目的探讨慢性肝囊型包虫持续感染患者外周血单核细胞的亚群组成及其功能状态。方法囊型包虫病患者(CE组)和健康者(NC组)各15例,采集外周血单核细胞(PBMC)后分成两部分,一部分应用流式细胞术分析其表面CD14、CD16的阳性表达率;另一部分分为未干预组和重组抗原B(rAgB)干预组,干预后ELISA法检测上清中IFN-γ、IL-12p70、IL-10、IL-4的浓度。结果 PBMC占外周血白细胞的比例在CE组、NC组分别为(13.42±3.90)%和(6.23±1.39)%,差异有统计学意义(t=6.17,P<0.05);CD14阳性表达率分别为(19.90±6.96)%和(45.61±14.25)%,差异有统计学意义(t=6.28,P<0.05);CD16阳性表达率分别为(22.75±8.99)%和(22.83±6.77)%,差异无统计学意义(t=0.03,P>0.05)。除NC组rAgB干预后的IL-4与未干预对照相比差异无统计学意义外,来源于NC组、CE组rAgB干预后的4种细胞因子浓度与未干预对照相比差异均有统计学意义(P均<0.05)。CE组与NC组相比,rAgB干预下的IFN-γ、IL-12p7...  相似文献

目的研究人外周血单核细胞(PBMC)人白细胞抗原DR的表达和单核细胞体外分泌细胞因子水平的增龄性改变。方法352例不同年龄的体检者按年龄分为A组120例(20-69岁)、B组85例(70-79岁)、c组98例(80-89岁)、D组49例(90~102岁)。用流式细胞仪检测PBMC表面人白细胞抗原(HLA)-DR的表达;每组随机抽取10人,用ELISA方法检测经内毒素(LPS)N激前后PBMC分泌肿瘤坏死因子-α(TNF—α)、白细胞介素(1L)-6、IL-10等细胞因子的水平,并比较各年龄组PBMC表面HLA—DR表达的变化以及分泌TNF-α、IL-6、IL-10等细胞因子水平的变化。结果A,B,C,D各组之间PBMC表面HLA—DR的表达有显著性差异(F=159.712,P=0.000),且不同年龄组人群PBMC表面HLA—DR的表达呈随龄性增加(r=0.744,P=0.000);LPS刺激前各年龄组人群PBMC分泌TNF-α,IL-6,IL-10细胞因子的水平无显著性差异(P〉0.05);LPS刺激后各年龄组人群PBMC分泌TNF-α,IL-6,IL-10细胞因子的水平较LPS刺激前均有明显增高(P〈0.05);将LPS刺激后各组PBMC分泌细胞因子的水平进一步行组内比较发现,与A组相比,B,C,D各组分泌TNF-α、IL-6的水平明显减低(P〈0.05),分泌IL-10的水平明显增高(P〈0。05)。结论人PBMC细胞免疫功能呈随龄性改变,其表面HLA—DR的表达呈随龄性增加;LPS刺激前各年龄组人群PBMC分泌TNF-α,IL-6,IL-10细胞因子的水平无显著性差异;而LPS刺激后,70岁以上老年人PBMC分泌TNF-α和IL-6的水平明显减低,分泌IL-10的水平明显增高。  相似文献

目的 通过检测囊性包虫病(cystic echinococcosis,CE)患者外周血中Treg细胞表面PD-1的表达与Treg细胞相关因子Foxp3、TGF-β和IL-10的表达情况,探讨负性共刺激分子PD-1与Treg细胞在CE患者中的关系及其对该疾病发生发展的作用。方法 选取30例CE患者和30例健康对照人群,用流式细胞术检测CE患者和健康对照人群外周血中PD-1+CD4+CD25+Treg细胞的比例;实时荧光定量PCR法分别检测CE患者和对照人群外周血单个核细胞中PD-1和Foxp3 mRNA的表达;ELISA法检测CE患者和对照人群血清中TGF-β和IL-10的水平;Pearson相关法分析CE患者外周血中PD-1的表达与Foxp3、TGF-β和IL-10的相关性。结果 与对照人群相比,CE患者外周血中PD-1+CD4+CD25+Treg细胞的比例均显著升高;外周血单个核细胞中PD-1和Foxp3 mRNA水平显著升高;血清中TGF-β和IL-10水平显著升高;CE患者外周血PD-1mRNA与Foxp3 mRNA表达水平正相关;PD-1+CD4+CD25+Treg细胞与TGF-β和 IL-10的水平均正相关。结论 负性共刺激分子PD-1在Treg细胞上高表达与Treg细胞及其因子TGF-β和 IL-10的产生和功能发挥相关,可能促进细粒棘球蚴的生长发展。  相似文献

Despite many years of studies on the mechanisms of immunological defence responses induced in host organisms by Toxoplasma, no satisfactory immunoprophylaxis or chemotherapy have yet been established for humans. Thus, alternative methods to prevent toxoplasmosis and to enhance the efficacy of currently used antitoxoplasmic drugs are under evaluation. In this work, we strove to determine the influence of human prolactin (endogenous present in serum--sPRL and recombinant--rhPRL) on the course of Toxoplasma infection of peripheral blood mononuclear cells (PBMC) originating from female hyperprolactinemia patients. This study revealed that exogenous rhPRL as well as autologous sPRL from inactivated sera significantly restricted intracellular growth of Toxoplasma in PBMC cultures. Moreover, analysis of IL-10 production by PBMC infected with Toxoplasma and cultured in the presence of sPRL showed a positive correlation between sPRL concentration and the level of IL-10. The obtained results could indicate the possible protective action of PRL in a host organism infected with Toxoplasma and suggest that a significant increase in the serum PRL level, during pregnancy for instance, might significantly limit the risk of Toxoplasma spreading and could play an important role in natural protection against toxoplasmosis. The mechanism of inhibitory effect of PRL needs further detailed study.  相似文献

目的 探讨肿瘤坏死因子α诱导的蛋白8-2(tumor necrosis factor-induced protein 8 like 2,TIPE2)在慢性阻塞性肺疾病(COPD)患者外周血单个核细胞(PBMC)中的表达及其在COPD发病中的作用.方法 选择42例符合标准的COPD患者及37名健康对照者,用荧光定量PCR法检测其TIPE2的表达水平,用ELISA法检测血清中肿瘤坏死因子α(TNF-α)、白介素6(IL-6)的含量,由定量数据分析其直线相关关系.结果 COPD组TIPE2表达水平明显低于正常组(11.25±1.37比4.50±1.04,P<0.01),而炎性细胞因子TNF-α、IL-6的含量明显高于正常组;COPD组TIPE2的表达水平与血清中TNF-α、IL-6的水平均呈负相关,而正常组二者之间无明显相关性.结论 COPD患者PBMC中TIPE2表达下降,且其表达水平与炎性细胞因子TNF-α、IL-6呈负相关,TIPE2可能在COPD的发病中发挥重要作用.  相似文献

目的探讨低氧对机体外周血单个核细胞(PBMC)迁移的影响及其临床意义。方法分离外周血单个核细胞,采用Transwell小室实验检测常氧和低氧条件下PBMC迁移能力的变化,流式细胞术(FCM)检测常氧和低氧条件下T、B淋巴细胞,NK细胞及单核细胞迁移的变化。结果低氧环境下PBMC的迁移率降低31%。对其细胞亚群的分析结果显示,NK细胞的迁移率降低31.7%,单核细胞的迁移率降低63.1%,T、B淋巴细胞的迁移率无显著变化。结论低氧能显著影响单核细胞及NK细胞的迁移功能;检测低氧条件下PBMC的迁移能力,可为以免疫细胞迁移功能为靶点的炎症等疾病提供新的治疗对策。  相似文献

目的 了解乙型肝炎患者外周血单个核细胞(PBMC)端粒酶活性的表达情况。方法 通过扩增端粒重复序列(TRAP)及光度酶联免疫法,分别检测健康人及各类乙型肝炎患者PBMC的端粒酶水平。结果 各组患者PBMC在植物血凝素(PHA)刺激前均有端粒酶活性的表达,以急性型肝炎组最高,重型肝炎组最低,二者差别具有显著性(P〈0.001)。PHA刺激后与刺激前比较各组端粒酶活性均有显著性升高(P〈0.001),刺激后的端粒酶水平以重型肝炎组为最低,与其他三组比较差别具有显著性(P〈0.05)。慢性重型乙型肝炎经胸腺五肽(TP5)治疗后端粒酶活性显著增高(P〈0.05)。结论 HBV急性感染期PBMC的端粒酶水平升高;慢性感染期PBMC的端粒酶水平在体内被抑制。TP5具有免疫调节作用,能使过低的端粒酶水平趋向于正常。  相似文献

目的 探讨Toll样受体(TLR)2、TLR4及其信号通路在痛风炎症反应中的作用.方法 应用实时荧光定量聚合酶链反应(RT-qPCR)法检测痛风性关节炎急性组32例、痛风性关节炎非急性组20例及健康对照组(健康体检者)32名外周血单个核细胞(PBMCs)TLR2 mRNA、TLR4 mRNA水平,Western-blot检测上述3组各8例PBMCs核蛋白核因子-κB p65,酶联免疫吸附试验(ELISA)检测3组血浆白细胞介素(IL)-1β含量;并将上述各指标水平与痛风患者及健康体检者血尿酸水平进行相关性分析.多组间比较采用单因素方差分析,两两比较采用q检验.结果 TLR4 mRNA、核因子-κB D65、血浆IL-1β及血尿酸水平在痛风性关节炎急性组[(5.0+1.2)、(7.11+0.18)、(283_+83)pg/ml、(585_+123)μmol/L]和非急性组[(2.3±O.4)、(0.63±0.06)、(134±29)pg/ml,(493±107)μmol/L]均显著高于健康对照组[(1.1± 0.6)、(0.52±0.12)、(97±17)pg/ml,(326±65)μmol/L](P均<0.01),痛风性关节炎急性组高于非急性组(P均<0.01);TLR2 mRNA在3组的表达差异无统计学意义(P>0.05).痛风患者TLR4 mRNA和IL-1B水平与血浆尿酸水平呈正相关(rs=0.876,0.779;P均<0.05),而TLR2 mRNA和IL-1β水平与血浆尿酸水平无相关性(P均>0.05).健康体检者TLR4、TLR2 mRNA与血尿酸、IL-1B水平均无相关性(P均>0.05).结论 原发性痛风性关节炎可通过胞膜型模式识别受体激活固有性免疫应答,TLR4-核因子-κB-IL-1β信号通路参与了痛风免疫及炎症反应调节,痛风患者体内尿酸盐晶体与TLR4及其信号通路激活有关.
Abstract:
objective The roles of TLRs and their signal pathway in gouty arthritis(GA)were explored.Methods TLR2 and TLR4 mRNA was measured using real-time quantitative polymerase chain reaction(RT-PCR)in PBMCs,IL-1β level was detected using ELISA in plasma,and NF-κB p65 protein level in PBMCs was measured using Western blot.Level of TLR2 mRNA,ILR4 mRNA,IL-1β,NF-κB p65protein was compared among acute GA,non-acute GA and healthy controls.Correlation between TLR2mRNA,TLR4 mRNA and serum uric acid,IL-1β level in GA patients was analyzed.One-way ANOVA was used to analyze data between multiple groups and q-test was used for two-two comparison.Spearman's analysis was applied for correlation analysis.Resuits The expression of TLR4 mRNA,NF-KB p65 protein,IL-1β arid serum uric acid level in patients with acute GA [(5.0±1.2), (7.11±0.18), (283±83)pg/ml,[585±123)μmol/L] was significantly increased compared to non-acute GA[(2.3±0.4),(0.63±0.06),(134±29)pg/ml,(493±107)μmol/Lj and healthy controls(1.1±0.6),(0.52±0.12),(97±17)pg/ml,(326±65)μmol/L](P<0.01,respectively).Significant diffefence was also observed between non-acute GA patients and healthy controls(P<0.05,respectively).The level of TLRR4 mRNA was positively correlated with uric acid and IL-1β level in GA patients(rs=0.876,0.779;P<0.05,respectively).Conclusion Innate immunity are activated by membrane-type pattern recognition receptors in primary GA.TLR4-NFκB p65-IL-1β signat transduction may participate in the inflammatory mechanisms of gout.Urate crystals in patients with gout may:be involved in the activation of TLR4 and its signal pathway.  相似文献

Lung disease in patients with cystic fibrosis (CF) is characterized by recurrent bacterial respiratory infections and intense airway inflammation. Pattern recognition receptors such as Toll‐like receptor 2 (TLR2) and TLR4 identify bacterial pathogens and activate the innate immune response. We therefore hypothesized that increased expression of these receptors would be found on circulating immune cells from children with CF. A cohort of 66 young children (median age 3 years) with CF was studied and compared to both healthy controls (n = 14) and children without CF who were being investigated for recurrent respiratory infections (non‐CF disease controls; n = 17) of a similar age. Surface expression of TLR2 and TLR4 on peripheral blood monocytes was analyzed using flow cytometry. TLR4 expression was significantly higher in patients with CF compared to healthy controls (P = 0.017) and non‐CF disease controls (P = 0.025) but did not vary according to the presence or absence of pulmonary infection with Gram‐negative or Gram‐positive bacteria (P = 0.387) in the CF group. In contrast, TLR2 expression was similar across all three study groups (P = 0.930). The increased surface expression of TLR4 seen in young children with CF appears to be related to having CF per se and not related to current pulmonary infection. Pediatr. 目的探讨慢性乙型肝炎(CHB)病人血清及外周血单个核细胞(PBMC)培养上清液白细胞介素一10(IL-10)检测的临床意义。方法分离15例CHB病人及6名正常人血清及PBMC。PBMC体外单独或与金黄色葡萄球菌场毒素B(SE)或重组HBcAg(rHBcA)共培养48小时后,用双抗体夹心ELISA方法检测血清及PBMC培养上清液IL-10水平。结果CHB病人血清IL-10水平为(123.11±13.89)ng/L,正常对照组平均为(95.97±11.68)ng/L,两者之间差异有非常显著性,P<0.01。PBMC体外培养48小时,培养上清液中IL-10水平均明显高于正常对照组,P<0.01。其中rHBCAde导病人PBMC产生的IL-10水平最高,达(369.5±30.52)ng/L。HBVDNA阳性病人血清及PBMC培养上清液IL-10水平均明显高于HBVDNA阴性组病人。慢性轻度和中度肝炎病人血清及PBMC培养上清液IL-10水平又高于慢性重度或慢性重型肝炎病人。结论CHB病人血清及PBMC培养上清液IL-10水平升高可能与HBV持续感染有关。  相似文献

Abstract The production rate of leukotriene B4 (LTB4) was measured using peripheral blood mononuclear cells (PBMC) in patients with fulminant hepatitis (FH) or other liver diseases. LTB4 in the culture media of PBMC under stimulation with Ca-ionophore was fractionated by HPLC and measured by radioimmunoassay. The production rate of LTB4 was elevated in 16 of 17 FH patients (3.3 ± 0.2 ng/106 cells for 5 min), while the production was below detectable level in patients with acute or chronic hepatitis and in healthy controls. In FH patients, the highest production rate of LTB4 was observed in the initial period of the disease. Enhanced LTB4 production may indicate the primed state of PBMC — the primed mononuclear cells are regarded as participating in the development of massive liver necrosis and of other organ failures in FH.  相似文献
