目的:探讨乳腺癌中bcl-2蛋白表达及DNA倍性与患者临床病理因素的关系,方法:采用流式细胞光度术(FCM)定量分析63例原发性乳腺癌癌组织、相应癌旁组织和22例乳腺良性肿瘤中bcl-2蛋白表达及DNA倍性结果:乳腺癌组织中bcl-2蛋白表达水平及DNA指数(DI)、S期比例(SPF)和增殖指数(PI)显著高于相应癌旁组织和乳腺艮性肿瘤(P<0.05),而后两者在上述指标上未见显著性差异(P>0.05)。浸润性非特殊类型癌患者DI、SPF和Pl明显高于早期浸润癌患者和浸润性特殊类型癌患者(P<0.05);有淋巴结转移组患者DI和SPF显著高于无淋巴结转移组患者(P<0.05)bcl-2蛋白表达与患者年龄、病理组织学类型、肿瘤大小、淋巴结转移、临床分期均无关(P>0.05)癌组织bcl-2蛋白表达水平与SPF和PI呈负相关(P<0.05)结论:bcl-2蛋白表达与乳腺癌发生、发展有关:DI和SPF与淋巴结转移有关;DNA含量可作为判断乳腺癌预后的重要指标  相似文献

The expression of lactoside-binding lectin L-31 was analyzed in normal mucosa and in primary and metastatic gastric carcinomas. Immunoblotting revealed L-31 lectin in extracts of normal and malignant gastric tissues from 26 patients. The L-31 level was higher in tumor than in normal tissue in 9/26 cases, similar in 14/26 cases, and lower in 3/26 cases. Anti-L-31 monoclonal antibodies (MAbs) were used in immunohistochemical analyses to compare lectin expression in specimens of primary gastric carcinomas and adjacent normal mucosa from 39 patients and in specimens of metastases and the corresponding primary gastric carcinomas from 74 patients. The lectin was detected in normal gastric epithelial cells and in all gastric carcinoma specimens, albeit in varying amounts. The L-31 level was significantly higher in the primary tumor than in adjacent normal tissue in 55% of the well-differentiated tubular carcinoma cases and in 50% of stage-III and -IV tumors. L-31 expression in liver metastases from well-differentiated tubular primary gastric carcinomas was higher in 31% of the cases relative to the corresponding primary cancers. Likewise, L-31 expression in metastases from poorly differentiated gastric carcinomas in lymph nodes was higher in 38% of the cases compared to the primary cancers. The higher expression of L-31 in primary cancers and metastases of certain types implicates this lectin in the metastatic phenotype, but the presence of L-31 in a primary cancer is not sufficient to allow the metastatic propensity of the tumor to be predicted.  相似文献

背景与目的:胃癌复发转移是术后化疗失败的主要原因,本文试图通过观察胃癌组织中EGFR、IL-6R、PCNA的表达、DNA指数(DI)及DNA倍体,并寻找它们在胃癌复发转移者组织中的分布规律,从而为判定胃癌是否有术前转移及术后复发提供可靠的依据。方法:应用免疫组织化学SP法及图像分析仪Fegulgen染色对199例胃癌、20例正常胃粘膜、20例不典型增生中的EGFR、IL-6R、PCNA、DI及DNA倍体进行检测。结果:有淋巴结及远处转移、残胃癌的癌组织中EGFR表达水平(阳性率分别为61.11%、66%、66.67%)较其它癌组织、正常胃粘膜、不典型增生(阳性率分别为47.83%、0%、35%)高(P<0.01);DNA倍体分布形式上,有转移胃癌、残胃癌的癌组织中非整倍体细胞百分率(56.9±13.3)%明显高于其它(无转移胃癌为(12.8±6.3)%,正常胃粘膜及不典型增生为0%,早期胃癌为11.2%),它们之间相比有显著性差异(P<0.01);DNA指数亦高(有转移胃癌及残胃癌DNA指数为2.91±0.33,无转移胃癌DNA指数为2.17±0.29,二者相比有显著差异P<0.01),但2C细胞检出率低(无转移胃癌为43.8±12.9,有转移胃癌及残胃癌为10.2±7.5,二者相比有显著差异P<0.01)。结论:EGFR、DI及DNA倍体可作为胃癌术前判断是否有复发、转移危险性的指标。  相似文献

 本文采用流式细胞术检测了54例乳腺肿瘤细胞的DI、PI和S期细胞比例等项参数,其中34例乳腺癌进行了AgNOR计数和5年~14年随访。经分析表明:DI、PI和S期细胞比例可区分肿瘤的良恶性,AgNOR计数、DI、PI和S期细胞比例与乳腺癌的预后有关。  相似文献

Although there have been many studies analyzing the heterogeneity of DNA ploidy either in primary breast cancer or between the primary and synchronous metastatic lesions of breast cancer, few studies have been made regarding a comparison of DNA ploidy between the primary and recurrent lesions of breast cancer. Flow cytometric analyses for DNA ploidy were performed on both the primary lesions and the matching synchronous metastatic lymph nodes of 104 breast cancers and on both the primary and matching recurrent lesions of 31 breast cancers. A concordance of DNA ploidy between the primary lesions and the matching metastatic lymph nodes was found in 80 (77%) cases, while a concordance of DNA ploidy between the primary and matching recurrent lesions was found in 22 (71%) cases. Forty-six (94%) of 49 cases showed less than an 8.0% difference in the DNA indices of the aneuploid peaks between the primary lesions and the metastatic lymph nodes, while 13 (87%) of 15 cases showed less than 15.0% difference in the DNA indices between the primary and recurrent lesions. The aneuploid group and the heterogeneity group, divided according to the comparison of DNA ploidy between the primary and synchronous metastatic lesions, had a worse disease-free survival than that of the diploid group (p=0.0426 and p=0.0503, respectively). In conclusion, the majority of DNA ploidies are consistent among the primary, synchronous metastatic and recurrent lesions of breast cancer. The existence of aneuploidy itself may also play a prognostic role in breast cancer.  相似文献

Galectin-3, a multifunctional beta-galactocide binding lectin possibly participates in a variety of biological events including cell proliferation, differentiation, and apoptosis. The implication of galectin-3 during malignancy progression has been suggested in several cancers, including colon, prostate, thyroid, and breast cancer, however, scarce data are available in gastric cancer. We examined the expression of galectin-3 in 86 primary gastric cancers and the 40 metastatic lymph nodes by immunohistochemistry to explore whether it is related to the malignant progression. Positive galectin-3 expression was observed in 84% of the gastric cancer cases. In enhanced cells of cancerous lesions, 48% showed stronger nuclear immunoreactivity than cytoplasmic one, whereas adjacent epithelial cells showed little or weak nuclear immunoreactivity. When galectin-3 expression in gastric carcinoma was compared with that in gastric tissues adjacent to the cancers, there was a significant difference. The degree of enhancement of immunoreactivity was different corresponding to various histopathological subtypes in cancer tissues. A significantly stronger expression of galectin-3 in cancer tissues was only observed in papillary and poorly differentiated adenocarcinoma. When galectin-3 expression and tumor progression (TNM staging) was compared, a significant correlation was observed in overall cases, and only in poorly differentiated adenocarcinoma the galectin-3 expression correlated with tumor progression among various subtypes. Galectin-3 expression was observed significantly stronger in metastatic lymph nodes than in the primary gastric cancers, and also in these cases among histological subtypes, only in poorly differentiated adenocarcinoma, the expression of galectin-3 in metastatic lymph nodes was stronger than the primary cancer. In conclusion galectin-3 might be a useful tumor marker for gastric cancers with respects to tumor progression and potentiality of lymph node metastasis especially in certain histological types of gastric cancer.  相似文献

A study has been conducted of the cellular DNA contents in the primary and metastatic lesions of 30 cases of lung-metastasized stomach cancers, in which each DNA Index (DI) was calculated and analysed comparatively. As a consequence, the rate of the diploid type in the nodular lung metastases was found to be higher than the rate of the others. Further, the DI's of 22 out of 30 (73%) metastatic lymph nodes corresponded with those of the primary lesions. Four lung metastatic lesions out of five differed from their primary lesions, but all five lesions simulated their metastatic lymph nodes. Thus understanding of the properties of the metastatic lymph nodes, rather than their primary lesions, would seen to be helpful in planning the therapy to combat recurrent cancers.  相似文献

目的 :研究Fas配体 (FasL)和Caspase 3表达在胃癌发生和演进中的作用。方法 :利用免疫组化方法检测FasL和Caspase 3在 113例胃癌旁上皮细胞、原发灶癌细胞和淋巴细胞以及转移灶癌细胞中的表达 ,比较原发灶癌细胞中两种蛋白表达与胃癌病理特征的关系 ,探讨原发灶癌细胞中FasL表达与原发灶癌细胞或浸润淋巴细胞中Caspase 3表达的关系。结果 :FasL在原发灶癌细胞中阳性表达率高于癌旁上皮细胞 ( 5 3 98%vs3 4 5 1% ) ,P <0 0 5 ,Caspase 3则有相反趋势 ( 3 2 74%vs 5 0 44 % ) ,P <0 0 5。70 80 %原发灶浸润淋巴细胞中Caspase 3呈阳性表达 ,显著高于原发灶癌细胞和癌旁上皮细胞中的阳性率 ,P <0 0 5。FasL在转移灶癌细胞中阳性表达率低于对应原发灶癌细胞 ( 5 1 16%vs 81 5 8% ) ,P<0 0 5 ,Caspase 3则有相反趋势 ( 5 8 14 %vs 3 4 2 1% ) ,P <0 0 5。原发灶癌细胞中FasL表达与胃癌肿块大小、浸润深度、转移、分化和lauren分型显著相关 ,P <0 0 5。原发灶癌细胞中FasL表达与其浸润淋巴细胞中Caspase 3表达呈正相关 ,P <0 0 5。结论 :FasL表达上调和Cas pase 3表达下调在胃癌发生中起到重要作用。FasL可能通过诱导浸润淋巴细胞凋亡而参与胃癌分化、浸润和转移 ,可作为一种反映胃癌生物学行为的有  相似文献

目的:研究食管鳞状细胞癌DNA含量的分布及倍体类型与其生物学行为的关系。方法:采用流式细胞技术对手术切除的106例食管鳞癌患者新鲜标本进行DNA分析,分析DNA含量、S期细胞分数、增殖指数及倍体类型与食管鳞癌浸润深度、淋巴结转移、病理分级及分期等生物学行为的关系。结果:食管鳞癌患者异倍体出现率为82.08%。DNA含量、S期细胞分数、增殖指数随TNM分期增加呈现逐渐增高的趋势,但与肿瘤浸润深度(T分期),病理分级不相关。有淋巴结转移组食管鳞癌S期细胞分数和增殖指数均高于无淋巴结转移组,DNA含量两组间差异并无统计学意义。异倍体食管鳞癌DNA含量、S期细胞分数、增殖指数及淋巴结转移均明显高于二倍体食管鳞癌。结论:食管鳞癌DNA含量的分布及倍体类型与其生物学行为相关,DNA分析是评估食管鳞癌预后、制定术后治疗方案的一个有价值的参数。  相似文献

目的: 探讨人胃癌组织中乙酰肝素酶(HPA)、上皮标志物E-cadherin、间质标志物N-cadherin和vimentin蛋白的表达及其与人胃癌临床病理指标的关系,以及HPA与E-cadherin、N-cadherin、vimentin蛋白之间的关系。方法: 应用免疫组织化学方法检测91例人胃癌组织中HPA、E-cadherin、N-cadherin和vimentin蛋白表达情况;采用卡方检验,检测这几种蛋白阳性表达率与人胃癌不同病理指标的关系;同时采用Spearman等级相关分析HPA与E-cadherin、N-cadherin、vimentin蛋白之间的关系。结果: 人胃癌组织中HPA、E-cadherin、N-cadherin和vimentin蛋白阳性表达率为75.82%、51.65%、54.95%和23.08%。91例人胃癌组织中HPA、E-cadherin、N-cadherin和vimentin蛋白的阳性表达率与胃癌患者年龄、性别、胃癌大小均无明显相关(P > 0.05),而与人胃癌发生部位、分化程度和有无淋巴结转移、侵袭程度有关(P < 0.05)。HPA、N-cadherin和vimentin蛋白在贲门部阳性表达率明显高于胃体、幽门及胃窦胃癌;低分化者、有淋巴结转移者和胃癌侵袭深处这3种蛋白阳性表达率明显高于高分化者、无淋巴结转移者和胃癌起始部位;E-cadherin蛋白在幽门及胃窦部胃癌阳性表达率明显高于贲门和胃体胃癌;高中分化者、无淋巴结转移者明显高于低分化者和有淋巴结转移者。E-cadherin蛋白阳性表达率在侵袭深处明显低于起始部位。经Spearman等级相关分析显示,人胃癌组织中HPA与E-cadherin蛋白的阳性表达率呈负相关(r=-0.341,P < 0.05);而与N-cadherin(r=0.366,P < 0.05)和vimentin(r=0.284,P < 0.05)蛋白阳性表达率呈正相关。结论: 人胃癌组织中HPA、N-cadherin和vimentin蛋白在低分化者、有淋巴结转移者和胃癌侵袭深处高表达;E-cadherin蛋白在高中分化胃癌,无淋巴结转移者和起始部位高表达。HPA与N-cadherin、vimentin蛋白阳性表达率呈正相关,HPA与E-cadherin蛋白的阳性表达率呈负相关,可见HPA蛋白与间质标志物N-cadherin和vimentin蛋白表达增强有关,而与上皮标志物E-cadherin蛋白缺失或减少有关,因此HPA可能诱导人胃癌组织发生上皮间质转化。  相似文献
