目的 了解小鼠感染旋毛虫后IgG抗体水平、IL-2分泌量和T淋巴细胞亚群的动态变化。方法 分别于旋毛虫感染后第7、14、21、28和35天,采用ELISA方法检测血甭特异性IgG抗体水平和IL-2含量,采用流式细胞仪检测CD4^+、CD8^+T细胞百分率。结果 IgG抗体水平在感染后逐渐上升,感染后35天达最高值;T淋巴细胞的变化表现为CD4^+细胞减少、CD8^+T细胞增多,CD4^+/CD8^  相似文献

芪黄冲剂对慢性乙型肝炎患者免疫功能的影响   总被引:16,自引:0,他引:16  
目的:观察芪黄冲剂对慢性乙型病毒性肝炎免疫功能的影响。方法:治疗组以芪黄冲剂治疗,对照组以乙肝清热解毒冲剂合乙肝养阴活血冲剂治疗。两组患者采用间接免疫荧光法检测,治疗前后免疫球蛋白IgG、IgM、IgA、T淋巴细胞亚群CD3^+、CD4^+、CD8^+、CD4^+/CD8^+、IL-2、IL-6、TNF、NK。结果:芪黄冲剂具有益气活血,滋养肝肾兼以清热解毒作用,能升高CD4^+、NK细胞,提高CD4^+/CD8^+比值及IL-2水平,降低CE8^+,尤其对CD4^+、CD4^+/CD8^+、NK细胞以及IL02的调整作用优于对照组(P〈0.05 ̄0.01),能显著降低IL-6及TNF水平(P〈0.05 ̄0.01)。同时还能降低IgG、IgM、IgS,其中对IgA的降低作用显著优于对照组。结论:芪黄冲剂不仅具有  相似文献

目的: 探讨紫外线减毒弓形虫ZS1 株在小鼠体内的免疫保护性和细胞免疫反应。方法: 用波长为2537°A 的紫外线照射弓形虫ZS1株滋养体, 照射高度为5 cm , 照射时间60 m in。小鼠于免疫后45 d 用同株滋养体攻击感染, 攻击后4 d 剖杀, 与单免疫组、单感染组及正常对照组小鼠比较其脾T淋巴细胞增殖反应及其亚群的变化。结果: 小鼠接种紫外线减毒弓形虫ZS1 株滋养体后能正常存活, 于接种后49 d 各组织未查见滋养体、包囊或假包囊; 免疫组攻击感染后存活时间较单感染组延长; 体外特异抗原刺激后, 可诱导免疫组及免疫攻击组强的脾T淋巴细胞增殖反应; 免疫攻击组CD4+ T细胞明显下降, CD4+ /CD8+ 比率倒置; 免疫组、免疫攻击组及感染组的NK细胞活性均明显增强。结论: 紫外线减毒弓形虫ZS1株滋养体疫苗能够诱导免疫小鼠产生一定的抗攻击感染保护力, 其中CD8+ T细胞和NK细胞可能发挥着重要作用。  相似文献

目的研究肝硬化和肝细胞癌患者IL-2、sIL-2R、IL-6、T细胞亚群的变化和相互关系。方法分别采用双抗体夹心ELISA法、ABS-ELISA法和红细胞花环实验,对46例肝炎肝硬化和肝癌进行了IL-2、IL-6、sIL-2R和T细胞亚群的测定,并与66例健康献血员进行了对照。结果肝炎肝硬化(LC)、肝细胞癌(HCC)患者血清IL-2、CD+4/CD+8水平明显低于正常对照(NC)组(P<001),而IL-6、sIL-2R水平明显升高,HCC组的平均IL-6水平高出正常10倍以上,较肝硬化组明显升高(P<001)。在肝炎肝硬化和肝癌患者均发现IL-2与CD+4/CD+8比值、IL-6与sIL-2R正相关,IL-6与CD+4/CD+8比值显著负相关,而与IL-2无相关关系。结论肝炎肝硬化和肝细胞癌患者存在免疫功能紊乱和低下、淋巴因子网络失衡,这与其病理生理机制有关,也为临床上LC、HCC应用IL-2治疗提供了理论依据。IL-6的显著增高有助于HCC的早期诊断  相似文献

本文观察8wk龄C57BL/6小鼠感染日本血吸虫尾蚴后不同时期脾细胞经SEA或ConA刺激,体外诱生的IL-2和IFN-γ活性变化。结果表明两种细胞因子活性均在感染后第4-6wk开始上升,第6—8wk达高峰,第12—14wk恢复至感染前水平。IFN-γ高峰时间略先于IL-2。非特异性刺激原诱生组和特异性刺激原诱生组各阶段细胞因子活性动态基本一致,前组活性高于后组。提示IFN-γ与IL-2活性与血吸虫卵肉芽肿的诱导、成熟与维持有密切关系。  相似文献

应用间接免疫荧光法,放射免疫分析法,微量细胞培养法及斑点杂交技术,检测了27例乙型慢性活动性肝炎(CAH一B)患者及7例正常对照者CD+4、CD+8及B细胞膜上白细胞介素一2受体(mIL-2R,CD25抗原为其β链),各亚群细胞培养上清及病人同期血清中可溶性白细胞介素一2受体(sIL-2R),CD+4细胞培养上清中IL一2活性。结果:CAH-B病人外周血CD25抗原阳性的CD+4、CD+8细胞及B细胞较正常对照者减少,且CD25抗原阳性的CD+4、CD+8细胞减少较B细胞明显;CD+细胞培养上清中IL一2活性较正常人降低,并与病情的轻重、CD+4细胞mIL一2R的表达呈正相关;CAH-B病人血清sIL-2R增高,但培养上清sIL-2R与正常对照无显著性差异。结果提示:CAH一B病人外周血CD+4、CD+8及B细胞mIL-2R表达均减弱;CAH-B病人体内T细胞免疫应答功能受损可能较B细胞受损明显;CAH-B病人体内IL一2产生水平低下与病情轻重有某种关联;CAH一B患者血清中增高的sIL一2R可能来自肝内浸润的免疫活性细胞mIL-2Rβ链脱落。  相似文献

Susceptibility/resistance of the intermediate host to alveolar echinococcosis (AE) seems to be based on hitherto unknown immunological mechanisms, possibly involving the activation of different CD4+ T cell immune responses (Th1/Th2). Mice of two strains previously characterized as 'susceptible' (C57BL/6 J) and 'resistant' (C57BL/10 J) to secondary AE were orally infected with eggs of Echinococcus multilocularis and the course of infection was analysed by macroscopical, pathohistological and immunohistochemical examinations of the lymphocytes and cytokines participating in the periparasitic granulomas and by serological examinations of cytokines and E. multilocularis-specific antibodies. Although differences in the extent of parasitic growth were seen between the two groups, the composition of the granulomas was quite similar with CD4+ cells being the dominant lymphocyte subpopulation, succeeded by B cells and CD8+ cells. Interferon (IFN)-gamma-, interleukin (IL)-2- and IL-4-expressing cells could not be detected in the lesions of the early phase of the infection, possibly indicating the host's immunosuppression, but were present at the end. IL-10 was the most prominent cytokine throughout the course of the disease. Serological analyses of the cytokine concentrations revealed small amounts at the beginning and high levels at the end of the infection. The pattern of cytokine response was similar for IL-4 in both strains, but different for IL-2 and IL-10 in the late phase, when the C57BL/10 J strain developed higher levels than the C57BL/6 J strain. Correspondingly only small amounts of immunoglobulin (Ig)M, IgG1, IgG2a and IgG3 could be detected at the beginning of disease, followed by higher levels at the end. The courses of antibody titres were similar in both groups except IgG3, which was more pronounced in the C57BL/10 J strain. Parasite-specific IgG2b could neither be detected in the C57BL/6 J nor in the C57BL/10 J strain by the test system used. The results of the study suggest both subsets of CD4+ T cells (Th1 and Th2) being involved in murine primary alveolar echinococcosis. A strict differentiation of mice in susceptible and resistant animals based on the activation of different CD4+ T cell immune responses (Th1 'resistant' and Th2 'susceptible') should be avoided.  相似文献

目的 探讨旋毛虫成虫可溶性抗原 (AWSAg)接种小鼠后诱发的免疫应答。  方法 制备旋毛虫AWSAg免疫小鼠 ,分别于攻击感染后 7、14、2 1、2 8和 3 5d ,动态观察免疫小鼠特异性IgG抗体水平、IL 2水平和T淋巴细胞亚群的变化。 结果 与非免疫鼠相比 ,免疫鼠血清特异性IgG抗体OD值在攻击感染后 7d明显升高 ,在观察期内一直高于非免疫鼠。感染后 7dIL 2水平明显增高 ,达观察期内最高值 ,2 1d后才逐渐减少 ,在感染后 2 8~ 3 5d仍高于正常组。免疫鼠CD4+ T细胞在攻击感染 7d明显增加并保持不变 ,非免疫鼠CD4+ T细胞及CD4/CD8比值明显较正常鼠减低。 结论 旋毛虫AWSAg免疫小鼠攻击感染后 ,出现细胞、体液免疫应答。  相似文献

BALB/c mice are highly susceptible to African trypanosomiasis, whereas C57BL/6 mice are relatively resistant. Other investigators have reported that the synthesis of IgG antibodies to purified membrane form of variant surface glycoprotein (mfVSG) of Trypanosoma brucei is CD1 restricted. In this study, we examine the role of the CD1d/NKT cell pathway in susceptibility and resistance of mice to infection by African trypanosomes. Administration of anti-CD1d antibodies to Trypanosoma congolense-infected BALB/c mice neither affects the parasitemia nor the survival time. Correspondingly, CD1d(-/-) and CD1d(+/+) BALB/c mice infected with T. congolense or T. brucei show no differences in either parasitaemia or survival time. The course of disease in relative resistant C57BL/6 mice infected with T. congolense is also not affected by the absence of CD1d. Parasitaemia, survival time, and plasma levels of IgG2a and IgG3 parasite-specific antibodies in infected CD1d(-/-) C57BL/6 are not different from those of infected CD1d(+/+) C57BL/6 mice. We conclude that CD1d-restricted immune responses do not play an important role in susceptibility/resistance of mice infected with virulent African trypanosomes. We speculate that virulent trypanosomes have an evasion mechanism that prevents the induction of a parasite-specific, CD1d-restricted immune response by the host.  相似文献

目的 研究细胞因子IFN γ和IL 4的抗卡氏肺孢子虫感染作用。 方法 分别用IFN γ或IL 4基因敲除的C5 7BL 6小鼠 (各 15只作为动物模型 )与未经基因敲除的C5 7BL 6小鼠 (15只作感染对照组 ) ,以密切接触的方式 ,使小鼠自然感染卡氏肺孢子虫 ;同时设非感染组 15只小鼠作阴性对照。分别于 3、5和 7wk后观察各组小鼠肺内虫体数量、肺部CD4+ 、CD8+ T细胞反应以及血清中卡氏肺孢子虫特异性IgG抗体滴度。分析基因敲除鼠对卡氏肺孢子虫的易感性。 结果 IFN γ基因敲除小鼠肺内虫体数量明显高于IL 4基因敲除小鼠 (P <0 .0 5 )和对照组小鼠 ,而IL 4基因敲除小鼠肺内虫体数量与对照组无显著性差异 (P >0 .0 5 )。T细胞亚群分析表明 ,两种基因敲除小鼠的CD4+ 和CD8+ T细胞反应曲线与感染度呈正相关。而IgG抗体滴度 ,3组接触感染鼠均逐渐呈不同程度上升。  结论 IFN γ在小鼠抗卡氏肺孢子虫中起重要作用 ,但缺少IL 4基因的小鼠对肺孢子虫感染的影响不明显。  相似文献

In order to evaluate the influence of route and dose of inoculation on interleukin 2 (IL2) production, C57BL/6 mice were infected either intravenously (I.V.) or subcutaneously (S.C.) with 10(5) or 10(8) Mycobacterium lepraemurium. The role of genetic factors on the production of IL2 during M. lepraemurium infection, was investigated in 7 inbred mouse strains (C57BL/6, DBA/2, F1 (C57BL/6 X DBA/2), DBA/1, BALB/c, CBA and A/J) after I.V. infection with 10(7) M. lepraemurium. At different times after M. lepraemurium inoculation, the number of AFB within the spleens of infected mice was counted and the ability of Con A-activated spleen cells to produce IL2 was studied. In S.C. inoculated C57BL/6 mice the increase in footpad thickness was measured during the progression of infection. After one month of infection heavily infected C57BL/6 mice (10(8) bacilli) showed an early and strong deficiency of IL2 production, regardless of the route of inoculation, whereas mice infected with a lower dose (10(5) bacilli) did not. In S.C. infected mice the decrease of IL2 production was observed when the footpad enlargement reached to the plateau phase. The data obtained from the numeration of AFB within the spleens of infected mice allowed to rank the infected mouse strains into 2 separated groups according to the pattern of the Bcg gene expression. An IL2 deficiency was only observed in C57BL/6, DBA/1, (C57BL/6 X DBA/2)F1 and DBA/2 infected mouse strains. No evident correlation could be shown between splenic IL2 activity upon Con A stimulation and the number of AFB recovered from the spleens of these 7 inbred mouse strains.  相似文献

目的: 探讨细胞因子在旋毛虫感染免疫中的调节作用。方法: 采用间接ELISA 和生殖力指数测定等方法对IL-2 作用下感染旋毛虫小鼠的IgG抗体水平及寄生虫感染力进行连续观察。结果: 不同剂量IL-2对不同地理株感染小鼠产生的影响存在较大差异: 对于黑龙江株猪型旋毛虫感染小鼠, IL-2 注射具有明显抗虫效果; 而对于美国株猪型旋毛虫感染的小鼠, IL-2 对成虫繁殖力无显著影响, 但大剂量IL-2 (每鼠2 000 U/次) 使新生幼虫的感染力降低。结论: IL-2 对黑龙江株猪型旋毛虫感染力有明显抑制作用。  相似文献

The impact of Toxocara canis infection on the proportion of CD4+ and CD8+ T splenocytes, the serum concentrations of cytokine IFN-gamma and IL-5, and the production of Toxocara-specific antibodies were studied in two C57BL6/J mouse groups and their offspring. The mice were infected with 1000 T. canis eggs on the day of mating (early infection) and on day 14 of pregnancy (late infection). Early infection resulted in a significant increase of CD4+ T-cell subtype, however, a decline in CD8+ in comparison with late infection, as well as with non-infected control. The IFN-gamma serum concentrations decreased in infected mothers after the birth when compared with non-infected mothers, while in the offspring this cytokine was barely or not detectable. In the mothers of both infected groups, the humoral immune response included both parasite-specific IgM and IgG2 antibodies. While IgG1 levels remained constant throughout the whole experiment in mothers with early infection, late-infected mothers became seropositive only 3 weeks after delivery. IgM was not detectable in any offspring. Pups from early-infected mothers had IgG1 antibodies. Conversely, IgG2 was detectable in pups of both experimental infection groups. A significant difference was observed in the amounts of pups/litter of the infected mothers in comparison with the non-infected ones. Only 56% of females after early infection and 79% of those after late infection had a successful pregnancy. However, all mice of the control group produced a litter. The first T. canis larvae were detected in the muscles of the offspring of both groups on day 5 after the birth. These data show the changes in regulatory and cytotoxic immunity mechanisms of the infected mothers and their offspring and the high level of pregnancy loss as a result of larval toxocariasis.  相似文献

目的 探讨不同品系小鼠对卡氏肺孢子虫 (P.c)的易感性及其免疫反应。 方法 以接触传播方式使BAL B/ c和 C5 7BL / 6小鼠 (各 15只 )感染 P.c。观察小鼠与传染源接触 4、5、6 wk后肺内虫数、支气管肺泡灌洗液中CD4 + T细胞和 CD8+ T细胞及血清 Ig G水平的变化。 结果 与传染源接触 4 wk,小鼠肺内均可检出 P.c,之后 ,BAL B/ c小鼠虫数继续增高 ,5 wk末达高峰。4 wk时 C5 7BL/ 6小鼠虫数无明显变化 ,5 wk时显著少于 BAL B/ c小鼠。两组小鼠支气管肺泡灌洗液内 CD4 + CD6 2 low T细胞和 CD8+ CD6 2 low T细胞数明显增多 ,血清 Ig G抗体水平显著上升。6 wk小鼠肺内 P.c均转阴。 结论  BAL B/ c比 C5 7BL / 6小鼠更易感染 P.c。 BAL B/ c和 C5 7BL / 6小鼠感染 P.c后5 wk可产生有效的细胞和体液免疫反应并清除肺内感染的 P.c  相似文献

We studied IL-4, IL-10 and IFN-gamma secretion by splenocytes and the plasma levels of different isotypes of antibodies against various antigens of Trypanosoma congolense in highly susceptible BALB/c and relatively resistant C57BL/6 mice during the early course of infection with T. congolense. The patterns of appearance of cytokine spotforming cells in the spleens were essentially similar in the two mouse strains although higher numbers were detected in the spleens of BALB/c than C57BL/6 mice on some days post-infection. However, the amount of IL-4, IL-10 and IFN-gamma secreted into the culture fluids was dramatically different. From day 4 forward, splenocytes from BALB/c mice secreted very high levels of these cytokines. In contrast, splenocytes from infected C57BL/6 mice did not secrete detectable levels of IL-4 throughout the period tested. The secretion of IL-10 and IFN-gamma by C57BL/6 splenocytes only became appreciable on day 6 and was down-regulated by day 8, when the first wave of parasitaemia was being controlled. At days 6-8, splenocytes from infected C57BL/6 mice secreted two-fold higher amounts of IL-12 p40 than those from BALB/c mice. Infected BALB/c mice mounted an earlier IgM antibody response to variant surface glycoprotein (VSG), formalin-fixed T. congolense and whole T. congolense lysates than did infected C57BL/6 mice. However, they failed to make any detectable IgG3 and IgG2a antibody responses to these antigens whereas infected C57BL/6 mice made strong IgG3 and IgG2a responses. We speculate that enhanced resistance against T. congolense infections in mice may be mediated by IL-12 dependent synthesis of IgG2 antibodies to VSG and possibly also common trypanosomal antigens.  相似文献

目的观察旋毛虫对C57BL/6小鼠体内Hepa1-6肝癌细胞生长的抑制作用。方法将C57BL/6小鼠随机分成8组,每组10只。第1和5、2和6、3和7组分别感染未处理旋毛虫、^60Co处理和紫外线处理旋毛虫,4和8组为不感染旋毛虫对照组,1~4组和5~8组分别于接种旋毛虫前7 d和接种后11 d接种Hepa1-6肝癌细胞。荷瘤后25 d后处死小鼠,测量肿瘤体积、重量及脾脏CD3^+、CD4^+、CD8^+淋巴细胞数量。结果未处理旋毛虫组、^60Co处理组和紫外线处理组小鼠肿瘤体积和重量均显著低于对照组(P〈0.05);脾脏CD3^+、CD4^+百分率和CD4^+/CD8^+、CD4^+/CD3^+的比值显著高于对照组(P〈0.01或P〈0.05)。结论未处理的旋毛虫、经^60Co和紫外线处理的旋毛虫对C57BL/6小鼠体的Hepa1-6肝癌细胞的生长均有抑制作用,未处理旋毛虫的抑瘤效果最好。  相似文献
