川芎嗪对急性放射损伤小鼠骨髓细胞粘附分子作用研究   总被引:1,自引:0,他引:1  
为探讨急性放射损伤小鼠骨髓细胞粘附分子表达及川芎嗪对其影响,将27只昆明小鼠平均分为3组,正常组,急性放射损伤对照组,川芎嗪组。将对照组及川芎嗪组习用8.0GY ^60Coγ射线照射后,分别胃饲生理盐水及25mg/ml川芎嗪注射液0.2ml/只,每天2次,连续天,第7天用流式细胞仪检测所有小鼠骨髓单个核细胞CD49d、CD49c的表达1的表达显著低于正常组(P〈0.01,P〈0.001),川芎唪组  相似文献

为探讨急性放射损伤小鼠骨髓细胞粘附分子表达及川芎嗪对其影响,将27只昆明小鼠平均分为3组:正常组,急性放射损伤对照组,川芎嗪组。将对照组及川芎嗪组小鼠用8.0Gy60Coγ射线照射后,分别胃饲生理盐水及25mg/ml川芎嗪注射液0.2ml/只,每d2次,连续6d。第7d用流式细胞仪检测所有小鼠骨髓单个核细胞CD49d、CD49e的表达水平,用免疫组织化学SABC方法检测骨髓基质细胞VCAM-1的表达、结果发现急性放射损伤对照组CD49d、VCAM-1的表达显著低于正常组(P<0.01,P<0.001),川芎嗪组CD49d、CD49e、VCAM-1的表达均显著高于对照组(P<0.01,P<0.001),且CD49d、CD49e高于正常组。结果显示川芎嗪能增强急性放射损伤小鼠骨髓造血细胞和基质细胞粘附分子的表达,有利于造血细胞的增生。  相似文献

给昆明小鼠8Gy60Coγ射线照射后,立即腹腔注射100%复方活血汤注射液(每ml含生药1g)0.2ml/只,每d2次,连续10d。分别于照射后第3、7、14d测定活体原位尺骨骨髓氧分压,观察相同部位骨髓切片组织学。结果表明,常用活血化瘀中药能促进急性放射损伤小鼠骨髓微血管恢复、增生,扩张其面积,使骨髓氧分压明显增高,增加骨髓微环境供氧,促进造血细胞增生。  相似文献

川芎嗪对大剂量化疗小鼠骨髓基质细胞损伤的保护作用   总被引:1,自引:1,他引:0  
目的:观察川芎嗪对大剂量甲氨喋呤(MTX)化疗所致骨髓基质细胞(BMSC)损伤的保护作用及其量效关系。方法:建立大剂量MTX化疗损伤BMSC的小鼠模型后,按每千克体重腹腔注射川芎嗪0mg(对照组)、20mg(Ⅰ组)、40mg(Ⅱ组),1次/d,第21天采取骨髓细胞进行体外BMSC培养,成纤维细胞集落(CFU-F)计数,BMSC层粘附率测定和BMSC层上造血细胞混合集落培养,结果:BMSC生长,CFU-F计数,BMSC层粘附率,BMSC层上造血细胞混合集落数目均表现为Ⅱ组>Ⅰ组>对照组(P<0.05,P<0.01)。结论:川芎嗪对大剂量MTX引起的BMSC损伤有明显的保护作用,且呈剂量依赖性。  相似文献

目的 观察刺五加注射液对放射损伤小鼠骨髓造血微环境的保护作用。方法 ①60 Coγ射线辐射小鼠建立骨髓微血管放射损伤模型 ;②取 6周龄左右雌性小鼠随机分为正常、模型、模型 刺五加注射液低剂量、模型 刺五加注射液高剂量和模型 复方丹参注射液 5组 ,采用分光光度计法分别测定提取的各组小鼠股骨骨髓血红蛋白溶液标本的光密度(OD)值 ;鼠尾采血测定外周血红细胞、白细胞数量和血红蛋白含量 ;并分别观测骨髓毛细动脉根数、面积和骨髓造血组织容量百分率。结果 刺五加注射液能明显降低放射损伤小鼠骨髓血红蛋白含量、提高外周血红细胞和白细胞的数量以及增加骨髓毛细动脉根数、面积和骨髓造血组织容量百分率。结论 刺五加注射液能明显修复放射损伤小鼠骨髓微血管 ,保护骨髓造血微环境  相似文献

取5.0Gyγ射线照射后3d和7d的小鼠骨髓体外液体培养14d和21d,显示骨髓基质细胞仍能贴壁生长,但骨髓基质细胞集落(CFU-F)数量显著低于正常,照后7d的CFU-F数量虽比照后3d的CFU-F数量有所增加,但恢复缓慢;延长培养时间,显示照射鼠的骨髓基质细胞增殖受抑。流式细胞仪检测结果表明G2+M期细胞和DNA含量显著低于正常对照组;随着照后时间的延长,S、G2+M期含量有所恢复,但仍显著低于正常对照组。说明骨髓造血基质细胞具有较高的辐射敏感性且对辐射损伤持续较久。  相似文献

Summary: The effect of ligustrazine on the expression of CD31 in syngenic bone marrow transplantation (BMT) mice was studied. Fifty-six Balb/c mice were divided into 3 groups: normal control group. BMT control group, and ligustrazine treated group. Syngenic BMT mouse models were established according to the literatures. In BMT control group and the ligustrazine treated group, the mice were given respecxively orally 0.2 mL saline and 2 mg ligustrazine twice a day. On the 7th, 14th, and 21st day after BMT, the mice were killed. The expression of CD31 on the surface of bone marrow nuclear cells (BMNC) was detected by flow cytometry. Peripheral blood leukocytes, platelets and BMNC were counted. Histological observation of bone marrow was made. The results showed thai in ligustrazine treated group the peripheral blood leukocylcs, platelets and BMNC counts, and the expression of CD31 on the day 7, 14, 21 after BMT were higher than in BMTcontrol group (P〈0.01 or P〈0.05). In conclusion, ligustrazine could obviously enhance the CD31 expression on the surface of BMNC after syngcnic BMT in mice, which may be one of the mecha- nisms underlying the ligustrazine accelerating hematopoietic reconstitution in syngenic BMT.  相似文献

目的:研究川芎嗪对人骨髓成纤维样基质细胞增殖及蛋白质表达的影响,探讨川芎嗪补血作用的分子机制。方法:分别采用流式细胞术和蛋白质组学方法测定川芎嗪对人骨髓成纤维样基质细胞周期及蛋白质表达的影响。结果:川芎嗪能促进人骨髓成纤维样基质细胞由G0/G1期进入S期,并使该细胞的9种蛋白质表达上调,6种下调。结论:川芎嗪能促进人骨髓成纤维样基质细胞增殖,作用于多个蛋白质靶点,间接发挥补血作用。  相似文献
