The present study was conducted to assess the risk to human health by heavy metals (Cd, Cu, Pb, Zn, Ni and Cr) through the intake of locally grown vegetables, cereal crops and milk from wastewater irrigated site. Milk is not directly contaminated due to wastewater irrigation, but is an important route of food chain transfer of heavy metals from grass to animals. Heavy metal concentrations were several fold higher in all the collected samples from wastewater irrigated site compared to clean water irrigated ones. Cd, Pb and Ni concentrations were above the ‘safe’ limits of Indian and WHO/FAO standards in all the vegetables and cereals, but within the permissible limits in milk samples. The higher values of metal pollution index and health risk index indicated heavy metal contamination in the wastewater irrigated site that presented a significant threat of negative impact on human health. Rice and wheat grains contained less heavy metals as compared to the vegetables, but health risk was greater due to higher contribution of cereals in the diet. The study suggests that wastewater irrigation led to accumulation of heavy metals in food stuff causing potential health risks to consumers.  相似文献

Multixenobiotic resistance mechanisms (MXR) related to the mammalian P-glycoprotein multidrug transporter protein (P-gp) are known to occur in several marine invertebrates. In the present work, we report on the induction of an MXR protein by various heavy metals in the gills of the freshwater clam Corbicula fluminea. The evaluation of the MXR protein level was assessed by Western blot using a specific monoclonal antibody raised against the human P-gp (C219). A field transplantation experiment, where clams were caged in a gradient relative to an industrial site, demonstrated a positive relationship between MXR levels and (a) metal pollution (Cd and Zn) in the environment and (b) metal bioaccumulation in the gills. To establish this correlative relationship, clams were exposed to different levels of cadmium (15-60 microg l(-1)) for up to 15 days in a controlled laboratory experiment. MXR protein levels increased in time for all treatments (including the control). However, the highest levels of MXR protein titer were expressed in clams that had been exposed to the lowest dose of cadmium. The causes for this observed inverse relationship between the exposure dose and the MXR induction is discussed. MXR protein titer was also shown to be induced by other heavy metals (zinc, inorganic mercury, and copper).  相似文献

Dairy cattle on a farm located in the vicinity of a lead and zinc-ore processing factory were studied over 21 mo and compared with cattle on a control farm. Mean daily intakes of lead from the diet were 4.3 mg/kg body weight, with great variations; mean daily zinc intakes were 5.6 mg/kg body weight; and mean daily cadmium intakes were 0.064 mg/kg body weight. The 3 major indicators of contamination were blood lead concentrations, with mean values of 50 micrograms/100 ml of blood, zinc protoporphyrin with mean values of 165 micrograms/100 ml blood, and lead concentrations in hair which averaged 10 micrograms/g. Blood zinc concentrations and zinc concentrations were not significantly increased. One cow developed fatal post-partum paralysis. Liver, kidney and bone lead concentrations and kidney cadmium concentrations were good "post-mortem" indicators of exposure.  相似文献

Vegetables (Beta vulgaris L., Abelmoschus esculentus L. and Brassica oleracea L.) from the production and market sites of India were tested for Cu, Cd, Zn and Pb. At market sites, the mean concentration of Cu in cauliflower, and of Zn and Cd in both palak and cauliflower had exceeded the PFA standard. Zn at the production sites also exceeded the PFA standard in cauliflower. Cd concentration in vegetables tested from both production and market sites was many folds higher than the EU standard. In contrast, Pb in vegetables tested from both production and market sites was below the PFA limit, but was considerably higher than the current EU and WHO standards. Heavy metals accumulation in vegetables tested are higher at market sites than those at the crop production sites. The contributions of these vegetables to dietary intake of Cu, Zn, Cd and Pb were 13%, 1%, 47% and 9% of provisional tolerable daily intake, respectively. The study concludes that the transportation and marketing systems of vegetables play a significant role in elevating the contaminant levels of heavy metals which may pose a threat to the quality of the vegetables with consequences for the health of the consumers of locally produced foodstuffs.  相似文献

The behavioural and electrocortical (ECoG) effects induced by a methanol extract of the skin of the Australian frog Pseudophryne coriacea, directly microinjected into several areas of the brain, were studied in freely moving rats. Administration of the P. coriacea extract (5, 10, 15 and 20 micrograms) into the dorsal hippocampus produced a dose-dependent and reversible behavioural stimulation and ECoG spikes lasting 20-140 min. Similar but less intense effects were elicited in rats receiving injections into the III cerebral ventricle, amygdala and caudate nucleus. In conclusion, the present experiments show that the skin extract of P. coriacea produces behavioural stimulation and ECoG spikes when injected into the rat brain, the most sensitive area being the hippocampus.  相似文献

Pathological changes induced by cadmium aerosol had features common to the changes evoked by oxidants. Female rats were exposed to fumes of lead, antimony, zinc and cadmium (15–100 nmoles/m3). One hour after termination of exposure, superoxide dismutase (SOD) activity in erythrocytes of the exposed rats lowered by 15–40%. SOD activity of lung lavage fluid also lowered by 20–35% at the 2nd day after the exposure. The inverse value of SOD activity (l/SOD) in erythrocytes and of lung lavage fluid were proportional to the molar exposure level adjusted by the particle size (Dixon plot), irrespective of the difference of the exposed substance. The ratio of dry weight to wet weight of the lung was 4.3–26% lower than the control value on the later period after the exposure. With the heavy metal exposure, the uptake of the exposed metal was found to be proportional to the endogenous zinc concentration, which was correlated well with the change of SOD in the lung and in erythrocytes. Traffic-related particulate matter (PM) is associated with adverse health effects. Quinones present in the traffic-related PM are hypothesized to contribute to these harmful effects through reactive oxygen species (ROS) generation. However, the impacts of the traffic-related PM and quinones on inflammatory processes and genotoxic damages are less well known. In present study we aimed to examine the genotoxic and inflammatory impacts of organic extracts from traffic-related PM (oTRP) in human lung epithelial A549 cells, and reveal the contributions from quinones. Significant cytotoxicity and DNA damage were caused by oTRP. The pro-inflammatory genes, interleukin-6 (Il-6), interleukin-8 (Il-8) and tumor necrosis factor (Tnf), and two aromatic hydrocarbon receptor-regulated genes, Cyp1a1 and 1b1, were significantly up-regulated by oTRP. A concomitant increase in ROS was observed, suggesting that oTRP may mediate genotoxic and inflammatory effects through oxidative stress pathway. Second, the effects from two typical airborne quinones, 9,10-anthraquinone (AQ) and 1,4-naphthroquinone (NQ) were compared. NQ, but not AQ, induced significant DNA damage in A549 cells. NQ up-regulated Il-8, Tnf, and Mcp-1 genes, while AQ induced the expression of Rantes gene. These results suggest that the NQ and AQ may participate in the pro-inflammatory responses through releasing different types of cytokines/chemokines.  相似文献

This report describes the effects of low levels of copper, nickel and lead salts on the concentrations of alpha-fetoprotein (AFP) in the sera and amniotic fluid of pregnant Nylar mice. During the early and mid-gestation (9-17 days), pregnant mice were injected intraperitoneally twice with heavy-metal salt solutions and were autopsied two days following the second injection. Maternal sera and amniotic fluid (AF) were collected and AFP levels were quantified by radial immunodiffusion. Metal levels determined by X-ray fluorescence spectroscopy in individual samples confirmed the presence of trace metals in the fetus. Low doses of nickel and copper were associated with elevated AFP levels in amniotic fluid in 15-17 day pregnant animals, while maternal serum AFP levels mostly remained unchanged. Decreased concentrations of maternal serum AFP occurred with increased doses of copper and lead in contrast to elevated concentrations of AFP in amniotic fluid. Furthermore, there was an increase in fetal wastage when higher doses of copper and lead were administered. A reduction of secondary litter size (F1 generation) with low dosage levels of lead was also observed. These results imply that the fetal-maternal transfer of AFP may either be impaired or reflect increased leakage or decreased placental permeability in the presence of sublethal doses of copper and lead. These findings suggest that the parallel measurements of AFP concentrations in sera and amniotic fluid might be employed for assessment of embryo- and fetotoxicity when heavy metal intake is suspected during pregnancy.  相似文献

A 1973 environmental accident in Michigan resulted in exposure of humans via the food chain to polybrominated biphenyl (PBB). To better characterize the toxicity of the halogenated polycyclic aromatic hydrocarbon class of chemicals, rodents were dosed with PBB and their target organs examined for morphological, histological, biochemical, and selected endocrine changes. Male and female rats and mice were given 125 po doses of PBB over a 6-month period at 0.1, 0.3, 1.0, 3.0, and 10.0 mg/kg of body weight/day (5 days/week). There was a dose-related decrease in body weight gain in both male and female rats and male mice. Thymus weights were significantly decreased in all rats exposed to 0.3 mg/kg or more of PBB. Dose-related hepatotoxic effects were observed in both rats and mice characterized by marked increase in liver weight with accentuation of hepatic lobular markings. Microscopically, there were moderate to marked swelling, disorganization, and single cell necrosis of hepatocytes, fatty infiltration, bile duct proliferation, and presence of atypical hyperplastic foci. Hepatic porphyrin levels were markedly increased in both rats and mice primarily in females. There was a significant decrease in serum thyroxine (T4) and triiodothyronine (T3) suggesting that PBB may interfere with thyroid hormone secretion. There was a significant dose-related increase in serum cholesterol and gamma-glutamyl transpeptidase, and a decrease in serum glucose.  相似文献

Sperm-positive Sprague-Dawley rats received one of four treatments for 20 days beginning within 24 hours of conception. One group received subcutaneous injections of 15 mg/kg cocaine twice daily (Cocaine-D); a second group received 15 mg/kg cocaine twice daily for two consecutive days at 5-day intervals (Cocaine-I); a third group received normal saline twice daily (Saline); and a fourth group received 1.5 mg/kg amfonelic acid (AFA), a dopamine reuptake inhibitor, once daily. Cocaine-D, Cocaine-I, and AFA dams were fed ad lib. An attempt was made to pair-feed the Saline dams with the Cocaine-D dams; however, the Saline dams did not eat as much as the Cocaine-D dams which resulted in dams in all groups essentially eating ad lib. The Cocaine-D pups showed a slightly delayed righting behavior and neophobia at 30 days of age, as evidenced by hypoactivity during the first 15 min of a 6-h activity test. The Cocaine-I pups were hypoactive during the 3-h dark phase of the 6-h activity test when tested at 30 days of age. These effects did not occur in the offspring exposed to AFA, a potent dopamine uptake inhibitor and CNS stimulant which indicate that one or more other sites for cocaine action may combine for its effects on the developing fetus.  相似文献

Mercury poisoning occurred after the acute, prolonged exposure of 53 construction workers to elemental mercury. Of those exposed, 26 were evaluated by clinical examination and tests of neuropsychological function. Patients received treatment with chelation therapy in the first weeks after exposure. Eleven of the patients with the highest mercury levels were followed in detail over an extended period. Observations included the evaluation of subjective symptoms of distress, using the 'Symptom Check List 90-Revised' (SCL-90R) and tests of visual-motor function such as 'Trailmaking Parts A and B', 'Finger Tapping', 'Stroop Colour Word Test' and 'Grooved Pegboard.' On day 85 +/- 11 (mean +/- s.d.) after exposure, these 11 men again received either 2,3-dimercaptosuccinic acid (DMSA) or N-acetyl-D, L-penicillamine (NAP) in a short-term study designed to compare the potential to mobilize mercury and the incidence of drug-induced toxicity of these two chelating agents. Rapidly resolving metal fume fever was the earliest manifestation of symptoms. CNS symptoms and abnormal performance on neuropsychological tests persisted over the prolonged period of follow-up. There were significant correlations between neuropsychological tests and indices of mercury exposure. Serial mercury in the blood and urine verified the long half-life and large volume of distribution of mercury. Chelation therapy with both drugs resulted in the mobilization of a small fraction of the total estimated body mercury. However, DMSA was able to increase the excretion of mercury to a greater extent than NAP. These observations demonstrate that acute exposure to elemental mercury and its vapour induces acute, inorganic mercury toxicity and causes long-term, probably irreversible, neurological sequelae.  相似文献

This study determined the heavy metals (HMs) accumulation in different vegetables in different seasons and attributed a serious health hazard to human adults due to the consumption of such vegetables in Jhansi. The total amounts of zinc (Zn), lead (Pb), nickel (Ni), manganese (Mn), copper (Cu), cobalt (Co), and cadmium (Cd) were analysed in 28 composite samples of soil and vegetables (Fenugreek, spinach, eggplant, and chilli) collected from seven agricultural fields. The transfer factor (TF) of HMs from soil to analysed vegetables was calculated, and significant non-carcinogenic health risks due to exposure to analysed heavy metals via consumption of these vegetables were computed. The statistical analysis involving Principal Component Analysis (PCA) and Pearson's correlation matrix suggested that anthropogenic activities were a major source of HMs in the study areas. The target hazard quotient of Cd, Mn, and Pb for fenugreek (2.156, 2.143, and 2.228, respectively) and spinach (3.697, 3.509, 5.539, respectively) exceeded the unity, indicating the high possibilities of non-carcinogenic health risks if regularly consumed by human beings. This study strongly suggests the continuous monitoring of soil, irrigation water, and vegetables to prohibit excessive accumulation in the food chain.  相似文献

An investigation of total arsenic in food composites, collected from the villagers, was carried out in arsenic-affected areas of the Murshidabad district, West Bengal where the agricultural system is mostly groundwater dependent. The shallow, large-diameter tubewells installed for agricultural irrigation contain an appreciable amount of arsenic (mean 0.085 mg/l, n=6). Even the soil is arsenic-contaminated (mean 11.35 mg/kg, n=36), so some arsenic can be expected in the food chain from crops cultivated in this area. The results revealed that the individual food composite and food groups containing the highest mean arsenic concentrations (microg/kg) are potato skin (292.62 and 104), leaf of vegetables (212.34 and 294.67), arum leaf (331 and 341), papaya (196.50 and 373), rice (226.18 and 245.39), wheat (7 and 362), cumin (47.86 and 209.75), turmeric powder (297.33 and 280.9), cereals and bakery goods (156.37 and 294.47), vegetables (91.73 and 123.22), spices (92.22 and 207.60) and miscellaneous items (138.37 and 137.80) for the Jalangi and Domkal blocks, respectively. Arsenic is absorbed by the skin of most of the vegetables. The arsenic concentration in fleshy vegetable material is low (mean 2.72 microg/kg, n=45). Higher levels of arsenic were observed in cooked items compared with raw. Daily dietary intakes of arsenic (microg) from the foodstuffs for adults are 171.20 and 189.13 and for children are 91.89 and 101.63 in the Jalangi and Domkal blocks, respectively.  相似文献

There are conflicting reports in the literature concerning the immunotoxic effects of polychlorinated biphenyls (PCB) on cell-mediated immune responses. In the studies reported here, we show that the in vivo generation of cytotoxic T lymphocytes (CTL) in response to allogeneic tumor challenge is sensitive to suppression by 3,4,5,3',4',5'-hexachlorobiphenyl[(345)2-HxCB], a poorly metabolized, toxic, Ah receptor-binding PCB isomer. C57B1/6 mice treated with a single oral dose of (345)2-HxCB two days prior to the i.p. injection of allogeneic P815 tumor cells exhibited a dose-dependent reduction in peak CTL activity in the spleen. When examined on a kinetic basis, the CTL response was reduced in magnitude on all days examined with no evidence for a shift in the kinetics of the response induced by (345)2-HxCB exposure. (345)2-HxCB exposure prior to antigen challenge (day -14, -7, or -1 relative to P815 injection on day 0) produced significant suppression of the CTL response. (345)2-HxCB exposure 6 weeks prior to antigen challenge was still significantly suppressive, although the reduced degree of suppression suggested that recovery was in progress. When (345)2-HxCB exposure occurred after antigen challenge, significant suppression was produced only when exposure occurred within the first three days of the response, suggesting that, as the CTL matured, their sensitivity to (345)2-HxCB diminished. Clearance of the allogeneic tumor cells from the peritoneal cavity was delayed in (345)2-HxCB-treated mice and was associated with an altered composition of the white blood cell infiltrate in the peritoneal cavity. Symptoms of overt toxicity as well as immunotoxicity were apparent at lower doses of (345)2-HxCB in male as compared to female mice. In addition, interactive effects of (345)2-HxCB exposure and P815 antigen challenge on body weight and thymic involution were observed in both male and female mice. Possible mechanisms for the altered CTL response in (345)2-HxCB-exposed mice are discussed.  相似文献

The effects of buffering ions, pH, temperature, and various metal cations on the activity of avian red blood cell aminolevulinic acid dehydratase (RBC-ALA-d) in blood hemolysates was studied. The pH optimum of the enzyme was approximately 6.6 in both citrate and morpholinethanesulfonic acid (MES) buffers, and maximal activity was comparable in both buffer systems. Enzyme activity was increased by about 20% at 42 degrees C (avian body temp.) relative to that at 37 degrees C. Pb2+ was 10-100-fold more potent than Cu2+, Cd2+, Hg2+ or CH3Hg+ as an inhibitor of RBC-ALA-d activity. The IC50 for Pb2+ was 0.03-0.04 mumol/ml blood. Zn2+ added to Pb-pretreated hemolysates was able to induce up to 65% recovery of enzyme activity. Pb2+ was a more effective inhibitor of delta-aminolevulinic acid dehydratase (ALA-d) activity in MES buffer than in citrate buffer, possibly because of chelation of Pb2+ by citrate.  相似文献

Tetrabromobisphenol A (TBBPA), brominated flame retardant, is produced in the largest amounts globally for use in plastics or building materials. TBBPA has been detected in sediment, air at the dismantling plant or human serum samples. In the present study, we examined the effects of prenatal and postnatal exposure to TBBPA in mice. TBBPA (99.1% pure) in diet was administered to pregnant ICR mice at doses of 0% (control), 0.01%, 0.1% or 1.0% from gestational day 0 to weaning at postnatal day 27. The average daily food intake and body weight of dams showed no significant differences between the control and treated groups. There were no dose-related effects on reproductive data. Serum concentrations of total-cholesterol and liver weights of treated dams and offspring were higher than those of the control mice. Histological findings in treated dams or offspring showed the increase of focal necrosis of hepatocytes and inflammatory cell infiltration in the liver, and increase of dilation or atrophy of renal tubules and cyst in the kidney. TBBPA was developed as a new, safe class of flame retardant and was not highly toxic. However, the present data suggested that TBBPA caused a lipid metabolic disorder and hepatic or kidney lesion, under these conditions.  相似文献

The Caloosahatchee Estuary has been exhibiting signs of impaired ecological health due to the extensive hydrological alteration, agricultural land use, and increasing watershed development. This project investigated the responses of the American oyster, Crassostrea virginica at five locations in the Caloosahatchee River in relation to salinity changes, levels of heavy metals, pesticides, and PCBs in the water as well as in the oyster tissue. Individual heavy metal and organochlorine pesticide concentrations in oysters varied significantly between sampling locations and sampling months. PCB concentrations in oyster tissues were below detection limits as were the metal, pesticide and PCB concentrations in water at all the sampling locations. Both heavy metal and pesticide concentrations decreased with increasing distance downstream indicating upstream source of contaminants. The highly pathogenic oyster parasite Perkinsus marinus infection intensity (level) and prevalence (% infected oysters), condition index, spat recruitment, and gonadal index showed a seasonal trend varying with spawning activity and increased downstream. However, juvenile oyster growth was higher at upstream estuarine locations. Oyster responses varied more with seasonal programming (salinity), rather than due to contaminant levels. While significant correlations were noted between some oyster responses and metal concentrations in oyster tissues, overall metal concentrations were low compared to national averages. It appears that oyster health in the Caloosahatchee River is influenced more by freshwater inflow and resulting salinity fluctuations, rather than due to the measured contaminants.  相似文献
