Objective To investigate the metabonomic variation between patients with esophageal cancer (EC) and healthy controls, and to analyze the variation between patients with EC. Methods 1H-MR and orthogonal partial least-squares discriminant analysis (OPLS-DA) was performed on 108 plasma specimens from EC patients and 50 health controls, and the metabonomic variation between patients with EC and healthy controls analyzed. Conclusion The results indicate that1H-MR spectra of plasma analyzed by OPLS-DA statistical methods might completely separate the EC patients from health controls. 目的探讨肝素结合蛋白(HBP)在早期细菌感染性疾病中的诊断价值。方法选取2017年7月-2018年2月该院收治的早期细菌感染性疾病患儿172例为研究对象,根据临床检查结果将其分为轻度感染组与重度感染组,同时选取同期该院诊断的非细菌感染性疾病患儿172例为非细菌感染组,另外选择同期在该院体检的正常儿童172例为对照组。检测所有研究对象超敏C-反应蛋白(hs-CRP)、降钙素原(PCT)和HBP水平。结果重度感染组、轻度感染组hs-CRP、PCT、HBP水平高于对照组与非细菌感染组,差异有统计学意义(P0. 05),其中重度感染组的hs-CRP、PCT、HBP水平最高。轻度感染组的hs-CRP曲线下面积为0. 632,阈值为3. 651,灵敏度为0. 551,特异度为0. 801,阴性似然值为0. 564,阳性似然值为2. 751; PCT曲线下面积为0. 785,阈值为0. 316,灵敏度为0. 601,特异度为0. 934,阴性似然值为0. 430,阳性似然值为8. 956; HBP曲线下面积为0. 845,阈值为6. 551,灵敏度为0. 726,特异度为0. 868,阴性似然值为0. 318,阳性似然值为5. 451。重度感染组hs-CRP曲线下面积为0. 992,阈值为3. 551,灵敏度为0. 851,特异度为0. 801,阴性似然值为0. 189,阳性似然值为0. 027; PCT曲线下面积为0. 850,阈值为0. 356,灵敏度为0. 976,特异度为0. 968,阴性似然值为4. 251,阳性似然值为29. 546; HBP曲线下面积为0. 998,阈值为12. 501,灵敏度为0. 996,特异度0. 982,阳性似然值为55. 333,阴性似然值为0. 004。鉴别轻、重度感染组时hs-CRP曲线下面积为0. 685,阈值为4. 951,灵敏度为0. 751,特异度为0. 576,阴性似然值为0. 436,阳性似然值为1. 766; PCT曲线下面积为0. 858,阈值为0. 396,灵敏度为0. 951,特异度为0. 676,阴性似然值为0. 075,阳性似然值为2. 924;HBP曲线下面积为0. 990,阈值为17. 751,灵敏度为0. 976,特异度为0. 951,阴性似然值为0. 027,阳性似然值为19. 501。回归系数中HBP的B值最大,说明HBP对轻、重度细菌感染的鉴别效能更高。结论 hs-CRP、PCT、HBP水平是检测早期细菌感染性疾病的敏感指标,且三者联合诊断效果更为准确。  相似文献

目的对比分析两种商品化培养液Cook和Vitrolife在辅助生殖中的临床结局。方法回顾性分析2014年5-6月在该院进行辅助生殖患者的临床资料。符合纳入标准的患者Cook组有43例,Vitrolife组有44例。比较两组的正常受精率、卵裂率、优质胚胎率、优质囊胚率、累积临床妊娠率、累积活产率、新生儿体质量和身长。结果两组患者一般资料比较,差异无统计学意义(P 0. 05)。Cook组与Vitrolife组各项临床结局数据比较,差异无统计学意义(P 0. 05),依次为正常受精率73. 26%vs. 68. 61%,卵裂率88. 89%vs. 90. 25%,优质胚胎率52. 96%vs. 48. 48%,优质囊胚率33. 59%vs. 36. 92%,累积临床妊娠率65. 12%vs. 65. 91%,累积活产率58. 14%vs. 52. 27%,单胎新生儿体质量(3. 4±0. 54) kg vs.(3. 15±0. 37) kg,双胎新生儿体质量(2. 53±0. 38) kg vs.(2. 54±0. 39) kg,单胎新生儿身长(49. 3±1. 22) cm vs.(49. 53±1. 92) cm,双胎新生儿身长(47. 1±2. 08) cm vs.(47. 00±2. 22) cm。结论两种商品化培养液的临床结局比较,差异无统计学意义。  相似文献

Objective To investigate the effects of methylation status of CpG islands of endogenous E-cadherin (CDH1) gene on the promoter activity of corresponding genes in reporter assays. Methods The methylation statuses of CpG island of CDHI in 8 different cell lines were detected by methylation-specific PCR. CDH1 protein was analyzed by Western blotting. Two sets of pGL3 reporter vectors with different genotypes/haplotypes of the CDH1 promoter were constructed [pGI3-A(-73)/-C(-73)pGL3-H1/-H4]and used to transfect these cell lines. The differences between these promoter reporter vectors were analyzed by t-test. Results (1) CDH1 CpG island was unmethylated in AGS, MCF7, MKN74, and PC-3 cell lines,expressed in MCF7, MKN74, and PC-3 ,but not in AGS. Expression of CDH1 was silenced by methylation in HeLa, BGC823, A549, and RKO cell lines. (2) In the four CDH1 -unmethylated MCFT, M KN74, PC-3, and AGS cell lines ,the promoter activities of pGI3-C(-73)(as 0. 78±0. 10,0. 17±0.01,0. 11±0. 01,1.19±0. 18)were significantly higher than those of pGL3-A(-73)(as 0. 30±0. 08,0. 07±0. 01,0. 07±0. 01,0. 39±0. 04) (t values are -6. 298, -12. 349, -8. 128, -7.388, and P<0. 01). Conclusion The methylation status of CpG island of the target gene in the tested cell lines affects the promoter activity in Reporter Assay significantly. Conclusion The methylation status of CpG island of the target gene in the tested cell lines affects the promoter activity in Reporter Assay significantly. Objective To investigate the metabonomic variation between patients with esophageal cancer (EC) and healthy controls, and to analyze the variation between patients with EC. 目的探讨红细胞体积分布宽度在4月龄内儿童筛查β地中海贫血的价值,并由此探讨改进新生儿地中海贫血筛查程序。方法回顾分析广东东莞地区1 196例4月龄内儿童静脉血样本,均进行全血细胞分析和基因验证,其中α、β地贫组各400例,常见α、β基因均阴性组396例。分别对3个组红细胞体积分布宽度标准差(RDW-SD)、分布宽度变异系数(RDWCV)进行对比,使用受试者工作曲线(ROC曲线)得出β地贫RDW-SD、RDW-CV的筛查诊断范围。结果α地贫、β地贫、阴性组静脉血样本RDW-CV值分别为17. 34±2. 40、20. 32±2. 77、15. 49±1. 68,RDW-SD值分别为46. 07±5. 23、60. 50±8. 63、49. 02±5. 99,各组差异均有统计学意义(P0. 05)。β、α地贫组RDW-CV最佳截断值18. 45,曲线下面积(AUC)为0. 797,灵敏度68. 8%,特异度74. 8%,约登指数(YI) 0. 435; RDW-SD最佳截断值53. 45,AUC 0. 917,灵敏度92. 5%,特异度86. 2%,YI 0. 788。β地贫组、阴性组RDW-CV最佳截断值17. 05,AUC 0. 942,灵敏度90. 0%,特异度88. 9%,YI 0. 789;RDW-SD最佳截断值54. 35,AUC 0. 879,灵敏度83. 8%,特异度84. 5%,YI 0. 683。α地贫组、阴性组RDW-CV最佳截断值16. 45,AUC 0. 745,灵敏度64. 3%,特异度82. 3%,YI 0. 466; RDW-SD最佳截断值48. 55,AUC 0. 666,灵敏度72. 3%,特异度55. 3%,YI 0. 276。结论 4月龄内儿童RDW-CV、RDW-SD能有效地将β地贫从α地贫和正常儿中区分。以MCV80fL和(或) MCH27 pg和(或) RDW-CV16. 45%为筛查条件筛查出可疑的α、β地贫患儿,如果怀疑是α、β地贫患儿可以RDWSD53. 45fL为条件得到可疑β地贫患儿,并行血红蛋白电泳,最终基因检测确诊。此筛查程序能更好的减少β地贫的遗漏,促进新生儿地贫筛查事业的发展,适合在基层医院推广。  相似文献
