兰州地区汉族人群HLA-A、B和DRB1等位基因多态性分析   总被引:1,自引:0,他引:1  
目的分析兰州地区汉族人群HLA-A、B和DRB1位点等位基因多态性特点。方法采用序列特异性引物聚合酶链反应技术对兰州地区200名健康无血缘关系的汉族个体HLA-A、B和DRB1基因座进行分型,并与西北、北方和南方汉族、西北回族、维吾尔族和藏族人群进行比较。结果兰州汉族人群中HLA-A基因座共检出14个等位基因,以A*02,A*11,A*24,A*33,A*30,A*01和A*31基因最常见;HLA—B基因座共检出32个等位基因,以B*40,B*15,B*46,B*13,B*51,B*60,B*58和B*44基因最为常见;HLA-DRB1基因座共检出13个等位基因,最多见的基因依次为DRB1*09.DRB*15,DRB1*12,DRB1*04,DRB1*11,DRB1*07,DRB1*08和DRB1*14,接近北方汉族而与南方汉族有差异,与西北回族无明显差异,但与西北维吾尔族和藏族差异有统计学意义。结论兰州地区汉族人群HLA-A、B和DRB1位点等位基因多态性与南、北汉族人群存在不同程度的差异,与西北维吾尔族和藏族差异显著。  相似文献

山西汉族人群HLA-A、-B、-DRB1基因多态性研究   总被引:5,自引:0,他引:5  
目的 调查山西汉族人群HLA-A、-B、DRB1基因多态性,获得完整准确的遗传学数据。方法 应用聚合酶链反应,序列特异性引物方法对7440名健康、无血缘关系的山西汉族个体进行HLA—A、-B、-DRB1基因型检测,并与不同人群等位基因进行比较。结果 检出A等位基因18个,B等位基因40个,DRB1等位基因13个,其中A*02、A*24、A*11、A*01、A*03、B*13、B*51、B*15、B*40、B*35、DRB1*15、DR*09、DR*1:2、DR*04、DR*07等位基因频率分布较高。结论 山西汉族人群HLA—A,-B,-DRB1基因具有中国北方汉族人群共有的遗传特征,但也有其自身的分布特点。  相似文献

目的:分析青海土族人群HLA-DRB1位点等位基因多态性特点。方法:采用序列特异性引物聚合酶链反应技术,对青海互助地区50名健康无血缘关系的土族个体HLA-DRB1基因座进行分型,并与国内其他地区少数民族人群进行比较。结果:青海土族人群中HLA-DRB基因座共检出16个等位基因,其中DRB1*04、DRB1*08、DRB1*14、DRB1*15、DRB3*、DRB4*基因频率较高;DRB1*06、DRB1*07、DRB1*09、DRB1*13、DRB1*16、DRB1*23基因频率较低。结论:青海土族HLA有独特性。青海土族基因频率与蒙古族有相似之处。  相似文献

目的 分析西藏门巴族HLA-A、B和DRB1 3个基因座的遗传特征,并研究其民族起源。方法应用序列特异性寡核苷酸探针反向斑点杂交技术对西藏自治区门巴族居住区47名门巴族无关个体HLA-A、B和DRB1基因座进行分型。对门巴族和我国其他11个群体(民族)的HLA-DRB1基因频率采用Neighbor-Joining(NJ)方法进行聚类分析。结果 在西藏门巴族人群中HLA-A基因座共检出23个等位基因,其中HLA-A*1101,A*2402,A*02011,A*0206基因最常见;HLA-B基因座共检出39个等位基因,其中HLA-B*3802,B*4001,B*4002,B*51011基因最常见;HLA-DRB1基因座共检出33个等位基因,其中HLA-DRB1*12021,DRB1*0403,DRB1*0701,DRB1*1201基因最常见,频率最高的等位基因分别是HLA-A*1101(0.2128),HLA-B*3802(0.1064)和HLA-DRB1*12021(0.0851)。结论 西藏门巴族人群HLA基因座具有高度遗传多态性,聚类分析门巴族与西藏藏族遗传关系较近。  相似文献

目的:从基因水平调查遵义地区汉族人群HLA-DRB1、DQB1等位基因频率,并了解其多态性分布状况。方法:应用聚合酶链反应-序列特异性引物(PCR-SSP)方法对遵义汉族200名健康个体进行HLA-DRB1、DQB1等位基因分型。结果:遵义地区汉族HLA-DRB1、DQB1等位基因频率分布,在中低分辨水平,共检出13个DRB1基因,7个DQB1基因,DRB1*09、DRB1*08及DQB1*05基因频率相对较高;DRB1*10及DQB1*04基因频率相对较低。与我国北方、南方汉族比较,更接近南方汉族人群。结论:遵义汉族人群HLA-DRB1、DQB1基因具有较为丰富的多态性,其分布符合南方汉族的特点,可能与重庆汉族人群有较为密切的民族融合。  相似文献

目的 分析济南地区汉族人群人类白细胞抗原(human leukocyte antigens,HLA)-A、B、DRB1座位等位基因的高分辨多态性.方法 采用PCR-测序分型(PCR-sequence-based typing,PCR-SBT)对鲁南地区483名无血缘关系的汉族健康个体进行HLA-A、B、DRB1座位高分辨基因分型,采用Arlequin3.5软件计算等位基因频率、单倍型频率,并就常见等位基因与其他人群进行比较.结果 济南汉族HLA-A、B、DRB1座位分别检出27、56和41个等位基因,其中等位基因频率分布最高的分别是A* 11∶01 (0.1615)、B*13∶02(0.1163)和DRB1* 07∶01 (0.1763);最常见的A*-B*-DRB1*单倍型是A*30∶01-B* 13∶02-DRB1* 07∶01 (0.0867).结论 济南地区汉族人群HLA-A、B、DRB1等位基因和单倍型具有较高的多态性.  相似文献

西北地区汉族人群HLA-A、-B、-DRB1基因座单倍型分析   总被引:3,自引:0,他引:3  
目的 分析西北地区汉族群体HLA-A、-B和-DRB1基因座等位基因频率和HIA-A-B、B-DRB1和A-B-DRB1单倍型,获得单倍型频率数据。方法 采用序列特异性寡核苷酸探针反向斑点杂交技术对西北地区62个家系和101个无关个体HLA-A、-B和-DRB1基因座进行基因分型,分析HLA单倍型。结果 在西北地区汉族人群中检出15个HLA-A等位基因,28个HLA-B等位基因,13个HLA-DRB1等位基因,A02、A11、A24、B13、B15、1340、DRB1*04、DRB1*07、DRB1*09和DRB1*15基因频率较高(〉10%),A02(0.3244)、B13(0.1200)和DRB1*15(0.1400)等位基因频率最高。分析得出HLA-A-B、B-DRB1、A-B-DRB1单倍型分别有122、147和278种,83种A-B-DRB1单倍型有至少两条以上相同的单倍型,占总单倍型数的18.44%(83/450)。A30-B13-DRB1*07、A02-B46-DRB1*09、A01-B37-DRB1*10、A24-B15-DRB*15、A02-B46-DRB1*08、A33-B58-DRB1*03是最常见的单倍型。结论 西北地区汉族群体HLA单倍型多态性较为丰富,等位基因频率和单倍型频率数据可用于骨髓移植供者的选择、法医学亲权鉴定以及人类学研究。  相似文献

目的 分析山东省烟台和威海地区汉族人群人类白细胞抗原(human leukocyte antigen,HLA)-A、B、DRB1等位基因的多态性分布特征,并探讨该人群与其他人群的亲缘关系.方法 应用聚合酶链反应-序列特异性寡核苷酸探针方法(polymerase chain reaction-sequence specific olignucleotide probe,PCR-SSOP)对山东省烟台和威海地区4062名无亲缘关系的汉族健康个体进行HLA-A、B、DRB1基因分型.采用Arlequin3.5软件计算HLA等位基因频率、单倍型频率和连锁不平衡参数,按内氏公式计算出不同人群之间的遗传距离,并利用Mega5.0软件构建系统发生树.结果 该人群HLA-A、B、DRB1等位基因分布均符合Hardy-Weinberg平衡(P>0.1).3个基因座分别检出18、33和13个等位基因,其中等位基因频率分布最高的分别是A* 02 (0.2935)、B* 15 (0.1485)和DRB1* 15 (0.1621);最常见的单倍型为A* 30-B* 13-DRB1* 07(0.0649),A* 33-B* 58、A*66-DRB1* 13、B*08-DRB1* 03呈现最强的连锁不平衡;山东省烟台和威海汉族人群与吉林省汉族人群遗传距离最小,为0.0034.结论 山东省烟台和威海地区汉族人群HLA-A、B、DRB1等位基因和单倍型具有较高的遗传多态性,该人群与吉林汉族人群亲缘关系最近.  相似文献

云南昆明彝族和汉族儿童HLA-DRB1等位基因的多态性研究   总被引:1,自引:0,他引:1  
目的研究昆明彝族和汉族儿童HLA-DRB1基因的多态性,探讨其在昆明彝族和汉族人群中的遗传特征。方法应用PCR-SSP基因分型技术,对云南昆明地区70名彝族和72名汉族健康儿童进行了HLA-DRB1位点的基因分型。结果昆明彝族儿童HLA-DRB1位点共检出了12种等位基因,其中以HLA-DRB1*12(33.57%)、DRB1*0901(11.43%)、DRB1*04(11.43%)较常见,其它基因频率大于5%的等位基因还有HLA-DRB1*01(8.57%)、DRB1*11(7.86%)、DRB1*14(7.14%)、DRB1*15(7.14%)、DRB1*08(5%);昆明汉族儿童HLA-DRB1位点共检出了12种等位基因,其中以HLA-DRB1*12(20.14%)、DRB1*0901(19.44%)、DRB1*04(18.06%)较常见,其他基因频率大于10%的等位基因还有HLA-DRB1*08(11.11%)、DRB1*15(10.42%);与北方汉族人群、南方汉族人群HLA-DRB1等位基因分布进行了比较,均有显著性差异(P<0.001)。结论昆明彝族和汉族HLA基因多态性分布有其特点,他们既不同于北方汉族人群也不同南方汉族人群,有其独特性。可能与复杂的民族迁移历史和民族融合及云南独特的地理环境有关。  相似文献

雷公藤甲素(triptolide,TPL)是从中草药雷公藤中提取的一种有效活性物质,已被用来治疗多种疾病,包括系统性红斑狼疮,类风湿性关节炎,肾病综合征等,TPL甚至有很强的抑制肿瘤的活性。近些年的研究显示,TPL具有抗血管新生的能力,TPL不仅可以抑制肿瘤的增殖,诱导细胞的凋亡,还可以抑制肿瘤的转移,可以增加其它化疗药物的抗肿瘤活性。本综述将讨论TPL在抗肿瘤血管新生方面的研究进展,以及初步探讨其潜在的作用机制。  相似文献

Total activities of neutral proteases in the cerebral, hepatic, and myocardial tissues of ground squirrel vary during hibernation: in autumn (before hibernation) activities of the enzymes in the brain and myocardium start increasing, while in the liver they do not change. A common feature for all tissues is minimum activity of active neutral proteases in the middle of hibernation month 1 bout, while the maximum activity is recorded before awakening. The optimal age for measles vaccination is an important health issue, since maternal antibodies may neutralize the vaccine antigen before a specific immune response develops, while delaying vaccination may increase the risk of complicated diseases in infants. However, measles vaccination impacts the duration of protection afforded by transplacental transfer of maternal antibodies: vaccination-induced maternal antibodies disappear faster than disease-induced antibodies. In order to maintain protection against measles in infants, it is important to monitor the dynamics of this phenomenon in vaccinated populations. To assess the current situation in France, a multicenter, prospective seroepidemiological study was conducted in seven French hospitals between October 2005 and January 2007. Maternal measles antibody concentrations from 348 infants 0 to 15 months old were measured using the plaque reduction neutralization assay. Geometric mean concentrations and the percentage of infants with maternal measles antibody concentrations above the protection threshold (≥120 mIU/ml) were assessed according to age. Results show that after more than 20 years of routine measles vaccination in France, maternal measles-neutralizing antibodies decrease dramatically in French infants by 6 months of age, from 1,740 mIU/ml for infants 0 to 1 month old to 223 mIU/ml for infants 5 to 6 months old, and that 90% of infants are not protected against measles after 6 months of age. Infant protection against measles could be optimized both by increasing herd immunity through an increased vaccine coverage and by lowering the age of routine vaccination from 12 to 9 months.  相似文献

In this study, the effect of endosulfan, an organochlorine pesticide, and the ameliorating effect of vitamin C on the livers of New Zealand white rabbits were studied. Livers of the rabbits were examined grossly and histopathologically, and caspase-3 activity was detected by immunohistochemical methods. A total of twenty-four rabbits were divided into four groups (n=6). Rabbits in Group I (END) were daily given a sublethal dose of endosulfan (1 mg/kg bw) in corn oil by oral gavage for 6 weeks. Group II (END+C) received the same dose of endosulfan and additionally Vit C (20 mg/kg bw) every other day during this period. Group III (OIL+C) received corn oil daily by oral gavage and vitamin C every other day for 6 weeks. Group IV (OIL), the control group, received only corn oil daily, by oral gavage throughout the experiment. The concentration of α-endosulfan in the END group was higher in livers (0.102±0.012 ppb) than the β-endosulfan (0.072±0.001 ppb). Decreased accumulation of α and β endosulfan was observed in the END+C group (0.025±0.003 and 0.016±0.002 ppb, respectively) (p<0.0001). The most prominent gross findings at the necropsy were seen in the END group, in which swollen and pale livers were commonly observed. Hemorrhages, degenerations, necrosis, and in some rabbits bile duct hyperplasia were the marked histopathological findings of the END group. Caspase-3 positive reaction was more severe in this group than in the others. An ameliorating effect of Vit C on gross, histopathological and immunohistochemical findings was observed in the END+C group. The results revealed that endosulfan is highly toxic for rabbit livers. However, toxicity was decreased by Vit C treatment, which reduced the accumulation of endosulfan in livers four-fold.  相似文献
