从景天科植物流苏瓦松的花叶的提得的瓦松甙使离体兔心房收缩振幅增高;使在体蟾蜍心输出量和作功显著增加;能增体兔心多种心肌收缩性能和泵血功能和泵血功能指标:增大左室压力变化速率峰值(LVdp/dt max)和在共同最高等容量收缩压时的心肌收缩分的缩短速度(Vce-CPIP),缩短左室从开始收缩到开始射血的时间(t~dp/dtmax),心率减慢,左室作功增加。 相似文献

铁筷子,别名黑毛七,小山桃七等,属毛茛科铁筷子属植物。产于我国陕西、甘肃、四川等省。民间主要用于治疗膀胱炎、尿道炎、疮痈肿毒、跌打损伤。其药理作用未见有报道,本实验证明其有洋地黄样强心作用。 材料 实验所用材料为干燥铁筷子根,在50℃下进一步烘干(约1小时),研细后按中国药典制成10%铁筷子酊,蒸去乙醇,用生理盐水稀释至10~20%,过滤后贮于冰箱备用。应用时再用生理盐水配成不同浓度。实 相似文献

用荧光法研究了黄芩甙、芸香甙、苦杏仁甙、丹皮酚和百里香酚对Fenton反应生成的、OH的清除作用。结果表明,以上化合物的清除作用的均强于.OH特异性清除剂甘露醇,以黄芩甙、芸香甙的清除作用特别显著,其EC50分别为0.039mmol/L和0.069mmol/L,根据清除地荧光的抑制,推导了清除剂与OH反应的速度常数。 相似文献

本文用临床通用剂量(20ml/kg,相当于成人一次使用1200ml)的氟碳代血液作慢性毒性病理学研究。狗在一次静脉注入本品后主要沉积在单核巨噬系统,并被固定和游离的组织细胞所吞噬,吞噬后的组织细胞转变为泡沫细胞。 注入后1月仍属潴留高峰,6月后除脾脏外各脏器均已基本排空并不留任何病理痕迹,12月后所有被检脏器在光镜下均已不能查见泡沫细胞,但在电镜下肝脾仍可偶见个别部位留有少量小泡状物质,作者认为此种极微量的潴留缺乏临床意义。 作者也认为氟碳代血液对靶细胞只是被吞噬和潴留,未出现任何明显病理学改变。 根据研究结果,作者认为本品属非原浆毒并为生物惰性物质,亦不存在由于本品在单核巨噬系统潴留而引起的继发性病理损害问题,并认为本剂量的临床应用是安全的。 相似文献

Background Ouabain and digoxin are important cardiac glycoside and related to many cardiovascular diseases.The purpose of this study was to investigate the changes of sodium pump α-subunit expression in rats and compare the effects of ouabain (OUA) and digoxin (DIG) on the development of hypertension.Methods In situ hybridization was performed.Specific sequence oligonucleotide probe tailing with a Dig-dUTP hybrid to target nucleic acids of the sodium pump α-subunit.According to counting positive particles sodium pump subunit expression was analyzed with statistical methods.Results On day 16 of drug administration,the blood pressure of rats increased significantly in the OUA group.In the DIG group,the blood pressure revealed no significant difference when compared to the control group.In addition,the effects of OUA and DIG on sodium pump α-subunit RNA expression in tissues differed.Conclusions OUA and DIG can not only change the configuration of the sodium pump to depress their activity,but also influence their gene expression which is important in the mechanism of hypertension.This may be a key point in the pathogenesis of hypertension in the manner in which OUA differs from DIG and changes the sodium pump gene expression in the arteries and kidneys of rats. 相似文献

目的 探索哇巴因对大鼠心脏超微结构及心功能的影响。方法 雄性SD大鼠随机分为哇巴因组和生理盐水对照组,分别给予哇巴因或生理盐水腹腔注射,每周测量收缩压。4周后,对两组动物进行心脏超声检查,并予有创血流动力学检查评估心功能,透射电镜观察心脏超微结构的变化。结果 SD大鼠腹腔注射哇巴因4周后,两组间血压尚无显著差异。心脏超声检查提示,哇巴因组大鼠左心室收缩及舒张末期内径、舒张末期左心室后壁厚度、室间隔厚度以及左心室质量增加,等容舒张时间延长,E/A值、射血分数及左心室短轴缩短率降低,与对照组相比有显著性差异(P〈0.05)。有创血流动力学检查显示,哇巴因组大鼠左心室收缩压、左心室内压最大上升/下降速率降低,左心室舒张压升高。透射电镜观察显示,哇巴因组大鼠心肌细胞肿胀,肌纤维断裂,Z线消失,线粒体增生肿胀,部分空泡化,细胞间胶原纤维增生。结论 哇巴因可引起大鼠左心室增大、室壁增厚、心脏收缩及舒张功能受损,心肌超微结构损伤,此效应发生在血压升高前,提示哇巴因可能有直接的心肌损害作用。 相似文献

Objective:To investigate the ouabain's effects on the ultrastructure and function of the rat heart. Methods: Male Sprague-Dawley (SD) rats were treated with ouabain and systolic blood pressure (SBP) were recorded weekly. After 4 weeks, echocardiography was performed, hemodynamic parameters were measured by invasive cardiac catheterization and changes in heart ultrastructure were analyzed using transmission electron microscopy. Results:After treated by ouabain for 4 weeks, there were no significant differences in the mean SBP of the two groups. However, cardiac systolic and diastolic performances were both worsened with ouabain treatment by echocardiography, left ventricular chamber diameters and wall thickness were significantly increased in the rats of ouabain group. Invasive monitoring indicated that left ventricular systolic pressures (LVSP), rate of pressure development (+dp/dt) and rate of pressure decay (-dp/dt) were significantly attenuated and left ventricular end-diastolic pressures (LVEDP) were increased in ouabain group (P〈0. 05). Disorganization of myofilaments, mitochondrial swelling, disruption and vacuolation, hyperplastic collagen fibers were found in ouabain group by transmission electron microscopy. Conclusion:It is suggested that ouabain induces alterations in cardiac ultrastructure and function, and the effects happened before the increase of blood pressure, which indicates that ouabain might damage rat heart independent of blood pressure. 相似文献

目的 观察哇巴因与地高辛对大鼠肝脏钠泵基因表达的影响 .方法 分别给大鼠注射哇巴因 (2 0 μg· kg- 1·d- 1 )、地高辛 (32 μg·kg- 1·d- 1 )和生理盐水 (1m L· kg- 1·d- 1 ) ,观察给药后 6wk大鼠血压的动态变化 ;并分别应用分子生物学 RT- PCR及免疫组织化学技术 ,探讨各组大鼠肝脏钠泵 α1 ,α2 及 α3 亚单位 m RNA及蛋白水平基因表达的改变 .结果 给予哇巴因 2 wk后大鼠血压开始升高 ,至 6 wk时明显高于生理盐水组 (17.7± 1.2 ) vs(15 .4± 1.1) k Pa,P<0 .0 1) ,而实验过程中地高辛组血压与对照组比较差异不明显 .无论是在 m RNA水平还是在蛋白水平 ,哇巴因对大鼠肝脏钠泵α1 亚单位表达无影响 ,而地高辛使α1 亚单位表达增强 ;两组大鼠 α2 亚单位表达均无改变 ;哇巴因使 α3 亚单位蛋白水平表达减弱 ,而 m RNA水平增强 ,地高辛组大鼠肝脏钠泵 α3 亚单位无论在 m RNA水平及蛋白水平表达均增强 .结论 哇巴因在高血压发病中可能起着重要作用 ;哇巴因与地高辛可导致不同的钠泵基因表达改变 ,为进一步揭示内源性哇巴因生理与病理作用以及洋地黄类药物药理与毒理作用的分子机制提供了理论及实验依据 相似文献
