目的 研究细胞外基质金属蛋白酶诱导因子(CD147)和基质金属蛋白酶-2(MMP-2)在大肠癌组织中的表达及其与大肠癌临床病理因素间的关系,探讨其与大肠癌侵袭和转移的相关性及二者间的相互联系.方法 应用免疫组织化学方法检测44例大肠癌患者癌组织、癌旁组织和正常对照组织中CD147和MMP-2的表达,以图象分析软件进行半定量分析.结果 大肠癌组织、癌旁组织和正常对照组织中CD147和MMP-2表达阳性率呈递减改变.CD147和MMP-2的表达部位和染色强度相似,半定量分析显示两者呈正相关(r=0.777,P<0.05).两者的表达与大肠癌患者年龄、肿瘤分型、肿瘤分化程度未见相关性(P<0.05),而在大肠癌患者伴转移组显著高于无转移组(P<0.05).结论 CD147在大肠癌患者组织中的表达增高与大肠癌侵袭转移密切相关,其促MMP-2的激活和表达增高的作用可能是其中一个重要因素. 相似文献

目的用质粒IL-3、CD40L基因在小鼠体内诱导肝脏B220^+/DEC205^+树突状细胞增殖。方法大容量快速小鼠尾静脉注射方法导入质粒IL-3和CD40L基因,Percoll密度梯度离心法分离肝脏非实质细胞,流式细胞仪标记并分离树突状细胞,Giemsa染色和扫描电子显微镜做形态学观察。结果实验组小鼠肝脏非实质细胞增殖,主要分布在小叶内、汇管区周围和汇管区。流式细胞仪标记并分离出B220^+/DEC205^+树突状细胞,实验组B220^+/DEC205^+细胞(16.0%)较对照组(1.1%)明显增多。Giemsa染色和扫描电子显微镜显示:B220^+/DEC205^+细胞胞核形状不规则,胞质内无颗粒,胞质具明显突起。结论大容量快速尾静脉注射将质粒IL-3和CD40L基因注入小鼠体内,可在肝脏诱导B220^+/DEC205^+树突状细胞增殖。 相似文献

目的 探讨狼疮性肾炎(LN)患者血浆及肾组织白细胞介素-13(IL-13)的表达及其临床意义。方法 采用ELISA法检测血浆IL-13水平,采用免疫组化ABC法检测肾组织IL-13的表达。结果 LN患者血浆IL-13水平、肾小球及肾小管间质区IL-13表达较对照组和非LN肾脏病对照组增高,以Ⅳ型LN患者肾小球区IL-13表达最高,而不同病理类型的LN肾小管间质区IL-13表达无显著性差异。LN患者血浆及肾小管间质区IL-13表达与Ccr和血清C3呈负相关,与肾小管间质病变活动指数和肾小球病变活动指数呈正相关。结论 IL-13参与LN的分子病理机制,血浆及肾组织IL-13水平可能有助于判断狼疮活动程度。 相似文献

目的探讨脓毒症患者血清中白细胞介素(IL)-17、IL-23和降钙素原(PCT)的表达及其临床意义。方法 70例脓毒症患者作为观察组,留取清晨静脉血并分离血清,30例无感染性疾病的健康体检的成人血清标本作为对照组。检测两组中IL-17、IL-23和PCT的表达。结果观察组中IL-17、IL-23和PCT的表达量明显高于对照组,观察组血清中IL-17、IL-23和PCT的表达量与是否伴有多器官功能障碍综合征(MODS)、预后密切相关。相关分析显示IL-17和IL-23、IL-17与PCT具有正相关性。结论脓毒症患者血清中IL-17、IL-23和PCT的表达升高,对病变的发生和进展有重要作用。 相似文献

目的检测老年女性骨质疏松(OP)患者血清白细胞介素(IL)-6、IL-12、降钙素(CT)及雌二醇(E2)表达及其意义。方法老年女性OP患者86例为观察组,老年健康女性50例为对照组,检测血清IL-6、IL-12、CT及E2含量。结果观察组IL-6、IL-12表达量显著高于对照组,观察组CT和E2表达明显低于对照组(P<0.01),线性相关分析显示观察组中IL-6、IL-12、CT及E2表达有相关性。结论老年女性OP患者血清中IL-6、IL-12、CT及E2异常表达,其在疾病进展中有重要作用,联合检测IL-6、IL-12、CT及E2可能是判断老年女性OP的重要诊断指标。 相似文献

为探讨白细胞介素-6(IL-6)和白细胞介素-8(IL-8)与冠心病(CHD)的相互关系及临床意义,采用双抗体夹心法(ELISA)检测了55例老年CHD患者血清IL-6、IL-8的水平变化并与25例健康老年人对照,结果显示CHD患者血清IL-6和IL-8水平明显高于对照组(P<0.05),且IL-6和IL-8之间呈正相关(γ=0.423,P<0.001),提示IL-6和IL-8的水平变化与CHD密切相关,IL-6和IL-8可能参与了CHD的发病及病理变化过程.监测其水平变化,对CHD的诊治具有一定的临床价值. 相似文献

目的研究自身免疫性疾病系统性红斑狼疮(SLE)中白细胞介素(IL)-23和IL-12的调节作用。方法78例SLE患者[以SLE疾病活动评分(SLEDAI)进行活动性评分]和正常对照36名,分别抽提SLE患者和正常对照外周血单个核细胞(PBMC)的mRNA,并反转录成cDNA,用实时定量聚合酶链反应(PCR)的方法对IL-23特异的亚单位P19、IL-12特异的亚单位P35以及它们共有的亚单位P40作定量分析,数据采用看家基因进行标化。结果未经治疗、已经治疗的SLE患者的IL-12特异性的亚单位P35表达水平显著低于正常对照(P值分别<0.05,<0.01)。与未经治疗相比,用类固醇激素或类固醇类激素加免疫抑制剂治疗的SLE患者显著抑制P40和P19的表达(P值分别<0.01和<0.05)。活动性SLE患者(SLEDAI>10)的P19、P40、P35表达水平显著高于非活动性SLE患者(SLEDAI≤10)(P值分别<0.01,<0.01和<0.05)。结论IL-12的表达下降或IL-23的表达上调也许参与SLE的发病过程,这两种细胞因子有可能是SLE患者的治疗靶点。 相似文献

目的:探讨白细胞介素-2(IL-2)与可溶性白细胞介素-2受体(sIL-2R)与乙型肝炎疫苗接种无、弱应答的关系。 方法:在HBsAg和植物血凝素(phytohaemagglutinin,PHA)两种刺激条件下,对21名乙型肝炎疫苗接种无、弱应答者、22名强应答者和21名HBsAg慢性携带者的外周血单个核细胞进行体外培养,并用酶联免疫法测定细胞培养上清液中IL-2和sIL-2R的水平。 结果:无、弱应答组的IL-2水平明显低于强应答组(t=8.80,P<0.001=,而与HBsAg慢性携带组相近(q=0.06, P>0.5)。无、弱应答组和强应答组的sIL-2R水平差异无显著意义。结论:外周血单个核细胞分泌IL-2 水平低下可能是乙型肝炎疫苗接种后无、弱应答发生的原因之一。 相似文献

目的探讨白细胞介素-5(IL-5)、可溶性白细胞介素-2受体(sIL-2R)、一氧化氮(N())及嗜酸性粒细胞(EOS)在哮喘发病中的意义。方法对治疗前后的急性发作期、稳定期哮喘组及对照组血清II-5、sIL-2R和NO水平及EOS计数进行了研究。结果哮喘急性发作组血清IL-5、sIL-2R、NO及EOS水平均明显高于哮喘缓解期组与对照组。哮喘发作期IL-5与EOS呈显著正相关。哮喘急性发作期组治疗后sIL-2R及NO水平较治疗前显著降低,IL-5与EOS水平下降但无统计学意义。结论血清IL-5、sIL-2R、NO及EOS水平升高,与哮喘发作密切相关。 相似文献

AIM: To clarify the clinicopathologic significance of COX-2 expression in human colorectal cancer. METHODS: A total of 128 surgically resected colorectal cancer specimens were immunohistochemically analyzed with the use of anti-COX-2, anti-VEGF and anti-MMP-2 antibodies. The relationship between the cyclooxygenase-2 expression in primary lesions of colorectal cancer and clinicopathoiogic parameters was evaluated by chi-square test. RESULTS: Among 128 cases of colorectal cancer, 87 (67.9%) were positive for cyclooxygenase-2. The expression of cyclooxygenase-2 was significantly correlated with the depth of invasion, stage of disease, and metastasis (lymph node and liver). Patients in T3-T4, stages Ⅲ-Ⅳand with metastasis had much higher expression of cyclooxygenase-2 than ones in T1-T2, stages Ⅰ-Ⅱ and without metastasis (P<0.05). Among 45 cases of colorectal cancer with lymph node metastasis, the COX-2-positive rate was 86.7% (39/45) for primary lesions and diffuse cytoplasmic staining for COX-2 protein was detected in cancer cells in 100% of metastatic lesions of the lymph nodes. VEGF expression was detected in 49 tumors (38.3%), and VEGF expression was closely correlated with COX-2 expression. The positive expression rate of VEGF (81.6%) in the cyclooxygenase-2-positive group was higher than that in the cyclooxygenase-2-negative group (18.4%, P<0.05). MMP-2 expression was detected in 88 tumors (68.8%), and MMP-2 expression was closely correlated with COX-2 expression. The positive expression rate of MMP-2 (79.6%) in the positive COX-2 group was higher than that in the negative COX-2 group (20.4%, P<0.05). CONCLUSION: Cyclooxygenase-2 may be associated with tumor progression by modulating the angiogenesis and cancer cell motility and invasive potential in colorectal cancer and it can be used as a possible biomarker. 相似文献

目的探讨micro RNA-124-2在CD137-CD137L信号通路调控小鼠血管平滑肌细胞(VSMC)活化T细胞核因子C1(NFATc1)表达中的作用。方法小鼠血管平滑肌细胞采用组织贴块法原代培养,细胞中micro RNA-124-2表达采用RT-PCR法检测;应用脂质体转染技术将micro RNA-124-2的模拟物(mimic)和抑制物(inhibitor)转染至小鼠VSMC内,细胞中NFATc1 m RNA及蛋白表达采用实时荧光定量PCR及Western blot方法检测;应用双荧光素酶报告检测micro RNA-124-2对NFATc1-3'UTR的作用。结果 anti-CD137特异性刺激CD137-CD137L轴后,平滑肌细胞micro RNA-124-2表达较对照组降低(0.29±0.13比1.00±0.00,P0.05),而anti-CD137L特异性阻断CD137-CD137L轴后,细胞中micro RNA-124-2表达较对照组增加(3.42±0.17比1.00±0.00,P0.05);与阴性对照组相比,升高或下调micro RNA-124-2的表达均可逆转CD137-CD137L轴对NFATc1的作用;双荧光素酶报告系统显示micro RNA-124-2对NFATc1-3'UTR有抑制作用(0.283±0.011比1.294±0.143,P0.001)。结论 CD137-CD137L受体-配体轴可以通过调控micro RNA-124-2进而影响NFATc1的表达。 相似文献

Purpose This study was conducted to evaluate the prognostic significance of CD44v5 and CD44v6 in resectable colorectal cancer.Materials and methods Membranous CD44v5 and CD44v6 levels were measured by an immunoenzymatic assay in tumors and surrounding mucosal samples obtained from 105 patients with resectable colorectal carcinomas.Results There were no significant differences of CD44v5 levels between tumors [median: 3.2 (range: 0.9–83.5) ng/mg protein) and surrounding mucosal samples (3 (3–146.2) ng/mg protein]. However, tumor samples showed significantly higher CD44v6 levels [19.5 (2.2–562.9) ng/mg protein] than mucosal samples [5 (5–230) ng/mg protein] (P=0.0001). Patients with higher CD44v5 or CD44v6 content in tumor samples had a considerably shorter relapse-free survival (P<0.05, for both). Patients with a higher CD44v6 content also had a shorter relapse-free and overall survival in the multivariate analysis (P<0.05).Conclusion The results of this study suggest a role of CD44v5 and CD44v6 in colorectal cancer progression. Immunotherapy encompasses a variety of interventions and techniques with the common goal of eliciting tumor cell destructive immune responses. Colorectal carcinoma often presents as metastatic disease that impedes curative surgery. Novel strategies such as active immunization with dendritic cells (DCs), gene transfer of cytokines into tumor cells or administration of immunostimulatory monoclonal antibodies (such as anti- CD137 or anti-CTLA-4) have been assessed in preclinical studies and are at an early clinical development stage. Importantly, there is accumulating evidence that chemotherapy and immunotherapy can be combined in the treatment of some cases with colorectal cancer, with synergistic potentiation as a result of antigens cross-presented by dendritic cells and/or elimination of competitor or suppressive T lymphocyte populations (regulatory T-cells). However, genetic and epigenetic unstable carcinoma cells frequently evolve mechanisms of immunoevasion that are the result of either loss of antigen presentation, or an active expression of immunosuppressive substances. Some of these actively immunosuppressive mechanisms are inducible by cytokines that signify the arrival of an effector immune response. For example, induction of 2, 3 indoleamine dioxygenase (IDO) by IFNγ in colorectal carcinoma cells. Combinational and balanced strategies fostering antigen presentation, T-cell costimulation and interference with immune regulatory mechanisms will probably take the stage in translational research in the treatment of colorectal carcinoma. 相似文献

AIM: To detect the expression of COX-2 and HER-2 in colorectal cancer and to analyze their correlation and clinical significance.METHODS: A total of 1026 colorectal cancer surgical specimens were collected from patients treated fromDecember 2002 to December 2007 at the First Affiliated Hospital of Anhui Medical University. All specimens were made into 4-μm slices. The expression of COX-2 and HER-2 were detected by immunohistochemistry using the streptavidin-biotin-peroxidase method. The correlations between COX-2 and HER-2 expression and colorectal cancer clinical features were analyzed.RESULTS: The positive rates of COX-2 and HER-2 expression in colorectal cancer were 77.97%(800/1026) and 46.20%(474/1026), respectively. There was a significant correlation between COX-2 and HER-2 expression in colorectal cancer(P 0.05). In patients with tumor size ≥ 5 cm, the positive rates of COX-2 and HER-2 expression were 81.48%(308/378) and 57.94%(219/378), respectively. In patients with serosal invasion, the positive COX-2 and HER-2 expression rates were 80.53%(612/760) and 49.21%(374/760), respectively. In patients with lymph node metastasis, the positive expression rates were 85.04%(506/595) and 54.62%(325/595), respectively, and the positive expression rates differed significantly between patients with lymph node metastasis and those without(P 0.05). In patients with Duke's C and D colorectal cancer, the positive COX-2 and HER-2 expression rates were 82.80%(443/535) and 57.94%(310/535), respectively. In patients with poorly differentiated colorectal cancer, the positive expression rates were 74.49%(210/282) and 52.84%(149/282), respectively(P 0.05). In patients with distant metastasis, the positive expression rates were 82.27%(116/141) and 53.90%(76/141), respectively(P 0.05). These findings suggest that COX-2 and HER-2 have synergistic effects in colorectal cancer. COX-2 and HER-2 expression had no significant correlation with sex, age, or tumor location. CONCLUSION: COX-2 and HER-2 are important markers for invasion and metastasis of colorectal cancer, and they act together to regulate the invasion and metastasis of colorectal cancer. 相似文献
