目的探讨冠心病患者血浆及外周血单个核细胞(PBMC)miR-328、miR-22、miR-147的表达变化及其临床意义。方法筛选100例冠心病患者及相对健康对照组,采用实时荧光定量PCR技术检测各组血浆及PBMC中miR-328、miR-147、miR-22的表达水平。结果冠心病患者血浆中miR-328、miR-22的表达较非冠心病患者明显升高,而miR-147的表达明显降低;在冠心病患者PBMC中,miR-328表达没有明显变化,miR-22表达略微增加,miR-147表达明显下降。在心肌梗死患者中,血浆中miR-328和miR-22表达水平明显增加,而miR-147表达明显下降;心肌梗死患者PBMC中miR-328表达略降低,miR-22表达增加,miR-147表达水平略有下降。结论 miR-328、miR-22及miR-147在冠心病患者中的差异表达,有望作为冠心病疾病中一种新的标记物或基因治疗靶点。  相似文献

[目的]检测miR-21和miR-155在结肠腺癌中的表达特征,分析其临床意义。[方法]选取2014年1月-2014年12月我院收治的95例结肠腺癌患者为观察组,选取距肿物边缘>5 cm的正常结肠黏膜69例作为对照组,应用实时荧光定量PCR法检测2组中miR-21和miR-155的表达。[结果]观察组中miR-21和miR-155的表达明显高于对照组,观察组中miR-21和miR-155的表达在不同分化程度、有无癌栓、浸润深度、淋巴结转移及TNM分期的表达中差异有统计学意义。miR-21和miR-155的表达均与增殖指数呈正相关。生存分析显示二者的表达与生存时间有关。[结论]结肠腺癌中miR-21和miR-155高表达是肿瘤形成的促进因素。二者可以促进肿瘤细胞的增殖,检测miR-21和miR-155的表达对判断预后可能有一定价值。  相似文献

目的探究miR-106a和miR-363-3p在溃疡性结肠炎(UC)患者血浆中的表达及其临床意义。方法选择2015年3月至2018年7月在重庆市开州区人民医院消化内科救治的78例UC患者(UC组)、53例CD患者(CD组)及同期46例健康体检者(NC组)作为研究对象。采用RT-PCR法检测3组受试者血浆miR-106a和miR-363-3p的表达水平。评估miR-106a和miR-363-3p表达水平鉴别CD和UC的效能。分析miR-106a和miR-363-3p表达水平与UC患者病情及预后的关系。结果 UC组患者血浆miR-106a和miR-363-3p相对表达量低于CD组,但高于NC组,差异均有统计学意义(P均0.05)。ROC曲线显示,miR-106a和miR-363-3p鉴别CD和UC的ROC曲线下面积(AUC)分别为0.906和0.846。miR-106a和miR-363-3p联合检测鉴别CD和UC的AUC大于各指标单独检测(P0.05)。活动期UC患者血浆miR-106a和miR-363-3p的相对表达量均高于缓解期UC患者,差异均有统计学意义(P均0.05)。单因素和多因素Logistic回归分析影响活动期UC患者预后的相关因素,结果显示miR-106a和miR-363-3p与活动期UC患者预后密切相关。miR-106a1.14、miR-363-3p1.05的活动期UC患者预后不良。ROC曲线显示,miR-106a和miR-363-3p及两者联合检测诊断活动期UC患者预后的AUC分别为0.836、0.867和0.860,差异无统计学意义(P0.05)。结论检测miR-106a和miR-363-3p表达水平可辅助鉴别CD和UC。miR-106a和miR-363-3p的表达水平可反映UC患者病情,并可评估UC患者预后。  相似文献

目的 观察冠心病患者血小板miR-126表达与冠状动脉粥样硬化指标的关系,探讨其在冠心病诊断、预测及预后分析中的应用价值。方法 随机选取2016年3月-2018年3月在沈阳市第四人民医院循环内科确定诊断为冠心病的患者60例为实验组,包括SAP(稳定性心绞痛)组(n=30)及ACS(急性冠状动脉综合征)组(n=30),健康人30例作为健康对照组(NC组),qRT-PCR检测血小板miR-126表达,评价miR-126在健康人群、急性冠状动脉综合征及稳定型心绞痛患者中的表达水平差异,miR-126与冠脉斑块CT值分型的关系,miR-126表达与冠脉造影病变程度、分型及病变支数相关性。结果 qRT-PCR显示,ACS组及SAP组miR-126表达高于NC组(P<0.01),ACS组高于SAP组(P<0.05);miR-126在冠脉斑块CT值分型中钙化型高于纤维型及软质型,纤维型高于软质型(P<0.05);miR-126表达水平在中度、重度及闭塞组高于轻度组(P<0.05),重度及闭塞组高于中度组(P<0.05);miR-126表达水平在2支及≥3支病变组高于1支病变组(P<0.05),≥3支病变组高于2支病变组(P<0.05)。结论 血小板miR-126表达于冠状动脉粥样硬化严重程度具有相关性,miR-126表达高者冠状动脉病变程度及支数均较高。  相似文献

目的探讨miR-126表达水平对人晶状体上皮细胞系HLEB3凋亡的影响及作用机制。方法使用H2O2构建HLEB3体外凋亡模型,qRT-PCR检测miR-126水平变化;使用脂质体2000分别将miR-126模拟物(mimic)和抑制物(inhibitor)以及二者相应的阴性对照转染到HLEB3中,分别上调、下调细胞中miR-126表达,转染48 h后qRT-PCR验证转染效率,流式细胞仪检测各组HLEB3凋亡水平;通过靶基因预测软件预测miR-126可能靶向调控KLF6基因的表达,检测各组HLEB3中KLF6 mRNA和蛋白表达水平。结果 HLEB3凋亡模型中miR-126相对表达量明显高于正常HLEB3,差异有统计学意义(P0.01)。模拟物组HLEB3凋亡率明显低于模拟物阴性对照组;抑制物组HLEB3凋亡率明显高于抑制物阴性对照组;差异均有统计学意义(均P0.05)。模拟物组HLEB3中KLF6 mRNA相对表达量明显低于模拟物阴性对照组,抑制物组KLF6 mRNA相对表达量明显高于抑制物阴性对照组,差异均有统计学意义(均P0.05);模拟物组HLEB3中KLF6蛋白相对表达量明显低于模拟物阴性对照组,抑制物组KLF6蛋白相对表达量明显高于抑制物阴性对照组,差异均有统计学意义(均P0.05)。结论 miR-126在HLEB3凋亡模型中表达上调,miR-126可通过调控KLF6基因表达来影响HLEB3凋亡,在白内障发病机制中发挥作用。  相似文献

目的探讨肝细胞肝癌患者组织中miR-21的表达水平及其预后意义。方法共收集50例肝细胞肝癌的患者,采用免疫组化法检测肝细胞肝癌及配对癌旁组织中miR-21的表达水平,分析miR-21蛋白的表达与患者临床病理特征及预后生存的相关性。结果肝细胞肝癌组织中miR-21表达水平显著高于癌旁组织(58.0%vs 6.0%,χ~2=31.07,P0.000 1)。miR-21高表达与临床T分期相关(χ~2=8.043,P=0.018);但miR-21蛋白表达水平与患者年龄、性别、肿瘤大小、临床分期、N分期及分化程度均无显著关联(P0.05)。高表达miR-21的肝细胞肝癌患者总生存时间明显降低(P=0.032),但患者无疾病进展生存期差异无统计学意义(P=0.116)。结论肝细胞肝癌患者miR-21高表达可能作为肿瘤患者预后不良的重要标志物之一,而与肿瘤的发生及发展的机制性研究尚需进一步验证。  相似文献

目的探讨2型糖尿病患者外周血血清中miR-126的表达水平及其临床意义。方法采用荧光定量PCR方法检测77例糖代谢异常患者血清miR-126的表达(其中糖调节受损组26例,初发2型糖尿病组23例,2型糖尿病伴有血管病变患者28例,和30例健康对照组)。结果 miR-126在健康对照组与糖调节受损组血清中的表达无统计学意义(P>0.05);初发糖尿病组与健康对照组及糖调节受损组之间均有统计学意义(P<0.01);糖尿病伴有血管病变组与健康对照组及糖调节受损组之间均有统计学意义(P<0.01),但与初发糖尿病组无统计学意义(P>0.05)。结论 2型糖尿病及合并有血管病变患者外周血血清中miR-126水平呈低表达。  相似文献

目的研究原发性肝癌(HCC)患者血清中微小RNA(miR)-21、miR-148b、miR-200a、miR-200b的表达及临床诊断意义。方法应用实时荧光定量逆转录-聚合酶链反应(qRT-PCR)检测西南医科大学附属医院收治的93例HCC患者(病例组)、48例肝脏良性病变(良性组)、48例慢性肝病患者(慢性肝病组)和50例健康体检者(对照组)血清中miR-21、miR-148b、miR-200a、miR-200b的表达。统计学分析各指标与临床病理特征之间的关系,受试者工作特征(ROC)曲线分析各指标的诊断价值。结果病例组血清miR-21与miR-200b表达显著高于良性组、慢性肝病组及对照组(均P<0.05),miR-200a与miR-148b表达显著低于良性组、慢性肝病组及对照组(均P<0.05)。HCC患者血清miR-21与miR-200b表达与病理分期、病理分级及肿瘤直径有关(均P<0.05),与性别、年龄、有无肝硬化及乙型肝炎病毒(HBV)抗原是否阳性无关(均P>0.05)。血清miR-200a与miR-148b表达与病理分期、病理分级有关(均P<0.05),与性别、年龄、肿瘤直径、有无肝硬化及HBV抗原是否阳性无关(均P>0.05)。miR-21、miR-148b、miR-200a、miR-200b及四项指标联合的ROC曲线下面积(AUC)分别为0.821、0.668、0.772、0.734、0.922。联合诊断的敏感性和特异性均高于任一单一指标。结论HCC患者血清miR-21与miR-200b表达上调,而miR-200a与miR-148b表达下调。并且其表达与病理分期、病理分级有关,联合检测血清miR-21、miR-148b、miR-200a、miR-200b有希望成为新的诊断HCC的肿瘤标志物。  相似文献

目的探讨冠心病(CHD)患者外周血miR-21的表达及与冠状动脉病变严重程度的相关性。方法行冠脉造影的患者145例中CHD患者77例,其中心绞痛(AP)45例(AP组)及急性心肌梗死(AMI)32例(AMI组),另外无冠脉病变68例为对照组。采用Gensni评分评价冠脉狭窄程度,应用荧光实时定量(qRT-PCR)方法检测外周血miR-21表达。结果三组血浆中miR-21表达水平、Gensini评分及冠脉狭窄程度差异显著(P<0.05),其中AMI组、AP组血浆中miR-21表达水平、Gensini评分及冠脉狭窄程度显著高于对照组,AMI组亦显著高于AP组(P<0.05);Spearman线性相关分析显示,CHD患者miR-21表达水平与Gensini评分、冠脉狭窄程度呈正相关性(r=0.895、0.888,均P<0.05)。结论 CHD患者外周血miR-21高表达,且与冠脉病变的严重程度具有正相关性,为CHD临床诊治及预后提供了理论依据。  相似文献

目的观察microRNA-4463(miR-4463)在下肢动脉硬化闭塞症(ASO)患者血浆和组织中的表达变化,推测其可能意义。方法采用miRNA芯片方法筛选ASO患者和对照人群血浆中差异表达miRNA,Real time PCR技术验证ASO患者和对照人群中miR-4463的表达水平,并进行临床分期比较。靶基因预测软件分析miR-4463靶基因,Gene Ontology和KEGG数据库分析靶基因的功能和信号通路。结果与对照组相比,ASO患者血浆中有51个miRNA变化超过1.5倍(P0.05)。miR-4463在ASO患者血浆和病变血管内膜组织中表达均显著降低,并随Fontaine分期呈逐渐下降趋势。生物信息学分析发现miR-4463靶基因与细胞迁移、脂质代谢、内吞作用等相关。结论 miR-4463参与了ASO的病程,其表达下降可能提示ASO的发生。  相似文献

BACKGROUND Esophageal squamous cell carcinoma(ESCC) is one of the main causes of human death. It is usually already in middle or advanced stage when diagnosed due to its hidden symptoms in early stage. Therefore, patients have already lost the best surgical timing when diagnosed. Radiotherapy and chemotherapy are standard treatment methods for ESCC clinically, but the efficacy and prognosis of patients from them are still unsatisfactory. Therefore, it is of great clinical significance to seek for biomarkers that can predict the radiotherapy and chemotherapy response and prognosis of ESCC patients.AIM To explore the clinical value of plasma miR-21 and miR-93 in ESCC.METHODS A total of 128 ESCC patients admitted to the First Affiliated Hospital of Zhenzhou University were enrolled as a study group and treated with concurrent radiotherapy and chemotherapy, and other 45 healthy people during the same period were enrolled as a control group. The expression of plasma miR-21 and miR-93 was determined using quantitative real-time polymerase chain reaction,and the correlation of expression of plasma miR-21 and miR-93 with clinical pathological parameters about the patients was analyzed. The receiver operating characteristic(ROC) curve was adopted to assess the diagnostic value of plasma miR-21 and miR-93 for clinical pathological features of ESCC patients, the Logistic regression analysis adopted to analyze the risk factors for radiotherapy and chemotherapy efficacy in ESCC patients, and the Cox regression analysis toidentify the prognostic factors for ESCC patients.RESULTS The study group showed significantly higher relative expression of plasma miR-21 and miR-93 than the control group(P 0.01). The area under the ROC curve(AUC) of plasma miR-21 for diagnosing T stage, N stage, M stage, and pathological differentiation of ESCC was 0.819, 0.758, 0.824, and 0.725,respectively, and that of plasma miR-93 for diagnosing T stage, N stage, and M stage of ESCC was 0.827, 0.815, and 0.814, respectively. The AUC of combined plasma miR-21 and miR-93 for predicting radiotherapy and chemotherapy efficacy before radiotherapy and chemotherapy was 0.894, and the AUCs of them for predicting the 3-year overall survival(OS) were 0.861 and 0.807, respectively.T stage(P 0.05), M stage(P 0.05), miR-21(P 0.01), and miR-93(P 0.05) were independent risk factors for radiotherapy and chemotherapy efficacy, and T stage(P 0.01), N stage(P 0.05), M stage(P 0.01), miR-21(P 0.01), and miR-93(P 0.01) were independent prognostic factors for ESCC patients.CONCLUSION MiR-21 and miR-93 can be adopted as effective biomarkers for predicting radiotherapy and chemotherapy efficacy in ESCC and the 3-year OS of ESCC patients.  相似文献

Objective to evaluate the effects of environmental factors and microRNAs (miRNAs) (miR-126, miR-143, and miR-145) on the risk of coronary heart disease (CHD). Methods A frequency-matched case-control study (450 patients, 450 controls) was conducted from April 2014 to December 2016 in Fuzhou City, China. Environmental factors were investigated using a self-administered questionnaire, and the expression levels of miR-126, miR-143, and miR-145 were determined by quantitative real-time Polymerase Chain Reaction (PCR) in peripheral blood mononuclear cells. Unconditional logistic regression models were used for statistical evaluation. Results Alcohol consumption, high-salt diets, high-intensity work, and lack of physical activity were significantly associated with increased CHD risk, whereas light diet was significantly associated with decreased risk. MiR-126, miR-143, and miR-145 were highly expressed in the CHD group compared with the control group. After adjustment for other environmental factors, unconditional logistic regression results revealed that miR-126, miR-143, and depression were the independent risk factors of CHD, and light diet was the independent protective factor of CHD. Conclusions Our data suggest that a family history of CHD, anxiety, and alcohol consumption was significantly associated with increased CHD risk, whereas light diet was significantly associated with decreased risk. Furthermore, miR-126 and miR-143 in combination with several risk factors, could play a joint role in the development of CHD. Therefore, it is necessary to manage patients with CHD in all directions and multiple level.  相似文献

目的 探讨胰腺癌组织织间质表皮转化因子(c-MET)的表达与循环miR-34a、miR-449水平的相关性及其临床意义。方法 收集2015年3月至2017年3月间嘉兴医学院附属第二医院行手术治疗并经病理证实的41例胰腺癌患者的临床资料。通过免疫组织化学染色检测胰腺癌组织及其匹配的癌旁正常胰腺组c-MET表达,根据测定结果将患者分为c-MET阳性组与c-MET阴性组。采集患者手术前和手术后3个月外周血,应用荧光定量PCR法检测循环miR-34a和miR-449水平。分析癌组织c-MET表达与患者临床病理参数、预后及循环miR-34a、miR-449水平的关系。分析循环miR-34a和miR-449水平对胰腺癌TNM分期、淋巴结转移和预后的评估价值。结果 胰腺癌组织c-MET阳性率明显高于癌旁正常组织(63.4%比24.4%),差异有统计学意义(P<0.05)。与c-MET阴性患者比较,c-MET阳性患者TNMⅢ/Ⅳ期者居多(73.1%比33.4%),淋巴结转移率高(76.9%比46.7%),生存时间短(29.5个月比35.0个月),生存率低(38.5%比53.3%),差异均有统计学意义(P值均<0.05)。术前c-MET阳性患者循环miR-34a、miR-449水平明显低于c-MET阴性患者(0.228±0.068比0.524±0.106、0.252±0.063比0.432±0.094,P<0.05),术后c-MET阳性患者循环miR-449水平仍明显低于c-MET阴性患者(0.414±0.088比0.512±0.114,P<0.05),但两组间miR-34a水平的差异无统计学意义。术前循环miR-34a、miR-449水平对胰腺癌TNM分期、淋巴结转移及预后有预测价值(P<0.05)。结论 miR-34a、miR-449靶向结合胰腺癌组织c-MET,有可能成为潜在的胰腺癌标志物。  相似文献

目的:探讨血清微小核糖核酸-208a(miR-208a)表达水平对急性ST段抬高型心肌梗死(STEMI)患者并发急性心力衰竭的预测关系。方法:连续性收集2018年1月至2019年6月,就诊于我科的152例急性STEMI患者纳入病例组,并随机选择同期在我院行健康体检的志愿者60例为对照组。根据发病48 h内是否发生急性心力衰竭(AHF)将病例组分为AHF亚组和非AHF亚组。采用实时荧光定量PCR(RT-qPCR)法检测血浆miR-208a的相对表达水平。结果:病例组血浆miR-208a相对表达水平显著高于对照组(Z=10.919,P=0.000)。AHF亚组血浆miR-208a相对表达水平显著高于非AHF亚组(Z=9.573,P=0.000)。Pearson相关性分析结果显示,病例组患者血浆miR-208a相对表达水平与BNP、hsCRP和cTnI均呈正相关关系(r=0.612,P=0.000;r=0.447,P=0.000;r=0.378,P=0.000)。二分类Logistic回归分析结果显示血浆miR-208a相对表达水平是急性STEMI患者并发AHF的危险因素(OR=2.118,95%CI 1.127~3.982,P=0.007)。血浆miR-208a预测AHF的AUC为0.896(0.844,0.948),cut-off值为32.00,对应的敏感性和特异性分别为88.4%和83.3%。结论:急性STEMI患者血浆miR-208a表达水平显著升高,且可能是并发AHF的危险因素。  相似文献

目的 比较血循环中心肌特异性miR-133a与肌钙蛋白T(cTnT)在急性心肌梗死(AMI)早期诊断中的价值.方法 收集67例AMI患者和32名非AMI对照者的血浆标本,通过定量逆转录聚合酶链反应(qRT-PCR)测定血浆中miR-133a水平,同时通过电化学发光法检测血浆cTnT的变化.结果 AMI患者血浆中miR-133a水平较非AMI对照者明显升高(P〈0.01),而当这些AMI患者康复出院时miR-133a水平已回落到基线.MiR-133a水平的波动与研究对象的自身特征无相关性(P&gt;0.05).受试者工作特征曲线(ROC)分析表明,miR-133a在早期诊断AMI方面具有显著的心肌特异性和敏感性,但并不优于cTnT.结论 血浆miR-133a可作为AMI早期诊断的标志物,但在诊断价值上并不优于cTnT.  相似文献

Objective

This work was implemented to elucidate the miR-126-5p expression in the plasma of patients with sepsis-induced acute lung injury (ALI) and its correlation with inflammation and immune function.

Methods

The peripheral blood of patients with sepsis-induced ALI was obtained, and the levels of inflammatory factors (interleukin-6 [IL-6], C-reactive protein [CRP], and procalcitonin [PCT]) were determined. Meanwhile, T lymphocyte subsets (CD3+, CD4+, and CD8+), and immunoglobulins (IgA, IgM, and IgG) were tested. miR-126-5p and TRAF6 mRNA expression in plasma was assessed. Receiver operating characteristic (ROC) curve was performed to assess the diagnostic accuracy of miR-126-5p in sepsis without ALI and sepsis with ALI. Correlation between miR-126-5p expression and clinical indicators was analyzed. The targets of miR-126-5p were predicted using the bioinformatics method, and the direct targets were verified through investigations.

Results

miR-126-5p expression in plasma of patients with sepsis-induced ALI was reduced than that of patients with sepsis without ALI. miR-126-5p expression was negatively correlated with IL-6, CRP, and PCT but positively correlated with IgA, IgM, and IgG as well as CD3+, CD4+, and CD8+ in patients with sepsis-induced ALI. ROC curve suggested that miR-126-5p (AUC: 0.777; 95%CI: 0.689–0.866) could distinguish patients with sepsis with ALI from patients with sepsis without ALI. TRAF6 expression in patients with sepsis-induced ALI was higher than that in patients with sepsis without ALI. TRAF6 was a target gene of miR-126-5p,

Conclusion

This research highlights that miR-126-5p is reduced in the plasma of patients with sepsis-induced ALI, and miR-126-5p relates to systemic inflammation and immune function indicators.  相似文献

目的探讨miR-221-3p在下肢缺血性疾病中的表达变化。方法临床试验:收集西南医科大学附属医院血管甲状腺外科2014年至2015年间55例动脉硬化闭塞症(ASO)患者和54例健康对照人群血浆,24例ASO截肢患者的正常血管及病变血管。选取3对年龄、性别相近的ASO患者和正常健康人血浆用Microarray筛选出ASO患者血浆中差异性表达在2倍以上的24种microRNA,其中miR-221-3p的表达量明显降低。用实时荧光定量PCR检测临床55例ASO患者和54例健康对照人群血浆及24例ASO截肢患者正常血管和病变血管中miR-221-3p的表达量。动物实验:选取SD大鼠,结扎组分离结扎大鼠股动脉制备下肢缺血模型,假手术组仅暴露分离股动脉不做结扎。分别于术前、术后即刻、术后7天、49天行激光多普勒扫描监测下肢皮肤血流变化。于术后7天、49天行激光多普勒扫描后处死各组大鼠,收集大鼠术侧腓肠肌及下腔静脉血,采用实时荧光定量PCR检测各组血浆和腓肠肌内miR-221-3p的表达量。结果临床试验显示:与健康对照组相比,ASO患者血浆中miR-22-3p的含量明显降低(1.078±0.119比0.617±0.121);与正常血管相比,病变血管中miR-221-3p的表达量也明显降低(1.017±0.113比0.625±0.136)。动物实验显示:与假手术组相比,结扎组7天时血浆和腓肠肌内miR-221-3p的含量明显降低(血浆1.04±0.07比0.31±0.07、腓肠肌1.01±0.07比0.64±0.09),49天时miR-221-3p的含量有所上升但仍低于对照组。结论 miR-221-3p在下肢缺血性疾病中明显降低,有望成为下肢缺血性疾病一个新的诊断方式及治疗靶点。  相似文献
