目的 探讨乌灵胶囊对卒中后抑郁大鼠海马P13K/Akt/mTOR通路和神经递质影响。方法 选取健康SD大鼠48只,随机分为对照组、模型组、低剂量组和高剂量组,每组12只。采用改良Zea-Longa法复制大脑中动脉缺血大鼠模型(MCAO模型),MCAO模型复制成功后皮下注射利血平复制卒中后抑郁模型。对照组和模型组给予等剂量双蒸水灌胃;低剂量组于模型复制成功后第2天给予40 mg乌灵胶囊;高剂量组于模型复制成功后第2天给予80 mg乌灵胶囊,1次/d。各组大鼠灌胃21 d。采用Western blotting检测大鼠海马组织海马组织PI3K、p-Akt和p-mTOR蛋白相对表达量;高效液相色谱法测定海马组织NE、DA和5-HT含量。结果 模型组、低剂量组和高剂量组大鼠Zea Longa评分高于对照组(P <0.05),海马组织PI3K、p-Akt和p-mTOR蛋白相对表达量低于对照组(P <0.05),海马组织NE、DA和5-HT含量低于对照组(P <0.05);而低剂量组和高剂量组大鼠Zea Longa评分低于模型组(P <0.05),海马组织PI3K、p-Akt和p-mTOR蛋白相对表达量高于模型组(P <0.05),海马组织NE、DA和5-HT含量高于模型组(P <0.05)。结论 乌灵胶囊可改善卒中后抑郁大鼠行为,且可调节海马P13K/Akt/mTOR通路,调节神经递质水平。  相似文献

Objective:To explore the effi cacy and mechanism of Lirukang Granule(利乳康颗粒,LRKG) in treating hyperplasia of mammary gland(HMG).Methods:One hundred patients with HMG were randomly assigned to two groups,50 in each group.The patients in the treated group were orally administered with LRKG thrice a day,one package each time,and those in the control group were given orally Rukuaixiao Tablet(乳块消,RKX) thrice a day,4 tablets each time.The therapeutic course for both groups was 4 months.The clinical eff icacy,pain alleviating rate,as well as changes of local sign and symptom scores were observed before and after treatment.The changes of serum estradiol(E2),progesterone(P),testosterone(T),follicle stimulating hormone(FSH),luteinizing hormone(LH) and prolactin(PRL) in some randomly selected patients(24 patients in the treated group and 24 in the control group) before and after treatment were measured with radioimmunoassay.Results:The total clinical effi cacy in the treated group was superior to that in the control group,signifi cant difference was shown between the two groups(P<0.01).The cure-effective rate and total effective rate in the treated group were 70.0%and 88.0% respectively,signif icantly higher than those in the control group(38.0% and 64.0%) respectively(P<0.01),and the pain alleviating rate in the treated group was also significantly higher in the former than that in the latter(88.0% vs 64.0%,P<0.05).Moreover,the treated group showed obvious superiority in improving the patients' symptom and sign scores(P<0.01),and abnormalities of gonadal hormone as compared with the respective items in the control group(P<0.01).Conclusion:LRKG has good effi cacy in the treatment of HMG,and its mechanism may be related to the regulation on endocrine and immune function.  相似文献

[目的] 夏枯草(Prunella vulgaris L.)在临床中已经应用于治疗自身免疫性甲状腺炎(AIT), 但是机制尚不明确。本研究通过建立实验性自身免疫性甲状腺炎(EAT)大鼠模型, 研究夏枯草水提液对EAT大鼠5-羟色胺(5-HT)、5-羟色胺转运体(SERT)和Th17的作用。[方法] 水提法提取夏枯草的有效成分并采用高效液相色谱法(HPLC)对其主要活性成分进行分析; 采用皮下注射甲状腺球蛋白(Tg)联合高碘水喂养的方式建立EAT大鼠模型, 随后将EAT大鼠分为模型对照组(EAT), 夏枯草低剂量组(E-LOW), 夏枯草中剂量组(E-MID), 夏枯草高剂量组(E-HIG), 连续灌胃12周。采用酶联免疫吸附检测法(ELISA)检测大鼠血清中辅助性T细胞17(Th17)和细胞因子水平; HE染色观察大鼠甲状腺形态; 流式细胞术检测大鼠外周血单个核细胞(PBMCs)中CD4+IL17A+(Th17)淋巴细胞的水平; qRT-PCR检测脾脏中SERT水平; ELISA法检测大鼠脾脏和血清中5-HT以及血清中IL-17A, IL-6含量。[结果] 迷迭香酸(Rosmarinic Acid)是夏枯草提取物的主要成分, 夏枯草水提液治疗12周可显著降低大鼠血清TgAb(P<0.05或P<0.01), 减小甲状腺体积(P<0.01), 减轻甲状腺内炎性浸润, 降低外周血Th17比例(P<0.01)和血清IL-17A, 抑制大鼠脾脏中SERT水平(P<0.01), 提高脾脏和血清5-HT水平(P<0.01)。[结论] 夏枯草可通过提高EAT大鼠中5-HT含量, 抑制Th17细胞分化, 发挥调节免疫作用, 该研究为夏枯草治疗EAT提供新的思路。  相似文献

[目的]探究四逆散对抑郁大鼠行为、中枢神经递质及ERK1/2-CREB-BDNF信号通路的影响。[方法]实验大鼠随机分为正常组、模型组、阳性对照组、四逆散低剂量组、四逆散中剂量组、四逆散高剂量组,每组20只。给药结束后,进行糖水消耗实验、旷场实验;Elisa试剂盒检测大鼠海马组织中多巴胺(DA)、去甲肾上腺素(NE)、5-羟色胺(5-HT);放免试剂盒检测血清中神经肽Y(NPY)、P物质(SP)、生长抑素(SS)含量;Western blot检测皮质中ERK1/2-CREB-BDNF信号通路关键蛋白表达情况。[结果]与正常组相比,模型组大鼠糖水偏爱度和旷场运动评分显著降低(P均<0.001),海马组织中DA、NE、5-HT含量显著下调(P<0.001),血清中NPY、SS显著下调(P<0.001),SP显著上调(P<0.001),皮质中ERK1/2、CREB蛋白磷酸化水平及BDNF蛋白表达显著下调(P<0.001);与模型组相比,四逆散给药组大鼠糖水偏爱度和旷场运动评分显著上调(P<0.001),海马组织中DA、NE、5-HT含量显著上调(P<...  相似文献

[目的] 对莫哈韦丝兰的化学成分进行系统研究。[方法] 利用正相硅胶、反相ODS、Sephadex LH-20及高效液相色谱等多种色谱法进行分离纯化,并通过理化性质及波谱分析鉴定化合物结构。[结果] 从莫哈韦丝兰70%乙醇提取物中分离鉴定了14个单体化合物,分别为咖啡酸(1)、β-羟基-3-甲氧基-4-羟基苯乙酮(2)、1-O-β-D-glu-copyranosyl-2-hydroxy-4-allylbenzene (3)、2-O-(4-hydroxybenzoyl)-2,4,6-trihydroxyphenylacetic acid (4)、对羟基苯甲酸(5)、原儿茶酸(6)、4-羟基-3-甲氧基苯甲酸(7)、丁香酸(8)、对羟基苯甲醛(9)、原儿茶醛(10)、麦芽酚(11)、5-hydroxymalto (l 12)、5-羟甲基糠醛(13)和4,4-二甲基-1,7-庚二酸(14)。[结论] 化合物2~4、7~14为首次从丝兰属植物中分离得到的化合物,5、6为首次从该种植物中分离得到的化合物。  相似文献

OBJECTIVE

To evaluate the efficacy of moxibustion, through stimulating acupoints of Danzhong (CV 17) and Ganshu (BL 18) in rats with hyperplasia of mammary gland (HMG) which induced by estrogen and progestogen.

METHODS

Thirty female Sprague-Dawley rats were randomly divided into saline control group, HMG model group, and HMG moxibustion group with 10 in each group. Saline control was the group injected by saline. HMG model were created by injection of estrogen and progestogen. Moxibustion group was also injected of estrogen and progestogen with moxibustion at the same time. The Changes of nipple diameter and height were measured. The rats’ skin temperature was recorded by an infrared thermal camera at the nipples, mammary areas, Danzhong (CV 17) and Ganshu (BL 18). Pathological changes of mammary gland in rats were also observed under light microscope.

RESULTS

The diameter and height of the nipples in model group were prominently bigger and higher than that in control group (P < 0.01). The diameter and height in moxibustion group were prominently smaller and lower than that in model group (P < 0.01), and there was no significant difference between moxibustion group and control group. Compared with control group, skin temperature of the nipples, mammary area, and acupoints Danzhong (CV 17) and Ganshu (BL 18) decreased prominently in model group (P < 0.01-0.05). Compared with model group, skin temperature of that in moxibustion group increased prominently (P < 0.05).

CONCLUSION

Treatment with moxibustion can effectively decrease the HMG rats’ nipple diameter and height, and increase the skin temperature in HMG model rats at the nipples, mammary areas, Danzhong (CV 17) and Ganshu (BL 18). This study convinces the therapeutic effect of moxibustion on mammary gland hyperplasia.  相似文献

目的 研究焦虑性抑郁模型大鼠海马、杏仁核、前额叶皮质内单胺递质的含量变化及脑内神经营养因子的表达趋势,探讨其可能的发病机制。方法 60只SD大鼠随机分为正常对照组、溶媒对照组、焦虑模型组、抑郁模型组、焦虑性抑郁模型组,每组12只。采用慢性束缚应激联合皮质酮注射的方法建立焦虑性抑郁大鼠模型,造模时间为21 d,造模结束后采用高架十字迷宫测试,旷场实验,强迫游泳实验评价大鼠的焦虑和抑郁样行为,HPLC-ECD法检测大鼠海马、杏仁核、前额叶皮质的单胺递质5-HT、NE、DA含量,蛋白印迹法检测大鼠各脑区神经营养因子BDNF、NT-3的含量。结果 焦虑性抑郁模型组大鼠在进入开臂的时间、次数、旷场中自主活动次数均与焦虑组相当,与对照组及抑郁组比较差异有显著性(P<0.01或P<0.05),在强迫游泳中的不动时间显著增加,与对照组及焦虑组对比差异有显著性(P<0.01);同时,与对照组比较,焦虑性抑郁模型组大鼠海马5-HT、杏仁核及前额叶皮质区的5-HT和NE含量均显著下降(P<0.01或P<0.05);此外,与对照组比较,焦虑性抑郁模型组大鼠各脑区BDNF、NT-3含量显著下降(P<0.01或P<0.05),同时与焦虑组比较,BDNF含量显著下降(P<0.05)。结论 焦虑性抑郁模型组大鼠具有显著的焦虑及抑郁样行为,其发病机制可能与脑内海马、杏仁核、前额叶皮质区域的单胺递质含量降低及神经营养因子BDNF、NT-3表达下调有关。  相似文献

目的:观察中药抑乳汤对女性原发性甲状腺机能减退 (简称甲减)患者血清泌乳素升高的治疗作用。方法:治疗组用抑乳汤配合甲状腺素片治疗;对照组用单纯甲状腺素片治疗。治疗前后用放射免疫法进行甲状腺素(T4)、游离甲状腺素 (FT4)、促甲状腺激素 (TSH)和泌乳素 (PRL)测定。结果:两组临床近期疗效总有效率无明显差异 (P>0.05),但治疗组PRL的下降率及远期疗效的复发率、不良反应率与对照组相比均有显着性差异 (P<0.05,P<0.01).结论:抑乳汤配合甲状腺素片是治疗原发性甲减伴高PRL血症的有效方法。PRL水平的升高与下丘脑-垂体-甲状腺轴的激素分泌失调有一定的内在联系,两者的关系值得探讨。  相似文献

Objective: To investigate the influence of Yiqi Huatan Decoction (益气化痰方, YHD) on a model of depression in rats under different pathological conditions. Methods: Thirty-two male SD rats were randomly divided into 4 groups of 8: normal, model, YHD, and maprotiline. The model group, YHD group and maprotiline group used separate feeding and rats were exposed to chronic and unpredictable stress to build the depression model. From day 2, the YHD group and maprotiline group were respectively given YHD (7 g/kg) and maprotiline (10 mg/kg) by gastrogavage once daily. The normal and model groups were given the same volume of drinking water. The medication duration were 21 days. At the end of the experiment, the serum levels of copper and zinc were determined by atomic absorption spectroscopy, plasma concentrations of adrenocorticotropic hormone (ACTH) and cortisol (COR) were detected by radioimmunoassay, and levels of norepinephrine (NE), dopamine (DA), and 5-hydroxytryptamine (5-HT) in the hypothalamus were analysed by high performance liquid chromatography-eletricochemistry. Results: Compared with the content of copper and zinc in the serum of rats in the normal group, serum copper levels in model rats were significantly increased and zinc content was significantly reduced (both P<0.05). Plasma concentrations of ACTH and COR in the model group were significantly increased compared with those in the normal group (P<0.05, P<0.01). The contents of NE, DA, and 5-HT in the hypothalamus of rats in the model group were significantly reduced compared with those of the normal group (P<0.05 or P<0.01). Compared with those in the model group, the serum copper content and plasma concentrations of ACTH and COR were significantly decreased (all P<0.05); meanwhile, serum zinc content and hypothalamic contents of NE, DA, and 5-HT were significantly increased in rats of the YHD group (all P<0.05). The same effects were also shown in the maprotiline group except for 5-HT (all P<0.05). Conclusion: The pharmacological actions of YHD for depression might be related to improving trace-element anomalies, reversing endocrine dysfunction, and modulating the disorders of monoaminergic neurotransmitters.  相似文献

目的 探讨老年功能性消化不良(FD)患者血浆神经肽S受体1(NPSR1)、5-羟色胺(5-HT4)的表达及其临床意义。方法 选取2020年2月—2022年8月在武汉市第一医院就诊的96例老年FD患者作为FD组,另取同期该院健康体检老年人80例作为对照组。对比FD组与对照组临床资料、血浆NPSR1、5-HT4水平。多因素逐步Logistic回归分析影响老年FD发生的相关因素。绘制受试者工作特征(ROC)曲线,分析血浆NPSR1、5-HT4及两者联合检测对老年FD的诊断效能。结果 FD组体质量指数、脂肪肝、睡眠障碍、焦虑、抑郁及IL-6、CCK及SS水平高于对照组(P <0.05),饮食规律、每周运动时间≥4 h占比及MTL水平低于对照组(P <0.05)。FD组血浆NPSR1水平低于对照组(P <0.05),5-HT4水平高于对照组(P <0.05)。多因素逐步Logistic回归分析结果显示,睡眠障碍[■=2.735(95%CI:1.299,5.756)]、NPSR1[■=3.203(95%CI:1.461,7.023)]、5-HT4[■=3.093(95%CI...  相似文献

目的:通过分析抑郁模型大鼠脑组织DA、5-HT及其代谢产物的浓度变化,探讨抑郁症发病的神经生化机制。方法:将24只雄性SD大鼠随机分成对照组、模型组和氟西汀组3组;采用慢性轻度不可预见性应激(CUMS)结合孤养建立抑郁大鼠模型,随后取脑并分离内侧前额皮质(mPFC)和海马,采用库仑阵列电化学高效液相色谱法测定上述脑区DA、5-HT及其代谢产物的浓度。结果:与对照组比较,模型组大鼠mPFC区DA、高香草酸(HVA)、5-HT及5-羟吲哚乙酸(5-HIAA)浓度均显著降低(P<0.01或0.05),海马DA、HVA和5-HT浓度显著降低(P<0.01或0.05);与模型组比较,氟西汀组大鼠mPFC区DA、HVA和5-HT浓度显著升高(P<0.01或0.05);海马DA、5-HT和5-HIAA浓度显著升高(P<0.05)。结论:CUMS抑郁模型大鼠mPFC和海马组织存在DA和5-HT递质系统功能紊乱。  相似文献

目的 探讨度洛西汀联合盐酸羟考酮对晚期癌性疼痛患者的疗效及焦虑抑郁情绪的影响。方法 选取2018年3月—2021年10月南通大学附属南通肿瘤医院收治的82例晚期癌痛患者作为研究对象。将患者分为对照组、研究组,各41例。对照组口服羟考酮治疗,研究组在对照组的基础上加用度洛西汀,两组均持续治疗4周后观察疗效。统计两组羟考酮人均日口服剂量,比较两组治疗前后疼痛评分、疼痛相关因子变化及焦虑抑郁状况,比较两组治疗前后神经递质表达状况,观察药物安全性。结果 研究组羟考酮人均日口服剂量低于对照组(P <0.05)。研究组与对照组治疗前、治疗2周后、治疗4周后静息状态下的BPI评分比较,经重复测量设计的方差分析,结果:(1)不同时间点的BPI评分有差异(P <0.05);(2)两组静息状态下BPI评分有差异(P <0.05),研究组治疗后较对照组低,相对镇痛效果较好;(3)两组BPI评分变化趋势有差异(P <0.05)。研究组与对照组治疗前、治疗2周后、治疗4周后的PEG2、SP比较,经重复测量设计的方差分析,结果:(1)不同时间点的PEG2、SP有差异(P <0.05)...  相似文献

目的 观察壬基酚(NP)对大鼠血浆和尿液中5-羟色胺(5-hydroxy tryptamine,5-HT)及血小板中5-HT和5-HT2A受体含量的影响,探讨壬基酚对大鼠5-HT和5-HT2A受体影响的毒效应机制。方法 将24只SD雄性大鼠分为阴性对照组和NP低、中、高剂量组[30、90、270mg/(kg·d)],隔日灌胃染毒28d后检测大鼠血浆中5-HT、血小板中5-HT和5-HT2A受体含量,并检测灌胃后收集到的24h尿液中5-HT的含量。结果 染毒28d后,随NP暴露的剂量增加,各组大鼠血浆、血小板及尿液中5-HT含量升高,血小板中5-HT2A受体表达则下降。NP暴露中、高剂量组大鼠血浆及血小板中5-HT含量均高于对照组(P<0.01;P<0.01),NP暴露高剂量组大鼠血小板5-HT2A受体表达低于对照组(P<0.01)。第4~28天,NP暴露低、中、高剂量组大鼠的尿液5-HT含量均高于对照组(P<0.01)。结论 NP暴露剂量与大鼠血小板5-HT、5-HT2A受体及血浆和尿液中5-HT含量均呈剂量-效应关系,提示NP通过影响大鼠5-HT水平和5-HT2A受体表达而产生毒效应。  相似文献

目的 观察夏天无总碱提取物对痴呆大鼠脑内 5羟色胺 (5 HT)和多巴胺 (DA)递质含量的影响。方法 采用D 半乳糖腹腔连续注射 6周 ,制备大鼠痴呆模型。Y电迷宫法测试对大鼠的学习、记忆能力 ,采用荧光分光光度计测定大鼠脑内 5 HT ,DA的含量。结果 夏天无总碱提取物用量为 0 .5mg/kg ,1.0mg/kg时可显著改善模型大鼠的学习记忆能力 (P <0 .0 1) ,显著增加痴呆大鼠脑内 5 HT含量 (P <0 .0 1,P <0 .0 5 ) ,夏天无总碱提取物用量为 0 .5mg/kg ,1.0mg/kg ,0 .2 5mg/kg时均可显著增加痴呆大鼠脑内DA含量 (P <0 .0 1;P <0 .0 5 )。 结论 夏天无总碱提取物提高痴呆大鼠学习记忆功能的作用 ,可能与增加脑内神经递质 5 HT和DA有关。  相似文献

目的 探讨生长抑素类似物奥曲肽(OCT)和三苯氧胺(TAM)的体内抗乳腺癌作用。方法 将接受致癌剂二甲基苯蒽(DMBA, 100mg/kg,一次性皮下注射)后8周的96只Wistar雌鼠,随机分为对照组(单纯诱发组),OCT组( 100μg/kg, 2次/日皮下注射,共14周),TAM组( 1mg/kg,5次/周皮下注射,共14周)和OCT加TAM组,每组24只。观察乳腺肿瘤发生的潜伏期、诱发率,每只大鼠乳腺肿瘤的个数和体积,并利用光镜、电镜进行肿瘤细胞形态学和超微结构研究。结果 (1)乳腺肿瘤平均潜伏期:TAM组(119d±19d)和OCT加TAM组(120d±16d)明显长于对照组(90d±17d)和OCT组(102d±15d) (均P<0 .05); (2)乳腺肿瘤诱发率:OCT加TAM组(23 .8% )、TAM组(45 .5% )、OCT组(52. 4% )均明显低于对照组(70% ) (均P<0 .05 ),而OCT加TAM组明显低于OCT组和TAM组(均P<0 .05); (3)乳腺肿瘤个数:对照组明显高于其余3组(均P<0 .05),OCT加TAM组明显低于OCT组和TAM组(均P<0 .05); (4)注射致癌剂后22周乳腺肿瘤平均体积:对照组(6434mm3 ±1536mm3 )明显高于OCT组(4366mm3 ±1213mm3 )、TAM组(41.38mm3 ±10.91mm3 )和OCT加TAM组(12.85mm3 ±3.29mm3 ) (均P<0 .05),OCT加TAM组明显低于TAM组和OCT组(均P<0 .01);实验18周时TAM组乳腺肿瘤体积(1126mm3 )明显低于OCT组(2139m  相似文献

目的 探讨镜像理论指导下的运动联合康复机器人对脑卒中患者手功能恢复的影响。方法 选取2020年1月—2022年1月广州医科大学附属第二医院收治的122例脑卒中手功能障碍患者作为研究对象。以随机数字表法分为对照组、研究组,每组61例。对照组患者给予康复机器人治疗,研究组患者在对照组患者基础上另给予镜像理论指导下的运动。两组患者持续治疗4周后评价疗效。对比两组患者治疗前后手最大握力、捏力及手指总主动活动情况,比较两组患者治疗前后腕关节等速肌力及手屈曲、背伸动作下屈指浅肌、伸指肌的积分肌电值情况,统计两组患者治疗前后上肢功能及日常生活活动功能变化。结果 研究组治疗前后的最大握力、捏力的差值高于对照组(P <0.05)。研究组手指总主动活动度优良率高于对照组(P <0.05)。研究组治疗前后的腕关节屈肌肌群手、腕关节伸肌肌群峰值力矩、最大总用功的差值高于对照组(P <0.05)。研究组治疗前后的手屈曲、手背伸动作下屈指浅肌、伸指肌的积分肌电值的差值高于对照组(P <0.05)。研究组治疗前后Fugl-Meyer运动功能量表、Barthel指数的差值高于对照组(P <...  相似文献

[目的] 观察针刀干预对肌筋膜疼痛综合征模型大鼠血清和触发点局部组织五羟色胺(5-HT)、去甲肾上腺素(NE)的影响,探讨针刀治疗肌筋膜疼痛综合征的机制。[方法] 将24只大鼠随机分为空白组、模型组、针刀组和注射组,每组6只。采用打击结合离心运动法建立肌筋膜疼痛综合征大鼠模型,造模8周。注射组、针刀组进行相应方法干预。用ELISA法检测各组大鼠血清及触发点局部肌肉组织5-HT、 NE的含量,采用SPSS 20.0统计学软件进行统计分析。[结果] 与空白组比较,模型组大鼠血清和局部组织5-HT、NE含量显着升高,差异有统计学意义(P<0.05),注射组大鼠血清和组织中5-HT含量显着升高(P<0.05);与模型组比较,注射组大鼠血清5-HT、NE的含量显着下降(P<0.05),局部肌肉组织5-HT含量与之比较未见统计学差异(P>0.05),NE含量显着下降(P<0.05);针刀组大鼠血清和局部肌肉组织5-HT、NE的含量显着下降(P<0.05).[结论] 肌筋膜疼痛综合征模型大鼠血清和局部组织5-HT、NE含量比正常大鼠显着升高,针刀干预可以降低血清和局部组织5-HT、NE含量,注射利多卡因可以降低血清NE、5-HT及局部组织NE含量;针刀及利多卡因注射对于肌筋膜疼痛综合征的治疗作用可能与其降低血清及局部组织5-HT、NE含量有关;且针刀对于局部组织5-HT的调节作用优于药物注射。  相似文献

[目的] 观察参竹精片对睡眠的影响。[方法] 选昆明小鼠随机分为正常对照组、参竹精片0.5、0.25、0.125 g/kg组,分别单次灌胃给药和连续7 d给药,末次给药后2 h,各组小鼠分别以30 mg/kg(阈下剂量)和50 mg/kg(阈剂量)腹腔注射戊巴比妥钠,观察小鼠入睡时间、入睡率及睡眠时间,测定脑干组织5-羟色胺(5-HT)、5-羟基吲哚乙酸(5-HIAA)含量。[结果] 与正常组比较,单次灌胃给药和连续7 d给药,参竹精片可明显增加戊巴比妥钠(阈下剂量)致小鼠的入睡率(0.5 g/kg,P<0.05),减少戊巴比妥钠(阈剂量)致小鼠的入睡时间(P<0.01),延长睡眠时间(P<0.01),增加脑干组织5-HT含量(P<0.05)。[结论] 参竹精片可促进脑组织分泌5-HT,减少入睡时间、提高入睡率、延长睡眠时间,具有一定改善睡眠的作用。  相似文献

Objective:To determine the mechanisms underlying the anti-depressant effects of Kaixin Jieyu Decoction(开心解郁汤,KJD)by investigating the effects of KJD on behavior,monoamine neurotransmitter levels,and serotonin(5-HT)receptor subtype expression in the brain in a rat model of depression.Methods:The rat depression model was established using chronic unpredictable mild stress(CUMS).Forty-eight Sprague Dawley rats were randomly divided into control,depression model(CUMS),CUMS±KJD(7.7 g/kg~(-1)·d~(-1)of crude drug),and CUMS+fluoxetine(2.4 mg/kg~(-1)·d~(-1))groups(n=12 in each group),and the treatments lasted for 21 days.We regularly evaluated body weight,sucrose consumption,and horizontal and vertical activity scores in open-field tests.The content of the monoamine neurotransmitters 5-HT,norepinephrine(NE),and dopamine(DA)and the DA metabolite homovanillic acid in the cerebral cortex,and 5-HT_(1A)and 5-HT_(2A)receptor mRNA in the cerebral cortex and the hippocampus,were determined respectively by high-performance liquid chromatography-coularray electrochemical detector and real-time polymerase chain reaction.Results:Compared with the control group,CUMS rats showed a variety of depression-like behavioral changes,including a significant reduction in body weight,sucrose consumption,and horizontal and vertical activity scores in open-field tests(P0.05 or P0.01),and a significant decrease in 5-HT and NE levels and 5-HT_(2A)receptor mRNA expression.In contrast,they showed a significant increase in 5-HT_(1A)receptor mRNA expression in the cerebral cortex.In the hippocampus,5-HT_(1A)receptor mRNA expression was lower whereas 5-HT_(2A)receptor mRNA expression was higher than in the control group(P0.05 or P0.01).Treatment with KJD or fluoxetine partially attenuated these changes(P0.05 or P0.01).Conclusion:KJD could normalize the levels of 5-HT and NE and adjust the balance of 5-HT_(1A)and 5-HT_(2A)receptor expression in rat cerebrum,and this may be one of mechanisms of antidepressant effects of KJD.  相似文献

目的:研究鹿宝养生饮品(LHD)对过度游泳训练所致疲劳大鼠的保护作用,并探讨其作用机制。方法:通过过度游泳训练 8 周建立大鼠疲劳模型,将雄性Wistar大鼠随机分成对照组、一般游泳训练组、过度负荷游泳模型组及LHD低、中、高剂量组,对照组、一般游泳训练组及过度负荷游泳模型组大鼠均灌胃蒸馏水10 mL.kg/d,LHD 3个剂量组大鼠分别灌胃不同浓度的LHD 10 mL/kg/d。给药 8 周后观察各组大鼠体质量变化,检测尿蛋白(UP)含量、血糖(BG)水平及血红蛋白(Hb)、血清睾酮(ST))和血浆皮质酮(PC)含量;以夹心ELISA法测定大鼠下丘脑多巴胺(DA)、去甲肾上腺素(NA)、5-羟色胺(5-HT)及γ-氨基丁酸(GABA)含量,并计算DA/5-HT、NA/5-HT及DA/NA比值。结果:与过度负荷游泳模型组比较,LHD中、高剂量组大鼠在游泳训练5~8周因过度游泳训练造成的体质量减轻得到改善,UP含量明显降低(P<0.05或P<0.01),BG水平及Hb、ST、PC含量均无明显变化。LHD中、高剂量组大鼠下丘脑5-HT含量明显降低, DA含量和DA/5-HT、NA/5-HT及DA/NA比值明显增高(P<0.05或P<0.01)。结论:LHD对过度游泳训练所致的大鼠超负荷疲劳具有明显改善作用,其作用机制可能与降低下丘脑5-HT含量,提高DA、GABA含量以及DA/5-HT、NA/5-HT和DA/NA比值有关。
