目的 研究白术莪术提取物对四氯化碳(CCl4)诱导的小鼠急性肝损伤的影响.方法 给予小鼠白术莪术提取物灌胃,连续7d,腹腔注射CCl4-花生油溶液造模急性肝损伤,16 h后测定血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)含量、肝组织中超氧化物歧化酶(SOD)活性及其肝脏、脾脏指数.结果 白术莪术提取物能显著降低急性肝损伤小鼠血清中ALT、AST含量及提高肝组织中SOD活性,减小肝脏指数(P<0.01).结论 白术莪术提取物对CCl4诱导的小鼠急性肝损伤具有很好的保护作用.  相似文献

目的研究诱导HO-1高表达对大鼠急性肝损伤的保护作用及其机制探讨。方法随机将大鼠分成4组,即正常对照组、四氯化碳染毒组(CCl4)、血晶素(hemin)组、hemin+CCl4组。采用western blot法测定HO-1蛋白的诱导表达情况:测定各组大鼠血清ALT、AST水平,肝组织MDA浓度和SOD活性以及Caspase-3活性和TNF-α水平变化;HE染色观察肝组织病理形态学改变。结果CCl4成功诱导了大鼠急性肝损伤,表现为染毒24 h后血清ALT、AST水平以及肝组织MDA浓度、Caspase-3活性和TNF-α水平均显著升高,SOD活性下降,和正常对照组相比差异有统计学意义(P〈0.01);病理学检测结果显示肝脏有严重损伤,大量肝细胞发生凋亡。给予hemin预处理能显著诱导大鼠肝脏HO-1的表达,并对肝脏产生明显的保护作用,表现为大鼠血清ALT、AST水平和MDA浓度明显降低,SOD活性升高,组织TNF-α水平和Caspase-3活性明显低于CCl4组;此外,hemin预处理亦使染毒大鼠的病理学指标得到明显改善。结论HO-1诱导表达对急性肝损伤大鼠产生明显的保护作用,提示HO-1在防治急性肝损伤的病理生理过程中占有重要地位;HO-1的保护机制可能与其减轻脂质过氧化反应,抑制Caspase-3活性和降低TNF-α水平有关。  相似文献

目的观察石榴多酚对小鼠肝损伤的防护作用。方法用四氯化碳(CCl4)致小鼠急性肝损伤模型,观察石榴多酚(200、400、600mg/kg)对小鼠血清丙氨酸转氨酶(ALT)、天门冬氨酸转氨酶(AST)活性和肝脏组织超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量的影响。结果石榴多酚能明显降低急性肝损伤小鼠血清ALT、AST水平;并可使肝脏中MDA含量降低,SOD活性升高。各实验组小鼠的肝体比值均低于肝损伤模型组。结论石榴多酚对肝损伤具有保护作用,其作用可能与抗氧化有关。  相似文献

[目的]观察茵陈紫金汤对四氯化碳(CCl4)诱导的小鼠急性肝损伤保护作用.[方法]以0.12?l4花生油溶液经腹腔注射,制备小鼠急性化学性肝损伤模型,测定给药后模型小鼠血清丙氨酸氨基转移酶(ALT)、天冬氨酸转氨酶(AST),肝组织匀浆超氧化物歧化酶(SOD)活性以及丙二醛(MDA)水平,计算肝脏指数,光镜下观察肝组织病理学变化.[结果]茵陈紫金汤高、中剂量组均能显著降低CCl4所致小鼠急性肝损伤血清ALT、AST活性,提高肝组织匀浆SOD活性,降低MDA及肝脏指数,明显改善肝组织损伤的程度,与模型组比较差异有统计学意义(P<0.05).[结论]茵陈紫金汤对CCl4诱导的小鼠急性肝损伤有保护作用,其作用机制可能与抗脂质过氧化有关.  相似文献

目的:观察中药肝脾调补方对大鼠急性酒精性肝损伤的防护作用.方法:采用白酒灌胃法建立大鼠急性酒精性肝损伤模型.通过肝脾调补方高、中、低剂量灌胃,观察肝脾调补方对各组大鼠血清ALT、AST、肿瘤坏死因子(TNF-α)、白细胞介素-6(IL-6)及肝组织匀浆超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、一氧化氮(NO)水平的影响.结果:肝脾调补方可明显降低大鼠血清ALT、AST、TNF-α、IL-6水平;降低肝组织匀浆的NO的含量,提高肝匀浆SOD、GSH-Px活性.结论:肝脾调补方对急性酒精性大鼠肝损伤有防护作用,能改善肝功能,其机制可能与减轻肝脏炎症、抗脂质过氧化有关.  相似文献

目的观察软坚护肝片(RJHGT)对四氯化碳(CCl4)所致小鼠急性肝损伤的保护作用,并探讨其作用机制。方法腹腔注射CCl4制备小鼠急性肝损伤模型,检测血清谷丙转氨酶(AST)、谷草转氨酶(ALT)、碱性磷酸酶(AKP)活性及总抗氧化能力(T-AOC);检测肝组织丙二醛(MDA)含量、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-PX)活性。结果 RJHGT各组能够减轻肝细胞损伤程度,与模型组比较,RJHGT能降低血清ALT、AST、AKP的活性及肝组织中MDA、GSH的含量(P0.01或0.05);提高T-AOC能力及SOD、GSH-PX的活力(P0.01或0.05)。结论 RJHGT对CCl4所致小鼠急性肝损伤具有明显的保护作用,其机制可能与其抗脂质过氧化作用有关。  相似文献

目的:观察清热利湿法对四氯化碳(CCl4)诱导的小鼠急性肝损伤的保护作用.方法:给小鼠以0.12?l4花生油溶液腹腔注射,制备急性化学性肝损伤模型,测定各组血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性,计算肝脏指数,光镜下观察肝组织病理学变化.结果:小鼠灌胃给予清香散后,清香散大、小剂量组均能显著降低CCl4所致小鼠急性肝损伤血清ALT、AST活性及肝脏指数(P<0.05),大剂量组能明显改善肝组织损伤的程度,与模型组比较差异有统计学意义(P<0.05).小剂量组虽有改善肝组织损伤的趋势,但与模型组比较差异无统计学意义(P>0.05).结论:清热利湿法对CCl4诱导的小鼠急性肝损伤有保护作用.  相似文献

目的:观察金菊花乙酸乙酯提取物(EAEDM)对肝损害的防护作用,方法:建立大鼠急性乙醇中毒肝损害模型,通过大鼠灌胃EAEDM,以对照组对照,测定血清过氧化脂质(MDA),谷胱甘肽过氧化物酶(GSH-PX),血清谷丙转氨酶(ALT)和血清谷草转氨酶(AST)的含量,结果:EAEDM能降低MDA,ALT和AST的活性,减少肝脏MDA的生成,增加GSH-PX的活性,结论:EAEDM对肝损伤具有防护作用。  相似文献

目的 研究叶黄素对四氯化碳(CCl4)所致急性化学性肝损伤小鼠的保护作用.方法 雄性昆明种小鼠60只,随机分为正常对照组、急性化学性肝损伤模型组(模型组)、联苯双酯阳性对照组(阳性对照组)(联苯双酯15 mg/kg)以及叶黄素低、中、高剂量组(10、15、20 mg/kg)共6组,每组10只.测定并比较各组小鼠肝脏系数,血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性与丙二醛(MDA)含量;测定肝组织中SOD、GSH-Px活性、MDA含量以及组织病理系数.结果 叶黄素各剂量组均能升高急性化学性肝损伤小鼠血清与肝组织SOD、GSH-Px活性(P<0.01),降低血清ALT、AST活性(P<0.01)和血清与肝组织MDA含量(P<0.01),并能不同程度地改善肝脏病理组织损伤.结论 叶黄素对CCl4所致急性化学性肝损伤具有预防性保护作用.  相似文献

目的探讨犁头草提取物对四氯化碳(CCl_4)诱导大鼠急性肝损伤的修复效果和抗氧化作用。方法体外抗氧化评价主要考察对DPPH、ABTS、羟自由基以及还原力的清除率,利用CCl_4诱导昆明小鼠肝损伤后,以不同剂量的犁头草提取物灌胃7 d并且测定血清中天冬氨酸转氨酶(AST)、碱性磷酸酶(ALP)、丙氨酸转氨酶(ALT)、谷氨酰转肽酶(GGT)、总胆红素(TB)和肝肾组织中的超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽(GSH)以及丙二醛(MDA)等指标,HE染色观察肝脏病理组织学变化。结果体外抗氧化实验证明犁头草提取物具有较强的抗氧化效果,各剂量组均能降低CCl_4致小鼠急性肝损伤血AST、ALP、ALT、GGT和TB的升高,降低肝肾中MDA含量,增强SOD、CAT和GSH活性,减轻CCl_4对肝组织的病理损伤。结论犁头草提取物对CCl_4致小鼠急性肝损伤具有一定的保护作用。  相似文献

Objective: To investigate the hepatoprotective efficacy of cranberry extract(CBE)against carbon tetrachloride(CCl4)-induced hepatic injury using in-vivo animal model.Methods: The hepatoprotective efficacy of CBE(200 and 400 mg/kg) was investigated against CCl4(4 m L/kg)-induced hepatotoxicity, elevated liver enzymes [ALT(alanine aminotransferase), AST(aspartate aminotransferase), and alkaline phosphatase(ALP)],and total protein(TP) contents in the serum. Moreover, CBE-aided antioxidant defense against hepatotoxic insult of CCl4 was measured by evaluating a number of anti-oxidative biomarkers including reduced glutathione(GSH), superoxide dismutase(SOD), catalase(CAT), and malondialdehyde(MDA) in the serum by using spectrophotometric analyses.Results: Results showed that the exposure of experimental animals to CCl4 did induce significant hepatotoxicity compared to the non-induced(untreated) group. The oral administration of CBE demonstrated a significant dose-dependent alleviation in the liver enzymes(AST, ALT, and ALP), increased antioxidant defense(GSH, SOD, and CAT),and reduced MDA levels in the serum of treated animals compared to the animals without treatment. The resulting data showed that the administration of CBE decreased the serum levels of ALT, AST, and ALP compared to the CCl4-induced group.Conclusions: The resulting data evidenced that CBE exhibits promising hepatoprotective potential against the chemical induced hepatotoxicity, maintains homeostasis in liver enzymes, and can provide significant antioxidant defense against free radicals-induced oxidative stress.  相似文献

目的观察丹酚酸A(SAA)抗过氧化肝损伤的作用.方法采用CCl4诱导大鼠肝损伤模型,观察SAA对肝脏病理、血清Alb含量与ALT,AST活性,肝组织MDA含量的影响.另以CCl4熏蒸诱导肝细胞损伤,观察SAA对ALT,AST,GSHPX的活性以及对MDA,GSH含量的变化.结果SAA能减轻模型鼠肝脏病理变化,降低ALT,AST活性与MDA含量;降低损伤肝细胞ALT,AST,SOD,GSHPX活性与MDA含量,增高GSH含量.结论SAA具有良好的抗过氧化肝损伤作用.  相似文献

AIM: To investigate the hepatoprotective effect of manual acupuncture at Yanglingquan (GB34) on CCl4-induced chronic liver damage in rats. METHODS: Rats were injected intraperitoneally with CC4l (1 mL/kg) and treated with manual acupuncture using reinforcing manipulation techniques at left GB34 (Yanglingquan) 3 times a week for 10 wk. A non-acupoint in left gluteal area was selected as a sham point. To estimate the hepatoprotective effect of manual acupuncture at GB34, measurement of liver index, biochemical assays including serum ALT, AST, ALP and total cholesterol, histological analysis and blood cell counts were conducted. RESULTS: Manual acupuncture at GB34 reduced the liver index, serum ALT, AST, ALP and total cholesterol levels as compared with the control group and the sham acupuncture group. It also increased and normalized the populations of WBC and lymphocytes. CONCLUSION: Manual acupuncture with reinforcing manipulation techniques at left GB34 reduces liver toxicity, protects liver function and liver tissue, and normalizes immune activity in CCl4-intoxicated rats.  相似文献

ObjectiveTo evaluate the protective effect of ethanol extract of Mollugo nudicaulis (M. nudicaulis) against perchloroethylene-induced hepatotoxicity.MethodsThe hepatoprotective activity of the ethanol extract of M. nudicaulis (200 mg/kg body wt) was studied in percholoroethylene (1 000 mg/kg body wt) induced hepatotoxicity in Wistar albino rats. The serum levels of AST, ALT, ALP, bilirubin and the liver content of SOD, CAT, GPx, GST, GSH, vitamin C were assessed to evaluate the hepatoprotective and antioxidant activities of the extract. The activity of the extract was compared with silymarin, a standard reference drug. In addition, serum urea, uric acid and creatinine levels were measured to evaluate the kidney function. The histopathological examination of the liver tissues was observed to support the biochemical parameters.ResultsThe results revealed that the extract significantly (P<0.05) restored the serum levels of AST, ALT, ALP, bilirubin and significantly (P<0.05) increased the antioxidant enzymes SOD, CAT, GPx, GST, GSH, vitamin C in perchloroethylene-induced rats to its normalcy. The biochemical observations were supported by the histopathological studies of the liver tissues.ConclusionsThe results led to the conclusion that M. nudicaulis possess hepatoprotective and antioxidant activites against perchloroethylene-induced hepatotoxicity in rats.  相似文献

Objective:This study was performed to investigate the protective effect of Thymus vulgaris(T.vulgaris) leaves 70%alcoholic extract against alcohol-mediate hepatotoxicity rats.Methods:The protective effect of extract was investigated at dose of 500 mg/kg/day orally against alcohol-mediate hepatotoxicity using adult male Wister albino rats during 21 days.Protective effect of T.vulgaris extract was evaluated comparing with silymarin standard drug al recommended dose(25 mg/kg/day) orally for 21 days.Results:Alcohol-mediate hepatotoxicity rats(alcohol-control) showed hepatocytes distortion represented as marked increment on liver biomarkers;alkaline phosphatase(ALP),aspartate transaminase(AST) and alanine transaminase(ALT) activities,as well as pronounced reduction on total protein and its fractions albumin and globulin corresponding to normal ranges.Addition to oxidative stress status as depletion on glutathione concentration,catalase(CAT),superoxide dismutase(SOD),glutathione reductase(GR),glutathione-S-transferase(GST) and glutathione peroxidase(GPx)activities,concurrence with augmentation oxidative stress parameters;malondyaldchydc(MDA) and hydrogen peroxide(H_2O_2) concentrations comparing to normal values.Alcohol administration elevated total cholesterol(TC),low density lipoprotein cholesterol(LDL-C) and high density lipoprotein cholesterol(HDL-C) comparing to normal ranges.Co-administration T.vulgaris extract with alcohol showed protective effect on hepatocytes manifested as minimizing on ALP.AST and ALT activities and increment on total protein,albumin and globulin production compared to alcohol-control.Antioxidant enzymes activities;CAT.SOD.GR,GST and GPx were significantly magnified,while MDA and H_2O_2 concentration were lessened corresponding to alcohol-control.Also,lipid profile was markedly improved and risk ratio was lowered compared to alcohol-control.These results were confirmed by normalization of degenerated and fibrotic liver tissue as of alcohol-control.Conclusion:T.vulgaris extract appeared hepatoprotective,hypolipidemic and antioxidant activities on alcohol-mediate hepatotoxicity rats compared to silymarin.  相似文献

AIM: The protective effects of Ginkgo biloba phytosomes (GBP) on carbon tetrachloride (CCl4)-induced hepatotoxicity and the probable mechanism(s) involved in this protection were investigated in rats. METHODS: Liver damage was induced in Wistar rats by administering a 1:1 (v/v) mixture of CCl4 and olive oil (1 ml/kg, i.p.) once daily for 7 days. GBP at 25 mg/kg and 50 mg/kg, i.p. and reference drug silymarin (200 mg/kg, p.o.) were administered for 10 days to CCl4-treated rats, this treatment beginning 3 days prior to the commencement of CCl4 administration. The degree of protection was evaluated by determining the marker enzymes (SGOT, SGPT and SALP), albumin (Alb) and total proteins (TP). Further, the effects of GBP on lipid peroxidation (LPO), glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX) and glutathione reductase (GR) were estimated in liver homogenates to evaluate antioxidant activity. RESULTS: GBP (25 and 50 mg/kg) and silymarin elicited significant hepatoprotective activity by decreasing the activities of serum marker enzymes and lipid peroxidation and elevated the levels of GSH, SOD, CAT, GPX, GR, Alb and TP in a dose-dependent manner. CONCLUSION: The present findings indicate that the hepatoprotective effects of GBP against CCl4-induced oxidative damage may be due to its antioxidant and free radical-scavenging activity.  相似文献

AIM: To explore the antioxidant and free radical scavenger properties of mistletoe alkali (MA). METHODS: The antioxidant effect of mistletoe alkali on the oxidative stress induced by carbon tetrachloride (CCl4) in rats was investigated. The rats were divided into four groups (n=8): CCl4-treated group (1 mL/kg body weight), MA -treated group (90 mg/kg), CCl4 MA-treated group and normal control group. After 4 wk of treatment, the level of malondialdehyde (MDA), a lipid peroxidation product (LPO) was measured in serum and homogenates of liver and kidney. Also, the level of glutathione (GSH), and activities of glutathione reductase (GR), glutathione peroxidase (GSPx), superoxide dismutase (SOD), and glutathione-S-transferase (GST) in liver and kidney were determined. Scavenging effects on hydroxyl free radicals produced in vitro by Fenton reaction were studied by ESR methods using 5,5-dimethyl-1-pyrroline-N-oxide (DMPO) as a spin trap reagent and H2O2/UV as the OH·source. Urinary 8-hydroxydeoxyguanosine (8-OHdG) was determined by competitive ELISA . RESULTS: In CCl4-treated group, the level of LPO in serum of liver and kidney was significantly increased compared to controls. The levels of GSH and enzyme activities of SOD, GSPx and GR in liver and kidney were significantly decreased in comparison with controls. In CCl4 MA-treated group, the changes in the levels of LPO in serum of liver and kidney were not statistically significant compared to controls. The levels of SOD, GSPx and GR in liver and kidney were significantly increased in comparison with controls. There was a significant difference in urinary excretion of 8-OHdG between the CCl4-treated and MA-treated groups. CONCLUSION: Oxidative stress may be a major mechanism for the toxicity of CCl4 MA has a protective effect against CCl4 toxicity by inhibiting the oxidative damage and stimulating GST activities. Thus, clinical application of MA should be considered in cases with carbon tetrachloride-induced injury.  相似文献

The intracellular levels of oxidant and antioxidant balances are gradually distorted during the aging process. An age associated elevation of oxidative stress occurring throughout the lifetime is hypothesized to be the major cause of aging. The present study was undertaken to evaluate the putative antioxidant activity of the fermented Panax ginseng extract (GINST) on lipid peroxidation and antioxidant status of major organs of aged rats compared to young rats. Increased levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), urea and creatinine were observed in the serum of aged rats. Increased levels of malondialdehyde (MDA) and significantly lowered activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GR) and glutathione-S-transferase (GST) were observed in the liver, kidneys, heart and lungs of aged rats, when compared with those in young rats. Quantitative analysis of the non-enzymatic antioxidants such as reduced glutathione (GSH), ascorbic acid and α-tocopherol levels showed significantly lower values in the liver, kidneys, heart and lungs of aged rats. On the other hand, administration of the fermented Panax ginseng extract (GINST) to aged rats resulted in increased activities of SOD, CAT, GPx, GR and GST as well as elevation in GSH, ascorbic acid and α-tocopherol levels. Besides, the level of MDA, AST, ALT, urea and creatinine were reduced on administration of GINST to aged rats. These results suggested that treatment of GINST can improve the antioxidant status during aging, thereby minimizing the oxidative stress and occurrence of age-related disorders associated with free radicals.  相似文献

目的:观察绞股蓝总皂苷对四氯化碳( CCl4)诱导的大鼠肝纤维化的防治作用.方法:采用CCl4诱导的大鼠肝纤维化模型,分为正常组(Z,n=6)、模型组(M,n=8)、绞股蓝总皂苷组(J,n=8)、秋水仙碱(Q,n=8).造模6周末开始给药(股蓝总皂苷200mg/kg体重、秋水仙碱0.1mg/kg体重),给药3周.观察:①大鼠体重、肝脾比值的变化;②血清丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、谷氨酰转肽酶(GGT)活性、白蛋白( Alb)、总胆红素(TBil)含量、肝组织羟脯氨酸(Hyp)含量;③肝组织超氧化物歧化酶(SOD)活性及丙二醛(MDA)、还原型谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)含量;④肝组织病理及胶原沉积情况.结果:①M组大鼠血清ALT、AST、GGT、TBil显著升高,Alb显著降低;J和Q组大鼠血清ALT、AST、GGT、TBil显著下降,Alb显著升高;②M组大鼠肝组织Hyp含量显著升高,J组及Q组大鼠肝组织Hyp含量显著下降;③肝组织HE染色显示:M组大鼠肝细胞脂肪变性,大量纤维结缔组织增生,假小叶形成.J组及Q组大鼠肝细胞脂肪变性减轻,纤维增生减少,少见完整假小叶结构.天狼星红染色显示:M组大鼠肝窦周胶原沉积明显,形成较厚汇管区和中央静脉间的纤维间隔,J组和Q组大鼠肝脏汇管区胶原纤维染较M组明显减轻;④M组大鼠肝组织SOD活性及GSH含量明显降低,MDA含量显著升高.J组大鼠肝组织SOD活性显著提高.结论:绞股蓝总皂苷具有显著抗CCl4诱导的大鼠肝纤维化及氧化损伤的作用.  相似文献

Free radical-induced oxidative damage is considered to be the most important consequence of the aging process. The activities and capacities of antioxidant systems of cells decline with increased age, leading to the gradual loss of pro-oxidant/antioxidant balance and resulting in increased oxidative stress. Our investigation was focused on the effects of cordycepin (3′-deoxyadenosine) on lipid peroxidation and antioxidation in aged rats. Age-associated decline in the activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GR), glutathione-S-transferase (GST), reduced glutathione (GSH), vitamin C and vitamin E, and elevated levels of malondialdehyde (MDA) were observed in the liver, kidneys, heart and lungs of aged rats, when compared to young rats. Furthermore, serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), urea, and creatinine were found to be significantly elevated in aged rats compared to young rats. Aged rats receiving cordycepin treatment show increased activity of SOD, CAT, GPx, GR and GST, and elevated levels of GSH, and vitamins C and E such that the values of most of these parameters did not differ significantly from those found in young rats. In addition, the levels of MDA, AST, ALT, urea and creatinine became reduced upon administration of cordycepin to aged rats. These results suggest that cordycepin is effective for restoring antioxidant status and decreasing lipid peroxidation in aged rats.  相似文献
