FoxP3+ regulatory T (Treg) cells comprise a highly dynamic population that restrains autoreactivity. Although complete or long‐term depletion of Foxp3+CD4+ Treg cells in adult mice has been shown to result in chronic inflammation and autoimmune disease, the impact of transient Treg‐cell depletion on self‐reactive responses is poorly defined. A new study published in this issue of the European Journal of Immunology [Eur. J. Immunol. 2014. 44: 3621–3631] shows that, although transient depletion of Treg cells in mice is swiftly followed by recovery of Treg‐cell numbers, the “rebounded” population fails to maintain tolerance, culminating in severe autoimmune gastritis. This commentary explores new questions about the quantitative and qualitative aspects of Treg‐cell function in immunological tolerance raised by this study and others.  相似文献

In vitro cell loading experiments are used to investigate stimulation of strain to cellular proliferation. As the flowing conditions of culture fluid in loading systems has been little known, strictly people can not detect the influence of strain to cellular proliferation exactly because shear flow can enhance cell proliferation either. Based on the working principle and cyclic loading parameters, we simplify Navier-Stokes equation to describe the flow of culture fluid on substrates of uniaxial and equi-biaxial fiat tensile loading systems and four point bending system. With approximate solutions, the distributions of velocity field and wall shear flow to cells are gained. Results show: shear flows are zero in the middle (or fixed point or line) of substrate for all systems, and they get larger proportionally to distance from middle and substrate elongate; the shear flow on the substrate of four point bending system is much greater than those of others. This shear flow in four point bending system, confirmed by Owan, I., et al with OPN mRNA increase in their experiment, could cause more influence to osteoblast-like cells than that caused by strain. We estimate the average magnitude of shear stress in Owan‘ s device, the results are consistent with other experimental data about shear flow. In conclusion our study makes it possible to differentiate the influence of strain on cellular proliferation to that of shear flow in loading experiments with the devices mentioned above quantitatively.  相似文献

Anatomical professionals know better than anyone else that donated bodies are a valuable asset to anatomical science and medical education. They highly value voluntary donations, since a dearth of bodies negatively affects their profession. With this in mind, we conducted a survey (n = 54) at the 171st scientific meeting of the Dutch Anatomical Society in 2009 to see to what extent anatomical professionals are willing to donate their own body. The results reveal that none of the survey participants are registered as a whole body donor and that only a quarter of them would consider the possibility of body donation. We argue that the two main constraints preventing Dutch anatomical professionals from donating their own body are their professional and their social environments. In contrast to the absence of registered body donors, half of the anatomical professionals are registered as an organ donor. This figure far exceeds the proportion of registered organ donors among the general Dutch population.  相似文献

Surface modification of biomedical polyurethane material by means of glowdischarge plasma was investigated in this study. After exposing the material to N. or N./H. plasma for thirty minutes, the hemocompatibilily of the material is significanlly improved according to the in-vitro evaluation presented in the article. The interracial free energy of the surface with water after modification enters the range of one to three dyn/cm after plasma being treated under the optimal technical parameters presented. The haemolytic percentage of the treated polyurethane is less than 5%, indicating that the treated material is a satisfactory candidate for clinical uses.  相似文献

Heavy metals have been shown to exert immunotoxic effects on humoral immunity. To ascertain the mechanisms by which these immunotoxic effects are exerted, the effects of CdCl2 and HgCl2 on the biology of murine B-lymphocytes were studied. It was shown that CdCl2 aand HgCl2 inhibited B-cell RNA and DNA synthesis. The IC50 (the concentration required to inhibit a specific B-cell function by 50%) for CdCl2 was 30 μM for RNA synthesis and DNA synthesis. The IC50 for HgCl2 was 50 and 120 nM for RNA and DNA synthesis, respectively. Cell cycle analysis revealed that B-cells were arrested throughout the cell cycle with CdCl2 and HgCl2. The inhibitory effects exerted by CdCl2 were rapid, inhibiting RNA synthesis within 2 h of activation. Differentiation to Ig secretion was inhibited by CdCl2 and HgCl2 in culture and there appeared to be selective effects on specific Ig isotypes. IgG3 production was most sensitive to inhibition by CdCl2 and HgCl2 followed by IgG1 and IgG2b and then IgM and IgG2a. Changes in the expression of B-cell surface antigens induced by LPS were also influenced by CdCl2. LPS-induced increases in class II MHC expression was inhibited by CdCl2, as was the constitutive expression of class I MHC antigen. A summary of the IC50 for CdCl2 are presented.In summary, both CdCl2 and HgCl2 exert early, inhibitory effects on B-cell activation. This is manifested by the inhibition of RNA, DNA and antibody synthesis. However, selective effects on the production of specific Ig isotypes by these metals may influence the ability of B-cells to mount effective immune responses to pathogens.  相似文献

We previously reported that interleukin-32 gamma (IL-32γ) has a direct effect on osteoclast differentiation and activation in vitro in the context of receptor activator of NF-κB ligand (RANKL) co-stimulation. However, the stage of osteoclast differentiation at which IL-32γ exerts its effect was not determined. Here, we demonstrated that IL-32γ plays an important role in the fusion of preosteoclasts to yield multinuclear osteoclasts, particularly large osteoclasts. The synergistic effect of IL-32γ on RANKL-induced formation of multinuclear osteoclasts was readily apparent when cells were treated with IL-32γ at the fusion stage. In addition, we demonstrated that IL-32γ induced the expression of dendritic cell-specific transmembrane protein (DC-STAMP) and nuclear factor of activated T cells cytoplasmic 1 (NFATc1), and NFATc1 inactivation by cyclosporine treatment attenuated the effect of IL-32γ. These results indicate that IL-32γ is a potential mediator of osteoclast fusion, likely through up-regulation of NFATc1 and DC-STAMP.  相似文献

Folic acid (or folate, FA) has attracted considerable attention for cancer therapy. As one small molecule, its receptor (folate receptor, FR) is significantly overexpressed on the surface of many human tumor cells compared with normal cells. In this work, physical and chemical coupled modification method, that is the combination of nanoimprinting technique and graft polymerization, was adopted to modify FA on nanopatterned polydimethylsiloxane (PDMS) surface for possible application in micro-nanofluidic cytology. The surface property of differently treated PDMS was characterized by FTIR, AFM and contact angle measurement. AO/PI double staining, cell counting and MTT method were performed to examine the potential influence of FA modified nanopatterned PDMS on human cervical carcinoma (HeLa) cell behavior. Both FA modification and nanostructure have positive effect on the growth and viability of HeLa cells. It is the first time that the small molecule-folic acid was used to immobilize on the surface of PDMS in order to improve its surface property.  相似文献

The linear relationship between movement time (MT) and index of difficulty (ID) for Fitts’ type tasks has proven ubiquitous over the last 50+ years. A reciprocal aiming task (IDs 3, 4.5, 6) was used to determine if an enlarged visual display (visual angle 5.1°, 7.4°, or 13.3°) would alter this relationship. With ID = 6, a condition typically associated with discrete action control, the largest visual display (13.3°) allowed the motor system to exploit features of cyclical action control, e.g., shorter dwell times, more harmonic motion, less time decelerating the limb. The large visual display resulted in a quadratic relationship between MT and ID. For the IDs of 3 and 4.5, the visual displays did not alter the underlying control processes. The results are discussed in terms of the preference of the motor system to assemble movements from harmonic basis functions when salient visual information is provided.  相似文献
