Detrimental Effect of Free Radicals on Tissues in Rat with Burn Shock

A model of burn shock was reproduced in rats and the changes of water content andmalondialdehyde(MDA)concentration in tissues were measured.The results showed that there wasa significant increase of MDA concentration and water content in lung and bumed skin post burnand a positive correlation was found between MDA level and water content.Pretreatment withsuperoxide dismutase(SOD)prevented effectively lung and skin from edema.The role of oxygen freeradicals in vascular permeability disturbance and development of edema in burns was discussed.

Expression of Heat Shock Protein 70 and 27 in Non-small Cell Lung Cancer and Its Clinical Significance

The heat shock proteins （HSPs） 70 and HSP 27 expression in patients with non-small cell lung cancer （NSCLC） was studied and the relation.ship between HSP 70 and HSP 27 with the clinicopathological features of NSCLC was investigated. The expression of HSP 70 and HSP 27 was detected in tumor tissues from 60 patients with NSCLC by S-P immunohistochemistry. The findings were analyzed in combination with the histological types, histopathological differentiation, lymph node metastasis, patients＇ clinical stages, smoking history and gender. The results showed that of the 60 NSCLC tissue specimens studied, the immunoreactivity of HSP 70 and HSP 27 was detected in 47 （78.3 %） and 43 （71.7 %） specimens, respectively. A positive correlation was found between the overexpression of HSP 70 and HSP 27. The histopathological differentiation, lymph node metastasis, clinical stages and smoking history were correlated to the expression of HSP 70, but not to the expression of HSP 27. No statistical significance was observed in histological types and gender with respect to both HSP 70 and HSP 27 expression. It is suggested that the HSP 70 expression is a powerful and significant prognostic indicator and is related to histopathological differentiation, lymph node metastasis, patients＇ clinical stages, smoking history, whereas HSP 27 expression is not.     

Micro-computerized Three-dimensional Reconstruction of the Palmar Arteries and Its Clinical Significance

Thirty adult upper extremities were used to study the arteries of the hand byangiography,cross-sectional dissection and three-dimensional reconstruction withmicrocomputer.The arterial skeleton of the palm was composed of superficial,deep anddorsal planes.There were three main anastomotic pathways between the radial and ulnararteries or their branches in the radio-ulnar direction,while in the palmar dorsal dirce-tion there were three anastomotic zones between the palrnar and dorsal arterial plancs.The branches of the deep palmar arch radiated distally,proximally,palmarly anddorsally,so that the arch acted as the center of the arterial network of the hand.Thetransverse arterial allocations in the carpo-metacarpal segment of the palm were four intypes,but in the mid-palmar and metacarpo-phalangcal segments were three only.Accord-ing to the sources,cross-sectional area,irrigation territories and anastomoses of the ar-teries in each segment,the clinical significance in the rcplantation of the amputatcd palmshas been discussed     

Expression of Telomerase Gene mTR in the Testis of SD Rats and Its Significance

Summary:To study the expression of mTR gene in the testis of SD rats with varied ages and its sig-nificance,in situ hybridization(ISH)techniques were applied to detect the expression of telomerasegene mTR mRNA in the testis of SD rats.The expression of mTR was found in testes of different-age male SD rats.There was a positive correlation between the expression of mTR and the location ofgerm cells(spermatogonia,spermatocyte,spermatid).In Setoli cells,leydig cell and spermatozoa,no telomerase mTR was detectable.Type A spermatogonia expressed the highest level of telomerasemTR mRNA.It was suggested that the expression of mTR gene in the testis of SD rats is of lifetimeand coincides with the telomerase activity.

Inhibitory Effect of Allopurinol on the Generation of Free Radicals in Reperfused Rat Brain

Free radicals in ischemic and reperfused rat brain were measured by electron spinresonance(ESR) spectrometer.The inhibitory effect of allopurinol on the free radical genera-tion in reperfused rat brain was observed.The experimental results revealed that the free radi-cal content of the brain in the ischemia group was markedly higher than that in the control group(P<0.01),that in the reperfusion group was markedly higher than that in the ischemia group(P<0.01)and that in the allopurinol group was markedly lower than that in the reperfusiongroup(P<0.01).These results suggest that free radicals increase.greatly after cerebralischemia-reperfusion and that allopurinol plays a certain inhibitory role in the free radical genera-tion in the reperfused rat brain.

Influence of Electroacupuncture on the mRNA of Heat Shock Protein 70 and 90 in Brain after Cerebral Ischemia/Reperfusion of Rats

To explore the anti-apoptotic role of electroacupuncture (EA) and its molecular mecha-nisms after cerebral ischemia/reperfusion (IR) of rats, by using animal model of middle cerebralartery occlusion (MCAO), the changes of the cleavage of PARP were observed by Western blot andthe mRNA of heat shock protein (Hsp) 70 and Hsp90β detected by competitive RT-PCR after cere-bral IR and EA treatment. The results were as follows: (1) The cleavage of PARP was increased inischemic hippocampus, and EA treatment could attenuate the level of the cleavage remarkably; (2)The mRNA expression of Hsp70 was increased in the ischemic cortex and hippocampus and was fur-ther increased after EA treatment; (3) The mRNA expression of Hsp90β was decreased in ischemiccortex and hippocampus and the decrease was relatively slight after EA treatment. The above resultsdemonstrated EA treatment could protect neurons from apoptosis after cerebral IR. One of the molec-ular mechanisms was the promotion of the inducible expression of Hsp7     

Expression and Clinical Significance of p27kipl Protein in Primary Liver Cancer

To investigate the expression and clinical significance of p27kip1 protein in primary liver cancer, the expression of p27kip1 protein and the relationship with clinicopathological factors were studied in primary liver cancer by using SABC immunohistochemical staining in specimens of 40 cases of primary liver cancer and 20 cases of liver cirrihosis. Our results showed that positive expression rate of p27kip1 protein in primary liver cancer was 37.5% (15/40), which was lower than that in benign lesion of liver 80.0% (16/20, P<0.01). The expression level of p27kip1 protein in primary liver cancer showed significant differences in tumor size, Edmonson histological grade, portal invasion, lymph node metastasis, TNM stage (P<0.05, for all), but not significantly correlated with patient's age and histological types. Log rank test showed that the p27kip1 expression was significantly related with prognosis of the patients (P<0.05), and the prognosis of the patients with p27kip1 positive expression was markedly better than that of those with p27kip1 negative expression. It is concluded that the expression of p27kip1 was significantly related clinicopathological factors of primary liver cancer. p27kip1 protein may be used as a novel tumor marker for primary liver cancer.
