目的测定鼻息肉病人鼻息肉组织和血清中超氧化物歧化酶(superoxidedismutase,SOD)活性、丙二醛(malondialdehyde,MDA)含量的变化并探讨其意义。方法通过生物化学方法分别测定30例鼻息肉病人鼻息肉组织和血清中SOD的活性与MDA含量,以20例鼻中隔偏曲病人的下鼻甲黏膜和血清作为对照组。结果①实验组SOD活性显著低于对照组(P<0.01)。②实验组MDA含量显著高于对照组(P<0.01)。结论①鼻息肉病人清除自由基能力下降,脂质过氧化反应活跃。②提示自由基代谢紊乱为鼻息肉的发病机制之一。  相似文献

目的 观察慢性鼻窦炎病变组织及鼻息肉中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量及其与病变程度的关系.方法 分别采用黄嘌呤氧化酶法和TBA法检测15例慢性鼻窦炎病变黏膜和鼻息肉组织术中及术后2周、1个月、2个月、3个月术区黏膜标本中SOD活性和MDA含量,其中Ⅰ、Ⅱ和Ⅲ型病例各5例;同时以5例正常中鼻甲黏膜作为对照组.结果 慢性鼻窦炎病变黏膜中SOD活性较对照组明显降低,且病变越重,SOD活性越弱;鼻息肉中SOD活性较同型病变黏膜更低.术后各型恢复期黏膜SOD活性随时间延长而逐渐增强.鼻窦炎病变黏膜中MDA含量显著高于鼻息肉和对照组黏膜,且随病变程度加重而增高;术后各型恢复期黏膜中MDA含量显著低于术中病变黏膜,且随术后时间延长而逐渐降低.SOD活性和MDA含量呈负相关.结论 自由基损伤程度与慢性鼻窦炎的病变严重性密切相关,自由基代谢紊乱可能在慢性鼻窦炎的发病机制中起重要作用.  相似文献

目的 观察银杏叶提取物一金纳多(Extract ofGinkgo Biloba Leaves Injection,EGB)对抗顺铂(Cisplatin,CDDP)耳毒性的作用。方法 将24只豚鼠随机分为CDDP组、EGB CDDP组及对照组,采用听性脑干反应(auditory brainstem response,ABR)、血清超氧化物歧化酶(superoxide dismutase,SOD)活性和丙二醛(MDA)含量、光镜及扫描电镜技术,观察用药前后各指标的变化。结果CDDP组动物ABR阈值较其它两组明显升高(P<0.01),其余两组间差异亦有显著性(0.010.05)。血清MDA含量以CDDP组升高最显著(P<0.01),EGB CDDP组较对照组略有升高,没有显著性差异(P>0.05)。耳蜗扫描电镜显示EGV CDDP组较CDDP组毛细胞受损程度明显减轻。结论EGB可有效减轻CDDP的耳毒性作用。  相似文献

对慢性扁桃体炎、扁桃体摘除术前1d及术后1周患者血清超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量进行了测定。结果示扁桃体炎患者血清T-SOD、Mn-SOD、CuZn-SOD活力降低,MDA含量增加,与正常对照组比较差异非常显著(P<0.001);术后1周血清T-SOD活力较术前1d明显增加、MDA含量下降与术前1d比较均有非常显著差异(P<0.001,P<0.01)提示扁桃体摘除术可使慢性扁桃体炎患者MDA含量降低、SOD活力增加。  相似文献

目的:探讨白细胞介素—5(IL—5)及嗜酸性粒细胞阳离子蛋白(ECP)在鼻息肉发病中的作用及相互关系。方法:采用pharmacia CAP荧光免疫系统和ELISA双抗体夹心法对30例鼻息肉患者(鼻息肉组)和8例鼻中隔偏曲或阻塞性睡眠呼吸暂停综合征(OSAS)患者(对照组)分别进行血清中ECP及组织匀浆中ECP、IL—5的检测。结果:鼻息肉组匀浆中IL—5的水平明显高于对照组,其差异有显著性意义(P<0.05);鼻息肉组血清与匀浆中的ECP含量明显高于对照组,其差异亦有显著性意义(P<0.05)。鼻息肉组匀浆中IL—5与血清中ECP水平呈明显正相关(r=0.598,P<0.05);与匀浆中的ECP水平也呈正相关(r=0.451,P<0.05)。结论:ECP是嗜酸性粒细胞活化的标志,也是导致鼻腔炎症发生的重要因子;IL—5在鼻息肉组织中高表达,并与血清和组织中ECP水平密切相关,共同促进鼻腔炎症过程的不断加重。  相似文献

目的:通过测定鼻息肉中核因子κB亚单位p50(NF-κB-p50)的活性及其与IL-4、γ-IFN表达的关系,来探讨p50对Th1/Th2细胞因子表达调节的可能机制.方法:采用SP免疫组织化学染色方法检测40例鼻息肉患者和20例正常钩突黏膜中IL-4、γ-IFN、p50的表达,以核染阳性率来评估p50活性.对p50活性与IL-4、γ-IFN因子进行直线相关分析.结果:①鼻息肉中p50细胞质、细胞核均有阳性表达,主要位于上皮细胞、炎症细胞及腺上皮细胞细胞质,p50活性显著增强(P<0.01),IL-4表达亦明显增强(P<0.01),γ-IFN表达无明显改变(P>0.05);②Pearson相关性分析,发现p50活性与IL-4表达呈直线正相关关系,r=0.70(P<0.01),而与γ-IFN表达无相关性,r=0.14(P>0.05).结论:鼻息肉中p50的活性明显增强,p50的激活上调IL-4的表达可能是鼻息肉中Th1/Th2因子失衡的重要环节之一.  相似文献

鼻息肉中IgE及GM-CSF与嗜酸粒细胞浸润的相关性研究   总被引:2,自引:0,他引:2  
目的:探讨鼻息肉组织中IgE和粒细胞巨噬细胞集落刺激因子(GM-CSF)的表达对嗜酸粒细胞浸润聚集的作用及二者的关系及意义。方法:采用免疫组织化学SP法检测30例鼻息肉标本(鼻息肉组)和11例下鼻甲部分切除术患者下鼻甲黏膜标本(对照组)组织中IgE阳性细胞分布情况;酶联免疫吸附法检测上述标本中GM-CSF含量,同时观测2组组织中嗜酸粒细胞的浸润程度。结果:鼻息肉组中IgE、GM-CSF水平及嗜酸粒细胞数均明显高于对照组(均P<0.01),且IgE与GM-CSF的水平分别与鼻息肉组织中嗜酸粒细胞浸润数呈正相关(r=0.65、0.62,均P<0.01),鼻息肉组中IgE与GM-CSF水平呈正相关(r=0.52,P<0.01)。结论:鼻息肉是以嗜酸粒细胞浸润为特征的疾病过程,IgE及GM-CSF则对鼻息肉中嗜酸粒细胞浸润发挥重要作用,IgE介导的Ⅰ型变态反应在鼻息肉的形成和发展中占有重要地位。  相似文献

目的 :测定鼻咽癌患者血清中活性氧 (ROS)、超氧化物歧化酶 (SOD)和谷胱甘肽过氧化物酶 (GSH PX)的活性水平 ,以探讨鼻咽癌患者体内自由基代谢状态及其在鼻咽癌发生、发展中的作用。方法 :分别采用Fenton反应显色法 ,黄嘌呤氧化酶法和二硫代二硝基苯甲酸法对 4 0例鼻咽癌患者 (鼻咽癌组 )和 2 0例健康体检者 (对照组 )血清中ROS、SOD和GSH PX水平进行检测。结果 :鼻咽癌组血清中ROS活性 [(11 32 7.835±4 83.777) /(U·ml-1) ]显著高于对照组 [(10 2 6 2 .712± 5 2 5 .2 87) /(U·ml-1) ](P <0 .0 5 ) ,SOD和GSH PX活性[(76 .6 19± 2 2 .2 83) /(NU·ml-1) ,(98.6 5 3± 4 6 .374 ) /AU ]显著低于对照组 [(15 4 .6 0 3± 2 7.2 4 1) /(NU·ml-1) ,(30 7.872± 116 .2 75 ) /AU](P <0 .0 5 ) ;不同病理类型鼻咽癌患者血清中ROS、SOD和GSH PX活性间的差异均无显著性意义 (P >0 .0 5 )。结论 :鼻咽癌患者体内自由基代谢紊乱 ,抗氧化损伤能力显著下降 ,给予抗氧化治疗有利于抑制鼻咽癌的发展  相似文献

The aim of this study was to investigate the role of nitric oxide (NO), free oxygen radicals, and scavenging enzymes in the development of nasal polyp (NP) disease. This study included 41 patients who underwent endoscopic surgery because of NPs. Control specimens were taken from the inferior turbinate of 32 patients who underwent septoplasty. The levels of malondialdehyde (MDA), NO, and superoxide dismutase (SOD) were measured in intraoperative specimens of polyp tissue and turbinate mucosa. The levels of tissue NO were 191.06 +/- 26.62 micromol/mg of protein in patients with NPs and 145.30 +/- 19.19 micromol/mg of protein (p < .001) in the control group. The levels of MDA in the study and control groups were 12.47 +/- 2.12 nmol/mg and 8.83 +/- 1.08 nmol/mg (p < .01), respectively. The levels of SOD in the study and control groups were 50.77 +/- 14.74 U/mg and 77.93 +/- 15.31 U/mg (p < .001), respectively. It was determined that the levels of MDA in plasma and erythrocytes were higher in the patients with NPs than in the control group (p < .05). The levels of NO in plasma and erythrocytes in both groups were similar. The levels of SOD in plasma and erythrocytes were lower in patients with NPs than in the control group (p > .05). Increases in the levels of tissue MDA and NO and decreases in scavenging enzymes in patients with NPs as compared to control groups may indicate the presence of free radical damage in patients with nasal NPs. New studies are needed to clarify the efficacy of using antioxidants in the treatment of NPs.  相似文献

The aim of this study was to investigate serum paraoxonase (PON) and arylesterase (ARE) activities, the total oxidant status (TOS), total antioxidant status (TAS), and oxidative stress index (OSI) in the patient with nasal polyp (NP) and control group to confirm the link between oxidative stress and NP. In this prospective study, 32 patients with NP (22 males and 10 females) and 29 healthy controls (19 males and 10 females) were enrolled. Preoperative serum levels of PON, ARE, TAS, and TOS were determined, and OSI was calculated. Serum PON activity, ARE activity, and TAS tend to be decreased in patients with NP compared to healthy controls (P < 0.0001). TOS and OSI levels were significantly higher in the NP patients than in the healthy controls (P < 0.01). Furthermore, there was established positive correlation between serum PON activity and serum ARE activity in patients with NP (r = 0.454; P = 0.009). Our results support the belief that altered PON and ARE activity may be involved in the pathogenesis of NP. Increased TOS and OSI, and decreased TAS indicated that NP is the pathologic consequence of oxidative stress.  相似文献

目的:探讨天然免疫蛋白腭、肺、鼻上皮克隆(PLUNC)在鼻息肉组织中的表达情况,分析PLUNC蛋白浓度与鼻息肉大小(鼻内镜记分)和手术后复发的关联性。方法:采用免疫组织化学和实时定量PCR检测鼻息肉组(28例鼻息肉患者,其中13例为术后复发患者)和对照组(16例钩突黏膜对照)PLUNC的组织染色定位和mRNA的表达情况,同时采用ELISA检测初发和复发鼻息肉组织中PLUNC的蛋白浓度差异,评估PLUNC蛋白浓度与息肉大小、鼻塞、流涕症状记分的相关性。结果:鼻息肉组织中PLUNC主要定位在黏膜上皮和腺体,染色强度记分显著低于对照的钩突组织(P〈0.01);PLUNCmRNA表达水平也显著低于对照的钩突组织(P〈0.01)。初发和复发鼻息肉组织中PLUNC蛋白OD值分别为0.33±0.11和0.15±0.05,差异有统计学意义(P〈0.01)。小型和大型鼻息肉PLUNC蛋白OD值分别为0.32±0.14和0.19±0.07,差异有统计学意义(P〈0.05)。鼻息肉组织中PLUNC蛋白浓度与鼻塞和流涕记分存在显著负相关(r=-0.51和r=-0.57,P〈0.01)。结论:PLUNC的表达降低提示鼻息肉的发生可能与天然免疫反应的减弱有关,因此,上调天然免疫分子如PLUNC等的表达可作为干预鼻息肉发病的一个新策略。  相似文献

目的明确单核细胞趋化蛋白1(monocyte chemotactic protein 1,MCP-1)和血管内皮生长因子(vascular endothelial growth factor,VEGF)在鼻息肉组织中的表达及其相关性,初步探讨MCP-1与鼻息肉发生的关系。方法取40例鼻息肉组织和25例下鼻甲组织,应用原位杂交和免疫组织化学等方法检测MCP-1和VEGF mRNA及蛋白质的表达。结果鼻息肉组织中MCP-1和VEGF mRNA及蛋白质的表达均高于对照组下鼻甲组织(P值均〈0.01);鼻息肉组织中MCP-1和VEGF蛋白质的表达呈正相关(r=0.871,P〈0.05)。结论鼻息肉组织中MCP-1和VEGF表达增加,二者协同作用可能是鼻息肉形成的原因之一。  相似文献

The aim of our study is to evaluate the status of selenium and zinc in nasal polyp tissues and to investigate the possible role of trace elements and antioxidants including superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in nasal polyps. In this study, the antioxidant enzyme and trace element levels measured in polyp tissues of 37 patients were compared with the levels measured in conchal mucosa of 27 control cases. The antioxidant enzyme and trace element levels in tissues were measured with graphite and flame spectrophotometry methods using Shimatsu UV.1601 spectrophotometer and Perkin Elmer atomic spectrometer. The mean tissue zinc and selenium levels were, respectively, 2.55 μg/g and 30.03 pg/g in patient group, 4.37 μg/g and 44.95 pg/g in control group. The mean tissue SOD and GSH-Px levels were, respectively, 4.27 and 0.69 U/mg protein in patient group, 7.09 and 0.77 U/mg protein in control group. When the measured levels in patients and control cases were compared, there were statistically significant differences between zinc, selenium, and SOD levels (P = 0.001). There was no significant difference between GSH-Px levels (P = 0.465). In conclusion, it has been revealed that the levels of zinc, selenium, and SOD in nasal polyps were significantly lower, and it may be concluded that this may have a role in the development of nasal polyps.  相似文献

Objectives

To underline the effect of oxidative stress in chronic otitis media with and without cholesteatoma and to compare the oxidative stress values in the serum and tissue specimens in these two forms.

Methods

The study included a total of 75 individuals, 35 cases with chronic otitis media (COM; 16 females and 19 males) and a healthy control group of 40 cases (20 females and 20 males). The COM patient group was comprised of 18 patients with cholesteatoma and 17 patients without cholesteatoma. All patients underwent mastoidectomy. Serum specimens were taken prior to surgery and diseased tissue specimens from the ear were obtained during surgery from all patients. Only serum specimens were taken from the healthy control cases. The malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GHPx) were measured in the serum and tissue samples of the patient group and in the serum specimens of the control group.

Results

The age ranged from 14 to 48 years in the patient group (mean age, 20.4±12.2 years) and from 19 to 40 years in the control group (mean age, 26.4±4.64 years). When the serum values of all COM patients were compared with those of the control group, in the patient group MDA, which reflects lipid peroxidation, was found to be significantly higher (P<0.01) whereas the antioxidant enzymes SOD, CAT, and GHPx were found to be significantly lower (P<0.01). When the serum and tissue MDA, SOD, CAT, and GHPx values in patients with and without cholesteatoma were compared, no significant difference was found these parameters (P>0.01).

Conclusion

Although oxidative stress plays a role in the pathogenesis of COM with or without cholesteatoma, it may not reflect the severity of the disease. In patients with COM, the evaluation of only serum oxidative stress values without tissue evaluation may be sufficient for assessing oxidative stress.  相似文献

目的 :探讨鼻息肉组织中免疫球蛋白E(IgE)、白细胞介素 5 (IL 5 )和粒细胞巨噬细胞集落刺激因子(GM CSF)的表达对嗜酸性粒细胞 (EOS)浸润聚集的作用及IgE与IL 5和GM CSF的关系及其意义。 方法 :应用连续两步免疫酶法 (Sandwich)检测 31例鼻息肉标本 (鼻息肉组 )和 11例下鼻甲黏膜标本 (对照组 )组织匀浆中IgE水平 ;应用酶联免疫吸附测定法检测两组组织匀浆中IL 5和GM CSF水平 ,同时观测两组组织中EOS的浸润程度。结果 :鼻息肉组IgE、IL 5和GM CSF水平均明显高于对照组 (圴 P <0 .0 1) ,且三者水平分别与鼻息肉组织中EOS浸润数呈正相关 (r分别为 0 .73、0 .5 4和 0 .4 8,均P <0 .0 1) ,且鼻息肉中IgE与IL 5和GM CSF水平呈正相关 (r分别为 0 .6 5和 0 .4 2 ,分别P <0 .0 1和P <0 .0 5 )。结论 :IgE局部水平的上调提示鼻息肉中存在局部变态反应 ,其对鼻息肉中IL 5和GM CSF的表达上调和EOS的聚集具有重要作用 ;IL 5和GM CSF在鼻息肉中高表达 ,对EOS的浸润聚集具有重要作用。  相似文献
