目的:检测鼻内应用肾上腺皮质激素(激素)治疗鼻息肉前后鼻息肉组织中的细胞间黏附分子-1(intercellular adhesion molecule-1,ICAM-1)和鼻分泌物中的可溶性细胞间黏附分子-1(soluble intercellular adhesion molecule-1,sICAM-1)表达水平的变化,探讨ICAM-1、sICAM-1两者间关系及其在鼻息肉发展过程中的作用及鼻内应用激素治疗鼻息肉的可能机制.方法:选择19例未用药的鼻息肉患者(未用药组)、14例经鼻内应用激素(氟替卡松)治疗后的鼻息肉患者(激素治疗组)、12名正常人(正常对照组),取其鼻息肉或鼻黏膜组织及鼻分泌物,用ELISA法检测其鼻息肉或鼻黏膜组织匀浆中ICAM-1和鼻分泌物中sICAM-1的含量并作比较.结果:未用药组鼻息肉组织匀浆中ICAM-1的含量及鼻分泌物中sICAM-1的含量,较激素治疗组及正常对照组为高(均为P<0.01),而激素治疗组与正常对照组鼻黏膜组织匀浆中ICAM-1的含量和鼻分泌物中sICAM-1的含量比较差异无统计学意义(P>0.05).未用药组鼻息肉组织匀浆中ICAM-1及鼻分泌物sICAM-1含量呈线性相关(r=0.843,P=0.000).结论:鼻内应用激素的治疗效果可能与其抑制ICAM-1的表达有关.在未受药物干扰的情况下,鼻分泌物中 sICAM-1的含量也能很好的反映鼻息肉组织中ICAM-1的表达情况.  相似文献

目的 探讨黏附分子CD44异构体6(CD44v6)、细胞间黏附分子-1(ICAM-1)和细胞钙黏附素E(E-cadherin)在子宫内膜异位症(EMs)发生、发展中的作用.方法 通过免疫组化方法分别检测CD44v6、ICAM-1和E-cadherin在子宫内膜异位症患者在位内膜、异位内膜和正常子宫内膜中的表达.结果 EMs患者中异位内膜CD44v6表达明显高于在位内膜和正常内膜(P<0.05);异位内膜中ICAM-1表达高于在位内膜(P<0.05),且其在位内膜高于正常内膜(P<0.05);异位内膜及在位内膜中E-cadherin表达显著低于正常内膜(P<0.05).结论 子宫内膜异位症异位内膜、在位内膜中CD44v6、ICAM-1的高表达和E-cadherin的低表达在子宫内膜异位症的发生、发展中起重要作用.  相似文献

【目的】探讨CD68及胰岛素样生长因子-1(IGF-1)在鼻息肉中的表达及其相关关系以分析其在鼻息肉发病中的作用。【方法】采用免疫组化SP法分别检测30例鼻息肉、20例中鼻甲黏膜组织中CD68及IGF-1表达情况。【结果】①鼻息肉组织中CD68及IGF-1水平均明显高于中鼻甲黏膜组织(P均〈0.01)。CD68阳性细胞主要位于黏膜下组织间隙。IGF-1阳性细胞主要集中于鼻息肉的黏膜上皮细胞、囊性扩张的腺体细胞及炎性细胞中。②鼻息肉组织中CD68与IGF-1呈明显正相关关系.相关系数r=0.843,P〈0.01。【结论】鼻息肉中细胞增殖活跃。尤其是上皮细胞、腺体细胞和组织中浸润的炎性细胞较正常明显增加。细胞增殖活跃在鼻息肉的形成和复发中起重要作用,而炎症反应则为上皮细胞增生的主要原因。  相似文献

目的 探讨核转录因子-κB(NF-κB)活化对急性肺损伤(AL1)大鼠P-选择素、细胞间黏附分子-1(ICAM-1)表达的影响,并观察低分子肝素(LMWH)和阿司匹林(ASA)对AL1的保护作用,并探讨其机制。方法尾静脉注射内毒素制备AL1大鼠模型。60只大鼠按随机数字表法分为正常对照组、AL1组、LMWH组和ASA组,每组15只。观察NF-κB、ICAM-1和P-选择素在肺组织中的表达情况。结果 ALI组肺组织NF-κB活性显著增强;ICAM-1、P一-选择素明显表达于血管内皮细胞和支气管上皮细胞膜(P均〈0.05);LMWH组和AsA组NF-κB、ICAM-1、P-选择素活性降低,炎症反应和病理损伤明显减轻,ASA组治疗效果优于LMWH组(P均〈0.05)。结论NF-κB活化在AL1发病机制中起作用,NF-κB参与活化中性粒细胞、血管内皮细胞等多种炎症细胞,并且调控ICAM-1、P-选择素基因的表达进而造成肺组织损伤。LMWH通过改善微循环,间接抑制NF-κB活化,减少中性粒细胞及血小板的黏附进而减轻肺损伤。ASA通过抗氧化特性直接抑制NF-κB活化而减轻肺损伤。  相似文献

目的:探讨子痫前期患者细胞间黏附分子-1(ICAM-1)、P-选择素(P-选择素)、D-二聚体(D-dimer)的水平变化及相互关系。方法:选择随机选取年龄、孕周、孕次相匹配的子痫前期孕妇90例为观察组,同期正常晚期妊娠孕妇92例为对照组,采用免疫组织化学SABC法检测两组研究对象胎盘组织中ICAM-1、P-选择素的表达水平;采用酶联免疫吸附ELISA法检测两组对象血浆ICAM-1、P-选择素和D-二聚体的表达水平。结果:子痫前期患者胎盘组织ICAM-1、P-选择素的表达水平明显高于正常对照组(P0.05),在子痫前期组二者之间存在正相关性(r=0.523,P0.05);两组孕妇血浆均有ICAM-1、P-选择素和D-二聚体的表达,子痫前期患者血浆ICAM-1、P-选择素和D-二聚体表达水平明显高于正常对照组(P0.05),在子痫前期组三者之间相互呈明显正相关关系(分别为r=0.489,r=0.561和r=0.517,均P0.05)。结论:子痫前期患者ICAM-1、P-选择素和D-二聚体的高表达可能与子痫前期的发生发展有关,监测这些指标对判断病情及指导治疗具有重要价值。  相似文献

目的观察树突状细胞(DC)在鼻息肉组织中的分布特点,探讨DC在鼻息肉发病机制中的作用。方法选择手术摘除鼻息肉30例、正常下鼻甲黏膜10例,采用免疫组织化学方法检测鼻息肉组织中S-100蛋白表达情况,免疫荧光双标记(CD1a/CD40)及低照度荧光显微镜图像分析检测方法,观察DC在鼻息肉组织及正常下鼻甲黏膜中的分布情况,并初步探讨鼻息肉组织中DC与T淋巴细胞之间的关系。结果鼻息肉组织中S-100阳性细胞及CD1a/CD40双阳性细胞数量较下鼻甲黏膜明显增多,其差异有显著统计学意义(P〈0.01);鼻息肉组织与下鼻甲黏膜中DC的密度分别为(7125±2575)个/cm^2和(589±456)个/cm^2,两者的差异有显著统计学意义(P〈0.01)。此外,鼻息肉组织中DC主要分布在黏膜下层,有从外向内递减的趋势,并且DC表面CD40分子为阳性。结论鼻息肉组织中DC浸润显著,DC可通过CD40/CD40L共刺激因子与T细胞产生相互作用,在鼻息肉发病及疾病发展过程中发挥重要作用。进一步探讨DC在鼻息肉发病机制中的作用,将会拓宽鼻息肉的研究视野,为鼻息肉的防治提供新的干预靶点。  相似文献

目的 探讨黏附分子P-选择素、ICAM-1及树突状细胞(DC)在大鼠肝和肾缺血-再灌注损伤中作用,以及抗P-选择素单抗的抗黏附抑制及其防治效果。方法 分别建立肝和肾缺血-再灌流损伤大鼠模型,随机分为P-选择素单抗治疗组(n=20)和非治疗组(n=20),按不同再灌注时间(1,3,6和24h)再分为4组,每组5只,另设两组另设假手术组(n=5)作为对照。采用免疫组化LSAB法和免疫双染与荧光图像分析法,分别观察和比较各组大鼠肝、肾组织P-选择素、ICAM-1表达及DC分布变化。结果 (1)缺血-再灌注1h起,P-选择素即以肝窦内皮细胞或肾小管上皮细胞为主于肝和肾内广泛高表达,至24h仍维持一定水平;ICAM-1于缺血-再灌注6h起持续以肝窦和肾血管为主表达上调和明显增强。(2)CDla^ CD80^ DC白再灌注1h起以肝窦和肾小管间质为主分布数量逐渐增多,至24h达峰,并与大鼠肝、肾功能密切相关。(3)经抗P-选择素单抗处理后,大鼠肝、肾组织P-选择素和ICAM-1表达受到抑制,CDla^ CD80^ DC分布及数量减少,组织病理损伤及肝、肾功能也相应减轻和改善。结论 DC可能通过黏附分子尤其P-选择素早期介导的肝、肾组织内迁移聚集,参与启动了肝、肾缺血-再灌注时的免疫病理损伤过程,而抗P-选择素单抗对此具有抗黏附调抑和防治效果。  相似文献

目的检测人β-防御素(h BD)2,3,4在不同分型的慢性鼻-鼻窦炎(CRS)患者鼻黏膜和正常鼻黏膜上皮细胞中的表达,并明确各组间的差异,分析其在CRS的病因及疾病进展中的作用。方法收集2015年3至12月在山西医科大学附属汾阳医院耳鼻喉科的住院患者,分为3组:正常对照组15例(鼻中隔矫正术时取中鼻甲外侧壁黏膜),CRS不伴鼻息肉组30例(取钩突黏膜),CRS伴鼻息肉组30例(取息肉组织),采用免疫组化染色法分别检测各组织中h BD的蛋白表达。结果 (1)CRS各型及正常对照组鼻黏膜中h BD-2,3,4的蛋白均有表达,3组间h BD的蛋白表达差异有统计学意义(P<0.05)。CRS伴鼻息肉组h BD-2,4蛋白表达水平高于CRS不伴鼻息肉组,在正常鼻黏膜中表达最低。CRS伴鼻息肉组患者h BD-3蛋白表达水平高于正常组,CRS不伴鼻息肉组表达最低。(2)免疫组化染色显示,h BD-2,3,4主要在腺上皮、间质细胞、炎性细胞中表达。结论 (1)鼻黏膜上皮细胞中h BD-2,3,4呈诱导性表达,CRS不伴鼻息肉组h BD-3表达最低,可能与激素抑制其表达有关。(2)免疫组化染色显示h BD-2,3,4蛋白主要在腺上皮、间质细胞及炎性细胞中广泛表达,这主要与细胞分泌防御素参与鼻黏膜免疫有关。  相似文献

目的探讨慢性鼻-鼻窦炎(CRS)鼻息肉内镜手术前后鼻黏膜组织中血管内皮生长因子(VEGF)和黏蛋白MUC5AC的表达情况。方法选择2012年1月-2015年1月台州市中心医院CRS鼻息肉内镜手术患者75例作为鼻息肉组和鼻骨骨折或鼻出血患者75例作为对照组,鼻息肉患者取术前鼻息肉标本、术后6周上颌窦口黏膜标本、对照组取下鼻甲上缘黏膜标本进行HE染色观察嗜酸性粒细胞计数、免疫组化染色测定VEGF和黏蛋白MUC5AC的表达情况。结果鼻息肉术前组和鼻息肉术后组标本中嗜酸性粒细胞均高于对照组(P0.05),鼻息肉术后组鼻黏膜嗜酸性粒细胞低于鼻息肉术前组(P0.05)。鼻息肉术前组和鼻息肉术后组标本中VEGF和黏蛋白MUC5AC阳性表达面积百分率均高于对照组(P0.05),鼻息肉术后组鼻黏膜VEGF和黏蛋白MUC5AC阳性表达面积百分率低于鼻息肉术前组(P0.05)。结论嗜酸性粒细胞计数和VEGF、黏蛋白MUC5AC表达水平在CRS鼻息肉内镜术前鼻黏膜组织中较高,术后6周时明显降低,VEGF和黏蛋白MUC5AC可能参与术后鼻黏膜的修复。  相似文献

目的研究黏附分子CD44和细胞间黏附分子1(ICAM-1)在肺炎支原体(MP)感染的BALB/c小鼠模型的表达,阐明其与MP感染的关系。方法采用MP鼻腔滴入法建立BALB/c小鼠MP模型,观察MP小鼠肺部的病理变化。采用ELISA法测定5,8,15,20和30天MP感染小鼠血清和肺泡灌洗液中黏附分子CD44和ICAM-1的表达水平。结果与模型组比较,MP感染BALB/c小鼠后肺部炎症明显,MP感染BALB/c小鼠后其血清和肺泡灌洗液中黏附分子CD44和ICAM-1的表达明显上调(t_(5天血清CD44)=53.64ng/L,t_(5天BALF CD44)=144.3ng/L;t_(5天血清ICAM-1)=73.72ng/L,t_(5天BALF ICAM-1)=165.06ng/L,均P值0.000;t_(8天血清CD44)=40.86ng/L,t_(8天BALF CD44)=21.31ng/L;t_(8天血清ICAM-1)=30.57ng/L,t_(8天BALF ICAM-1)=19.61ng/L,均P值0.000)。结论 MP感染使小鼠黏附分子CD44和ICAM-1的表达水平升高,用替加环素治疗MP小鼠后CD44和ICAM-1表达下调,CD44和ICAM-1表达上调可能与MP肺炎的严重程度有关。  相似文献

Tissue eosinophilia is a characteristic feature of a number of inflammatory diseases including asthma and nasal polyposis. Eosinophil migration into tissues is controlled in part by interactions between eosinophil adhesion receptors and counter-structures on the vascular endothelium. To determine the receptors used by eosinophils to adhere to vascular endothelium in allergic inflammation we have adapted the Stamper-Woodruff frozen section assay (FSA) to study eosinophil adhesion to nasal polyp endothelium. Immunohistology indicated that intercellular adhesion molecule 1 (ICAM-1), E-selectin and P-selectin were well expressed by nasal polyp endothelium, whereas expression of vascular cell adhesion molecule 1 (VCAM-1) was weak or absent. Unstimulated human peripheral blood eosinophils adhered specifically to nasal polyp endothelium. Adherence was temperature and divalent cation- dependent and saturable at cell densities > 5 x 10(6) cells/ml. Eosinophil adhesion was almost completely inhibited by a monoclonal antibody (mAb) against P-selectin and by a chimeric molecule consisting of the Fc portion of human IgG and the lectin binding domain of P- selectin, which binds to the P-selectin ligand on leucocytes. Anti-Mac- 1 mAb partially inhibited eosinophil adhesion whereas mAb against E- selectin, L-selectin, ICAM-1, VCAM-1, very late activation antigen 4, and lymphocyte function-associated antigen 1 had no effect. P-selectin is stored in intracellular granules within the endothelial cell and in vitro is only transiently expressed. To determine if P-selectin was expressed on the membrane of the nasal polyp endothelium we compared P- selectin expression in normal skin and nasal polyps after acetone fixation, which permeabilizes cells, and paraformaldehyde, which only allows staining of membrane expressed receptors. In the skin, good expression was seen with acetone fixation but no expression was seen after paraformaldehyde treatment, whereas in nasal polyps, similar expression was observed with both fixatives. In addition immunofluorescence with confocal microscopy demonstrated lumenal staining of nasal polyp endothelium indicating that P-selectin was located on the surface of endothelial cells while in skin only an intracellular granular distribution was apparent. Lastly, whereas eosinophils bound consistently to nasal polyp endothelium, no binding was observed to blood vessels in normal skin further supporting the idea that eosinophils were binding to membrane expressed and not intracellular P-selectin. The importance of P-selectin in eosinophil adhesion to nasal polyp endothelium suggests that P-selectin antagonists may be effective at inhibiting eosinophil accumulation at sites of allergic inflammation.  相似文献

目的研究Survivin在鼻息肉组织中表达及其与Ki67表达的相关性,探讨Survivin在鼻息肉发病过程中的作用。方法收集28例鼻息肉组织(鼻息肉组)和12例正常下鼻甲组织(对照组),用免疫组化法检测鼻息肉组与对照组组织中Survivin与Ki67的表达,观察比较两组之间的表达差异。结果免疫组织化学染色显示Survivin、Ki67免疫阳性细胞数及着色强度均明显高于对照组。图像分析显示,鼻息肉组Survivin、Ki67的积分光密度(IOD,103/HP)分别为50.21±6.32、18.25±2.71,与对照组5.67±0.58、3.80±0.37相比,有显著性差异(P0.05);Spearman等级相关分析显示,鼻息肉中Survivin表达与Ki67表达密切相关(r=0.650,P0.01)。结论鼻息肉组织中Survivin、Ki67表达均上调,且二者有协同表达关系,鼻息肉可能是通过凋亡抑制基因Survivin使炎性细胞凋亡受到抑制,从而促进炎性细胞浸润生长,导致一种慢性炎症反应,并造成局部组织增生。  相似文献

Adhesion molecules are known to be an important part of leukocyte migration and extravasation in both homeostatic and inflammatory conditions. Intracellular adhesion molecule-1 (ICAM-1 or CD54) is constitutively expressed on endothelial cells and is up-regulated during acute and chronic inflammation. We investigated the efficacy and consequences of interfering with CD54 after administration of an antisense oligonucleotide to ICAM-1 (CD54) in the transgenic HLA-B27/beta2 microglobulin rat model. One hundred percent of the HLA-B27 transgene + animals will spontaneously develop chronic inflammation (some more severely than others) in the gastric mucosa, cecum, and colon. We carried out two studies, i.p. injection and rectal administration of antisense. Following i.p. and rectal treatment, there were significant decreases in colonic mucosal wall thickness, histologic inflammation, CD54 expression in the colon and peripheral blood, and the percentage of colon weight per end body weight. Furthermore, decreased expression of CD49d, CD18, and tumor necrosis factor-alpha was observed in antisense treated rats. Therefore, the HLA-B27 transgenic model of spontaneous and chronic inflammatory bowel disease, which has increased expression of adhesion molecules, responds to both routes of administration of ICAM-1 antisense oligonucleotides. These studies support the regulatory role of adhesion molecules in chronic intestinal inflammation, the need for an understanding of how the route of drug delivery can alter the dose and area affected, and finally the role of antisense oligonucleotides as a therapeutic modality in chronic spontaneous inflammatory bowel diseases.  相似文献

目的探讨人鼻息肉组织标本白细胞介素(interleukin,IL)-17表达水平及临床意义。方法48例鼻息肉患者术后鼻息肉组织标本(鼻息肉组)与10例正常下鼻甲组织(对照组),采用反转录一聚合酶链反应方法检测2组IL-17mRNA的表达水平。结果鼻息肉组IL-17mRNA光密度值(1.82±0.47)明显高于对照组(0.65±0.08)(P〈0.01);鼻息肉组织中IL-17mRNA表达水平在性别、年龄、息肉初发或复发上差异无统计学意义(P〉0.05)。结论IL-17mRNA在鼻息肉组织中呈高表达,与鼻息肉的发生、发展有关。  相似文献

OBJECTIVES: Antimicrobial agents are important risk factors for infusion phlebitis, but the risk varies between different antibiotics. Erythromycin and dicloxacillin are known to induce phlebitis frequently, as well as to exert toxic effects on cultured endothelial cells. The pathogenesis of infusion phlebitis is unclear, but chemical toxicity is thought to lead to inflammation and subsequent thrombosis. In the present study, endothelial cells were exposed to antibiotics at the range of concentrations used for intravenous administration, followed by analysis of pro-inflammatory and pro-coagulant surface molecules. METHODS: Primary human umbilical vein endothelial cells (HUVEC) and the endothelial hybrid cell line EaHy926 were exposed to dicloxacillin, erythromycin, benzylpenicillin and cefuroxime (all at 6250 mg/L) for 60 min, followed by washing. After 5 or 24 h additional incubation, cells were analysed for E-selectin (CD62E), tissue factor (TF) or intercellular adhesion molecule 1 (ICAM-1, CD54) density by flow cytometry. RESULTS: Despite constitutive expression of ICAM-1 (34%) in HUVEC, 6250 mg/L of dicloxacillin or erythromycin significantly increased the number of cells with ICAM-1 expression by 37% and 30%, respectively. In contrast, cefuroxime and benzylpenicillin did not up-regulate ICAM-1 above background levels. A similar pattern was seen with the endothelial cell line EaHy926. The E-selectin and TF density were not affected by the antibiotics examined. CONCLUSIONS: The results of this study support the theory that endothelial cells that are affected by high concentrations of antibiotics may initiate an inflammatory response through expression of ICAM-1. This is a novel finding in the pathogenesis of infusion phlebitis.  相似文献

This study was aimed at understanding the histopathologic changes that occur in the nasal mucosa of patients with perennial allergic rhinitis after high-intensity focused ultrasound (HIFU) treatment. Biopsy specimens of the inferior turbinate mucosa were taken from 11 PAR patients before, immediately after and 1 y after HIFU treatment. Morphometric analysis revealed that the density of eosinophils and other inflammatory cells increased immediately after treatment and then were decreased significantly 1 y post-treatment. Submucosal glands were swollen and venous sinusoids were dilated, but there was no statistically significant change in their density, immediately after treatment. However, both glands and venous sinusoids significantly decreased in number 1 y after HIFU treatment. The ciliated epithelium or basement membrane of the nasal mucosa was well preserved at all stages. In conclusion, HIFU is a tolerable and effective treatment to reduce inflammation of the inferior turbinate mucosa in patients with perennial allergic rhinitis.  相似文献

目的:探讨细胞间黏附分子-1(intercellular adhesion molecule-1,ICAM-1)在家兔急性鼻窦炎上颌窦黏膜中的表达及意义。方法:健康成年新西兰白兔24只,随机分成鼻腔置入Merocel!高分子膨胀海绵+鼻腔注入肺炎链球菌组(A组,8只)、单纯鼻腔置入Merocel!高分子膨胀海绵组(B组,8只)及空白对照组(C组,8只)。10d后将动物处死,完整取出实验侧上颌窦,脱钙3周后采用免疫组化检测上颌窦黏膜中ICAM-1的表达。结果:ICAM-1在A组家兔上颌窦黏膜上皮细胞、腺上皮细胞、血管内皮细胞及固有层炎性细胞中大量表达;B组上颌窦黏膜ICAM-1的表达量也明显增加,但阳性表达强度弱于A组;C组上颌窦黏膜上皮细胞、腺上皮细胞极少量表达ICAM-1,3组间比较差异均有统计学意义(P<0.05)。结论:ICAM-1在实验性急性鼻窦炎鼻窦黏膜中大量表达,在急性鼻窦炎炎症进程中起重要作用。  相似文献

目的:观察红霉素对上皮细胞凋亡相关基因Bax和Bcl-2表达的影响,进一步探讨上皮细胞凋亡在鼻息肉组织形成中的意义。方法:将30例鼻息肉组织及6例下鼻甲组织的上皮细胞均分成两组,其中一组加红霉素(红霉素组),进行原代上皮细胞的培养,另一组加蒸馏水(对照组),以免疫组化法检测细胞凋亡基因Bax、抑凋亡基因Bcl-2。结果:鼻息肉上皮细胞Bcl-2和Bax表达均显著强于下鼻甲(均P〈0.01);培养3d后鼻息肉上皮细胞红霉素组Bax表达显著强于对照组(P〈0.05),Bcl-2表达虽强于对照组,但差异无显著性;下鼻甲上皮细胞红霉素组与对照组的Bcl-2与Bax阳性表达差异均无显著性(P〉0.05);红霉素干预5d后鼻息肉上皮细胞显示明显的促凋亡趋势(P〈0.05)。结论:红霉素能显著提高Bax蛋白的表达,有显著的促鼻息肉上皮细胞凋亡的作用。  相似文献
