目的从EST鉴定入手利用生物信息学方法筛选验证PCBP2基因内部UC.338基因座位附近的长非编码RNA。方法首先利用生物信息学方法筛选包含内含子转录区域的ESTs,经CPC分析和统计学比较,共筛选到4个(人、鼠各两个)可能的lncRNAs,用PCR方法进行鉴定,克隆到pGEM-T载体测序验证,并用PCR,real-time PCR的方法检测各ESTs在人源细胞系或小鼠不同组织,小鼠发育不同时间点大脑中的表达谱。结果筛选到4个ESTs中有3个ESTs均具有一定的非编码特征,并最终测序正确:1个人源,2个鼠源,并且其中有些lncRNAs具有一定的细胞、组织特异性,有些lncRNAs具有广谱表达模式。结论使用了一种新的寻找lncRNAs的方法,从EST鉴定入手,对现有数据库进行了挖掘。找到了可能在小鼠大脑发育过程中起作用的lncRNAs。  相似文献

It was previously reported that several kinds of intercellular adhesion molecules are closely related to chronic HBV infection. The complex of CD2 and CD58 plays an important role in enhancing the adhesion of T lymphocytes to target cells, and promoting hyperplasia and activation of T lymphocytes. In this study, we detected the level of CD2 expressed on the surface of PBMC, the expression level of CD2 mRNA in PBMC and the percentage of CD2 positive cells in PBMC of patients with chronic HBV infection and compared them with the expression level of normal controls. We also determined the level of serum HBV DNA from patients with chronic HBV infection and from normal controls. The clinical characteristics of hepatic function were tested as well. The results showed that the expression of CD2 significantly increased with the severity of chronic HBV infection, which suggested that CD2 might contribute to the hepatocyte damage in chronic HBV infection. 目的探讨长链非编码RNA Linc00467在结直肠癌中的表达和意义。方法实时荧光定量PCR检测Linc00467在结直肠癌组织和结肠癌细胞系中的表达水平。通过Transwell迁移实验、MTS实验和克隆形成实验检测Linc00467对结肠癌细胞迁移、增殖等行为的影响。统计纳入的67例患者的临床病理资料,分析Linc00467的表达水平和患者临床病理学特征之间的关系。结果 Linc00467在结直肠癌组织和细胞系中高表达。敲低Linc00467可以抑制结肠癌细胞的迁移和增殖。Linc00467的表达丰度与结直肠癌患者的临床病理特征之间无显著相关性。结论 Linc00467在结直肠癌中高表达,可以促进结肠癌的迁移和增殖。  相似文献

Epstein-Barr virus (EBV) is a prevalent γ-herpesvirus associated with a variety of cancers, including epithelial nasopharyngeal carcinoma (NPC) and gastric carcinoma (GC). Long noncoding RNAs (lncRNAs) are emerging as powerful regulators that have demonstrated to play crucial roles in cancer development. In this approach, based on genome-wide RNA sequencing, we examined the lncRNA expression profiles in four EBV genome-infected and EBV-negative 293 cells. A series of lncRNAs were found to be dysregulated in a comparative analysis. Then, eight typical lncRNAs were selected for validation of expression levels by real-time quantitative polymerase chain reaction and were detected in EBV-positive or EBV-negative GC and NPC cells. The differential expression patterns of the eight lncRNAs for validation were fundamentally identical to those revealed in RNA sequencing. Particularly, the differential expression of these lncRNAs in GC and NPC cells indicated their possible roles in EBV infection and tumorigenesis. In addition, a predicted lncRNA-microRNA-messenger RNA network suggested their potential interactions. This study reveals the first lncRNAome related to EBV infection in the epithelial cells and provides novel clues for the study of viral role in epithelial cancers through the interaction between EBV and host lncRNAs.  相似文献

长链非编码RNA(lncRNA)是指长度超过200个核苷酸的非编码RNA,其通过与DNA、RNA和蛋白质等分子相互作用,从表观遗传学、转录水平以及转录后水平等3个层面发挥重要调控作用.近年研究表明,lncRNAs在许多肿瘤的发生、发展过程中发挥着促癌或抑癌作用,部分lncRNAs因其特殊的生物学特性有希望成为新型肿瘤标记物和肿瘤治疗的新靶点.本文总结了近年来在肿瘤中低表达或呈表达缺失的lncRNAs,探讨其与肿瘤发生的相关机制,评价其在肿瘤诊断和治疗方面的临床应用前景.  相似文献

Sera of 1 55 chronic hepatitis (CH) patients in Argentina were tested for the presence of HBsAg, anti-HBs, anti-HBc, and anti-HAV. Our purpose was to define the role that both virus A and B might play in the etiology and pathogenesis of this condition. The patients were divided into two groups: group I (57) HBsAg-negative; group II (98) HBsAg-positive. The control group consisted of 1,209 healthy blood donors from Banco Central de Sangre de Rosario; 286/ 1,209 (24%) had viral markers for HBV. In group I, 38/57 (67%) had anti-HBs and/or anti-HBc, but none had anti-HBs alone. Group II showed a higher percentage of males (P < 0.05). We found similar incidence of anti-HAV among group I, group II, and the control group.  相似文献

Due to a lack of standardized tests, it is difficult to obtain prevalence data and define the real impact of occult HBV infection (OBI) in Western countries. The present review article addresses the prevalence of OBI, defined as presence of hepatitis B virus (HBV) DNA in liver tissue or plasma in HBsAg-negative subjects, in Western countries. This varies in different studies according to the different methodologies used (based on serology vs virology), to the sample analyzed for the diagnosis (liver tissue vs plasma), to the different populations studied, to the different geographical variations in the HBV spread, to the host characteristics (age, gender, risk factors for acquiring HBV infection) and to the presence of other parenteral infections (hepatitis C virus and/or human immunodeficiency virus [HIV] infections). Considering the different liver diseases analyzed, that is in patients with cryptogenic cirrhosis or advanced liver fibrosis, the prevalence of OBI ranges 4% to 38%. Considering the different populations studied, in the case of parenteral blood exposure it is about 45%, in patients with chronic hepatitis C it is estimated at about 52%, in HIV-infected patients it ranges from 0% to 45%, in blood donors from 0% to 22.7% and in hemodialysis patients it ranges from 0% to 54%. In conclusion, OBI is a virological entity to be considered when performing the patient's evaluation for immunosuppressive diseases, liver pathologies, or for blood transfusions. Knowing the prevalence and clinical impact of OBI will allow better patient management.  相似文献

Hepatitis B virus (HBV) is a virus that infects about 350,000,000 people worldwide with a clinical spectrum of acute hepatitis, the healthy carrier state, cirrhosis and hepatocellular carcinoma (HCC). The outcome of HBV infection is the result of complicated viral-host interactions. As in other infections with non-cythopatic viruses, the immune response is thought to play a crucial role in disease pathogenesis but there is increasing evidence that a variety of viral mechanisms, some depending on the function of virally encoded proteins, have a profound impact on the infected hepatocytes, the liver microenvironment, and host anti-viral responses. Indeed, the virus has evolved multiple mechanisms to ensure its success in infecting a susceptible host. The essential aspects of the life cycle of HBV and the host immune response are reviewed and recent new developments in the molecular virology of HBV, including experimental animal models, in the role of accessory viral proteins in disease pathogenesis and HCC development and in the characterisation of the T cell response in the control of HBV infection, are highlighted.  相似文献
