carbohydrates, proteins and polyphenols further, the FT-IR analysis of TCP showed the presence of functional groups of sugar moiety, uronic acids and sulfate groups. TCP showed maximum LPO, NO and DPPH inhibition of 78.04%, 38.82% and 80.21% at a concentration of 1000, 125 and 500 μg/ml respectively. Oral administration of TCP (2.5, 5, 10, 20 mg/kg) reduced the paw edema considerably (p < 0.05) in a dose dependent manner compared to carrageenan induced rats. Similarly, oral administration of TCP (3, 10, 30 mg/kg) evoked a significant (p < 0.05) dose dependent inhibitory effect on vascular permeability in mice. Altogether, these results suggest that the crude polysaccharide of T.ornata could be considered as a potential antioxidant and anti-inflammatory agent.  相似文献

The effect of extracts obtained from Teucrium ramosissimum leaves on genotoxicity and SOS response induced by aflatoxin B1 (0.5 μg/assay) as well as nitrofurantoin (5 μg/assay) was investigated in a bacterial assay system, i.e., the SOS chromotest with Escherichia coli PQ37. The T. ramosissimum tested extracts exhibited no genotoxicity either with or without the external S9 activation mixture. However, all the extracts, particularly the total oligomers flavonoids (TOF) extract significantly decreased the genotoxicity induced by aflatoxin B1 and nitrofurantoin. Antioxidant capacity of the tested extracts was evaluated using the enzymatic (xanthine/xanthine oxidase assay) (X/XOD) and the non-enzymatic (NBT/Riboflavine assay) systems. TOF extract was the most effective one in inhibiting both xanthine oxidase activity and NBT reduction. Our findings emphasize the potential of T. ramosissimum to prevent mutations and also its antioxidant effect.  相似文献

Methanol extract of Symplocos cochinchinensis S. Moore leaves was evaluated for its in vitro and in vivo antioxidant activity. The total phenolic content of the extract was 230 mg of gallic acid equivalents/g extract. The extract showed very good scavenging activity on 2,2-diphenyl-picrylhydrazyl (DPPH) (IC50 620.30 ± 0.14 μg/ml), hydroxyl (IC50 730.21 ± 1.05 μg/ml), nitric oxide (IC50 870.31 ± 0.19 μg/ml) radicals, as well as high reducing power. The extract also showed strong suppressive effect on lipid peroxidation. In in vivo study CCl4 induced oxidative stress produced significant increase in SGOT, SGPT and LDH levels along with reduction in liver SOD, CAT, GSH and GPx levels. Pre-treatment of rats with the extract (250 and 500 mg/kg) for 7 days showed significant reduction in the levels of SGOT, SGPT and LDH compared to CCl4 treated rats. SOD, CAT, GSH and GPx levels were increased significantly due to treatment with the extract. The activity of the extract was comparable to the standard drug, silymarin (25 mg/kg). The results suggest that the leaves of S. cochinchinensis are a source of natural antioxidants.  相似文献

Hydromethanolic extracts from 18 Algerian medicinal plants were screened for their phenolic contents and radical scavenging activities. The phenolic extract of Cleome arabica (Capparaceae) was found to be the most active one. Purification of this extract by semi-preparative high performance liquid chromatography led to the isolation and identification of new steroid derivative. The structure of the active principle is proposed as (17-(4-hydroxy-1,5-dimethylhexyl)-2,3,7-(acetyloxy) gona-1,3,5(10)-trien-15-ol). Compared to six other standard antioxidants which were ascorbic acid, α-tocopherol, Trolox, (+) catechin, p-coumaric acid and gallic acid, the isolated compound was found to be significantly more active in the radical scavenging assay using 2,2-diphenyl-1-picrylhydrazyl (DPPH). Similar results were obtained in the hemolysis assay. The antioxidant capacities of the methanolic extract of C. arabica and its principle compound indicate that this plant may be an important source of chemopreventive and chemotherapeutic natural products activity.  相似文献

Roses are one of the most important groups of ornamental plants and their fruits and flowers are used in a wide variety of food, nutritional products and different traditional medicines. The antioxidant activity of methanolic extracts from fresh flowers of three rose species (Rosa damascena, Rosa bourboniana and Rosa brunonii) was evaluated by 1,1-diphenyl-2-picryl hydrazyl (DPPH) free-radical method. The ability to scavenge DPPH radical was measured by the discoloration of the solution. The methanolic extract from R. brunonii exhibited maximum free-radical-scavenging activity (64.5 ± 0.38%) followed by R. bourboniana (51.8 ± 0.46%) and R. damascena (43.6 ± 0.25%) at 100 μg/ml. Simultaneously, ultra-performance liquid chromatography coupled with electrospray ionization-quadrupole time-of-flight mass spectrometry (UPLC-ESI-QTOF-MS) was used to study phenolic composition in the methanolic extracts from the fresh flowers of rose species. The phenolic constituents were further investigated by direct infusion-ESI-QTOF-MS/MS in negative ion mode. Characteristic Electrospray ionization tandem mass spectrometry (ESI-MS/MS) spectra with other diagnostic fragment ions generated by retro Diels–Alder (RDA) fragmentation pathways were recorded for the flavonoids. Distinct similarities were observed in the relative distribution of polyphenolic compounds among the three species. The dominance of quercetin, kaempferol and their glycosides was observed in all the three species.  相似文献

Melissa officinalis (lemon balm) infusions are used worldwide for digestive, analgesic and other pharmaceutical applications. Herein, the nutraceuticals production and antioxidant potential in garden cultivated, in vitro cultured and two commercial samples (bags and granulated) of lemon balm was compared. The profile of in vitro cultured lemon balm is closer of garden cultivated sample than of both commercial samples (bag or granulate). It presented the highest levels of proteins and ash, and the lowest energetic value. The most favorable n6/n3 ration, as also the highest PUFA (mostly α-linolenic acid), tocopherols (including α-, γ- and δ-isoforms) and ascorbic acid contents were also observed in this sample. Nevertheless, it was the commercial bag lemon balm that gave the highest antioxidant activity and the highest levels of phenolics and flavonoids. As far as we kwon, this is the first comparison of nutraceuticals and antioxidant potential of cultivated, in vitro cultured and commercial lemon balm samples. Moreover, it proved that in vitro culture might be used to stimulate vitamins production.  相似文献

The essential oil of Artemisia campestris and the ethanol-water, hexane and water extracts of A. campestris and Thymelaea hirsuta collected in southern of Tunisia were investigated for their antioxidant (DPPH, ABTS and beta-carotene methods) and antitumor growth inhibition of human colon cancer HT-29 cells using MTT test activities.All the A. campestris extracts tested at high concentrations (100 μg/ml) showed activity ranging from 19.5% for essential oil to 64.4% of negative control growth for infusion extract, except the hexane extract. With T. hirsuta, all the extracts tested (hexane and ethanol-water), except the infusion extract, also exhibited antitumor activity (58.2% and 65.5% of control growth respectively).The ethanol-water and infusion extracts of A. campestris showed higher antioxidant activity, polyphenol and flavonoid contents than those of T. hirsuta. These results show that there is a positive correlation between the antitumor activity and the antioxidant activity, and of these two activities and with the levels of polyphenols and flavonoids.The essential oil and the other extracts of A. campestris, which exhibited significant antitumor activity against the HT-29 cells deserve further research into the chemoprevention and treatment of colon cancer.  相似文献
