目的观察钙结合蛋白S100A2在原发性肝癌中的表达及与肿瘤细胞增殖的关系,探讨两者对肝癌患者预后的影响。方法采用免疫组织化学链霉菌抗生物素蛋白-过氧化酶(SP)法检测62例肝癌标本中S100A2的表达,同时用计算机图象分析技术对其DI值(DNA含量指数)进行检测。结果高/中分化组,无门脉/胆管浸润或淋巴结/远处转移的病例中,S100A2阳性表达率分别为77.27%、76.92%、80.00%;DI值分别为1.68±0.20、1.73±0.23、1.76±0.23;但随着肝癌分化程度的降低,出现门肘胆管浸润或发生淋巴结/远处转移,S100A2阳性表达率降低(50.00%、47.22%、50.00%)(Χ^2=4.39、5.53、5.07,P均〈0.05),而DI值明显升高(1.93±0.29、1.99±0.31、2.01±0.33)(t=3.612、3.794、3.379,P均〈0.01)。并且在短/中期生存患者中,S100A2阳性表达率(50.00%)明显低于长期生存者(100%)(Χ^2=10.05,P〈0.01),而DI值(1.88±0.25)却明显高于长期生存者(1.69±0.21)(t=2.606,P〈0.05)。S100A2的表达与DI值呈明显负相关(t=3.023,P〈0.05)。结论S100A2蛋白表达降低和高的DI值提示肝癌的低分化、高侵袭转移能力及预后不良,两者的联合检测对判断肝癌的恶性程度和预后具有重要的临床意义。  相似文献

目的 探讨血清中MMP-9、S100A12及CRP水平与强直性脊柱炎(ankylosing spondylitis, AS)之间的联系,为AS的诊治提供临床依据。方法 选取2017年6月~2019年10月在我院接受治疗的122例AS患者为观察组,选取同时期122例健康体检者为对照组,对受试者血清中的MMP-9、S100A12及CRP水平进行测定并进行比较。结果 观察组患者血清中MMP-9、S100A12及CRP水平明显高于对照组(P<0.05);BASDAI≥5分组中的患者血清MMP-9、S100A12及CRP水平明显偏高(P<0.05);BASFI≥5分组中的患者血清MMP-9、S100A12及CRP水平明显偏高(P<0.05)。结论 AS患者血清中MMP-9、S100A12及CRP均异常表达,且与病情联系密切,可依据血清中相关指标变化对病情进行评估。  相似文献

目的观察人骨肉瘤中S100A6蛋白的表达,探讨其与肿瘤转移的关系以及对预后的影响。方法采用免疫组织化学技术对30例骨软骨瘤和60例骨肉瘤石蜡包埋标本进行检测,对9例骨软骨瘤和7例骨肉瘤泳冻标本进行Western-blot检测,分析其表达与转移和预后的关系。结果与骨软骨瘤(10%)相比,S100A6蛋白显著表达于人骨肉瘤(85%)中(P〈0.01),其表达与性别、肿瘤的分期、分型无关(P〉0.05),而与肿瘤的转移(P〈0.05)和术后生存时间(P〈0.01)密切相关。结论S100A6蛋白选择表达于骨肉瘤中,可作为肿瘤的早期诊断标志之一,也是判断骨肉瘤转移和预后的重要指标。  相似文献

目的 探讨白血病抑制因子(LIF)在不同时期骨性关节炎(OA)组织中的表达变化规律及临床相关性。方法取35只日本兔制作兔右膝OA模型,左膝为对照组行假手术;于术后第3、7、14、28、42、56及84天各处死5只动物,免疫组化检测LIF在关节组织中的表达变化。另取32例OA患者自愿捐赠的73具关节病变组织标本,应用逆转录-多聚酶链反应及酶联免疫吸附反应分别检测不同病变阶段OA组织中LIF mRNA的表达和含量。结果LIF在正常关节组织中少量表达;OA关节软骨、滑膜均见LIF表达,实验性OA术后28d为LIF表达高峰期。人中度OA软骨下骨中LIF mRNA表达量最高,其次为软骨和滑膜。重度OA软骨中LIF mRNA表达量最高,软骨下骨最低。结论LIF在兔及人关节组织中的表达呈一定时间及空间分布规律;在OA早中期,LIF可能对软骨下骨病变过程具有重要作用。  相似文献

目的 探讨髋关节骨性关节炎的临床特征及全髋关节置换术的疗效。方法 明确诊断髋关节骨性关节炎 81例 ,其中男4 9例 ,女 32例 ,年龄 4 8~ 72岁 ,平均 5 5岁。 1 4例髋关节置换术中 ,其中 2例行双侧全髋置换术。术前Harris评分平均 5 1分 (38~ 6 4分 )。结果  81例髋关节骨性关节炎据临床特征均得到确诊。术后 1 4例病人均无髋脱位 ,髋关节疼痛消失。X线平片示假体位置良好。术后平均随访 2 8个月 (6~ 76个月 ) ,患者Harris评分平均 85分 (81~ 88分 )。结论 对髋关节骨性关节炎患者只要根据其临床特征和采取正确的诊断方法及标准 ,均能确诊。对髋关节骨性关节炎晚期病人 ,采取正确的全髋置换技术 ,均能恢复患者的髋关节功能 ,提高生活质量  相似文献

目的探讨患者关节液和血清中钙结合蛋白(S100A12)浓度和膝关节骨性关节炎严重程度相关性,以评估它在骨性关节炎(osteoarthritis,OA)发病中的作用。方法 40例骨性关节炎(OA组)与40例需关节镜手术的半月板、韧带损伤的患者及行髓内钉固定的股骨及胫骨骨折患者(对照组);对两组患者静脉采血及取关节液,采用ELISA法测S100A12含量;采用Kellgren-Lawrence(K-L)标准对膝关节X线片进行评估分级。结果 OA组的血清中S100A12水平(35.10±26.43)ng/ml显著高于对照组含量(22.13±15.67)ng/ml,有统计学差异(P=0.026);OA组关节液S100A12含量(14.09±8.08)ng/ml显著高于对照组关节液中的含量(9.30±6.85)ng/ml(P=0.02)。OA组血清中的S100A12水平显著高于关节滑液中的水平(P0.01)。关节液中的S100A12含量和血液中的S100A12水平显著正相关(r=0.42,P=0.02),但相关关系并不密切。OA组关节滑液中S100A12与K-L分级成正相关(r=0.65,P0.01)。血清中的S100A12与K-L分级关联不显著(r=0.29,P=0.24)。结论 S100A12在OA患者的血液及关节液中都有表达;关节液的S100A12表达与患者骨关节炎严重程度成正相关;检测关节液中的S100A12有助于骨性关节炎的早期诊断,提示病变严重程度,为临床治疗提供参考。  相似文献

目的探讨髋关节发育不良(developmental dysplasia of the hip,DDH)患者血清及其滑膜组织中的P物质(substance P,SP)、钙结合蛋白(S100 calcium binding protein A12,S100A12)表达及其意义。方法本研究采取回顾性研究方法,选取我院2013年4月至2018年4月收治的182例DDH患者,其中单纯的DDH患者114例(DDH组)、DDH合并骨关节炎(osteoarthritis,OA)的患者68例(OA组),同时选取髋部外伤骨折实施手术的40例患者作为对照组,分别采用ELISA法、免疫组化法检测三组的血清及滑膜组织中的SP、S100A12表达情况并进行比较。结果OA组患者的血清SP、S100A12表达水平显著高于DDH组和对照组(P<0.05),DDH组患者的血清SP、S100A12表达水平显著高于对照组(P<0.05);采用ELISA方法检查,OA组患者的髋关节滑膜组织中SP、S100A12表达水平显著高于DDH组和对照组(P<0.05),DDH组患者的髋关节滑膜组织中SP、S100A12表达水平显著高于对照组(P<0.05);采用免疫组化方法检查,OA组患者的髋关节滑膜组织中SP、S100A12表达的OD值高于DDH组和对照组(P<0.05),DDH组患者的髋关节滑膜组织中SP、S100A12表达OD值显著高于对照组(P<0.05)。结论DDH患者血清及髋关节滑膜组织中SP、S100A12表达较正常髋关节滑膜组织显著增高,合并OA的DDH患者SP、S100A12表达的升高更显著,可能与关节退变及关节的炎性改变有关。  相似文献

目的探讨滑膜切除对全膝关节置换术治疗骨性关节炎临床效果的影响。方法回顾性分析自2011-07—2014-12诊治的105例膝关节骨性关节炎,分为试验组和对照组,试验组(56例)行膝关节置换术与滑膜切除术,对照组(49例)只行膝关节置换术。记录并比较2组手术时间、术后引流量、隐性失血量、输血率及住院时间等,术前和术后随访时进行VAS评分、KSS评分及测量关节活动度。结果试验组获得随访(17.0±4.2)个月,对照组获得随访(15.0±3.1)个月。试验组手术时间、隐性失血量、术后引流量较对照组多,差异均有统计学意义(P0.05),2组住院时间及输血率差异无统计学意义(P0.05)。2组术后4周及1年随访时VAS评分、KSS临床评分、KSS功能评分差异无统计学意义(P0.05)。2组术后3 d及术后7 d关节活动度比较差异无统计学意义(P0.05)。结论与单纯行膝关节置换术相比,膝关节置换同时行滑膜切除术在治疗骨性关节炎上无优势,然而行滑膜切除术会导致更多的体内失血,延长手术时间,从而增加了手术风险。  相似文献

目的探讨钙结合蛋白S100A8/A9复合物对人宫颈癌上皮细胞癌细胞系CasK/细胞骨架及表面超微结构的影响。方法应用20μg/ml S100A8/A9复合物作用于CasKi细胞,对细胞骨架F-肌动蛋白进行染色,应用快速共聚焦荧光显微镜观察细胞骨架的改变。在生理条件下,应用原子力显微镜(atomic force microscopy,AFM)对活体细胞进行高分辨成像,观察细胞表面超微结构及应力纤维。结果S100A8/A9复合物作用24h后宫颈癌细胞系CasKi细胞的F-肌动蛋白排列出现紊乱,主要分布于细胞的周边,细胞中央F-肌动蛋白含量明显减少。CasKi细胞经过20μg/ml S100A8/A9蛋白复合物处理前细胞核部位F-肌动蛋白光密度为92.42±5.16,经过S100A8/A972h处理后,F-肌动蛋白的光密度降为57.67±3.70,两者相比较差异具有显著的统计学意义(t=5.268,P=0.000)。AFM成像显示S100A8/A9复合物作用后细胞边缘卷曲,高度增加,伪足减少,细胞膜下应力纤维结构遭到破坏。结论S100A8/A9蛋白复合物可以对宫颈癌细胞系CasKi细胞表面的超微结构及应力纤维产生影响,并且这种影响主要是通过肌动蛋白的重新分布实现的。  相似文献

目的 检测S100钙结合蛋白A13(S100A13)和人成纤维细胞生长因子-1(FGF-1)在甲状腺癌、甲状腺腺瘤、正常甲状腺组织中的表达,探讨两者与甲状腺癌发生发展的关系.方法 采用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶(SP)方法检测甲状腺癌、甲状腺腺瘤、正常甲状腺组织中两者的表达,比较两者在两种不同组织中的表达.结果 甲状腺癌组织中,S100A13的表达阳性率为93.3%(56/60),在甲状腺腺瘤组织中S100A13的表达阳性率为66.7% (20/30),在正常甲状腺组织中的表达阳性率为60.0%(12/20),S100A13在甲状腺癌组织的表达阳性率明显高于甲状腺腺瘤及正常甲状腺组织,差异有统计学意义(P<0.05),但是在甲状腺腺瘤及正常甲状腺组织中其表达差异无统计学意义(P>0.05);FGF-1在甲状腺癌组织中的表达阳性率为81.7%(49/60),在甲状腺腺瘤组织中FGF-1的表达阳性率为63.3%(19/30),在正常甲状腺组织中的表达阳性率为55.0%(11/20),FGF-1在甲状腺癌组织的表达阳性率明显高于甲状腺腺瘤及正常甲状腺组织,差异有统计学意义(P<0.05),但是在甲状腺腺瘤及正常甲状腺组织中其表达差异无统计学意义(P>0.05);S100A13与FGF-1表达阳性与甲状腺癌的淋巴结转移相关(P<0.05).结论 S100A13与FGF-1与甲状腺癌的发生及发展密切相关.  相似文献

目的 检测胃腺癌组织中钙结合蛋白S100A2、reprimo的表达情况,探讨它们在胃腺癌发生发展过程中的作用以及相互作用.方法 应用免疫组织化学SP法,检测 100例胃腺癌组织和20例正常胃黏膜组织中S100A2和reprimo蛋白的表达,分析其与胃癌临床病理特征之间的关系.结果 在20例正常胃黏膜组织中S100A2、reprimo均呈阳性表达,在100例胃腺癌组织中,其阳性表达率分别为48%、35%,差异均有统计学意义(χ2值分别为18.35、28.36,P<0.01).S100A2和reprimo的表达与浸润深度、淋巴结转移密切相关(χ2值分别为4.54、10.53、15.71、16.22,P<0.05或P<0.01),S100A2的表达还与胃腺癌分化程度有关(χ2值为6.73,P<0.01).S100A2蛋白的表达与reprimo蛋白的表达呈显著正相关(χ2值为14.91,P<0.01). 结论胃腺癌组织S100A2、 reprimo蛋白表达下调可能与胃腺癌发生发展有关.低的S100A2、 reprimo阳性表达率常意味着肿瘤高的浸润转移能力.S100A2、 reprimo的表达可以作为胃腺癌预后的判断指标.  相似文献

作者采用随机区组设计比较了126名老年人中髋关节骨关节炎及骨质疏松患者与正常对照组的Singh指数及骨关节炎程度。结果表明,三组之间的Singh指数及骨关节炎程度均相差非常显著(P<0.01),其中骨关节炎组的Singh指数高于股骨颈骨折组(P<0.01)和正常对照组(P<0.05)。股骨颈骨折组的骨关节炎程度则轻于骨关节炎组(P<0.01),但与正常对照组相比并无显著之统计学意义(P>0.05)。研究结果提示:髋关节的骨关节炎与骨质疏松之间存在着十分明显的负相关。  相似文献

IntroductionPrimary synovial osteochondromatosis (SOCM) or Synovial chondromatosis (SC) of the hip is a benign metaplastic condition of the synovium that is rare and may present with a spectrum of clinical features and radiological findings.Patients and methodsA retrospective search using the keyword ‘Synovial chondromatosis’ (SC) of the hip was performed at a tertiary care orthopaedic referral centre in the UK and a hospital in India. The radiology images were collected from our Picture Archiving and Communication System (PACS) and Radiology Information System (RIS), over 15 years. The patient's data was collated with Electronic Patient Records (EPR), RIS, and correlated with histo-pathology laboratory records where available. The demographic details of the patients, their clinical symptoms, imaging details, and management outcomes were collected.ResultsWe found 15 cases, with a mean age of 36.53 years (range: 14–50 years). There were 9 male and 6 female patients. The follow-up ranged from 1 year to 6 years. Predominantly unilateral presentation with insidious onset of symptoms was found. A spectrum of radiological Imaging was undertaken. Management strategies included supervised observation, arthroscopic or open synovectomy, and hip arthroplasty. No malignant transformation was found in the analysed cohort.ConclusionPrimary ‘Synovial chondromatosis’ of the hip had a male preponderance in our cohort, presenting with a range of clinical features. Radiologically, Magnetic Resonance Imaging (MRI) was the commonest modality of cross-sectional imaging utilised and crucial for the diagnosis, evaluating underlying articular involvement including guiding appropriate patient management presenting with Primary ‘Synovial chondromatosis’ of the hip.  相似文献

The distribution and localization of S-100 protein (S-100) and its subunits (S100-alpha and S100-beta) in the testis of swamp-type water buffalo were investigated using immunohistochemistry. S-100 was detected in the Sertoli cells in the convoluted seminiferous tubules, modified Sertoli cells lining the terminal segment of the seminiferous tubules and in the intratesticular excurrent ducts (straight tubules and rete testis). S100-beta showed the same distribution and localization with that of S-100. However, the cytoplasmic extension of the Sertoli cells in S100-beta staining showed less staining intensity compared with that of S-100. S100-alpha showed a positive staining only in the modified Sertoli cells of the terminal segment of the seminiferous tubule. Endothelial cells of blood vessels were also positive with the proteins while the Leydig and spermatogenic cells showed a negative reaction. The localization of S-100 in the testis of the water buffalo was in parallel with that of other artiodactyls which supports the hypothesis that this protein is a multifunctional protein. S100-beta in the Sertoli cells suggests that this protein is involved in establishing blood-testis barrier. Its presence in the modified Sertoli cells and in the epithelium of the excurrent ducts suggest secretory and absorptive function, respectively. Meanwhile, S100-alpha, which was detected only in the modified Sertoli cells, is involved in the secretory activity of these cells that are related to exocrine function.  相似文献

目的探讨钙结合蛋白S100A12在小鼠重症急性胰腺炎治疗中的潜在作用。方法采用连续7次腹腔注射雨蛙素50μg/kg(每次间隔1 h)及脂多糖建立小鼠急性胰腺炎模型。将160只SPF级雌性ICR小鼠随机分为对照组(A组,生理盐水)、轻型组(B组,雨蛙素)、重症组(C组,雨蛙素+脂多糖)、干预组(D组,S100A12重组抗体+雨蛙素+脂多糖),每组40只。在首次注射雨蛙素后8 h、12 h和24 h采血,分别检测各时段血清中S100A12、淀粉酶(AMY)、C反应蛋白(CRP)、白介素(IL-1β、IL-6)、肿瘤坏死因子(TNF-α)的含量,观察小鼠胰腺的病理形态并进行评分。结果与C组相比,D组各时段血清中AMY、CRP、IL-1β、IL-6、TNF-α、S100A12含量明显降低(P0.05);与B组相比,D组各时段血清S100A12浓度明显降低(P0.05),胰腺病理形态也有明显改善。结论 S100A12重组抗体能够有效降低小鼠急性胰腺炎的严重程度,S100A12可以作为重症急性胰腺炎治疗的一个有效靶点。  相似文献

BACKGROUND: To evaluate the diagnostic performance of plasma S100A8, S100A9, and the S100A8/A9 complex as novel markers to discriminate between benign and malignant prostatic diseases as recently suggested for S100A9. METHODS: The study included 90 prostate cancer (PCa) patients (pN0M0, n = 50; pN1M0, n = 27; M1, n = 13), 50 controls without PCa, and six patients within 72 hr after radical prostatectomy for repeated measurements. The S100 proteins were analyzed with specific ELISAs. Comparisons were made to the prostate-specific antigen (tPSA) and the ratio of free to tPSA (%fPSA). RESULTS: The plasma concentrations of the S100 proteins in controls had either significantly higher values (S100A8; P = 0.020) or the tendency to higher values compared with the results in PCa patients. Differences between the three PCa groups were almost negligible. No correlation could be found between S100 protein levels and PSA concentration (r(s) = -0.110 to 0.433, P = 0.317-0.433) or prostate volume (r(s) = -0.038 to 0.018, P = 0.676-0.844). Tumor stage and tumor grade had no observed effect on S100 protein concentrations. After prostatectomy, there were discordant elimination kinetics for PSA and the S100 proteins, as the S100 proteins partially increased while PSA continuously decreased. Analyses of receiver-operating curves showed that, compared with PSA, S100A8, S100A9, and S100A8/A9 did not improve the differentiation between patients with and without PCa, while the discrimination ability was significantly lower than that of %fPSA. CONCLUSIONS: Our re-evaluation study showed that S100A8, S100A9, and the complex S100A8/A9 were less indicative than %fPSA and that they are not suitable to replace PSA.  相似文献
