Objective To investigate the expression and relationship of TIP30(HIV-1 Tat interactive protein 2), vascular endothelial growth factor (VEGF) and MVD (detected by CD34) in the angiogenesis of human brain astrocytomas. Methods Expression of TIP30, VEGF and CD34 in 19 cases of normal brain tissue and 71 cases of astrocytoma were immunohistochemically examined with Elivision plus two-step method. Results The positive expression of TIP30 could be seen in cytoplasm of neuroglial cells and neurons of 19 normal brain tissues. The positive expression rate of TIP30 in 71 cases of astrocytoma was 33.80 % (24/ 71). The positive expression rate of TIP30 in astrocytoma of different grades was 52 % for grade Ⅱ, 34.78 % for grade Ⅲ and 13.04 % for grade Ⅳ. The positive expression rate of TIP30 in high grade (Ⅲ+Ⅳ) of astrocytoma was found significantly lower than that in low grade(Ⅱ) (χ2=5.71, P <0.05); The expression of VEGF and MVD detected by CD34 in astrocytomas were higher than that in normal brain tissue and increased as the tumor grade increased; In astrocytoma, the negtive correlation was found between the expression of TIP30 and VEGF (r=-0.428, P<0.05); no correlation was found between TIP30 and MVD(r=-0.065, P 0.05); the positive correlation was found between VEGF and MVD(r=0.684, P<0.01). Conclusion The positive expression rate of TIP30 in normal brain tissue is significantly higher than that in astrocytoma. The positive expression rate of TIP30 significantly decreases as the pathological grade of the astrocytoma increases; The expression of TIP30 and VEGF is negatively correlated in astrocytoma.  相似文献

 目的 探讨P2 7kip1、cyclinE的表达与子宫内膜癌发生发展的关系。 方法 用免疫组化方法检测P2 7kip1和cyclinE在 4 0例子宫内膜癌、10例子宫内膜不典型增生、2 0例正常子宫内膜组织中的表达。结果 P2 7kip1在正常子宫内膜、内膜不典型增生、子宫内膜癌中的表达率分别为 90 %、70 %、6 2 .5 % ,其中子宫内膜癌与正常子宫内膜差异有显著性 (P <0 .0 5 )。cyclinE表达阳性率子宫内膜癌组显著高于内膜不典型增生组和正常子宫内膜组 (P <0 .0 5 )。P2 7kip1表达与子宫内膜癌的组织学分级、手术分期有关 (P <0 .0 5 )。P2 7kip1与cyclinE表达呈负相关。 结论 P2 7kip1、cyclinE在子宫内膜癌发生中可能起一定作用 ,P2 7kip1还与子宫内膜癌的进展有关 ,并可能成为判断预后的有用指标。  相似文献

目的:探讨缺氧诱导因子1α(hypoxia inducible factor 1α,HIF-1α)在乳腺正常组织、不典型增生、原位癌、浸润性癌组织中的表达水平,分析其与浸润性乳腺癌临床病理指标及肿瘤血管生成之间的关系。方法:收集20例正常乳腺组织、23例不典型增生、34例原位癌、80例浸润性乳腺癌组织标本,用免疫组化SP法检测各组中HIF-1α的表达,分析其在浸润性乳腺癌中的表达与肿瘤大小、腋窝淋巴结受累情况、TNM分期、组织学分级、激素受体等临床病理指标之间的关系。并用CD34标记肿瘤新生血管,分析HIF-1α表达对肿瘤血管生成的影响。结果:HIF-1α在正常乳腺组织中未见阳性表达,在乳腺不典型增生组织、原位癌组织、浸润性癌组织中表达阳性率分别为8.7%(2/23)、47.1%(16/34)及73.8%(59/80),表达率依次增高,乳腺原位癌组织与不典型增生组织之间、乳腺浸润癌组织与原位癌组织之间差异具有统计学意义(P<0.01)。在浸润性乳腺癌中,HIF-1α的表达在不同肿瘤直径、组织学分级及淋巴结转移分组中差异具有统计学意义(P均<0.05)。浸润性导管癌HIF-1α阳性组MVD值(11.74±4.398)明显高于HIF-1α表达阴性组(9.093±2.856),差异有统计学意义(P<0.05)。HIF-1α表达与MVD值呈正相关性(r=0.633,P<0.05)。结论:HIF-1α在浸润性乳腺癌组织中表达水平升高,其表达上调与乳腺癌新生血管生成密切相关。  相似文献

目的 研究环氧化酶 2 (COX 2 )在子宫内膜癌组织中的表达及其与血管生成的关系。方法 用免疫组化法检测42例子宫内膜癌和15例正常子宫内膜组织中COX 2的表达,并检测微血管密度(MVD)。结果 子宫内膜癌组织中COX 2、MVD明显高于正常子宫内膜组织(P <0 0 5 ) ,COX 2高表达与子宫内膜癌组织学分级和浸润深度有关(P <0 0 5 ) ,与临床分期、淋巴结转移和组织学分型无关(P >0 0 5 ) ,COX 2表达阳性者MVD明显高于COX 2表达阴性者(P <0 0 5 )。结论 COX 2过表达可能参与子宫内膜癌发生发展过程,且与子宫内膜癌新生血管形成有关。  相似文献

背景与目的:子宫内膜癌发病率呈逐年上升趋势,但其确切发病机制及预后因素尚不明确。本实验旨在研究增殖细胞核抗原(proliferating cell nuclear antigen.PCNA)指数、微血管密度microvessel density.MVD雪在子宫内膜癌组织中的表达,并结合肿瘤的手术病理分期、病理分级,进一步判断其价值。方法:采用免疫组织化学SP法,检测62例子宫内膜腺癌、22例正常子宫内膜组织的PCNA指数和MVD,结合肿瘤的临床分期、病理分级,对结果进行统计学分析。结果:子宫内膜癌组织中PCNA指数(54.49±9.29)和MVD(48.97±8.56)较正常子宫内膜明显增高(P<0.05);子宫内膜癌的期别越晚、分化越差,PCNA指数和MVD越高(P<0.05);在子宫内膜癌组织中,PCNA指数与MVD呈正相关。结论:PCNA指数、MVD增高与子宫内膜癌的发展有关;血管生成促进肿瘤细胞增殖。  相似文献

目的建立乳腺非典型增生病动物模型,探讨非典型增生与血管生成的关系。方法采用灌服二甲基苯蒽+颈部枷具方法复制大鼠肝郁血瘀型乳腺非典型增生动物模型,并给予中药乳复汤治疗。常规染色观察大鼠乳腺组织病理组织形态,免疫组化方法检测MVD、VEGF和bFGF的表达。结果病证模型组乳腺非典型增生率90%,较单纯造模组高,P=0·0415;中药乳复汤治疗组非典型增生率较病证模型组低,P=0·0459;随着一般增生到非典型增生程度加重,MVD计数增多,VEGF、bFGF的表达升高,P<0·05。结论病证结合造模更适合中医动物实验要求,中药乳复汤具有抑制血管生成作用。  相似文献

目的:探讨结直肠癌组织中FGF20的表达及其与微血管密度之间的相关性。方法:免疫组织化学染色和Western Blot方法检测结直肠癌组织及正常肠黏膜组织中FGF20的表达或CD34的表达,分析FGF20在癌组织及正常肠黏膜组织中的表达差异以及与临床病理之间的相关性,探讨FGF20的表达与结直肠癌组织中微血管密度的相关性。结果:免疫组织化学和Western Blot结果均提示结直肠癌组织中FGF20表达显著高于正常肠黏膜组织(P<0.01);癌组织中FGF20的表达与肿瘤浸润(P<0.05)及淋巴结转移(P<0.05)密切相关,与分化程度及肝转移无明显相关(P>0.05);癌组织中FGF20的表达与癌组织中微血管密度呈显著正相关(P<0.05)。结论:FGF20在结直肠癌微血管形成中发挥重要作用,进而促进结直肠癌的发生、发展及远处转移,为明确FGF20在结直肠癌微血管形成中的作用以及深入探讨FGF20促进微血管形成的分子机制提供实验依据。  相似文献

Objective:To investigate vascular endothelial growth factor(VEGF) expression and microvessel density(MVD) on the fibromyoma uteri treated with mifepristone.Methods:VEGF expression and MVD were counted on 60 cases of the fibromyoma uteri by SP method,including 40 cases of mifepristone treated and 20 cases of untreated patients as controlled group.Results:VEGF positive expression rate and MVD in treated group were 37.5% and 9.90 ± 5.95,which were lower than those in controlled group(80% and 16.36 ± 2.07;P < 0.05).Meanwhile,in treated group,those in marked shrink in tumor size sub-group were lower than in not obvious sub-group(12.0% and 7.89 ± 4.36 vs.80% and 11.29 ± 3.10;P < 0.05).Conclusion:VEGF and MVD expression decreases in the fibromyoma uteri after treatment with mifepristone,suggesting that mifepristone could inhibit angiogenesis and blood supply resulting in tumor shrink.  相似文献

目的:研究乙酰肝素酶(heparanase,Hpa)在子宫内膜癌中的表达,并探讨其临床意义.方法:采用免疫组织化学SP法检测48例子宫内膜癌组织中Hpa蛋白的表达情况.结果:Hpa蛋白在子宫内膜癌中阳性率显著高于正常子宫内膜、子宫内膜增生症(P>0.05).单因素分析表明浸润不超过内1/2肌层者的Hpa蛋白阳性率显著低于浸润达到外1/2肌层者(P>0.05).有淋巴转移组的Hpa蛋白阳性率显著高于无淋巴转移组(P>0.05).Ⅰ-Ⅱ期组的Hpa蛋白阳性率显著低于Ⅲ-Ⅳ期组(P>0.05).弥漫型内膜癌中Hpa蛋白阳性率高于局限性(P>0.05).中、低分化内膜癌中Hpa表达与细胞分化无关(P>0.05).多因素Logistic回归分析结果表明Hpa蛋白表达只与淋巴转移、临床分级及肌层浸润显著相关(P<0.05).结论:Hpa在子宫内膜癌的阳性率显著高于正常子宫内膜、子宫内膜增生症.Hpa表达与转移、临床分期显著相关.  相似文献

目的:探讨核呼吸因子1(NRF-1)与线粒体转录因子A(mtTFA)在子宫内膜癌组织中表达的相关性研究。方法:采用SP免疫组化染色法检测NRF-1与mtTFA蛋白在41例子宫内膜癌组织和23例非典型增生子宫内膜组织中的表达,同时选择21例正常子宫内膜组织作为对照。结果:NRF-1和mtTFA在子宫内膜癌中的表达阳性率均明显高于正常子宫内膜组织和非典型增生子宫内膜组织,从正常子宫内膜组织到非典型增生内膜组织,再到子宫内膜癌组织,其表达阳性率均呈逐渐上升趋势,差异有统计学意义,χ2=24.564,P=0.000 1;χ2=33.372,P=0.000 1。NRF-1和mt-TFA的表达与临床分期、组织学分级、肌层浸润及淋巴结转移情况均有明显相关性,且差异有统计学意义,P<0.05。子宫内膜癌组织中mtTFA与NRF-1的表达呈正相关,rs=0.806,P=0.004。结论:NRF-1和mtTFA蛋白的表达异常可能在子宫内膜癌变过程中产生一定的作用。  相似文献
