目的研究SEA-Fab'对肿瘤细胞的凋亡作用及单克隆抗体于肿瘤定向治疗作用.方法实验分为三组SEA-Fab'、SEA及对照组,三组肿瘤细胞分别用SEA-Fab'、SEA及PBMC+Walker-256细胞处理,在24~72 h内观察肿瘤细胞的凋亡情况.结果SEA-Fab'组、SEA组的肿瘤细胞指数分别为(34.6%~68.9%)和(15.5%~31.9%)高于对照组(5.5%~12.5%),差异存在显著性,SEA-Fab'组高于SEA组差异显著.结论SEA-Fab'在激活T细胞和杀死肿瘤细胞效果好于SEA,将来有可能利用单克隆抗体来作定向靶位治疗.  相似文献

目的 探讨了CKD 3~5期患者外周血T淋巴细胞CD154表达能力的变化。 方法 以健康人外周血为对照组,以未接受血透析治疗的慢性肾脏病(CKD)3~5期(未透析组)及CKD 5期维持性血液净化治疗的患者(透析组)外周血为实验组,检测各组受试者外周血T淋巴细胞表达频率、CD3+CD8-T细胞CD154的表达比例。 结果 与对照组相比,CKD 3~5期未透析患者外周血淋巴细胞中T细胞所占频率下降,差异有统计学意义(P<0.05);CKD5期透析组与对照组比较差异无统计学意义(对照组为72.05%±2.65%;未透析组为63.06%±3.37%;透析组为68.92%±3.22%)。离子霉素联合佛波酯刺激全血后,各组表达CD154的CD3+CD8- T细胞占CD3+CD8- T细胞比例:未血液净化组(73.61%±1.91%)较对照组(79.78%±2.00%)及血液净化组(78.94%±2.47%)明显下降,差异有统计学意义(P<0.05);提取PBMC后用离子霉素联合佛波酯刺激,各组间CD154表达差异无统计学意义(P>0.05)。 结论 CKD 3~5患者存在CD3+CD8- T细胞CD154表达能力下降,可能影响患者的免疫功能。  相似文献

目的 本研究旨在探讨18F-FDG PET-CT与乳腺癌新辅助化疗疗效的关联性。方法 2014年1月~2015年12月,收集笔者医院收治的行新辅助化疗的乳腺癌患者98例。前瞻性分析18F-FDG PET-CT与乳腺癌新辅助化疗疗效的关联性。结果 与病理无效组相比,病理有效组患者SUVmax0显著增加(9.54±3.11 vs 6.12±3.21,P=0.015);SUVmax1变化率显著增加(53%±17% vs 29%±11%,P=0.000);SUVmax2变化率显著增加(61%±21% vs 34%±12%,P=0.000)。与未pCR组相比,pCR组患者SUVmax0显著增加(10.54±4.11 vs 5.87±2.21,P=0.000);SUVmax1变化率显著增加(57%±17% vs 28%±10%,P=0.000);SUVmax2变化率显著增加(64%±23% vs 33%±11%,P=0.000)。结论 18F-FDG PET-CT可作为评价乳腺癌新辅助化疗效果的一种影像学方式,值得临床进一步推广。  相似文献

Summary The curcumin prodrugs, which could be selectively activated in tumor cells, were prepared to establish a basis for the targeted chemotherapy for cancer. On the basis of the molecular structure of curcumin, the N-maleoyl-L-valine-curcumin (NVC), N-maleoyl- glycine-curcumin (NGC) were chemically synthesized and identified by IR and NMR spectroscopy. After treatment with these two prodrugs for 6–24 h, the rates of growth inhibition on human bladder cancer EJ cells and renal tubular epithelial (HKC) cells were detected by MTT colorimetry. Our results showed that after the treatment with 20 μmol/L–40 μmol/L NVC and NGC for 6–24 h, the growth inhibitory effects on EJ cells were 6.71%–65.13% (P<0.05), 10.96%–73.01% (P<0.05), respectively, in both dose- and time-dependent manners. When compared with the curcumin of same concentrations, the growth inhibitory effects of these two prodrugs on HKC cells were significantly decreased (P<0.01). It is concluded that activation of curcumin prodrugs via hydrolysis functions of cellular esterase could inhibit the growth activities of tumor cells, and reduce the side effects on normal diploid cells. 目的 通过建立大鼠门脉高压背景下肝肿瘤模型,观察病理性脾脏的切除对肝肿瘤发生发展的影响.方法 按照四氯化碳复合、梯段用药法制备大鼠肝硬化门脉高压模型,后将Walker-256肿瘤块种植在大鼠肝脏左外叶上,随机分组的A组大鼠在种植瘤块同时切除脾脏,B组大鼠保留脾脏.10 d后分别测定两组大鼠的T细胞亚群CD4、CD8及CD4/CD8;切除大鼠肝脏左外叶,测量种植肿瘤块最大直径,并用免疫组织化学染色方法检测肿瘤组织Ki-67的表达.结果 门脉高压模型建成后,大鼠的血常规以及肝功能指标出现明显异常.A组大鼠的CD4值和CD4/CD8比值均高于B组大鼠(0.36±0.01 vs0.35±0.02,1.33±0.08 vs 1.24±0.05;P<0.05或P<0.01),而CD8值则低于B组大鼠(0.27±0.01 vs0.29±0.02,P<0.01).A组大鼠的肝脏肿瘤直径与B组大鼠比较差异无统计学意义.A组大鼠肿瘤组织的Ki-67阳性率较B组降低,差异具有统计学意义(P<0.05).结论 门脉高压状态下病理性脾脏的切除既有利于提高机体抗肿瘤的免疫力,又可以在一定程度上减弱肝肿瘤的侵袭能力.  相似文献

目的 通过观察TLR4在乙型肝炎相关性肝癌患者外周血CD14+单核细胞表面的表达,探讨其在疾病发生、发展中的作用。方法 实验共选取慢性乙型肝炎患者25例(慢性乙型肝炎组)、乙型肝炎并肝癌23例(乙型肝炎相关性肝癌组)、健康对照16例(对照组),利用流式细胞术检测研究对象外周血CD14+单核细胞表面TLR4阳性细胞表达百分率和平均荧光强度的几何均数(GMF),并分析TLR4阳性细胞表达百分率和GMF与内毒素水平及CRP、PCT的相关性。结果 乙型肝炎相关性肝癌组TLR4阳性细胞表达百分率和GMF分别为79.56%±8.12%、54.14±12.28,较慢性乙型肝炎组(46.15%±5.35%、32.27±6.46)及正常对照组(14.89%±8.21%、15.21±6.87)显著升高,差异有统计学意义(P<0.05)。乙型肝炎相关性肝癌患者TLR4阳性细胞百分率和TLR4 GMF与内毒素水平呈正相关(P<0.05),与临床指标PCT、CRP无显著相关性(均P>0.05)。结论 TLR4在乙型肝炎相关性肝癌患者外周血CD14+单核细胞表面表达有显著升高,提示TLR4可能在乙型肝炎相关性肝癌的发生和发展中具有促进作用。  相似文献
