Corni fructus has been used as a tonic, analgesic, and diuretic in Korean herbal medicine. The present investigation was undertaken to evaluate the antioxidative effect of corni fructus and its capacity to protect cells against oxidative damage. The radical scavenging activity of corni fructus extracts was measured using 1,1-diphenyl-2-picrylhydrazyl (DPPH), and the antioxidant activity was determined by measuring the peroxide value in a linoleic acid emulsion system. In addition, human umbilical vein endothelial cells (HUVECs) were treated with corni fructus extracts and incubated with H(2)O(2) to investigate protection against apoptosis induction. Both ethanol and water extracts of corni fructus produced higher DPPH radical scavenging activity than hexane, chloroform, and ethyl acetate extracts. Strong antioxidative activities of both water and ethanol extracts were observed in an emulsion system containing linoleic acid and phosphate buffer. The incubation of HUVECs with the addition of ethanol extract significantly decreased H(2)O(2)-initiated damage of endothelial cells, but the water extract did not. The pretreatment with ethanol extract, but not with water extract, significantly decreased apoptotic damage of the H(2)O(2)-treated HUVECs and kept the morphological normality. This study demonstrates that corni fructus is a potent antioxidant substance, and suggests that further investigation is needed to characterize the difference between extract types and to identify its antioxidant compounds.  相似文献

Antioxidant properties of selected tropical wild edible mushrooms   总被引:1,自引:0,他引:1  
Selected species of wild edible mushrooms were obtained from the interior areas of East Malaysia to determine the total phenolics and antioxidant properties, including free radical scavenging, reducing power and metal chelating activities. The in vitro antioxidant activities of petroleum ether (PE) and methanolic extracts of the edible wild mushrooms were comparable to the cultivated oyster mushroom. The radical scavenging activity was the highest in PE extract of Pleurotus porrigens (angel's wings) (85%), while methanolic extract of Hygrocybe conica (witch's hat mushroom) exhibited the highest (94%) chelating effect at 20 mg/ml. PE extracts were more effective than methanolic extract in scavenging ability on DPPH radicals, whereas methanolic extracts were more effective in reducing power and chelating ability on ferrous ions as evidenced by their lower EC50 values. Principal component analysis (PCA) indicated phenolic group was the primary factor contributing to the metal chelating ability for PE extract although phenolic was better correlated with reducing power in methanolic extracts.  相似文献

Waste obtained from six genotypes of tomato was subjected to evaluation as potential sources of hydrophilic antioxidants on the basis of content of total phenolics (TPh; from 11.7 to 18.6 mg/g), total flavonoids (TFl; from 7.62 to 12.1 mg/g) and ascorbic acid (AA; from 0.51 to 1.89 mg/g), and antioxidant activities. The antioxidant activities of tomato waste extracts were evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, ferrous ion chelating activity (CA) and reducing power (RP) assay. The O(2) tomato waste extract was found to have the strongest antioxidant activity in scavenging DPPH radicals (effective concentration, EC(50) = 0.18 mg/ml). The Saint Pierre tomato waste extract showed the best performance in RP assay (EC(50) = 1.57 mg/ml). The Rutgers tomato waste extract exhibited a strongest CA (EC(50) = 1.49 mg/ml). The linear correlation analysis produced moderate-to-high correlation coefficients between the antioxidant activities and TPh, TFl and AA.  相似文献

Antioxidant properties of brown seaweed (Sargassum siliquastrum) extracts were evaluated using various antioxidant measurements, i.e., inhibitory effect on thiobarbituric acid-reactive substances (TBARS), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, metal chelating effect, reducing power effect, and total phenolic compounds. When the extraction solvents n-hexane, chloroform, ethanol, and water were compared, the water extract showed the highest yield in extracted mass. Total phenolic compounds were the highest in the ethanol extract, with 127.4 mg/g. The TBARS inhibition of chloroform and ethanol extracts at 10 mg/mL was 90.9% and 80.9%, respectively. DPPH radical scavenging capacity was more than 90% in all extracts at 1 mg/mL. The chloroform extract exhibited the highest metal ion chelating ability of 69.6% at 10 mg/mL. The reducing power was found to be the highest in the ethanol extract at 10 mg/mL, showing an effect similar to ascorbic acid. Thus, the ethanol extract of S. siliquastrum has potential as a natural antioxidant.  相似文献

The aim of the study was to investigate antioxidant activities of Ganoderma lucidum and Funalia trogii. Ethanol and water crude extracts from G. lucidum and F. trogii were investigated for their antioxidant capacity in some different assays, namely, the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, metal chelating activity against ferrous ions, and plasma lipid peroxidation inhibitory. In addition, the amounts of total phenol, ascorbic acid, β-carotene, and lycopene components in the extracts were determined. Among the four mushroom extracts, G. lucidum water extract and G. lucidum ethanol extract showed the highest scavenging activity against DPPH radicals (50% inhibitory concentration?=?0.055?±?0.001?mg/mL). Total phenol was the major antioxidant component found in the mushroom extracts. These results showed that G. lucidum may be used in pharmaceutical applications because of its effective antioxidant properties.  相似文献

The aim of this study was to investigate the mechanism of antioxidant and prooxidant effects of epigallocatechin-3-gallate (EGCG) using chemiluminescence analysis. Results showed that EGCG scavenged superoxide radical and H2O22 in a dose dependent manner. EGCG scavenged 50% of superoxide radical at 0.31 mM and scavenged 50% of H2O22 at 0.09 mM, demonstrating that EGCG has a stronger reactive oxygen species (ROS) scavenging activity than ascorbic acid. Effects of EGCG on free radical-induced DNA oxidative damage were investigated. EGCG had protective effect on DNA at low concentrations (2-30 mM), but it enhanced the DNA oxidative damage at higher concentrations (>60 mM), exhibiting a prooxidant effect on DNA. EGCG showed a greater reducing power on iron ions, reducing Fe3+ to Fe2+, which accelerates the generation of hydroxyl radical from the Fenton reaction. At low concentrations, ROS scavenging activity of EGCG might predominate over its reducing power and lead to its protective effect on DNA. However, relatively higher reducing power of EGCG at higher concentrations may gradually predominate over its ROS scavenging activity and result in the prooxidant effect of EGCG on DNA.  相似文献

Herbs and spices not only variety and racy flavour to Korean foods, they also are the richest source for antioxidant power. The present study evaluates the radical scavenging-linked antioxidant activities of hot water extracts from commonly used herbs and spices in Korea. 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical and superoxide anion scavenging activities of bay extract were 39.5% and 22.1%, respectively. The hydroxyl radical scavenging activity was in order of dill (50.0%) > bay (31.3%) > garlic (27.9%) > white pepper and black pepper (15.1–15.3%) > onion (10.1%) extracts. Bay extract had the highest total phenolic content (17.86 μg CE/g). High correlation coefficients were found between the total phenol content and DPPH radical scavenging activity (R = 0.9162). These results indicate that herbs and spices had high antioxidant activity that is partly due to the phenolic compounds and provide basic data for further development of processed food products.  相似文献

In this study, antioxidant and free radical scavenging activities of the natural antioxidative compound, pyrogallol-phloroglucinol-6,6''-bieckol (PPB) isolated from brown algae, Ecklonia cava was assessed in vitro by measuring the radical scavenging activities (DPPH, alkyl, hydroxyl, and superoxide) using electron spin resonance (ESR) spectrometry, intracellular reactive oxygen species (ROS) scavenging activity, and DNA damage assay. According to the results of these experiments, the scavenging activity PPB against difference radicals was in the following order: DPPH, alkyl, hydroxyl, and superoxide radicals (IC50; 0.90, 2.54, 62.93 and 109.05 µM). The antioxidant activities of PPB were higher than that of the commercial antioxidant, ascorbic acid. Furthermore, PPB effectively inhibited DNA damage induced by H2O2. These results suggest that the natural antioxidative compound, PPB, can be used by the natural food industry.  相似文献

Water and ethanol crude extracts from black pepper (Piper nigrum) were investigated for their antioxidant and radical scavenging activities in six different assay, namely, total antioxidant activity, reducing power, 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) free radical scavenging, superoxide anion radical scavenging, hydrogen peroxide scavenging, and metal chelating activities. Both water extract (WEBP) and ethanol extract (EEBP) of black pepper exhibited strong total antioxidant activity. The 75 microg/ml concentration of WEBP and EEBP showed 95.5% and 93.3% inhibition on peroxidation of linoleic acid emulsion, respectively. On the other hand, at the same concentration, standard antioxidants such as butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT) and alpha-tocopherol exhibited 92.1%, 95.0%, and 70.4% inhibition on peroxidation of linoleic acid emulsion, respectively. Also, total phenolic content in both WEBP and EEBP were determined as gallic acid equivalents. The total phenolics content of water and ethanol extracts were determined by the Folin-Ciocalteu procedure and 54.3 and 42.8 microg gallic acid equivalent of phenols was detected in 1 mg WEBP and EEBP.  相似文献

This study aims to evaluate the antioxidant activity (total antioxidant and free radical scavenging activities) of seaweeds commercially available in the Malaysian supermarket. Four types of seaweeds namely Nori (Porphyra sp.), Kumbu (Laminaria sp.), Wakame (Undaria sp.) and Hijiki (Hijikia sp.) were used in the study. The extracts were prepared with water and ethanol, respectively. The b-carotene bleaching and 1,1-diphenyl-2-picrylhydrazyl (DPPH) assays were used to determine antioxidant properties of seaweeds by measuring the decrease in absorbance at 470 and 517 nm. In water extract, Kumbu showed the highest total antioxidant activity of 63% compared with other samples. Kumbu, Nori and Hijiki exhibited higher radical scavenging activity than Wakame when extracted with water. Wakame exhibited the highest antioxidant and free radical scavenging activities in ethanolic extract with 58% and EC50 = 0.42 mg/ml respectively. The results of ANOVA analysis show significant differences (p<0.05) in the means of total antioxidant and free radical scavenging activities of the seaweeds. The results showed that processed commercial seaweeds exhibited varying degrees of antioxidant properties.  相似文献

Cyperus rotundus (Family Cyperaceae) is used both as a functional food and as a drug. In this study, the antioxidative potential of a hydroalcoholic extract of C. rotundus (CRE) was evaluated by various antioxidant assays, including antioxidant capacity by the phosphomolybdenum method, total antioxidant activity in linoleic acid emulsion systems, 1,1-diphenyl-2-picrylhydrazyl (DPPH), superoxide, hydroxyl radicals, and nitric oxide (NO) scavenging. We further evaluated the reducing potential of the extract as well as Fe(2+)/ascorbate-induced lipid peroxidation in rat liver homogenate. These various antioxidant activities were compared to standard antioxidants such as butylated hydroxytoluene, tocopherol, L-ascorbic acid, and catechin. Total phenolic and flavonoid content of CRE was also determined by a colorimetric method. The extract exhibited high reduction capability and powerful free radical scavenging, especially against DPPH and superoxide anions as well as a moderate effect on NO. CRE also showed inhibited lipid peroxidation in rat liver homogenate induced by Fe(2+)/ascorbate and prevented deoxyribose degradation in both non-site-specific and site-specific assays showing the hydroxyl radical scavenging and metal chelating activity of the hydroalcoholic extract. Moreover, the peroxidation inhibiting activity of CRE was demonstrated in the linoleic acid emulsion system. These results clearly established the antioxidative potency of C. rotundus, which may account for some of the medical claims attributed to this plant.  相似文献

Oxidative stress is considered an important factor that promotes cell death in response to a variety of pathophysiological conditions. This study investigated the antioxidant properties of allicin, the principle ingredient of garlic, on preventing oxidative stress-induced injury. The antioxidant capacities of allicin were measured by using 1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay and hydrogen peroxide (H2O2)-induced cell damage on H9c2 cardiomyoblasts. Allicin (0.3–10?μM) pre-incubation could concentration-dependently attenuate the intracellular reactive oxygen species (ROS) increase induced by H2O2 on H9c2 cells. It could also protect H9c2 cells against H2O2-induced cell damage. However, the DPPH free radical scavenging activity of allicin was shown to be low. Therefore, it is believed that the protective effect of allicin on H9c2 cells could inhibit intracellular ROS production instead of scavenging extracellular H2O2 or free radicals. For the observed protective effect on H9c2 cells, allicin might also be effective in reducing free radical-induced myocardial cell death in ischemic condition.  相似文献

A strong radical-scavenging activity against a stable radical compound, 1,1-diphenyl-2-picrylhydrazyl (DPPH) was found in the hot water extract of Japanese rice bran. When the extract was treated with ethanol, a dominant radical-scavenging activity was observed in the ethanol-soluble (ES) fraction in a dose-dependent manner, but a weak radical-scavenging activity was detected in the ethanol-precipitable (EP) fraction. Their activities were proportional to the amounts of phenolic substances in each fraction. The phenolic substances in the ES fraction were efficiently separated by Amberlite XAD column chromatography and high performance liquid chromatography using an ODS column. The four major phenolic acids (ferulic, para-coumaric, para-hydroxybenzoic and vanillic acids) and four minor phenolic acids (caffeic, gentisic, protocatechuic and syringic acids) were detected in the HPLC system. Among these phenolic acids, protocatechuic, caffeic, ferulic and gentisic acids showed relatively strong radical scavenging activities (EC50: 8, 9, 29 and 75 microM, respectively) compared with the control antioxidants such as ascorbic acid and alpha-tocopherol (EC50: 93 and 134 microM). Para-coumaric, syringic and vanillic acids exhibited weak but significant radical-scavenging activities (EC50: 780, 2640 and 3250 microM). However, para-hydroxybenzoic acid did not show any significant effects even at 5 mM. Furthermore, a simulated mixture combined with these phenolic acids in comparable amounts in the ES fraction showed slightly weak radical-scavenging activity compared with that of rice bran extract. However, all the phenolic acids detected in the ES fraction did not show significant antioxidant activities against hydroperoxide generation in lipid peroxidation compared with that of a typical antioxidant such as ascorbic acid, which was estimated by the alminum chloride method. These results suggest that Japanese rice bran has a potent radical-scavenging activity against DPPH radical and this activity is associated with some phenolic acids in the ES fraction. The significance of this finding is discussed from the viewpoint of the protective role of rice bran against oxygen radical-induced chronic diseases.  相似文献

目的 : 对 6种芦荟提取物的抗氧化活性进行比较研究 ,进一步纯化并探讨抗氧化物对动物红细胞的保护功能。方法 : 采用分别产生氧自由基 (O-2· )和羟自由基 (·OH)的化学模型体系研究清除自由基的活性 ,进一步利用硅胶柱和薄层层析法对龙山芦荟 (Aloe berebihoria)的提取物进行纯化 ,并用反相高压液相色谱法分析其纯度 ,采用体外红细胞体系研究抗氧化物对动物细胞的保护功能。结果 : 龙山芦荟 (Aloe berebihoria)提取物的清除两种自由基的能力最强 ,清除率分别为 3 2 .1 9%和 2 6 .0 5 % ,经纯化得到组分 A(比移值 Rf为 0 .497) ,高压液相色谱分析为单一组分 ,保留时间 (Rt)为 1 5 .95 6 min,醋酸镁甲醇显色反应表明该物质为具有酚羟基的蒽醌类物质。A组分对由活性氧引起的红细胞溶血有较高的抑制作用 ,半抑制量 (IC50 )为 0 .85 6 g/L。结论 : 龙山芦荟提取物的组分 A对 O-2· 和· OH具有强烈的清除作用 ,从而抑制红细胞膜脂质过氧化 ,保护红细胞膜的生理功能  相似文献

The aims of this study are to determine and compare the antioxidant levels and activities (i.e. primary and secondary) between selected seeds of Malaysian tropical fruits - guava (Psidium guajava), mango (Mangifera indica L.) and papaya (Carica papaya L.). Seeds are among byproducts from the processing of fruitsbased products. Instead of discarding seeds as waste, seeds with high potential as antioxidants could be utilised for commercial purposes. Accordingly, the selected seeds of Malaysian tropical fruits were tested in this study for total phenolic content (TPC), free radical scavenging activity by 1, 1- diphenyl-2-picrylhydrazyl (DPPH) assay and metal ion chelating effect by ferrous ion chelating (FIC) assay. Extraction of antioxidant compounds from sample was done with 70% ethanol. TPCs of the seeds were expressed as gallic acid equivalents (GAE) in mg per 100 g fresh seed weight. TPC assay showed that mango seeds had the highest TPC (i.e. 32 ± 0.001 mg GAE) followed by guava seeds (i.e. 20 ± 0.001 mg GAE) and papaya seeds (8 ± 0.003 mg GAE). For DPPH assay, IC50 data showed that mango seed extract scavenged 50% DPPH radicals at the lowest concentration (0.11 ± 0.01 mg/mL) followed by the positive control BHA (0.13 ± 0.01 mg/mL), guava seed extract (0.26 ± 0.01 mg/mL) and papaya seed extract (0.34 ± 0.01 mg/mL). Interestingly, all seed extracts showed higher free radical scavenging activities than BHA after sample concentration of 0.60 mg/mL. However, FIC assay indicated that metal ion chelating effects of all seed extracts were weaker than BHA suggesting that the fruit seeds are not sources of good metal ion chelators. Overall, present results suggest that TPC of the seeds show strong negative correlation with their primary antioxidant activity (r= -0.985, R2= 0.970), and not all compounds in extracts which could scavenge DPPH radicals are good metal ion chelators. Mango seeds relatively showed the highest antioxidant level and primary antioxidant activity followed by guava seeds and papaya seeds.  相似文献

The phenolic contents of methanolic extracts of emblica (Phyllanthus emblica L.) fruit from six regions in China were measured in this work. The antioxidant activities of these extracts were also evaluated. Total phenolic content was ranged from 81.5 to 120.9 mg gallic acid equivalents (GAE)/g and the flavonoid content was varied from 20.3 to 38.7 mg quecetin equivalents (QE)/g, while proanthocyanidin content was ranged from 3.7 to 18.7 mg catechin equivalents (CE)/g. Among all the methanolic extracts analyzed, the Huizhou sample exhibited a significantly higher phenolic content than other samples (P<0.05). The antioxidant activities were evaluated by in vitro experiments using scavenging assays of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, hydroxyl radicals, and superoxide anion radicals, chelating ability of ferrous ion, reducing power, and inhibition capability of Fe (II)-induced lipid peroxidation, respectively. The Huizhou sample was found to have the strongest antioxidant activities in scavenging DPPH radicals, superoxide anion radicals, and had the highest reducing power, while the Chuxiong sample showed the best performance in chelating iron and inhibiting lipid peroxidation. Furthermore, the Chuxiong sample exhibited a stronger inhibition activity of the hydroxyl radicals compared with other samples. The high correlation coefficient was existed between the phenolic content and superoxide anion radical scavenging activity, but no significant correlation was found between the former and hydroxyl radical scavenging activity. Methanolic extracts of emblica fruit from some selected regions exhibited stronger antioxidant activities compared to those of the commercial compounds (quercetin and BHA). This study investigated the antioxidant activity of methanol (MeOH) and water extracts from roots of Cirsium japonicum in vitro. MeOH extract showed a stronger free radical scavenging activity than water extract. However, both of extracts showed a concentration dependent hydroxyl radical scavenging activity, reducing power and metal chelating ability. MeOH extract had greater phenolic and flavonoid contents than water extract. The antidiabetic activity of these two extracts was evaluated by the α-glucosidase inhibition assay. The water extract showed a considerable α-glucosidase inhibitory activity. To our knowledge, this may be the first time to report the antioxidant and antidiabetic activities in Cirsium japonicum roots.  相似文献

Antioxidant activity was studied for anthocyanins extracted from purple black rice (PBR) by a 3% aqueous trifluoroacetic acid solution (TFA), as well as for anthocyanins extracted from blueberry (Bluetta, high bush type). Capillary zone electrophoresis revealed that the PBR extract contained almost exclusively a single anthocyanin, which was identified as cyanidin 3-O-beta-D-glucoside (Cy 3-Glc) after purification by polyvinylpyrrolidone column chromatography. In contrast, 11 anthocyanins were identified in the blueberry extract. PBR extract showed slightly weaker superoxide scavenging and crocin bleaching activities than blueberry extract did. Both PBR and blueberry extracts, however, showed 10 to 25 times stronger activity than the same concentration of Trolox used as a reference antioxidant. It was further noted that the purified Cy 3-Glc from PBR extract retained approximately 74% of the antioxidant activity (both crocin bleaching and superoxide scavenging) observed in the original TFA extract. The hydroxyl radical scavenging activity of both extracts was several times weaker than that of the same concentration of Trolox, although the PBR extract showed approximately two times stronger activity than blueberry extract did. The hydroxyl radical scavenging activity of the purified Cy 3-Glc from PBR, however, decreased to approximately 20% of that of the original PBR extract. These results indicate that the anthocyanin Cy 3-Glc contributes to the antioxidant activity of PBR through its strong superoxide radical but not hydroxyl radical scavenging activity.  相似文献
