目的 探索食管癌细胞EC9706及人正常食管上皮细胞HEEC中USP39分子的表达差异及沉默USP39对食管癌细胞EC9706增殖的影响。方法 利用qRT-PCR及Western blot法检测细胞中USP39表达情况;设计并合成USP39的siRNA (USP39-siRNA)及对照(USP39-NC)转染食管癌EC9706细胞,利用qRT-PCR、Western blot法检测EC9706细胞中USP39的表达变化;MTT、平板克隆实验检测EC9706细胞的增殖。结果 USP39在EC9706细胞中的表达高于人正常食管上皮细胞HEEC的表达,USP39-siRNA下调了EC9706细胞中USP39基因水平与蛋白水平的表达,并抑制了细胞的增殖能力。结论 USP39-siRNA能够下调USP39的表达,并有效抑制食管癌EC9706细胞的增殖,为以USP39为靶点的食管癌基因治疗奠定基础。  相似文献

目的 采用重组慢病毒CIAPIN1表达载体和CIAPIN1沉默载体感染食管癌EC9706细胞,观察调控CIAPIN1基因表达对细胞增殖、细胞周期、细胞凋亡及体外侵袭转移的影响。 方法 构建CIAPIN1表达慢病毒载体和CIAPIN1 siRNA慢病毒载体,包装得到重组慢病毒;分别感染食管癌EC9706细胞,筛选得到稳定CIAPIN1基因高表达和低表达的细胞。实验分为CIAPIN1表达组、CIAPIN1 siRNA组、无关siRNA对照组和空白对照组。采用RT-PCR和蛋白质印迹法分析CIAPIN1基因和蛋白的表达,MTT实验检测细胞增殖,流式细胞术(FCM)分析细胞周期及细胞凋亡的变化, Boyden小室检测细胞侵袭转移。 结果 与空白对照组、无关siRNA对照组和CIAPIN1 siRNA组比较,CIAPIN1高表达组细胞生长受到抑制(P<0.05);S和G2/M期细胞比例减少,G0/G1期细胞比例增加(P<0.05);平均细胞凋亡率增加(P<0.05),穿透Matrigel的平均细胞数减少(P<0.05)。 结论 以慢病毒为载体介导的CIAPIN1基因高表达可有效抑制食管癌EC9706细胞增殖、促进细胞凋亡和降低细胞侵袭迁移能力。  相似文献

目的 观察香芹酚对前列腺癌细胞增殖、凋亡以及侵袭的影响,并从丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)信号通路探讨其可能的作用机制。方法 采用80 μmol/L浓度的香芹酚干预前列腺癌DU145细胞(香芹酚组)24 h或者48 h,同时设置不加香芹酚的阴性对照组。CCK-8法检测细胞在第0、1、2、3、4、5天的生长情况,绘制细胞生长曲线;流式细胞术检测细胞凋亡情况;Transwell侵袭实验检测细胞侵袭能力;RT-PCR以及蛋白质印迹分析检测基质金属蛋白酶9(MMP-9)及其抑制剂TIMP-1的表达;蛋白质印迹分析检测聚腺苷二磷酸核糖聚合酶(PARP)、半胱天冬氨酸蛋白酶9(caspase-9)的表达以及细胞外信号调节激酶(ERK)、p38信号通路的激活情况。结果 与阴性对照组比较,香芹酚作用后前列腺癌细胞DU145活性明显受到抑制,作用的第3天起细胞增殖能力降低(P<0.05,P<0.01)。香芹酚作用后,细胞凋亡率升高(P<0.05),细胞侵袭能力降低(P<0.01),细胞TIMP-1、caspase-9表达升高,PARP发生裂解,p38信号被激活,同时MMP-2表达降低,ERK信号通路受到抑制。结论 香芹酚能够抑制前列腺癌细胞的增殖和侵袭,诱导其凋亡,其作用与MAPK信号通路有关。  相似文献

目的 :探讨HOTAIR/CSTF2/CD44轴调控食管癌(esophageal cancer,EC)侵袭转移的机制。方法 :收集2020年9月―2022年6月43例非特异性侵袭性EC患者的EC组织样本和邻近正常组织,分为对照组和EC组。培养EC细胞系KYSE150,EC109和TE-1以及人正常食管上皮细胞系HEEC细胞,分为control、si-HOTAIR、CSTF2模拟物和si-HOTAIR+CSTF2模拟物等4组。采用实时荧光定量PCR检测HOTAIR、CD24和CSTF2的表达,CCK8测定细胞活力,流式细胞术检测细胞凋亡,随后检测细胞的迁移和侵袭,蛋白印迹检测细胞凋亡蛋白(Ki67、PCNA、裂解的半胱天冬酶-3和半胱天冬酶-9)和迁移蛋白(VEGF和MMP-9)。监测小鼠模型肿瘤生长的迹象。结果 :HOTAIR和CSTF2在EC组织中的表达显著高于正常组织;与HEEC细胞相比,HOTAIR和CSTF2在EC细胞系EC109,KYSE150和TE-1中表达增加;与control组比较,HHOTAIR敲低显著降低了细胞增殖、迁移和侵袭,并加速了细胞凋亡;与CSTF2相互作用...  相似文献

目的探讨miR-338-3p影响食管癌细胞增殖、迁移、侵袭及凋亡的分子机制及其对WNK1的靶向调控作用。方法体外培养人正常食管上皮细胞株Het-1A与人食管癌细胞Eca-109、EC-1、EC9706,采用q RT-PCR与Western blot分别检测miR-338-3p、WNK1的表达水平;以EC9706细胞为研究对象,分别将miR-con、miR-338-3p mimics、si-con、si-WNK1、miR-338-3p mimics与pc DNA、miR-338-3p mimics与pc DNA-WNK1转染至EC9706细胞;MTT法检测细胞增殖、流式细胞术检测细胞凋亡、Transwell小室实验检测迁移及侵袭能力;双荧光素酶报告实验检测miR-338-3p与WNK1的靶向关系。结果与Het-1A比较,食管癌细胞Eca-109、EC-1、EC9706中miR-338-3p的表达显著下调(P0.05),WNK1 m RNA及蛋白表达显著上调(P0.05);转染miR-338-3p mimics或si-WNK1可明显降低EC9706细胞的活力、迁移及侵袭细胞数(P0.05),增加EC9706细胞的凋亡率(P0.05);双荧光素酶报告实验证实miR-338-3靶向结合WNK1;共转染miR-338-3p mimics与pc DNA-WNK1可明显逆转转染miR-338-3p mimics对细胞增殖、迁移、侵袭及凋亡的作用。结论 miR-338-3p可靶向下调食管癌中的WNK1,进而抑制食管癌细胞的增殖、迁移及侵袭,和诱导细胞凋亡。  相似文献

目的 建立耐紫杉醇食管癌(EC)细胞系EC9706/PTX,观察姜黄素对EC9706/PTX细胞上皮间质转化(EMT)的抑制作用并探讨其机制,为耐药EC的治疗提供理论依据.方法 用中等浓度间歇作用法建立耐紫杉醇EC细胞EC9706/PTX,四甲基偶氮唑蓝(MTT)法测定细胞耐药指数及交叉耐药性,检测不同浓度姜黄素对EC9706/PTX细胞增殖的抑制.使用细胞骨架染色、划痕实验、Transwell侵袭实验检测姜黄素对EC9706/PTX细胞形态变化、迁移和侵袭能力的影响.荧光定量PCR及蛋白免疫印迹(Western blot)检测姜黄素对EC9706/PTX细胞中EMT相关分子标志物E-钙黏蛋白、N-钙黏蛋白、波形蛋白、纤维连接蛋白在mRNA和蛋白水平的表达影响.Western blot检测姜黄素对EC9706/PTX细胞中转录因子NF-κB p65及Snail在蛋白水平的表达影响.结果 EC9706/PTX对紫杉醇的耐药指数为28.4,对顺铂、阿霉素产生交叉耐药性,姜黄素能够抑制EC9706/PTX细胞的增殖.在20 μmol/L浓度的姜黄素作用下,EC9706/PTX细胞的迁移和侵袭能力明显降低.荧光定量PCR及West-ern blot检测显示,细胞耐药后E-钙黏蛋白的表达明显下调,而N-钙黏蛋白表达则明显上调,姜黄素逆转了这一现象.Westernblot检测提示,EC细胞发生耐药及EMT后,NF-κB p65及Snail蛋白的表达增强,姜黄素阻断了这一作用.结论 姜黄素能够抑制紫杉醇耐药EC细胞的增殖并且能够逆转紫杉醇耐药EC细胞的EMT现象,其机制可能是通过抑制NF-κB-Snail信号通路实现的.  相似文献

【目的】 探讨泰山银杏外种皮多糖(Ginkgo biloba exocarp polysaccharides,GBEP)对体外培养的食管癌细胞增殖和凋亡的影响及其作用机制。 【方法】 通过沸水浸提,有机溶剂沉淀,并通过sevage法除去蛋白,获得GBEP。利用MTT法检测不同浓度的GBEP对食管癌细胞EC-9706增殖的影响;流式细胞术检测细胞周期的分布;Annexin-V/PI双染分析凋亡率;蛋白质印迹法检测与细胞周期及细胞凋亡相关的蛋白表达变化。 【结果】 GBEP显著抑制EC-9706细胞的增殖,并呈时间、剂量依赖性。其半数抑制浓度(IC50)为138.9 μg/mL。细胞周期分析结果表明,GBEP处理后G1期细胞比例由52.25%增加至69.76%,蛋白质印迹结果显示细胞周期蛋白D1的表达呈浓度依赖性下降。说明GBEP诱导EC-9706细胞G1期阻滞。流式细胞术分析结果表明,GBEP诱导的EC-9706细胞凋亡随剂量和作用时间增加而增加。同时GBEP抑制抗凋亡蛋白Bcl-2的表达,增加促凋亡蛋白Bax的表达,促进caspase3的激活和PARP的剪切。 【结论】 泰山银杏外种皮多糖抑制食管癌细胞的增殖,诱导细胞G1期阻滞,并激活内源性细胞凋亡。  相似文献

目的:探讨硒蛋氨酸对食管癌细胞系EC9706环氧合酶-2(COX-2)表达的影响。方法:采用MTT比色法、细胞生长曲线描绘观察硒蛋氨酸对食管癌细胞系EC9706增殖的影响,琼脂糖凝胶电泳法检测DNA ladder,细胞免疫化学方法检测EC9706COX-2的表达。结果:硒蛋氨酸呈时间、剂量依赖性方式抑制EC9706细胞增殖,抑制食管癌细胞系EC9706COX-2的表达,诱导细胞凋亡。结论:硒蛋氨酸可能通过抑制食管癌细胞系EC9706COX-2的表达从而抑制EC9706细胞增殖,诱导细胞凋亡。  相似文献

目的 通过RNA干扰(RNAi)技术沉默DNA聚合酶polβ的表达,观察其对食管癌细胞顺铂(cDDP)耐药性的影响。方法 构建靶向polβ基因的siRNA重组慢病毒pRNAT-U6.2/Lenti-polβ1、pRNAT-U6.2/Lenti-polβ2及阴性对照载体pRNAT-U6.2/Lenti-polβC,将其感染人食管鳞癌顺铂耐药细胞系EC9706/cDDP,通过RT-PCR、蛋白质印迹分析和免疫荧光实验观察细胞polβ的表达情况,采用MTT法观察不同浓度顺铂对细胞生长的抑制作用并计算50%细胞生长抑制所需的药物浓度(IC50)和耐药指数(RI)。结果 靶向polβ的siRNA重组慢病毒pRNAT-U6.2/Lenti-polβ1和pRNAT-U6.2/Lenti-polβ2感染EC9706/cDDP后均可下调polβ的mRNA和蛋白表达水平,其中前者效果更为显著。顺铂以剂量依赖方式抑制EC9706/cDDP细胞增殖,感染pRNAT-U6.2/Lenti-polβ1、pRNAT-U6.2/Lenti-polβC的EC9706/cDDP细胞及未感染EC9706/cDDP细胞对顺铂的IC50分别为55.71、62.41、63.11 μg/mL,耐药指数分别为13.9、15.5、15.7,其中前者与后二者相比差异有统计学意义(P<0.05)。结论 polβ的表达与食管鳞癌细胞系EC9706/cDDP顺铂耐药性有关,沉默其表达可部分逆转EC9706/cDDP细胞对顺铂的耐药性。  相似文献

目的 采用RNAi抑制食管癌EC9706细胞株MTA1基因的表达,观察对食管癌细胞侵袭和迁移的影响.方法 用脂质体包裹转染沉默MTA1表达的siRNA表达载体人食管癌细胞EC9706.定量RT-PCR和免疫印迹分别检测MTAl mRNA和蛋白表达情况;划痕损伤实验和细胞体外侵袭实验分别测定迁移和侵袭的影响.结果 定量RT-PCR检测显示,MTA1基因的表达水平明显降低;转染沉默MTA1的siRNA表达载体组的食管癌细胞划痕未愈合,穿透Matrigel的细胞数量明显减少,与未转染组、转染空载体组和转染无关序列siRNA载体组比较差异有显著性(P<0.05).结论 RNA干扰介导的MTA1基因表达沉寂可有效降低食管癌细胞侵袭和迁移;MTA1基因可能成为肿瘤治疗上有前途的分子靶点.  相似文献

Objective: To evaluatel the value of D-dimers in patients with acute aortic dissection (AAD). Methods: This study consisted of 16 patients with AAD and 27 non-AAD patients. Serum D-dimets were measured by Sta-Liatest D-DI immunoturbidimetric assay. Results: D-dimer level was higher (P < 0.001) in patients with AAD(7.91 ± 5.52 μg/ml) than that in non- AAD group(1.57±1.24 μg/ml). D-dimer was positive (>0.4 μg/ml) in all patients with AAD and in 10 control group patients (37%). Among patients with acute AAD, D-dimers tended to be higher in Stanford A than in Stanford B (8.67 ± 4.31 μg/ml vs. 3.24±1.27 μg/ml, P <0.01). D-dimer values tended to be higher in more extended disease(3.84 ± 1.65 μg/ml, 8.57 ± 3.58 μg/ml and 11.87 ± 5.69 μg/ml in thoracic aorta, thoracic and abdominal aorta, thoracic and abdominal aorta and iliacal arteries, respectively, P < 0.05 for both 8.57 ± 3.58 and 11.87 ± 5.69 vs. 3.84 ± 1.65 ). Including the control group into the analysis, we found a sensitivity of 100%, a negative predictive value of 100%, and a specificity of 66% and a positive predictive value of 64% for D-dimer in diagnosis of AAD in our patients with suspected AAD. Conclusion: D-dimer was elevated in patients with AAD. A negative D-dimer test result could be useful in excluding AAD.  相似文献

Objective: To set up a simple and reliable rat model of combined liver-kidney transplantation. Methods: SD rats served as both donors and recipients. 4℃ sodium lactate Ringer's was infused from portal veins to donated livers,and from abdominal aorta to donated kidneys, respectively. Anastomosis of the portal vein and the inferior vena cava (IVC) inferior to the right kidney between the graft and the recipient was performed by a double cuff method, then the superior hepatic vena cava with suture. A patch of donated renal artery was anastomosed to the recipient abdominal aorta. The urethra and bile duct were reconstructed with a simple inside bracket. Results: Among 65 cases of combined liver-kidney transplantation, the success rate in the late 40 cases was 77.5%. The function of the grafted liver and kidney remained normal. Conclusion: This rat model of combined liver-kidney transplantation can be established in common laboratory conditions with high success rate and meet the needs of renal transplantation experiment.  相似文献

Objective To observe blood pressure change with age in salt-sensitive teenagers whose salt sensitivity were determined by repeated testing.Methods Salt sensitivity was determined through intravenous infusion of normal saline combined with volume-depletion by oral diuretic furosemide in 55 teenagers. After five years, salt sensitivity was re-examined and subject blood pressure was followed up. Blood pressure changes in salt-sensitive teenagers were compared to that of non-salt sensitive teenagers over five years.Results After 5 years, the repetition rate of salt sensitivity determined by intravenous saline loading is 92.7%. In teenagers with salt sensitivity on the baseline, both the systolic blood pressure increments and increment rates were much higher than non-salt sensitive teenagers (12.7±12.1 mmHg vs. 2.8±5.2 mmHg, P< 0.01; 12.2%± 12.0% vs. 2.5% ±4.4%, P< 0.001,respectively). There was a similar trend for diastolic blood pressure (8.4 ± 6.4 mmHg vs. 3.7 ± 6.4 mmHg, P = 0.052; 13.2% ±10.6 % vs. 6.8%± 10.1%, P = 0.053, respectively).Conclusions Salt sensitivity determined by intravenous saline loading showed good reproducibility. Blood pressure increments with age were much higher in salt-sensitive teenagers than non-salt sensitive teenagers, especially in terms of systolic blood pressure.  相似文献

目的:评价使用安心颗粒对急诊经皮冠状动脉介入术(PPCI)术后生活质量的影响.方法:将160例接受PPCI的急性ST段抬高型心肌梗死患者随机分为安心颗粒组(术前顿服安心颗粒8.8g,术后安心颗粒4.4 g/次,每日2次)和对照组(仅接受基础药物治疗).所有患者均服用阿司匹林、氯吡格雷和阿托伐他汀.分别在入院时、出院前1d、出院后180 d时,应用心肌梗死多维度量表(MIDAS)、中文版SF-36评价量表对患者生活质量评分.并观察术后30 d以内的出血并发症、血小板减少症发生情况.结果:入院时和出院前1d,两组患者的心肌梗死MIDAS、SF-36量表评分比较无差异(P>0.05);出院后180 d时,与对照组比较,安心颗粒组MIDAS、SF-36评分明显减低(P<0.05);组内与入院时比较,两组出院前1d、出院后180 d时,MIDAS、SF-36评分均降低(P<0.05).两组患者在随访期间均无大量出血、少量出血、重度和极重度血小板减少症发生,安心颗粒组有4例、对照组有7例发生不明显出血(P>0.05).两组发生轻度血小板减少症的患者数比较无差异(P>0.05).结论:PPCI使用安心颗粒,能改善急性ST段抬高型心肌梗死患者的生活质量,且不增加出血风险.  相似文献

Objective:To investigate the influences of urapidil and nicardipine on rabbit sinus function,atrio-ventricular node function and hemodynamics.Methods:Thirty-two Angora's rabbits were selected and randomly divided into four groups.U1 group:urapidil 0.25 mg/kg;U2 group:urapidil 0.5 mg/kg;N1 group:nicardipine 10 μg/kg;N2 group:nicardipine 20 μg/kg.All these medicine were administrated within 30 seconds.Measurements were taken before and after the administration of urapidil or nicardipine for the following data:mean blood pressure(MAP),heart rate(HR),sino-atrial conduction time(SACT),maximal sinoatrial recovery time(SNRTmax)corrected sinus node recovery time(CSNRT),index of sinus node recovery time(SNRTI),Wenckebach A-V conduction frequency (WB),and P-R interval.Results:Significant MAP and HR changes were identified in all of the four groups before and after administration of both urapidil and nicardipine.No significant changes could be found in the rest of the parameters.Intergroup analysis showed that SACT and CSNRT of N1 and N2 groups were shorter than those of the U2 group(P<0.01);the MAP decreased(P<0.01)and the HR increased drastically(P<0.01).Conclusions:Neither urapidil(0.25 mg/kg,0.5 mg/kg)nor nicardipine(10μg/kg,20μg/kg)has any significant influence on rabbit sinus function or rabbit atrio-ventricular node function.Nicardipine could be a better choice than urapidil for parafunctional sinus node patients.  相似文献

Objective:To investigate the gene expression of osteoprotegerin(OPG) and osteoclast differentiation factor(ODF) in the bone tissue of patients with hip fracture due to osteoporosis. Methods:OPGmRNA and ODFmRNA in the bone tissue in 50 cases of osteoporosis sufferers(over 50 years old) with hip fracture(Observer Group) and 30 cases of hip facture sufferers with no osteoporosis(Control group) were analyzed with the Semi-Quantitative RT-PCR method. Results:The mRNA expressed of ODF, OPG were both high in the patients with hip fracture. In the control group, the expression of OPG mRNA was observed, while the expression of ODF mRNA was very slight. Conclusion:Aged patients contained all signals including OPG, ODF that are essential for inducing osteoclastogenesis and promoting bone resorption.  相似文献

Objective:To investigate the clinical features, pathological characteristics and immunophenotype of solid-pseudopapillary tumor of the pancreas(SPTP). Methods:Nine surgically treated cases of SPTP were retrospectively reviewed. Hematoxylin and Eosin(HE) staining and immunohistochemical staining were used to analyze all cases, and the general clinical data was collected. Results:Six patients were asymptomatic except for a palpable mass. Two patients complained of vague-epigastric pain. One patient appeared jaundice. The tumor was encapsulated and solid tissues alternately with cystic tissues. Histologically, the histological structure of solid portion was pseudopapillary with a fibrovascular core. Tumor cells were uniform and medium-sized which were arranged in sheets ets or nests or pseudopapillary patterns. Immunohistochemical studies demonstrated that SPTP proved positive in vimentin(9/9 cases), AAT(9/9 cases), NSE(9/9 cases), ACT(7/9 cases), CK20(2/9 cases), CgA(1/9 cases), S-100(3/gcases), PR(4/gcases), Syn(3/9 cases) and CD56(5/9cases), negative in CEA and ER. Conclusion:SPTP is a tumor predominantly occurring in young women frequently without special symptoms. This tumor has various characteristical histological patterns with different immunophenotype.  相似文献

Objective:To probe into the influence of changes of ovarian hormones on the pathogenesis of the specific sub-type premenstrual syndrome(PMS)and reveal partial microcosmic mechanisms of adverse flow of liver-qi.Methods:Estradiol(E2)and progesterone(P)levels in serum were determined at different phases of menstrual cycle by radioimmunoassay.Results:In the group of PMS with adverse flow of liver-qi.the secretive peak value Of E2 and P at the follicular phase significantly decreased,and the secretive peak value at the luteal phase did not come into being.Conclusions:Low E2 and P secretive peak at the follicular phase and absence of secretive peak at the luteal phase is one of the microcosmic mechanisms of PMS with adverse flow of liver-qi.One of the pathophysiologic mechanisms of specific sub-type PMS is probably the continuous low level of E2and P.  相似文献

Real-time three-dimensional echocardiography (RT3DE)is a new ultrasound technique that enables dynamic threedimensional visualization and quantification of the heart in real time. Investigation of feasibility and methodology of RT3DE in determining left ventricular (LV) and right ventricular (RV) volumes, RT3DE was performed in 35 normal adults using Philips SONOS 7500 system with a 2-4 MHz matrix array transducer. The 60°×60° "pyramid" volume database was obtained and analyzed on a TomTec echo workstation. Both LV and RV volumes were calculated with four 3DE methods (i.e. apical 2, 4, 8, and 16-plane) through manually tracing ventricular endocardial borders in end diastole and end systole. Stroke volumes were then calculated. LV volume was also measured by 2DE Simpson's rule using GE VIVID 7 ultrasound machine.  相似文献

Increasing maternal age is the only etiological factor unequivocally linked to Down's syndrome in humans. The occurrence rate of newborns with Down's syndrome is about 1/220 in women over 35 years old. However, the occurrence rate in embryos fertilized in vitro, of the elder woman is unclear. Using FISH we screened the number of chromosome 21 in preimplanted embryos of 5 elderly women (average age, 38.4 years) to study the feasibility and necessity of screening trisomy 21 in embryos in patients over 35 years old at the in vitro fertilization (IVF) center.  相似文献
