??Objective    To discuss the expression of inflammation factors COX-2 and MMP-9 in periodontal tissue and its significance in the experimental acute cerebral ischemia.  Methods    The study was conducted in the laboratory of Stomatological hospital of Hebei Medical University in June 2012. Totally 30 male Wistar rats of 300 g or so were randomly divided into 3 groups?? 10 rats in each group. Four vessel occlusion was used to establish the model of cerebral ischemia in rats. The 3 groups of rats were respectively killed immediately and 3 d and 7 d after modeling?? and the periodontal tissue of 5 μm thick was produced in the maxillary first molar. HE staining was performed to observe the periodontal tissues?? and the expression of MMP-9 and COX-2 in periodontal tissues was observed by immunohistochemical staining ??SP method??. Image analysis of immunohistochemical staining of the specimens of each group were performed by professional image analysis software. Results    The expression of COX-2 and MMP-9 in the 3-day group after the acute cerebral ischemia model rats was significantly higher than those in the immediate death group ??P < 0.05??. The expression of COX-2 and MMP-9 in the 7-day group was significantly higher than those in the 3-day group??P < 0.05??. Conclusion    After onset of acute cerebral ischemia?? the inflammatory cytokines COX-2 and MMP-9 in periodontal tissues are increased?? and periodontal destruction has begun.  相似文献

目的    从转录水平探讨牙龈组织中白细胞介素-8(IL-8)与慢性牙周炎的关系。方法    选择2000年3—9月四川大学华西口腔医学院门诊就诊的全身无系统性疾病的患者32例,分为牙周健康组和慢性牙周炎组,各16例。采用原位杂交法检测32例患者牙龈组织中IL-8 mRNA表达,分析IL-8 mRNA的检出率及IL-8分泌细胞在牙龈组织中的分布。结果    慢性牙周炎组IL-8 mRNA检出率高于牙周健康组,IL-8阳性细胞分布于牙龈上皮和结缔组织中。结论    IL-8 mRNA不仅存在于牙周炎组织中,在正常组织中也有检出,IL-8 mRNA致炎的具体机制尚有待于进一步研究。  相似文献

Methods    Primary cultures of hPDLCs were established and obtained from the extracted premolars of 3 patients for orthodontic treatment. hPDLCs of were respectively treated with 0.1% absolute ethyl alcohol ??control group???? 10 μg/mL Pg-LPS ??simple Pg-LPS group??A???? 10-10 mol/L 1??25D ??low 1.25 D group??B???? 10-8 mol/L 1??25D ??High 1.25 D group??C???? 10-10 mol/L 1??25D+10 μg/mL Pg-LPS ??group D???? 10-8mol/L 1??25D+ 10 μg/mL Pg-LPS ??group E?? for 24 h and 48 h. The supernatant fluid samples were collected and the IL-8 and IL-6 concentration was determined with enzyme-linked immunosorbent assay ??ELISA?? method. Results    ??1?? The 10 μg/mL Pg-LPS exerted the highest promotion effect on the expression level of IL-8 by 38.86-folds at 48 h ??P??0.001?? and that of IL-6 by 6.19-folds at 24 h ??P??0.001?? respectively. ??2?? 1??25D down-regulated the IL-8 level in a dose- and time- dependent manner. The highest inhibition effect occurred at 48 h?? the IL-8 level of cells treated with 10-8mol/L 1??25D was 49.94% of that of the control cells ??P??0.001??. ??3?? 1??25D significantly decreased the LPS-induced IL-8 and IL-6 production. At 48 h?? 10-8mol/L 1??25D combined with Pg-LPS significantly down-regulated the IL-8 level in hPDLCs?? 71.98% of that in the cells treated with Pg-LPS alone??P??0.001??. ??4?? At 24 h?? the level of IL-6 in cells treated with Pg-LPS combined with 10-8mol/L 1??25D was 84.51% of that in cells treated with Pg-LPS alone ??P=0.003??. Conclusion    Our results suggest that vitamin D may suppress the periodontal inflammation partly by inhibiting the expressions of innate IL-8 and Pg-LPS-induced IL-8 and IL-6 in hPDLCs.  相似文献

目的 探讨牙龈卟啉单胞菌(Porphyromonas gingivalis,P.gingivalis)W83、ATCC33277刺激人牙周膜成纤维细胞(HPDLFs)分泌基质金属蛋白酶-1(MMP-1)、金属蛋白酶组织抑制剂-1(TIMP-1)的变化.方法 本研究于2010年9月至2011年6月在中国医科大学口腔医学院中心实验室进行.将P.gingivalis W83、ATCC33277作用于HPDLFs0、6、12、24、48h后,运用酶联免疫吸附法(ELISA)检测细胞上清液中MMP-1、TIMP-1质量浓度变化,并计算MMP-1/TIMP-1值.结果 P.gingivalis感染HPDLFs的MMP-1和TIMP-1表达均增强,并呈时间依赖性;且MMP-1/TIMP-1值明显高于未感染的对照组(P<0.05).P.gingivalis W83感染后MMP-1/TIMP-1值明显高于P.gingivalis ATCC33277(P< 0.05).结论 P.gingivalis具有促进HPDLFs分泌MMP-1、TIMP-1的作用,且可造成牙周组织破坏;P.gingivalis W83降解细胞外基质的能力高于P.gingivalisATCC33277.  相似文献

目的 探讨双氯芬酸钠对脂多糖(LPS)诱导的人牙周膜成纤维细胞(HPDLFs)白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)表达的影响。方法 本研究于2013年3—5月在山西医科大学口腔实验室进行。将复苏培养冻存的HPDLFs进行形态学鉴别后,用10 μg/mL的LPS进行诱导,分别以0.1、1、10、50、100 mg/L的双氯芬酸钠进行干预,酶联免疫吸附试验法(ELISA法)检测HPDLFs表达IL-1β和TNF-α水平的变化。结果 对同一质量浓度LPS诱导的HPDLFs而言,双氯芬酸钠能下调HPDLFs表达IL-1β和TNF-α的水平,并且随着双氯芬酸钠质量浓度的增加,IL-1β和TNF-α的表达量呈逐渐减弱的趋势(P<0.05)。结论 双氯芬酸钠可能对于LPS诱导的HPDLFs IL-1β和TNF-α的表达有重要的抑制作用,这为牙周病的临床治疗提供了新的思路。  相似文献

目的:探讨在不同强度及不同时间的静压力作用下,髁突软骨细胞中MMP-13表达的变化及意义。方法 :体外分离培养SD大鼠髁突软骨细胞,用甲苯胺蓝及Ⅱ型胶原免疫细胞化学染色进行鉴定;分别用0、50、100、150、200 k Pa静压力进行2 h的加压处理。另外在200 k Pa静压力下,分别对髁突软骨细胞进行0、1、2、4、8和12 h的加压,采用Western blot免疫印迹及实时荧光定量PCR,分析髁突软骨细胞在不同强度及不同时间的静压力加载后,MMP-13的m RNA和蛋白表达变化。结果 :在50~200 k Pa的静压力范围内,随着压力的增大,MMP-13蛋白及m RNA表达水平均呈下降趋势,实验组表达水平均降至对照组(0 k Pa组)以下,且差异有统计学意义(P<0.05)。在200 k Pa的静压力下,随着加压时间的延长,MMP-13表达呈先上升后下降的趋势,且4 h实验组达最高水平(P<0.05)。结论:髁突软骨细胞对压力微环境的改变具有较好的适应能力,压力的改变可能影响颞下颌关节的适应性改建。  相似文献

??Abstract??Objective To discuss the effect of PG0839 of Porphyromonas gingivalis ??P.gingivalis?? on inflammatory cytokine expression?? including interleukin 1β and interleukin 6?? in liver tissues and serum of mice. Methods The study was completed in the central laboratory of School of Stomatology?? China Medical University from Jun. 2013 to Nov. 2013. Group 1 was challenged with P.gingivalis W83 strain at the doses of 1×109 CFU/ml cells. Group 2 was challenged with PG0839-defective mutant at the same dose. All mice in both groups were challenged with subcutaneous injections of 0.1 mL of bacterial suspension at two sites on the dorsal surface. Control group was challenged with 0.1 mL sterile PBS only. After 5 days?? all mice were sacrificed?? and the liver and serum were stored. The levels of IL-1β and IL-6 in mice liver tissue and serum were detected using ELISA. Analyze all result data with statistics software SPSS 13.0. Results IL-1β expression in the liver tissue and serum of mice was lower in group 2 than in group 1 ??P < 0 05??. Compared with the control group?? IL-1β and IL-6 expressions in the liver tissue and serum of mice were significantly increased in group 1 and 2 ??P < 0 05??. Conclusion These results suggest that PG0839 may participate in the process of P.gingivalis inducing systemic inflammation in mice.  相似文献

目的:观察大鼠实验性牙周炎基础治疗前后血清中IL-18与sTREM-1水平的变化,进而探讨IL-18与sTREM-1水平变化与牙周炎治疗效果间的关系。方法:选取2个月大的健康雄性Wistar大鼠30只,同等条件下饲养,随机分为3组,每组10只。正常对照组(A组)不做其他处理;牙周炎组(B组)与牙周炎治组(C组),将消毒尼龙丝线放置在双侧上颌第二磨牙牙颈部,同时配合牙周炎食谱诱导大鼠牙周炎模型。建模成功后C组去除丝线进行基础治疗,2周后对大鼠进行腹主动脉取血并处死,采用酶联免疫吸附法(ELISA)测定大鼠血清中IL-18和sTREM-1水平。结果:通过HE染色观察各组牙周组织学改变,牙周炎组(B组)大鼠血清中IL-18及sTREM-1水平高于正常对照组(A组)和牙周炎治疗组(C组)(P<0.05),且牙周炎治疗组大鼠血清中IL-18及sTREM-1水平高于对照组(P<0.05)。结论:牙周炎时IL-18、sTREM-1水平明显增高,且经基础治疗后明显降低。表明IL-18、sTREM-1水平可以为牙周炎的诊断和预测提供指导,为临床工作者提供准确有效的信息,同时也为临床治疗提供新的治疗靶点。  相似文献

目的:研究上颌快速扩弓前后龈沟液(gingival crevicular fluid,GCF)中白细胞介素-1β(interleukin-1β,IL-1β)及金属基质蛋白酶-8(matrix metalloproteinases-8,MMP-8)水平的变化,及其与上颌牙周组织改建的关系。方法:34例需上颌快速扩弓的正畸患者,随机分为对照组(戴入Hyrax扩弓器但不加力)和实验组(持续扩弓1周),各17例。分别在扩弓前(基线)、扩弓后24h、1周,保持1、3、5、7、9周时进行临床检查,并检测GCF量及IL-1β、MMP-8的水平,从而进行统计学分析。结果:与基线比较,实验组GCF量、IL-1β及MMP-8水平呈动态变化,扩弓后24h开始升高(P〈0.05),扩弓后1周达峰值(P〈0.001),此后逐渐下降,但一直保持较高水平(P〈0.05)。组间比较显示,整个实验过程中,两组间临床指标差异无统计学意义,从扩弓后24h至保持7周,实验组GCF量、IL-1β及MMP-8水平与对照组比较有统计学差异(P〈0.05),而至保持9周时无统计学差异。结论:上颌快速扩弓过程中,GCF中IL-1β及MMP-8水平发生了动态变化,在一定程度上可以反映快速扩弓后牙周组织的改建。  相似文献

目的探讨种植体周围龈沟液中骨桥蛋白(OPN)水平与种植体周围炎的关系。方法选择山西医科大学第一附属医院口腔科接受牙种植术戴冠后12个月的患者30例(40枚种植体),按照临床指标标准分为炎症种植体组[探诊深度(PD)>3 mm、牙龈出血指数(SBI)>2 mm,28枚种植体]和健康种植体组(PD≤3 mm、SBI≤2,12枚种植体);同时选择患者同名健康牙(40颗)为对照组。检查并记录所有患者PD、SBI;检测龈沟液量及其中OPN质量浓度(ABC-ELISA法)。结果 (1)炎症种植体组的PD值、SBI值、龈沟液量、OPN质量浓度均高于对照组和健康种植体组(均P<0.05);对照组和健康种植体组间各指标差异无统计学意义(均P>0.05)。(2)种植体周围龈沟液中OPN质量浓度同各临床指标间均呈正相关关系。结论种植体龈沟液中OPN质量浓度可在一定程度上反映种植体周围组织的健康状态。  相似文献

Expression of interleukin-2 receptor (IL2R) and HLA-DR on lymphocytes of gingival crevicular fluid (GCF) was examined by two-color flow cytometric analysis. GCF from 15 patients with periodontitis was collected by crevicular washing. Mononuclear cells were isolated by Ficoll-paque gradient centrifugation from inflamed gingival tissue (GT) and peripheral blood (PB) sampled from each of the 15 patients. Lymphocyte subsets were detected by using monoclonal antibodies (mAb) of Leu 12 (CD19), Leu 4 (CD3), Leu 3a (CD4) and Leu 2a (CD8) directed to B cells, T cells, helper/inducer T cells (Th) and suppressor/cytotoxic T cells (Ts), respectively. Anti-IL2R (CD25) and anti-HLA-DR were used as lymphocyte activation markers. IL2R- or HLA-DR-positive fractions in Th, Ts and B cells were calculated. Percentage of IL2R-positive fraction in Th (IL2R+ Th) of GCF (34.0%) was significantly higher than those of GT (18.4%) and PB (13.7%). IL2R-positive fraction in B cells (IL2R+ B) of GCF was the highest among the three groups (23.9% in GCF, 12.5% in GT, 6.3% in PB). Ts did not express IL2R regardless of the origin of the samples. Compared with PB and GT, GCF showed significantly higher HLA-DR expression on Th and Ts in GCF (PB: 8.7% and 27.1%; GT: 27.9% and 50.3%; GCF: 44.7% and 65.3%). These results suggest that lymphocytes in GCF were highly activated and are related to the local host immune response in periodontitis.  相似文献

目的检测慢性牙周炎患者牙周基础治疗前后龈沟液中白细胞介素-4(IL-4)的质量浓度,探讨IL-4与牙周炎的关系及其在牙周炎发病机制、病情进展等方面所起的作用。方法用滤纸条浸润法采集成年健康者和牙周炎患者治疗前后的龈沟液样本,用酶联免疫吸附测定检测样本中IL-4的质量浓度。结果慢性牙周炎患者龈沟液中IL-4的质量浓度低于健康对照组(P<0.05)。经牙周基础治疗1个月后,IL-4的质量浓度无明显变化,治疗前后的差异无统计意义(P>0.05);IL-4的质量浓度与探诊深度呈显著负相关,与牙龈指数和附着丧失无明显相关性。结论IL-4缺乏可能会导致牙周病的发生,IL-4可作为早期诊断牙周病和检测易患人群的敏感性指标。  相似文献

目的:探讨前列腺素E2(Prstaglandin E2,PGE2)在种植体周龈沟液中的含量水平与种植牙牙周组织临床指数-菌斑指数(plaque index,PI),牙龈指数(gingival index,GI)和牙周探诊深度(probing pocket depths,PPD)之间的关系.方法:检查实验组36颗和对照组36颗种植体牙周情况,实验组为有明显炎症的种植体,牙周探诊深度均超过3mm.同时试纸收集种植体周围龈沟液,ELISA法检测其龈沟液中的PGE2含量,所得数据用t检验和Pearson相关分析进行统计学处理.结果:PGE2表达和患种植体周围炎的种植体牙周指数呈显著相关(P<0.05),且实验组和对照组的PGE2表达统计学差异明显(P<0.05).结论:龈沟液中PGE2含量可为种植体周围炎病变的诊断提供客观参考.  相似文献

INTRODUCTION: Cytokines are of major importance in periodontal disease progression. Interleukin-12 (IL-12) stimulates interferon-gamma production by T helper type 1 (Th1) cells while IL-18 induces Th1 responses when present with IL-12 but Th2 responses in the absence of IL-12. IL-1beta has been correlated with periodontal disease destruction. This study determined the local concentrations of these cytokines in sites of gingivitis and periodontitis. METHODS: Gingival crevicular fluid was collected from two sites in each of 10 gingivitis patients and from two gingivitis sites and two periodontitis sites from each of 10 periodontitis patients. Serum samples were also collected. IL-1beta, biologically active IL-12 p70, the IL-12 p40 subunit and IL-18 concentrations were determined by enzyme-linked immunoabsorbent assay. RESULTS: IL-1beta and IL-18 concentrations were higher in the gingival crevicular fluid from periodontitis patients than in that from gingivitis patients; IL-18 concentrations were higher than those of IL-1beta. Very little IL-12, either p40 or p70, was detected in the gingival crevicular fluid samples. In the serum, very low levels of cytokines were found. The level of serum IL-12 p40, however, was higher than in the fluid from periodontitis sites of periodontitis patients. CONCLUSION: The local production of IL-1beta and IL-18 in the gingival crevicular fluid increased with increasing inflammation and IL-18 was the predominant cytokine at both gingivitis and periodontitis sites. Very little IL-12 was detected with levels decreasing with increasing inflammation. These results suggest that there is an association between severity of periodontal disease and levels of IL-1, IL-12 and IL-18.  相似文献

目的:探讨不同牙科材料对龈沟液中白介素-1(IL-1)表达水平的影响。方法:收集牙体缺损需全冠修复的病例256例(312个牙),分别采用镍铬合金烤瓷冠、金合金烤瓷冠及ISP Empress2全瓷冠进行修复。完成修复6个月后,用Whatman滤纸采集龈沟液标本,用酶联免疫吸附法测定龈沟液中IL-1的含量。结果:三种全冠对龈沟液中IL-1的影响:完成修复6个月后镍铬合金烤瓷冠和对照组相比有差异性(p〈0.05),且高于金合金烤瓷冠和全瓷冠的水平,而金合金烤瓷冠和全瓷冠与对照组相比均无统计学意义(p〉0.05)。结论:不同牙科修复材料对龈沟液中IL-1的含量有影响,从而为临床全冠材料的选择提供了具有生理意义的理论依据。  相似文献

目的 比较广泛型侵袭性牙周炎患者与健康人龈沟液中白介素-17(IL-17)的表达水平及与临床指标的关系。方法 选择广泛型侵袭性牙周炎患者18例和健康人16例,共68颗牙,记录临床牙周检查指标,用滤纸条法收集龈沟液。采用抗体夹心ELISA法测定广泛型侵袭性牙周炎患者治疗前、治疗后3个月及健康对照者龈沟液中IL-17总量和浓度。结果 广泛型侵袭性牙周炎患牙治疗前龈沟液中IL-17的总量和浓度高于牙周健康牙,基础治疗3个月后广泛型侵袭性牙周炎患牙龈沟液中IL-17的总量和浓度较治疗前下降。广泛型侵袭性牙周炎患牙龈沟液中IL-17浓度与探诊深度、附着丧失和出血指数呈正相关关系,而IL-17总量仅与探诊深度呈正相关关系。结论 龈沟液中IL-17浓度可能与广泛型侵袭性牙周炎牙周破坏及炎症的严重程度相关。?  相似文献

目的分析种植体组不同时期龈沟液(peri-implant sulcular fluid,PISF)中(interleukin,IL)-17A和IL-35水平改变,探讨其在种植体周围炎或黏膜炎发展过程中的作用和相互关系。方法收集2008—2013年在中山大学孙逸仙纪念医院口腔科行种植修复患者共40例为研究对象,分为种植体健康组20例和种植体炎症组20例,将种植体炎症组经过治疗后作为对照组。在种植体中有单冠修复、联冠修复。采集不同患者龈沟液并定量,对标本采用双抗体夹心酶联免疫吸附方法 (Enzyme Linked Immunosorbent Assay,ELISA)测定各组龈沟液中IL-17A和IL-35表达水平。并分别记录健康组、种植体炎症组、治疗后种植体炎症组探诊出血指数,改良出血指数,牙龈指数,并行统计学分析。结果种植体炎症组IL-17A,IL-35水平比健康组IL-17A,IL-35水平高(P<0.05),炎症组IL-17A水平治疗前比治疗后高(P<0.05),炎症组IL-35水平治疗后比治疗前升高(P<0.05),Pearson相关检验示,IL-35和IL-17A表达成负相关关系(P<0.05)。结论 IL-35和IL-17A的表达与种植体黏膜炎或者周围炎的发生发展密切,IL-17A和IL-35是辅助种植体周围炎诊治和评估抗炎疗效的潜在分子生物学指标。  相似文献
