用冻干浓缩液的圆周免疫扩散法观察了牛胰岛素对 Hep G2 细胞分泌载脂蛋白apo AⅠ、apo AⅡ、apo B100、apo CⅢ及apo E的影响。结果发现,当培养液中的胰岛素浓度为1μg/m l时,apo AⅠ、apo E的分泌无明显改变,apo AⅡ的分泌有降低的趋势,apo CⅢ的分泌有增加的趋势,apo B100 的分泌降低21.2% ( P< 0.01); 当培养液中胰岛素的浓度为 10 μg/m l时,apo AⅠ的分泌减少6.9% ( P< 0.05),apo AⅡ的分泌减少 44.3% ( P< 001),apo B100 的分泌减少29.3% ( P< 0.01),而apo CⅢ的分泌增加51.8% ( P< 0.01),apo E的分泌无明显改变。该研究结果支持刘秉文提出的中国人内源性高甘油三酯血症发病机理的设想中的部分环节。  相似文献

为探讨成都地区汉族人群载脂蛋白(apo)E多态性情况及其与血脂、apo 之间的关系,作者随机抽取了成都地区健康体检的362名汉族成人,采用等电聚焦免疫印迹分析法对其血清apoE表型进行了测定,同时对受检者空腹血TG、TC及apoAⅠ,B100,CⅡ,CⅢ,E进行了全面分析。结果显示:成都地区汉族人群apoE表型分布频率为:E3/3 72.93% ,E2/3 12.98% ,E3/4 11.33% ,E2/4 1.38% ,E4/4 1.38% ,E2/2未检测到。ApoE等位基因频率为:ε3 0.8508,ε2 0.0718,ε4 0.0774。362名汉族成人不同apoE表型的血脂和载脂蛋白水平显示:与apoE3 (E3/3)或apoE4(E3/4 + E4/4 )组比较,apoE2 (E2/3 + E2/2 )组血清TC及apoB100 明显降低(P< 0.05),apoE水平明显升高(P<0.001),TG、apoAⅠ、apoCⅡ、apoCⅢ水平无统计学差异;与apoE3 组比较,apoE4 组TG、TC、apoAⅠ、B100、CⅡ、CⅢ和E均无统计学差异。  相似文献

用直线相关及多元逐步回归的方法对20例正常人及20例内源性高甘油三酯血症患者血浆、VLDL、LDL及HDL的脂质与载脂蛋白的关系进行了分析。单因素分析结果表明:TG及vLDL、TG分别与血浆apoCⅡ、CⅢ、E,LDLapoB100,VLDL中apoCⅡ、CⅢ、E.B100呈正相关,与HDL3poAⅠ呈负相关;HDL-C与血浆apoAⅠ、HDL中apoAⅠ、CⅢ呈正相关,与血浆apoCⅢ、E.LDLapoB100及VLDL中apoB100、CⅡ、CⅢ、E呈负相关;TC与apoB100、LDLapoB100呈正相关。多因素分析结果表明:有7种因素对TG有显著性影响,其中VLDLapoCⅢ,血浆apoCⅢ、E及apoB100/AⅠ与TG呈正相关,HDL中apoCⅡ.E及CⅡ/CⅢ与TG呈负相关;只有血浆apoCⅢ、VLDLapoCⅢ、LDLapoB100三种因素对VLDLTG有显著性影响;对HDL-C有显著性影响的因素是:apoAⅠ、HDLapoAⅠ及HDLanoCⅡ呈正相关,VLDLapoE及HDLapoAⅡ呈负相关;apoB100及apoCⅡ与TC呈正相关。文内还对所得结果进行了讨论。  相似文献

为研究血清apoB100与血清TG、TC、HDL-C、LDL-C及apoAⅠ、AⅡ、CⅡ、CⅢ和E的关系,对438例40~70岁正常男性中老年人空腹12~14小时的血清TG、TC、HDL-C、LDL-C及apoB100、AⅠ、AⅡ、CⅡ、CⅢ和E的水平进行了全面的分析。结果表明,血清apoB100水平为742.5±165.8mg/L,且有随年龄上升的趋势。其中5组及6组的apoB100水平比1、2、3、4组均显著升高(P<0.05)。血清脂质水平(x±s,mmol/L):TG为1.39±0.45、TC为4.79±0.82、HDL-C为1.62±0.40、LDL-C为2.65±0.81,且5组及6组的TG、TC、LDL-C水平均比1、2、3、4组显著升高(P<0.01)。相关分析表明:血清apoB100与血清TG、TC、LDL-C及apoCⅡ、CⅢ和E均呈显著正相关(P<0.001);与HDL-C呈显著负相关(P<0.01)。以上结果提示:中老年男性,尤其是60~70岁年龄段的血清apoB100水平与血清脂质及apoCⅡ、CⅢ和E有密切的关系。  相似文献

观测22例正常人及33例非透析治疗尿毒症患者血清各种载脂蛋白(apo)变化。结果显示,尿毒症患者apoAⅠ、apoAⅡ低于正常;apoB100较正常人高;apoCⅡ、apoCⅢ无明显变化;AI/B100、AⅠ/CⅢ值较正常人低;CⅢ/CⅡ较正常人高。提示非透析尿毒症患者血清apo的变化主要表现为AⅠ、AⅡ、B100以及AⅠ/B100、AⅠ/CⅢ、CⅢ/CⅡ比率的改变。  相似文献

本文介绍测定人血清载脂蛋白AⅡ(apoAⅡ)的酶联免疫法(ELISA)。特异抗人apoAⅡ血清由免疫山羊获得,按过碘酸钠法制备辣根过氧化物酶羊抗人apoAⅡγ球蛋白交联物。用羊抗人apoAⅡγ球蛋白(20μg/ml)包被聚苯乙烯酶标板。结果表明:apoAⅡ抗血清与apoAⅠ、apoCs,anoB100(LDL)以及人血清白蛋白无交叉反应,最小检测量为50Ong,标准曲线工作范围为0.25~8.00mg/dl,板内及板间变异系数分别为5.0%~9.2%及6.8%~11.7%,回收率106.0±2.1%(n=4)。应用本法测得41例血脂正常者apoAⅡ含量为24.4±5.9mg/dl,与用RID法测得的结果26.7±4.6mg/dl相比(r=0.8000,P<0.001),无显著性差异。本法特异性强,灵敏度高,简便快速。  相似文献

内源性高甘油三酯血症(HTG)常伴血浆HDL-C含量下降及HDL组成异常。为了研究HTG患者血浆HDL亚类的改变,作者建立了密度梯度聚丙烯酰胺梯度凝胶电泳法(GGE),对25例HTG患者(血浆TG>2.26mmol/L,TC<6.72mmol/L)及25例年龄、性别匹配的血脂正常者(血浆TG<1.69mmol/L,TC<5.17mmol/L)的HDL亚类——HDL2a、HDL2b、HDL3a、HDL3b和HDL3c进行了分析,同时对血浆及HDL中脂质及载脂蛋白(apo)AⅠ、AⅡ、B100、CⅡ、CⅢ及E进行了测定。结果发现,HTG患者空腹血浆TG、apoB100、apoCⅡ、apcCⅢ及apoE显著增加(P<0.001),HDL-C及apoAⅠ显著降低(P<0.001及P<0.05);患者血浆HDL组成异常,HDL中TG含量显著增加(P<0.001),TC、apoAⅠ、apoCⅡ、apoCⅢ及apoE含量下降(P<0.01);患者血浆HDL亚类含量改变,HDL2b显著降低(P<0.02),HDL2a及HDL3a增加(P<0.05),且随血浆TG含量的升高而升高,HDL3b及HDL3c则未见改变  相似文献

对20例正常人及20例内源性高甘油三酯血症(HTG)患者的血浆及血浆VLDL,LDL、HDL的TG、FC、CE、PL以及apoAⅠ、AⅡ、B100、CⅡ、CⅢ、E的组成及含量进行了全面分析。结果表明:内源性HTG患者空腹血浆TG、apoCⅡ、CⅡ及E显著增加,HDL-C及apoAⅠ显著降低,而且其血浆VLDL、LDL、HDL的组成明显异常,即VLDLTC、CE、FC、PL以及VLDLapoB100、CⅡ、CⅡ、E均显著增加,HDLTG增加,CE及apoAⅠ、CⅡ、CⅢ和E显著降低LDLTG及apoB100增加,文内还对载脂蛋白的功能及其与脂质代谢的关系进行了讨论.探讨了内源性HTG患者脂质及载脂蛋白异常的一些原因,为揭示HTG的发生机理提供了有价值的资料。  相似文献

OBJECTIVE: This study inquired into the role of retinoic acid in lipoprotein metabolism. METHODS: We observed the effect of retinoic acid on the secretion of apolipoproteins A I, A II, C III, B100 and E by cultured HepG2 cells. The apolipoprotiens contents in culture media were measured by radioimmunodiffusion assay (RID) kits developed by our research laboratory. 20-fold lyophilizely condensed culture media were used for the assays. RESULTS: Retinoic acid increased the secretion of apoA I, B100, C III and A II, and it inhibited the secretion of apoE. The effect of retinoic acid was strengthened in a dose-dependent manner. When the concentration of retinoic acid in cultured media was 2 x 10(-4) mol/l, the secretion of apoA I, A II, B100 and C III increased by 14.3% (P < 0.01), 23.8% (P < 0.05), 16.1% (P < 0.01) and 47.6% (P < 0.01) respectively, and the secretion of apoE decreased by 37.2% (P < 0.01). CONCLUSION: These findings indicate that retinoic acid does not have a general effect on apolipoprotein secretion in HepG2 cells.  相似文献

OBJECTIVE: To gain an insight into the relation of plasma glucose levels to lipids and apolipoproteins levels. METHODS: Fasting 12-14 h blood glucose (BG), high density lipoprotein cholestrol(LDL-C), apolipoproteins(apo) A I, A II, B100, C II, C III and E levels were measured in 735 men aged 40-70 in Chengdu area. The subjects studied were divided into three groups by the plasma glucose levels: 1. normal group (S1), 2. decreased glucose tolerance group (S2), 3. diabetes mellitus (DM) group (S3). RESULTS: Among 735 cases studied, groups S2 and S3 had more hyperlipidemia than group S1 (64%, 45% and 32.2% respectively). The fasting serum mean TG levels in groups S2 (3.62 +/- 2.52 mmol/L) and S3 (4.45 +/- 3.82 mmol/L) group were significantly higher than that in group S1 (1.92 +/- 1.39 mmol/L) (P < 0.01 and P < 0.01), and the mean TG levels in groups S2 (5.68 +/- 1.15 mmol/L) and S3 (5.68 +/- 1.12 mmol/L) were higher than that in group S1 (5.13 +/- 1.06 mmol/L) (P < 0.05, P < 0.05). The serum apo B100, C II, C III and E levels in groups S2 and S3 were significantly higher as compared with that in the normal group (P < 0.01, P < 0.05). The correlation analysis indicated that there was a positive correlation of BG with serum TG in groups S1, S2 and S3 whose correlation coefficients (r) were 0.1800, 0.1436 and 0.3212 respectively (P < 0.001); and a negative correlation with HDL-C levels in groups S2 and S3 (r = -0.2534 and -0.1477, P < 0.001). The plasma glucose levels were positively correlated with apo B100, C III and E levels in groups S1, S2 and S3, P < 0.001), with apo C II in groups S1 and S2 (P < 0.001, P < 0.01); and negatively correlate to apoA I only in group S2 (r = -0.1182, P < 0.01). CONCLUSION: The results suggest that serum TG, TC, HDL-C, apoB100, C II, C III and E levels be the main factors that related to plasma glucose levels.  相似文献

目的 探讨视黄酸在脂蛋白代谢中的作用。方法 采用冻干浓缩培养液载脂蛋白 RID测定法 ,分别观察 4种不同浓度的视黄酸对培养的人肝癌细胞系 Hep G2细胞载脂蛋白 ( apo) A 、A 、C 、B10 0及 E分泌的影响。结果 视黄酸对 Hep G2细胞 apo A 、A 、B10 0及 C 的分泌均有促进作用 ,对 apo E的分泌则有抑制作用。在本研究条件下 ,当视黄酸的浓度为 2× 10 - 4 m ol/ L时 ,其对 Hep G2细胞 apo分泌的影响作用最大 ,此时apo A 、A 、B10 0和 C 的分泌分别增加 14.3% ( P<0 .0 1)、2 3.8% ( P<0 .0 5 )、16 .1% ( P<0 .0 1)、47.6 % ( P<0 .0 1) ,而 apo E的分泌减少 37.2 % ( P<0 .0 1)。结论 视黄酸对 Hep G2细胞 apo A 、A 、C 、B10 0及 E的分泌有不同的影响  相似文献

This study was conducted in 438 normal men aged 40-70. The age groups were: 40-44, 45-49, 50-54, 55-59, 60-64, and 65-70. The results showed that the serum apolipoproteins levels (mean+/- s, mg/L) were: B100 levels 742.5 +/- 165.8, A I 1301.2 +/- 219.9, A II 299.4 +/- 49.0, C II 42.7 +/- 21.4, C III 113.3 +/- 34.0 and E 38.7 +/- 8.2. The levels of serum apoB100 increased with age. The serum apoB100 levels in the 5th and 6th age groups were significantly higher as compared with the groups 1, 2, 3 and 4 (P < 0.05 and P < 0.05). The fasting serum mean TG (1.39 +/- 0.45 mmol/L), TC(4.79 +/- 0.82 mmol/L) and LDC-C(1.62 +/- 0.40 mmol/L levels in the groups 5 and 6 were significantly higher as compared with the groups 1, 2, 3 and 4 (P < 0.01). Serum apoC II, C III and E levels were significantly increased in the groups 5, 6 (P < 0.05). The correlation analysis indicated that there was a positive correlation of apoB100 with serum TG, TC, LDL-C, apoC II, C III and E respectively (P < 0.01); and a negative correlation with HDL-C levels (r = 0.1312); and apoB100 correlated negatively with apoA I (r = -0.0706). The results suggest that serum TG, TC, LDL-C, apoC II, C III and E are the main factors related with the serum apoB100 levels.  相似文献

The effects of gemfibrozil on apolipoproteins C II and C III (apo C II, C III) were observed in 20 NIDDM hyperlipidemic patients. All of the patients continued their anti-diabetic treatment, gemfibrozil 900 mg/day for 4 weeks. The results revealed no significant change in fasting plasma glucose (FPG) and HbA1 before and after the study. But after the treatment with gemfibrozil, the following parameters changed significantly: total cholesterol (TC) decreased by 18.64% (P less than 0.01), total triglyceride (TG) decreased by 65.05% (P less than 0.001), VLDL-C decreased by 63.19% (P less than 0.001), HDL-C increased by 44.23% (P less than 0.001), apo C III decreased by 31.38% (P less than 0.02), and the ratio of apo C III/C II reduced by 35.49% (P less than 0.01). These findings suggest that gemfibrozil has excellent effect on decreasing apo C III and the ratio of apo C III/C II, thus facilitates the metabolism of chylomicron and decreases TG level in hyperlipidemic diabetic patients.  相似文献

This paper deals with the correlation between insulin release curves and syndrome types of TCM in 142 cases of diabetes. The syndromes were divided into 3 types: I, Qi-Yin deficiency, 94 cases; II, Yin deficiency with interior heat overabundance, 36 cases; and III, deficiency of both Yin and Yang, 12 cases. The results showed that fasting for 60 min, the postprandial (M +/- SD) of serum insulin were 18.27 +/- 1.10 mu/L and 45.21 +/- 2.82 mu/L in type I 25.47 +/- 1.88 mu/L and 60.53 +/- 0.25 mu/L in Type II; 8.29 +/- 0.93 mu/L and 19.20 +/- 1.67 mu/L in type III respectively. It was obvious that serum insulin level was Type II greater than Type I greater than Type III. But there was no difference between the aged and the pre-aged with diabetes (P greater than 0.05); even though the former was lower than the latter. According to the analysis of insulin release curve types (insulin secretion level and peak time), there were 39 cases (41.5%) of the low secretion type, 7 cases (7.5%) of the high secretion type among the 94 cases of Type I. Of the 36 cases of Type II, there were 7 cases (19.4%) of high secretion, more than that of Type I, and the highest value reached 155-106 mu/L. The 12 cases of Type III are all of the lower secretion type, with the fasting mean value being 47.5 mu/L and 10.93 mu/L and the postprandial highest value 21.69 mu/L and 20.73 mu/L, much lower than those of Types I and II. The results also showed that correlation between insulin secretion level and blood glucose value was significantly negative. The paper also discussed the mechanism of insulin secretion condition of 3 syndrome types according to the theory of TCM.  相似文献

目的 比较不同硬膜外给药方式对子宫切除手术患者术后镇痛效果的影响。方法 对75例择期子宫全切手术的患者 ,随机单盲分为 3组 ,每组 2 5例 ;Ⅰ组为空白对照组 ,切皮前 10min于硬膜外中加用生理盐水 10ml。Ⅱ组切皮前 10min于硬膜外中加用 2 %利多卡因 10ml(其中每ml含芬太尼 2 5 μg) ,后于硬膜外中持续泵入罗哌卡因和芬太尼混合液 (罗哌卡因 0 12 5 % ,芬太尼 2 μg/ml)至手术结束 (速度 9ml/h)。Ⅲ组切皮前 10min于硬膜外中加用 2 %利多卡因 10ml(其中每ml含芬太尼 2 5 μg ,氟哌啶 0 15mg) ,后于硬膜外中持续泵入罗哌卡因和芬太尼的混合液 (罗哌卡因 0 12 5 % ,芬太尼 2 μg/ml) ,至手术结束 (速度 9ml/h)。所有患者均于气管插管全麻下手术。 3组患者均于手术结束后自控硬膜外镇痛 (PCEA) ,微电脑泵为BaxterAP Ⅱ。自控镇痛液为罗哌卡因和芬太尼的混合液 (罗哌卡因浓度 0 12 5 % ,芬太尼 2 μg/ml) ,预充 5ml,背景剂量 3ml/h ,单次自控镇痛 (PCA)剂量 2ml/次 ,锁定时间 10min ,限量 16ml/h ,术后镇痛维持 4 8h。结果 第 1个 2 4h自控镇痛的视觉模拟(VAS)评分静息时Ⅰ组为 2 2分± 1 5分 ,Ⅱ组为 1 4分± 0 78分 ,Ⅲ组为 1 1分± 1 1分 ;Ⅰ组与Ⅱ组比较P <0 0 5 ,I组与Ⅲ组比较P <0 0 1  相似文献

OBJECTIVE: To investigate the changes of the apolipoproteins(apoA I, A II, B100, C II, C III, E) in patients with type 2 diabetes mellitus. METHODS: The levels of fasting plasma glucose(FBG), insulin, TG, TC, apoA I, A II, B100, C II, C III, E were all measured in 127 non-diabetic subjects and 143 type 2 diabetic patients (20 associated with coronary heart disease (CHD) and 66 associated with hypertriglyceridemia(HTG), 55 associated with hypercholesterolemia). RESULTS: In male type 2 diabetic patients, the levels of FBG, WHR and apoC II were significantly higher (P < 0.05) and apoA I, A II levels were significantly lower (P < 0.05) than those in male non-diabetic subjects. In female type 2 diabetic patients, the levels of FBG, BMI, WHR, TG significantly elevated while HDL-C, apoA I, A II levels significantly decreased as compared with those in female non-diabetic subjects. In type 2 diabetic group, the levels of WHR, FBG and TG in HTG patients were elevated significantly as compared with those without HTG, and the levels of HDL-C, apoA I and apoA II were decreased; the levels of WHR, TG, TC, apoB100, C II, C III, E in patients with HTC were significantly higher than those whose cholesterol levels were normal. In patients with CHD, the levels of fasting insulin, apoB100, apoC II and apoE were significantly higher than those in patients without CHD, and the levels of HDL-C and apoA II were decreased significantly. CONCLUSION: Abnormal changes of apo(s) in type 2 diabetes mellitus may be a cause of type 2 diabetes associated with HTG and CHD.  相似文献

This investigation was conducted to observe the frequency distribution of apoE phenotypes and alleles and to explore the relationship between apoE polymorphism and plasma lipids or apolipoproteins in Chinese population. ApoE phenotypes were assayed by isoelectric focusing and immunoblotting with serum. Serum lipids and apoA I, B100, C II, C III, E were determined in a random subset of 362 subjects including 268 males and 94 females with a mean age of 43.7 +/- 12.3 yrs from a population of Han Nationality in Chengdu area. The results showed that the frequencies of apoE phenotypes and alleles were: E3/3 72.93%, E2/3 12.98%, E3/4 11.33%, E2/4 1.38%, E4/4 1.38%, E2/2 0.00%; epsilon 3 0.8508, epsilon 2 0.0718, epsilon 4 0.0774. The results also showed that the apo E2(E2/3 + E2/2) group had lower levels of serum TC and apoB100 (P < 0.05) and a higher level of serum apoE (P < 0.001) when compared with the apoE3(E3/3) or apoE4(E3/4 + E4/4) group. No significant difference was observed in TG, apoA I, apoC II, and apoC III levels among the apoE2, E3 and E4 groups (P > 0.05).  相似文献
