目的了解保山市献血员中主要民族的ABO、Rh血型分布,建立本地稀有血型献血员档案,满足无偿献血员的招募和临床用血需要。方法用微板法对ABO、Rh血型进行检测。抽取124名RhD阴性者,用试管法对C/c、E/e进行筛选。结果 7个主要献血民族中,汉、彝、白、傈僳、回族ABO血型分布为A>O>B>AB;布朗、傣族为O>B>A>AB;Rh阴性比率:汉族0.33%,彝族0.35%,白族0.09%,布朗族0.00%,傈僳族0.32%,傣族0.21%,回族0.18%;Rh表型主要有ccdee(60.5%)、Ccdee(33.1%)、CCdee(4.0%)、ccdEe(2.4%),没有发现其他表型。结论保山市汉、彝、白、傈僳、回族献血员ABO血型分布率与中国汉族人群的分布相符;布朗、傣族则与其不同;Rh阴性频率处于0.00%~0.35%之间,体现了血型分布在本市的地域和民族特点。  相似文献

目的探讨1例罕见AB3亚型的分子机制。方法对1例ABO血型正反定型不符患者的血型通过血清学鉴定、PCR-SSP、ABO基因直接测序、TA克隆单体型分析等方法分析其分子机制。结果该患者血型鉴定为比较少见的A102/B301亚型,该亚型是由于ABO基因第七外显子存在1054C/T杂合导致R352W氨基酸改变所致。结论 ABO基因存在1054CT,该突变可能是导致B301亚型的分子遗传基础之一。关健词:  相似文献

目的探讨尿道下裂与ABO血型之间的相关性。方法对1997年1月1日至2008年9月1日中山大学第二附属医院收治的633例尿道下裂患者血型进行回顾性分析,并与健康男性进行对照,两组均采用试管凝集法检测其ABO血型,统计出两组的ABO血型分布、频率及相对危险率,用卡方(χ2)检验进行统计学处理。结果尿道下裂患者血型比例分别为A型27.33%、B型28.44%、AB型6.63%、O型37.60%,ABO血型分布为OBAAB,与对照组ABO血型分布特征差异有统计学意义(P=0.0020.05),经分割χ2检验发现患者组和对照组两组人群的O型血的频率分布差异有统计学意义(P=0.0040.0167);相对危险率统计分析结果显示,O型血是尿道下裂的易感因素,与其他几种血型比较差异有统计学意义(P0.001)。结论尿道下裂的发生与ABO血型存在一定的相关性,O型者可能易患尿道下裂。  相似文献

目的研究ABO血型B亚型系统Bw亚型与B糖基转移酶的关系。方法运用血型血清学鉴定方法鉴定1个家庭2例ABO血型的Bw亚型,运用聚合酶链式反应的方法扩增糖基转移酶1~7号外显子,送到试剂公司测序。结果直接测序发现2例ABO血型的Bw亚型,其中B糖基转移酶基因第721位C〉T的转变,导致糖基转移酶多肽链Arg241Trp的转变。结论糖基转移酶基因第721位的C〉T的突变引起糖基转移酶活性的消失或者减弱,导致Bw亚型。  相似文献

目的评估婴儿ABO血型的鉴定方法,找出一种更适宜的鉴定方法。方法采用微柱凝胶血型卡、抗人球蛋白卡和试管法对本院2009年10月~2010年1月60名婴儿全血标本做ABO血型鉴定,同时比较60名标本抗体检出率在血型和年龄分布中的差异。结果用微柱凝胶血型卡、抗人球蛋白卡和试管法鉴定时,正定型结果一致,反定型时,抗体检出率分别为70%、91.67%、81.67%,三种方法在婴儿抗体检出率中差异有统计学意义;在血型为A型、年龄小于4个月的婴儿中抗体检出率差异也有统计学意义。结论抗人球蛋白卡在检测弱抗体时灵敏度更高。  相似文献

[目的]了解学生群体中的ABO血型分布情况,有利于临床医务人员及相关工作人员掌握血源概况,为血液血型贮备对策提供科学依据,并分析造成批量ABO血型错检的原因。[方法]运用玻片凝集法和试管离心法对8860名学生进行ABO血型鉴定,对可疑血液样本做盐水试管法的正反定型试验。[结果]血型分布为A型占27.75%,B型占31.65%,O型占29.65%,AB型占10.95%。[结论]8860名学生血型分布为B〉O〉A〉AB。当发现血型鉴定出现可疑时,首先应重复做试验一次,严格执行操作规程,细心观察结果,按步骤一步步排除错检的可能性。  相似文献

目的了解云南红河哈尼族无偿献血人群ABO及Rh血型分布规律以及基因频率。方法采用血站ABO血型正反2次定型及Rh(D)筛查及鉴定程序,对云南红河哈尼族6 817例无偿献血人群进行血型鉴定。结果云南红河哈尼族A型、B型、O型及AB型构成比分别为25.08%、27.92%、39.81%及7.19%;期望值构成比为25.19%、28.08%、40.02%及6.73%,基因频率P=0.1749,q=0.1925,r=0.6326,观察值与期望值无显著差异(P0.05),分布符合Hardy-Weinberg平衡。ABO血型分布为O型B型A型AB型。6 817例云南红河哈尼族共检出Rh D(-)23例,占0.37%,其中CCdee为21.74%,Ccdee为47.83%,ccdee为30.43%,其期望值构成比为20.87%、49.57%及29.57%,基因频率Cde为0.4566,cde为0.5434,观察值与期望值无显著差异(P0.05),分布符合Hardy-Weinberg平衡。结论对哈尼族无偿献血人群ABO及Rh血型分布规律以及基因频率的研究对本地区采供血计划的制定及安全用血均有重要的指导意义。  相似文献

目的研究ABO血型A201等位基因表达A抗原活性。方法运用血型血清学方法对1家2代3人的唾液及血液标本的ABO血型进行检测;运用Identifiler法医基因分析试剂盒作为亲缘关系鉴定;PCR扩增ABO基因后,对所有外显子做DNA测序分析,运用PCR-RFLP法进行同步检测A201等位基因中的序列变异。结果亲缘关系鉴定在Identifiler的15个遗传标记系统中显示,父母可以将所有的等位基因提供给子女,确定具有亲缘关系;然而运用不同来源的3份抗体对ABO血型进行检验时,却出现了正定型否定了母子关系,反定型无法排除母子关系的现象。3个标本ABO基因的测序:父、母、子的基因型分别为O01/O01、A201/B101及A201/O01,符合反定型结果,这就说明A201等位基因在A表型及AB表型中表达的A抗原活性差异显著。运用PCR-RFLP法可将A201等位基因中的2个主要变异点1059-1061del C和467C/T。结论 ABO血型A201基因在AB及A表型中表达A抗原活性,可由于检测抗体不同出现一定的差异;基因测序与PCR-RFLP法进行同步分析能够快速简便的鉴定A201等位基因中的1059-1061del C和467C/T。  相似文献

Background

Toxoplasmosis is a cosmopolitan parasitic disease caused by Toxoplasma gondii (T. gondii). Blood transfusion is a probable route of T. gondii transmission. Due to lack of information about seroprevalence of T. gondii in healthy blood donors, this study was aimed to determine the chronic and acute infection using serological and molecular methods.

Material and methods

In this cross-sectional investigation, 380 samples were collected from donated bloods. Anti-Toxoplasma IgG and IgM antibodies were examined using enzyme-linked immunosorbent assay (ELISA). Also, all IgG positive samples were tested by IgG avidity test. Eventually, to detection of active infection, DNA was extracted from IgM positive and low IgG avidity samples and then tested using nested-polymerase chain reaction (PCR).

Results

Among 380 blood donors, 131 (34.47%) were positive for only anti-T. gondii IgG, 2 (0.5%) were positive for only anti-T. gondii IgM, and 11 (2.9%) were positive for both IgG and IgM antibodies. Then, 142 samples (131 IgG + and 11 IgG +IgM +) were evaluated using IgG avidity test. Of these, 115 (81%) had high avidity IgG indicates past infection; 16 (11.26%) had low avidity IgG representing recent infection, and 11 (7.74%) were equivocal. With nested PCR, 20 samples of 50 seropositive samples were diagnosed positive.

Conclusion

Detected active infection using nested-PCR draws attention to the possibility of T. gondii infection via blood transfusion which emphasizes the importance of parasite DNA screening before donation of blood in high risk groups such as: multi-transfused persons, immunosuppressed patient, and pregnant women.  相似文献

目的探讨Beckman Coulter PK7300全自动血型分析仪与微板法在ABO血型筛查中的应用效果。方法选取2016年1-5月无偿献血者EDTA抗凝全血标本12 000例,采用Beckman Coulter Pk7300全自动血型分析仪和STAR加样微板手工比色法分别进行检测分析。结果两种方法检测ABO血型的准确率差异无统计学意义(P0.05)。在ABO血型筛查中,Beckman Coulter Pk7300全自动血型分析仪ABO亚型和抗体减弱的检出率均高于微板法。3例ABO血型正反定型在Beckman Coulter Pk7300全自动血型分析仪检测中一致而在微板法中不一致,4例ABO血型正反定型在Beckman Coulter Pk7300全自动血型分析仪检测中不一致而在微板法中一致。结论 Beckman Coulter Pk7300全自动血型分析仪能够安全有效对献血者进行ABO血型筛查,检测结果可疑的标本仍需结合试管法进行人工判读。  相似文献

目的:了解茂名市无偿献血者 HCV 感染人群特征,为无偿献血者招募和 HCV 感染防控提供依据。方法选取茂名市中心血站无偿献血者标本,采用不同厂家试剂、不同操作人员进行 HCV 抗体检测。对初次检测具反应性标本采用双孔进行复试确认,对检测结果进行统计分析。结果茂名地区无偿献血者 HCV 感染率为0.74%;女性感染率略高于男性,但差异无统计学意义(P >0.05);公务员及医务人员感染率相对较低;重复献血者 HCV 感染率明显低于首次献血者,差异有统计学意义(P <0.05)。结论茂名地区无偿献血人群 HCV 感染率处于较低水平,符合献血条件的小于35岁人群 HCV 感染风险较低,建立一支固定的无偿血者队伍,有利于保障临床用血安全。宜加强对于公务员和医务人员之外的群体进行 HCV 知识的宣教和防控,有效控制人群 HCV 感染。  相似文献

目的:探讨核酸检测(NAT)在血液筛查中的作用,为选择血液筛查策略提供科学依据。方法该中心在2012年1月至2013年4月对68662例无偿献血标本同步进行血清学和核酸检测;29例血清学阴性 HBV DNA 阳性样本进行乙型肝炎血清标志物测试;部分抗 HIV 阳性样本送疾控中心做确认实验。结果68662例标本中核酸单独阳性样本120例,输血残余风险为0.175%;乙型肝炎血清标志物检测,以 HBcAb 阳性模式居多;HIV 确认阳性11例,全部为核酸阳性标本。结论核酸检测能够降低输血残余风险,保证输血安全。NAT 和血清学检测相互补充,不能替代,应选择适合的筛查策略。  相似文献

目的 探讨幽门螺杆菌(Hp)感染病人中Hp细胞毒相关蛋白(CagA)、细胞空泡毒素(VacA)抗体在胃十二指肠疾病的比例,并评估不同类型幽门螺杆菌感染对胃窦粘膜炎症程度的影响。方法 应用免疫印迹法测定Hp感染者病人体内的CagA、VacA抗体。结果 73%的Hp感染者病人中出现CagA抗体,61%Hp感染者病人中出现VacA抗体。CagA抗体和VacA抗体在各疾病之间差异无显著性(P>0.05)。在胃溃疡、十二指肠溃疡和复合性溃疡三类病人中,具CagA和VacA双阳性的比例较CagA和VacA双阴性的比例差异有显著性(P<0.05)。胃窦萎缩病变在CagA和VacA双阳性的病人中比CagA和VacA双阴性的病人严重(P<0.05)。结论 具有高毒力因子(CagA和VacA)的Hp是胃肠疾病病人感染的常见菌株,具有CagA和VacA的Hp可能在诱导胃粘膜发展到萎缩病变过程中起重要作用。但不能作为特异性指标来判断胃十二指肠疾病。  相似文献

Since 1901, more than 20 distinct blood group systems have been identified and characterized. Yet, the ABO System, the first described, remains the most clinically significant in blood transfusion and organ transplantation medicine. The ABO antigens are the only cellular antigens which consistently produce a potent, naturally occurring antithetical antibody which circulates in the plasma of healthy individuals. ABO antigens are expressed on most blood cells, organs, and tissues and in most body fluids. Expression of the antigens results from the interaction of several separate, but closely related genes: ABO. H. and Secretor (Se). Blood group specificity depends upon the inheritance of the ABO and H genes, and the subsequent expression of these antigens on the red blood cells. The “D” or Rh0 antigen is the most clinically significant blood group antigen next to the ABO antigens. The D antigen belongs to the Rh System, which consists of a trio of genes that are so closely linked that they are inherited as a package. The antigens are an integral part of the red blood cell membrane. They lend stability to the membrane structure when they are normal, but result in decreased red cell survival when absent. Rh antibodies are immune antibodies requiring a stimulus and can cause significant transfusion and childbearing complications if present. © 1992 Wiley-Liss, Inc.  相似文献

This study aimed to characterize anti-A and anti-B hemolysins, IgM, and IgG titers in Thai blood donors. Altogether, 300 serum samples from group O donors at the National Blood Centre, Thai Red Cross Society, were screened for anti-A and anti-B hemolysins and treated with 0.01 M dithiothreitol to characterize IgM and IgG titers by standard tube technique. Antibody titers were compared with hemolysis grade. Male and female ratio = 1:1.3 and ages ranged from 17 to 60 years. The overall prevalence of anti-A and anti-B hemolysins was 69%. Anti-A and anti-B hemolysins comprised 18.3% and 16.7%, respectively and 34% had both antibodies. High titers of anti-A hemolysins were associated with females (P< 0.05), and only anti-B IgM titers were associated with age (P< 0.05). Interestingly, the association of anti-A IgM titers, anti-A IgG titers, and hemolysin grade was demonstrated (P< 0.05). A significant association between hemolysin grade and anti-B IgM titers was found (P< 0.05). The prevalence of anti-A and anti-B hemolysins and high titers of IgM and IgG in Thais are high. Hemolysin grade showed significant associations with IgM titers; therefore, when providing ABO-incompatible platelet transfusion, especially for female plateletpheresis donors, IgM high titers of anti-A and anti-B screening is suggested.  相似文献

目的 了解揭阳市无偿献血者人类免疫缺陷病毒(HIV)的感染情况,以确定低危献血人群,确保血液安全.方法 采用两种不同厂家ELISA试剂,对2007年1月至2010年12月87 454例献血者血液做抗-HIV抗体初筛,对首次呈反应性的标本再重新留取血袋辫子标本与原试管标本进行双孔复试,仍呈反应性的标本送揭阳市疾病控制中心进行确证.结果 抗-HIV抗体初筛阳性共计100例,确证阳性11例,不确定8例,其余确证为阴性.2007~2010年抗-HIV抗体确证阳性数依次为1、2、4、4例.结论 揭阳市无偿献血人群中HIV感染呈逐年上升趋势.  相似文献

目的 探讨ABO血型基因分型技术应用于临床检验的可行性.方法 随机抽样100例江西健康汉族个体,并收集20例正反定型不符的标本,采用血清学和序列特异性引物PCR法(PCR-SSP)鉴定血型.结果 100例健康个体的基因分型结果与血清学分型结果一致,19例正反定型不符标本的基因分型结果准确.分型结果表明江西地区以O型为多,100例健康个体中,O型36例(36%),A型32例(32%),B型24例(24%),AB型8例(8%),基因频率:p(A)基因为0.225 3,q(B)基因为0.175 3,r(O)基因为0.599 8,且符合Hardy-Weinberg平衡.结论 基因分型技术应用于临床检验具有可行性,在鉴定疑难血型方面可以作为血清学分型的补充.  相似文献

The goal of this project was to find and collect high concentrations of endotoxin-specific antibodies for therapeutic IgG- or IgM-enriched preparations. Various enzyme-linked immunosorbent assays (ELISAs) were developed to perform longitudinal studies of the serological response to a large panel of smooth and rough purified lipopolysaccharide (LPS) extracts in a population of healthy blood donors. To accomplish this, 1612 human serum samples from volunteer blood donors collected by seven different blood banks in Belgium were screened and specific IgM and IgG activities were measured. Approximately 17% of the donors had anti-LPS concentrations higher than 40 mg L−1. Of these, 10.9% had anti-smooth LPS antibodies, 3.7% had anti-rough LPS antibodies and 2.8% were found to be positive towards both types of LPS. The mean anti-LPS antibody concentration was 8 mg L−1 for rough LPS and 14 mg L−1 for smooth LPS. Age- and sex-related distributions of the activities indicated that the greatest prevalence of high anti-LPS concentration was in women aged 40–49 years and in men older than 60 years. Differential absorption experiments showed that the pooled serum of selected blood donors contained a mixture of specific and cross-reacting antibodies. We detected predominantly anti-LPS activities due to the IgG1 and IgG2 subclasses. The range of specificities to different LPS was increased by the pooling of selected sera. It was concluded that pools of naturally occurring specific anti-LPS immunoglobulin antibodies may be obtained in Belgium by screening blood donors using ELISAs that we have developed.  相似文献
