Micafungin is one of three FDA-approved echinocandins, a novel class of antifungal agents that target 1,3-beta-D-glucan in the fungal cell wall. Its spectrum of activity and favorable safety profile have made it an attractive option in the treatment of invasive Candida and Aspergillus infections. Since its approval in 2005 in the US, a variety of studies describing micafungin's use in the pediatric population have been published. As with many drugs, the pharmacokinetic profiles observed in the pediatric population differ from those seen in adult studies. A thorough understanding of the unique characteristics of this drug in the pediatric population is essential in order to optimize treatment outcomes for this diverse population of patients. 相似文献

p53, the "guardian of the genome" and the most mutated gene in cancer presents a considerable therapeutic opportunity as well as a challenge. In the past decade, several therapeutic strategies have been developed that aim to take advantage of a wealth of knowledge about p53, including insights into the biology and patho-biology of p53. Nevertheless, considerable challenges remain, not least as a result of tissue- and cancer-specific differences in p53 regulation and/or function. p53 does not act in the same manner in all tissues or in the cancers arising from them. Nor is p53 regulated in the same way in the wide variety of tissues from which cancers develop. Therefore, potential strategies for therapeutic targeting need to be tailored to each tumour/tissue type. This review summarises some of these tissue- and cancer-specific issues to suggest how different strategies are required for cancers arising from different tissues and to illustrate the complexities of therapeutic targeting of p53. 相似文献

Heavy metals are commonly associated with the generation of reactive oxygen species (ROS), which may cause oxidative damage to several cellular macromolecules and organelles. In an attempt to correlate biomarker responses to oxidative stress, caged mussels (Mytilus galloprovincialis) were exposed for 30 days in a relatively clean site and two areas (Stations 1 and 2) unevenly polluted by heavy metals in Gulf of Patras (Greece). Three periods of caging were: one in winter, the second in spring, and the third in autumn. Heavy metal content was determined in digestive glands of the exposed mussels as a measure of metal pollution, metallothionein content as an adaptive and detoxifying index, lysosomal membrane stability as a biomarker of general stress, superoxide radical production and lipid peroxidation as indicators of oxidative stress, and micronucleus frequency in gill cells as an index of chromosomal damage. Considering that protein-synthesizing machinery is one of the candidate targets for ROS, the in vivo activity of ribosomes in digestive glands was also tested. Compared with the reference samples, mussels transplanted to Station 1 showed increased levels of heavy metals and metallothionein in digestive glands, lower lysosomal membrane stability, higher values in oxidative stress indices, reduced activity of ribosomes, and increased chromosomal damage in gill cells. In addition, run-off ribosomes isolated from mussels transplanted to Station 1 were less efficient at initiating protein synthesis in a cell-free system than those from mussels in the reference site. Mussels transplanted to Station 2 exhibited similar but less pronounced responses. Statistical analysis revealed a strong positive correlation of ribosomal activity with lysosomal membrane stability, as well as a significant negative correlation with the oxidative stress indices, metallothionein content, micronucleus frequency, and the digestive gland content in Cr, Cu and Mn. Silver nanoparticles, chemically neutral particles in the size range of 1–100 nm, express strong antimicrobial activity and therefore have a broad range of applications. The increased use of consumer products with nanosilver (nanoAg) may result in its release into the environment, and may particularly affect aquatic systems. The mechanisms of the harmful effects of nanoAg against aquatic organisms are unclear. Therefore, in the present study we investigate the pro‐oxidative potential of these nanoparticles in experimentally exposed crayfish Orconectes limosus. Markers of oxidative stress and parameters of the antioxidant cell defense system such as total glutathione, glutathione reductase and the level of sulfhydryl groups were examined in the hepatopancreas of both sexes of O. limosus collected seasonally from Bia?e Lake (Poland) and subsequently exposed to nanoAg particles for 2 weeks. Exposure to nanoAg led to a high concentration‐dependent increase in the rate of lipid peroxidation and a decrease of protein‐bound SH groups which indicates protein oxidation. These markers of oxidative stress were accompanied by decreased levels of thiols and reduced activity of glutathione reductase. These results indicate a deficiency of reduced glutathione and suggest that the exposed organisms have less efficient antioxidative mechanisms available to counter ROS‐mediated cellular stress. Furthermore, we find that confocal microscopy is of limited utility in monitoring the presence of silver nanoparticles in tissues of exposed crayfish. © 2013 Wiley Periodicals, Inc. Patients with treatment-refractory obsessive-compulsive disorder (OCD) are sometimes considered for surgical interventions. The identification of reliable predictors of outcome following such interventions would be of great clinical importance, as it would lead to stricter selection of suitable patients, thus avoiding unnecessary surgery and improving the overall response rate. We analyzed data from 24 severe treatment-resistant patients who underwent capsulotomy for OCD and were carefully followed-up one year after the surgery and at long term (mean 10.8 years after surgery). The Yale-Brown Obsessive Compulsive Scale Symptom Checklist was administered to assess the lifetime presence of the most common symptom types. We applied an algorithm to calculate the patients' scores on 4 well-established symptom dimensions: Contamination/cleaning, forbidden thoughts, symmetry/order and hoarding. Multiple regression models were employed to examine whether scores on certain symptom dimensions were predictive of long-term outcome. The presence and number of lifetime symptoms in the symmetry/order domain were associated with greater severity of OCD, depression and anxiety, as well as greater impairment in various functional domains like work, social and family life at both one-year and long-term follow-ups. These results remained consistently significant after controlling for preoperative psychopathology, scores on other OCD symptom dimensions, sex, age, age of onset, duration of follow-up, type of surgical procedure, number of operations and lesion volume. The results could have implications for existing ablative and deep brain stimulation protocols and challenge our current conceptualization of OCD as a unitary diagnostic entity with a single neurobiological substrate. 相似文献

The purpose of this study was to examine the toxic effects of cadmium-telluride (CdTe) quantum dots on freshwater mussels. Elliption complanata mussels were exposed to increasing concentrations of CdTe (0, 1.6, 4 and 8 mg/L) and cadmium sulfate (CdSO(4), 0.5mg/L) for 24h at 15 degrees C. After the exposure period, they were removed for assessments of immunocompetence, oxidative stress (lipid peroxidation) and genotoxicity (DNA strand breaks). Preliminary experiments revealed that CdTe dissolved in aquarium water tended to aggregate in the particulate phase (85%) while 15% of CdTe was found in the dissolved phase. Immunotoxicity was characterized by a significant decrease in the number of hemocytes capable of ingesting fluorescent beads, and hemocyte viability. The cytotoxic capacity of hemocytes to lyse mammalian K-562 cells was significantly increased, but the number of circulating hemocytes remained unchanged. Lipid peroxidation was significantly increased at a threshold concentration of 5.6 mg/L in gills and significantly reduced in digestive glands at a threshold concentration <1.6 mg/L CdTe. The levels of DNA strand breaks were significantly reduced in gills at <1.6 mg/L CdTe. In digestive glands, a transient but marginal increase in DNA strand breaks occurred at the lowest concentration and dropped significantly at the higher concentrations. A multivariate analysis revealed that the various response patterns differed based on the concentration of CdTe, thus permitting the identification of biomarkers associated with the form (colloidal vs. molecular) of cadmium. 相似文献

Fenthion is one of the most widely used organophosphate insecticides for the control of many varieties of pests in Nigeria. The genotoxic effect of the pesticide was evaluated in the blood erythrocytes of Clarias gariepinus using the micronucleus (MN) test. The oxidative stress parameters were also studied in the liver and gill tissues. Fish were exposed to 2.0, 4.0, and 8.0?mgL?1 of fenthion and sampling was done on days 1, 7, 14, 21 and after 7-day recovery. Micronuclei induction was highest (7.55) on day 14 at all concentrations in the peripheral blood cells. Oxidative stress was evidenced by increased lipid peroxidation (LPO). Maximum LPO values of 62.47% and 71.17% were observed in the gill and liver tissues respectively in C. gariepinus exposed to 8.0?mgL?1 concentration of fenthion. There were alterations in the values of reduced glutathione (GSH), glutathione reductase (GR), glutathione peroxidase (GPx), superoxide dismutase (SOD), and catalase (CAT) during the exposure and recovery periods. The 7-day recovery period was not adequate to eliminate fenthion-induced changes as LPO, CAT, and GR activity remain elevated. However, MN frequency and activity of SOD, GSH, and GPx (except at 8.0?mgL?1) recovered. The present findings give further credence on the integrated use of MN test and oxidative stress parameters in risk assessment of pollutants in aquatic ecosystem. 相似文献

The present study was carried out to elucidate whether lysosomal size reduction in digestive cells of mussels Mytilus galloprovincialis constitutes a selective response against a particular group of organic chemical compounds, in contrast to the lysosomal enlargement characteristic of general stress response. Mussels were treated with di(2-ethylhexyl) phthalate (DEHP), benzo(a)pyrene (B[a]P), and the water accommodated fraction (WAF) of a lubricant oil, which were daily applied by either injection through the adductor muscle for 7 days or water-exposure for 21 days. Control mussels were either kept untreated in clean sea water, or treated with acetone (injection and water-exposure), vehicle used for DEHP and B[a]P. A third set of controls consisted of mussels with pierced shell kept in clean seawater. Digestive glands were excised at various treatment days and beta-glucuronidase activity was demonstrated in 8-microm cryotome sections. Lysosomal volume, surface and numerical densities, and surface-to-volume ratio were quantified by image analysis. Other sections were stained with oil red 0 to demonstrate neutral lipids and changes in lipid levels were quantified by image analysis. Neutral lipid accumulation in digestive cells was used as a complementary indication of exposure to organic chemicals. It resulted to be a very prompt and all-or-nothing response, which reached a plateau before 1 day of treatment with WAF, DEHP and B[a]P after both injection and water-exposure. DEHP-treatment induced a general stress response characterised by lysosomal enlargement in digestive cells, which was already induced after 1 day. Treatment with either WAF or B[a]P elicited a comparable biphasic response. A transient lysosomal enlargement, shorter with WAF than with B[a]P treatment, was evidenced after both injection and water-exposure. Further, under water-exposure conditions, WAF reduced the endo-lysosomal system in size more markedly than B[a]P. Such lysosomal size reduction constitutes a transient response after exposure to diverse organic xenobiotics (acetone, WAF and B[a]P). In addition, this lysosomal size reduction might be followed by a further lysosomal enlargement, which later might yet again give rise to an apparent lysosomal size reduction under chronic exposure conditions. As a whole, the lysosomal response is intricate and cannot be simply interpreted in environmental pollution monitoring programmes. Increasing evidence suggests that oxidative stress may play a significant role in causing microcystin (MCs) toxicity not only in mammals, but also in fish. MCs are a family of cyclic peptide toxins produced by some species of freshwater cyanobacteria (blue-green algae). Among the microcystins, MC-LR is the most extensively studied. In the present study the differential response of the antioxidant enzymes superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione reductase (GR) as well as lipid peroxidation (LPO) as a biomarker of oxygen-mediated toxicity were assessed in liver, kidney and gill tissues of tilapia (Oreochromis sp.) exposed to MCs. Fish were injected intraperitoneally (i.p.) with a single dose of 500 microg/kg MC-LR or 500 microg/kg MC-RR and sacrificed after 7 days. The results show that MCs exposure induces adaptive responses such as increase in the antioxidant enzymatic activities, mainly those of SOD and CAT, as well as in LPO values. With regard to LPO values, the liver was the most affected organ by MC-LR. MC-RR, however, did not affect this parameter in the liver of the exposed fish. Oxidative stress biomarkers, therefore, are valuable tools in the assessment of early responses of fish to the increasing occurrence of cyanobacterial blooms worldwide. 相似文献

1. The effects of a single oral dose (100 mg) of flosequinan on systemic and regional (forearm, splanchnic and renal) vascular responses to simulated orthostatic stress (lower body negative pressure, LBNP) were investigated in nine healthy male volunteers, in a double-blind, placebo-controlled crossover study. 2. Forty-five minutes after its administration and before LBNP, flosequinan induced a significant decrease in total peripheral and in forearm vascular resistances without any concomitant change in arterial pressure, in heart rate and in the investigated biological parameters (plasma catecholamines, arginine vasopressin and renin activity). 3. After flosequinan and placebo, LBNP induced similar decreases in central venous pressure at all levels of LBNP (-10, -20 and -40 mm Hg) and in pulse pressure at LBNP -40 mm Hg. LBNP-induced increase in forearm vascular resistance was significantly more marked after flosequinan than after placebo at all levels of LBNP, and this was also true for splanchnic vascular resistance but at LBNP -40 mm Hg only. However, inasmuch as the basal values of these two parameters before LBNP were lower after flosequinan than after placebo, their final values after LBNP -40 mm Hg were similar. Finally, LBNP-induced changes in renal vascular resistance, glomerular filtration rate and filtration fraction as well as in plasma catecholamines, arginine vasopressin and renin activity were similar after flosequinan and placebo at all levels of LBNP. 4. Despite the availability of analytic data, little is known about the toxicity of salicylic acid (SA) on aquatic non-target organisms. The present study aimed at evaluating the impact of SA through a short-term exposure of the Mediterranean mussel Mytilus galloprovincialis to five environmentally relevant concentrations of SA. A set of suitable biomarkers was applied at selected time-points on mussel digestive glands, including histological observations and expression of oxidative stress related genes. The obtained results showed a conspicuous hemocytic infiltration among mussel digestive tubules, as confirmed also by a flow cytometric approach that revealed an increase of halinocytes and granulocytes. Interestingly, a significant dose and time dependent decrease in the expression levels of oxidative stress related genes was found in mussels exposed to SA except for the glutathione S-transferase gene that was significantly up-regulated in a time-dependent manner confirming its important role against oxidant species and in the metabolism of pharmaceuticals. 相似文献

Oxidative stress by increased production of reactive oxygen species has been implicated in the toxicity of many pesticides. Therefore, the aim of the present study was to investigate the effect of a broad spectrum insecticide, composed of a mixture of organophosphate plus pyrethroids (fenitrothion 25%, lambda cyhalothrin 2.5% and piperonyl butoxide 6%), on antioxidant status and oxidative stress biomarkers in rat brain. Different insecticide concentrations (0, 0.1, 1, 10, 100 and 1000 mM) were incubated with brain homogenate at 37 °C for time intervals (0, 30, 60, 120, 180 and 240 min). Exposure to insecticide mixture resulted in a significant increase (p < 0.05) in thiobarbituric acid reactive substances (TBARS), which might be associated with decreased levels of reduced glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), glutathione S-transferase (GST) and acetylcholinesterase activities and beside protein content in rat brain. However, a significant induction of lactate dehydrogenase (LDH) activities was observed. The response was concentration and time dependent. Results showed that the used insecticides had the propensity to cause significant oxidative damage in rat brain, which is associated with marked perturbations in antioxidant defense system in addition to antioxidant enzymes can be used as potential biomarkers of toxicity associated with pesticides exposure. 相似文献

Exposure to cement dust is one of the most common occupational dust exposures worldwide, but the mechanism of toxicity has not been fully elucidated. Cement dust (N) and clinker (C) samples collected from Nigeria and another sample of cement dust (U) collected from USA were evaluated using alveolar macrophage (NR8383) cell culture to determine the contribution of different sources of cement dust in the severity of cement dust toxicity. Cement dust particles internalization and morphologic alterations using transmission electron microscopy (TEM), cytotoxicity, apoptotic cells induction, intracellular reactive oxygen species generation, glutathione reduction, TNF-α, IL-1β, and CINC-3 secretion in alveolar macrophages (NR8383) exposed to cement dust and clinker samples were determined. Particles were internalized into the cytoplasmic vacuoles, with cells exposed to U showing increased cell membrane blebbing. Also, NR8383 exposed to U show more significant ROS generation, apoptotic cells induction and decreased glutathione. Interleukin-1β and TNF-α secretion were significantly more in cells exposed to both cement dust samples compared with clinker, while CINC-3 secretion was significantly more in cells exposed to clinker (p < 0.05). Endocytosis, oxidative stress induced-apoptosis and induction of pro-inflammatory cytokines may be key mechanisms of cement dust immunotoxicity in the lung and toxicity may be factory dependent. 相似文献

Oxidative stress derived from ultraviolet (UV) light in sunlight induces different hazardous effects in the skin, including sunburn, photo-aging and DNA mutagenesis. In this study, the protein-bound lipid peroxidation products 4-hydroxy-2-nonenal (HNE) and the oxidative DNA damage marker 8-hydroxy-2'-deoxyguanosine (8OHdG) were investigated in chronically sun-exposed and sun-protected human skins using immunohistochemistry. The levels of antioxidative enzymes, such as heme oxygenase 1 and 2, Cu/Zn-SOD, Mn-SOD and catalase, were also examined. Oxidative stress is also implicated in the activation of signal transduction pathways, such as mitogen-activated protein kinase (MAPK). Therefore, the expression and distribution of phosphorylated p38 MAPK, phosphorylated Jun N-terminal kinase (JNK) and phosphorylated extracellular signal-regulated kinase (ERK) were observed. Skin specimens were obtained from the surgical margins. Chronically sunlight-exposed skin samples were taken from the ante-auricular (n = 10) and sunlight-protected skin samples were taken from the post-auricular (n = 10). HNE was increased in the chronically sunlight-exposed skin but not in the sunlight-protected skin. The expression of heme oxygenase-2 was markedly increased in the sunlight-exposed skin compared with the sun-protected skin. In contrast, the intensity of immunostaining of Cu/Zn-SOD, Mn-SOD and catalase was not different between the two areas. Phosphorylated p38 MAPK and phosphorylated JNK accumulated in the ante-auricular dermis and epidermis, respectively. These data show that particular anti-oxidative enzymes function as protective factors in chronically sunlight-exposed human skin. Taken together, our results suggest (1) antioxidative effects of heme oxygenase-2 in chronically sunlight-exposed human skin, and that (2) activation of p38 MAPK may be responsible for oxidative stress. 相似文献
