AIM: To determine the antimicrobial efficacy of ozonated water, gaseous ozone, sodium hypochlorite and chlorhexidine in human root canals infected by Enterococcus faecalis. METHODOLOGY: Thirty human maxillary anterior teeth were prepared and inoculated with E. faecalis for 60 days. Eppendorf tubes were connected to the coronal portion of the teeth. Urethane hoses were attached to the tubes and to the entrance of a peristaltic pump. The exit of the apparatus corresponded to the apical portion of the root canals. The test irrigating solutions were ozonated water, gaseous ozone, 2.5% sodium hypochlorite (NaOCl), 2% chlorhexidine that circulated at a constant flow of 50 mL min(-1) for 20 min. Samples from the root canals were collected and immersed in 7 mL Letheen Broth (LB), followed by incubation at 37 degrees C for 48 h. Bacterial growth was analysed by turbidity of the culture medium and subculture on a specific nutrient broth. A 0.1 mL inoculum obtained from LB was transferred to 7 mL of brain heart infusion and incubated at 37 degrees C for 48 h. Bacterial growth was checked by turbidity of the culture medium carried out in triplicate. RESULTS: No solution used as an irrigant over a 20-min contact time demonstrated an antimicrobial effect against E. faecalis. CONCLUSION: The irrigation of infected human root canals with ozonated water, 2.5% NaOCl, 2% chlorhexidine and the application of gaseous ozone for 20 min was not sufficient to inactivate E. faecalis.  相似文献

Ozone is known to act as a strong antimicrobial agent against bacteria, fungi, and viruses. In the present study, we examined the effect of ozonated water against Enterococcus faecalis and Streptcoccus mutans infections in vitro in bovine dentin. After irrigation with ozonated water, the viability of E. faecalis and S. mutans invading dentinal tubules significantly decreased. Notably, when the specimen was irrigated with sonication, ozonated water had nearly the same antimicrobial activity as 2.5% sodium hypochlorite (NaOCl). We also compared the cytotoxicity against L-929 mouse fibroblasts between ozonated water and NaOCl. The metabolic activity of fibroblasts was high when the cells were treated with ozonated water, whereas that of fibroblasts significantly decreased when the cells were treated with 2.5% NaOCl. These results suggest that ozonated water application may be useful for endodontic therapy.  相似文献

This study evaluated the in vitro antibacterial activity of conventional and experimental endodontic irrigants against Enterococcus faecalis. The following substances were evaluated by direct contact test: 2.5% sodium hypochlorite (NaOCl); 2% chlorhexidine (CHX); 1% peracetic acid. After different contact periods (30 s, 1, 3, and 10 min), a neutralizing agent was applied. Serial 10-fold dilutions were prepared and plated onto tryptic soy agar (TSA) and the number of colony-forming units per milliliter (CFU/mL) was determined. Sterile saline was used as a negative control. Both 2.5% NaOCl and 2% CHX eliminated E. faecalis after 30 s of contact. Peracetic acid reduced the bacterial counts by 86% after 3 min and completely eliminated E. faecalis after 10 min. These results allow us to conclude that 1% peracetic acid is effective against E. faecalis, despite its slower action compared with 2.5% NaOCl and 2% CHX.  相似文献

The effectiveness of 4.0% sodium hypochlorite (NaOCl) used with three irrigation methods in the elimination of Enterococcus faecalis from the root canal was tested in vitro . Root canals contaminated with E. faecalis were treated as follows: (i) irrigation with 2 mL of NaOCl solution and agitation with hand files; (ii) irrigation with 2 mL of NaOCl solution and ultrasonic agitation; (iii) irrigation with NaOCl alternated with hydrogen peroxide. Contaminated canals irrigated with sterile saline solution served as the control. Paper points used to sample bacteria from the root canals were transferred to tubes containing 5 mL of brain heart infusion (BHI) broth. Tubes were incubated and the appearance of broth turbidity was indicative of bacteria remaining in the root canal. There were no statistically significant differences between the experimental groups. However, NaOCl applied by the three methods tested, was significantly more effective than the saline solution (control group) in disinfecting the root canal.  相似文献

The purpose of this investigation was to determine the antimicrobial effect of MTAD as a final irrigant on eight strains of Enterococcus faecalis (E. faecalis) and to measure the minimum inhibitory concentration (MIC) and the minimum lethal concentration (MLC) of MTAD. The roots of 240 extracted human teeth were instrumented using 1.3% NaOCl and 17% EDTA. The roots were divided into eight groups and contaminated with one of eight strains of E. faecalis. After irrigating with 1.3% NaOCl, the root canal and the external surfaces were exposed to MTAD for 5 minutes. Roots or dentin shavings were cultured to determine the growth of E. faecalis. The results showed that this treatment regimen was effective in completely eliminating growth in seven of eight strains of E. faecalis. The MIC/MLC tests showed that MTAD inhibited most strains of E. faecalis growth when diluted 1:8192 times and killed most strains of E. faecalis when diluted 1:512 times.  相似文献

Pulp and periradicular diseases are of microbial origin. To effectively clean the root canal system a disinfecting agent must be able to penetrate into difficult-to-reach areas and kill microorganisms with minimal damage to the host tissues. The purpose of this investigation was to test the ability of a mixture of a tetracycline isomer, an acid, and a detergent (MTAD) to kill Enterococcus faecalis and compare its efficacy to that of sodium hypochlorite (NaOCl) and ethylene diamine tetraacetic acid (EDTA). The zones of inhibition and minimum inhibitory concentrations were measured for these solutions. Measurement of zones of inhibition and determination of the minimum inhibitory concentrations showed that MTAD is as effective as 5.25% NaOCl and significantly more effective than EDTA (p < 0.0001). Furthermore, MTAD is significantly more effective in killing E. faecalis than NaOCl when the solutions are diluted (p < 0.0001). Measurement of the minimum inhibitory concentrations demonstrated that although MTAD is still effective in killing E. faecalis at 200x dilution, NaOCl ceases to exert its antibacterial activity beyond 32x dilution. EDTA did not exhibit any antibacterial activity. Based on the results of this study, it seems that MTAD is an effective solution in eradicating E. faecalis.  相似文献

目的评价次氯酸钠对根管内粪肠球菌的杀菌效果。方法将45个离体前磨牙的感染根管标本分为6组,1、2组用5.25%及2.5%次氯酸钠冲洗,3组用0.9%NaCl冲洗,4、5组在根管预备时辅以5.25%及2.5%次氯酸钠冲洗,6组在根管预备时辅以0.9%Nacl冲洗。冲洗前、冲洗后即刻及冲洗后72h分别取样培养。结果6组根管内的细菌量均显著下降。1、2组差异无统计学意义(P〉0.05),但均好于3组(P〈0.05)。4、5和6组差异均有统计学意义(P〈0.05)。根管冲洗后培养72h均有细菌生长。结论2.5%次氯酸钠基本可达到更高浓度的灭菌效果,但是经过机械预备和次氯酸钠化学消毒后的根管内仍有细菌残留。  相似文献

The aim of this study was to evaluate the antibacterial activity of 2% iodine potassium iodide (IKI) used as a final rinse after the cleaning and shaping procedures in mesial root canals of mandibular molars infected with Enterococcus faecalis. Seventy two mandibular first molars were used. The root canals were infected with Enterococcus faecalisfor 30 days. After the infection procedures, the root canals were cleaned and shaped by using the Pro Taper rotary system and manual files. The teeth were randomly assigned to four experimental groups (N = 18). In group 1, the root canals were irrigated with sterile distilled water (control). In group 2, the root canals were irrigated with 1% Sodium hypochlorite (NaOCl) during instrumentation. In group 3, root canals were irrigated with 1% NaOCl during instrumentation and a five minute final irrigation using 2% IKI. In group 4, the root canals were irrigated with 1% NaOCl during instrumentation and a 15 minutes final irrigation with 2% IKI. Bacteria colony-forming units (CFU) from root canals were semi-quantified and the presence of negative cultures among the groups was compared using Fisher's test (p < 0.05). The order of effectiveness was: 1% NaOCI plus 2% IKl for 15 minutes (95%), 1% NaOCl plus 2% IKl for 5 minutes (44%), 1% NaOCl (17%) and sterile distilled water (0%). Fisher's exact test showed a significant difference among the groups (p < 0.05). It was concluded that under in vitro conditions, IKI was able to eliminate the Enterococcus faecalis from infected dentin significantly in a 15-minute time frame after the cleaning and shaping procedures.  相似文献

Was evaluated the minimum inhibitory concentration (MIC) and the antibacterial effect (AE) of 2.5% NaOCl, 0.2% chlorhexidine gluconate (CHX) and 17% EDTA on Enterococcus faecalis. The antibacterial capacity was assessed by difusion in agar. The AE was evaluated on contaminated root dentin, employing apical and middle portions of human roots, sterilized and contaminated with Enterococcus faecalis, immersed in the irrigation solutions and incubated at 37 degrees C. Viable cells were counted at 0, 4, 8 and 24 hours. MIC: NaOCl and CHX: 0.2%, EDTA below 5%. Diffusion in agar: NaOCl 2.5% = 21 mm. CHX 0.2% = 14 mm. EDTA 17% = 20 mm. Effect on root dentin: NaOCl 2.5%: Enterococcus faecalis was totally inhibited for 24 hours in the apical area, and for 8 hours in the middle area. CHX 0.2% elicited a reduction of more than 5 log CFU and EDTA 17% induced a reduction of more than 3 log CFU at all the time points examined in the apical and middle areas.  相似文献

目的:探讨不同浓度大蒜素对照品二烯丙基化三硫(dially trisulfide,DATS)体外对粪肠球菌生物膜的作用。方法:体外培养形成粪肠球菌生物膜,分别经1280、2560、5120 mg/L浓度的DATS作用后,采用MTT法观察DATS对生物膜粪肠球菌的抑菌率。结果:1280 mg/L DATS对对数期和饥饿期的生物膜粪肠球菌抑菌率约71%,显著高于对稳定期生物膜粪肠球菌的抑菌率58%,差异具有统计学意义(P<0.05),随着药物浓度的增高,抑菌率增高。结论:DATS对生物膜中的粪肠球菌具有明显杀灭或抑制作用。  相似文献

Abstract

Objectives. To evaluate the antimicrobial efficacy of curcumin against Enterococcus faecalis bio?lm formed on tooth substrate in vitro. Sodium hypochlorite (NaOCl) and chlorhexidine (CHX) served as standards for comparison. Materials and methods. Biofilms of E.faecalis were formed on instrumented, extracted human teeth (n = 96). At the end of the 2nd day, 2nd and 8th weeks, specimens were treated for 30 min with one of the test solutions or saline (control) and the surviving colony-forming units (CFU/mL) was recorded. Results were analyzed by Kruskal-Wallis test and Dunnet test for pair-wise comparison with Bonferroni correction (p = 0.05). Results. Only NaOCl showed complete eradication of bacteria at all time periods. In the 2-day and 2nd week biofilms, curcumin and NaOCl showed complete inhibition, which was significantly lower than the CFU recovered in the CHX and saline groups (p < 0.05). In 8 week biofilms, samples treated with curcumin showed 553 ± 137.6 CFU/mL, which was significantly higher than NaOCl (0 CFU/mL), but significantly lower than CHX (2551 ± 129.8) and saline control (1.42 × 1011 ± 2.12 × 1010; p < 0.05). Conclusions. Sodium hypochlorite (3%) showed maximum antibacterial activity against E.faecalis bio?lm formed on the tooth substrate, followed by curcumin and CHX. Considering the potential for undesirable properties of NaOCl, the use of herbal alternatives in endodontics might prove to be advantageous.  相似文献

Given the importance of bacteria in the development of periradicular lesions, the eradication of the root canal infection is paramount in endodontic treatment. This study evaluated the in vitro intracanal bacterial reduction produced by instrumentation and irrigation with 1%, 2.5%, and 5.25% sodium hypochlorite (NaOCl) or saline solution. Root canals inoculated with Enterococcus faecalis were instrumented and irrigated with the solutions tested. Canals were sampled before and after preparation. After serial dilution, samples were plated onto Mitis salivarius agar, and the colony-forming units grown were counted. Inhibitory effects of the three NaOCl solutions on E. faecalis were also evaluated by means of the agar diffusion test. All test solutions significantly reduced the number of bacterial cells in the root canal (p < 0.05). There was no significant difference between the three NaOCl solutions tested (p > 0.05). Nonetheless, all NaOCl solutions were significantly more effective than saline solution in reducing the number of bacterial cells within the root canal (p < 0.05). The three NaOCl concentrations showed large zones of inhibition against E. faecalis. The results of this study suggest that regular exchange and the use of large amounts of irrigant should maintain the antibacterial effectiveness of the NaOCl solution, compensating for the effects of concentration.  相似文献

目的:比较本实验室研制的根管润滑剂与不同浓度次氯酸钠组合用于粪肠球菌感染根管预备冲洗的抗菌效果。方法:将145颗单根管人离体牙消毒,随机选取5颗置于无菌培养基中培养作为阴性对照,其余140颗接种粪肠球菌4周,按照不同的冲洗组合随机分为14组,使用ProTaper器械按照冠根向深入法进行机械预备,分别在预备冲洗前、预备冲洗中和预备冲洗后进行细菌取样培养计数,并进行统计学分析。结果:与双氧水联合生理盐水组以及蒸馏水对照组相比,各处理组在冲洗过程中与冲洗结束后,均不同程度的降低了根管中的细菌的数量,其中含5.25%次氯酸钠的四种组合处理后菌数减少最为明显,较其它各组差异显著。结论:本实验室研制的根管润滑剂与5.25%次氯酸钠组合抗菌效果最佳,与G lyde凝胶配合次氯酸钠组没有显著差异。  相似文献

AIM: The aim of this study was to assess, in vitro, the effectiveness of several concentrations of NaOCl (0.5%, 1%, 2.5%, 4% and 5.25%) and two forms of chlorhexidine gluconate (gel and liquid) in three concentrations (0.2%, 1% and 2%) in the elimination of E. faecalis. METHODOLOGY: A broth dilution test using 24-well cell culture plates was performed and the time taken for the irrigants to kill bacterial cells was recorded. Isolated 24 h colonies of pure cultures of E. faecalis grown on 10% sheep blood plus Brain Heart Infusion (BHI) agar plates were suspended in sterile 0.85% NaCI solution. The cell suspension was adjusted spectrophotometrically to match the turbidity of a McFarland 0.5 scale. One mL of each tested substance was placed on the bottom of wells of 24-well cell culture plates (Corning, NY), including the control group (sterile saline). Six wells were used for each time period and irrigant concentration. Two mL of the bacterial suspension were ultrasonically mixed for 10 s with the irrigants and placed in contact with them for 10, 30, and 45 s; 1, 3, 5, 10, 20, and 30 min; and 1 and 2 h. After each period of time, 1 mL from each well was transferred to tubes containing 2 mL of freshly prepared BHI + neutralizers in order to prevent a residual action of the irrigants. All tubes were incubated at 37 degrees C for 7 days. The tubes considered to have positive growth were those which presented medium turbidity during the incubation period. Data were analysed statistically by the Kruskal-Wallis test. with the level of significance set at P < 0.05. RESULTS: All irrigants were effective in killing E. faecalis. but at different times. Chlorhexidine in the liquid form at all concentrations tested (0.2%, 1% and 2%) and NaOCI (5.25%) were the most effective irrigants. However, the time required by 0.2% chlorhexidine liquid and 2% chlorhexidine gel to promote negative cultures was only 30 s and 1 min, respectively. CONCLUSIONS: Even though all tested irrigants possessed antibacterial activity, the time required to eliminate E. faecalis depended on the concentration and type of irrigant used.  相似文献

The purpose of this investigation was to compare the antimicrobial efficacy of irrigating with 1.3% NaOCl/Biopure MTAD versus irrigation with 5.25% NaOCl/15% EDTA in the apical 5 mm of roots infected with Enterococcus faecalis. Bilaterally matched human teeth were sterilized and inoculated with E. faecalis. After chemomechanical root canal preparation, the root-ends were resected and pulverized in liquid nitrogen to expose E. faecalis in dentinal tubules or other recesses away from the main root canal system. The number of colony forming units (CFU) of E. faecalis per mg was determined from the pulverized root-ends. No significant differences were seen (t = 0.70, p = 0.495) between the number of colony forming units of E. faecalis for teeth irrigated with 5.25% NaOCl/15% EDTA (mean 131 +/- 291 CFU/mg) versus those teeth irrigated with 1.3% NaOCl/Biopure MTAD (mean 187 +/- 237 CFU/mg). This study demonstrated that there is no difference in antimicrobial efficacy for irrigation with 5.25% NaOCl/15% EDTA versus irrigation with 1.3% NaOCl/Biopure MTAD in the apical 5 mm of roots infected with E. faecalis.  相似文献

Objective: The antimicrobial activity of lactic acid (LA) alone or in combination with chlorhexidine (CHX) and cetrimide (CTR) against three Enterococcus faecalis strains, E. faecalis ATCC 29212, E. faecalis EF-D1 and E. faecalis U-1765, one Enterococcus durans strain and one dual-species biofilm was investigated. Study Design: The irrigating solutions tested were 20%, 15%, 10%, 5% and 2.5% LA, alone and in combination with 2% CHX and with 0.2% CTR. The biofilms were grown in the MBECTM high-throughput device for 24 hours and exposed to the solutions for 30 seconds and 1 minute. "Eradication" was defined as 100% bacterial kill. Results: Twenty percent LA eradicated all enterococci biofilms after 30 seconds contact time. The association of LA + 0.2% CTR achieved better results than LA alone, in contrast with the results obtained using LA + 2% CHX. E. durans was eradicated by all the tested solutions at 1 minute. The dual-species biofilm, E. faecalis ATCC 29212 + E. durans, gave intermediate values of the pure cultures. Conclusions: LA is capable of eradicating enterococci biofilm at a concentration of 20%. The combination of lower concentrations with 0.2% CTR achieved eradication after 1 minute.  相似文献

The aim of this study was to evaluate the antimicrobial capacity of sodium hypochlorite (1% and 5%) and chlorhexidine (0.12%, 0.5% and 1%) with or without the addition of organic material (bovine serum albumin, BSA) against some bacterial samples (Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, Porphyromonas gingivalis and Fusobacterium nucleatum) using two activity tests (contact and diffusion agar tests). In the contact test (first model), bacterial samples were kept in contact with each irrigating solution for different time intervals: immediately (t(0)), 5 min (t(5)), 15 min (t(15)) and 30 min (t(30)). The agar diffusion test was the second model used. In half the specimens, 0.5% BSA was added to simulate organic tissue present in the root canal. Bacterial growth was evaluated for each microorganism and activity test. Each test was repeated 10 times. In the contact test, 0.12% chlorhexidine solution (CHX) did not eliminate E. faecalis at any tested time. CHX at 0.5% eliminated all strains except E. faecalis after immediate contact. All strains were eliminated by 1% CHX, 1% NaOCl and 5% NaOCl. BSA did not interfere with the antimicrobial activity of the irrigating solutions. In the agar diffusion test, all solutions exhibited zones of antimicrobial activity; however, BSA interfered with the antimicrobial activity of NaOCl and CHX. Under the condition of the contact test, the 0.12% CHX was ineffective in eliminating E. faecalis, while 0.5% CHX, 1% CHX, 1% NaOCl and 5% NaOCl showed antibacterial effectiveness against all the tested bacterial strains. The addition of an organic load interfered with the accuracy of the agar diffusion test.  相似文献

目的比较超声、Er,Cr:YSGG激光及Er:YAG激光辅助1%次氯酸钠(NaOCl)溶液冲洗对人离体牙根管表面粪肠球菌生物膜及根尖区不同深度牙本质小管内粪肠球菌的杀灭效果。 方法单直根管下颌前磨牙42颗,建立粪肠球菌生物膜感染的根管模型,采用随机字表法随机抽取2颗确定建成粪肠球菌生物膜感染根管模型,剩余40颗牙采用随机数字表法按随机对照原则分为5组,每组8颗。A组:5.25% NaOCl联合17%乙二胺四乙酸(EDTA)注射器冲洗;B组:0.9%氯化钠溶液注射器冲洗;C组:1% NaOCl溶液超声荡洗;D组:1% NaOCl溶液Er:YAG激光活化冲洗;E组:1% NaOCl溶液Er,Cr:YSGG激光活化冲洗。按分组进行处理后取样菌落计数,计算灭菌率。使用SPSS 17.0统计软件对实验数据进行方差齐性及正态性检验比较各组间和组内的灭菌率。 结果(1)组间比较:对根管壁表面,各组冲洗方法灭菌率比较,B组(5.74%)相似文献

Aim To assess the antimicrobial efficacy of aqueous (1.25–20 μg mL?1) and gaseous ozone (1–53 g m?3) as an alternative antiseptic against endodontic pathogens in suspension and a biofilm model. Methodology Enterococcus faecalis, Candida albicans, Peptostreptococcus micros and Pseudomonas aeruginosa were grown in planctonic culture or in mono‐species biofilms in root canals for 3 weeks. Cultures were exposed to ozone, sodium hypochlorite (NaOCl; 5.25%, 2.25%), chlorhexidine digluconate (CHX; 2%), hydrogen peroxide (H2O2; 3%) and phosphate buffered saline (control) for 1 min and the remaining colony forming units counted. Ozone gas was applied to the biofilms in two experimental settings, resembling canal areas either difficult (setting 1) or easy (setting 2) to reach. Time‐course experiments up to 10 min were included. To compare the tested samples, data were analysed by one‐way anova . Results Concentrations of gaseous ozone down to 1 g m?3 almost and aqueous ozone down to 5 μg mL?1 completely eliminated the suspended microorganisms as did NaOCl and CHX. Hydrogen peroxide and lower aqueous ozone concentrations were less effective. Aqueous and gaseous ozone were dose‐ and strain‐dependently effective against the biofilm microorganisms. Total elimination was achieved by high‐concentrated ozone gas (setting 2) and by NaOCl after 1 min or a lower gas concentration (4 g m?3) after at least 2.5 min. High‐concentrated aqueous ozone (20 μg mL?1) and CHX almost completely eliminated the biofilm cells, whilst H2O2 was less effective. Conclusion High‐concentrated gaseous and aqueous ozone was dose‐, strain‐ and time‐dependently effective against the tested microorganisms in suspension and the biofilm test model.  相似文献

The aim of this in vitro study was to compare the antimicrobial substantivity of 2% chlorhexidine gluconate (CHX), 100 mg ml(-1) doxycycline and 2.6% sodium hypochlorite (NaOCl) in bovine root dentine. Eighty dentine tubes prepared from bovine incisors were infected in vitro for 14 days with Enterococcus faecalis. The specimens were divided into five groups as follows: doxycycline HCl; CHX; NaOCl; infected dentine tubes (positive control); and sterile dentine tubes (negative control). Dentine chips were collected with round burs into tryptic soy broth. After culturing, the number of colony-forming units (CFU) was counted. In all experimental groups, the number of CFU was minimum in the first cultures, and the results obtained were significantly different from each other at any time period (P < 0.05). In the first culture, the NaOCl group and doxycycline HCl group showed the lowest and highest number of CFU, respectively. In each group, the number of CFU increased significantly by time-lapse (P < 0.05). In conclusion, the substantivity of CHX was significantly greater than NaOCl and doxycycline.  相似文献
