目的研究TGF-β1和/或TNF-α反义硫代寡核苷酸(PS-ODNS)对造血干/祖细胞体外扩增的调节作用.方法采用免疫磁珠分离技术从新鲜脐带血中分离CD34+细胞,用淋巴细胞分离液从骨髓血中分离单个核细胞,应用液体培养及造血祖细胞集落分析检测TGF-β1和/或TNF-α反义PS-ODNS对CD34+细胞数及多向性造血祖细胞(CFU-mix)、粒-单祖细胞(CFU-GM)、红系祖细胞(BFU-E和CFU-E)集落数的影响.结果培养体系中加入TGF-β1反义PS-ODNS后CD34+细胞数、CFU-mix、CFU-GM、BFU-E和CFU-E集落数分别是对照组(仅加细胞因子组)的4、2.6、2.7、1.8、2.1倍;加入TNF-α反义PS-ODNS后CD34+细胞数、CFU-mix、CFU-GM、BFU-E和CFU-E集落数分别是对照组的4、2.9、2.6、1.7、1.8倍;同时加入TGF-β1和TNF-α反义PS-ODNS后CD34+细胞数、CFU-mix、CFU-GM、BFU-E和CFU-E集落数分别是对照组的5.3、2.1、2.7、1.9、1.8倍.结论采用反义PS-ODNS阻断内源性TGF-β1和TNF-α是造血干/祖细胞体外扩增的有效措施之一.  相似文献

血管内皮生长因子(vascularendothelial growth factor,VEGF)是内皮细胞特异性的生长因子,大多数关于VEGF的研究都是致力于其在血管生长方面的作用,而近年来有大量文献报道VEGF具有神经营养和促神经发生作用,它能够直接作用于神经元细胞和神经胶质细胞甚至是神经干细胞,促进其生长及存活。VEGF的多种功能使其和多种神经退行性疾病相关,如阿茨海默病,肌萎缩侧索硬化症,帕金森病等。导入VEGF基因能够改善肌萎缩侧索硬化症、帕金森病动物模型的病情。  相似文献

Curcumin (diferuloylmethane), found in the spice turmeric, exhibits anti-inflammatory, antioxidant, and chemopreventive activities. However, the effect of curcumin on the immunological responses largely remains unknown. In this study we have investigated the effect of curcumin on mitogen (phytohaemagglutinin; PHA) stimulated T-cell proliferation, natural killer (NK) cell cytotoxicity, production of cytokines by human peripheral blood mononuclear cells (PBMCs), nitric oxide (NO) production in mouse macrophage cells, RAW-264.7. Furthermore, we have carried out an electromobility shift assay to elucidate the mechanism of action of curcumin at DNA protein interaction level. We observed that curcumin inhibits PHA-induced T-cell proliferation, interleukin-2 production, NO generation, and lipopolysachharide-induced nuclear factor-κB (NF-κB) and augments NK cell cytotoxicity. Our results suggest that curcumin most likely inhibits cell proliferation and cytokine production by inhibiting NF-κB target genes involved in the induction of these immune parameters.  相似文献

目的 研究雌二醇(estradiol17β,E2)对颌下腺促性腺激素释放激素(gonadotropin relesaing hormoe GnRH)和生长激素(growth factor,GH)生成的影响。方法 采用免疫组织化学的ABC法。结果 雌二醇促进颌下腺GnRH的生成而对GH的产生有抑制作用。结论 雌二醇可能对颌下腺GnRH和GH的合成起重要的调节作用。  相似文献

Here we review the results of experimental and clinical observations concerning the gonadotoxic effect of antitumor chemotherapy. Previous experiments showed that antiblastic preparations with various mechanisms of cytostatic action produce damage to the sex glands that differs in the severity, stage of manifestation, and period of reparative regeneration. These differences are related to different sensitivity of epitheliocytes in the testes and structural and functional elements in the ovaries. Morphological changes in the gonads are accompanied by dysfunction of the reproductive system. We compared damages to the reproductive system and its individual components produced by various antitumor preparations. The offspring was examined after cytostatic treatment of one of the parent animals.  相似文献

The most easily discernible and characteristic result of interaction of LPS with the immune system are the activation of B cells to divide and/or synthesize and secrete immunoglobulin. LPS as an antigen or carrier for chemically defined haptens induces a T independent antibody response, while antibody responses to unrelated T dependent antigens are modulated by LPS. Such regulation can be explained both by increased susceptibility of B cells to regulation after interaction with LPS, and increased activity of regulatory cells after their interaction with LPS. Studies of these mechanisms have exploited the availability of several strains of mice which are genetically poorly responsive to LPS and thus have helped to define the genetic basis of LPS responses. In addition, these studies have demonstrated the existence of other bacterial cell wall components which have some of the immunological properties of LPS but are not susceptible to the same genetic control. Future studies on the molecular nature of the interaction of LPS with target cells and the biochemical basis of cell triggering should yield interesting information on the cell biology of cell activation.  相似文献

瘦素是重要的脂肪细胞因子。近年发现瘦素及其受体参与了胰岛素抵抗(IR)。瘦素本身可造成脂肪组织的IR,减少脂肪合成,增加脂肪分解,从而引起肝脏和肌肉的IR,使胰岛素依赖的糖处理效率受到削弱,与2型糖尿病的形成与发展密切相关。  相似文献

目的:检测血小板生成素(TPO)及TPOⅡ在体外对人血小板活化的影响。方法:取正常健康成人富血小板血浆,分别与血小板生成素(rhTPO)、TPOⅡ及磷酸盐溶液孵化,通过荧光单克隆抗体标记血小板表面CD62P及CD41分子,用流式细胞术测定CD62P/CD41比率,观察各组血小板活化率。结果:rhTPO、TPOⅡ组血小板活化率与PBS组无明显差异,P>0.05。结论:血小板生成素不会导致血小板异常活化,可应用于血小板生成减少的各种情况。  相似文献
